Gastric cancer (GC) is the 5th most common cancer and the 3rd leading cause of cancer-related death worldwide. It is of great significance to study the underlying molecular mechanism of GC, and targeting glycolysis is a good strategy to treat GC. SET domain containing 5 (SETD5) contains a catalytic methyltransferase SET domain, which is known as a lysine methyltransferase that affects the progression of multiple cancers. However, its possible role in GC was still unclear. Here, we revealed that SETD5 was highly expressed in GC and was associated with a poor prognosis. Further through the in vitro experiments, we revealed that the downregulation of SETD5 inhibited the proliferation and migration of GC cells. Knockdown of SETD5 inhibited glucose consumption and glycolysis. Further studies have shown that SETD5 knockdown restrained the Akt signaling pathway. Therefore, we thought that SETD5 could act as a GC target.

The occurrence of Maillard reaction in heated Torreya grandis cv. Merrilli seed oil was verified by detecting browning index and the contents of the Maillard reaction products (MRPs) 3-deoxyglucosone, methylglyoxal and 5-(hydroxymethyl)furfural, and the effects of the MRPs on the quantification of total phenolics using the Folin-Ciocalteu and Fast Blue BB assays were explored. The Folin-Ciocalteu assay showed that the content of total phenolics in the oil increased after treatment at 150 ℃ for 90 to 120 min, while the Fast Blue BB assay showed the opposite result. It was also observed that the absorbance of the oil at a wavelength of 294 nm increased after long-term low-temperature or high-temperature heat treatment. 3-Deoxyglucosone (0.21–0.47 μg/g) and 5-(hydroxymethyl)furfural (0.06–0.40 μg/g) were detected only in the 150 ℃ treated oil, while methylglyoxal (0.67–1.73 μg/g) existed in both oil samples. In the Folin-Ciocalteu assay, the absorbance at a wavelength of 765 nm of 3-deoxyglucosone and methylglyoxal linearly increased with an increase in their concentrations, and the decreasing order of the absorbance of the MRPs at the same concentration was 3-deoxyglucosone > methylglyoxal > 5-(hydroxymethyl)furfural. In the presence of 3-deoxyglucosone, the Folin-Ciocalteu result was greater than the actual value, and the degree of interference was not related to the content of total phenolics in samples, but instead was positively correlated with the concentration of 3-deoxyglucosone. In the Fast Blue BB assay, there was no significant difference in absorbance at 420 nm among the three MRPs. For food matrices prone to the formation of 3-deoxyglucosone and methylglyoxal during processing, the Fast Blue BB assay can be selected instead of the Folin-Ciocalteu assay to mitigate the interferences from the two substances in the quantification of total phenolics.

To explore the anti-inflammatory mechanism of Camellia oleifera seed oil, the metabolites in ethanol extracts from 15 batches of C. oleifera seeds were analyzed by ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) and the inhibitory activity of the ethanol extracts on enteritis in Drosophila was evaluated by the “Smurf” test. The correlation between small molecular metabolites in the ethanol extracts and their anti-inflammatory activity was investigated and molecular docking with lipoxidase (LOX) was conducted to identify the key metabolites. Finally, the key metabolites were verified by the “Smurf” test. The results showed that the ethanol extract of C. oleifera seeds had good enteritis inhibitory activity, with percentage inhibition of 15.06%–61.71%, and its metabolites mainly included polyphenols, amino acids, organic acids, nucleotides, sugars and lipids. Seven polyphenolic compounds were identified in C. oleifera seeds for the first time. The monomeric compounds with good anti-enteritis activity were preliminarily identified as bruceine B, miltirone, 8-geranyloxypsoralen, cedrelone, wighteone, kaempferitrin and kaempferol-3-O-rutinoside. The anti-inflammatory activity of wighteone was reported for the first time in this study, and at 0.1 mg/mL, the percentage inhibition of enteritis was 64%, much greater than the positive control sulfasalazine (27.99%). This study lays the foundation for further exploring the anti-enteritis activity and mechanism of C. oleifera seed oil in vitro and in vivo.

The Precipitation nowcasting can provide high-resolution forecasts of rainfall and hydrometeors in 2 hours and play an important role in risk management for flash flood and debris flow events, but it is a very challenge work. This study proposed a new method which combine of XGBoost method and the PPLK model for precipitation nowcasting (XGB-PPLK). The proposed method was assessed in four different types of storms, and the experimental results show that the XGB-PPLK can improve the probability of detection (POD), as while as reducing the normalized mean square error (NMSE), and maintain the basically equivalent false-alarm ratio (FAR).

