Chromatin-based epigenetic memory relies on the accurate distribution of parental histone H3-H4 tetramers to newly replicated DNA strands. Mcm2, a subunit of the replicative helicase, and Dpb3/4, subunits of DNA polymerase ε, govern parental histone H3-H4 deposition to the lagging and leading strands, respectively. However, their contribution to epigenetic inheritance remains controversial. Here, using fission yeast heterochromatin inheritance systems that eliminate interference from initiation pathways, we show that a Mcm2 histone binding mutation severely disrupts heterochromatin inheritance, while mutations in Dpb3/4 cause only moderate defects. Surprisingly, simultaneous mutations of Mcm2 and Dpb3/4 stabilize heterochromatin inheritance. eSPAN (enrichment and sequencing of protein-associated nascent DNA) analyses confirmed the conservation of Mcm2 and Dpb3/4 functions in parental histone H3-H4 segregation, with their combined absence showing a more symmetric distribution of parental histone H3-H4 than either single mutation alone. Furthermore, the FACT histone chaperone regulates parental histone transfer to both strands and collaborates with Mcm2 and Dpb3/4 to maintain parental histone H3-H4 density and faithful heterochromatin inheritance. These results underscore the importance of both symmetric distribution of parental histones and their density at daughter strands for epigenetic inheritance and unveil distinctive properties of parental histone chaperones during DNA replication.

Heavy metal contamination has been regarded as an environmental variable that affects the efficiency of pest biological control, but the parasitic fitness of parasitoids under heavy metal stress is poorly understood. Herein, the effect of Cd exposure through the host pupa of Hyphantria cunea on the parasitic fitness of Chouioia cunea was investigated, and the mechanism by which Cd exposure affects the interaction between H. cunea and C. cunea from the perspective of innate immunity in host insect and the oxidative status in the parasitoid offspring was explored. Our results indicated that Cd can be transferred from the H. cunea pupae to the parasitoid offspring, and the transfer coefficient reflected biological amplification. There were no significant differences in the rates of parasitism success and offspring emergence between the untreated and Cd-treated groups. However, after parasitizing Cd-accumulated pupae, the parasitic fitness of offspring wasps (e.g., the number, individual size and life span) decreased significantly. Under Cd exposure, the cellular and humoral immunity of H. cunea pupae decreased significantly. Compared with the untreated group, the H2O2 content of parasitoid offspring in the Cd-treated group was significantly increased. Cd exposure significantly inhibited superoxide dismutase activity in parasitoid offspring, but the contents of ascorbic acid and glutathione were significantly increased by Cd stress. Taken together, these results indicate that Cd exposure reduces the cyclic utilization efficiency of C. cunea on H. cunea pupae. The oxidative status of parasitoid offspring triggered by Cd exposure could be responsible for the reduced parasitic fitness of C. cunea on Cd-accumulated H. cunea pupae.

