Photosynthesis is the most temperature-sensitive process in the plant kingdom, but how the photosynthetic pathway responds during low-temperature exposure remains unclear. Herein, cold stress (4°C) induced widespread damage in the form DNA double-stranded breaks (DSBs) in the mesophyll cells of rice (Oryza sativa), subsequently causing a global inhibition of photosynthetic carbon metabolism (PCM) gene expression. Topoisomerase genes TOP6A3 and TOP6B were induced at 4°C and their encoded proteins formed a complex in the nucleus. TOP6A3 directly interacted with KU70 to inhibit its binding to cold-induced DSBs, which was facilitated by TOP6B, finally blocking the loading of LIG4, a component of the classic non-homologous end joining (c-NHEJ) pathway. The repression of c-NHEJ repair imposed by cold extended DSB damage signaling, thus prolonging the inhibition of photosynthesis in leaves. Furthermore, the TOP6 complex negatively regulated 13 crucial PCM genes by directly binding to their proximal promoter regions. Phenotypically, TOP6A3 overexpression exacerbated the γ-irradiation-triggered suppression of PCM genes and led to the hypersensitivity of photosynthesis parameters to cold stress, dependent on the DSB signal transducer ATM. Globally, the TOP6 complex acts as a signal integrator to control PCM gene expression and synchronize cold-induced photosynthesis inhibition, which modulates carbon assimilation rates immediately in response to changes in ambient temperature.

During the drilling of shale gas wells, the shape of shale cuttings cut by polycrystalline diamond compact bits has an obvious flaky structure. The accumulation of such flaky cuttings is one of the main reasons for high drilling torque and drag and serious back pressure, which affect the drilling speed of the horizontal section of shale gas wells. Therefore, predicting the thickness of the flaky cuttings bed and restraining it are essential for ensuring safe and efficient drilling of the horizontal section of shale gas wells. However, most of the previous research on migration laws for cuttings were based on spherical particles, which affected the accuracy of cuttings bed thickness prediction. In this study, through visualization experiments combined with computational fluid dynamics numerical simulation, the hole cleaning laws of shale gas wells in the long horizontal section were studied, and a set of cuttings bed thickness prediction models and cuttings bed removal process parameter optimization methods were established. Field application was conducted in the Changning 209H24-1 well in the southern Sichuan Basin, China. The results revealed the following: (1) There are significant differences in the migration laws of flaky cuttings and spherical cuttings in the horizontal section. (2) The fitting accuracy of the established long horizontal fragmentation layer distribution model considering multiple factors and the experimental data is as high as 0.973. (3) Based on this model, the drilling parameters of the Changning 209H24-1 well were optimized, which played a good role in wellbore cleaning and ensured the safe and smooth implementation of the later casing operation.

Purpose:The limited efficacy of chimeric antigen receptor (CAR) T-cell therapies with solid malignancies prompted us to test whether epigenetic therapy could enhance the antitumor activity of B7-H3.CAR T cells with several solid cancer types.Experimental Design:We evaluated B7-H3 expression in many human solid cancer and normal tissue samples. The efficacy of the combinatorial therapy with B7-H3.CAR T cells and the deacetylase inhibitor SAHA with several solid cancer types and the potential underlying mechanisms were characterized with in vitro and ex vivo experiments.Results:B7-H3 is expressed in most of the human solid tumor samples tested, but exhibits a restricted expression in normal tissues. B7-H3.CAR T cells selectively killed B7-H3 expressing human cancer cell lines in vitro. A low dose of SAHA upregulated B7-H3 expression in several types of solid cancer cells at the transcriptional level and B7-H3.CAR expression on human transgenic T-cell membrane. In contrast, the expression of immunosuppressive molecules, such as CTLA-4 and TET2, by T cells was downregulated upon SAHA treatment. A low dose of SAHA significantly enhanced the antitumor activity of B7-H3.CAR T cells with solid cancers in vitro and ex vivo, including orthotopic patient-derived xenograft and metastatic models treated with autologous CAR T-cell infusions.Conclusions:Our results show that our novel strategy which combines SAHA and B7-H3.CAR T cells enhances their therapeutic efficacy with solid cancers and justify its translation to a clinical setting.

