Abstract Background The aim of this study was to identify the ferroptosis induced tumor microenvironment (FeME) landscape in bladder cancer (BCa) for mRNA vaccine development and selecting suitable patients for precision treatment. Methods Gene expression profiles and clinical information of 1216 BCa patients were extracted from TCGA-BLCA, three GEO databases and IMvigor210 cohort. We comprehensively established the FeME landscape of 1216 BCa samples based on 290 ferroptosis related genes (FRGs), and systematically correlated these regulation patterns with TME cell-infiltrating characteristics. Besides, we identified the patients’ ferroptosis risk index (FRI) to predict the prognosis of BCa for precise treatment. Results Six over-expressed and mutated tumor antigens associated with poor prognosis and infiltration of antigen presenting cells were identified in BCa. Furthermore, we demonstrated the evaluation of FeME within individual tumors could predict stages of tumor inflammation, subtypes, genetic variation, and patient prognosis. Then, 5-lncRNA signature was mined to produce the FRI. Low FRI was also linked to increased mutation load, better prognosis and enhanced response to anti-PD-L1 immunotherapy. Besides, an immunotherapy cohort confirmed patients with lower FRI demonstrated significant therapeutic advantages and clinical benefits. Conclusions TFRC, SCD, G6PD, FADS2, SQLE, and SLC3A2 are potent antigens for developing anti-BCa mRNA vaccine. Establishment of FRI will contribute to enhancing our cognition of TME infiltration characterization and guiding more effective immunotherapy strategies and selecting appropriate patients for tumor vaccine therapy.

Irritable bowel syndrome (IBS) is a gastrointestinal disorder with no structural damage and biochemical abnormality, and its pathogenic mechanism is still unclear. Variety of drugs, psychotherapy, diet regulation and other methods have been used by modern medicine to relieve symptoms, but the effect is not satisfactory. In recent years, TCM treatment of IBS has attracted widespread attention, especially for constipation type and diarrhea type. Among the many experiments, most of the traditional Chinese medicine experiments aimed at the 5-HT signaling pathway have achieved good results. Therefore, this paper summarizes the main mechanisms and results of traditional Chinese medicine based on 5-HT in the treatment of IBS, in order to provide a reference for subsequent research.

Tourist satisfaction is an important reference in destination management decisions, but on-site validation is often lacking. By exploring the relationship between the current situation of recreational fishery and tourism satisfaction evaluation, we can accurately understand tourists’ consumption preferences and obtain product/service quality information. After on-site verification, such validation is conducive to promoting the high-quality development of regional recreational fisheries. The study area is located on Sandu Island, Ningde City, Fujian Province. This study examines how each item of service quality may exhibit different impacts on customer satisfaction by using a questionnaire survey of consumers in the recreational fisheries. This study combines the Kano model and the Better–Worse index method to investigate the critical service quality. The results show that among the 19 quality indicators of visitor satisfaction, environmental and landscape quality elements are the most important. In addition, through expert verification, it can be confirmed that the improvement of the water environment is of great help to the industry. To improve and maintain customer satisfaction, the above attributes are combined with underlying quality attributes as strategic weapons to highlight competitive advantages.

Obstructive sleep apnea-hypopnea syndrome (OSAHS) is a disease marked by apnea, hypopnea, decreased oxygen saturation, and disordered sleep structure, which is a major risk for cardiovascular disease. Recent studies have found that OSAHS patients have an increased risk of hypertension, coronary atherosclerotic heart disease, insulin resistance, type 2 diabetes, metabolic syndrome, non-alcoholic fatty liver disease, etc. And these patients have a high prevalence of obvious glucolipid metabolism disorder (GMD) , which plays an important role in cardiovascular morbidity and mortality in OSAHS. We reviewed the latest advances in the association of GMD and OSAHS, and the potential pathogenesis of OSAHS-induced GMD and insulin resistance, aiming at providing new ideas for clinical treatment of GMD in OSAHS.

