Aiming to resolve the problem where the reliability of gearbox bearings of wind turbines is easily affected by random impact, this paper puts forward a reliability evaluation method for the competitive failure of gearbox bearings of wind turbines considering the fault incubation point. Firstly, we use Weibull distribution to simulate the fault latent process of wind turbine gearbox bearings. Secondly, a natural degradation model of gearbox bearing is established based on the Wiener process. Then, we model the random impact arrival frequency and impact intensity through a homogeneous Poisson process and normal distribution, respectively. Finally, based on considering the fault incubation point, the natural degradation of the bearing, the instantaneous degradation caused by impact, and the decline in the impact resistance of the bearing, a reliability evaluation model of gearbox bearings of wind turbines is established. A high-speed bearing of a gearbox from a wind farm in northern China is selected for simulation analysis. The results show that the proposed method can better describe the reliability decline process of the gearbox bearings of wind turbines, which has a specific guiding significance for the maintenance of wind turbines.

Purpose. Acupuncture and moxibustion techniques have been increasingly used to treat peripheral neuropathic pain (PNP). However, there is a paucity of comparative information and cost-effectiveness assessment for techniques on PNP management. Patients and Methods. Randomized controlled trials studying the acupuncture or moxibustion treatments on PNP were identified from electronic databases. The quality of the included studies and the potential risk of bias was evaluated using the ROB 2.0 assessment tool. The primary outcome was at least 20% pain relief. The treatment effects were pooled through a frequentist-based network meta approach. Subsequently, the cost-effectiveness measured by incremental cost per additional responder (ICPR) was calculated. Results. One three-arm trial and 15 two-arm trials comprising 1308 participants that satisfy the eligibility criteria were identified. Among the included studies, 12.5% were at low risk of bias, 68.75% had some concerns about the risk of bias, and 18.75% were at high risk of bias. The major sources of bias originated from the randomization processes of the studies. The patients were assigned to seven different acupuncture or moxibustion interventions and two pharmaceutical treatments. Except for acupoint injection, all the included acupuncture and moxibustion techniques showed superior improvements in PNP and were more cost-effective as compared to pharmaceutical treatments. Warm needling, fire needling, and moxibustion were the most effective treatments. Fire needling showed the lowest ICPR relative to the nonsteroidal anti-inflammatory drugs in the cost-effectiveness analysis of direct and indirect costs. Conclusion. Acupuncture and moxibustion techniques are beneficial and cost-effective approaches for easing PNP and hence can be considered for PNP management.

Peptide-based therapeutics are natural candidates to desirable wound healing. However, enzymatic surroundings largely limit its stability and bioavailability. Here, we developed a tetrahedral framework nucleic acids(tFNA)-based peptide delivery system, that is, p@tFNAs, to address deficiencies of healing peptide stability and intracellular delivery in diabetic wound healing.AGEs (advanced glycation end products) were used to treat endothelial cell to simulate cell injury in diabetic microenvironment. The effects and related mechanisms of p@tFNAs on endothelial cell proliferation, migration, angiogenesis and ROS (reactive oxygen species) production have been comprehensively studied. The wound healing model in diabetic mice was photographically and histologically investigated in vivo.Efficient delivery of healing peptide by the framework(tFNA) was verified. p@tFNAs helped overcome the angiogenic obstacles induced by AGEs via ERK1/2 phosphorylation. In the meantime, p@tFNA exhibited its antioxidative property to achieve ROS balance. As a result, p@tFNA improved angiogenesis and diabetic wound healing in vitro and in vivo.Our findings demonstrate that p@tFNA could be a novel therapeutic strategy for diabetic wound healing. Moreover, a new method for intracellular delivery of peptides was also constructed.

