Abstract Background Diabetic wounds are one of the long-term complications of diabetes, with a disordered microenvironment, diabetic wounds can easily develop into chronic non-healing wounds, which can impose a significant burden on healthcare. In diabetic condition, senescent cells accumulate in the wound area and suppress the wound healing process. AMPK, as a molecule related to metabolism, has a close relationship with aging and diabetes. The purpose of this study was to investigate the effects of AMPK activation on wound healing and explore the underlying mechanisms. Methods AMPK activator A769662 was topically applied in wound models of diabetic mice. Alterations in the wound site were observed and analyzed by immunohistochemistry. The markers related to autophagy and ferritinophagy were analyzed by western blotting and immunofluorescence staining. The role of AMPK activation and ferritinophagy were also analyzed by western blotting. Results Our results show that AMPK activation improved diabetic wound healing and reduced the accumulation of senescent cells. Intriguingly, we found that AMPK activation-induced ferroptosis is autophagy-dependent. We detected that the level of ferritin had deceased and NCOA4 was markedly increased after AMPK activation treatment. We further investigated that NCOA4-mediated ferritinophagy was involved in ferroptosis triggered by AMPK activation. Most importantly, AMPK activation can reverse the ferroptosis-insensitive of senescent fibroblast cells in diabetic mice wound area and promote wound healing. Conclusions These results suggest that activating AMPK can promote diabetic wound healing by reversing the ferroptosis-insensitive of senescent fibroblast cells. AMPK may serve as a regulatory factor in senescent cells in the diabetic wound area, therefore AMPK activation can become a promising therapeutic method for diabetic non-healing wounds.

Squall lines often produce extremely disastrous gales and play a crucial role in the safety of hytropower stations.This article explores a squall line event over the lower reaches of Jinsha River on June 4， 2016， which brought 13-level wind at the canyon of Baihetan Hytropower Station.This research detect the formation mechanism of this squall line from environmental conditions and medium-scale meteorology.The following conclusions are obtained： （1） Before the squall line arrived， the ground pressure dropped steadily by 9 hPa because of the low pressure， and the temperature of the canyon rises up above 14 ℃.As the squall line arrived， the pressure rises sharply with thunderstorm high pressure， and the temperature dropped more than 10 ℃.At the same time， the relative humidity reached as higher as 70%， accompanied by short-term precipitation.After the squall line left， the aforementioned meteorological elements restored to their original values.（2） Analysis on the synoptic scale circulation， we finds that Baihetan hytropower station located in the south of southwest vortex， which induced a wind convergence background at the low level.Meanwhile， the station lied in right side of the jet stream inlet at upper level， and strong vertical motion formed by the wind shear.As a result， the squall line to come into being.Furthermore， in the troposphere before the squall line， the atmospheric circulation displayed a dry-cold advection in the middle-upper level superimposed over a warm-wet advection and higher ground temperature in the lower levels， increasing the temperature lapse rate.t As a result， the strong static instability and convection accumulated， and the conditions were favorable for the squall line to live a long time.The analyses from vertical wind speed showed that the subsidence movement made the momentum spread down， and promoted the uplift in the front side of line， and a positive feedback effect developed， promoting a stable mechanism for the strong storm.（3） Radar of Zhaotong detection indicates that echo cell that greater than 30 dBZ aggregated costly into a band and embed with the strong echo cores of greater than 50 dBZ， as a cylindrical convective cloud of higher than 10 km.On the radical velocity field， the meso-scale convergence line coinciding with the banded strong echo， as well as MARC （Mid-Altitude Radial Convergence） characteristics of the mid-altitude radial convergence， were the trigger mechanism for the squall line.As the cloud structure of the storm， strong upward movement in front of the squall line leaded to the dome.As the squall line arrived， the echo core dropped rapidly， and the extreme gale of downburst formed， resulted the new convective monomers come in being along the canyon.