The oriental fruit fly, Bactrocera dorsalis (Hendel), is a devastating pest of citrus fruits. After successful mating, adult females insert their eggs into the ripened fruit, resulting in moldy and rotten fruit and causing great economic losses for the citrus industry. In the field, flies initiate copulatory behaviors as twilight approaches, and decreasing light intensity in this period is the normal stimulus for copulation. In this study, ten light intensities ranging from 0–30 000 lux were set to identify the typical intensity that strongly regulates the copulation behavior of B. dorsalis. Three light intensities found to regulate the copulation behavior were then selected to verify their effects on adult male wing fanning and female chemotaxis towards 2,3,5-trimethylpyrazine (TMP). At last, strong light and complete darkness were artificially combined in the lab to verify whether they could prevent copulation to inform behavioral manipulation of oriental flies in the future. Objective： Optimized the drying process of jujube slices and improved the product quality. Methods： Taking jujube slices as the research object, the correlation between conversion moisture content, infrared temperature and slice thickness and drying time and drying rate was studied, and the change rule of effective moisture diffusion coefficient of jujube slices in FD-IRD with conversion moisture content, infrared temperature and slice thickness was calculated, and the drying activation energy of jujube slices in FD-IRD was calculated according to the test data. Results： The lower the conversion moisture content is, the shorter the infrared drying time was. But the lower the conversion moisture content would greatly increase the freeze drying time. Proper increase of infrared drying temperature was beneficial to increase the effective diffusion coefficient of water. The thinner the slice is, the higher the drying rate was. Reducing the slice thickness can improve the effective water diffusion coefficient and shorten the drying time. The front and rear two stages were both speeds reducing drying processes. The effective moisture diffusion coefficients of freeze-drying and infrared drying under different drying conditions by solving Fick's second law were 3.39×10-9~9.47×10-9 and 3.34×10-9~2.01×10-8 m2/s, respectively; The drying activation energy of the infrared drying stage was 59.03 kJ/mol calculated by Anius formula. Conclusion： The conversion moisture content is 30%, and the infrared temperature is 60 ℃. Under the condition of slice thickness of 6 mm, the freeze infrared combined drying technology has short drying time and high efficiency.

Oncogenic histone lysine-to-methionine mutations block the methylation of their corresponding lysine residues on wild-type histones. One attractive model is that these mutations sequester histone methyltransferases, but genome-wide studies show that mutant histones and histone methyltransferases often do not colocalize. Using chromatin immunoprecipitation sequencing (ChIP-seq), here, we show that, in fission yeast, even though H3K9M-containing nucleosomes are broadly distributed across the genome, the histone H3K9 methyltransferase Clr4 is mainly sequestered at pericentric repeats. This selective sequestration of Clr4 depends not only on H3K9M but also on H3K14 ubiquitylation (H3K14ub), a modification deposited by a Clr4-associated E3 ubiquitin ligase complex. In vitro, H3K14ub synergizes with H3K9M to interact with Clr4 and potentiates the inhibitory effects of H3K9M on Clr4 enzymatic activity. Moreover, binding kinetics show that H3K14ub overcomes the Clr4 aversion to H3K9M and reduces its dissociation. The selective sequestration model reconciles previous discrepancies and demonstrates the importance of protein-interaction kinetics in regulating biological processes.

This work describes a strategy to employ complex carbides for high-yield microalloyed 4Mn steels processed by quenching and reversion (Q&R). The nano/microstructural details are investigated by using electron microscopy and atom probe. It is found that complex carbides are classified into two types and contribute to yield strength in different ways. First, coarse carbides (MC1), which are retained after austenization and quenching, provide the Zener pinning effect and contribute to strengthening by microstructure refinement and precipitation hardening. Second, nanoscale carbides (MC2), which precipitate in martensite during austenite reversion, provide precipitation hardening. Furthermore, the precipitation of MC1 accelerates austenite reversion due to the finer martensite microstructure. Hence, the strengthening effects and TRIP effect can be tuned by controlling the proportion of the two types of carbide. This concept is validated in Ti-Mo 4Mn steel, which exhibits a high yield strength of about 850 MPa, a high ultimate tensile strength of about 950 MPa, and excellent elongation of over 20%. The complex carbide strategy is able to increase yield strength without sacrificing much ductility for Q&R medium-Mn steels.