Introduction: Nowadays, it is indicated that miRNA was anomaly expressed in tumour. Previous studies have shown that miRNAs can regulate the proliferation, invasion, and migration of cancer cell-related processes. Meanwhile, current investigations show that RAB23 also plays an important role in cancer cell-related processes. But the potential mechanism remains unclear. Material and methods: SW579 cells were selected and transfected with miR-597-3p mimics. Then the expression of miR-597-3p and RAB23 were measured by quantitative real-time polymerase chain reaction (PCR) and western blotting, respectively. Subsequently, the abilities of proliferation, invasion, and migration of SW579 cells were researched. For further study, the Luciferase reporter assay proved that miR-597-3p could target the expression of RAB23, and the proteins of invasion and migration were also measured to clear the mechanism. Results: After being transfected with miR-597-3p mimics, the expression of miR-597-3p was remarkably increased and RAB23 was significantly decreased. The abilities of proliferation, invasion, and migration also decreased significantly. The miRTarase Database predicated and Luciferase reporter assay proved that RAB23 was the target gene of miR-597-3p. The expression of matrix metalloproteinase (MMP)-2, MMP-9, and N-cadherin was down-regulated, and the expression of E-cadherin was up-regulated. Conclusion: miR-597-3p could reduce the proliferation, invasion and migration abilities of SW579 cells, which may be related to the targeted inhibition of RAB23 expression and down-regulation of the expression levels of MMP-2, MMP-9 and N-cadherin proteins of SW579 cells.

Fibroblast-like synoviocytes (FLSs) are key effector cells in the pathogenesis of rheumatoid arthritis (RA) and display a unique aggressive tumor-like phenotype with remarkable hyperplasia, increased cell migration and invasion. How FLSs undergo these changes in RA remains unknown. We previously reported a novel function of transcription factor SOX5 in RA-FLSs that promote cell migration and invasion. In this study, we found that miR-15a/16 directly targets the SOX5 3'UTR and suppresses SOX5 expression. Moreover, miR-15a/16 is significantly down-regulated in RA-FLSs, which negatively correlates with SOX5 expression. Transfection with miR-15a/16 mimics in RA-FLSs inhibits cell migration, invasion, IL-1β and TNFα expression. Overexpression SOX5 in RA-FLSs decreases miR-15a/16 expression and rescues miR-15a/16-mediated inhibitory effect. Furthermore, RA patients with the lower baseline serum miR-15a/16 level present poor response of 3 months disease-modifying antirheumatic drugs (DMARDs) therapy. Collectively, this study reveals that miR-15a/16/SOX5 axis functions as a key driver of RA-FLSs invasion, migration and inflammatory response in a mutual negative feedback loop and correlates with DMARDs treatment response in RA.

Papillary thyroid cancer (PTC) is a frequent thyroid malignancy. With the significant regulatory role in tumor progression, more attention has been employed to investigate mechanism of long noncoding RNAs (lncRNAs) in progression of PTC. We prospectively explored the mechanism whereby lncRNA SET‐binding factor 2‐antisense RNA1 (SBF2‐AS1) is implicated in pathogenesis of PTC. First, differentially expressed SBF2‐AS1 between PTC and normal adjacent thyroid tissues was determined, and result indicated a higher SBF2‐AS1 expression in PTC tissues than adjacent normal tissues. Moreover, highly SBF2‐AS1 expression predicted a poor prognosis in PTC patients. Second, SBF2‐AS1 overexpression promoted cell viability and cycle of PTC, while inhibited cell apoptosis. However, SBF2‐AS1 downregulation reduced viability and cycle, while promoted cell apoptosis. Moreover, SBF2‐AS1 could bind with miR‐431‐5p and showed negative correlation with miR‐431‐5p in PTC patients. Furthermore, miR‐431‐5p bind with cyclin‐dependent kinase (CDK) 14 and showed negative correlation with CDK14 in PTC patients. Finally, overexpression of CDK14 counteracted with the inhibitory role of SBF2‐AS1 downregulation on cell viability, cycle, and apoptosis of PTC. In conclusion, SBF2‐AS1 exhibited oncogenic property in PTC, and knockdown of SBF2‐AS1 could be a therapeutic strategy for PTC.

In a motor control system, the parameters tuning of speed and position controller depend on the value of the moment of inertia. A new moment of inertia identification scheme for permanent magnet motor system was proposed in this paper. This is an extension of the existing acceleration deceleration methods, which solves the large moment of inertia identification error caused by variable angular acceleration, large calculation error of inertia torque, and large measurement noise in the acceleration process. Based on the fact that the angular acceleration is not constant and the sampling signal is noisy, the integral chain differentiator was used to calculate the instantaneous angular acceleration at any time and suppress the sampling signal noise at the same time. The error function with instantaneous angular acceleration and inertia torque as parameters was designed to estimate the moment of inertia. In order to calculate the inertia torque accurately, viscous friction torque was considered in the calculation of inertia torque, and Kalman filter was used to estimate the total load torque to solve the problem of under rank of motor motion equation. Simulation and experimental results showed that the proposed method could effectively identify the moment of inertia in both noisy and noiseless environments.

