The reconstruction of visual stimuli from fMRI signals, which record brain activity, is a challenging task with crucial research value in the fields of neuroscience and machine learning. Previous studies tend to emphasize reconstructing pixel-level features (contours, colors, etc.) or semantic features (object category) of the stimulus image, but typically, these properties are not reconstructed together. In this context, we introduce a novel three-stage visual reconstruction approach called the Dual-guided Brain Diffusion Model (DBDM). Initially, we employ the Very Deep Variational Autoencoder (VDVAE) to reconstruct a coarse image from fMRI data, capturing the underlying details of the original image. Subsequently, the Bootstrapping Language-Image Pre-training (BLIP) model is utilized to provide a semantic annotation for each image. Finally, the image-to-image generation pipeline of the Versatile Diffusion (VD) model is utilized to recover natural images from the fMRI patterns guided by both visual and semantic information. The experimental results demonstrate that DBDM surpasses previous approaches in both qualitative and quantitative comparisons. In particular, the best performance is achieved by DBDM in reconstructing the semantic details of the original image; the Inception, CLIP and SwAV distances are 0.611, 0.225 and 0.405, respectively. The cryptographic system is the foundation of blockchain security mechanisms. This paper constructs a blockchain security mechanism based on the Chinese national cryptographic SM2 algorithm, involving the generation of public–private key pairs and the signing of block information using key pairs. However, the original SM2 algorithm has a high computational complexity, which cannot meet the requirements of low on-chain delay in blockchain applications. To solve this problem, this paper optimizes the SM2 digital signature algorithm. The scalar multiplication operation in the digital signature algorithm has a large computational complexity, which affects the efficiency of a digital signature. To reduce its computational complexity, this paper improves the traditional window non-adjacent form (w-NAF) algorithm for the scalar multiplication operation and proposes a low computational complexity Fw-NAF algorithm based on the prime number precomputation window, which greatly reduces its computational complexity. Background: Evaluation of vestibular graviceptive pathway (VGP) in patients with unilateral peripheral vestibular dysfunction (UPVD) has received increasing attention from researchers. The study aimed to investigate the value of VGP evaluation in the diagnosis of UPVD. Methods: Ninety‐five UPVD patients were divided into attack and remission phase groups. VGP evaluation‐related indicators, including subjective visual vertical (SVV), subjective visual horizontal (SVH), head tilt, ocular torsion (OT), and skew deviation (SD), were measured, and their correlations with cochleovestibular function test results were analyzed. The possible etiologies of contralesional VGP (c‐VGP) were analyzed. Results: Positive rates of SVV, SVH, OT, and SD were significantly higher, and the degrees of SVV, SVH, and OT were significantly greater in the attack phase group than the remission phase group. The sides with abnormal VGP evaluation results were correlated with the sides with hearing loss, abnormal caloric, and video head impulse test (vHIT) results. A total of 14 patients showed c‐VGP, and possible etiologies included contralateral benign paroxysmal positional vertigo (n = 4), bilateral hearing loss (n = 8), bilateral vHIT gain reduction (n = 1), autoimmune diseases (n = 6), vascular risk factors (n = 6), lacunar infarction (n = 3), and endolymphatic hydrops (n = 3). Conclusions: Alterations in SVV, SVH, OT, and SD were noted in UPVD patients in different phases, which are presumed to be related to dynamic vestibular compensation; correlations between VGP evaluation results and cochleovestibular function test results indicate that VGP evaluation may be helpful for the diagnosis of the side affected in UPVD; the presence of c‐VGP may be related to bilateral labyrinth lesions or endolymphatic hydrops on the affected side; and the involvement of autoimmune mechanisms also deserves attention. Summary: Wheat fixes CO2 by photosynthesis into kernels to nourish humankind. Improving the photosynthesis rate is a major driving force in assimilating atmospheric CO2 and guaranteeing food supply for human beings. Strategies for achieving the above goal need to be improved. Here, we report the cloning and mechanism of CO2ASSIMILATION RATE AND KERNEL‐ENHANCED 1 (CAKE1) from durum wheat (Triticum turgidum L. var. durum). The cake1 mutant displayed a lower photosynthesis rate with smaller grains. Genetic studies identified CAKE1 as HSP90.2‐B, encoding cytosolic molecular chaperone folding nascent preproteins. The disturbance of HSP90.2 decreased leaf photosynthesis rate, kernel weight (KW) and yield. Nevertheless, HSP90.2 over‐expression increased KW. HSP90.2 recruited and was essential for the chloroplast localization of nuclear‐encoded photosynthesis units, for example PsbO. Actin microfilaments docked on the chloroplast surface interacted with HSP90.2 as a subcellular track towards chloroplasts. A natural variation in the hexaploid wheat HSP90.2‐B promoter increased its transcription activity, enhanced photosynthesis rate and improved KW and yield. Our study illustrated an HSP90.2–Actin complex sorting client preproteins towards chloroplasts to promote CO2 assimilation and crop production. The beneficial haplotype of Hsp90.2 is rare in modern varieties and could be an excellent molecular switch promoting photosynthesis rate to increase yield in future elite wheat varieties. Objectives: Oogonial stem cells (OSCs) are germ cells that can sustain neo‐oogenesis to replenish the pool of primary follicles in adult ovaries. In lower vertebrates, fresh oocytes are produced by numerous OSCs through mitosis and meiosis during each reproduction cycle, but the OSCs in adult mammals are rare. The birds have retained many conserved features and developed unique features of ovarian physiology during evolution, and the presence of OSCs within avian species remain unknown. Materials and Methods: In this study, we investigated the existence and function of OSCs in adult chickens. The chicken OSCs were isolated and expanded in culture. We then used cell transplantation system to evaluate their potential for migration and differentiation in vivo. Results: DDX4/SSEA1‐positive OSCs were identified in both the cortex and medulla of the adult chicken ovary. These putative OSCs undergo meiosis in the reproductively active ovary. Furthermore, the isolated OSCs were expanded in vitro for months and found to express germline markers similar to those of primordial germ cells. When transplanted into the bloodstream of recipient embryos, these OSCs efficiently migrated into developing gonads, initiated meiosis, and then derived oocytes in postnatal ovaries. Conclusions: This study has confirmed the presence of functional OSCs in birds for the first time. Neutralizing antibodies (nAbs) are important assets to fight COVID-19, but most existing nAbs lose the activities against Omicron subvariants. Here, we report a human monoclonal antibody (Ab08) isolated from a convalescent patient infected with the prototype strain (Wuhan-Hu-1). Ab08 binds to the receptor-binding domain (RBD) with pico-molar affinity (230 pM), effectively neutralizes SARS-CoV-2 and variants of concern (VOCs) including Alpha, Beta, Gamma, Mu, Omicron BA.1 and BA.2, and to a lesser extent for Delta and Omicron BA.4/BA.5 which bear the L452R mutation. Of medical importance, Ab08 shows therapeutic efficacy in SARS-CoV-2-infected hACE2 mice. X-ray crystallography of the Ab08-RBD complex reveals an antibody footprint largely in the β-strand core and away from the ACE2-binding motif. Negative staining electron-microscopy suggests a neutralizing mechanism through which Ab08 destructs the Spike trimer. Together, our work identifies a nAb with therapeutic potential for COVID-19. The NAC (NAM, ATAF1/2 and CUC2) is a large gene family of plant-specific transcription factors that