Split-type flying car will play an important role in the future transportation. This paper adopts a guidance method that couples visual information and depth information, and improves the docking accuracy through the mutual cooperation of the drone and the vehicle. Firstly, a multilevel docking marker is designed to achieve adaptive target matching within different distances during the docking process. The marker has strong robustness and can adapt to complex scenes such as occlusion, strong light, and large angle tilting, providing the redundant corner points required for machine vision detection pose information accurately. Secondly, a three-dimensional pose estimation algorithm is proposed, which can introduce depth information to correct the homography matrix. The algorithm combines the advantages of strong robustness to multilevel marker detection and high accuracy of depth information, and can output millimeter-level precision pose information in different environments, different inclination angles, and different occlusions. Finally, a flying car model experiment was carried out, and the results showed that the guidance technology can obtain millimeter-level precise pose information during the entire process of long distance-near distance-completion of docking, thus realizing precise docking.

Adequate substance exchange is an important prerequisite for tissue repair, and how to efficiently establish mature vascular networks to enhance the graft-host substance exchange is a challenge. Herein, inspired by the special structure of the aquatic plant “Ludwigia sedioides”, ultra-thin microporous scaffolds (TMS) of bovine Achilles tendon derived collagen were fabricated as “floating leaves” via pyridine reaction, and cleverly combined with platelet-rich plasma (PRP) after the Schiff base and Michael additions reactions of polydopamine to construct a biomimetic “Ludwigia sedioides” spacer (PTMS@P), allowing blood vessels (as a “rhizome”) freely shuttling and substance exchange. In this spacer, growth factors were efficiently released to peak concentrations in the first 12 h, thereby facilitating rapid blood vessel growth. In the subsequent 96 h, these slow-released growth factors retained at 11.6 % to 45.8 % of the peak concentrations, sustainedly promoting vascular networks shuttling through the spacer, graft-host substance exchange and extracellular matrix synthesis in fibroblasts. In vitro, the spacer significantly fostered fibroblasts and endothelial cells’ proliferation, adhesion and migration, which were fundamental for revascularization and substance exchange. Immunohistochemical staining of CD31 and Ki67 in vivo demonstrated that abundant vascular networks shuttled through PTMS@P and extended from flap base to the farther flap body, promoting more robust substance exchange and cellular metabolism, which significantly enhanced the survival of ischemic flaps in nude mice. Altogether, this “Ludwigia sedioides” inspired spacer PTMS@P with strengthened revascularization and substance-exchange functionality holds promising applications for tissue repair and regeneration.

By anchoring the lower rollers and dynamically adjusting the upper roller’s downfeed of a pyramid roll bender, one can achieve the precise bending of a workpiece into a desired planar form characterized by variable curvature. To ensure the seamless processing of individual cross sections without impinging upon adjacent areas, critical roller spacings are identified through theoretical mechanics analyses. The reaction force exerted on the top roller is calculated by integrating the desired curvature into the elastoplastic constitutive equation and subsequently deriving a dynamic adjustment of the downfeed from quasi-static finite element simulations. This preliminary downfeed protocol undergoes refinement to mitigate discrepancies between the targeted and the actual curvatures. Numerical instances demonstrate that the application of roller configurations, as outlined herein, yields a product profile that closely mirrors the intended curve. This congruence can be further improved with an additional iteration; however, subsequent iterations are seen to yield negligible improvements, indicating a rapid convergence of this algorithm.

To solve the wave propagation problems of the Euler–Bernoulli beam in an unbounded domain effectively and efficiently, a new local artificial boundary condition technology is proposed. It replaces the residual right-hand side of the truncated discrete equation with an equivalent linear algebraic system. First, the equivalent Schrodinger equation is discussed. Its artificial boundary condition is obtained by first rationalizing the Dirichlet-to-Neumann condition in the frequency domain with a Pade approximation and then inverse transforming each Pade term back into the time domain by introducing auxiliary degrees of freedom. Frequency shifting is employed such that it performs better near a prescribed frequency. Then, the artificial boundary condition of the finite element Euler–Bernoulli beam is obtained by simple algebraic manipulations on that of the corresponding Schrodinger equation. This method only makes local changes to the original truncated discrete dynamic system and thus is very efficient and easy to use. The accuracy of the proposed method can be improved by using more Pade terms and a proper shift frequency. The numerical example shows, with only a few additional degrees of freedom, the proposed artificial boundary condition effectively eliminates the spurious reflection. The idea of the proposed method can also be used in other dispersive wave systems.

