We present ALMA archival data for 219-235 GHz continuum and line observations toward the hot molecular core (HMC) W49N MCN-a (UCHII region J1) at a resolution of ~0."3. The dust continuum emission, showing an elongated structure of 1."40x0."95 (PA=43.5deg) perpendicular to the outflow seen in SiO and SO, represents a rotating flattened envelope, or torus, with a radius of 7,800au inclined at 47.5deg or larger. The emissions from CH3CN and 11 molecular lines exhibit a consistent velocity gradient as a result of rotation. The magnitude of each velocity gradient is different, reflecting that each line samples a specific radial region. This allows us to derive a rotation curve as Vrot prop R^0.44+-0.11 for 2,400au < R < 14,000au, giving the dynamical mass as Mdyn = 57.0+24.5-17.1 (R [au]/3, 000)^1.88 Msun. Histopathologic analysis through biopsy has been one of the most useful methods for the assessment of malignant neoplasms. However, some aspects of the analysis such as invasiveness, evaluation range, and turnaround time from biopsy to report could be improved. Here, we report a novel method for visualizing human cervical tissue three-dimensionally, without biopsy, fixation, or staining, and with sufficient quality for histologic diagnosis. Near-infrared excitation and nonlinear optics were employed to visualize unstained human epithelial tissues of the cervix uteri by constructing images with third-harmonic generation (THG) and second-harmonic generation (SHG). THG images enabled evaluation of nuclear morphology in a quantitative manner with six parameters after image analysis using deep learning. It was also possible to quantitatively assess intraepithelial fibrotic changes based on SHG images and another deep learning analysis. Using each analytical procedure alone, normal and cancerous tissue were classified quantitatively with an AUC ≥0.92. Moreover, a combinatory analysis of THG and SHG images with a machine learning algorithm allowed accurate classification of three-dimensional image files of normal tissue, intraepithelial neoplasia, and invasive carcinoma with a weighted kappa coefficient of 0.86. Our method enables real-time noninvasive diagnosis of cervical lesions, thus constituting a potential tool to dramatically change early detection.Significance:This study proposes a novel method for diagnosing cancer using nonlinear optics, which enables visualization of histologic features of living tissues without the need for any biopsy or staining dye.

We present high resolution observations of CS(J=1-0), H13CO+ (J=1-0), and SiO(v=0:J=1-0) lines, together with the 49GHz and 86GHz continuum emissions, toward W49N carried out with Nobeyama Millimeter Array. We identified 11 CS, 8 H13CO+, and 6 SiO clumps with radii of 0.1-0.5pc. The CS and H13CO+ clumps are mainly divided into two velocity components, one at 4kms-1 and the other at 12kms-1, while the SiO clumps have velocities between the two components. The SiO emission is distributed toward the UCHII ring, where the 4kms-1 component clumps of CS and H13CO+ also exist. The 12kms-1 component clumps of CS are detected at the east and west of the UCHII ring with an apparent hole toward the ring. The clump masses vary from 4.4x10^2 M_SUN to 4.9x10^4 M_SUN with the mean values of 0.94x10^4M_SUN, 0.88x10^4M_SUN, and 2.2x10^4M_SUN for the CS, H13CO+, and SiO clumps, respectively. The total masses derived from CS, H13CO+, and SiO clumps are 1.0x10^5M_SUN, 0.70x10^5M_SUN, and 1.3x10^5 M_SUN, respectively, which agree well with the corresponding virial masses of 0.71x10^5M_SUN, 1.3x10^5M_SUN, and 0.88x10^5M_SUN, respectively. The average molecular hydrogen densities of the clumps are 0.90x10^6 cm-3, 1.4x10^6cm-3, and 7.6x10^6 cm-3 for the CS, H13CO+ and SiO clumps, respectively. The density derived from the SiO clumps seems significantly higher than those from the others, probably because the SiO emission is produced in high density shocked regions. The free fall time scale of the clumps is estimated to be ~3x10^4 yr, which gives an accretion rate of 3x10-3-1M_SUN yr-1 onto a stellar core. The observed clumps are, if they are undergoing free fall, capable of producing dozens of massive stars in the next 10^5 yr. Osteoimmunology highlights the reciprocal interactions between the skeletal and immune systems. Over the past two decades, many molecules that link the two have been identified, including cytokines, receptors and transcription factors, leading to successful translation of research into therapeutic approaches to autoimmune diseases such as rheumatoid arthritis. The development of an intravital imaging system using two-photon microscopy, combined with a variety of fluorescent probes and reporter mouse strains, has provided valuable insights into the real-time dynamics of osteoclasts and immune cells in the bone marrow. This technique is now applied to the synovial tissue of arthritic mice to investigate the pathogenesis of osteoimmune diseases and enables direct observation of complex biological phenomena in vivo. In addition, rapid progress in the next-generation sequencing technologies has provided important insights into the field of osteoimmunology through characterizing individual cells in the synovial microenvironment. Single-cell RNA sequencing (scRNA-seq) dissects cellular heterogeneity within a biological system and enables the identification of specific cells differentiating into mature osteoclasts within the previously defined ‘osteoclast precursor-containing population’. In this review, we will explain the cellular interactions and cytokine milieu involved in inflammatory bone destruction and update how the novel technologies, such as scRNA-seq and intravital imaging, have contributed to better understand the pathogenesis of bone destruction in arthritis.