play a pivotal role in various physiological processes and abiotic stresses. Due to the lack of genome-wide characterization, intraspecific and interspecific synteny, and drought-responsive expression pattern of NAC genes in poplar, the functional characterization of drought-related NAC genes have been scarcely reported in Populus species. Here, we identified a total of 170 NAC domain-containing genes in the P. trichocarpa genome, 169 of which were unevenly distributed on its nineteen chromosomes. These NAC genes were phylogenetically divided into twenty subgroups, some of which exhibited a similar pattern of exon–intron architecture. The synteny and Ka/Ks analysis indicated that the expansion of NAC genes in poplar was mainly due to gene duplication events occurring before and after the divergence of Populus and Salix. Ten PdNAC (P. deltoids × P. euramericana cv.'Nanlin895') genes were randomly selected and cloned. Their drought-responsive expression profiles showed a tissue-specific pattern. The transcription factor PdNAC013 was verified to be localized in the nucleus. Our research results provide genomic information for the expansion of NAC genes in the poplar genome, and for further characterizing putative poplar NAC genes associated with water-deficit. QT interval length is an important risk factor for adverse cardiovascular outcomes; however, the genetic architecture of QT interval remains incompletely understood. We conducted a genome-wide association study of 76,995 ancestrally diverse Kaiser Permanente Northern California members enrolled in the Genetic Epidemiology Research on Adult Health and Aging cohort using 448,517 longitudinal QT interval measurements, uncovering 9 novel variants, most replicating in 40,537 individuals in the UK Biobank and Population Architecture using Genomics and Epidemiology studies. A meta-analysis of all 3 cohorts (n = 117,532) uncovered an additional 19 novel variants. Conditional analysis identified 15 additional variants, 3 of which were novel. Little, if any, difference was seen when adjusting for putative QT interval lengthening medications genome-wide. Using multiple measurements in Genetic Epidemiology Research on Adult Health and Aging increased variance explained by 163%, and we show that the 6 measurements in Genetic Epidemiology Research on Adult Health and Aging was equivalent to a 2.4 increase in sample size of a design with a single measurement. The array heritability was estimated at 17%, approximately half of our estimate of 36% from family correlations. Heritability enrichment was estimated highest and most significant in cardiovascular tissue (enrichment 7.2, 95% CI= 5.7–8.7, P = 2.1e-10), and many of the novel variants included expression quantitative trait loci in heart and other relevant tissues. Brain decoding is to predict the external stimulus information from the collected brain response activities, and visual information is one of the most important sources of external stimulus information. Decoding functional magnetic resonance imaging (fMRI) based on visual stimulation is helpful in understanding the working mechanism of the brain visual function regions. Traditional brain decoding algorithms cannot accurately extract stimuli features from fMRI. To address these shortcomings, this paper proposed a brain decoding algorithm based on a graph convolution network (GCN). Firstly, 11 regions of interest (ROI) were selected according to the human brain visual function regions, which can avoid the noise interference of the non-visual regions of the human brain; then, a deep three-dimensional convolution neural network was specially designed to extract the features of these 11 regions; next, the GCN was used to extract the functional correlation features between the different human brain visual regions. Furthermore, to avoid the problem of gradient disappearance when there were too many layers of graph convolutional neural network, the residual connections were adopted in our algorithm, which helped to integrate different levels of features in order to improve the accuracy of the proposed GCN. The proposed algorithm was tested on the public dataset, and the recognition accuracy reached 98.67%. Compared with the other state-of-the-art algorithms, the proposed algorithm performed the best. The third-generation epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs), osimertinib, aumolertinib, and furmonertinib represent a new treatment option for patients with EGFR p.Thr790 Met (T790 M)-mutated non-small cell lung cancer (NSCLC). Currently, there are no studies reporting the simultaneous quantification of these three drugs. A simple ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS) method was developed and validated for the simultaneous quantitative determination of osimertinib, aumolertinib, and furmonertinib concentrations in human plasma, and it was applied for therapeutic drug monitoring (TDM). Plasma samples were processed using the protein precipitation method (acetonitrile). A positive ion monitoring mode was used for detecting analytes. D3-Sorafenib was utilized as the internal standard (IS), and the mobile phases were acetonitrile (containing 0.1% formic acid) and water with gradient elution on an XSelect HSS XP column (2.1 mm × 100.0 mm, 2.5 µm, Waters, Milford, MA, USA) at a flow rate of 0.5 mL·min−1. The method's selectivity, precision (coefficient of variation of intra-day and inter-day ≤ 6.1%), accuracy (95.8–105.2%), matrix effect (92.3–106.0%), extraction recovery, and stability results were acceptable according to the guidelines. The linear ranges were 5–500 ng·mL−1, 2–500 ng·mL−1, and 0.5–200 ng·mL−1 for osimertinib, aumolertinib, and furmonertinib, respectively. The results show that the method was sensitive, reliable, and simple and that it could be successfully applied to simultaneously determine the osimertinib, aumolertinib, and furmonertinib blood concentrations in patients. These findings support using the method for TDM, potentially reducing the incidence of dosing blindness and adverse effects due to empirical dosing and inter-patient differences. We propose an alternative approach to Lüscher's formula for extracting two-body scattering phase shifts from finite volume spectra with no reliance on the partial wave expansion. We use an effective-field-theory-based Hamiltonian method in the plane wave basis and decompose the corresponding matrix elements of operators into irreducible representations of the relevant point groups. The proposed approach allows one to benefit from the knowledge of the long-range interaction and avoids complications from partial wave mixing in a finite volume. We consider spin-singlet channels in the two-nucleon system and pion-pion scattering in the ρ-meson channel in the rest and moving frames to illustrate the method for non-relativistic and relativistic systems, respectively. For the two-nucleon system, the long-range interaction due to the one-pion exchange is found to make the single-channel Lüscher formula unreliable at the physical pion mass. For S-wave dominated states, the single-channel Lüscher method suffers from significant finite-volume artifacts for a L = 3 fm box, but it works well for boxes with L > 5 fm. However, for P-wave dominated states, significant partial wave mixing effects prevent the application of the single-channel Lüscher formula regardless of the box size (except for the near-threshold region). Using a toy model to generate synthetic data for finite-volume energies, we show that our effective-field-theory-based approach in the plane wave basis is capable of a reliable extraction of the phase shifts. For pion-pion scattering, we employ a phenomenological model to fit lattice QCD results at the physical pion mass. Wearable devices are increasingly popular for health monitoring via electrocardiograms (ECGs) as they can portably monitor heart conditions over a long time. However, so far there are no publicly available ECG data sets collected from wearable devices. Most ECG analysis algorithms target ECG data collected by hospital equipment. In the present study, we used the IREALCARE2.0 Flexible Cardiac Monitor Patch as the wearable device to collect ECG signals and formed