In present research, a potent fungal strain was isolated from paper mill effluent (black liquor) in order to investigate its potential for the biodegradation of lignin. Two step strategy was used to screen most efficient fungal strain having ability to growin MSM-black liquor medium and to degrade alkali lignin.The results of initial screening indicated that the strain M-2 produced comparatively higher ligninolytic zone on MSN agar plates supplemented with black liquor (BL) and alkali ligninase compared to the other isolates.The results of 18S rRNA gene sequencing revealed that strain M-2 showed ≥ 99% sequence homology with Dipodasceus australiansis.The process for the biodegradation of lignin was optimized using Taguchi Orthogonal Array design. Under optimized conditions of pH 9, 40 °C and 4% inoculum, a maximum of 89% lignin was degraded with 41% color reduction after 8 days of incubation period by Dipodasceus australiansis M-2. The pH and temperature were found to be significant terms with the p-values of 0.002 and 0.001 respectively. The laccase activity of the Dipodascus australiensis was found to be maximum of 1.511 U/mL. The HPLC analysis of lignin biodegradation indicated sharp transformation of peaks as compared to the control. Our results suggested that the strain Dipodascus australiensis M-2 possess excellent lignin degradation and color reduction capability and can be applied in waste treatment systems for pulp and paper mill effluent. In present work we are reporting first hand information regarding biodegradation of lignin by a potent strain of Dipodascus australiensis and statistical optimization of the bioprocess.

We investigated the properties of the low molecular weight thermo-alkali-stable and mercury ion-tolerant xylanase production from Thermomyces dupontii KKU-CLD-E2-3. The xylanase was purified to homogeneity by ammonium sulfate, Sephadex G–100 and DEAE–cellulose column chromatography which resulted 27.92-fold purification specific activity of 56.19 U/mg protein and a recovery yield of 2.01%. The purified xylanase showed a molecular weight of 25 kDa by SDS–PAGE and the partial peptide sequence showed maximum sequence homology to the endo-1,4-β-xylanase. The optimum temperature and pH for its activity were 80 °C and pH 9.0, respectively. Furthermore, the purified xylanase can maintain more than 75% of the original activity in pH range of 7.0–10.0 after incubation at 4 °C for 24 h, and can still maintain more than 70% of original activity after incubating at 70 °C for 90 min. Our purified xylanase was activated by Cu2+ and Hg2+ up to 277% and 235% of initial activity, respectively but inhibited by Co2+, Ag+ and SDS at a concentration of 5 mM. The Km and Vmax values of beechwood xylan were 3.38 mg/mL and 625 µmol/min/mg, respectively. Furthermore, our xylanase had activity specifically to xylan-containing substrates and hydrolyzed beechwood xylan, and the end products mainly were xylotetraose and xylobiose. The results suggested that our purified xylanase has potential to use for pulp bleaching in the pulp and paper industry.

A potential bacterium Bacillus flexus RMWW II has been isolated from rice mill effluent, and examined for its decolorizing potential for lignin-mimicking dyes. The biodegradation of alkali lignin by the rod-shaped, Gram-positive, oxidase and catalase-positive Bacillus flexus RMWW II bacteria is due to its uptake of lignin as the sole carbon source. The lignin degradation was 100% at a lignin concentration of 50 mg L−1 but the degradation reduced to 20% at 400 mg L−1. The bacterial-mediated biodegradation of alkali lignin was suitably explained by the Edward kinetics model with a maximal specific biodegradation rate (qmax) of 0.056 h−1 and true specific biodegradation rate (q*) of 0.042 h−1. The non-toxic nature of the metabolites of alkali lignin after bacterial degradation was illustrated by phytotoxicity studies. This bacterium was utilized to treat complex rice mill wastewater, as lignin is one of the major components of the effluent. A considerable reduction of 84% of chemical oxygen demand (COD) was observed in a batch reactor in 70 h of operation. The bacterial treatment results for the actual rice mill effluent indicate that Bacillus flexus RMWW II could be a promising agent for microbial remediation of lignin-laden raw rice mill wastewater.