We have developed a method to make a spectral-line-based survey of hot cores, which represent an important stage of high-mass star formation, and applied the method to the data of the FUGIN (FOREST Unbiased Galactic plane Imaging survey with the Nobeyama 45 m telescope) survey. First, we select hot core candidates by searching the FUGIN data for the weak hot core tracer lines (HNCO and CH3CN) by stacking, and then we conduct follow-up pointed observations on these candidates in C34S, SO, OCS, HC3N, HNCO, CH3CN, and CH3OH J = 2–1 and J = 8–7 lines to confirm and characterize them. We applied this method to the l = 10°–20° portion of the FUGIN data and identified 22 “HotCores” (compact sources with more than two significant detections of the hot core tracer lines, i.e., SO, OCS, HC3N, HNCO, CH3CN, or CH3OH J = 8–7 lines) and 14 “DenseClumps” (sources with more than two significant detection of C34S, CH3OH J = 2–1, or the hot core tracer lines). The identified HotCores are found to be associated with signposts of high-mass star formation such as ATLASGAL clumps, WISE H ii regions, and Class II methanol masers. Many of the FUGIN HotCores are identified with the Herschel Hi-GAL clumps with a median mass of 6.8 × 102 M⊙ and a median bolometric luminosity of 7.4 × 103 L⊙. Five of the seven HotCores with stronger CH3CN lines exhibit elevated gas temperatures of 50–100 K. These observations suggest that FUGIN HotCores are closely related to the formation of stars with medium to high mass. For those associated with ATLASGAL clumps, their bolometric luminosity to clump mass ratios are consistent with the star formation stages centered at the hot core phase. The catalog of FUGIN HotCores provides a useful starting point for further statistical studies and detailed observations of high-mass star forming regions.

Osteoclastic bone resorption and osteoblastic bone formation/replenishment are closely coupled in bone metabolism. Anabolic parathyroid hormone (PTH), which is commonly used for treating osteoporosis, shifts the balance from osteoclastic to osteoblastic, although it is unclear how these cells are coordinately regulated by PTH. Here, we identify a serine protease inhibitor, secretory leukocyte protease inhibitor (SLPI), as a critical mediator that is involved in the PTH-mediated shift to the osteoblastic phase. Slpi is highly upregulated in osteoblasts by PTH, while genetic ablation of Slpi severely impairs PTH-induced bone formation. Slpi induction in osteoblasts enhances its differentiation, and increases osteoblast–osteoclast contact, thereby suppressing osteoclastic function. Intravital bone imaging reveals that the PTH-mediated association between osteoblasts and osteoclasts is disrupted in the absence of SLPI. Collectively, these results demonstrate that SLPI regulates the communication between osteoblasts and osteoclasts to promote PTH-induced bone anabolism., The mechanism by which parathyroid hormone mediates the switch from bone resorption to bone formation is unclear. Here, the authors show that SLPI regulates the communication between osteoblasts and osteoclasts to promote the anabolic effect of parathyroid hormone.