ECG data sets. Wearable ECG data tended to contain more interference and be large in size. This article proposed a deep CNN approach, named time–spatial convolutional neural networks (TSCNNs), for the automatic classification and analysis of ECG signals from wearable devices. First, the original long-term ECG signals were divided into separate heartbeats and input into the TSCNN. Second, we applied convolution over time and spatial filtering for each heartbeat to extract abundant features. Finally, the cascaded small-scale kernel convolution was applied to improve classification performance and reduce the number of network parameters. To avoid overfitting, some regularized methods such as dropout and batch normalization were adopted. In the experiments, the method proposed in this letter is compared with other eight ECG classification algorithms. Our method attained the highest classification accuracy. The experimental results indicated that the proposed method can achieve better performance for wearable ECG data and can effectively monitor whether the wearer has an abnormal ECG. Dairy cows with mastitis are frequently treated with antibiotics. The potential effect of antibiotics on the milk microbiome is still not clear. Therefore, the objective of this research was to investigate the effect of 2 commonly used cephalosporins on the milk microbiota of dairy cows and the antibiotic resistance genes in the milk. The milk samples were collected from 7 dairy cows at the period before medication (d 0), medication (d 1, 2, 3), withdrawal period (d 4, 6, 8), and the period after withdrawal (d 9, 11, 13, 15). We applied 16S rRNA sequencing to explore the microbiota changes, and antibiotic resistance patterns were investigated by quantitative PCR. The microbiota richness and diversity in each sample were calculated using the Chao 1 (richness), Shannon (diversity), and Simpson (diversity) indices. The cephalosporins treatment lowered the Simpson diversity value at the period of withdrawal. Members of the Enterobacter genera were the most affected bacteria associated with mastitis. Meanwhile, antibiotic resistance genes in the milk were also influenced by antibiotic treatment. The cephalosporins treatment raised the proportion of bla TEM in milk samples at the period of withdrawal. Therefore, the treatment of cephalosporins led to change in the milk microbiota and increase of β-lactam resistance gene in the milk at the time of withdrawal period. [ABSTRACT FROM AUTHOR]

Background: Freezing stress is the key factor that affecting the cell activity and fermentation performance of baker's yeast in frozen dough production. Generally, cells protect themselves from injury and maintain metabolism by regulating gene expression and modulating metabolic patterns in stresses. The Snf1 protein kinase is an important regulator of yeast in response to stresses. In this study, we aim to study the role of the catalytic subunit of Snf1 protein kinase in the cell tolerance and dough leavening ability of baker's yeast during freezing. Furthermore, the effects of SNF1 overexpression on the global gene expression and metabolite profile of baker's yeast before and after freezing were analysed using RNA-sequencing and untargeted UPLC − QTOF-MS/MS, respectively. Results: The results suggest that overexpression of SNF1 was effective in enhancing the cell tolerance and fermentation capacity of baker's yeast in freezing, which may be related to the upregulated proteasome, altered metabolism of carbon sources and protectant molecules, and changed cell membrane components. SNF1 overexpression altered the level of leucin, proline, serine, isoleucine, arginine, homocitrulline, glycerol, palmitic acid, lysophosphatidylcholine (LysoPC), and lysophosphatidylethanolamine (LysoPE) before freezing, conferring cells resistance in freezing. After freezing, relative high level of proline, lysine, and glycerol maintained by SNF1 overexpression with increased content of LysoPC and LysoPE. Conclusions: This study will increase the knowledge of the cellular response of baker's yeast cells to freezing and provide new opportunities for the breeding of low-temperature resistant strains. Background. S100 family genes exclusively encode at least 20 calcium-binding proteins, which possess a wide spectrum of intracellular and extracellular functions in vertebrates. Multiple lines of evidences suggest that dysregulated S100 proteins are associated with human malignancies including colorectal cancer (CRC). However, the diverse expression patterns and prognostic roles of distinct S100 genes in CRC have not been fully elucidated. Methods. In the current study, we analyzed the mRNA expression levels of S100 family genes and proteins and their associations with the survival of CRC patients using the Oncomine analysis and GEPIA databases. Expressions and mutations of S100 family genes were analyzed using the cBioPortal, and protein-protein interaction (PPI) networks of S100 proteins and their mutation-related coexpressed genes were analyzed using STRING and Cytoscape. Results. We observed that the mRNA expression levels of S100A2, S100A3, S100A9, S100A11, and S100P were higher and the level of S100B was lower in CRC tissues than those in normal colon mucosa. A high S100A10 levels was associated with advanced-stage CRC. Results from GEPIA database showed that highly expressed S100A1 was correlated with worse overall survival (OS) and disease-free survival (DFS) and that overexpressions of S100A2 and S100A11 were associated with poor DFS of CRC, indicating that S100A1, S100A2, and S100A11 are potential prognostic markers. Unexpectedly, most of S100 family genes showed no significant prognostic values in CRC. Conclusions. Our findings, though still need to be ascertained, offer novel insights into the prognostic implications of the S100 family in CRC and will inspire more clinical trials to explore potential S100-targeted inhibitors for the treatment of CRC. Background: The Saccharomyces cerevisiae Snf1 complex is a member of the AMP-activated protein kinase family and plays an important role in response to environmental stress. The α catalytic subunit Snf1 regulates the activity of the protein kinase, while the β regulatory subunits Sip1/Sip2/Gal83 specify substrate preferences and stress response capacities of Snf1. In this study, we aim to investigate the effects of SNF1 overexpression on the cell tolerance and glucose consumption of S. cerevisiae in high glucose, ethanol, and heat stresses and to explore the valid Snf1 form in the light of β subunits in these stresses. Results: The results suggest that overexpression of SNF1 is effective to improve cell resistance and glucose consumption of S. cerevisiae in high glucose, ethanol, and heat stresses, which might be related to the changed accumulation of fatty acids and amino acids and altered expression levels of genes involved in glucose transport and glycolysis. However, different form of β regulatory subunits dominated in stresses with regard to cell tolerance and glucose utilization. The Sip1 isoform was more necessary to the growth and glucose consumption in ethanol stress. The glucose uptake largely depended on the Sip2 isoform in high sugar and ethanol stresses. The Gal83 isoform only contributed inferior effect on the growth in ethanol stress. Therefore, redundancy and synergistic effect of β subunits might occur in high glucose, ethanol, and heat stresses, but each subunit showed specificity under various stresses. Background: This study was to investigate of the mechanism by which histone deacetylase (HDAC) 8 inhibitor ameliorated airway hyperresponsiveness (AHR) and allergic airway inflammation.Methods: Mice were sensitized and then treated with budesonide (BUD) or PCI-34051 (PCI) prior to exposing to normal saline (NS) or ovalbumin (OVA). The raw264.7 cells were treated with interleukin (IL)-4 and PCI or shRNA alone. Repetitive measurements of enhanced pause (Penh) were executed by increasing concentrations of acetyl-β-methacholine chloride (0 - 50 mg/ml). Cells in bronchoalveolar lavage fluid (BALF) and pathological changes of lungs were examined, respectively. The expression levels of HDAC8, Galecitn (Gal)-3, CD68, CD86, CD163, Arg1 