Abstract Cellular senescence describes a state of permanent proliferative arrest in cells. Studies have demonstrated that diabetes promotes the pathological accumulation of senescent cells, which in turn impairs cell movement and proliferation. Historically, senescence has been perceived to be a detrimental consequence of chronic wound healing. However, the underlying mechanism that causes senescent cells to remain in diabetic wounds is yet to be elucidated. Ferroptosis and ferritinophagy observed in diabetes are due to iron metabolism disorders, which are directly associated with the initiation and progression of diabetes. Herein, we reveal that senescent fibroblasts in diabetic wounds are resistant to ferroptosis and that impaired ferritinophagy may be a contributing cause. Further, the expression of NCOA4, a key factor that influences ferritinophagy, is decreased in both diabetic wound tissue and high glucose-induced senescent fibroblasts. Moreover, NCOA4 overexpression could render senescent fibroblasts more vulnerable to ferroptosis. A faster wound healing process was also linked to the induction of ferroptosis. Thus, resistance to ferroptosis impedes the removal of senescent fibroblasts; promoting ferritinophagy could reverse this process, which may have significant implications for the management of diabetic wounds.

Citrus, one of the most widely cultivated fruit crops in the world, relies on arbuscular mycorrhizal fungi (AMF) to absorb nutrients and water from soil. However, the molecular mechanism of AM symbiosis (AMS) in citrus in general have largely been understudied. Here, using a TMT labeling proteomic approach, we identified 365 differentially expressed proteins (DEPs) in roots of Poncirus trifoliata (a common citrus rootstock) upon Rhizophagus irregularis colonization as compared with uninoculated roots, of which 287 were up-regulated and 78 were down-regulated. GO analysis revealed that the DEPs were mainly involved in biological processes such as negative regulation of endopeptidase inhibitor activity, negative regulation of endopeptidase, one-carbon metabolic process and carbohydrate metabolic process. KEGG enrichment analysis indicated that the DEPs were mainly involved in regulating metabolic pathways such as fatty acid biosynthesis, phenylpropanoid biosynthesis and carbon metabolism. Furthermore, 194 of the 365 DEPs were found to be associated with AMS-responsive genes by association analysis with our previous transcriptomes data, which highlighted the important roles of these proteins in AMS. One of the 194 DEPs, neutral ceramidase (PtNCER), was further chosen for function analysis via RNAi interfering its homologous gene MtNCER in a mycorrhizal model plant Medicago truncatula, which confirmed a positive role of NCER in AM establishment. Our results provided basic data and key candidate genes for genetic improvement of efficient nutrient uptake through AM establishment in citrus and other crops.

This study addressed the autonomous planning of three-dimensional (3D) underwater inspection paths for autonomous underwater vehicles (AUVs) in marine ranching by integrating differential evolution and particle swarm optimization (PSO) algorithms. First, a modified PSO algorithm incorporating swap operators, mutation, and crossover strategies was employed to enable autonomous obstacle avoidance during the inspection of offshore net cages in fish farms situated within a 3D marine environment. This approach addresses the problem of planning full-traversal paths for multiple inspection points. Second, the performance of the proposed algorithm was assessed through comparative tests with other algorithms. The proposed algorithm demonstrated significant improvements in convergence speed, accuracy, and stability under complex scenarios involving multiple optima and intense oscillations. To validate the superiority and overall planning proficiency of the modified method, an experimental setup comprising of two distinct 3D marine cage environments with a series of checkpoints was utilized. The experimental results demonstrated the ability of the proposed algorithm to generate an optimal path while traversing all inspection points of fish farm offshore net cages. By ensuring the safety of AUVs and closely adhering to the surfaces of offshore net cages during the inspection process, the algorithm exhibits remarkable adaptability to specific application scenarios, effectively mitigating concerns related to local optima and premature convergence.