Our study explored the effects of parental social integration on migrant children’s vaccination status in China. Using data obtained from the 2014 China Migrants Dynamic Survey, a total of 4915 participants were included in this study. Social integration was measured by economic, social, cultural, and internal identity. Univariate chi-square testing was used to calculate associations between all variables and migrant children’s vaccination status. Binary logistic regression was employed to calculate the impacts of social integration on migrant children’s vaccination status. In total, 94.7% of migrant children had complete vaccinations for their age. Migrants who had medical insurance, spoke the native language when communicating with locals, lived mainly with locals, and did not perceive discrimination were more likely to have their children completely vaccinated. Social integration was positively associated with migrant children’s vaccination status. Our study indicated that to improve vaccination coverage of migrant children, more policy support for migrant employment and housing, promotion of health services for migrants, and language support in health institutions is needed.

Neuroblastoma (NB) is an rare type of tumor that almost affects children age 5 or younger due to its rapid proliferation ability. The overall survival rate of patients with advanced NB is not satisfactory. Ribosomal proteins (RPs) play a critical role in the development and progress of cancer. However, the contribution of RPL35 in NB has not been proven. In this study, we reveal that RPL35 is upregulated in NB tissues and the upregulation of RPL35 promotes proliferation and migration of NB while RPL35 knockdown significantly restrained the proliferation of NB cells. In terms of mechanism, glycolysis was decreased and the mitochondrial respiration was increased with knockdown of RPL35 in NB cells, indicating that RPL35 function as a positive regulator in aerobic glycolysis. Importantly, our data indicated that RPL35 deficiency decreased HIF1α expression both in mRNA and protein levels. Western blot analysis showed that RPL35 knockdown has a negative regulatory effect on the ERK pathway, and RPL35 modulated aerobic glycolysis in part through its regulation of the RPL35/ERK/HIF1α axis. Overall, RPL35 functions as a positive regulator of aerobic glycolysis, and the RPL35/ERK/HIF1α axis could be a potential therapeutic target for the therapy of NB.

In recent years, with the improvement of the sensitivity of examination equipment and the change of people's living environment and diet, the rate of thyroid cancer has risen rapidly, which has increased nearly five folds in 10 years. The pathogenesis, clinical manifestation, biological behavior, treatment and prognosis of thyroid carcinoma of different pathological types are obviously different. Papillary thyroid carcinoma (PTC) can develop at any age, which accounts for about 90% of thyroid cancer. It progresses slowly and has favourable prognosis, but lymph node metastasis appears easily. Whether PTC is accompanied by lymph node metastasis has an important impact on its prognosis and outcome. The Raf murine sarcoma viral oncogene homolog B(BRAF)gene mutation plays a crucial role in PTC lymph node metastasis. Having an in-depth understanding of the specific role and mechanism of BRAF gene mutation in PTC is expected to provide new ideas for diagnosis and treatment of PTC.近年来随着检查设备敏感性的提高及人们生活、饮食环境的改变，甲状腺癌（TC）的发病率直线上升，10 年内增加了近 5 倍，不同病理类型甲状腺癌发病机制、临床表现、生物学行为、治疗方法和预后有明显不同。甲状腺乳头状癌（PTC）任何年龄均可发病，约占甲状腺癌的 90%，疾病进展缓慢，预后良好，但易出现淋巴结转移。是否伴有淋巴结转移对于 PTC 的预后转归有着重要影响，抗鼠科肉瘤病毒癌基因同源物 B1（BRAF）基因突变在 PTC 的淋巴结转移中发挥着至关重要的作用，深入了解 BRAF 基因突变在 PTC 的具体作用及机制，有望对 PTC 提供新的诊疗思路。.