and NOS2 in lungs were measured. Co-regulation of HDAC8 and Gal-3 proteins was observed by immunofluorescence staining and co-immunoprecipitation assay (Co-IP).Results: Significant increases in Penh and IL-4 level were detected with a large inflammatory infiltrate, comprised predominantly of macrophages and eosinophils, into the BALF in OVA-exposed lungs. HDAC8, Gal-3, CD68, CD86, CD163, Arg1 and NOS2 proteins were over-expressed with the significant changes in the Arg1 and NOS2 mRNA levels in the lungs and the IL-4-treated cells. PCI intervention obviously reduced the counts of CD163+ cells. Furthermore, Gal-3 knockdown suppressed Arg1 expression in the cells. Immunofluorescence staining displayed simultaneous changes in HDAC8 and Gal-3 expression in the investigated samples. Treatment with PCI resulted in synchronous reduction of HDAC8 and Gal-3 expression in the Co-IP complexes.Conclusions: The HDAC8 inhibitor ameliorates AHR and airway inflammation in animal model of allergic asthma through reducing HDAC8-Gal-3 interaction and M2 macrophage polarization. Neospartoneae are a small tribe in Verbenaceae, native to temperate South America and comprising seven species in three genera: Diostea, Lampayo, and Neosparton. Diostea and Neosparton share an ephedroid habit and the presence of a staminode. Diostea and Lampayo share a schizocarpous fruit and bilobed stigma. Previous chloroplast DNA phylogenetic studies first identified Neospartoneae as a clade. However, evolutionary relationships within Neospartoneae remain unclear. In this study, nine loci from both chloroplast and nuclear genomes were used to reconstruct phylogeny with almost complete taxon sampling. Maximum likelihood and Bayesian analyses were conducted using single-locus and concatenated datasets to generate gene trees. A species tree was reconstructed by Bayesian multispecies coalescent analyses. The generic relationships are well resolved and confirm the monophyly of the Neospartoneae and each genus. The topologies show that Neosparton is sister to a clade comprised of Diostea and Lampayo. This study presents a first species-level phylogeny of Neospartoneae and provides insight into morphological character evolution of this tribe. The presence of a staminode shared by Diostea and Neosparton, and dry schizocarpous fruits shared by Diostea and Lampayo, are inferred to be plesiomorphic traits in Neospartoneae and not indicative of close relationship. The ephedroid habit and curved corolla tubes shared by Diostea and Neosparton may have evolved in the common ancestor of Neospartoneae and subsequently reversed in the ancestor of Lampayo, or have evolved in parallel in the two genera. Silver nanowires (AgNWs) are a promising substitute for indium tin oxide (ITO) transparent conductive films (TCFs). However, AgNWs suffer the insufficiencies in contact resistance between the nanowires and light loss. Here, graphene oxide and moth-eye-like antireflection nanostructure have been introduced to physically "weld" the AgNWs junctions and modulate the AR properties, respectively. The sandwich structure AgNWs TCFs achieved resistance of 26.26 Ω sq−1 and transmittance of 98.67%, exhibiting promoted photoelectronic performance compared with the pure AgNWs films. The stability of sandwich structure AgNWs TCFs was improved, and their wettability was also discussed with water contact angle changed from 92.55° to 69.18°, switched from hydrophobic to hydrophilic. Though a simple solution process without mechanical or heat post-treatment, we proposed a stable sandwich structure to obviously improve the electrical and optical capabilities of AgNWs. The mechanisms leading to self-assembly of misfolded proteins into amyloid aggregates have been studied extensively in the test tube under well-controlled conditions. However, to what extent these processes are representative of those in the cellular environment remains unclear. Using super-resolution imaging of live cells, we show here that an amyloidogenic polyglutamine-containing protein first forms small, amorphous aggregate clusters in the cytosol, chiefly by diffusion. Dynamic interactions among these clusters limited their elongation and led to structures with a branched morphology, differing from the predominantly linear fibrils observed in vitro. Some of these clusters then assembled via active transport at the microtubule-organizing center and thereby initiated the formation of perinuclear aggresomes. Although it is widely believed that aggresome formation is entirely governed by active transport along microtubules, here we demonstrate, using a combined approach of advanced imaging and mathematical modeling, that diffusion is the principal mechanism driving aggresome expansion. We found that the increasing surface area of the expanding aggresome increases the rate of accretion caused by diffusion of cytosolic aggregates and that this pathway soon dominates aggresome assembly. Our findings lead to a different view of aggresome formation than that proposed previously. We also show that aggresomes mature over time, becoming more compacted as the structure grows. The presence of large perinuclear aggregates profoundly affects the behavior and health of the cell, and our super-resolution imaging results indicate that aggresome formation and development are governed by highly dynamic processes that could be important for the design of potential therapeutic strategies. We have developed a facile one-step hydrothermal strategy to synthesize pyramid-like NiSe2 nanostructures, serving as electrode materials for battery-supercapacitor hybrid (BSH) devices. The NiSe2 nanopyramid electrode exhibits superior electrochemical performances, including a high specific capacity of 240.83 mAh g−1 at current density of 1 A g−1 and a low internal resistance of 0.85 Ω. The all-solid-state hybrid devices have been assembled with NiSe2 as the battery-type electrode and activated carbon as the capacitor-type electrode. The hybrid device exhibits a high energy density of 0.196 mWh cm−2 at power density of 1.60 mW cm−2. The internal resistance of 1.52 Ω further reveals the nature of low resistance and high conductivity for the hybrid devices. Connecting two hybrid devices in series is able to drive a red LED for more than 3 min after charging for 9 s. This work has demonstrated that the pyramid-like NiSe2 nanostructure is expected to be an ideal high specific capacity electrode for BSH devices, especially for all-solid-state energy storage devices and portable electronic devices. Taking advantage of the formation of stereocomplex (SC) crystallites between poly(L-lactide) (PLLA) and poly(D-lactide) (PDLA), we propose a facile strategy to prepare PLA microspheres with tunable morphology and crystalline structure by compounding PLLA and PDLA. With increasing PDLA content, the crystallinity of SC-PLA rose gradually until the ratio of PLLA and PDLA reached 1:1 and then fell. Correspondingly, the morphology of the microspheres were varied (smooth, porous, golf-ball like, guava like) and higher crystallinity of SC-PLA would lead to a more coarse and porous structure. Finally, three typical kinds of Rifampicin-loaded microspheres with different ratio of PLLA and PDLA (7:3, 3:7, 10:0, sorted by porosity from high to low) were prepared and the release behavior was compared. At 30 h, the cumulative release of 7:3, 3:7 and 10:0 microspheres were 32.6%, 17.8% and 6.0% respectively, indicating that the release profiles were generally determined by the porosity of the microspheres. Our findings not only provide a new strategy to prepare PLA microspheres with controllable morphology but offer additional possibilities for the applications of SC-PLA products in biomedical area. Strain localization is a common phenomenon existing in various multiscale materials or structures, e.g., the bulking bands of thin-walled structures, the local collapse of porous materials, and the crack in solid materials, etc. However, this phenomenon cannot be captured by the conventional homogenization methods, such as mean-field homogenization (e.g., Mori–Tanaka method), first-order or high-order computational homogenization (e.g., the Finite Element Square method), etc., due to