The human body is comprised of hundreds of bones, which are constantly regenerated through the interactions of two cell types: osteoblasts and osteoclasts. Given the difficulty of analyzing their intravital dynamics, we have developed a system for intravital imaging of the bone marrow cavity using two-photon microscopy, to visualize the dynamic behaviors of living bone cells without sectioning. Combined with the newly developed chemical fluorescent probes to detect localized acidification caused by osteoclasts, we identified two distinct functional states of mature osteoclasts, i.e., “bone-resorptive” and “non-resorptive”. Here, we focus on the dynamics and functions of bone cells within the bone marrow cavity and discuss how this novel approach has been applied to evaluate the mechanisms of action of drugs currently in clinical use. We further introduce our recent study that identified arthritis-associated osteoclastogenic macrophages in inflamed synovium and revealed their differentiation trajectory into the pathological osteoclasts, which together represent to a new paradigm in bone research.

We present 850 $��$m polarimetric observations toward the Serpens Main molecular cloud obtained using the POL-2 polarimeter on the James Clerk Maxwell Telescope (JCMT) as part of the B-fields In STar-forming Region Observations (BISTRO) survey. These observations probe the magnetic field morphology of the Serpens Main molecular cloud on about 6000 au scales, which consists of cores and six filaments with different physical properties such as density and star formation activity. Using the histogram of relative orientation (HRO) technique, we find that magnetic fields are parallel to filaments in less dense filamentary structures where $N_{H_2} < 0.93\times 10^{22}$ cm$^{-2}$ (magnetic fields perpendicular to density gradients), while being perpendicular to filaments (magnetic fields parallel to density gradients) in dense filamentary structures with star formation activity. Moreover, applying the HRO technique to denser core regions, we find that magnetic field orientations change to become perpendicular to density gradients again at $N_{H_2} \approx 4.6 \times 10^{22}$ cm$^{-2}$. This can be interpreted as a signature of core formation. At $N_{H_2} \approx 16 \times 10^{22}$ cm$^{-2}$ magnetic fields change back to being parallel to density gradients once again, which can be understood to be due to magnetic fields being dragged in by infalling material. In addition, we estimate the magnetic field strengths of the filaments ($B_{POS} = 60-300~��$G)) using the Davis-Chandrasekhar-Fermi method and discuss whether the filaments are gravitationally unstable based on magnetic field and turbulence energy densities., 18 pages, accepted for publication in ApJ

Tao-Chung Ching et al., We present 850 μm dust polarization observations of the massive DR21 filament from the B-fields In STar-forming Region Observations (BISTRO) survey, using the POL-2 polarimeter and the SCUBA-2 camera on the James Clerk Maxwell Telescope. We detect ordered magnetic fields perpendicular to the parsec-scale ridge of the DR21 main filament. In the subfilaments, the magnetic fields are mainly parallel to the filamentary structures and smoothly connect to the magnetic fields of the main filament. We compare the POL-2 and Planck dust polarization observations to study the magnetic field structures of the DR21 filament on 0.1–10 pc scales. The magnetic fields revealed in the Planck data are well-aligned with those of the POL-2 data, indicating a smooth variation of magnetic fields from large to small scales. The plane-of-sky magnetic field strengths derived from angular dispersion functions of dust polarization are 0.6–1.0 mG in the DR21 filament and ∼0.1 mG in the surrounding ambient gas. The mass-to-flux ratios are found to be magnetically supercritical in the filament and slightly subcritical to nearly critical in the ambient gas. The alignment between column density structures and magnetic fields changes from random alignment in the low-density ambient gas probed by Planck to mostly perpendicular in the high-density main filament probed by James Clerk Maxwell Telescope. Hwang et al., We present and analyze observations of polarized dust emission at 850 μm toward the central 1 × 1 pc hub-filament structure of Monoceros R2 (Mon R2). The data are obtained with SCUBA-2/POL-2 on the James Clerk Maxwell Telescope (JCMT) as part of the B-fields in Star-forming Region Observations survey. The orientations of the magnetic field follow the spiral structure of Mon R2, which are well described by an axisymmetric magnetic field model. We estimate the turbulent component of the magnetic field using the angle difference between our observations and the best-fit model of the underlying large-scale mean magnetic field. This estimate is used to calculate the magnetic field strength using the Davis–Chandrasekhar–Fermi method, for which we also obtain the distribution of volume density and velocity dispersion using a column density map derived from Herschel data and the C18O (J = 3 − 2) data taken with HARP on the JCMT, respectively. We make maps of magnetic field strengths and mass-to-flux ratios, finding that magnetic field strengths vary from 0.02 to 3.64 mG with a mean value of 1.0 ± 0.06 mG, and the mean critical mass-to-flux ratio is 0.47 ± 0.02. Additionally, the mean Alfvén Mach number is 0.35 ± 0.01. This suggests that, in Mon R2, the magnetic fields provide resistance against large-scale gravitational collapse, and the magnetic pressure exceeds the turbulent pressure. We also investigate the properties of each filament in Mon R2. Theoretically, misalignment between the magnetic field and rotational axis in a dense core is considered to be dynamically important in the star formation process, however, extent of this influence remains observationally unclear. For a sample of 32 Class 0 and I protostars in the Perseus Molecular Cloud, we analyzed gas motions using C$^{18}$O data from the SMA MASSES survey and the magnetic field structures using 850 $\mu$m polarimetric data from the JCMT BISTRO-1 survey and archive. We do not find any significant correlation between the velocity gradients in the C$^{18}$O emission in the protostellar envelopes at a 1,000 au scale and the misalignment between the outflows and magnetic field orientations in the dense cores at a 4,000 au scale, and there is also no correlation between the velocity gradients and the angular dispersions of the magnetic fields. However, a significant dependence on the misalignment angles emerges after we normalize the rotational motion by the infalling motion, where the ratios increase from $\lesssim1$ to $\gtrsim1$ with increasing misalignment angles. This suggests that the misalignment could prompt angular momentum transportation to the envelope scale but is not a dominant factor in determining the envelope rotation, and other parameters, like mass accretion in protostellar sources, also play an important role. These results remain valid after taking into account projection effects. The comparison between our estimated angular momentum in the protostellar envelopes and the sizes of the known protostellar disks suggests that significant angular momentum is likely lost between radii of $\sim$1,000-100 au in protostellar envelopes., Comment: Accepted by ApJ

