14. Stigmasterol alleviates allergic airway inflammation and airway hyperresponsiveness in asthma mice through inhibiting substance-P receptor.

Abstract

Stigmasterol has significant anti-arthritis and anti-inflammatory effects, but its role in immune and inflammatory diseases is still unclear. The potential advantages of stigmasterol in asthma were explored in IL-13-induced BEAS-2B cells and asthmatic mice. The optimal target of stigmasterol was confirmed in asthma. After detecting the cytotoxicity of stigmasterol in BEAS-2B cells, 10 μg/mL and 20 μg/mL stigmasterol were incubated with the BEAS-2B cell model for 48 h, and anti-inflammation and antioxidative stress were verified. Asthmatic mice were induced by OVA and received 100 mg/kg stigmasterol for 7 consecutive days. After 28 days, lung tissues and BAL fluid were collected for the following study. To further verify the role of NK1-R, 0.1 μM WIN62577 (NK1-R specific antagonist), and 1 μM recombinant human NK1-R protein were applied. NK1-R was the potential target of stigmasterol. When the concentration of stigmasterol is 20 μg/mL, the survival rate of BEAS-2B cells is about 98.4%, which is non-toxic. Stigmasterol exerted anti-inflammation and antioxidant stress in a dose-dependent manner and decreased NK1-R expression in IL-13-induced BEAS-2B. Meanwhile, in vivo assay also indicated the anti-inflammation and antioxidant stress of stigmasterol after OVA challenge. Stigmasterol inhibited inflammation infiltration and mucus hypersecretion, and NK1-R expression. The protective effect of stigmaterol on asthma and its underlying mechanism have been discussed in depth, providing a theoretical basis and more possibilities for its treatment of asthma. Abstract

This work described the one-pot synthesis of orange pectin encapsulated Fe3O4 nanoparticles (Fe3O4@Pectin NPs) which is prepared by co-precipitation of Fe(II/(III) ions in alkaline solution mediated by pectin. This process led to formation of magnetic nanoparticles within the network of pectin. Physicochemical characterization of the as-synthesized Fe3O4@Pectin NPs was carried out through Fourier transformed infrared spectroscopy (FT-IR), electron microscopy (SEM and TEM), energy dispersive X-ray spectroscopy (EDX), vibrating sample magnetometer (VSM) and X-ray diffraction (XRD). The in vitro cytotoxic and anti-liver cancer effects of biologically synthesized Fe3O4@Pectin NPs against pleomorphic hepatocellular carcinoma (SNU-387), hepatic ductal carcinoma (LMH/2A), morris hepatoma (McA-RH7777), and novikoff hepatoma (N1-S1 Fudr) cancer cell lines were assessed. The anti-liver cancer properties of the Fe3O4@Pectin NPs could significantly remove pleomorphic hepatocellular carcinoma (SNU-387), hepatic ductal carcinoma (LMH/2A), morris hepatoma (McA-RH7777), and novikoff hepatoma (N1-S1 Fudr) cancer cell lines in a time and concentration-dependent manner by MTT assay. The IC50 of the Fe3O4@Pectin NPs were 8, 13, 10, and 7 µg/mL against pleomorphic hepatocellular carcinoma (SNU-387), hepatic ductal carcinoma (LMH/2A), morris hepatoma (McA-RH7777), and novikoff hepatoma (N1-S1 Fudr) cancer cell lines. The antioxidant activity of Fe3O4@Pectin NPs was determined by DPPH method. The Fe3O4@Pectin NPs showed the high antioxidant activity according to the IC50 value. Abstract

