Introduction: WhiA is a conserved protein found in numerous bacteria. It consists of an HTH DNA-binding domain linked with a homing endonuclease (HEN) domain. WhiA is one of the most conserved transcription factors in reduced bacteria of the class Mollicutes. Its function in Mollicutes is unknown, while it is well-characterized in Streptomyces. Here, we focused on WhiA protein from Mycoplasma gallisepticum. Methods: We used a combination molecular dynamics, EMSA, MST and AFM to study the DNA-binding and ATP-binding properties of WhiA from M. gallisepticum. The transcriptional repressor function of WhiA was demonstrated using gene knockdown, reporter constructs and proteome analysis. Results: We demonstrate that WhiA homolog from M. gallisepticum binds a conserved sequence of the GAYACRCY core (Y = C or T, R = A or G), which is located in the promoter of an operon coding for ribosomal proteins and adenylate kinase (rpsJ operon). We show that WhiA in M. gallisepticum is a repressor of rpsJ operon and a sensor of ATP. HTH domain binds to the core motif and HEN domain binds to the auxiliary motif GTTGT that is located downstream to the core motif. We show that binding by both domains to DNA is required to fulfill the transcription repressor function. Knockdown of whiA does not affect actively growing M. gallisepticum , but leads to the growth retardation after freezing. Discussion: We propose the following model for M. gallisepticum WhiA function. WhiA remains bound to the core motif at any conditions. At low ATP concentrations (starvation) HEN domain binds auxiliary motif and represses rpsJ operon transcription. At high ATP concentrations (nutrient-rich conditions) HEN domain binds ATP and releases auxiliary motif. It leads to the de-repression of rpsJ operon and increased production of ribosomal proteins. We have demonstrated a simple all-fiber thulium (Tm) laser Q-switched by stimulated Brillouin scattering (SBS). The maximum output pulse energy was 80 μJ. This allowed us to generate a broadband spectrum directly at the laser outputs. For the first time, we measured the fine structure of the output pulses with a resolution of less than 100 ps. It was found that the SBS Q-switched laser is capable of generating bunches of picosecond pulses. The effect of modulation instability on the pulse decay is discussed. The potential application of the investigated laser radiation for producing destructive effects on soft biological tissues has been demonstrated. [ABSTRACT FROM AUTHOR]

The spectral and spatial output parameters of a two-channel, Yb-doped fiber laser operating in the intracavity spectral beam combining mode were investigated. We showed that by using active media with slightly different gain spectra, it is possible to implement either the spectral combining mode of the independent laser channels or the mode of collective lasing of the channels. The difference in the gain spectra of the active media was realized due to the difference in the threshold inverse population in the Yb-doped fibers. [ABSTRACT FROM AUTHOR]

We propose a simple dumbbell-shaped scheme of a Holmium-doped fiber laser incorporating a minimum number of optical elements. Mode-locking regimes were realized with the help of polymer-free single-walled carbon nanotubes (SWCNTs) synthesized using an aerosol (floating catalyst) CVD method. We show that such a laser scheme is structurally simple and more efficient than a conventional one using a ring cavity and a similar set of optical elements. In addition, we investigated the effect of SWCNT film transmittance, defined by the number of 40 nm SWCNT layers on the laser's performance: operating regimes, stability, and self-starting. We found that three SWCNT layers with an initial transmittance of about 40% allow stable self-starting soliton mode-locking at a wavelength of 2076 nm with a single pulse energy of 0.6 nJ and a signal-to-noise ratio of more than 60 dB to be achieved. Results of the study of the lateral surface of single-crystal (SC) sapphire fibers grown along crystallographic directions [0001] and [1120] by the LHPG method are presented. The appearance or absence of faceting of the lateral surface of the fibers depending on the growth direction is analyzed. The crystallographic orientation of the facets is investigated. The microstructure of the samples is investigated with the help of an optical microscope and a JSM-5910LV scanning electronic microscope (JEOL). The crystallographic orientations of the facets on the SC sapphire fiber surface are determined by electron backscatter diffraction (EBSD). The seed orientation is studied by means of XRD techniques. We present the results on the interaction of an all-fiber Holmium-doped laser CW radiation at a wavelength of 2100 nm with soft tissues and compare it with the other results obtained by the most used solid-state laser systems. Ex-vivo single spot experiments were carried out on the porcine longissimus muscles by varying the laser impact parameters in a wide range (average output power 0.3, 0.5 and 1.1 W; exposure time 5, 30 and 60 s). Evaluation of the laser radiation exposure was carried out by the size of coagulation and ablation zones on the morphological study. Exposure to a power of 0.3 W (1.5–18 J of applied energy) caused only reversible changes in the tissues. The highest applied energy of 66 J for 1.1 W and a 60-s exposure resulted in a maximum ablation depth of approximately 1.2 mm, with an ablation efficiency of 35%. We have shown that it is not necessary to use high powers of CW radiation, such as 5–10 W in the solid-state systems to provide the destructive effects. Similar results can be achieved at lower powers using the simple all-fiber Holmium laser based on the standard single-mode fiber, which could provide higher power densities and be more convenient to manufacture and use. The obtained results may be valuable as an additional experimental point in the field of existing results, which in the future will allow one to create a simple optimal laser system for medical purposes. Broadband NIR photoluminescence (from 1000 to 2500 nm) was observed from partially reduced AlCl₃/ZnCl₂/BiCl₃ glass, containing subvalent bismuth species. The luminescence consists of three bands, assigned to Bi⁺ , Bi₂⁴⁺, and Bi₅³⁺ ions. The physical and optical characteristics of these centers and possible contribution to NIR luminescence from bismuth-doped oxide glasses are discussed.