Purpose:The overall biological roles and clinical significance of most long noncoding RNAs (lncRNA) in chemosensitivity are not fully understood. We investigated the biological function, mechanism, and clinical significance of lncRNA NR_034085, which we termed miRNA processing–related lncRNA (MPRL), in tongue squamous cell carcinoma (TSCC).Experimental Design:LncRNA expression in TSCC cell lines with cisplatin treatment was measured by lncRNA microarray and confirmed in TSCC tissues. The functional roles of MPRL were demonstrated by a series of in vitro and in vivo experiments. The miRNA profiles, RNA pull-down, RNA immunoprecipitation, serial deletion analysis, and luciferase analyses were used to investigate the potential mechanisms of MPRL.Results:We found that MPRL expression was significantly upregulated in TSCC cell lines treated with cisplatin and transactivated by E2F1. MPRL controlled mitochondrial fission and cisplatin sensitivity through miR–483-5p. In exploring the underlying interaction between MPRL and miR–483-5p, we identified that cytoplasmic MPRL directly binds to pre–miR–483 within the loop region and blocks pre–miR–483 recognition and cleavage by TRBP–DICER-complex, thereby inhibiting miR–483-5p generation and upregulating miR–483-5p downstream target-FIS1 expression. Furthermore, overexpression or knockdown MPRL altered tumor apoptosis and growth in mouse xenografts. Importantly, we found that high expression of MPRL and pre–miR–483, and low expression of miR–483-5p were significantly associated with neoadjuvant chemosensitivity and better TSCC patients’ prognosis.Conclusions:We propose a model in which lncRNAs impair microprocessor recognition and are efficient of pre-miRNA cropping. In addition, our study reveals a novel regulatory network for mitochondrial fission and chemosensitivity and new biomarkers for prediction of neoadjuvant chemosensitivity in TSCC.These findings uncover a novel mechanism by which lncRNA determines mitochondrial fission and cisplatin chemosensitivity by inhibition of pre-miRNA processing and provide for the first time the rationale for lncRNA and miRNA biogenesis for predicting chemosensitivity and patient clinical prognosis.

miRNAs that translocate from the nucleus to mitochondria are referred to as mitochondrial microRNAs (mitomiR). mitomiRs have been shown to modulate the translational activity of the mitochondrial genome, yet their role in mitochondrial DNA (mtDNA) transcription remains to be determined. Here we report that the mitomiR-2392 regulates chemoresistance in tongue squamous cell carcinoma (TSCC) cells by reprogramming metabolism via downregulation of oxidative phosphorylation and upregulation of glycolysis. These effects were mediated through partial inhibition of mtDNA transcription by mitomiR-2392 rather than through translational regulation. This repression required specific miRNA–mtDNA base pairing and Argonaute 2. mitomiR-2392 recognized target sequences in the H-strand and partially inhibited polycistronic mtDNA transcription in a cell-specific manner. A retrospective analysis of TSCC patient tumors revealed a significant association of miR-2392 and regulated mitochondrial gene expression with chemosensitivity and overall survival. The clinical relevance of targeted mitochondrial genes was consistently validated by The Cancer Genome Atlas RNA sequencing in multiple types of cancer. Our study revealed for the first time the role of mitomiR in mtDNA transcription and its contribution to the molecular basis of tumor cell metabolism and chemoresistance.Significance: These findings uncover a novel mechanism by which mitomiRNA regulates mitochondrial transcription and provide rationale for use of mitomiRNA and mtDNA-encoded genes to predict chemosensitivity and patient clinical prognosis.

Recognizing various human actions in video is a challenging task, which is also one of the key tasks in computer vision. It has received extensive attention from AI researchers Bakker et al. (2003), Bruderlin and Williams (1995), Cardle et al. (2003), Carlsson (1996, 1999), Clausen and Kurth (2004). It has important applications in human behavior analysis, artificial intelligence, and video surveillance. Compared with still image classification, the time component of video provides important clues for recognition, so multiple human actions can be recognized based on motion information. In addition, video provides natural data enhancement for individual images. For motion recognition from videos, appearance and temporal dynamics are two key and complementary cues. In this work, we formulate a human motion recognition framework based on 2D spatial feature fusion of Kinect bone data. This method utilizes the human body structure and spatial geometry to represent human body structure and extract features. Meanwhile, it combines the active action and auxiliary action features of two spatial dimensions by layering, and leverages the pervasively used support vector machine and hidden Markov model to classify human body structure. Noticeably, it is difficult to extract its information due to the limitations of background clutter, viewpoint change, scale change, different lighting conditions and camera movement. In this way, while learning the classification information of human behavioral categories, designing effective representations is the key to deal with these challenges. In this work, a method for human motion recognition in video based on ResNext network is proposed. Based on our proposed ResNeXt network, using the data of RGB and optical flow, we can extract more appearance characteristics and temporal features of human actions, so as to better achieve the classification of actions. The utilization of video time segmentation method can capture the long range of time in video, in order to make better use of the longer range of time information in video. Comprehensive experimental results have shown that the proposed method improves the performance of UCF101 and HMDB51 motion recognition data sets to a certain extent.

