Abstract

This article discusses the potential for increasing the flavonoid content in tomato fruits through the disruption of a gene called SlSPL-CNR. Flavonoids are important compounds that contribute to the color and health benefits of fruits. The researchers found that the SlSPL-CNR gene negatively regulates flavonoid biosynthesis by repressing the transcriptional activity of another gene called SlMYB12. By using the CRISPR/Cas9 gene editing system to disrupt the SlSPL-CNR gene, the researchers were able to increase the flavonoid content in tomato fruits. Abstract

Hemorrhagic transformation (HT) is one of the most serious complications after endovascular thrombectomy (EVT) in acute ischemic stroke (AIS) patients. The purpose of this study is to develop and validate deep‐learning (DL) models based on multiparametric magnetic resonance imaging (MRI) to automatically predict HT in AIS patients. Multiparametric MRI and clinical data of AIS patients with EVT from two centers (data set 1 for training and testing: n = 338; data set 2 for validating: n = 54) were used in the DL models. The acute infarction area of diffusion‐weighted imaging (DWI) and hypoperfusion of perfusion‐weighted imaging (PWI) was labeled manually. Two forms of data sets (volume of interest [VOI] data sets and slice data sets) were analyzed, respectively. The models based on single parameter and multiparameter models were developed and validated to predict HT in AIS patients after EVT. Performance was evaluated by area under the receiver‐operating characteristic curve (AUC), accuracy (ACC), sensitivity, specificity, negative predictive value, and positive predictive value. The results showed that the performance of single parameter model based on MTT (VOI data set: AUC = 0.933, ACC = 0.843; slice data set: AUC = 0.945, ACC = 0.833) and TTP (VOI data set: AUC = 0.916, ACC = 0.873; slice data set: AUC = 0.889, ACC = 0.818) were better than the other single parameter model. The multiparameter model based on DWI & MTT & TTP & Clinical (DMTC) had the best performance for predicting HT (VOI data set: AUC = 0.948, ACC = 0.892; slice data set: AUC = 0.932, ACC = 0.873). The DMTC model in the external validation set achieved similar performance with the testing set (VOI data set: AUC = 0.939, ACC = 0.884; slice data set: AUC = 0.927, ACC = 0.871) (p > 0.05). The proposed clinical, DWI, and PWI multiparameter DL model has great potential for assisting the periprocedural management in the early prediction HT of the AIS patients with EVT. 30. Effect of humanized care in the treatment of neonatal jaundice and its effect on oxygen saturation

Abstract

Objective: This study aimed to investigate the effect of humanized care in the treatment of neonatal jaundice and its effect on oxygen saturation. Methods: A total of 202 infants with neonatal jaundice admitted to our hospital from January 2018 to June 2020 were divided into group A (n=102) and group B (n=100) according to their parents’ choice. Group A received humanized care and group B received routine nursing. The clinical efficacy, serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), total bilirubin (TBIL) levels and arterial blood oxyhemoglobin saturation (SaO(2)), cerebral oxygen saturation (rSO(2)), mean arterial pressure (MAP) levels were determined between the two groups. Results: Compared with group B, group A had significantly shorter time of fetal stool turning yellow, time of jaundice regression and duration of blue light irradiation (P < 0.05), lower serum AST, ALT and TBIL levels (P < 0.05), higher levels of SaO(2), rSO(2) and MAP (P < 0.05), higher average sleep time per day and mean daily milk consumption (P < 0.05). The incidence of adverse events in group A was significantly lower than that in group B (P < 0.05). Parental satisfaction with care in group A was significantly higher than that in group B (P < 0.05). Conclusion: Humanized care can significantly improve the prognosis and recovery speed and is conducive for SaO(2) to return to normal level, and can reduce the adverse reactions with high parental satisfaction.