Lung adenocarcinoma, which is the second most prevalent cancer in the world, has a poor prognosis and a low 5-year survival rate. The MS4A protein family is crucial to disease development and progression, particularly for cancers, allergies, metabolic disorders, autoimmune diseases, infections, and neurodegenerative disorders. However, its involvement in lung adenocarcinoma remains unclear. In this study, we found that 11 MS4A family genes were upregulated or downregulated in lung adenocarcinoma. Furthermore, we described the genetic variation landscape of the MS4A family in lung adenocarcinoma. Notably, through functional enrichment analysis, we discovered that the MS4A family is involved in the immune response regulatory signaling pathway and the immune response regulatory cell surface receptor signaling pathway. According to the Kaplan–Meier curve, patients with lung adenocarcinoma having poor expression of MS4A2, MS4A7, MS4A14, and MS4A15 had a low overall survival rate. These four prognostic genes are substantially associated with immune-infiltrating cells, and a prognosis model incorporating them may more accurately predict the overall survival rate of patients with lung adenocarcinoma than current models. The findings of this study may offer creative suggestions and recommendations for the identification and management of lung adenocarcinoma.

BackgroundThe most commonly acknowledged non-scarring alopecia are androgenetic alopecia (AGA) and alopecia areata (AA). Previous studies have revealed various risk factors associated with alopecia. However, the relationship between leukocyte telomere length (LTL) and non-scarring alopecia remains unclear.MethodsA two-sample Mendelian randomization (MR) analysis was performed to evaluate the causality between genetically predicted LTL and the risk of non-scarring alopecia. MR analyses were performed using the inverse variance-weighted (IVW) method and complemented with other MR methods.ResultsThe summary statistics of the genome-wide association studies (GWAS) for AGA and AA were obtained from the FinnGen biobank, which included 119,185 and 211,428 individuals, respectively. A total of 126 single nucleotide polymorphisms (SNPs) with genome-wide significance were selected as the instrumental variables for LTL. The MR analyses suggested a causal relationship between LTL and AGA, and the risk of AGA increased by 3.19 times as the genetically predicted LTL was shortened by one standard deviation in log transformed form under the IVW method (OR = 4.19, 95% CI = 1.20–14.61, p = 0.024). The other MR methods also demonstrated a similar trend of the effect of LTL on AGA. There was no causal relationship between LTL and AA (p > 0.05). Sensitivity analyses further demonstrated that the current results were less likely to be affected by confounders and bias.ConclusionOur results suggested a potential causal relationship between LTL and AGA, and shortened LTL was associated with an increased risk of AGA.

Although employed to release growth factors (GFs) for regenerative medicine, platelet-rich plasma (PRP) has been hindered by issues like burst effect. Based on collagen sponge scaffolds (CSSs) modified with polydopamine (pDA), a novel dermal regeneration template (DRT) was designed. However, whether it could efficiently deliver PRP and even foster wound healing remained unclear. In this work, after PRP was prepared and pDA-modified CSSs (pDA-CSSs) were fabricated, microscopic observation, GFs release assay and in-vitro biological evaluations of pDA-CSSs with PRP (pDA-CSS@PRP) were performed, followed by BALA-C/nu mice full-thickness skin defects implanted with pDA-CSS@PRP covered by grafted skins (termed as a One-step strategy). As a result, scanning electron microscope demonstrated more immobilized platelets on pDA-CSS′ surface with GFs’ controlled release via enzyme-linked immunosorbent assay, compared with CSSs. In line with enhanced in-vitro proliferation, adhesion and migration of keratinocytes & endothelial cells, pDA-CSS@PRP were histologically revealed to accelerate wound healing with less scar via rapid angiogenesis, arrangement of more mature collagen, guiding cells to spread, etc. In conclusion, pDA-CSSs have potential to serve as a novel DRT capable of delivering PRP, which may foster full-thickness skin defect healing by means of a One-step strategy.

Nanofiber scaffolds have emerged as a revolutionary drug delivery platform for promoting wound healing, due to their unique properties, including high surface area, interconnected porosity, excellent breathability, and moisture absorption, as well as their spatial structure which mimics the extracellular matrix. However, the use of nanofibers to achieve controlled drug loading and release still presents many challenges, with ongoing research still exploring how to load drugs onto nanofiber scaffolds without loss of activity and how to control their release in a specific spatiotemporal manner. This comprehensive study systematically reviews the applications and recent advances related to drug-laden nanofiber scaffolds for skin-wound management. First, we introduce commonly used methods for nanofiber preparation, including electrostatic spinning, sol–gel, molecular self-assembly, thermally induced phase separation, and 3D-printing techniques. Next, we summarize the polymers used in the preparation of nanofibers and drug delivery methods utilizing nanofiber scaffolds. We then review the application of drug-loaded nanofiber scaffolds for wound healing, considering the different stages of wound healing in which the drug acts. Finally, we briefly describe stimulus-responsive drug delivery schemes for nanofiber scaffolds, as well as other exciting drug delivery systems.