We previously reported that adiponectin (AD) promotes naïve T cell differentiation into Th17 cells and participates in synovial inflammation and the bone erosion process in patients with rheumatoid arthritis. Here, we use a T cell lineage adiponectin receptor 1 (AdipoR1) conditional knockout model to investigate the role of AdipoR1 in Th17 differentiation. RNA-sequencing (RNA-seq) demonstrated that AdipoR1 knockout reduced the expression of a variety of T cell related genes, with Rorc showing the greatest level of down-regulation. AdipoR1 deficiency inhibited Th17 cell differentiation in vitro and ameliorated joint inflammation in antigen-induced arthritis mice. Moreover, AdipoR1-deficent CD4+T cells displayed reduced Hypoxia-Inducible Factor-1α expression leading to glycolysis inhibition during naïve CD4+T cell differentiation into Th17 cells. We describe a novel function of AdipoR1 in regulating Th17 cell differentiation through modulating HIF-1α-dependent glycolysis.

Low-cost and highly effective catalysts are crucial to the electrocatalytic hydrogen evolution reaction (HER). Among non-noble catalysts, molybdenum carbides are promising candidates because of their high reserves, stability, low cost, and structural diversity. In this work, we report a simple method to fabricate a hollow porous Mo2C@C nanoball through a hydrothermal preparation process of molybdenum precursors at high temperatures. Specifically, we have combined interfacial polymerization and the chelation effect to synthesize the Mo-polydopamine (Mo-PDA) precursor. As a result, Mo2C@C-3 only requires an ultralow Tafel slope (~55 mV dec−1) and low overpotential (η50 ≈ 167 mV) in a 0.5 M H2SO4 solution with long-term cycling stability. Besides, it also exhibits outstanding activity and stability under extensive HER testing in alkaline media. This study is promising for the development of advanced molybdenum carbide electrocatalysts toward electrochemical applications.

This paper incorporates macroeconomic determinants into the forecasting model of industry-level stock return volatility in order to detect whether different macroeconomic factors can forecast the volatility of various industries. To explain different fluctuation characteristics among industries, we identified a set of macroeconomic determinants to examine their effects. The Clark and West (J Econom 138(1):291–311, 2007) test is employed to verify whether the new forecasting models, which vary among industries based on the in-sample results, make better predictions than the two benchmark models. Our results show that default return and default yield have a significant impact on stock return volatility.

Objectives Due to the instability of microRNAs, the applications of microRNA are currently limited. Thus, we utilized tetrahedral framework nucleic acids and a targeted microRNAs to form a stable nanocomposite to explore whether this nanocomposite can promote apoptosis of tumour cells. Materials and methods In our study, the survivin gene, which is expressed only in tumour cells and embryonic cells, was selected as the target gene; miRNA‐214‐3p, which can reduce the expression of survivin, was modified onto tetrahedral framework nucleic acid, thereby producing a reduction in the expression of survivin upon intracellular delivery and eventually leading to tumour cell apoptosis. Results By comparing the stability of microRNAs with that of microRNA‐tetrahedral framework nucleic acid, we proved the superiority of this carrier system. The results of flow cytometry showed that after treated with this complex, the ratio of A549 cells in both late and early period of apoptosis in miRNA‐214‐3p‐tetrahedral framework nucleic acid group had doubled and the cell cycle in the G2‐M phase had declined. The decrease in the expression of anti‐apoptotic protein and the increase in the expression of pro‐apoptotic protein indicate that the ability of this complex to function in cells also makes it attractive as a new targeted therapy for cancer. Conclusion The unique expression of survivin in tumour cells and embryonic cells makes microRNA‐tetrahedral framework nucleic acid a new targeted therapy. In addition, due to the functional diversity of microRNAs, this delivery system approach can be applied to a wide variety of fields, such as targeted therapy and tissue regeneration.

Background: Acupuncture is a well known treatment for ischemic stroke recovery, but evidence of its effectiveness remains limited. Methods: Rehabilitation training was provided to a control group. In Arm 1 and Arm 2, acupoint schemes were determined by a consensus of acupuncture experts and previous clinical research. Arm 1 was based on an acupoint summary of ancient literature, and Arm 2 was based on the summary of effective acupuncture points from modern RCT literature. Acupuncture was given on the acupoint. After acupuncture, the needles were left in for 30 minutes, and checked every 10 minutes. The three groups received treatment once a day, 5 times a week, for 2 weeks. The primary outcome was the National Institute of Health stroke scale (NIHSS), and the secondary outcomes were the Barthel index (BI) and the modified Ashworth scale (MAS). Outcomes were observed in patients both before and after treatment. Results: A total of 497 patients with ischemic stroke were randomized into either Arm 1 (159 cases), Arm 2 (173 cases) or a control group (165 cases). After 2 weeks of treatment, the NIHSS scores for Arm 1 were lower than those of the control group (P=0.017); the BI scores were higher in Arm 2 than they were in the control group (P=0.020). Additionally, there was no significant difference between Arm 1 and the control group for either the BI scores or the modified Ashworth scale scores (P>0.05). Conclusion: The clinical efficacy of Arm 1 and Arm 2 (acupuncture groups) was superior to that of the control group.