Based on Recursive Radial Basis Function (RRBF) neural network, the Reduced Order Model (ROM) of compressor cascade was established to meet the urgent demand of highly efficient prediction of unsteady aerodynamics performance of turbomachinery. One novel ROM called ASA-RRBF model based on Adaptive Simulated Annealing (ASA) algorithm was developed to enhance the generalization ability of the unsteady ROM. The ROM was verified by predicting the unsteady aerodynamics performance of a highly-loaded compressor cascade. The results show that the RRBF model has higher accuracy in identification of the dimensionless total pressure and dimensionless static pressure of compressor cascade under nonlinear and unsteady conditions, and the model behaves higher stability and computational efficiency. However, for the strong nonlinear characteristics of aerodynamic parameters, the RRBF model presents lower accuracy. Additionally, the RRBF model predicts with a large error in the identification of aerodynamic parameters under linear and unsteady conditions. For ASA-RRBF, by introducing a small-amplitude and high-frequency sinusoidal signal as validation sample, the width of the basis function of the RRBF model is optimized to improve the generalization ability of the ROM under linear unsteady conditions. Besides, this model improves the predicting accuracy of dimensionless static pressure which has strong nonlinear characteristics. The ASA-RRBF model has higher prediction accuracy than RRBF model without significantly increasing the total time consumption. This novel model can predict the linear hysteresis of dimensionless static pressure happened in the harmonic condition, but it cannot accurately predict the beat frequency of dimensionless total pressure.

Supersonic shear layer development affects mixing between the primary and secondary fluids in a supersonic ejector substantially. The splitter plate plays a key role in shear layer development in the near-trailing-edge region. In this paper, supersonic shear layers with various splitter plate thicknesses are calculated and analyzed. Two types of instability modes are found when various splitter plate thicknesses are used. The shock wave strength and absolute unstable region determine the instability modes. Shock waves can limit instability development but an absolute unstable region promotes instability development. Velocity thickness growth rates are found to be the result of the merging process between two or more adjacent vortices in the transition region. The number of vortices that merge depends on the ratio of the vortex shedding and nature frequencies. The velocity thickness growth rates are larger under the local instability mode than under the global instability mode since three vortices merge under the former mode but two vortices merge under the latter. This study may provide a helpful reference for the design of supersonic ejectors.

The transforming growth factor-β (TGF-β)/Smads signal plays an important role in cancer metastasis by mediating the epithelial-mesenchymal transition (EMT) in cancer cells. lnc-TSI is a recently identified long noncoding RNA that negatively regulates the TGF-β/Smads signal. The present study was conducted to test the hypothesis that lnc-TSI inhibits metastasis in clear cell renal cell carcinoma (ccRCC) by regulating the TGF-β/Smad3 pathway. Herein, we show that lnc-TSI was upregulated in ccRCC cells and tissue and was associated with activation of the TGF-β/Smads signal. Depleting lnc-TSI enhanced tumor cell invasion and metastasis in vitro and ccRCC lung metastasis in vivo, whereas overexpressing lnc-TSI inhibited ccRCC cell invasion and tumor metastasis. Mechanistic studies indicated that lnc-TSI specifically inhibited the phosphorylation of Smad3 and subsequent EMT by binding with the MH2 domain of Smad3 to block the interaction between Smad3 and TGF-β receptor I in ccRCC cells. In a cohort of 150 patients with ccRCC, expression of lnc-TSI in tumors was negatively correlated with phosphorylated (p)Smad3 and activated EMT markers. Patients with expression of tumor lnc-TSI greater than or equal to the median at radical nephrectomy had a higher survival rate compared to those with lnc-TSI below the median during follow-up. These findings reveal a new regulatory mechanism of ccRCC metastasis and suggest a potential molecular target for the development of anti-cancer drugs., Graphical Abstract, TGF-β/Smad3 signaling has been found to play an important role in cancer metastasis. Wang et al. show that lncRNA lnc-TSI inhibits cancer cell invasion and tumor metastasis via specifically inhibiting the phosphorylation of Smad3 and subsequent EMT, which could be potentially applied in treatments for cancer metastasis.