the high strain gradient associated with strain localization. Aiming at this, a hybrid Direct FE2 method is proposed by combining the D-FE2 (Direct FE2) and the traditional FE methods, while the multiscale structure is modeled using the D-FE2 method in the region exhibiting low deformation gradient, and the other region displaying high deformation gradient is modeled using the traditional FE method. Moreover, a node displacement constraint and an overall node displacement constraint derived from the multilevel equilibrium equations using the Gauss–Ostrogradsky theorem are respectively prescribed to the interface between the D-FE2 model and the FE model of the multiscale structure, to enforce the energy equilibrium and deformation continuity. The proposed hybrid D-FE2 method is then applied to predict the strain localization behavior of multiscale materials or structures, including local bulking of honeycomb structures, in-situ crack propagation, and localized plastic deformation in fiber reinforced composites, etc. Comparison of the simulation results obtained from the hybrid D-FE2 method and the traditional FE method validates the accuracy, efficiency and ease of numerical implementation of the proposed hybrid D-FE2 method. • A hybrid D-FE2 method was proposed for m ultiscale modeling of strain localization. • Displacement constraints were derived for d eformation continuity at the interface. • The hybrid D-FE2 method is accurate and more e fficient compared to DNS. Octamer-binding transcription factor 4 (OCT4) is a transcription factor with a well-defined role in stem cell pluripotency. Two OCT4 isoforms, OCT4A and OCT4B, tend to be downregulated as normal cells differentiate. However, OCT4, particularly OCT4B, may become reactivated in cancer cells. Despite this observation, the exact function of OCT4B re-expression in cancer is unclear. In the present study, the role of OCT4 in breast cancer cells was determined. In particular, the ability of OCT4 to regulate key genes involved in cellular proliferation and apoptosis, two pathways that are frequently deregulated in cancer, was examined. The cyclin-dependent kinase inhibitor 2A locus encodes p16INK4a and p14ARF, two important cell cycle inhibitors. The tumor suppressor p53 also has well characterized roles in suppressing proliferation and promoting apoptosis. The present study demonstrated, via overexpression and genetic knockdown techniques, that OCT4B regulates the expression of several of these genes and ultimately regulates the rate of apoptosis of MCF-7 breast cancer cells. It was also observed that, while OCT4B and OCT4A regulate one another, it is OCT4B that serves a more prominent role in regulating the transcription of downstream genes. Taken together, the present results suggest that OCT4B is re-expressed in a number of breast cancer cell lines, where it affects both the transcription of cell cycle genes and the rate of apoptosis. Commercial milk products worldwide come not only from cows, but also from goats, buffaloes, camels, and yaks. Milk from non-bovine animals is important culturally and economically. Pseudomonas spp. are frequently linked to milk spoilage under storage temperatures. The objectives of this study were to identify Pseudomonas spp. isolated from goat, buffalo, camel, and yak milks, and to measure proteolytic activity of Pseudomonas spp. under different storage temperatures. Raw milk samples of goat (n = 50), buffalo (n = 25), camel (n = 25), and yak (n = 25) were collected from 5 provinces in China. Pseudomonas spp. were analyzed by Pseudomonas-specific 16S, universal 16S rRNA, and rpoB gene sequence analyses. Proteolytic activity on milk agar, quantification via the trinitrobenzenesulfonic acid assay at 2°C, 4°C, 7°C, 10°C and 25°C, as well as alkaline peptidase gene (aprX) identification were performed to ascertain the proteolytic activity of these isolates. Pseudomonas spp. were found in 46 samples out of total 125 samples. A total of 67 Pseudomonas spp. were identified. Of Pseudomonas isolates, we obtained extracellular peptidase activity in 7 (10.4%) at 2°C, 17 (25.4%) at 4°C, 24 (35.8%) at 7°C, 39 (58.2%) at 10°C, and 41 (61.2%) at 25°C. The results revealed that a wide diversity of Pseudomonas spp. were present in different non-bovine raw milks, with the ability to produce peptidases at storage temperatures. In this work, we investigate the chiral corrections to the magnetic moments of the spin- $$3\over 2$$ doubly charmed baryons systematically up to next-to-next-to-leading order with the heavy baryon chiral perturbation theory. In recent years, sequential tests for detecting structural changes in time series have been adapted for deforestation monitoring using satellite data. The input time series of such sequential tests is typically a vegetation index (e.g., NDVI), which uses two or three bands and ignores all other bands. Being limited to a vegetation index will not benefit from the richer spectral information provided by newly launched satellites and will bring two bottle-necks for deforestation monitoring. Firstly, it is hard to select a suitable vegetation index a priori. Secondly, a single vegetation index is typically affected by seasonal signals, noise and other natural dynamics, which decrease its power for deforestation detection. A novel multispectral time series change monitoring method that combines dimension reduction methods with a sequential hypothesis test is proposed to address these limitations. For each location, the proposed method automatically chooses a “suitable” index for deforestation monitoring. To demonstrate our approach, we implemented it in two study areas: a dry tropical forest in Bolivia (time series length: 444) with strong seasonality and a moist tropical forest in Brazil (time series length: 225) with almost no seasonality. Background/Aims: The aminolycoside Gentamicin is a widely used antibiotic, applied in equine medicine. Despite its clinical use, concerns remain regarding the potential toxic side-effects, such as nephrotoxicity. Early detection of renal damage is critical in preclinical drug development. This study was aimed to determine whether kidney injury molecule-1 (KIM-1) and neutrophil gelatinase-associated lipocalin (NGAL) may be early indicators in the assessment of Gentamycin-induced nephrotoxicity. Methods: In our study, a model of Gentamicin-induced nephrotoxicity in male Sprague Dawley rats treated for up to 7 days at 50 or 100mg/kg/day was used to monitor the expressions of novel biomarkers of renal toxicity during the progression of acute kidney injury (AKI). Additionally, biomarkers were assessed in human kidney proximal epithelial cells (HK-2) treated with Gentamicin for 2, 6, 12, 24, 36 or 48h in vitro. Results: Repeated administration of Gentamicin to rats for 1, 3, or 7 days resulted in a dose- and time-dependent increase in the expression of KIM-1 and NGAL. The expressions of the two biomarkers changed prior to renal tubule damage and increases in serum creatinine (SCr) and blood urea nitrogen (BUN) levels, suggesting their usefulness for predicting Gentamicin-induced acute kidney injury (AKI) in vivo. Conclusions: In contrast, no significant increase in the expression of the biomarker genes and proteins were evident in HK-2 cells after treated by Gentamycin for up to 48h, suggesting that they may not be suitable endpoints for sensitive detection of nephrotoxic effects in vitro. Either arbuscular mycorrhizal (AM) fungi or earthworms as soil organisms promote plant growth, while their interaction on nitrogen (N) content in plants, soil, and glomalin-related soil protein (GRSP) is not known. In this study, an AM fungus (Funneliformis mosseae) and an earthworm (Pheretima guillelmi), either in single or in combination, were introduced into soil growing white clover to analyze their effects on plant growth, chlorophyll, soluble protein, N contents in leaves, roots, GRSP, and soil, and the contribution of N in purified GRSP to soil total N. The four-week introduction of earthworms significantly increased root mycorrhizal colonization rate, and accelerated an improved AM effect on chlorophyll and plant growth. The single introduction of earthworms significantly increased