Membrane/water partition coefficient (Km/w) is a vital parameter used to characterize the membrane permeability of compounds. Considering the Km/w value is difficult to observe experimentally for real biological membranes, liposome/water partition coefficient (Klip/w) is employed to approximate Km/w. Here, quantitative structure property relationship (QSPR) models for logKlip/w of the neutral organic chemicals and the neutral form of ionogenic organic chemicals (IOCs) (logKlip/w-neutral), ionic form of IOCs (logKlip/w-ionic), the speciation-corrected liposome–water distribution ratios at a pH = 7.40 (logDlip/w-(pH=7.40)) were developed. In the modeling, two modeling methods (multiple linear regressions (MLR) and k-nearest neighbor (kNN)) were used. The predictive variables employed here could be calculated from the molecular structure directly. For logKlip/w-neutral and logDlip/w-(pH=7.40), the logKOW and logDOW-based, non-logKOW and non-logDOW-based kNN-QSPR and MLR-QSPR models were developed, respectively. The evaluation results implied that the predictive performance of kNN-QSPR models is better than that of MLR-QSPR models. For logKlip/w-ionic, only one acceptable MLR-QSPR model was developed for cation and anion, respectively. The model quality of the derived models was evaluated following the OECD QSPR models validation guideline. The determination coefficient (R2), leave-one-out cross validation Q2 (Q2LOO) and bootstrapping coefficient (Q2BOOT), the external validation coefficient (Q2EXT) of all the models met the acceptable criteria (Q2 > 0.600, R2 > 0.700); while the root-mean-square error (RMSE) range from 0.351 to 0.857. All the results implied that the models had good goodness-of-fit, robustness and predictive ability. Therefore, the developed models could be used to fill the data gap for substances within the applicability domain on their missing logKlip/w-neutral, logKlip/w-ionic, logDlip/w-(pH=7.40) values.

Background:Regulatory T cells (Tregs) play an essential role in maintaining self-tolerance and immune homeostasis. Abnormalities in the quantity or function of Treg cells are believed in RA patients, contributing to the inability to suppress autoimmunity and proinflammatory cytokines. Forkhead box P3 (Foxp3) is a crucial transcription factor for the development and differentiation of Tregs. How Tregs lose Foxp3 expression under inflammatory milieu remains largely unknown. Peptidase inhibitor 16 (PI16) is a member of the CAP (Cysteine-rich secretory proteins, Antigen 5, and Pathogenesis-related 1) protein family and its function are largely poor understood. In a genome-wide expression profiling study for identifying human Foxp3 target genes revealed PI16 was expressed on the cell surface of >80% of resting human CD25+ Foxp3+ Tregs. In the inflamed joint of juvenile idiopathic arthritis revealed a low number of PI16+ Tregs but high number of Th17 cells. However, little is known the function role of PI16 on Tregs or on RA development.Objectives:To investigate the role of peptidase inhibitor 16 (PI16) on the key T regulatory (Tregs) cells transcription factor Foxp3 expression and on the development of autoimmune arthritis.Methods:The expression of PI16 in blood, synovial fluid, inflamed joints were examined in Rheumatoid arthritis (RA) patients and in arthritic mice. Arthritis symptom, histological features and Foxp3 expression in PI16 transgenic (PI16Tg) arthritic mice were examined. Posttranslational mechanisms on PI16-mediated Foxp3 expression were analyzed. The specific role PI16 on Foxp3 expression was validated in conditional knockout (KO) mice.Results:The expression of PI16 was significantly increased in PBMC, serum, synovial tissue from RA patients or arthritic mice compared with controls. PI16Tg arthritic mice exhibited obvious inflammation, synovial hyperplasia and articular cartilage destruction in the joints compared with those in wild-type mice (WT) arthritic mice.Foxp3 is downregulated in splenic T cells and synovial tissue from PI16Tg arthritic mice. Naïve T cells derived from PI16Tg arthritic mice showed the decreased capacity to differentiate into Tregs. Polycomb-group (PcG) proteins complex molecule of Bmi-1 was significant increase in Tregs and joint tissue from PI16Tg arthritic mice. A direct interaction between 1-95AA domains of PI16 and 169 and 436 domains of Bmi-1 in Tregs promoter was observed. The binding of PI16 with Bmi-1 in the Foxp3 promoter inhibit the K48-linked polyubiquitin degradation of Bmi-1 at lysine site 72 and 153 region, which prompts the repressive histone modification of H3K27me3 and H2AK119ub, and inhibits the active histone modification of H3K4me3. Furthermore, conditional knockout of PI16 in Tregs retarded Foxp3 loss and blunted disease progression in experimental arthritis.Conclusion:PI16 represses Foxp3 expression by mediating histone modification via inhibiting K48-linked polyubiquitin degradation of Bmi-1 in Foxp3 promoter, contributing to disease progression in arthritic mice.Disclosure of Interests:None declared.