Head and neck squamous cell carcinoma (HNSCC), which occurs frequently worldwide, is characterized by high risk of metastasis. MicroRNAs (miRNAs) play crucial roles in tumorigenesis and cancer development. In this study, miR-29c-5p was identified using three high throughput microarrays. We measure miR-29c-5p expression in HNSCC tissues and cell lines. To determine the function of miR-29c-5p in HNSCC, we evaluated its effects in vitro on cell proliferation, the cell cycle, apoptosis, and cell migration. We employed a mouse tumor xenograft model to determine the effects of miR-29c-5p on tumors generated by HNSCC cell lines. The miR-29c-5p expression was lower in HNSCC tissues than in normal tissues. Upregulated miR-29c-5p expression in HNSCC cells inhibited migration and arrested cells in the G2/M phase of the cell cycle. Further, upregulated miR-29c-5p expression inhibited the proliferation of HNSCC cells in vivo and in vitro. In addition, transmembrane protein 98 (TMEM98) was identified as a direct target of miR-29c-5p by using a luciferase reporter assay. These findings provide new insights that link the regulation of miR-29c-5p expression to the malignant phenotype of HNSCC and suggest that employing miR-29c-5p may serve as a therapeutic strategy for managing patients with HNSCC.

Laryngeal carcinoma (LCC) is a common malignant tumor with low radiosensitivity and generally poor response rates. The ubiquitin protein ligase E3 component n-recognin 5 (UBR5) has prognostic implications in several neoplasms; however, its role in LCC and radiotherapy sensitivity remains unknown. Immunohistochemistry and bioinformatics analyses were performed to measure UBR5 protein and mRNA expression in LCC and adjacent non-tumor tissues. The gene and protein expression of UBR5 in LCC and HuLa-PC cell lines were measured using quantitative PCR and western blot analyses. Following transfection with small interfering RNA or UBR5 overexpression plasmid in LCC cells, the proliferation, cell cycle distribution, invasion, migration and radiosensitivity of LCC cells were analyzed. UBR5-related lncRNA, targeted miRNA and protein-protein interaction networks were analyzed using bioinformatics. Finally, the expression of the p38/mitogen-activated protein kinase (MAPK) pathway was evaluated following UBR5 silencing in M2E cells treated with radiation. Increased UBR5 expression was observed in LCC tissues compared with adjacent non-tumor tissues, and it was correlated with poor overall survival of LCC patients. After overexpression or silencing of UBR5 in M2E and M4E LCC cells, cell proliferation and radiosensitivity were significantly increased or decreased, respectively, compared with the control groups. The percentage of S phase cells decreased in the UBR5 si-RNA group compared with that in the control group, while overexpression of UBR5 exerted no effect on the cell cycle. In addition, the expression of Bcl-2 and p38 was decreased in the si-UBR5 combined with radiation groups. The level of phosphorylated p38 expression was increased after combination of si-UBR5 with radiation. The small molecule inhibitor of p38/MAPK signaling, SB203580, decreased the viability of UBR5-overexpressing cells and the survival fraction when cells were exposed to radiation. These findings demonstrated that UBR5 may be involved in regulating cell proliferation and sensitivity to radiotherapy in LCC via the p38/MAPK pathway, thereby highlighting its possible value for the development of new therapeutic strategies and targets for the treatment of this disease.

