Aims: The electroencephalography (EEG) microstates are indicative of fundamental information processing mechanisms, which are severely damaged in patients with prolonged disorders of consciousness (pDoC). We aimed to improve the topographic analysis of EEG microstates and explore indicators available for diagnosis and prognosis prediction of patients with pDoC, which were still lacking. Methods: We conducted EEG recordings on 59 patients with pDoC and 32 healthy controls. We refined the microstate method to accurately estimate topographical differences, and then classify and forecast the prognosis of patients with pDoC. An independent dataset was used to validate the conclusion. Results: Through optimized topographic analysis, the global explained variance (GEV) of microstate E increased significantly in groups with reduced levels of consciousness. However, its ability to classify the VS/UWS group was poor. In addition, the optimized GEV of microstate E exhibited a statistically significant decrease in the good prognosis group as opposed to the group with a poor prognosis. Furthermore, the optimized GEV of microstate E strongly predicted a patient's prognosis. Conclusion: This technique harmonizes with the existing microstate analysis and exhibits precise and comprehensive differences in microstate topography between groups. Hop (Humulus lupulus) is a perennial plant with commercial values. Here, we reported the complete chloroplast genome for a local hop cultivar (Humulus lupulus cv. 'Fubei-1') from Xinjiang, China. The chloroplast genome is 153,614 bp long with an A + T-biased base composition, and contains a total of 113 gene species, including 79 protein-coding, 30 tRNA, and four rRNA gene species. Nineteen gene species are duplicated, and 18 gene species harbor one or two introns. Phylogenetic analysis revealed close relatedness among the three hop cultivars ('Saazer', 'Hallertauer', and 'Fubei-1'). [ABSTRACT FROM AUTHOR]

Eight healthy multiparous Saanen dairy goats (41.50 ± 1.84 kg) were assigned to a double 4 × 4 Latin square design. The four treatment diets were: (i) negative control, rice straw (NC); (ii) first positive control, sticky corn stover silage (PC1); (iii) second positive control, PC1 with 1 g/d commercial purple corn pigment (PC2); and (iv) anthocyanin-rich purple corn stover silage (PSS; AR). DMI did not differ (P > 0.05) among the treatments. Goats receiving the NC tended (P < 0.05) to reduce nutrient apparent digestibility, nitrogen (N) absorption, N retention, and volatile fatty acid production relative to the other groups. The levels of 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and superoxide dismutase (SOD) in plasma were greater (P < 0.05) in goats fed PC2 and AR compared with NC and PC1. The inclusion of PC2 and AR increased (P < 0.05) the abundance of nuclear factor (erythroid-derived 2)-like 2 (NFE2L2), but decreased (P < 0.05) the level of tumor necrosis factor in the mammary gland. Moreover, goats receiving AR tended to increase (P < 0.05) the levels of SOD2, GPX1, and GPX2 mRNA expression in the mammary gland. There were significant (P < 0.05) positive correlations between DPPH scavenging activity, total antioxidant capacity, SOD, catalase enzymes in plasma, and the abundance of NFE2L2 in the mammary gland. In addition, stronger (P < 0.05) positive correlations were noted between the expression of several inflammation related and antioxidant genes. Collectively, the results from the current study indicated that the consumption of anthocyanin-rich PSS by dairy goats had the potential to enhance antioxidant potential by improving antioxidant capacity in plasma and by modulating the abundance of several inflammation related and antioxidant genes in the mammary gland. Context2a,-3a,-24-Trihydroxyurs-12-en-28-oic acid (TEO, a corosolic acid analogue) is a triterpenoid saponin isolated fromActinidia valvataDunn (Actinidiaceae), a well-known traditional Chinese medicine. ObjectiveThis study investigated the anti-proliferation and inducing apoptosis effects of TEO in three human hepatocellular carcinoma (HCC) cell lines. Materials and methodsCytotoxic activity of TEO