Immunological adjuvants are essential for successful cancer vaccination. However, traditional adjuvants have some limitations, such as lack of controllability and induction of systemic toxicity, which restrict their broad application. Here, we present a light-activable immunological adjuvant (LIA), which is composed of a hypoxia-responsive amphiphilic dendrimer nanoparticle loaded with chlorin e6. Under irradiation with near-infrared light, the LIA not only induces tumour cell lysis and tumour antigen release, but also promotes the structural transformation of 2-nitroimidazole containing dendrimer to 2-aminoimidazole containing dendrimer which can activate dendritic cells via the Toll-like receptor 7-mediated signaling pathway. The LIA efficiently inhibits both primary and abscopal tumour growth and induces strong antigen-specific immune memory effect to prevent tumour metastasis and recurrence in vivo. Furthermore, LIA localizes the immunological adjuvant effect at the tumour site. We demonstrate this light-activable immunological adjuvant offers a safe and potent platform for in situ cancer vaccination., Immunological adjuvants are a crucial component of cancer vaccines. Here the authors design a light-activable immunological adjuvant, based on hypoxia-responsive amphiphilic dendrimer nanoparticles loaded with a photodynamic agent, promoting anti-tumor immune responses in preclinical cancer models.

Abstract

Quantitative and direct evidence for tetragonal to monoclinic martensitic transformation toughening was revealed by electron backscatter diffraction (EBSD) and in-situ nanoindentation, using plasma-sprayed 3 mol% Y 2 O 3 –ZrO 2 coatings. On the basis of EBSD phase distribution, four zooms with different phase compositions and microstructure were selected. The tetragonal grains, which were surrounded by large pores, completely transformed into a monoclinic phase, but were then crushed when subjected to a loading of 10 mN. Moreover, the critical excitation stress σ c for the martensitic transformation was estimated to be about 4.2 GPa. According to the displacement curves, the ratio of reduced modulus to hardness ( E r /H ) , which directly indicates the toughness of a material, was quantitively calculated. This experimentally demonstrated that both the elastic and plastic deformation capacity of the partially transformed grain were significantly improved, compared with the untransformed tetragonal grains. These findings will provide a fundamental insight into martensitic transformation toughening.

29. Correlation between the microstructure and low thermal conductivity of plasma-sprayed Yb2O3–Y2O3co-doped strontium zirconate coatings

Abstract

Yb2O3–Y2O3co-doped strontium zirconate coatings (Sr(Zr0.9Yb0.05Y0.05)O2.95) were investigated as promising materials for thermal barrier coating (TBC) applications. Two types of Sr(Zr0.9Yb0.05Y0.05)O2.95coatings prepared under suitable atmospheric plasma spraying parameters have low thermal conductivity, approximately 0.60 W m−1 K−1at 1100 °C. The low thermal conductivities of these two coatings can be attributed to their microstructures, such as small grain size, solid solution, and special grain boundary structures. In both Sr(Zr0.9Yb0.05Y0.05)O2.95coatings, Yb3+and Y3+ions aggregated at the grain boundaries, hinder grain growth and reduce grain size. The rare-earth element distribution and transmission electron microscopy analysis of the coatings indicated that the dopant atoms replaced some of the Zr4+ions at the grains and grain boundaries, successfully introducing oxygen vacancies and strain fields. These are two important reasons for the low thermal conductivity of Sr(Zr0.9Yb0.05Y0.05)O2.95coatings. In addition, the lower thermal conductivity of the Sr(Zr0.9Yb0.05Y0.05)O2.95-B coating is attributed to the presence of wider grain boundary structures composed of tightly packed nanocrystals.