The present work investigated the vortex structure and fluctuation frequency characteristics generated by boundary layer separation of a high-load compressor cascade using modal decomposition methods. The dominant modes and dynamic behaviors of unsteady flow in the cascade were obtained, and the differences of three modal decomposition methods (Proper Orthogonal Decomposition, Dynamic Mode Decomposition and Spectral Proper Orthogonal Decomposition) in feature recognition of cascade flow were discussed. The results show that:(1) The POD method can accurately extract the dominant spatial structure of the flow field, but the modal coefficients are multi-frequency coupled, which makes the dominant modal characteristics of cascade flow unclear. (2) The standard DMD method can obtain the spatial-temporal single frequency mode of cascade flow, as well as their growth rates and frequencies. However, this method is likely to capture the suboptimal mode of large amplitude with large attenuation rate, and fails to obtain the high-frequency coherent structure, which makes it impossible to obtain the dominant feature with limited mode number. (3) The SPOD method, based on spectral characteristics, can obtain spatial and temporal single frequency modes, and there is no modal screening problem. The use of spectral estimation method (SPOD) reduces the sensitivity to numerical noise. This method can obtain the low-rank behavior of cascade flow, which is helpful to understand cascade flow mechanisms. Therefore, SPOD method is more advantageous for the modal analysis of unsteady separated flow in high-load compressor cascade.

This paper has established an evaluation system for the sensitivity analysis of cascade/compressor aerodynamic performance to geometric deviations. The system includes blade profile parameterization with non-uniform rational B-spline (NURBS), uniform Latin hypercube sampling in association with Monte Carlo algorithm and statistical investigation methods. The sensitivity of a subsonic cascade performance to its geometric deviations is investigated under different typical inflow conditions. Results show that there is an approximately linear relationship between the geometric deviations and the aerodynamic parameters. The total pressure loss coefficient is the most sensitive to the leading edge profile deviation at i=2°. However the suction profile deviation and the leading edge profile deviation dominate the total pressure loss when the incidences are i = −4° and i = −8°. The blocking effect of the increased leading edge profile moves forward the separation point of the boundary layer. Therefore the scale of the separation vortex is enlarged, the total pressure loss is increased at i = 2° condition. Regarding i = −8°, the enlarged suction profile deviation reduces the circumferential static pressure gradient, resulting in an increase of the scale of separation vortex on the pressure side. The work is helpful to robust design of compressor blade profile and manufacturing & maintenance techniques of compressor blades.

When injectors are used in petroleum industries or sea water desalination system, it is a special case that the induced gas is accompanied by water, the ejector is in two-phase operation. In the present study, the ejector performance at two phase flow is investigated experimentally and numerically. The results indicate that: the entrainment ratio would decrease when the water is injected into the induced flow, as well as the pressure ratio. The maximum decrement of entrainment ratio is around 40.5% at sub-critical mode when the motive pressure is 1.0 MPa, the induced pressure is 0.2 MPa, liquid-air ratio is 22%. For the pressure ratio, the maximum decrement is 62.5% when the motive pressure is 1.0 MPa, the induced pressure is 0.5 MPa, liquid-air ratio is 2.87%. However, the lower the induced pressure is, the more obvious the effect of mass flow rate on entrainment rate is, while the effect on pressure ratio is just opposite. Moreover, the performance becomes worse in sub-critical mode than that in critical mode. The effect of NXP on ejector performance also has been analyzed. There exists an optimum NXP whether the two phase flow ejector is in critical mode or sub-critical mode. The optimum NXP for entrainment ratio enlarges from 3.6 mm to 4.4 mm with the increase of water mass flowrate, and is bigger than that of single phase ejector (the optimum NXP is 2.8 mm). This study may provide a beneficial reference for the operation of supersonic ejectors on two phase flow.

To provide a high-strength, non-resin organic plugging agent for water shutoff in carbonate fracture and cave reservoirs, a high-strength gel was prepared with polyvinyl alcohol 1788 as gelant and p-phthalaldehyde as crosslinker in this study. By selecting latent acids rather than strong acid such as hydrochloric acid and sulfuric acid as catalysts, and by adding clay and sulphonated phenolic resin as stabilisers to the gelation, the drawbacks of short gelation time of polyvinyl alcohol and high dehydration rate of the formed gel soaking in brine were overcame. The evaluation results showed that the gellant composed of 7wt% polyvinyl alcohol, 4wt% clay, 0.3wt% aldehyde crosslinker, 0.25wt% latent acid, 1.5wt% sulfonated phenolic resin, and 0.3wt% hexamethylenetetramine had a gelation time of 2 h at 130 °C. The storage modulus of the gel was up to 2000 Pa, and the dehydration rate of the gel after soaking in synthetic brine for 30 days was less than 15%. Furthermore, the breakthrough pressure gradient in a slim tube with a diameter of 3mm reached 15 MPa / m, which indicated that the gel is a temperature and salinity resistant plugging agent with strong plugging ability.