We present 250~GHz continuum and H29alpha line data toward W49N:A2, a hypercompact HII region ionized by an O9 star. The data obtained with ALMA at a resolution of ~0"05 (600 au) confirmed the presence of an ionized ring with a radius of~700 au inclined by ~50degree (0degree for pole-on). It has a width of ~1000 au and is relatively flat with a scale height of less than several hundred au. The tilted ring, or the apparent ellipse, has a prominent velocity difference between its NW and SE ridges along the minor axis, suggesting that it is expanding in the equatorial plane at a velocity of 13.2 km/s. The ring also shows a hint of rotation at 2.7 km/s, which is significantly (2.5sigma) smaller than the Kepler velocity of 5.2 km/s at its radius around the 20 M$_sun star. This can be interpreted that the ring gas has been transported from the radius of ~170 au by conserving its original specific angular momentum that it had there. The ionized ring may thus be a remnant of the accretion disk that fed the O9 star, whose radiation or magnetic activities became so strong that the disk accretion was reversed due to the intense thermal or magneto-hydrodynamic pressure around the star. The data has revealed a rare example of how a massive star terminates its accretion at the end of its formation, transforming a hypercompact HII region into an ultracompact HII region., Comment: 8 pages, 7 figures, 1 Table

We present the results of simultaneous 450 $\mu$m and 850 $\mu$m polarization observations toward the massive star forming region NGC 2071IR, a target of the BISTRO (B-fields in Star-Forming Region Observations) Survey, using the POL-2 polarimeter and SCUBA-2 camera mounted on the James Clerk Maxwell Telescope. We find a pinched magnetic field morphology in the central dense core region, which could be due to a rotating toroidal disk-like structure and a bipolar outflow originating from the central young stellar object, IRS 3. Using the modified Davis-Chandrasekhar-Fermi method, we obtain a plane-of-sky magnetic field strength of 563$\pm$421 $\mu$G in the central $\sim$0.12 pc region from 850 $\mu$m polarization data. The corresponding magnetic energy density of 2.04$\times$10$^{-8}$ erg cm$^{-3}$ is comparable to the turbulent and gravitational energy densities in the region. We find that the magnetic field direction is very well aligned with the whole of the IRS 3 bipolar outflow structure. We find that the median value of polarization fractions, 3.0 \%, at 450 $\mu$m in the central 3 arcminute region, which is larger than the median value of 1.2 \% at 850 $\mu$m. The trend could be due to the better alignment of warmer dust in the strong radiation environment. We also find that polarization fractions decrease with intensity at both wavelengths, with slopes, determined by fitting a Rician noise model, of $0.59 \pm 0.03$ at 450 $\mu$m and $0.36 \pm 0.04$ at 850 $\mu$m, respectively. We think that the shallow slope at 850 $\mu$m is due to grain alignment at the center being assisted by strong radiation from the central young stellar objects., Comment: 17 pages, 11 figures, 4 tables

