Background: Bellamya bengalensisis an important bioresource of the freshwater ecosystem of India. This edible mollusc is filter feeder, indigenous diet of human, poultry and fishery. Natural habitat of this freshwater gastropod mollusc bears the risk of contamination by agricultural pesticide fenvalerate, a synthetic type II pyrethroid. Hemocytes are the immuno effector cells of molluscs which are affected by environmental toxins or pathogenic microorganisms. Hemocytes perform various types of immunological functions such as phagocytosis, cytotoxic response etc. Results: Experimental exposure of fenvalerate resulted in impairment of phagocytic efficacy and alteration in the generation of phenoloxidase in B. bengalensis. Conclusion: The alteration of phagocytic response and generation of phenoloxidase of hemocytes, exposed to experimental concentrations of fenvalerate under static laboratory condition have been determined to establish as biomarker of aquatic toxicity in toxin contaminated natural habitat. Background: This study aims to analyze the work done on leukemia detection and diagnosis using machine learning, deep learning and different image processing techniques from 2011 to 2020, using the bibliometric methods. Methods: different articles on leukemia detection were retrieved using one of the most popular database-Scopus. The research articles are considered between 2011 to 2020. Scopus analyzer is used for getting some analysis results such as documents by year, source, county and so on. VOSviewer Version 1.6.15 is used for the analysis of different units such as coauthorship, co-occurrences, citation analysis etc. Results: In our study, a database search outputs a total of 617 articles on leukemia detection from 2011 to 2020. Statistical analysis and network analysis shows the maximum articles are published in the years 2019 and 2020 with India contributed the largest number of documents. Network analysis of different parameters shows a good potential of the topic in terms of research. Conclusions: Scoups keyword search outcome has 617 articles with English language having the largest number. Authors, documents, country, affiliation etc are statically analyzed and indicates the potential of the topic. Network analysis of different parameters indicates that, there is a lot of scope to contribute in the further research in terms of advanced algorithms of computer vision, deep learning and machine learning. The textile industries play a major role in the economy of India. In addition to this, they exert acute and/or chronic effects on environment by discharge of effluent in the environment. This study was designed to assess the cyto genotoxicity of Reactive Red 120 (RR120) on freshwater fish Catla catla, using DNA damage in gill cells and blood cells as sensitive biomarkers. For this, fingerlings were exposed to three sublethal concentrations of RR120 (0.35, 0.7, 3.5 mg L-1). Samples (gills and peripheral blood) were collected and analyzed at fixed intervals (10, 20 and 30 days) and were compared with those of the control. Some nuclear changes were observed, such as micronuclei (MN), nuclear buds (NB), fragmented-apoptotic (FA) and bi-nucleated (BN) cells in both peripheral blood cells and gill cells. All the abnormalities showed dose- and time-dependent increase and were higher in the gill cells. The results clearly revealed the most significant change for MN, followed by FA, BN and NB in both samples. Hence, RR120 has a potential to damage DNA in cells and exerts genotoxic impact on fish. These findings also suggest the possible toxicity of RR120 to humans by direct or indirect exposure. [ABSTRACT FROM AUTHOR]

Effective vaccine design relies on accurate knowledge of protection against a pathogen, so as to be able to induce relevant and effective protective responses against it. An ideal Human Immunodeficiency virus (HIV) vaccine should induce humoral as well as cellular immune responses to prevent initial infection of host cells or limit early events of viral dissemination. A Phase I HIV-1 prophylactic vaccine trial sponsored by the International AIDS Vaccine Initiative (IAVI) was conducted in India in 2009.The trial tested a HIV-1 subtype C vaccine in a prime-boost regimen, comprising of a DNA prime (ADVAX) and Modified Vaccine Ankara (MVA) (TBC-M4) boost. The trial reported that the vaccine regimen was safe, well tolerated, and resulted in enhancement of HIV-specific immune responses. However, preliminary immunological studies were limited to vaccine-induced IFN-γ responses against the Env and Gag peptides. The present study is a retrospective study to characterize in detail the nature of the vaccine-induced cell mediated immune responses among volunteers, using Peripheral Blood Mononuclear Cells (PBMC) that were archived during the trial. ELISpot was used to measure IFN-γ responses and polyfunctional T cells were analyzed by intracellular multicolor flow cytometry. It was observed that DNA priming and MVA boosting induced Env and Gag specific bi-functional and multi-functional CD4+ and CD8+ T cells expressing IFN-γ, TNF-α and IL-2. The heterologous prime-boost regimen appeared to be slightly superior to the homologous prime-boost regimen in inducing favorable cell mediated immune responses. These results suggest that an in-depth analysis of vaccine-induced cellular immune response can aid in the identification of correlates of an effective immunogenic response, and inform future design of HIV vaccines. Background: QuantiFERON-TB Gold In-Tube (QFT) is an IFNγ-release assay used in the diagnosis of Mycobacterium tuberculosis (MTB) infection. The risk of TB progression increases with the magnitude of the MTB-specific IFNγ-response. QFT reversion, also associated with low Tuberculin Skin Test responses, may therefore represent a transient immune response with control of M. tuberculosis infection. However, studies at the single cell level have suggested that the quality (polyfunctionality) of the T-cell response is more important than the quantity of cytokines produced. Objective: To explore the quality and/or magnitude of mycobacteria-specific T-cell responses associated with QFT reversion and persistent QFT-positivity. Methods: Multi-color flowcytometry on prospectively collected peripheral blood mononuclear cells was applied to assess mycobacteria-specific T-cell responses in 42 QFT positive Indian adolescents of whom 21 became QFT negative (reverters) within one year. Ten QFT consistent negatives were also included as controls. Results: There was no difference in the qualitative PPD-specific CD4+ T-cell response between QFT consistent positives and reverters. However, compared with QFT consistent positives, reverters displayed lower absolute frequencies of polyfunctional (IFNγ+IL2+TNFα+) CD4+ T-cells at baseline, which were further reduced to the point where they were not different to QFT negative controls one year later. Moreover, absolute frequencies of these cells correlated well with the magnitude of the QFT-response. Conclusion: Whereas specific polyfunctional CD4+ T-cells have been suggested to protect against TB progression, our data do not support that higher relative or absolute frequencies of PPD-specific polyfunctional CD4+ T-cells in peripheral blood can explain the reduced risk of TB progression observed in QFT reverters. On the contrary, absolute frequencies of these cells correlated with the QFT-response, suggesting that this readout reflects antigenic load. [ABSTRACT FROM AUTHOR]