Electron beam irradiation (EBI) is an alternative treatment for intrinsic viscosity (IV) control in cellulose pulps, but has never been integrated in full bleaching sequences for comparison to conventional methods. Both eucalyptus kraft (EK) paper pulp and beech sulfite (BS) dissolving pulp were subjected to totally chlorine free (TCF) bleaching sequences comprising either EBI, ozone, or both for IV control. Additionally, effects of EBI on hexenuronic acid (HexA) and xylan were investigated. IV was adjusted to 450–500 mL g−1 and properties including carbonyl content, kappa, brightness, alkali-resistance and sugar composition were compared. Pulps produced with EBI had a higher alkali-resistance, uniformity and less cellulose oxidation. However, the degree of bleaching (DoB) was low without the use of ozone. HexA content in a birch pulp was halved by EBI. Isolated xylans were more resistant to irradiation than cellulose with little decrease of molar masses and moderate oxidation.

Cellulase (mainly endoglucanase, EG) has been used in pulp modification for improving paper quality through environmentally friendly process. But low activity in alkaline pH and high filter paper activity (FPA) were still obstacles for extending the cellulase application in papermaking industry. In the study, an alkali-tolerant EG gene of Bacillus subtilis Y106 was homologous over-expressed for obtaining suitable enzyme used in pulp modification. The engineering strain could produce the crude enzyme with more alkali-tolerant EG and little PFA. Potential of the crude enzyme in modification of different pulps were investigated. It was found that the enzyme could be used for improving drainage and strength properties of pulps from softwood, hardwood and non-wood materials, especially non-wood pulp such as wheat straw pulp. The underlying mechanisms of pulp modification and different effects on various types of pulps by the EG treatment were also discussed by studying the change in fibers characteristics and fiber bonding.

The present contribution evaluates the methods of degradation and stabilization of alum-containing paper with a focus on the alkaline environment achieved by deacidification procedures. In terms of reviewed subjects, the contribution focuses on alum-rosin sized paper, which is still used as a carrier of knowledge and information; however, it also mentions cellulose itself and other brands of paper. The contribution summarizes the results on the homogeneity of the distribution of alum and rosin in the paper mass and on the paper surface. It provides the knowledge gained in the field of alkaline hydrolysis and oxidation with special regard to transition metal species. It shows the values of alkaline reserves achieved in the main mass-deacidification processes. On the basis of the acquired knowledge, the contribution emphasizes the procedures of paper stabilization. Criteria of “increased mechanical permanence and lifetime prolongation” adopted to evaluate and compare the efficacy of individual mass-deacidification processes were applied and corresponding data are introduced. The contribution also draws attention to the existence of open issues in the area of paper degradation and stabilization.

Bacteriophages find applications in agriculture, medicine, and food safety. Many of these applications can expose bacteriophages to stresses that inactivate them including acidic and basic pH. Bacteriophages can be stabilized against these stresses by materials including paper, a common material in packaging and consumer products. Combining paper and bacteriophages creates antibacterial materials, which can reduce the use of antibiotics. Here we show that adsorption on paper protects T4, T5, and T7 bacteriophage from acidic and basic pH. We added bacteriophages to filter paper functionalized with carboxylic acid (carboxyl methyl cellulose) or amine (chitosan) groups, and exposed them to pH from 5.6 to 14. We determined the number of infective bacteriophages after exposure directly on the paper. All papers extended the lifetime of infective bacteriophage by at least a factor of four with some papers stabilizing bacteriophages for up to one week. The degree of stabilization depended on five main factors (i) the family of the bacteriophage, (ii) the charge of the paper and bacteriophages, (iii) the location of the bacteriophages within the paper, (iv) the ability of the paper to prevent bacteriophage-bacteriophage aggregation, and (v) the sensitivity of the bacteriophage proteins to the tested pH. Even when adsorbed on paper the bacteriophages were able to remove E. coli in milk. Choosing the right paper modification or material will protect bacteriophages adsorbed on that material against detrimental pH and other environmental challenges increasing the range of applications of bacteriophages on materials.

Wood shortages, environmental pollution and high energy consumption remain major obstacles hindering the development of today’s pulp and paper industry. Energy-saving and environmental friendly pulping processes are still needed, especially for non-woody materials. In this study, soda-oxygen pulping of bagasse was investigated and a successful soda-oxygen pulping process for bagasse at 100 °C was established. The pulping parameters of choice were under active alkali charge of 23%, maximum cooking temperature 100 °C, time hold at maximum temperature 180 min, initial pressure of oxygen 0.6 MPa, MgSO4 charge 0.5%, and de-pithed bagasse consistency 12%. Properties of the resultant pulp were screened yield 60.9%, Kappa number 14, viscosity 766 dm3/kg, and brightness 63.7% ISO. Similar pulps were also obtained at 110 °C or 105 °C with a cooking time of 90 min. Compared with pulps obtained at higher temperatures (115–125 °C), this pulp had higher screened yield, brightness, and acceptable viscosity, while the delignification degree was moderate. These results indicated that soda-oxygen pulping at 100 °C, the lowest cooking temperature reported so far for soda-oxygen pulping, is a suitable process for making chemical pulp from bagasse. Pulping at lower temperature and using oxygen make it an environmental friendly and energy-saving pulping process.