Superhydrophobic wearable textile electronics are of great promising devices due to their broad application in healthcare monitoring and artificial intelligence, etc. MXenes, the emerging two‐dimensional (2D) carbides and carbonitrides, are ideal candidates for fabricating wearable electronic devices. However, in an effort to maintain its highly conductive and long-term stability of cotton fabrics under harsh condition, it is an imperative demand to prepare superhydrophobic MXenes-textile sensor. Herein, a waterproof, highly sensitive, and wearable multi-mode sensor was successfully fabricated by the construction of 2D MXene nanosheets and zero-dimension (0D) silicon nanoparticles (SiNPs) onto a cotton fibers substrate (MX@SiNPs cotton). The as-cultivated MX@SiNPs cotton exhibited an integration of high-performance sensing toward pression, bending, and torsion, superior sensitivity (S1 = 12.23 kPa−1), a stable response under press-relaxing cycles, and broad sensing ranges (pressure: 8.8 Pa – 70 kPa, bending: 0 – 180°, torsion: 0 – 628 rad m−1). By virtue of its hierarchical structure and low surface energy of the SiNPs layer, the MXenes-textile sensor can maintain intrinsic conductivity under wet and corrosive conditions. The waterproof and highly sensitive MXene-reinforced cotton fabrics lay a basis for the broad-range application and large-scale preparation of the sensor and prolong its service life.

Background Herpes zoster, is one of the most familiar skin diseases in conducted a systematic review and meta-analysis in 2018 showed that fire needle acupuncture can relieve the pain caused by herpes zoster quickly and prevent the outcome of postherpetic neuralgia (PHN), with little side effects. The purpose of this study is to update the systematic review with the latest evidence. Methods Four English (PubMed, Embase, the Cochrane Library, the Web of Science) and 4 Chinese databases (CNKI, Wanfang, VIP, and CBM) will be searched dating until 30 June 2020 for randomized controlled trials with no language restrictions. In addition, a hand search of the reference lists of included studies will also be done. Adults (aged 18-70) with acute herpes zoster (less than 7 days) using fire needle acupuncture will be included. Pairs of researchers will independently conduct the search, screen titles and abstracts, retrieve full texts of potentially eligible studies, assess the risk of bias, and conduct date extraction and synthesis. If there is any discrepancy in the whole process, consult a third researcher. For meta-analysis, the primary outcome is the pain intensity (visual analogue scale [VAS] pain scale; pain relieve of 30%, duration of pain), and the second outcome is incidence of PHN. A sequential analysis will be done to test the robustness of results of meta-analysis. The quality of evidence will be assessed using GRADE system. Results The results will be published in a peer-reviewed journal. Conclusion This study will provide the latest systematic review and meta-analysis of fire needle acupuncture for acute herpes zoster and prevention of PHN.