The shortage of freshwater resources is becoming increasingly serious. A thermal power plant coupled with a low-temperature multi-effect distillation seawater desalination (single-stage thermal vapor compressor (TVC)) has been widely used. To reduce the water production cost effectively, a two-stage TVC is proposed in this study. The simulation work for the system performance under single- and two-stage TVC is analyzed. Results show that the performance of the system coupled with the two-stage TVC is superior under most conditions, and an optimal intermediate pressure (16 kPa) of the two-stage TVC is found for minimum coal consumption. When the power plant load is higher than 70% THA, the cogeneration system performs well, and the motive steam of the two-stage TVC comes from extraction point No. 5. Otherwise, the motive steam that comes from extraction point No. 4 is a suitable choice. The maximum coal consumption reduction value is 1.48 t/h. To deal with the low operation load of power plants in China, 60% THA load of power plant is a suitable choice for two-stage TVC of power-water cogeneration system.

As the evaluation of distributed energy system (DES) is a complex problem, multi-criteria is necessary to fully evaluate the performance for different systems. In this paper, a novel multi-criteria evaluation method was established to assess the comprehensive performance of DES from economic, energy and environment criterions. Before constructing a multi-level indexes system, index correlation analysis was carried out to ensure the independence of indexes. Considering the advantages and disadvantages of subjective and objective methods, AHP-entropy method was proposed by integrating analytic hierarchy process (AHP) and information entropy method together to assign weights to the indexes. Finally, the technique for order preference by similarity to ideal solution (TOPSIS) method was applied to determine the optimal DES scheme. The case study indicated that the evaluation results of this method were in accordance with the reality, which could provide a reference for the selection of DES solutions.

MicroRNAs (miRNAs) have been recently found in the mitochondria, and were named “mitomiRs”, but their function has remained elusive. Here, we aimed to assess the presence and function(s) of mitomiRs in tongue squamous cell carcinoma (TSCC). Methods: miRNA microarray was performed in paired TSCC cell lines, Cal27 and its chemoresistant counterpart, Cal27-re. Decreased expression of mitomiRs in chemoresistant cells was characterized. The functions of mitomiRs were investigated by a series of in vitro and in vivo experiments. Results: Differential microarray analysis identified downregulation of mitomiR-5787 in Cal27-re cells. We knocked down mitomiR-5787 in parental cells and upregulated its expression in cisplatin-resistant cells. The sensitivity of TSCC cells to cisplatin was regulated by miR-5787. The glucose metabolism assay suggested that reduced expression of miR-5787 changed the balance of glucose metabolism by shifting it from oxidative phosphorylation to aerobic glycolysis. Xenograft experiments in BALB/c-nu mice further verified the in vitro results. Reduced expression of miR-5787 contributes to chemoresistance in TSCC cells by inhibiting the translation of mitochondrial cytochrome c oxidase subunit 3 (MT-CO3). The prognostic analysis of 126 TSCC patients showed that the patients with low expression of miR-5787 and/or MT-CO3 had poor cisplatin sensitivity and prognosis. Conclusions: Mitochondrial miR-5787 could regulate cisplatin resistance of TSCC cells and affect oxidative phosphorylation and aerobic glycolysis. Downregulation of miR-5787 inhibited the translation of MT-CO3 to regulate cisplatin resistance of TSCC. Mitochondrial miR-5787 and MT-CO3 can be used as predictive biomarkers or therapeutic targets for cisplatin chemotherapy resistance.

The geometrical aberration of an optical system is usually measured through wavefront aberration. We propose a very direct experimental ray-tracing measurement method in this paper. Two fiber point-light-sources are used to simulate an arbitrary incident ray in space, the diffracting wavefronts of the two fibers pass through the lens under test and interfere on two photographic planes. The outgoing ray intersects the photographic planes at the extrema of their phase maps, thus can be traced by connecting the two intersections. We measure four kinds of geometrical aberrations of the lens and compare them with Zemax simulation results. This method provides a new solution for the geometrical aberration measurement of an optical system.