Rheumatoid arthritis is a chronic form of arthritis that causes bone destruction in joints such as the knees and fingers. Over the past two decades, the clinical outcomes of rheumatoid arthritis have improved substantially with the development of biological agents and Janus kinase inhibitors. Osteoclasts are myeloid lineage cells with a unique bone-destroying ability that can lead to joint destruction. On the other hand, osteoclasts play an important role in skeletal homeostasis by supporting bone remodeling together with osteoblasts in the bone marrow under steady-state conditions. However, the same osteoclasts are considered to participate in physiological bone remodeling and joint destruction. We found that pathological osteoclasts have different differentiation pathways and regulatory transcription factors compared to physiological osteoclasts. We also identified arthritis-associated osteoclastogenic macrophages (AtoMs), which are common progenitors of pathological osteoclasts in mice and humans that develop specifically in inflamed synovial tissue. This review presents details of the newly identified AtoMs and the original intravital imaging systems that can visualize synovial tissue and pathological osteoclasts at the pannus-bone interface.

We present the first 850 $\mu$m polarization observations in the most active star-forming site of the Rosette Molecular Cloud (RMC, $d\sim$1.6 kpc) in the wall of the Rosette Nebula, imaged with the SCUBA-2/POL-2 instruments of the JCMT, as part of the B-Fields In Star-Forming Region Observations 2 (BISTRO-2) survey. From the POL-2 data we find that the polarization fraction decreases with the 850 $\mu$m continuum intensity with $\alpha$ = 0.49 $\pm$ 0.08 in the $p \propto I^{\rm -\alpha}$ relation, which suggests that some fraction of the dust grains remain aligned at high densities. The north of our 850 $\mu$m image reveals a "gemstone ring" morphology, which is a $\sim$1 pc-diameter ring-like structure with extended emission in the "head" to the south-west. We hypothesize that it might have been blown by feedback in its interior, while the B-field is parallel to its circumference in most places. In the south of our SCUBA-2 field the clumps are apparently connected with filaments which follow Infrared Dark Clouds (IRDCs). Here, the POL-2 magnetic field orientations appear bimodal with respect to the large-scale Planck field. The mass of our effective mapped area is $\sim$ 174 $M_\odot$ that we calculate from 850 $\mu$m flux densities. We compare our results with masses from large-scale emission-subtracted Herschel 250 $\mu$m data, and find agreement within 30%. We estimate the POS B-field strength in one typical subregion using the Davis-Chandrasekhar-Fermi (DCF) technique and find 80 $\pm$ 30 $\mu$G toward a clump and its outskirts. The estimated mass-to-flux ratio of $\lambda$ = 2.3 $\pm$ 1.0 suggests that the B-field is not sufficiently strong to prevent gravitational collapse in this subregion., Comment: 15 pages (12+3), 9 figures, 1 table. We have obtained sensitive dust continuum polarization observations at 850 μm in the B213 region of Taurus using POL-2 on SCUBA-2 at the James Clerk Maxwell Telescope as part of the B-fields in STar-forming Region Observations (BISTRO) survey. These observations allow us to probe magnetic field (B-field) at high spatial resolution (∼2000 au or ∼0.01 pc at 140 pc) in two protostellar cores (K04166 and K04169) and one prestellar core (Miz-8b) that lie within the B213 filament. Using the Davis–Chandrasekhar–Fermi method, we estimate the B-field strengths in K04166, K04169, and Miz-8b to be 38 ± 14, 44 ± 16, and 12 ± 5 μG, respectively. These cores show distinct mean B-field orientations. The B-field in K04166 is well ordered and aligned parallel to the orientations of the core minor axis, outflows, core rotation axis, and large-scale uniform B-field, in accordance with magnetically regulated star formation via ambipolar diffusion taking place in K04166. The B-field in K04169 is found to be ordered but oriented nearly perpendicular to the core minor axis and large-scale B-field and not well correlated with other axes. In contrast, Miz-8b exhibits a disordered B-field that shows no preferred alignment with the core minor axis or large-scale field. We found that only one core, K04166, retains a memory of the large-scale uniform B-field. The other two cores, K04169 and Miz-8b, are decoupled from the large-scale field. Such a complex B-field configuration could be caused by gas inflow onto the filament, even in the presence of a substantial magnetic flux.