Abstract

Summary: Dynamic chemical modifications in eukaryotic messenger RNAs (mRNAs) constitute an essential layer of gene regulation, among which N6‐methyladenosine (m6A) was unveiled to be the most abundant. m6A functionally modulates important biological processes in various mammals and plants through the regulation of mRNA metabolism, mainly mRNA degradation and translation efficiency. Physiological functions of m6A methylation are diversified and affected by intricate sequence contexts and m6A machineries. A number of studies have dissected the functional roles and the underlying mechanisms of m6A modifications in regulating plant development and stress responses. Recently, it was demonstrated that the human FTO‐mediated plant m6A removal caused dramatic yield increases in rice and potato, indicating that modulation of m6A methylation could be an efficient strategy for crop improvement. In this review, we summarize the current progress concerning the m6A‐mediated regulation of crop development and stress responses, and provide an outlook on the potential application of m6A epitranscriptome in the future improvement of crops. Abstract

Jasmonates accumulate rapidly and act as key regulators in response to mechanical wounding, but few studies have linked receptor-like cytoplasmic kinases (RLCKs) to wound-induced jasmonic acid (JA) signaling cascades. Here, we identified a novel wounding-induced RLCK-XII-2 subfamily member (SlZRK1) in tomato (Solanum lycopersicum) that was closely related to Arabidopsis HOPZ-ETI-DEFICIENT 1 (ZED1)-related kinases 1 based on phylogenetic analysis. SlZRK1 was targeted to the plasma membrane of tobacco mesophyll protoplasts as determined by transient co-expression with the plasma membrane marker mCherry–H+-ATPase. Catalytic residue sequence analysis and an in vitro kinase assay indicated that SlZRK1 may act as a pseudokinase. To further analyse the function of SlZRK1, we developed two stable knock-out mutants by CRISPR/Cas9. Loss of SlZRK1 significantly altered the expression of genes involved in JA biosynthesis, salicylic acid biosynthesis, and ethylene response. Furthermore, after mechanical wounding treatment, slzrk1 mutants increased transcription of early wound-inducible genes involved in JA biosynthesis and signaling. In addition, JA accumulation after wounding and plant resistance to herbivorous insects also were enhanced. Our findings expand plant regulatory networks in the wound-induced JA production by adding RLCKs as a new component in the wound signal transduction pathway. [ABSTRACT FROM AUTHOR]

However, the underlying mechanism is still unknown. A549 resistance to DDP (A549/DDP) was established. Microarray was used to analyze microRNA (miRNA) expression profiles of A549 cells, A549/DDP cells, A549 exosomes, and A549/DDP exosomes. There was a strong correlation of miRNA profiles between exosomes and their maternal cells. A total of 11 miRNAs were significantly upregulated both in A549/DDP cells compared with A549 cells and in exosomes derived from A549/DDP cells in contrast to exosomes from A549 cells. A total of 31 downregulated miRNAs were also observed. miR-100–5p was the most prominent decreased miRNA in DDP-resistant exosomes compared with the corresponding sensitive ones. Downregulated miR-100–5p was proved to be involved in DDP resistance in A549 cells, and mammalian target of rapamycin (mTOR) expression was reverse regulated by miR-100–5p. We examined whether epithelial-to-mesenchymal transition affects gefitinib resistance in non-small cell lung cancer (NSCLC) cells. Cell viability was detected by CCK-8 assay, VIM expression levels were determined by quantitative real-time polymerase chain reaction. Western blot and immunocytochemistry were performed to determine the protein expression level of vimentin. We observed morphologic differences between gefitinib-sensitive and -insensitive cells. Compared with the sensitive parental cell line, HCC827, vimentin expression levels were increased in HCC827 cells with acquired gefitinib resistance. Vimentin expression was also markedly upregulated in cells with intrinsic gefitinib resistance, and upregulated vimentin expression was correlated with gefitinib sensitivity. Our previous study demonstrated that coadministration of gefitinib and GW3965 resulted in decreased cell proliferation and induced apoptosis. Therefore, we investigated the relationship among GW3965, vimentin, and gefitinib resistance in NSCLC cells by analysis of the expression of vimentin in cells treated with a combination of gefitinib and GW3965. Gefitinib treatment led to increased levels of intracellular vimentin, while combined treatment with gefitinib and GW3965 resulted in decreased vimentin expression levels through reduction of gefitinib drug resistance in NSCLC cells.