root N and soil total N contents, while the single introduction of AM fungi distinctly elevated N contents in leaves, roots, easily extractable GRSP, and soil (nitrate N and total N). The N in total GRSP was 5.78–7.70 mg g−1, accounting for 3.46 %–5.45 % of the soil total N, of which the contribution of N from easily extractable GRSP and difficult-to-extract GRSP was 1.84 %–3.07 % and 1.34 %–2.39 %, respectively. AM fungi, but not earthworms, significantly increased the contribution of N in GRSP to soil total N, and the introduction of earthworms further accelerated the increased effect of mycorrhizas on the contribution of N in easily extractable and total GRSP to soil total N. These results demonstrated that the introduction of earthworms into mycorrhizosphere can facilitate N storage in GRSP and thus a contribution to soil total N. • Effects of earthworms and AMF on N content of soil and plants were studied. • Dual earthworms and AMF addition recorded higher plant growth and chlorophyll than single addition. • Earthworms increased root N and soil total N, while AMF elevated N in plants, EE-GRSP, and soil NO 3 −−N and total N. • Dual addition recorded higher N contents in GRSPs than single addition. • Earthworm accelerated the AMF effect on the contribution of N in EE-GRSP and T-GRSP to soil total N. Lipids have been identified as important carbon sources for Mycobacterium tuberculosis (Mtb) to utilize in vivo. Thus gluconeogenesis bears a key role for Mtb to survive and replicate in host. A rate-limiting enzyme of gluconeogenesis, fructose 1, 6-bisphosphatase (FBPase) is encoded by the gene glpX. The functions of glpX were studied in M. marinum, a closely related species to Mtb. The glpX deletion strain (ΔglpX) displayed altered gluconeogenesis, attenuated virulence, and altered bacterial proliferation. Metabolic profiles indicate an accumulation of the FBPase substrate, fructose 1, 6-bisphosphate (FBP) and altered gluconeogenic flux when ΔglpX is cultivated in a gluconeogenic carbon substrate, acetate. In both macrophages and zebrafish, the proliferation of ΔglpX was halted, resulting in dramatically attenuated virulence. Intracellular ΔglpX exhibited an elongated morphology, which was also observed when ΔglpX was grown in a gluconeogenic carbon source. This elongated morphology is also supported by the observation of unseparated multi-nucleoid cell, indicating that a complete mycobacterial division in vivo is correlated with intact gluconeogenesis. Together, our results indicate that glpX has essential functions in gluconeogenesis, and plays an indispensable role in bacterial proliferation in vivo and virulence of M. marinum. Reaction mechanisms between ozone and extracellular polymeric substances (EPS) can be the key of understanding the improvements in microbial aggregates properties by low-concentration ozonation. In this study, EPS are extracted from activated sludge and treated continuously by ozone gas at 270 ± 41 ppm. The reaction between ozone and EPS was investigated by observation of EPS component concentrations, functional groups and molecular weight distributions using UV–Vis spectrometry, excitation-emission matrix fluorescence spectroscopy (EEM), high performance size-exclusion chromatography (HPSEC) and gas chromatography-mass spectrometry (GC–MS). In a 12-hour-ozonation experiment, significant ozone consumption was observed in the first 4 h and protein concentration in EPS solution was reduced by 30 ± 12%. However, the polysaccharides concentration only had a slightly decrease at the end of the ozonation process. UV–Vis spectra and EEM spectra results suggest that ozone removed protein and fluorescent matters (SMP and tryptophan-like aromatic protein) rapidly by attacking specific amino acid residues on polypeptide chain. After ozonation, the molecular weight of polysaccharide and protein dropped by 4 orders of magnitude according to HPSEC results. TOC concentration of EPS solution was reduced by 13 ± 2% after ozonation. The loss in TOC could be explained by the observation of volatile organic compounds such as carboxylic acids, aldehydes and ketones in the off-gas detected by GC–MS. The quasicrystalline alloy powders with the composition of Ti 40.83 Zr 40.83 Ni 18.34 are prepared by mechanical alloying (MA) techniques, and the bulk part is obtained by Spark Plasma Sintering (SPS) method. The structure, mechanical properties, friction and wear properties of the bulk part are investigated. The results showed that the structure of the bulk part is pure icosahedral quasicrystalline phase (I-phase). The Vickers Hardness of the bulk part is 7.03 GPa. The maximum compression strength, the elastic deformation and Young modulus is (662 ± 50) MPa, 2.7 ± 0.1% and (30 ± 3) GPa, respectively. The lowest value of friction coefficient is 0.40 at a normal load of 100 N, a sliding velocity of 10 mm/s, a sliding time of 10 min and an ambient temperature of 293.15 K. [ABSTRACT FROM AUTHOR]

Juxtanuclear aggresomes form in cells when levels of aggregation- prone proteins exceed the capacity of the proteasome to degrade them. It is widely believed that aggresomes have a protective function, sequestering potentially damaging aggregates until these can be removed by autophagy. However, most in-cell studies have been carried out over a few days at most, and there is little information on the long term effects of aggresomes. To examine these long term effects, we created inducible, singlecopy cell lines that expressed aggregation-prone polyglutamine proteins over several months. We present evidence that, as perinuclear aggresomes accumulate, they are associated with abnormal nuclear morphology and DNA double-strand breaks, resulting in cell cycle arrest via the phosphorylated p53 (Ser-15)- dependent pathway. Further analysis reveals that aggresomes can have a detrimental effect on mitosis by steric interference with chromosome alignment, centrosome positioning, and spindle formation. The incidence of apoptosis also increased in aggresome-containing cells. These severe defects developed gradually after juxtanuclear aggresome formation and were not associated with small cytoplasmic aggregates alone. Thus, our findings demonstrate that, in dividing cells, aggresomes are detrimental over the long term, rather than protective. This suggests a novel mechanism for polyglutamine-associated developmental and cell biological abnormalities, particularly those with early onset and non-neuronal pathologies. [ABSTRACT FROM AUTHOR]

Transplantation of olfactory ensheathing cells (OEC) is a promising therapy in spinal cord injury (SCI) treatment. However, the therapeutic efficacy of this method is unstable due to unknown reasons. Considering the alterations in the culture environment that occur during OEC preparation for transplantation, we hypothesize that these changes may cause variations in the curative effects of this method. In this study, we compared OEC cultured in medium containing different types and concentrations of serum. After purification and passage, the OEC were cultured for 7 days in different media containing 5%, 10%, 15% or 20% fetal bovine serum (FBS) or rat serum (RS), or the cells were cultured in FBS-containing medium first, followed by medium containing RS. In another group, the OEC were first cultured in 10% FBS for 3 days and then cultured with rat spinal cord explants with 10% RS for another 4 days. An MTT assay and P75 neurotrophin receptor immunofluorescence staining were used to examine cell viability and OEC numbers, respectively. The concentration of neurotrophin-3 (NT-3), which is secreted by OEC into the culture supernatant, was detected using the enzyme-linked immunosorbent assay (ELISA). RT-PCR was applied to investigate the NT-3 gene expression in OEC according to different groups. Compared with FBS, RS reduced OEC proliferation in relation to OEC counts (x² = 166.279, df = 1, p < 0.01), the optical density (OD) value in the MTT assay (x² = 34.730, df = 1, p < 0.01), and NT-3 concentration in the supernatant (x² = 242.997, df = 1, p < 0.01). OEC cultured with spinal cord explants secreted less NT-3 than OEC cultured alone (F = 9.611, df = 5.139, p < 0.01). Meanwhile, the order of application of