Bone metabolism is regulated by the cooperative activity between bone-forming osteoblasts and bone-resorbing osteoclasts. However, the mechanisms mediating the switch between the osteoblastic and osteoclastic phases have not been fully elucidated. Here, we identify a specific subset of mature osteoblast-derived extracellular vesicles that inhibit bone formation and enhance osteoclastogenesis. Intravital imaging reveals that mature osteoblasts secrete and capture extracellular vesicles, referred to as small osteoblast vesicles (SOVs). Co-culture experiments demonstrate that SOVs suppress osteoblast differentiation and enhance the expression of receptor activator of NF-κB ligand, thereby inducing osteoclast differentiation. We also elucidate that the SOV-enriched microRNA miR-143 inhibits Runt-related transcription factor 2, a master regulator of osteoblastogenesis, by targeting the mRNA expression of its dimerization partner, core-binding factor β. In summary, we identify SOVs as a mode of cell-to-cell communication, controlling the dynamic transition from bone-forming to bone-resorbing phases in vivo.

Optical stellar polarimetry in the Perseus molecular cloud direction is known to show a fully mixed bi-modal distribution of position angles across the cloud (Goodman et al. 1990). We study the Gaia trigonometric distances to each of these stars and reveal that the two components in position angles trace two different dust clouds along the line of sight. One component, which shows a polarization angle of -37.6 deg +/- 35.2 deg and a higher polarization fraction of 2.0 +/- 1.7%, primarily traces the Perseus molecular cloud at a distance of 300 pc. The other component, which shows a polarization angle of +66.8 deg +/- 19.1 deg and a lower polarization fraction of 0.8 +/- 0.6%, traces a foreground cloud at a distance of 150 pc. The foreground cloud is faint, with a maximum visual extinction of < 1 mag. We identify that foreground cloud as the outer edge of the Taurus molecular cloud. Between the Perseus and Taurus molecular clouds, we identify a lower-density ellipsoidal dust cavity with a size of 100 -- 160 pc. This dust cavity locates at l = 170 deg, b = -20 deg, and d = 240 pc, which corresponds to an HI shell generally associated with the Per OB2 association. The two-component polarization signature observed toward the Perseus molecular cloud can therefore be explained by a combination of the plane-of-sky orientations of the magnetic field both at the front and at the back of this dust cavity., Accepted for publication in ApJ

We report the first high spatial resolution measurement of magnetic fields surrounding LkH$\alpha$ 101, a part of the Auriga-California molecular cloud. The observations were taken with the POL-2 polarimeter on the James Clerk Maxwell Telescope within the framework of the B-fields In Star-forming Region Observations (BISTRO) survey. Observed polarization of thermal dust emission at 850 $\mu$m is found to be mostly associated with the red-shifted gas component of the cloud. The magnetic field displays a relatively complex morphology. Two variants of the Davis-Chandrasekhar-Fermi method, unsharp masking and structure function, are used to calculate the strength of magnetic fields in the plane of the sky, yielding a similar result of $B_{\rm POS}\sim 115$ $\mathrm{\mu}$G. The mass-to-magnetic-flux ratio in critical value units, $\lambda\sim0.3$, is the smallest among the values obtained for other regions surveyed by POL-2. This implies that the LkH$\alpha$ 101 region is sub-critical and the magnetic field is strong enough to prevent gravitational collapse. The inferred $\delta B/B_0\sim 0.3$ implies that the large scale component of the magnetic field dominates the turbulent one. The variation of the polarization fraction with total emission intensity can be fitted by a power-law with an index of $\alpha=0.82\pm0.03$, which lies in the range previously reported for molecular clouds. We find that the polarization fraction decreases rapidly with proximity to the only early B star (LkH$\alpha$ 101) in the region. We present new observations of the active star formation region NGC 1333 in the Perseus molecular cloud complex from the James Clerk Maxwell Telescope B -Fields In Star-forming Region Observations (BISTRO) survey with the POL-2 instrument. The BISTRO data cover the entire NGC 1333 complex (∼1.5 pc × 2 pc) at 0.02 pc resolution and spatially resolve the polarized emission from individual filamentary structures for the first time. The inferred magnetic field structure is complex as a whole, with each individual filament aligned at different position angles relative to the local field orientation. We combine the BISTRO data with low- and high- resolution data derived from Planck and interferometers to study the multiscale magnetic field structure in this region. The magnetic field morphology drastically changes below a scale of ∼1 pc and remains continuous from the scales of filaments (∼0.1 pc) to that of protostellar envelopes (∼0.005 pc or ∼1000 au). Finally, we construct simple models in which we assume that the magnetic field is always perpendicular to the long axis of the filaments. We demonstrate that the observed variation of the relative orientation between the filament axes and the magnetic field angles are well reproduced by this model, taking into account the projection effects of the magnetic field and filaments relative to the plane of the sky. We herein describe a case of an 83-year-old man who presented with epigastralgia, vomiting, and abdominal distention. The physical abdominal examination revealed mild tenderness. Computed tomography revealed intramural gastric gas spread throughout the stomach, intraabdominal free gas, and hepatic portal venous gas. We diagnosed gastric emphysema with intraabdominal free gas and hepatic portal venous gas. We selected a wait-and-watch approach because physical examination did not show any peritoneal signs, although the radiological examinations showed remarkable findings. As a result, he received conservative therapy with fasting, intravenous infusion of antibiotics, and gastric decompression by nasogastric intubation. The patient was relieved of the symptoms, and follow-up computed tomography showed that all the abnormal gas disappeared soon after the treatment. In conclusion, the intramural gastric gas even with both intraabdominal free gas and hepatic portal venous gas does not always require surgical intervention. In case clinicians including general surgeons and physicians encounter intraabdominal free gas with hepatic portal venous gas, gastric emphysema should be considered in the different diagnosis. Lack of knowledge may lead to misdiagnosis, which may result in unnecessary surgical intervention.