Abstract Background Three-dimensional (3D) cultivation with biomaterials was proposed to facilitate stem cell epithelial differentiation for wound healing. However, whether human adipose-derived stem cells (hASCs) on collagen sponge scaffold (CSS) better differentiate to keratinocytes remains unclear. Methods 3D cultivation with CSS on hASC epidermal differentiation co-cultured with HaCaT cells at air-liquid interface (ALI) was compared with two-dimensional (2D) form and cultivation without “co-culture” or “ALI.” Cellular morphology, cell adhesion, and growth condition were evaluated, followed by the protein and gene expression of keratin 14 (K14, keratinocyte specific marker). Results Typical cobblestone morphology of keratinocytes was remarkably observed in co-cultured hASCs at ALI, but those seeded on the CSS exhibited more keratinocyte-like cells under an invert microscope and scanning electron microscope. Desired cell adhesion and proliferation were confirmed in 3D differentiation groups by rhodamine-labeled phalloidin staining, consistent with H&E staining. Compared with those cultured in 2D culture system or without “ALI,” immunofluorescence staining and gene expression analysis revealed hASCs co-cultured over CSS expressed K14 at higher levels at day 15. Conclusions CSS is positive to promote epithelial differentiation of hASCs, which will foster a deeper understanding of artificial dermis in skin wound healing and regeneration.

Two-dimensional materials have opened up extensive applications for traditional materials. In particular, heterostructures can further create fantastic performances. In this investigation, the lateral heterostructure was constructed using Janus MoSSe and WSSe monolayers with armchair and zigzag interfaces. Performing first-principles calculations and molecular dynamics simulation method, the thermal stability and the semiconductor characteristics with the type-II band structure to separate the photogenerated charges of such Janus MoSSe/WSSe heterostructure are presented, which suggests the potential application of acting as a photocatalyst for water splitting. Importantly, the asymmetric interface of the Janus MoSSe/WSSe heterostructure can result in natural bending, which limits the heat flow transport. Smaller heat flow and the interfacial thermal resistance of the lateral MoSSe/WSSe heterostructure with a zigzag edge interface are mainly due to suppressed acoustic branches. These structural symmetry and interface-dependent properties show the future applications in photovoltaic and thermoelectric devices.

Deformations in lithium-ion batteries, which may lead to thermal runaway, can occur during storage and transportation handling, as well as in road use. In this study, both radial and axial compression deformation were produced experimentally to analyze their influence on the performance and safety of lithium-ion batteries. In the radial plate compression experiment, the battery was loaded to different displacements and then charge–discharge cycles were performed. It was found that the greater the deformation of the battery, the smaller the initial capacity and the faster the capacity decay. Under axial loading, the voltage of low state of charge (SOCs) batteries showed a clear step-drop phenomenon. The battery was compressed until the first voltage drop, loading was stopped, and the voltage gradually dropped to 0 V. For high-SOC lithium-ion batteries, there was almost no voltage step-down, and a small deformation could cause thermal runaway in the battery. The results showed that the small deformation in the radial direction only reduced the capacity of the battery, but had little impact on its safety, whereas a small deformation in the axial direction was more likely to cause an internal short circuit (ISC).

The information resources organization and management model were discussed. Firstly, based on the principles of information service, a network and big data service integration model was built. Secondly, the information integration, sharing and access mechanism were introduced respectively. Finally, agriculture information sharing service and platform implementation were given, which provided the theoretical and application demonstration for intelligence agriculture factory information construction and realization.

The characteristics of the small and medium-sized manufacturing enterprises information system in China were analyzed. Combined with the“cloud manufacture”technology and service concept, an intelligence information service based on cloud model was introduced. The overall system architecture of manufacturing resources layer, the network layer, the virtual service layer, the core service layer and the wisdom application layer were put forward. Also the virtual service mechanism and service delivery methods were explained.