The aim of this study was to explore the characteristics of the combined application of organic fertilizer and inorganic fertilizer using different tillage methods to delay the senescence of maize leaves. The yield and activities of GDH, CAT, APX, GR, and GSH enzymes in maize leaves were measured at different growth stages by using two tillage methods, three organic and inorganic combined applications (P1, P2, and P3), and four control treatments. (1) During the growth period, the R + S and R treatments were P1 treatments, with the highest enzyme activities noted for GDH, CAT, APX, GR, and GSH, which were 36.79–103.22% higher than those of CK. (2) The average yield of all R + S treatments was higher than that of R treatments, and the average yield of P1 treatment was the highest under R + S, which was 13,663.79 kg hm−2, which was 6.39%, 7.90%, and 14.67% higher than that of P2, P3, and CK, respectively, which was lower than that of R. The yield of P1 treatment was 2.53% higher. (3) There was a significant positive correlation between APX activity, CAT activity, GR activity, GDH activity, GSH activity, grain number per ear, ear length, and 100-grain weight of maize leaves at the grain filling stage, and a significant negative correlation between bald tip length and yield. The treatment details had the strongest enzyme activity and the highest yield when using the rotary tillage + subsoiling (R + S) P1 method, which was the most suitable tillage method and the best fertilizer ratio combination, which could be demonstrated and popularized in a large area in the dry farming area of spring maize in Shanxi Province. Species richness is the most basic concept of diversity and is crucial to biodiversity conservation and sustainable fisheries. To understand the fish species richness of the Minjiang Estuary and its adjacent waters, eight documents and surveyed data were collected and compared from 1990–2021. To obtain suitable analysis data, the content of the data was compared and evaluated. Explore the suitability of data based on several criteria. Among them, the bottom trawling survey carried out in 2006–2007, and non-parametric estimation methods such as Chao 2, Jackknife 1, Jackknife 2 and Bootstrap were used to estimate the fish species richness. The results of this case show that a total of 153 species of fish were caught in the trawling survey in the fourth quarter, belonging to 14 orders, 57 families and 101 genera. The 2006–2007 cruise is more complete for studying species richness. The Estimable expectations of fish species richness are: 250 (Chao 2), 204 (Jackknief 1), 241 (Jackknief 2) and 174 (Bootstrap). The number of fish species was significantly higher in summer and autumn than winter and spring. To manage fishery resources and sustainability in the sea area of Fujian Province, biological information and stock assessment are required. This meaningful information, especially for endemic and economically important species such as can set a baseline. Once species change exceeds the baseline range, it provides decision-making basis for marine biodiversity conservation and fisheries management.

Objective To explore the effect of miR-133a-3p on the apoptosis and inflammatory response of bronchial epithelial cells infected by respiratory syncytial virus (RSV) and its regulatory effect on CD2-associated protein (CD2AP). Methods The bronchial mucosal tissue specimens from 59 children with bronchial asthma were collected as the AsP group and 59 bronchial mucosal tissue specimens from non-asthmatic children with bronchiec- tasis were selected as the NC group. RT-qPCR method was used to detect the expression of miR-133a-3p and CD2AP in the tissues. RSV infected bronchial epithelial cells (16-HBE) were used to establish a cell injury model (RSV group). miR-NC, miR-133a-3p mimics, si-NC, si-CD2AP, pcDNA-NC, pcDNA-CD2AP, miR-133a-3p mimics and pcDNA-NC, miR-133a-3p mimics and pcDNA-CD2AP were respectively transferred into 16-HBE cells infected with RSV. RT-qPCR and Western blot were used to detect the expression of miR-133a-3p and CD2AP in the cells. ELISA was used to detect the level of TNF-α, IL-6, and IL-1α. Flow cytometry was used to detect apoptosis . The dual luciferase reporter experiment was used to detect the targeting relationship between miR-133a-3p and CD2AP. Western blot method was used to detect the protein expression of cleaved-caspase-3. Results Compared with NC group, the expression of miR-133a-3p in the bronchial mucosa tissue of the AsP group was decreased (P＜0.05), and the expression of CD2AP mRNA was increased (P＜0.05). The expression of miR-133a-3p in RSV-infected 16-HBE cells was decreased (P＜0.05), and the expression level of CD2AP, TNF-α, IL-6 and IL-1α was increased (P＜0.05), the apoptosis rate and the protein level of cleaved-caspase-3 were increased(P＜0.05). Transfection of miR-133a-3p mimics or transfection of si-CD2AP into RSV-infected 16-HBE cells significantly reduced the level of TNF-α, IL-6, IL-1α, the apoptosis rate and the protein level of cleaved-caspase-3 (P＜0.05). The dual luciferase report experiment confirmed that CD2AP was the target gene of miR-133a-3p. Co-transfection of miR-133a-3p mimics and pcDNA-CD2AP significantly reversed the effects of miR-133a-3p mimics transfection on 16-HBE cell apoptosis and inflammation. Conclusions The over-expression of miR-133a-3p inhibits the inflammatory response and apoptosis of RSV-infected bronchial epithelial cells by negatively regulating the expression of CD2AP and thereby reducing cell damage.