Sudo et al. demonstrate that group 2 innate lymphoid cells (ILC2s) in the bone marrow become activated following chemotherapy-induced stress and support hematopoietic recovery by producing GM-CSF. Adoptive transfer of ILC2s has therapeutic potential for myelosuppressive conditions., The cell-cycle status of hematopoietic stem and progenitor cells (HSPCs) becomes activated following chemotherapy-induced stress, promoting bone marrow (BM) regeneration; however, the underlying molecular mechanism remains elusive. Here we show that BM-resident group 2 innate lymphoid cells (ILC2s) support the recovery of HSPCs from 5-fluorouracil (5-FU)–induced stress by secreting granulocyte-macrophage colony-stimulating factor (GM-CSF). Mechanistically, IL-33 released from chemo-sensitive B cell progenitors activates MyD88-mediated secretion of GM-CSF in ILC2, suggesting the existence of a B cell–ILC2 axis for maintaining hematopoietic homeostasis. GM-CSF knockout mice treated with 5-FU showed severe loss of myeloid lineage cells, causing lethality, which was rescued by transferring BM ILC2s from wild-type mice. Further, the adoptive transfer of ILC2s to 5-FU–treated mice accelerates hematopoietic recovery, while the reduction of ILC2s results in the opposite effect. Thus, ILC2s may function by “sensing” the damaged BM spaces and subsequently support hematopoietic recovery under stress conditions.

Histopathologic analysis through biopsy has been one of the most useful methods for the assessment of malignant neoplasms. However, some aspects of the analysis such as invasiveness, evaluation range, and turnaround time from biopsy to report could be improved. Here, we report a novel method for visualizing human cervical tissue three-dimensionally, without biopsy, fixation, or staining, and with sufficient quality for histologic diagnosis. Near-infrared excitation and nonlinear optics were employed to visualize unstained human epithelial tissues of the cervix uteri by constructing images with third-harmonic generation (THG) and second-harmonic generation (SHG). THG images enabled evaluation of nuclear morphology in a quantitative manner with six parameters after image analysis using deep learning. It was also possible to quantitatively assess intraepithelial fibrotic changes based on SHG images and another deep learning analysis. Using each analytical procedure alone, normal and cancerous tissue were classified quantitatively with an AUC ≥0.92. Moreover, a combinatory analysis of THG and SHG images with a machine learning algorithm allowed accurate classification of three-dimensional image files of normal tissue, intraepithelial neoplasia, and invasive carcinoma with a weighted kappa coefficient of 0.86. Our method enables real-time noninvasive diagnosis of cervical lesions, thus constituting a potential tool to dramatically change early detection. Significance: This study proposes a novel method for diagnosing cancer using nonlinear optics, which enables visualization of histologic features of living tissues without the need for any biopsy or staining dye.