was determined by the MTT assay at various concentrations from 2.5 to 40 μg/mL in BEL-7402, BEL-7404 and SMMC-7721 cell lines. Cell morphology was assessed by acridine orange/ethidium bromide and 4′-6-diamidino-2-phenylindole dihydrochloride staining and fluorescence microscopy. Cell-cycle distribution and DNA damage were determined by flow cytometry and comet assay. Mitochondrial dysfunction was assessed by JC-1 staining and transmission electron microscopy. Apoptosis changes were explored by Western blot, TNF-α and caspase-3, -8, -9 assays. ResultsTEO exhibited inhibition effects on BEL-7402, BEL-7404 and SMMC-7721 cells treated for 24 h, the IC50values were 34.6, 30.8 and 30.5 μg/mL, respectively. TEO (40 μg/mL)-treated three cell lines increased by more than 21% in the G1 phase and presented the morphological change and DNA damage. TEO also declined the mitochondrial membrane potential and altered mitochondrial ultra-structure. Furthermore, caspase-3, caspase-8, caspase-9 and TNF-α were also activated. Mechanism investigation showed that TEO could decrease anti-apoptotic Bcl-2 protein expression, increase proapoptotic Bax and Bid proteins expressions and increase Bax/Bcl-2 ratio. ConclusionOur results demonstrate for the first time that TEO inhibited growth of HCC cell lines and induced G1 phase arrest. Myocardial ischemia/reperfusion (I/R) injury is a key factor in deterioration of myocardial function. The c-Jun NH2-terminal kinase (JNK) activation and the transcription factor nuclear factor-kappaB (NF- κ B) nuclear translocation have been found in I/R injury. 6-Gingerol, an important bioactive ingredient of ginger, has been reported to have cardiovascular pharmacological effects. However, the molecular mechanism through which it is beneficial is unclear. In this work, I/R induced the increase in the apoptosis and reactive oxygen species level in AC16 cardiomyocytes. 6-Gingerol administration decreased cardiomyocyte apoptosis and improved oxidative stress indexes. 6-Gingerol administration also inhibited I/R-induced HMGB2 expression upregulation and JNK activation and reduced Cleaved Poly(ADP-ribose) polymerases (PARP) and Caspase-3 expression. HMGB2 treatment mimicked the effect of I/R-induced cell damage, which was reversed by 6-gingerol administration. On the other hand, transcriptional activity of NF- κ B was reduced in 6-gingerol treated cells. Thus, overall results indicated that 6-gingerol administration protected I/R-induced cardiomyocytes apoptosis via JNK/NF- κ B pathway in the regulation of HMGB2. This work supported the efficacy of 6-gingerol on cardiovascular disease and partially revealed its mechanism, which was helpful for understanding the therapeutic effects of this natural drug.

The protective activity of the total saponins from Actinidia valvata Dunn root (TSAV) was studied against carbon-tetrachloride- (CCl(4)-) induced acute liver injury in mice. Mice were orally administered TSAV (50, 100, and 200 mg/kg) for five days and then given CCl(4). TSAV pretreatment significantly prevented the CCl(4)-induced hepatic damage as indicated by the serum marker enzymes (AST, ALT, and ALP). Parallel to these changes, TSAV also prevented CCl(4)-induced oxidative stress by inhibiting lipid peroxidation (MDA) and restoring the levels of antioxidant enzymes (SOD, CAT, GR, and GPX), GSH and GSSG. In addition, TSAV attenuated the serum TNF-α and IL-6 levels and inhibited the serum iNOS and NO levels. Liver histopathology indicated that TSAV alleviated CCl(4)-induced inflammatory infiltration and focal necrosis. TSAV (200 mg/kg) also significantly decreased Bak, Bax mRNA and Fas, FasL, p53, and NF-κB p65 protein expressions and increased Bcl-2 mRNA and protein expressions. Meanwhile, TSAV significantly downregulated caspase-3 and caspase-8 activities and prevented CCl(4)-induced hepatic cell apoptosis. In addition, TSAV exhibited antioxidant activity through scavenging hydroxyl and DPPH free radicals in vitro. These results indicated that TSAV could protect mice against CCl(4)-induced acute liver damage possibly through antioxidant, anti-inflammatory activities and regulating apoptotic-related genes.