Objective Management of chondral defects has long been a challenge due to poor self-healing capacity of articular cartilage. Many approaches, ranging from symptomatic treatment to structural cartilage regeneration, have obtained very limited satisfactory results. Cartilage tissue engineering, which involves optimized combination of novel scaffolds, cell sources and growth factors, has emerged as a promising strategy for cartilage regeneration and repair. In this study, the aim was to investigate the role of poly(e-caprolactone)-poly(ethylene glycol)-poly(e-caprolactone) (PCL-PEG-PCL, PCEC) PCEC scaffold in cartilage repair. Materials and methods First, PCEC film was fabricated, and its characteristics were tested using SEM and AFM. Cell (rASC – rat adipose-derived stem cells, and mASCs – green fluorescent mouse adipose-derived stem cells) morphologies on PCEC film were observed using SEM and fluorescence microscopy, after cell seeding. Tests of cell viability on PCEC film were conducted using the CCK-8 assay. Furthermore, full cartilage defects in rats were created, and PCEC films were implanted, to evaluate their healing effects, over 8 weeks. Results It was found that PCEC film, as a biomaterial implant, possessed good in vitro properties for cell adhesion, migration and proliferation. Importantly, in the in vivo experiment, PCEC film exhibited desirable healing outcomes. Conclusions These results demonstrated that PCEC film was a good scaffold for cartilage tissue engineering for improving cell proliferation and adhesion and could lead to excellent repair of cartilage defects.

Objectives This study aimed to investigate the influence of hypoxia on angiogenesis in a 3D gel, with co-culturing adipose-derived stromal cells (ASCs) and endothelial cells (ECs). Materials and methods ASCs from green fluorescent protein-labeled mice and ECs from red fluorescent protein-labeled mice were co-cultured in 3D collagen gels at 1:1 ratio, in normal and hypoxic oxygen conditions, and morphology of angiogenesis was observed using confocal laser scanning microscopy. To discover changes in growth factors between monoculture ASCs and ECs, transwell co-cultures of ASCs and ECs were applied. Semi-quantitative PCR was performed to explore mRNA expression of growth factors. Results Enhanced angiogenesis was observed in 3D gels implanted with 1:1 mixture of ASCs and ECs after 7 days hypoxia. Genes including VEGFA/B, EGF-1, HIF-1a, IGF-1, PDGF, TGF-β1 and BMP-2/4 in ECs, both monoculture and co-culture, were significantly enhanced after being cultured under hypoxia. In comparison, genes VEGFA/B, EGF-1, HIF-1a, TGF-β1 and BMP-2 in ASCs increased. In all, factors VEGFA/B, EGF-1, HIF-1a, TGF-β1 and BMP-2 increased in both ASCs and ECs after being cultured in hypoxia no matter whether as monoculture or co-culture. Conclusions Co-culture of ASCs and ECs at 1:1 ratio in a 3D gel under hypoxia promoted angiogenesis. Those growth factors which were increased in both ASCs and ECs, indicate that VEGFA/B, EGF-1, HIF-1a, TGF-β1 and BMP-2 might be responsible for enhancement in angiogenesis triggered by hypoxia.

Objectives Physiological oxygen tension plays a critical role in homoeostatic maintenance and development of endochondral bone. Based on the proximity between uncalcified cartilage and subchondral bone, and microchannels that serve as a message delivery network between them, we aimed to explore the influence of low oxygen tension on soluble factor secretion in both chondrocytes and osteoblasts, after co-culture. Materials and methods Contact co-culture was achieved for morphological observation using red fluorescent protein (RFP)-labelled chondrocytes and green fluorescent protein (GFP)-labelled osteoblasts, and non-contact co-culture achieved by transwell chambers. This was used to screen genetic variation of growth factors in hypoxia, including respective phenotypic markers, factors involving hypoxia and angiogenesis relationships, bone morphogenetic family proteins, and other general factors. Results We observed a significant increase in chondrocyte size following co-culture, in both normoxia and hypoxia, but not of osteoblasts. Expression of Aggrecan in chondrocytes and alkaline phosphatase in osteoblasts was down-regulated under hypoxia following co-culture. Under hypoxia, we found that expression of hypoxia-inducible factor-1α, vascular endothelial growth factor-A/B, VE-cadherin, bone morphogenetic protein-2, and insulin-like growth factor-1 in chondrocytes, increased, but HIF-1α, platelet-derived growth factor, BMP-5/-6 and fibroblast growth factor-1 in osteoblasts, decreased. Conclusions These results not only indicate the importance of crosstalk between chondrocytes and osteoblasts but also improve our understanding of the mechanisms underlying homoeostatic maintenance of endochondral bone.