[Abridged] Filaments and hubs have received special attention recently thanks to studies showing their role in star formation. While the column density and velocity structures of both filaments and hubs have been studied, their magnetic fields (B-field) are not yet characterized. We aim to understand the role of the B-field in the dynamical evolution of the NGC 6334 hub-filament network. We present new observations of the dust polarized emission at 850$\mu$m towards NGC 6334 obtained with the JCMT/POL-2. We study the distribution and dispersion of the polarized intensity ($PI$), the polarization fraction ($PF$), and the B-field angle ($\theta_{B}$). We derive the power spectrum of the intensity and $\theta_{B}$ along the ridge crest. Our analyses show a complex B-field structure when observed over the whole region ($\sim10$ pc), however, at smaller scales ($\sim1$ pc), $\theta_{B}$ varies coherently along the filaments. The observed power spectrum of $\theta_{B}$ can be well represented with a power law function with a slope $-1.33\pm0.23$, which is $\sim20\%$ shallower than that of $I$. This result is compatible with the properties of simulated filaments and may indicate the processes at play in the formation of filaments. $\theta_{B}$ rotates from being mostly perpendicular to the filament crests to mostly parallel as they merge with the hubs. This variation of $\theta_{B}$ may be tracing local velocity flows of matter in-falling onto the hubs. Our analysis suggests a variation of the energy balance along the crests of these filaments, from magnetically critical/supercritical at their far ends to magnetically subcritical near the hubs. We present the first release of the data and compact-source catalogue for the JCMT Large Program SCUBA-2 Continuum Observations of Pre-protostellar Evolution (SCOPE). SCOPE consists of 850-um continuum observations of 1235 Planck Galactic Cold Clumps (PGCCs) made with the Submillimetre Common-User Bolometer Array 2 on the James Clerk Maxwell Telescope. These data are at an angular resolution of 14.4 arcsec, significantly improving upon the 353-GHz resolution of Planck at 5 arcmin, and allowing for a catalogue of 3528 compact sources in 558 PGCCs. We find that the detected PGCCs have significant sub-structure, with 61 per cent of detected PGCCs having 3 or more compact sources, with filamentary structure also prevalent within the sample. A detection rate of 45 per cent is found across the survey, which is 95 per cent complete to Planck column densities of $N_{H_{2}}$ $>$ 5 $\times$ 10$^{21}$ cm$^{-2}$. By positionally associating the SCOPE compact sources with YSOs, the star formation efficiency, as measured by the ratio of luminosity to mass, in nearby clouds is found to be similar to that in the more distant Galactic Plane, with the column density distributions also indistinguishable from each other., 16 pages, 13 figures, 1 table. We present the 850 $\mu$m polarization observations toward the IC5146 filamentary cloud taken using the Submillimetre Common-User Bolometer Array 2 (SCUBA-2) and its associated polarimeter (POL-2), mounted on the James Clerk Maxwell Telescope (JCMT), as part of the B-fields In STar forming Regions Observations (BISTRO). This work is aimed at revealing the magnetic field morphology within a core-scale ($\lesssim 1.0$ pc) hub-filament structure (HFS) located at the end of a parsec-scale filament. To investigate whether or not the observed polarization traces the magnetic field in the HFS, we analyze the dependence between the observed polarization fraction and total intensity using a Bayesian approach with the polarization fraction described by the Rice likelihood function, which can correctly describe the probability density function (PDF) of the observed polarization fraction for low signal-to-noise ratio (SNR) data. We find a power-law dependence between the polarization fraction and total intensity with an index of 0.56 in $A_V\sim$ 20--300 mag regions, suggesting that the dust grains in these dense regions can still be aligned with magnetic fields in the IC5146 regions. Our polarization maps reveal a curved magnetic field, possibly dragged by the contraction along the parsec-scale filament. We further obtain a magnetic field strength of 0.5$\pm$0.2 mG toward the central hub using the Davis-Chandrasekhar-Fermi method, corresponding to a mass-to-flux criticality of $\sim$ $1.3\pm0.4$ and an Alfv\'{e}nic Mach number of $, Comment: 24 pages, 15 figures, accepted for publication in ApJ