This paper understands the personality traits and cluster types of physical education (PE) teachers in elementary schools. A total of 393 elementary PE teachers volunteered to participate in this study. The Personality Trait Questionnaire for PE teachers in elementary school was summarized and compiled on the basis of the theory of the Big Five personality traits. The researchers used the discriminant analysis method to analyze and obtain the data. The results show that the Big Five personality traits of PE teachers in elementary schools was agreeableness. Classification of the Big Five personality traits through discriminant analysis revealed a Wilk’s λ value of .199, an eigenvalue of 3.254, and 94.7% of the variance explained. Use of Ward’s minimum variance method indicated that the participants’ personalities were characterized by steady and pioneering type, sensitive and cautious type, and moderate and peaceful type. There were more males than females in the steady and pioneering type, more steady and pioneering type under the age of 30 years old, and more moderate and peaceful type with more than 16 years of work experience. In conclusion, most the personality traits of PE teachers in elementary school were agreeableness, moderate and peaceful type, which shows that most PE teachers have calm emotions, appropriate expressions of emotions, regularity, optimism, and enjoy interacting and collaborating with others.

(1) Background: The accurate diagnosis of esophageal strictures is quite critical for optimizing medical intervention. However, the diagnosis of suspicious malignant esophageal strictures with intact mucosa appearance and negative biopsy results is challenging. This study aimed to evaluate the role of endoscopic ultrasound-guided fine needle aspiration (EUS-FNA) in the diagnosis of suspicious esophageal strictures. (2) Methods: We retrospectively analyzed the cases with suspicious malignant esophageal strictures that underwent EUS-FNA, with or without rapid on-site evaluation (ROSE), in our hospital from April 2017 to September 2022. Their clinical manifestations, imaging examinations, gastroscopic examinations, EUS-FNA results, and therapeutic strategies were retrospectively recorded and analyzed. (3) Results: A total of 23 patients (15 male and 8 female) were enrolled in this study. Based on EUS-FNA results, 18 patients were diagnosed with malignancies, including 16 cases of primary esophageal cancer (13 squamous carcinomas and 3 adenocarcinomas), 1 case of mediastinal cancer, and 1 case of metastatic esophageal cancer; 1 case of tuberculosis was also confirmed by EUS-FNA. Among 4 cases of ambiguous diagnosis with EUS-FNA, 1 was diagnosed with an esophageal glomus tumor after surgical removal, and 2 patients survived for several years without medical intervention, which hinted at the possibility of benign esophageal strictures. No major complications, including bleeding or perforation, were observed. (4) Conclusions: EUS-FNA may serve as a safe and effective diagnostic tool in suspicious malignant esophageal strictures with accurate specimen acquisition, especially for biopsy-negative cases.

Leber's hereditary optic neuropathy (LHON) is the most common mitochondrial disease. Mitochondrial modifiers are proposed to modify the phenotypic expression of primary LHON-associated mitochondrial DNA (mtDNA) mutations. In this study, we demonstrated that the LHON susceptibility allele (m.14502T > C, p. 58I > V) in the ND6 gene modulated the phenotypic expression of primary LHON-associated m.11778G > A mutation. Twenty-two Han Chinese pedigrees carrying m.14502T > C and m.11778G > A mutations exhibited significantly higher penetrance of optic neuropathy than those carrying only m.11778G > A mutation. We performed functional assays using the cybrid cell models, generated by fusing mtDNA-less ρo cells with enucleated cells from LHON patients carrying both m.11778G > A and m.14502T > C mutations, only m.14502T > C or m.11778G > A mutation and a control belonging to the same mtDNA haplogroup. These cybrids cell lines bearing m.14502T > C mutation exhibited mild effects on mitochondrial functions compared with those carrying only m.11778G > A mutation. However, more severe mitochondrial dysfunctions were observed in cell lines bearing both m.14502T > C and m.11778G > A mutations than those carrying only m.11778G > A or m.14502T > C mutation. In particular, the m.14502T > C mutation altered assemble of complex I, thereby aggravating the respiratory phenotypes associated with m.11778G > A mutation, resulted in a more defective complex I. Furthermore, more reductions in the levels of mitochondrial ATP and increasing production of reactive oxygen species were also observed in mutant cells bearing both m.14502T > C and m.11778G > A mutation than those carrying only 11778G > A mutation. Our findings provided new insights into the pathophysiology of LHON that were manifested by interaction between primary and secondary mtDNA mutations.

