Observing how an action is done by others allows the observer to understand the cognitive and emotion state of the agent. This information, carried by the kinematics of the observed action, has been defined by Daniel Stern “vitality forms”. The expression and the capacity to understand the vitality forms is already present in infants, a finding indicating their importance for the development of social attunement. It has been proposed that, well before developing linguistic abilities, infants are actively engaged in non-verbal exchanges with their caregivers. This ability denotes a primordial way to relate to and understand others and presumably represents a constitutive element of interpersonal relations, namely intersubjectivity. In the present neuroimaging (fMRI) study we presented participants with videos showing hand actions performed with different velocities and asked them to judge their vitality form (gentle, neutral, rude) or their velocity (slow, medium, fast). Previous studies showed that the dorso-central insula is selectively active both during vitality form observation and execution. The aim of the present study was to assess, using multi-voxel pattern analysis (MVPA), whether in the insula there are voxels discriminating vitality form from velocity. Results showed that, consistently across subjects, in the dorso-central sector of the insula there are voxels selectively tuned to vitality forms. Supporting previous findings, these results confirm that the dorso-central insula is involved in processing the vitality forms of an action, both when carryied out in the first person and when observed in other individuals. This supports the idea that the understanding of others' behavior in terms of affective content is mediated by an automatic activation system that allows the recipient to tune in and respond to another individual's emotional state without necessarily having "formal" knowledge of what is being observed. Knowledge about upcoming actions is fundamentally important to anticipate rather than react to the actions of other individuals. Influential theoretical models suggest that the human motor system is “designed” to be an anticipation device and that it predicts what others are doing by implementing an inner motor simulation process. Multimodal, perceptuomotor, multiple-duty cells (mirror neurons) may also play an important function in action anticipation. Data indicate that the superior predictive ability of individuals who achieve excellence in a given cognitive-motor domain is inherently linked to a modulatory, fine-tuning activity that expertise exerts on somatosensory and motor systems. Phenomena and neural correlates of the anticipatory action mapping in elite athletes and expert pianists are reviewed, providing support for the hypothesis that one’s own sensorimotor representations are used for the predictive readout of actions and intentions of others. In addition, the possibly crucial role of predictive coding in social motor interactions is highlighted.

Children with autism spectrum disorders (ASD) and children with Williams syndrome (WS) show divergent social phenotypes, but also several similarities in their socio-cognitive deficits. Cross-syndrome direct comparisons could lead to a better understanding of mechanisms that determine deficits in social cognition in the two syndromes. A fundamental factor for social cognition is the ability to understand and predict others' actions (e.g. what action is being done and why it is being done when observing a goal-related act). Here we compared the understanding of others' actions in children with ASD, WS and in children with typical development. Comprehension of what motor act was being done and of why it was being done was assessed with or without contextual cueing using a computer-based task. The results showed that what understanding was impaired in the WS group, but not in the ASD group, which showed mental-age appropriate performance. While there have been several studies investigating the neural correlates of action observation associated with hand grasping movements, comparatively little is known about the neural bases of observation of reaching movements. In two experiments, using functional magnetic resonance imaging (fMRI), we defined the cortical areas encoding reaching movements and assessed their sensitivity to biological motion and to movement velocity. In the first experiment, participants observed video-clips showing either a biological effector (an arm) or a non-biological object (rolling cylinder) reaching toward a target with a biological and a non-biological motion, respectively. In the second experiment, participants observed video-clips showing either a biological effector (an arm) or a non-biological object (an arrow) reaching toward a target with the same biological motion profiles. The results of the two experiments revealed activation of superior parietal and dorsal premotor sites during observation of the biological motion only, independent of whether it was performed by a biological effector (reaching arm) or a non-biological object (reaching arrow). These areas were not activated when participants observed the non-biological movement (rolling cylinder). To assess the responsiveness of parietal and frontal sites to movement velocity, the fMRI repetition-suppression (RS) technique was used, in which movement was shown with same or different velocities between consecutive videos, and observation of identical stimuli was contrasted with observation of different stimuli. Regions of interest were defined in the parietal and frontal cortices, and their response to stimulus repetition was analyzed (same vs. different velocities). The results showed an RS effect for velocity only during the observation of movements performed by the biological effector and not by the non-biological object. In a previous functional magnetic resonance imaging (fMRI) study, where we investigated the neural correlates of esthetic experience, we found that observing canonical sculptures, relative to sculptures whose proportions had been modified, produced the activation of a network that included the lateral occipital gyrus, precuneus, prefrontal areas, and, most interestingly, the right anterior insula. We interpreted this latter activation as the neural signature underpinning hedonic response during esthetic experience. With the aim of exploring whether this specific hedonic response is also present during the observation of non-art biological stimuli, in the present fMRI study we compared the activations associated with viewing masterpieces of classical sculpture with those produced by the observation of pictures of young athletes. The two stimulus-categories were matched on various factors, including body postures, proportion, and expressed dynamism. The stimuli were presented in two conditions: observation and esthetic judgment. The two stimulus-categories produced a rather similar global activation pattern. Direct comparisons between sculpture and real-body images revealed, however, relevant differences, among which the activation of right antero-dorsal insula during sculptures viewing only. Along with our previous data, this finding suggests that the hedonic state associated with activation of right dorsal anterior insula underpins esthetic experience for artworks.

Observing actions made by others activates the cortical circuits responsible for the planning and execution of those same actions. This observation-execution matching system (mirror-neuron system) is thought to play an important role in the understanding of actions made by others. In an fMRI experiment, we tested whether this system also becomes active during the processing of action-related sentences. Participants listened to sentences describing actions performed with the mouth, the hand, or the leg. Abstract sentences of comparable syntactic structure were used as control stimuli. The results showed that listening to action-related sentences activates a left fronto-parieto-temporal network that includes the pars opercularis of the inferior frontal gyrus (Broca's area), those sectors of the premotor cortex where the actions described are motorically coded, as well as the inferior parietal lobule, the intraparietal sulcus, and the posterior middle temporal gyrus. These data provide the first direct evidence that listening to sentences that describe actions engages the visuomotor circuits which subserve action execution and observation.

Positron emission tomography (PET) was used to localize brain regions that are active during the observation of grasping movements. Normal, right-handed subjects were tested under three conditions. In the first, they observed grasping movements of common objects performed by the experimenter. In the second, they reached and grasped the same objects. These two conditions were compared with a third condition consisting of object observation. On the basis of monkey data, it was hypothesized that during grasping observation, activations should be present in the region of the superior temporal sulcus (STS) and in inferior area 6. The findings in humans demonstrated that grasp observation significantly activates the cortex of the middle temporal gyrus including that of the adjacent superior temporal sulcus (Brodmann's area 21) and the caudal part of the left inferior frontal gyrus (Brodmann's area 45). The possible functional homologies between these areas and the monkey STS region and frontal area F5 are discussed.

Regional cerebral blood flow was measured using positron emission tomography (PET) in normal subjects while performing simple aimless proximal and distal arm movements. The aim of the experiment was to compare the somatotopic organization of precentral and mesial (the so called supplementary motor area, SMA) motor cortices and to evaluate whether in man, as in the monkey, the rostral and caudal sectors of SMA are functionally different. The results showed that proximal and distal arm movements are to a large extent segregated in the precentral motor cortex, but not in the SMA.