30. Effect of Dy2O3doping on phase compositions, microstructures, thermophysical properties and fracture toughness of YSZ coatings

Abstract

Yttria-stabilized zirconia (YSZ) coatings with different Dy2O3doping contents were prepared via plasma spraying. The effects of phase compositions, microstructures and lattice defects on the thermal conductivity, fracture toughness and thermal shock resistance were analyzed. The result indicates that the Dy2O3doping significantly affects thermophysical properties of YSZ coatings. The Dy2O3doping can improve the insulation performance of the YSZ coating and the minimum thermal conductivity of the coating has reached 0.89 W/K·m. A decrease in thermal conductivity with increasing Dy2O3doping content can be attributed to the lattice distortion, ions substitution, oxygen vacancies and reducing grain size. Meanwhile, the Dy2O3doping causes the fracture toughness of YSZ coatings to decrease. Consequently, the Dy2O3-doped YSZ coatings exhibit weaker thermal shock resistance because the Dy2O3doping promotes the formation of the cubic YSZ phase. The tetragonal phase in plasma-sprayed coatings is beneficial for service lifetime owing to its phase transformation and ferroelasticity-induced toughening under stress. Although the Dy2O3-doped YSZ coatings undergo more severe sintering owing to a decrease in the activation energy of grain growth caused by lattice distortion, they exhibit better phase stability than the YSZ coating under high temperatures because the Dy2O3doping inhibits the diffusion of Y. Finally, a Dy2O3-doped ZrO2coating composed mainly of the tetragonal ZrO2phase was designed, which exhibited a better thermal shock resistance.

Abstract

A simple hydrofluoric acid-free one-pot method was developed for the preparation of three functionalized magnetic MIL-101 composites (Fe3O4–NH2@MIL-101, Fe3O4–COOH@MIL-101 and Fe3O4/C@MIL-101). As magnetic solid-phase extraction materials, they exhibited a remarkable preconcentration ability for estrone (E1), 17β-estradiol (E2), estriol (E3) and bisphenol-A (BPA). However, because the incorporated magnetic particles were decorated with different surface functional groups, the three magnetic MIL-101 composites exhibited certain differences in extraction efficiency of the four target estrogens. Herein, the Fe3O4–NH2@MIL-101 composite as an adsorbent was applied for magnetic solid-extraction coupled with high performance liquid chromatography (HPLC) to determine the estrogens content in aqueous samples. The dominant parameters affecting enrichment efficiency including extraction time, sample pH, extraction temperature and desorption conditions were investigated. Under the optimized conditions, the developed method showed good linearity within the range of 0.2–100 μg L−1, low detection limits (0.06–0.22 μg L−1) and high repeatability (RSD, 5.9–8.3%). The recoveries were between 80.6 and 99.6% with the spiked level of 10 μg L−1. The proposed method was successfully applied to determine the estrogens content in real water samples.

39. Dopamine Functionalized S,N Co-doped Carbon Dots as a Fluorescent Sensor for the Selective Detection of Fe3+and Fe2+in Water

Abstract

In current work, novel functionalized carbon dots have been designed and synthesized by covalently linking dopamine to the surface of S,N co-doped carbon dots (DA-S,N-CDs) for the selective detection of Fe3+and Fe2+in water. The as-synthesized DA-S,N-CDs emit blue fluorescence peaked at 470 nm and exhibit excitation-dependent tunable emissions. The tolerance towards pH, salt, and UV irradiation of synthesized carbon dots reveals excellent stability. Upon exposure to Fe3+or Fe2+, the fluorescence of DA-S,N-CDs was selectively quenched, while other competitive cations did not change significantly. Under the optimal experimental conditions, the fluorescence intensity of DA-S,N-CDs showed a good linear relationship with the concentrations of Fe3+and Fe2+(5–200 μM for Fe3+and 5–300 μM for Fe2+), and the limit of detection was 2.86 and 2.06 μM, respectively. Furthermore, considering the excellent stability and anti-interference, DA-S,N-CDs have been successfully used for the detection of Fe3+and Fe2+in environmental water.