(1) Background: Circulating tumor DNA (ctDNA) has emerged as a promising biomarker for many kinds of tumors. However, whether ctDNA could be an accurate diagnostic biomarker in colorectal cancer (CRC) remains to be clarified. The aim of this study was to evaluate the diagnostic accuracy of ctDNA in CRC. (2) Methods: PubMed, Web of Science, and Cochrane databases were searched to identify studies reporting the use of ctDNA to screen and diagnose CRC, and all relevant studies published until October 2022 were enrolled for our analysis. These studies were divided into three primer subgroups: the subgroup of quantitative or qualitative analysis of ctDNA and the subgroup of septin9 (SEPT9) methylation assay. (3) Results: A total of 79 qualified articles with 25,240 subjects were incorporated into our meta-analysis. For quantitative studies, the combined sensitivity (SEN), specificity (SPE), and diagnostic odds ratio (DOR) were 0.723 (95% CI: 0.623–0.803), 0.920 (95% CI: 0.827–0.966), and 23.305 (95% CI: 9.378–57.906), respectively, yielding an AUC of 0.860. The corresponding values for qualitative studies were 0.610 (95% CI: 0.566–0.651), 0.891 (95% CI: 0.878–0.909), 12.569 (95% CI: 9.969–15.848), and 0.823, respectively. Detection of SEPT9 methylation depicted an AUC of 0.879, with an SEN of 0.679 (95% CI: 0.622–0.732), an SPE of 0.903 (95% CI: 0.878–0.923), and a DOR of 20.121 (95% CI:14.404–28.106), respectively. (4) Conclusion: Blood-based ctDNA assay would be a potential novel biomarker for CRC screening and diagnosis. Specifically, quantitative analysis of ctDNA or qualitative analysis of SEPT9 methylation exhibited satisfying diagnostic efficiency. Larger sample studies are needed to further confirm our conclusions and to make the ctDNA approach more sensitive and specific.

Objective: A method for determining the residues of 30 antiparasitic drugs in animal-derived food using ultrahigh- performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was established. Methods: The samples were extracted with acetonitrile and 1% ammonia acetic ether and purified by QuEChERS. After purification, Waters ACQUITY UPLCTM BEH C18 column was used for separation, and a gradient elution of 10 mmol/L ammonium formate (containing 0.1% formic acid) aqueous solution, acetonitrile:methanol (50:50, v:v) was performed as the mobile phase. Detection was carried out using electrospray ionization (ESI) in both positive and negative ion modes using multiple reaction monitoring (MRM). Matrix-matched external standard quantification was used. Results: Under the optimized conditions, the 30 antiparasitic drugs showed good linearity within their respective linear ranges, with coefficient of determination (r2) greater than 0.99. The recoveries ranged from 70.1% to 111%, and the relative standard deviations were between 0.10% and 9.1% (n=6). The method detection limit ranged from 0.001 to 0.3 μg/kg, and the method quantification limit ranged from 0.004 to 1 μg/kg. Conclusion: The method is sensitive, accurate, and exhibits good repeatability and stability, making it suitable for detecting various antiparasitic drug residues in animal-derived food. Lung cancer is a leading cause of cancer‐related deaths worldwide. Recent studies have identified pyroptosis, a type of programmed cell death, as a critical process in the development and progression of lung cancer. In this study, we investigated the effect of EEBR, a new compound synthesized by our team, on pyroptosis in non‐small cell lung cancer cells (NSCLC) and the underlying molecular mechanisms. Our results demonstrated that EEBR significantly reduced the proliferation and metastasis of NSCLC cells in vitro. Moreover, EEBR‐induced pyroptosis in NSCLC cells, as evidenced by cell membrane rupture, the release of cytokines such as interleukin‐18 and interleukin‐1 beta and the promotion of Gasdermin D cleavage in a Caspase‐1‐dependent manner. Furthermore, EEBR promoted the nuclear translocation of NF‐κB and upregulated the protein level of NLRP3. Subsequent studies revealed that EEBR‐induced pyroptosis was suppressed by the inhibition of NF‐κB. Finally, EEBR effectively suppressed the growth of lung cancer xenograft tumours by promoting NSCLC pyroptosis in animal models. Taken together, our findings suggest that EEBR induces Caspase‐1‐dependent pyroptosis through the NF‐κB/NLRP3 signalling cascade in NSCLC, highlighting its potential as a candidate drug for NSCLC treatment.