‘Candidatus Liberibacter asiaticus (CLas)’, which causes citrus Huanglongbing (HLB) disease, has not been successfully cultured in vitro to date. Here, a rapid multiplication system for CLas was established through in vitro regeneration of axillary buds from CLas-infected ‘Changyecheng’ sweet orange (Citrus sinensis Osbeck). Stem segments with a single axillary bud were cultured in vitro to allow CLas to multiply in the regenerating axillary buds. A high CLas titer was detected in the regenerated shoots on an optimized medium at 30 days after germination (DAG). This titer was 28.2-fold higher than in the midribs from CLas-infected trees growing in the greenhouse. To minimize contamination during in vitro regeneration, CLas-infected axillary buds were micrografted onto seedlings of ‘Changyecheng’ sweet orange and cultured in a liquid medium. In this culture, the titers of CLas in regenerated shoots rapidly increased from 7.5×104 to 1.4×108 cells μg–1 of citrus DNA during the first 40 DAG. The percentages of shoots with >1×108 CLas cells μg–1 DNA were 30 and 40% at 30 and 40 DAG, respectively. Direct tissue blot immunoassay (DTBIA) indicated that the distribution of CLas was much more uniform in regenerated plantlets than in CLas-infected trees growing in the greenhouse. The disease symptoms in the plantlets were die-back, stunted growth, leaf necrosis/yellowing, and defoliation. The death rate of the plantlets was 82.0% at 60 DAG. Our results show that CLas can effectively multiply in citrus plantlests in vitro. This method will be useful for studying plant-HLB interactions and for rapid screening of therapeutic compounds against CLas in citrus.

Heortia vitessoides Moore, 1885 (Lepidoptera: Crambidae) is an economically important lepidopteran pest that caused severe damage to the plantation area of Aquilaria sinensis (Lour.) Gilg, 1825 (Thymelaeaceae), resulting in extensive defoliation of the trees during an epidemic. In this study, we used scanning electron microscopy (SEM) to analyze the external morphology and ultrastructure of sensilla on various body parts of H. vitessoides. Specifically, seven, four, four, and five types of sensilla were found, respectively, on the antennae, proboscis, labial palps, and legs. We described the types, distributions, and sexual dimorphism of these sensilla on antennae, and found that the number and size of sensilla differed significantly between males and females. This study provides crucial information for future investigations into the function of these sensilla in H. vitessoides.

Deep learning is a promising solution for computer vision at present. To solve the computer vision problem, it requires massive and high-quality image training datasets. Collecting and accurately labeling image datasets is a very time-consuming and expensive process. As computer vision applications become more widespread, it makes this problem even more pronounced. Image augmentation technologies are technical methods to effectively solve the problem of deep learning training under the condition of small-scale or low-quality training data. These technologies are continually accompanied with the development of deep learning and computer vision. This paper first reviews these image augmentation researches from the perspective of augmentation objects, operation spaces, label processing methods, and augmentation strategies and then concludes corresponding paradigms of current image data augmentation methods. After that, this paper proposes a taxonomy for current image data augmentation guided by the above paradigms, and reviews corresponding representative methods of each image data augmentation category. Finally, this paper makes conclusions on existing image data augmentation, points out the problems existing in the current image augmentation research and presents promising directions for future research.

Abstract Prostate cancer is a major cause of death in males. Cyproterone acetate (CPA), the steroidal anti-androgen for part of androgen deprivation therapy, may block the androgen-receptor interaction and then reduce serum testosterone through its weak anti-gonadotropic action. In addition to CPA inducing hepatitis, CPA is known to cause liver tumors in rats also. Aryl hydrocarbon receptor (AhR) is a cytoplasmic receptor and regulates multiple physiological functions. CYP1A1 is an AhR-targeted gene. We found that CPA induced CYP1A1 expression, transcriptional activity of the aryl hydrocarbon response element (AHRE), and the nuclear localization of AhR in mouse Hepa-1c1c7 cells. However, CPA suppressed CYP1A1 mRNA expression and the transcriptional activity of AHRE in human HepG2 and MCF7 cells, and also decreased AhR ligand-induced CYP1A1 protein expression and transcriptional activity of AHRE in HepG2 cells. In summary, CPA is an AhR agonist in mouse cells, but an AhR antagonist in human cells. Accordingly, CPA potentially plays a role as an endocrine disruptor of the AhR. This study helps us to understand why CPA induces acute hepatitis, gene mutation, and many other side effects. In addition, it may trigger further studies investigating the relationships between CPA, glucocorticoid receptor and castration-resistant prostate cancer in the future.