Abstract

As an intensely studied computed tomography (CT) contrast agent, gold nanoparticle has been suggested to be combined with fluorescence imaging modality to offset the low sensitivity of CT. However, the strong quenching of gold nanoparticle on fluorescent dyes requires complicated design and shielding to overcome. Herein, we report a unique nanoprobe (M-NPAPF-Au) co-loading an aggregation-induced emission (AIE) red dye and gold nanoparticles into DSPE-PEG(2000) micelles for dual-modal fluorescence/CT imaging. The nanoprobe was prepared based on a facile method of "one-pot ultrasonic emulsification". Surprisingly, in the micelles system, fluorescence dye (NPAPF) efficiently overcame the strong fluorescence quenching of shielding-free gold nanoparticles and retained the crucial AIE feature. In vivo studies demonstrated the nanoprobe had superior tumor-targeting ability, excellent fluorescence and CT imaging effects. The totality of present studies clearly indicates the significant potential application of M-NPAPF-Au as a dual-modal non-invasive fluorescence/X-ray CT nanoprobe for in vivo tumor-targeted imaging and diagnosis.

Abstract

The purpose of this study was to explore a composite thermosensitive in situ gelling formulation using the distribution of solid lipid nanoparticles (SLNs) among poloxamer-based hydrogels as a potential carrier for novel ocular drug delivery. SLNs containing the model drug Resina Draconis were prepared using a melt-emulsion ultrasonication method. A central composite design (CCD) was adopted to screen the thermosensitive hydrogel (THG) formulation. After aqueous SLNs were dispersed into the THG matrices, the physicochemical properties of the SLNs were characterized before and after their incorporation into hydrogels. The in vitro corneal penetration experiment, ocular irritant test and transcorneal mechanism across the cornea have been previously described to predict the feasibility for the proposed ophthalmic application. Finally, the optimal THGs consisted of 27.8% (w/v) poloxamer 407 and 3.55% (w/v) poloxamer 188. The particle size of the SLNs remained within the colloidal range. In vitro corneal penetration studies revealed a nearly steady sustained drug release. The hen's egg test-chorioallantoic membrane (HET-CAM) test indicated that all of the tested polymer systems were non-irritant. Coumarin-6 labeled SLNs formulated into THGs displayed a more homogeneous fluorescence with a deeper penetration intensity into the cornea at various times. Taken together, these results suggest that the SLN-based THG system can be used as a potential vehicle for ocular application.

Abstract

Rare‐earth hafnates are gaining attention due to their excellent high‐temperature phase stability and low thermal conductivity. However, they still have shortcomings of low thermal expansion and poor calcium‐magnesium‐aluminum‐silicate (CMAS) corrosion resistance. In this study, we employed high‐entropy engineering and component design to synthesize three high‐entropy hafnates (La0.2Ce0.2Nd0.2Gd0.2T0.2)2Hf2O7 (T = Dy, Ho, Tm) as well as a single‐component hafnate Nd2Hf2O7, with the aim of preparing thermal barrier coatings with an excellent comprehensive performance. Test results indicate that the high‐entropy compositions have excellent thermal properties. The focus is on elucidating the corrosion process and failure mechanism of CMAS at 1300°C. Moreover, the analysis of residual CMAS and corrosion products was conducted to evaluate the discrepancies in CMAS corrosion behavior among the various compositions. [ABSTRACT FROM AUTHOR]

Abstract

• Mo/HZSM-22 bi-functional catalysts were prepared by utilizing MoO 3 sublimation. • The sublimation of the MoO 3 could promote the dispersion of Mo species. • High reduction temperature enhances more Mo4+ and Mo0 formation. • Mo/HZSM-22 catalysts achieved complete deoxygenation of palmitic acid. • High selectivity of C 16 alkanes and iso -alkanes were achieved. The optimal preparation conditions of Mo/H-ZSM-22 bi-functional catalysts were obtained via the sublimation phenomenon of MoO 3 at high calcination temperatures, which was beneficial for the well-dispersion of MoO x species with 5−10 nm particles. High reduction temperature enhanced the reduction from Mo6+ to Mo4+ and even from Mo4+ to Mo°, which would be beneficial for iso -alkanes formation with higher carbon reservation. Importantly, 89.3 % selectivity of C 16 alkanes of which 61.7 % were iso -C 16 alkanes were obtained with complete deoxygenation of palmitic acid, which was the highest selectivity of C 16 alkanes over Mo/H-ZSM-22 bi-functional catalyst prepared at a calcination temperature of 550 °C and at a reduction temperature of 600 °C. 49. Effects of compatibility of Clostridium butyricum and Bacillus subtilis on growth performance, lipid metabolism, antioxidant status and cecal microflora of broilers during the starter phase.

