Abstract

This thesis investigates Gu Hongming's English translation of three Confucian classics Lunyu (1898), Zhongyong (1906) and Daxue (1915). Rather than regarding Gu's work as extreme domestication, this study reinterprets it as a hybrid translation by considering his ambiguous view of culture in a specific historical context. The focus is on how Gu translated China to the Anglophone world in the late 19th and early 20th centuries, which is examined through a transdisciplinary approach of postcolonialism, cultural studies, diaspora studies and comparative literature, and a combination of contextual and textual analyses. This study also draws on a variety of primary materials, such as Gu's English writings, periodicals, and archival materials of his private correspondence with James Stewart Lockhart and Richard Wilhelm, in order to unfold what role translation played in re-envisioning the cultural exchange between China and Anglophone countries. This thesis explores the multi-layered dimensions of Gu's hybrid translation from three perspectives: diasporic, postcolonial and linguo-cultural. It examines the role of the translator by analyzing how Gu's diasporic experience structured his ambiguous and ambivalent cultural consciousness, which influenced the way in which he translated Confucian classics. Through a postcolonial reading of Gu's motivation, thought, as well as religious and political rendition of Confucian texts, this thesis investigates the complexity of hybrid translation as a negotiating process involving tension, resistance, ambiguity, interplay and creation. This study further analyzes how Gu's distinctive hybrid strategy reveals the dynamics of translation, and indicates the ethical appeal of Self and Other. Moreover, this thesis explores how Gu's hybrid translation reflects on various themes such as linguo-cultural (in)commensurability, (un)translatability, and the relationship between foreignization and domestication. As a case study, this thesis probes into hybridity as a constructive yet ruptured term in translation studies.

6. M1 Microglia-Derived Exosomes Promote A1 Astrocyte Activation and Aggravate Ischemic Injury via circSTRN3/miR-331-5p/MAVS/NF-κB Pathway.

Abstract

Background: After ischemic stroke (IS), microglia and astrocytes undergo polarization, transforming into a pro-inflammatory phenotype (M1 or A1). According to previous studies, exosomes might play an important role in the interplay between M1 microglia and A1 astrocytes after IS. Methods: We used the microglial oxygen-glucose deprivation/reperfusion (OGD/R) model and ultracentrifugation to extract M1 microglial exosomes (M1-exos). Subsequently, we identified circSTRN3 enriched in exosomes through RNA sequencing and detected the role of circSTRN3 in astrocyte activation based on bioinformatics analysis, immunofluorescence, Western blotting, and polymerase chain reaction analysis. We validated these findings in the middle cerebral artery occlusion/reperfusion (MCAO/R) model of adult male C57BL/6J mice. Finally, we confirmed the correlation among circSTRN3, miR-331-5p, and stroke severity score in exosomes isolated from peripheral blood of IS patients. Results: Our findings revealed that M1-exos promoted A1 astrocyte activation. CircSTRN3 was abundant in M1-exos, which could sponge miR-331-5p to affect mitochondrial antiviral signaling protein (MAVS), activate NF-κB pathway, and participate in A1 astrocyte activation. In addition, overexpressed circSTRN3 augmented the infarct size and neurological dysfunction in MCAO/R models, while miR-331-5p mimics reversed the effect. Furthermore, circSTRN3 in IS patients was positively correlated with stroke severity score (R2 = 0.83, P < 0.001), while miR-331-5p demonstrated a negative correlation with the same score (R2 = 0.81, P < 0.001). Conclusion: Taken together, our research indicated that circSTRN3 from M1-exos could promote A1 astrocyte activation and exacerbate ischemic brain injury via miR331-5p/MAVS/NF-κB axis. We sought to explore whether serum NSE concentration in ischemic stroke patients undergoing mechanical thrombectomy (MT) is related to 3-month functional outcome and symptomatic intracranial hemorrhage (sICH).Patients and Methods: We retrospectively collected the data of acute ischemic stroke patients with anterior circulation infarction receiving MT within 6 h in our stroke center. Favorable outcome and poor outcome at 3 months were defined as modified Rankin Scale (mRS) score 0– 2 and 3– 6, respectively. sICH was defined according to the Heidelberg bleeding classification. We used multivariate logistic regression model and receiver operating characteristic curves to investigate the correlation between NSE and clinical outcomes.Results: Among the 426 patients enrolled, 40 (9.4%) patients developed sICH. This study aimed to determine the relationship between the novel metabolic score for insulin resistance (METS-IR) and symptomatic intracranial hemorrhage (sICH) after endovascular thrombectomy (EVT) in stroke patients.Methods: We retrospectively included patients with large artery occlusion in the anterior circulation and treated by EVT from 2 stroke centers (Nanjing First Hospital from September 2019 to April 2022, and Jinling Hospital from September 2019 to July 2021). The METS-IR was used as an alternative marker of insulin resistance and calculated using laboratory data after admission. sICH was diagnosed according to the Heidelberg Bleeding Classification.Results: Of the 410 enrolled patients (mean age, 69.8 ± 11.7 years; 60.7% men), 50 (12.2%) were diagnosed as sICH. After adjusting for demographic characteristics, poor collateral status, and other potential confounders, higher METS-IR was revealed to be independently associated with sICH (odds ratio, 1.076; 95% confidence interval, 1.034− 1.120; P = 0.001). Abstract

Organ transplantation has become a powerful strategy for the treatment of malignant diseases. Nevertheless, graft rejection is one of the main factors affecting graft survival after organ transplantation. Under this circumstance, the transplant-related mortality still keeps up. This invention includes the precise medication guidance of Tacrolimus (FK506) inapplicable population, against the side-effects of this drug. This invention, based on second-generation sequencing, has the advantages of relatively low cost and high sequencing throughput. During the design process, we collect the data of single nucleotide polymorphism (SNP) concerning the adverse drug reactions of Tacrolimus. Then we filter and summarize fifteen SNPs basing on importance degree (level >key enzyme>race). Thenceforth, after the process of analyzing the raw extract by operating BWA, Picard-tools, GATK, and Perl, we annotate SNPs by Annovar. 15. Non-coding RNAs in acute ischemic stroke: from brain to periphery.

Abstract

Acute ischemic stroke is a clinical emergency and a condition with high morbidity, mortality, and disability. Accurate predictive, diagnostic, and prognostic biomarkers and effective therapeutic targets for acute ischemic stroke remain undetermined. With innovations in high-throughput gene sequencing analysis, many aberrantly expressed non-coding RNAs (ncRNAs) in the brain and peripheral blood after acute ischemic stroke have been found in clinical samples and experimental models. Differentially expressed ncRNAs in the post-stroke brain were demonstrated to play vital roles in pathological processes, leading to neuroprotection or deterioration, thus ncRNAs can serve as therapeutic targets in acute ischemic stroke. Moreover, distinctly expressed ncRNAs in the peripheral blood can be used as biomarkers for acute ischemic stroke prediction, diagnosis, and prognosis. In particular, ncRNAs in peripheral immune cells were recently shown to be involved in the peripheral and brain immune response after acute ischemic stroke. In this review, we consolidate the latest progress of research into the roles of ncRNAs (microRNAs, long ncRNAs, and circular RNAs) in the pathological processes of acute ischemic stroke-induced brain damage, as well as the potential of these ncRNAs to act as biomarkers for acute ischemic stroke prediction, diagnosis, and prognosis.