Some Chilean copper slag dumps from the nineteenth century still remain, without a proposed use that encourages recycling and reduces environmental impact. In this paper, the copper slag abandoned in landfills is proposed as a new building material. The slags studied were taken from Playa Negra and Puquios dumps, both located in the region of Atacama in northern Chile. Pozzolanic activity in lime and Portland cement systems, as well as the alkali activation in pastes with copper slag cured at different temperatures, was studied. The reactivity of the slag was measured using thermogravimetric analysis (TGA), scanning electron microscopy (SEM), X-ray diffraction (XRD), electrical conductivity and pH in aqueous suspension and Fourier Transform Infrared Spectroscopy (FTIR). Furthermore, copper slag-Portland cement mortars with the substitution of 25% (by weight) of cement by copper slag and alkali-activated slag mortars cured at 20 and 65 °C were made, to determine the compressive strength. The results indicate that the ancient copper slags studied have interesting binding properties for the construction sector.

Paper mill sludge (PS), a solid waste from pulp and paper industry, was investigated as a feedstock for acetone-butanol-ethanol (ABE) production by simultaneous saccharification and fermentation (SSF). ABE fermentation of paper sludge by Clostridium acetobutylicum required partial removal of ash in PS to enhance its enzymatic digestibility. Enzymatic hydrolysis was found to be a rate-limiting step in the SSF. A total of 16.4-18.0g/L of ABE solvents were produced in the SSF of de-ashed PS with solid loading of 6.3-7.4% and enzyme loading of 10-15FPU/g-glucan, and the final solvent yield reached 0.27g/g sugars. No pretreatment and pH control were needed in ABE fermentation of paper sludge, which makes it an attractive feedstock for butanol production. The results suggested utilization of paper sludge should not only consider the benefits of buffering effect of CaCO3 in fermentation, but also take into account its inhibitory effect on enzymatic hydrolysis.

Alkaline xylanases from alkaliphilic Bacillus strains NCL (87-6-10) and Sam III were compared with the commercial xylanases Pulpzyme HC and Biopulp for their compatibility with detergents and proteases for laundry applications. Among the four xylanases evaluated, the enzyme from the alkaliphilic Bacillus strain NCL (87-6-10) was the most compatible. The enzyme retained its full activity (40 degrees C for 1 h) in the presence of detergents, whereas Pulpzyme HC and Sam III showed only 30% and 50% of their initial activity, respectively. Biopulp, though stable to detergents, had only marginal activity (5%)at pH 10. However, all four enzymes retained significant activity (80%) for 60 min in the presence of the proteases Alcalase and Conidiobolus protease. Supplementation of the enzyme enhanced the cleaning ability of the detergents.

Rice straw is a widely available lignocellulosic waste with potential for energy recovery through anaerobic digestion. Lignin slows the hydrolysis phase, resulting in low methane recovery and long digestion periods. Although pretreatment is effective, it often requires high energy inputs or chemicals that are not feasible for farm-scale systems. This study investigates a unique co-digestion strategy to improve methane yields and reduce digestion times for farm-scale systems.By adding both piggery wastewater and paper mill sludge, specific methane yields in laboratory-scale digesters reached the theoretical value for rice straw (i.e. 330LNCH4/kgVS) over the 92-day period. Accelerated hydrolysis of the straw was directly related to the quantity of sludge added. The most stable digester, with sufficient buffering capacity and nutrients, contained equal parts of straw, wastewater and sludge. This approach is feasible for farm-scale applications since it requires no additional energy inputs or changes to existing infrastructure for dry systems. © 2013 Elsevier Ltd.

A commercial variety of Leucaena leucocephala K360 was used for pulp production and papermaking employing the soda-anthraquinone process. Also, the chemical and energy contents of the resultant black liquors were determined to simultaneously optimize: pulp and paper production and energy generation. A process temperature of (185 °C), an operating time of (120 min) and an active alkali concentration of (21%) provided sheets of paper with good strength (tensile index of 12.12 N m/g, burst index of 0.38 kPa m 2 /g, tear index of 1.29 mN m 2 /g and a Kappa number of 20.5) and black liquor with a greater calorific value (14.1 MJ/kg) than that obtained with higher active alkali concentrations. However, reducing the active alkali concentration to a level in the low operation range led to less marked degradation of cellulose and allowed paper sheets with good properties to be obtained and energy to be optimally produced from the black liquor.