In this paper, a Susceptible-Exposed-Asymptomatic-Infectious-Recovered (SEAIR) epidemic model with application to COVID-19 is established by capturing the key features of the disease. The global dynamics of the model is analyzed by constructing appropriate Lyapunov functions utilizing the basic reproduction number R0 as an index. We obtain that when R0 < 1, the disease-free equilibrium is globally asymptotically stable. While for R0 > 1, the endemic equilibrium is globally asymptotically stable. Furthermore, we consider the pulse vaccination for the disease and give an impulsive differential equations model. The definition of the basic reproduction number R0 of this system is given by utilizing the next generation operator. By the comparison theorem and persistent theory, we obtain that when R0 < 1, the disease-free periodic solution is globally asymptotically stable. Otherwise, the disease will persist and there will be at least one nontrivial periodic solution. Numerical simulations to verify our conclusions are given at the end of each of these theorems. Objective: To determine the effects of varying positive end-expiratory pressures (PEEPs) on right ventricular function, hemodynamics, oxygenation, and the incidence of acute cor pulmonale (ACP) in patients with moderate-to-severe acute respiratory distress syndrome (ARDS)., Methods: This prospective paired-design study involved patients with moderate-to-severe ARDS in the ICU. Participants received lung-protective ventilation and hemodynamic monitoring. During the study, mechanical ventilation was administered with PEEPs of 5 cmH2O, 10 cmH2O, and 15 cmH2O, while maintaining an end-inspiratory plateau pressure ≤ 30 cmH2O. Various assessments, including transthoracic echocardiography, cardiac output measurement, and blood gas analysis, were conducted at baseline and after 1 h of ventilation at each PEEP. Subsequently, variations in ventilation oxygenation, echocardiographic parameters, and hemodynamic indicators under different PEEPs were analyzed to explore the potential effects of PEEP on right ventricular function and hemodynamics, as well as the incidence of ACP., Results: A total of 317 ARDS patients were screened. Among them, 104 met the diagnostic criteria for moderate-to-severe ARDS, and 52 completed the study. The baseline PEEP of these 52 participants, acquired before commencement, was 11.5 ± 1.7 cmH2O, and the incidence of ACP was 25.0% (13/52). Intensive care unit mortality, overall hospital mortality, and 28-day mortality rates were 19.2% (10/52), 21.2% (11/52), and 32.7% (17/52), respectively. During the study, ACP incidences at PEEPs of 5 cmH2O, 10 cmH2O, and 15 cmH2O were 17.3% (9/52), 21.2% (11/52), and 38.5% (20/52), respectively. Meanwhile, the PaO2/FiO2 ratio improved with increasing PEEP, reaching 162.0 (140.9, 174.0), 171.0 (144.0, 182.0), and 176.5 (151.0, 196) mmHg at PEEPs of 5 cmH2O, 10 cmH2O, and 15 cmH2O, respectively. In addition, higher PEEPs were associated with a slight increase in PaCO 2 , showing statistically significant differences compared to moderate and low PEEPs. Compared to a PEEP of 5 cmH2O or 10 cmH2O, right ventricular function exhibited substantial changes at 15 cmH2O PEEP, manifested as increased pulmonary artery systolic pressure, enlarged right ventricular end-diastolic area, and decreased tricuspid annular plane systolic excursion, all with significant differences. Conversely, variations in left ventricular end-diastolic area and ejection fraction were not statistically significant. In terms of hemodynamics, increasing PEEP resulted in a decline in cardiac index (CI), with statistically significant differences between different PEEPs. Specifically, compared to the value at a PEEP of 5 cmH2O, the CI at a PEEP of 15 cmH2O decreased by 14.3% (2.63 [2.20, 2.95] vs. 3.07 [2.69, 3.67], p  < 0.001). The decline in the stroke volume index with PEEP was more obvious (22.1 [18.4, 27.1] vs. 27.0 [24.2, 33.0], p  < 0.001), reaching 18.1%. Additionally, both end-diastolic volume index and extravascular lung water index decreased significantly with increasing PEEP, while the pulmonary vascular permeability index remained unaffected., Conclusion: Different PEEPs can affect the incidence of ACP in patients with moderate-to-severe ARDS. High PEEP improves oxygenation and reduces extravascular lung water without significantly affecting the pulmonary vascular permeability index and left ventricular systolic function.