different sera was not influential. There was statistically significant difference in NT-3 gene expression among different groups when the serum concentration was 15% (x² = 64.347, df = 1, p < 0.01). Earthworms and arbuscular mycorrhizal fungi (AMF) are beneficial organisms in the soil and play an important role in improving soil fertility and plant growth. The objective of this study was to analyze the response of single versus dual inoculation of earthworms (Pheretima guillelmi) and AMF (Funneliformis mosseae) on mycorrhizal growth, root morphology, and rhizosphere properties of white clover (Trifolium repens). Addition of earthworms significantly increased the degree of root AMF colonization, while decreased hyphal length and spore number in the soil. Single or dual inoculation of F. mosseae and P. guillelmi significantly increased root traits (total length, area, volume, and average diameter), soil phosphatase activities (acid, neutral, and alkaline), easily extractable and total glomalin-related soil protein content, the percentage of soil water-stable aggregates at the size of 2–4 mm, 1–2 mm and 0.5–1 mm, aggregate stability, Bray-P, and soil organic carbon (SOC) contents. Among them, the dual inoculation further amplified the effect on glomalin production, phosphatase activity, and aggregate stability, but did not show a superposition effect on root improvement, Bray-P, and SOC. A special topography and ultra-high atmospheric boundary layer conditions in the Tarim Basin (TB) lead to the unique spatial–temporal distribution characteristics of dust aerosols. A typical dust storm with persistent floating dust over the TB from 27 April to 1 May 2015 was used to investigate the characteristics of the dust radiative effect using the Weather Research and Forecasting Model with Chemistry (WRF-Chem). Based on reasonable evaluations involving in situ sounding observations, as well as remotely sensed MODIS observations of meteorology, dust aerosols, and the ultra-high atmospheric boundary layer, the simulation characterized the complete characteristics of the dust direct radiative effect (DDRE) during the dust storm outbreak stage and persistent floating dust stage over the TB. During the daytime, the shortwave (SW) radiative effect heated the atmosphere and cooled the land surface (SUR), whereas the longwave (LW) radiative effect had the opposite effect on the TB. Regarding low-level dust, the LW radiative effect was greater than the SW DDRE in the atmosphere, while for high-level dust the situation was reversed. During the nighttime, the LW DDRE at the top of the atmosphere (TOA), at the SUR, and in the atmosphere was less than that during the daytime, when the DDRE at the SUR was the most significant. In contrast to the daytime, the near-surface dust aerosols exerted an LW warming effect in the atmosphere during the nighttime; however, the dust LW radiative effect had a cooling effect from above a 100 m altitude until the top of the dust layer. In contrast, the DDRE heating rate peaked at the top of the dust layer within the TB. The event-averaged net DDRE was 0.53, −5.90, and 6.43 W m−2 at the TOA, at the SUR, and in the atmosphere over the TB, respectively. The dust SW radiative effect was stronger than the dust L4W radiative effect over the TB at the SUR and in the atmosphere. Moreover, the DDRE at the TOA was weaker than that at the SUR. In order to have a better understanding of unexplained heritability for complex diseases in conventional Genome-Wide Association Studies (GWAS), aggregated association analyses based on predefined functional regions, such as genes and pathways, become popular recently as they enable evaluating joint effect of multiple Single-Nucleotide Polymorphisms (SNPs), which helps increase the detection power, especially when investigating genetic variants with weak individual effects. In this paper, we focus on aggregated analysis methods based on the idea of Principal Component Analysis (PCA). The past approaches using PCA mostly make some inherent genotype data and/or risk effect model assumptions, which may hinder the accurate detection of potential disease SNPs that influence disease phenotypes. In this paper, we derive a general Supervised Categorical Principal Component Analysis (SCPCA), which explicitly models categorical SNP data without imposing any risk effect model assumption. We have evaluated the efficacy of SCPCA with the comparison to a traditional Supervised PCA (SPCA) and a previously developed Supervised Logistic Principal Component Analysis (SLPCA) based on both the simulated genotype data by HAPGEN2 and the genotype data of Crohn's Disease (CD) from Wellcome Trust Case Control Consortium (WTCCC). Our preliminary results have demonstrated the superiority of SCPCA over both SPCA and SLPCA due to its modeling explicitly designed for categorical SNP data as well as its flexibility on the risk effect model assumption. In this study, a method for detection of an ssRNA viral pathogen that causes viral flacherie in the silkworm, Bombyx mori (L.) (Lepidoptera: Bombycidae), was used for the detection of B. mori infectious flacherie virus (BmIFV). A combination of nested and reverse transcriptase polymerase chain reaction was used for detection. Although BmIFV has been reported in almost all the sericultural regions of the world, there had been no reports of BmIFV incidence in India. Therefore, the confirmation of the presence of BmIFV in Karnataka, India, is of great significance. The present method is advantageous because it can be used to detect the virus by using samples from infected midgut tissues, thus simplifying and avoiding laborious genome isolation procedures. Thickness of tundish cover flux (TCF) plays an important role in continuous casting (CC) steelmaking process. Traditional measurement method of TCF thickness is single/double wire methods, which have several problems such as personal security, easily affected by operators, and poor repeatability. To solve all these problems, in this paper, we specifically designed and built an instrumentation and presented a novel method to measure the TCF thickness. The instrumentation was composed of a measurement bar, a mechanical device, a high-definition industrial camera, a Siemens S7-200 programmable logic controller (PLC), and a computer. Our measurement method was based on the computer vision algorithms, including image denoising method, monocular range measurementmethod, scale invariant feature transform(SIFT), and image gray gradient detectionmethod. Using the present instrumentation and method, images in the CC tundish can be collected by camera and transferred to computer to do imaging processing. Experiments showed that our instrumentation andmethod worked well at scene of steel plants, can accurately measure the thickness of TCF, and overcome the disadvantages of traditionalmeasurement methods, or even replace the traditional ones. High temperature conventional triaxial compression test of shale were carried out by using the MTS815 servo-controlled testing machine, based on the results of the tests, the relationships between confining pressure and the characteristic of shale stress-strain curve, peak strength, elastic modulus, Poisson's ratio, peak strain were investigated,and the general regulation is obvious. According to the characteristics of volumetric strain,the stress-strain curves were classified into three types. They are dilatation type,compression-dilatation type,compression type,and the confining pressure has conspicuous dilatation effect on shale. At the same temperature, when confining pressure rises from 5 MPa to 25 MPa, shale peak strength (σ1 - σ3) is comparatively lower, indicating stronger plastic deformation', and peak strength and elastic modulus increase with uprising confining pressure. When confining pressure is lower than 15 MPa, shale Poisson' s ratio increases with increasing confining pressure while peak axial strain and peak circle strain decreases with increasing confining pressure. When confining pressure is higher than 15 MPa, shale Poisson's ratio decreases slightly with the increasing confining pressure, while peak axial strain and peak circle strain increase slightly with the