International audience; AMDES (aminopropylmethyldiethoxysilane) was used to investigate the physicochemical and chemical events related to the introduction of aminoalkylalkoxysilanes in cellulosic materials. Using 29Si CP-MAS and 1H NMR to study the reactivity and structural modification of AMDES in the paper it was shown that polymerization occurs in situ. The distribution of the active compound on the surface of the fibers and throughout the fibers’ thickness was visualized by SEM-EDS. A relation between moisture content, fiber swelling, and uptake of AMDES was found. To better represent old and brittle documents, the paper was predegraded by oxidation with sodium hypochlorite. XRD confirmed the advanced destruction of the amorphous areas of cellulose. Adding AMDES in the oxidized paper resulted in improved mechanical properties, a roughly unmodified degree of polymerization of cellulose, but a slight increase in the yellowing, probably due to several possible reaction products such as imines, amine, amides, and Maillard reactions products. The deacidification efficacy was established and the strengthening effect was shown to arise from the interaction of AMDES with the cellulose fibers.

The aim of this work was to evaluate the response to biobleaching of steam exploded kraft pulps and to compare the results with the controls. For this end, a laccase-mediator treatment using commercial laccase (Novozyme 51003) and a natural mediator (acetosyringone) were assayed, followed by alkaline extraction and hydrogen peroxide stages. Our approach resulted in exploded biobleached pulps with lower kappa number and improved optical properties compared to controls, even after subjecting pulps to accelerated ageing. Additionally, use of hydrogen peroxide was reduced. The LMS (laccase-mediator system) had a smaller impact on the properties of the bleached pulps and on hydrogen peroxide consumption than the steam explosion process did. © 2010 Elsevier Ltd.

The effect of hot-water extraction on alkaline pulping was investigated. The properties of black liquor and pulp strength of bagasse were analyzed. The extraction was conducted at 160 degrees C for 30min where 13.2% of the mass was dissolved in the extraction liquor. Untreated bagasse and extracted bagasse were digested by soda and soda-AQ processes at 17% and 15.5% (with 0.1% AQ) alkali charge (NaOH). Cooking temperatures were 160 degrees C and 155 degrees C respectively. The pulp from extracted bagasse had a lower Kappa number and a higher viscosity compared to the pulp from the untreated bagasse. The black liquor from pulping extracted bagasse had a lower solid content, a lower viscosity and a lower silica content, but a higher heating value than that from pulping of untreated bagasse. Hot-water extraction resulted in a significant decrease in bleaching chemical consumption and the formation of chlorinated organics. Pulp strength properties such as the tensile index and the burst index were found to be lower, but the tear index, bulk, opacity and pulp freeness were found to be higher when hot-water extraction was applied.

Anaerobic bioconversion of newsprint and waste office paper was performed in bench-scale reactors with three inocula sources: landfill, rumen, and anaerobic digester. Office paper bioconversion was nearly complete within 20 days but continued for about 165 days with methane yield efficiencies ranging from 71-85% of potential chemical oxygen demand (COD) conversion. Average newsprint methane conversion efficiencies ranged from 32-41% of total COD under strictly anaerobic conditions for 300 days. Mass balance calculations revealed that more than 80% of newsprint cellulose was biodegraded. The apparent limiting factor for anaerobic bioconversion of newsprint was the physical association between lignin and cellulose. After proper acclimation, the three inocula tested equally well for methane production under strictly anaerobic conditions. Testing of ground, shredded strips, and whole paper pieces showed no effects of feedstock size on bioconversion rate or extent. Alkali pretreatment with NaOH concentration up to 10% significantly improved newsprint biodegradability. Treatment for longer duration or at elevated temperatures increased the solubilization of lignin, but did not improve bioconversion of newsprint to methane. Neutralizing treated samples with carbon dioxide gave higher methane yields compared to sulfuric or hydrochloric acids, suggesting that digester neutralization could be combined with biogas scrubbing.

Central corneal alkali burn injuries were induced in rabbits by applying five NaOH concentrations on uniformly soaked 7 mm filter paper discs. Clinical parameters were evaluated daily by microscopic examination and photography, and corneal myeloperoxidase levels were measured periodically. Satisfactory alkali burn models of corneal inflammation, vascularization and ulceration were developed by manipulating the alkali concentration.