Screening and identifying the active compounds in foods are important for the development and utilization of functional foods. In this study, the anti-enteritis activity of ethanol extract from Camellia oleifera oil (PECS) was quickly evaluated using a Smurf Drosophila model and the metabolomics approach, combined with molecular docking techniques, were performed to rapidly screen and identify compounds with potential anti-enteritis activity in PECS. PECS showed good anti-enteritis activity and inhibited the activity of 5-lipoxygenase (LOX), cyclooxygenase 2 (COX-2) and inducible nitric oxide synthase (iNOS). In particular, wighteone and p-octopamine were newly identified in C. oleifera oil and were proven to have good anti-enteritis activity. The inhibitory activity of kaempferitrin (IC50 = 0.365 mmol L−1) was higher than that of wighteone (IC50 = 0.424 mmol L−1) and p-octopamine (IC50 = 0.402 mmol L−1). Of note, the IC50 value of salazosulfapyridine was 0.810 mmol L−1. Inhibition of LOX activity is likely one of the anti-enteritis mechanisms of PECS. These new findings lay the foundation for further investigations into the underlying mechanisms of anti-enteritis activity in C. oleifera oil. [ABSTRACT FROM AUTHOR]

Abstract: Apricot kernels were roasted at various temperatures (120–180 °C) for 10 min and changes in the fatty‐acid profiles, oxidative stability, and antioxidant activity, as well as the total phenolic contents (TPC) of the oils and skin (seed coat), were monitored. Roasting has no obvious influence on profiles and contents of fatty acid, induction period (IP), browning index, TPC, and antioxidant activity (2,2‐diphenyl‐1‐picrylhydrazyl (DPPH), 2,2‐azinobis(3‐ethylbenzothiazoline‐6‐sulfonic acid), (ABTS) and Oxygen Radical Absorbance Capacity (ORAC) of oils obtained from apricot naked‐kernel, but increases IP, TPC, and oxidative stability in oils obtained from apricot kernel with skin. All results in the present work demonstrated that thermal treatment accelerated the production and transference of alcohol‐soluble phenolics into the oil, and improved the oil oxidative stability. It is not Maillard reaction products but alcohol‐soluble phenolic compounds in skins that play a role in improving the oxidative stability and antioxidant activity of oils, and inhibition for primary peroxide production was more effective than secondary peroxide production at a low roasting temperature and a short roasting time. The present findings can advance knowledge on the conditions used for utilization of coproducts (skin) of apricot kernel and facilitate large‐scale production of stable oil against oxidation. Distal tibiofibular syndesmosis injury (DTS) occurs frequently with ankle sprains. Current treatments pose several limitations including causing soft tissue irritation, bringing damage to fixation secondary to weight-bearing, and requiring follow-up surgeries. Here, we investigated the clinical effects of a new technique, titanium cable isotonic annular fixation, for the treatment of DTS injury. From January 2015 to June 2017, 36 patients with ankle fractures and DTS injuries had their fractures repaired with the titanium cable isotonic annular fixation system. Recovery was scored by the AOFAS ankle function score system. We also assessed the differences in ankle motion between healthy and operative joints, and recorded the complications. All patients recovered from surgery without any serious complications. We followed all the cases for 18-25 months with an average follow-up of 21.26±3.23 months. 12 months after the operation, X-ray images showed that the titanium cables were fixed in the correct position without any fracture or loosening. Additionally, no degeneration or traumatic arthritis was observed in the ankle joint. There were no incision or bone mineral density changes between the titanium fix and tibiofibular bones. Nearly all patients recovered well except for three who developed inflammation and infection. However, these three patients recovered following 1 week of intravenous antibiotics and local radiofrequency physiotherapy. According to the AOFAS scoring system, all patients achieved satisfactory recovery 12 months post operation. Our titanium cable isotonic annular fixation system has both the advantages of elastic and rigid fixations. It can restore isotonic strength of the distal tibiofibular joint, and its biomechanical performance approaches normal physiological function. After the operation, patients tolerated weight-bearing exercise and recovered joint mobility. Finally, there is no need to remove the distal tibiofibular implant after 12 weeks.