To study the imaging features of leukoaraiosis (LA) and hemorrhage in cerebral amyloid angiopathy (CAA) patients.

IntroductionIntegrating training on health equity of sexual and gender minorities (SGM) in medical education has been challenging globally despite emphasis on the need for medical students to develop competence to provide adequate care for diverse patient groups. This study elicits Taiwanese medical students' perceptions of their values and preparedness to care for Lesbian, Gay, Bisexual, or Transgender (LGBT) patients using a qualitative approach that considers broader societal changes, and more focused topics such as the provision of relevant training in medical education.MethodsEighty-nine medical students/trainees from two southern Taiwanese medical schools (one public and one private) participated in focus groups (n = 70) and individual interviews (n = 19). Qualitative analysis was conducted using inductive thematic analysis.ResultsParticipants (i) expressed wide social acceptance and openness toward LGBT individuals, but were unsure of ways to communicate with LGBT patients; (ii) confirmed that stigmatization and biases might be developed during their training; (iii) recognized gender stereotypes could have negative impacts on clinical reasoning; (iv) considered themselves prepared to care for LGBT patients, yet equated non-discriminatory attitudes to preparedness; (v) acknowledged a lack of relevant professional skills; (vi) implicated curriculum did not address LGBT issues systematically and explicitly.ConclusionThis study has identified the insufficiencies of current medical training and inadequate preparedness of medical students/trainees to provide better care for LGBT patients. It provides insights for medical educators to design and implement effective medical curriculum and training, and faculty development programs to equip medical students/trainees with self-awareness and competencies to more readily provide holistic care for SGM, in keeping up with social progress, and promote health equity for a more diverse patient population.

鑑於金融機構之經濟重大影響性，本文自金融機構角度切入，介紹揭弊者保護理論及相關規範，以凸顯我國揭弊者保護仍為不足之現況，並援引美國法案例與制度，以對我國法務部廉政署研擬，並於 2020 年 4 月 8 日經立委提案之「公益揭弊者保護法草案」做一檢視。本文依據揭弊者與其他行為人間之身分關係，將揭弊者劃分為不同類型，因應各類型揭弊者所面臨之困境，提出相應之保護措施，並以金融機構之產業特性及現存弊病，說明金融機構中揭弊者保護制度建立之必要性。由於我國揭弊者保護相關法制闕漏，且美國法之金融機構吹哨者保護相關法制齊備，如沙賓法Sarbanes-Oxley Act）、華爾街改革與消費者保護法（Dodd-Frank Wall Street Reform and Consumer Protection Act）即詳盡規範相關保護、獎勵措施，且相關案例繁多可供借鑑，因此本文援引美國法案例及制度，以輔助檢視揭弊者保護法草案，以期完善揭弊者保護制度，並對金融機構及主管機關於揭弊者法制建構後，提出相關應行作為建議。 This study focuses on the whistleblower protection policy in Taiwan and compares Taiwan’s policy with the U.S. laws. The financial industry in U.S. has a long history and developed successfully, which could provide Taiwan’s legal reform a reference model. In Taiwan, whistleblower bears unfair burdens and culture stigma. Lacking of sufficient protection forces whistleblower to take risks of personal safety and retaliation in workplace. On the contrast, the U.S. legal system provides whistleblower protection comprehensively including identity’s confidentiality, prevention from workplace retaliation and legal remedies. Corporations are required to establish compliant system and protection mechanism. Due to the insufficiency of current whistleblower protection policy, Taiwan’s Agency Against Corruption at Ministry of Justice proposed Whistleblower Protection Act in the end of 2018. The bill in now under the review at the Congress. This article will analyze and give comments to the bill as well. Hopefully, Taiwan could own its whistleblower protection law as soon as possible.
