Background: Characterization of early breast cancer circulating tumor cells (CTCs) may provide valuable information on tumor metastasis. Methods: We used immunomagnetic nanospheres to capture CTCs from the peripheral blood of eight early breast cancer patients and then performed single‐cell RNA sequencing using our proposed bead‐dd‐seq method. Results: CTCs displayed obvious tumor cell characteristics, such as the activation of oxidative stress, proliferation, and promotion of metastasis. CTCs were clustered into two subtypes significantly correlated with the lymph node metastasis status of patients. CTCs in subtype 1 showed a strong metastatic ability because these CTCs have the phenotype of partial epithelial‐mesenchymal transition and enriched transcripts, indicating breast cancer responsiveness and proliferation. Furthermore, DNA damage repair pathways were significantly upregulated in subtype 1. We performed in vitro and in vivo investigations, and found that cellular oxidative stress and further DNA damage existed in CTCs. The activated DNA damage repair pathway in CTCs favors resistance to cisplatin. A checkpoint kinase 1 inhibitor sensitized CTCs to cisplatin in mouse models of breast cancer metastasis. Conclusion: The present study dissects the molecular characteristics of CTCs from early‐stage breast cancer, providing novel insight into the understanding of CTC behavior in breast cancer metastasis. Background: Gastrin is an important gastrointestinal hormone produced primarily by G-cells in the antrum of the stomach. It normally regulates gastric acid secretion and is implicated in a number of human disease states, but how its function affects breast cancer (BC) development is not documented. The current study investigated the suppressive effects of gastrin on BC and its underlying mechanisms.Methods: Serum levels of gastrin were measured by enzyme-linked immunosorbent assay (ELISA) and correlation between gastrin level and development of BC was analyzed by chi-square test. Inhibitory effects of gastrin on BC were investigated by CCK-8 assay and nude mice models. Expressions of CCKBR/ERK/P65 in BC patients were determined through immunohistochemistry (IHC) and Western blot. Survival analysis was performed using the log-rank test.Results: The results indicated that the serum level of gastrin in BC patients was lower compared with normal control. Cellular and molecular experiments indicated that reduction of gastrin is associated with inactivation of cholecystokinin B receptor (CCKBR)/ERK/P65 signaling in BC cells which is corresponding to molecular type of estrogen receptor (ER) positive BC. Furthermore, we found that low expression of gastrin/CCKBR/ERK /P65 was correlated to worse prognosis in BC patients. Gastrin or ERK/P65 activators inhibited ER+ BC through CCKBR-mediated activation of ERK/P65. Moreover, combination treatment with gastrin and tamoxifen more efficiently inhibited ER+ BC than tamoxifen alone.Conclusions: We concluded that low serum gastrin is related to increased risk of ER+ BC development. The results also established that CCKBR/ERK/P65 signaling function is generally tumor suppressive in ER+ BC, indicating therapies should focus on restoring, not inhibiting, CCKBR/ERK/P65 pathway activity. Objective To explore the arousal of people with high stress-related sleep vulnerability and the effect of music intervention on hyperarousal. Methods A total of 33 healthy subjects from China Rehabilitation Research Center were enrolled in this study. Based on the median value (17 score) of Ford Insomnia Response to Stress Test-Chinese Version (FIRST-C), they were divided into 2 groups: 16 cases of high stress-related sleep vulnerability and 17 cases of low stress - related sleep vulnerability. Multi-parameter physiological indexes monitoring system of Thought Technology Ltd. was used to gather physiological indicators [blood volume pulse (BVP), skin temperature, skin conduction, respiration rate, respiration amplitude and heart rate], and brain wave [δ wave, θ wave, low α wave and high α wave, α wave, sensory-motor rhythm (SMR), low β wave and high β wave]. Results Compared with low vulnerability subjects, high vulnerability subjects had significantly higher skin conduction (P = 0.003, 0.001), amplitude of SMR (P = 0.015, 0.031) and low β wave (P = 0.000, 0.001) before and after treatment, while had significantly increased high β wave after treatment (P = 0.004). After treatment, all subjects had significantly reduced BVP (P = 0.000), increased skin temperature (P = 0.000), increased respiration rate (P = 0.008) and reduced heart rate (P = 0.000). Compared with before treatment, high vulnerability subjects had significantly reduced skin conduction (P = 0.001), respiratory amplitude (P = 0.032), high α wave (P = 0.017) and low β wave (P = 0.013) after treatment. Compared with before treatment, low vulnerability subjects had significantly reduced skin conduction (P = 0.039), low α wave (P = 0.035), high α wave (P = 0.031), α wave (P = 0.044) and low β wave (P = 0.015) after treatment. Conclusions Both physiological and cortical hyperarousal are presented in people with high stress-related sleep vulnerability in resting state. Music intervention can improve the hyperarousal of high vulnerability people, especially their physiological indicators. It can be used as a health management technique to prevent people with high stress-related sleep vulnerability from developing into chronic insomnia. [ABSTRACT FROM AUTHOR]

It has been shown that inflammation and oxidative stress are important factors in postoperative atrial fibrillation (POAF). Angiotensin converting enzyme (ACE) and apelin have a close relationship with inflammation and oxidative stress. The effect of ACE and apelin on POAF after off-pump coronary artery bypass grafting (OPCABG) remains a question. The concentrations of serum ACE, angiotensin II (Ang II), apelin, bradykinin (BK), malondialdehyde (MDA), and C reactive protein (CRP) were measured in the perioperative period of OPCABG. The levels of serum ACE in the POAF group were higher than in the no POAF group both preoperatively and postoperatively. Apelin in the POAF group was lower than in the no POAF group. There was a correlation between serum ACE and apelin. Postoperatively, CRP and MDA in the POAF group were higher than in the no POAF group; however, there was no difference before the operation. Preoperative ACE and apelin were both significant and independent risk factors for POAF. In conclusion, the high ACE and low apelin preoperatively led to CRP and MDA being increased postoperatively, which was probably associated with POAF after OPCABG. α-Amylase as an important industrial enzyme has been widely used in starch processing, detergent, and paper industries. To improve expression efficiency of recombinant α-amylase from Bacillus licheniformis (B. licheniformis), the α-amylase gene from B. licheniformis was optimized according to the codon usage of Pichia pastoris (P. pastoris) and expressed in P. pastoris. Totally, the codons encoding 305 amino acids were optimized in which a total of 328 nucleotides were changed and the G+C content was increased from 47.6 to 49.2%. The recombinants were cultured in 96-deep-well microplates and screened by a new plate assay method. Compared with the wild-type gene, the optimized gene is expressed at a significantly higher level in P. pastoris after methanol induction for 168 h in 5- and 50-L bioreactor with the maximum activity of 8100 and 11000 U/mL, which was 2.31- and 2.62-fold higher than that by wild-type gene. The improved expression level makes the enzyme a good candidate for α-amylase production in industrial use. [ABSTRACT FROM AUTHOR]