Abstract

Objective: This study aimed to determine the effects of compatibility of Clostridium butyricum and Bacillus subtilis on growth performance, lipid metabolism, antioxidant status and cecal microflora of broilers during the starter phase., Methods: A total of 600 1-day-old Ross 308 broilers were randomly divided into two groups with six replicates in each group. Chickens in the control group were fed a basal diet, while chickens in the experimental group were fed a diet supplemented with 2 × 108 CFU/kg of C. butyricum and 1 × 109 CFU/kg of B. subtilis. The experimental period was 21 days., Results: Addition of C. butyricum and B. subtilis significantly increased (p<0.05) the body weight and liver NADP-malic enzyme (NADP-ME) activity of broilers, enhanced (p<0.05) the average daily gain and average daily feed intake of broilers. However, the addition of C. butyricum and B. subtilis did not significantly affect the concentrations of triglyceride and total cholesterol in the serum, the activities of fatty acid synthase and acetyl-CoA carboxylase in the liver, the total antioxidant capacity, glutathione peroxidase activity and malondialdehyde content in the serum and liver. Besides, microbial analysis revealed that supplementation of C. butyricum and B. subtilis increased (p<0.05) the abundance of Firmicutes such as CHKCI001 and Faecalibacterium, decreased (p<0.05) the abundance of Bacteroidota such as Bacteroides and Alistipes. Spearman correlation analysis confirmed that the above cecal microbiota were closely related to the growth performance of broilers (p<0.05). In addition, simultaneous supplementation of C. butyricum and B. subtilis significant affected (p<0.05) 33 different functional pathways such as lipid metabolism and carbohydrate metabolism. This explains the phenomenon of increased growth performance and liver NADP-ME activity in the probiotics group., Conclusion: The compatibility of C. butyricum and B. subtilis could improve the growth of broilers during the starter phase by changing the cecal microflora.

Abstract

Introduction: Ox-LDL (oxidized low-density lipoprotein)-induced endothelial cell injury and dysfunction of vascular smooth muscle cells play critical roles in the development of atherosclerosis (AS). Tropomyosin 2 (TPM2) has been implicated in cardiac diseases, but its critical role and regulatory mechanism in AS progression have not yet been elucidated., Material and Methods: The expression of TPM2 was investigated in AS tissues. Ox-LDL was used to construct an AS in vitro model based on endothelial and vascular smooth muscle cells (HAECs and VSMCs). An overexpression assay was performed to evaluate the role of TPM2 in AS. Meanwhile, the involvement of the RhoA pathway in TPM2-mediated AS progression was evaluated using narciclasine., Results: Tropomyosin 2 was dramatically upregulated in both AS tissues and ox-LDL-induced HAECs. Overexpression of TPM2 attenuated ox-LDL-stimulated cell growth depression, inflammatory and adhesive responses in HAECs, as well as oxidative stress and mitochondrial dysfunction. Additionally, VSMCs, impacted by TPM2-overexpressed HAECs, showed alleviated cellular processes which were abnormally activated by ox-LDL. Furthermore, depressed activation of the RhoA pathway was found in TPM2-overexpressed HAECs and activating the signaling rescued these effects of TPM2 exerted on ox-LDL-stimulated HAECs and VSMCs., Conclusions: TPM2 had an advantageous impact on ox-LDL-induced AS progression in vitro by mediating the RhoA pathway.