increasing confining pressure. The protein structural entries grew far slower than the sequence entries. This is partly due to the bottleneck in obtaining diffraction quality protein crystals for structural determination using X-ray crystallography. The first step to achieve protein crystallization is to find out suitable chemical reagents. However, it is not an easy task. Exhausting trial and error tests of numerous combinations of different reagents mixed with the protein solution are usually necessary to screen out the pursuing crystallization conditions. Therefore, any attempts to help find suitable reagents for protein crystallization are helpful. In this paper, an analysis of the relationship between the protein sequence similarity and the crystallization reagents according to the information from the existing databases is presented. We extracted information of reagents and sequences from the Biological Macromolecule Crystallization Database (BMCD) and the Protein Data Bank (PDB) database, classified the proteins into different clusters according to the sequence similarity, and statistically analyzed the relationship between the sequence similarity and the crystallization reagents. The results showed that there is a pronounced positive correlation between them. Therefore, according to the correlation, prediction of feasible chemical reagents that are suitable to be used in crystallization screens for a specific protein is possible. [ABSTRACT FROM AUTHOR]

Sequence alignment depends on the scoring function that defines similarity between pairs of letters. For local alignment, the computational algorithm searches for the most similar segments in the sequences according to the scoring function. The choice of this scoring function is important for correctly detecting segments of interest. We formulate sequence alignment as a hypothesis testing problem, and conduct extensive simulation experiments to study the relationship between the scoring function and the distribution of aligned pairs within the aligned segment under this framework. We cut through the many ways to construct scoring functions and showed that any scoring function with negative expectation used in local alignment corresponds to a hypothesis test between the background distribution of sequence letters and a statistical distribution of letter pairs determined by the scoring function. The results indicate that the log-likelihood ratio scoring function is statistically most powerful and has the highest accuracy for detecting the segments of interest that are defined by the statistical distribution of aligned letter pairs. • Anthropogenic nitrogen (N) addition may substantially alter the terrestrial N cycle. However, a comprehensive understanding of how the ecosystem N cycle responds to external N input remains elusive. • Here, we evaluated the central tendencies of the responses of 15 variables associated with the ecosystem N cycle to N addition, using data extracted from 206 peer-reviewed papers. • Our results showed that the largest changes in the ecosystem N cycle caused by N addition were increases in soil inorganic N leaching (461%), soil NO3- concentration (429%), nitrification (154%), nitrous oxide emission (134%), and denitrification (84%). N addition also substantially increased soil NH4+ concentration (47%), and the N content in belowground (53%) and aboveground (44%) plant pools, leaves (24%), litter (24%) and dissolved organic N (21%). Total N content in the organic horizon (6.1%) and mineral soil (6.2%) slightly increased in response to N addition. However, N addition induced a decrease in microbial biomass N by 5.8%. • The increases in N effluxes caused by N addition were much greater than those in plant and soil pools except soil NO3-, suggesting a leaky terrestrial N system. Background: Monocyte recruited into the tumor and maturation to tumor-associated macrophage (TAM). Interleukin-10(IL-10) is a potent immunosuppressive cytokine, which can be secreted from both primary tumor and stromal cells. However, there are controversies regarding its role in the progression of cancer. So it is important to isolate TAM from tumor cells to study the role of IL-10 in the progress of cancer. The aim of our study was to determine whether IL-10 expressed by TAM correlated with clinicopathological factors in NSCLC. Methods: TAM in NSCLC was isolated by short-term culture in serum free medium with the modification to literature reports. The mRNA expression levels of IL-10, cathepsin B, cathepsin S, which were closely related with TAM according to the literatures, were evaluated by Quantitative real-time RT-PCR in 63 NSCLC. The relationships between their expression levels and clinicopathological features were investigated. Results: We successfully achieved up to 95% purity of TAM, derived from 63 primary lung cancer tissues. TAM expressed high levels of IL-10, cathepsin B in NSCLC. High levels of IL-10 in TAM significantly correlated with stage, tumor size, lymph node metastasis, lymphovascular invasion or histologic poor differentiation. Conclusions: Our results revealed that TAM with high levels of IL-10 expression may play an important role in the progression of non-small cell lung cancer. The data also suggested that TAMs may involve in tumor immunosuppression through overexpressed IL-10. Background: Gallbladder cancer (GBC) is the most common and lethal malignancy of the biliary tract that lacks effective therapy. In many GBC cases, infiltration into adjacent organs or distant metastasis happened long before the diagnosis, especially the direct liver invasion, which is the most common and unfavorable way of spreading. Methods: Single-cell RNA sequencing (scRNA-seq), spatial transcriptomics (ST), proteomics, and multiplexed immunohistochemistry (mIHC) were performed on GBC across multiple tumor stages to characterize the tumor microenvironment (TME), focusing specifically on the preferential enrichment of neutrophils in GBC liver invasion (GBC-LI). Results: Multi-model Analysis reveals the immunosuppressive TME of GBC-LI that was characterized by the enrichment of neutrophils at the invasive front. We identified the context-dependent transcriptional states of neutrophils, with the Tumor-Modifying state being associated with oxidized low-density lipoprotein (oxLDL) metabolism. In vitro assays showed that the direct cell-cell contact between GBC cells and neutrophils led to the drastic increase in oxLDL uptake of neutrophils, which was primarily mediated by the elevated OLR1 on neutrophils. The oxLDL-absorbing neutrophils displayed a higher potential to promote tumor invasion while demonstrating lower cancer cytotoxicity. Finally, we identified a neutrophil-promoting niche at the invasive front of GBC-LI that constituted of KRT17+ GBC cells, neutrophils, and surrounding fibroblasts, which may help cultivate the oxLDL-absorbing neutrophils. Conclusions: Our study reveals the existence of a subset of pro-tumoral neutrophils with a unique ability to absorb oxLDL via OLR1, a phenomenon induced through cell-cell contact with KRT17+ GBC cells in GBC-LI. To remedy the contradiction between the equal strain assumption and the flow continuity assumption at a soil–column interface, the traditional flow continuity assumption was abandoned and the radial and vertical flows within a stone column were incorporated to consider the column consolidation and deformation in a coupled fashion. Moreover, two possible variation patterns of the horizontal permeability coefficient of soil within the disturbed soil zone were included, to reflect the detrimental influence on the surrounding soil due to column construction. In addition, a linearly changed total vertical stress along the column depth was assumed, to achieve a more realistic stress distribution in practice. By considering the above mentioned characteristics, the governing equations for this type of consolidation problem were developed. The solutions for the governing equations were subsequently derived by using a new initial condition obtained from the equilibrium condition and equal strain assumption. On the basis of this, the average degree of consolidation of the composite foundation was obtained and discussed.
