In this paper, an adaptive fault-tolerant method is proposed to synthesize a compensation controller for the uncertain nonlinear pure-feedback systems possessing dead-zone actuators and stochastic failures. Each actuator’s failure mode is described by a scalar Markovian type function, which is not only much more practical in control engineering but also challenging in control theory. By exploring the adaptive backstepping methodology, a compensation scheme for actuator failure is presented to guarantee that the solution of the closed-loop system is a unique and bounded in probability. Importantly, the proposed control method can achieve arbitrarily small tracking error in the presence of nonlinear actuators with random failures. Background: Severe hypoxia induces a series of stress responses in mammals; however, subterranean rodents have evolved several adaptation mechanisms of energy metabolisms and O2 utilization for hypoxia. Mammalian brains show extreme aerobic metabolism. Following hypoxia exposure, mammals usually experience irreversible brain damage and can even develop serious diseases, such as hypoxic ischemic encephalopathy and brain edema. To investigate mechanisms underlying the responses of subterranean rodents to severe hypoxia, we performed a cross-species brain transcriptomic analysis using RNA sequencing and identified differentially expressed genes (DEGs) between the subterranean rodent Lasiopodomys mandarinus and its closely related aboveground species L. brandtii under severe hypoxia (5.0% O2, 6 h) and normoxia (20.9% O2, 6 h). Results: We obtained 361 million clean reads, including 69,611 unigenes in L. mandarinus and 69,360 in L. brandtii. We identified 359 and 515 DEGs by comparing the hypoxic and normoxia groups of L. mandarinus and L. brandtii, respectively. Gene Ontology (GO) analysis showed that upregulated DEGs in both species displayed similar terms in response to severe hypoxia; the main difference is that GO terms of L. brandtii were enriched in the immune system. However, in the downregulated DEGs, GO terms of L. mandarinus were enriched in cell proliferation and protein transport and those of L. brandtii were enriched in nuclease and hydrolase activities, particularly in terms of developmental functions. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed that upregulated DEGs in L. mandarinus were associated with DNA repair and damage prevention as well as angiogenesis and metastasis inhibition, whereas downregulated DEGs were associated with neuronal synaptic transmission and tumor-associated metabolic pathways. In L. brandtii, upregulated KEGG pathways were enriched in the immune, endocrine, and cardiovascular systems and particularly in cancer-related pathways, whereas downregulated DEGs were associated with environmental information processing and misregulation in cancers. Conclusions: L. mandarinus has evolved hypoxia adaptation by enhancing DNA repair, damage prevention, and augmenting sensing, whereas L. brandtii showed a higher risk of tumorigenesis and promoted innate immunity toward severe hypoxia. Background: Subterranean rodents have evolved many features to adapt to their hypoxic environment. The brain is an organ that is particularly vulnerable to damage caused by exposure to hypoxic conditions. To investigate the mechanisms of adaption to a hypoxic underground environment, we carried out a cross-species brain transcriptome analysis by RNA sequencing and identified genes that are differentially expressed between the subterranean vole Lasiopodomys mandarinus and the closely related above-ground species Lasiopodomys brandtii under chronic hypoxia [10.0% oxygen (O2)] and normoxia (20.9% O2). Results: A total of 355 million clean reads were obtained, including 69,611 unigenes in L. mandarinus and 69,360 in L. brandtii. A total of 235 and 92 differentially expressed genes (DEGs) were identified by comparing the hypoxic and control groups of L. mandarinus and L. brandtii, respectively. A Gene Ontology (GO) analysis showed that upregulated DEGs in both species had similar functions in response to hypoxia, whereas downregulated DEGs in L. mandarinus were enriched GO terms related to enzymes involved in aerobic reactions. In the Kyoto Encyclopedia of Genes and Genomes pathway analysis, upregulated DEGs in L. mandarinus were associated with angiogenesis and the increased O2 transport capacity of red blood cells, whereas downregulated DEGs were associated with immune responses. On the other hand, upregulated DEGs in L. brandtii were associated with cell survival, vascular endothelial cell proliferation, and neuroprotection, while downregulated genes were related to the synaptic transmission by neurons. Conclusions: L. mandarinus actively adapts its physiological functions to hypoxic conditions, for instance by increasing O2 transport capacity and modulating O2 consumption. In contrast, L. brandtii reacts passively to hypoxia by decreasing overall activity in order to reduce O2 consumption. These results provide insight into hypoxia adaptation mechanisms in subterranean rodents that may be applicable to humans living at high altitudes or operating in other O2-poor environments. With three-dimensional (3D) microelectrodes used to machine microcavities in microelectrical discharge machining (micro-EDM), the processing efficiency can be greatly improved. However, the surfaces of a laminated 3D microelectrode always have stair steps, which will be further copied to the machined surfaces of the microcavities, reducing the processed surface quality. To solve the problems, the paper proposed using layer-by-layer scanning microelectrochemical machining (micro-ECM) to eliminate the stair-stepping effect of the laminated 3D microelectrodes. First, wire electrical discharge machining (WEDM) was chosen to cut Cu foils for obtaining multilayer two-dimensional (2D) microstructures. Second, the multilayer 2D microstructures were applied to thermal diffusion bonding to form 3D microelectrode blanks. Then, the 3D microelectrode blanks were processed via layer-by-layer scanning micro-ECM to eliminate the stair steps on the surfaces. The influences of applied voltage, offset distance, and layer thickness on the elimination of the stair-stepping effect were investigated in details. Experimental results show that the stair steps on the laminated 3D microelectrode surfaces were eliminated obviously under 4.0 V applied voltage, 200 μm offset distance, and 20 μm layer thickness. Finally, the laminated 3D microelectrodes without stair steps on the surfaces were applied to micro-EDM to process microcavities. Two typical microcavities without the stair steps were obtained. Abstract: Orbitally shaken bioreactors (OSRs) are commonly used for the cultivation of mammalian cells in suspension. To aid the geometry designing and optimizing of OSRs, we conducted a three‐dimensional computational fluid dynamics (CFD) simulation to characterize the flow fields in a 10 L cylindrical OSR with different vessel diameters. The liquid wave shape captured by a camera experimentally validated the CFD models established for the cylindrical OSR. The geometry size effect on volumetric mass transfer coefficient (kLa) and hydromechanical stress was analyzed by varying the ratio of vessel diameter (d) to liquid height at static (hL), d/hL. The highest value of kLa about 30 h−1 was observed in the cylindrical vessel with the d/hL of 6.35. Moreover, the magnitudes of shear stress and energy dissipation rate in all the vessels tested were below their minimum values causing cells damage separately, which indicated that the hydromechanical‐stress environment in OSRs is suitable for cells cultivation in suspension. Finally, the CFD results suggested that the d/hL higher than 8.80 should not be adopted for the 10 L cylindrical OSR at the shaking speed of 180 rpm because the “out of phase” state probably will happen there. In micro scale metal forming processes, the plastic deformation behaviors are complicated and difficult to predict because of size effects. In this study, micro cylinder compression tests have been carried out to determine the plastic deformation behaviors in micro bulk metal forming. The effects of specimen height-to-diameter ratio and strain on the plastic deformation behaviors have been investigated extensively by compressing pure nickel micro cylinders. It is found that both the flow stress and friction coefficient decrease with the increase of the specimen height-to-diameter ratio. Friction coefficient decreases with the increase of compression ratio for specimens with the same height or decreases with the increase of the specimen height-to-diameter ratio under the same compression ratio. A relationship between normal pressure of lubricant friction, strain and height-to-diameter ratio ( H/D ) is established. A modified friction model is proposed based on the assumption of open-close lubricant pockets by introducing the specimen geometrical parameters. The calculated friction coefficients by the developed model and their tendencies are in good accordance with the experimental ones. It is also found that the ratio of the real contact areas (RCAs) decreases with increase of the specimen height-to-diameter ratio under the same compression ratio which can explain the decrease of the equivalent friction stress between the tool and the specimen. Micro wire electrical discharge machining (micro-WEDM) provides a promising alternative for micro gear machining. However, the processing defect of non-intact tooth profile often appears due to the influence of micro gear gravity, flushing force and explosive force in micro-WEDM process. This paper presents a process change of micro-WEDM to achieve the precise fabrication of micro gear with intact tooth profile, i.e. firstly reserving a sharp corner on the gear teeth where the processing defect exists and then removing it by the second processing of micro-WEDM. The locating and clamping of the micro gear with sharp corner has been achieved using a novel self-centering flexible fixture. The machining error during the second processing is analyzed, and whilst the process datum is accurately determined. Appropriate processing parameters are obtained by performing preliminary experiments on X153CrMoV12 workpiece, indicating that high open voltage and discharge capacitance cause large kerf width, moderate reference voltage and feed rate lead to narrow kerf width. The static contact between the core jig of the fixture and the micro gear with sharp corner is theoretically analyzed using ABAQUS software and experimentally proved to be in good condition. To verify the feasibility and repeatability of the proposed approach, five micro gears with 10 gear number, 0.1 mm module and 2 mm tooth width are machined using 50 μm diameter wire electrode, exhibiting excellent conformity, machining error with less than 1.5 μm and surface roughness Ra with 0.9 μm. With the challenges of arid transboundary river water management among Central Asian countries, it is important to evaluate the water stress and propose practicable climate adaptation strategies. This study used the Syr Darya Basin (SDB) in a Central Asia data scarce region as the study area and, conducted the first quantified analysis of the spatiotemporal characteristics of water supply and requirements at the basin and irrigation district scale; identified its driving mechanisms based on systematic monitoring data concerning reservoir regulation and complex crop structure from 2000 to 2018; and a climate change scenario combined with a crop restructuring scenario was applied to evaluate water conservation effects and propose a climate adaptation strategy from 2019 to 2030. The results showed that runoff increased by 7.45 billion m3 but the water withdrawal decreased by 3.42 billion m3 after reservoir regulation in the basin during 2000–2018. Due to the unified water allocation, it was challenging to conduct and usually exceeded the plan by 0.4–8.5 %. The total crop water requirement (TCWR) mainly derived from cotton (61.5 %), winter wheat (12.1 %) and rice (13.9 %) increased by 3.15 billion m3 (+30.7 %) and the influence of crop area (380 million m3/a) was greater than that of climate (130 million m3/a). The wind speed (27.5 %) and rice area (19.2 %) also significantly contributed to the TCWR variation. The contradiction between water supply and requirement resulted in water scarcity in the basin. From 2019–2030, the water conservation effect of winter wheat (+20 %) under the restructuring scenarios reached 28.7 mm, which helped relieve water stress under the scenario of a 0.51–0.91 °C increase. Thus, priority planting of a winter wheat rotation and low water consuming crops is a practicable approach for coping with water scarcity and adapting to climate change in arid transboundary rivers of Centra Asia. Mandarin vole ( Lasiopodomys mandarinus ) is a subterranean rodent that is often used as a model for studying subterranean hypoxic stress in mammals. However the taxonomy of this species is still in dispute. Mitochondrial DNA (mtDNA) has long been used for phylogenetic reconstruction and, in this study, the complete mitochondrial genome of L. mandarinus mandarinus was sequenced. Our results showed that the mitochondrial genome of L. m. mandarinus is a circular molecule of 16,367 bp, which contains 13 protein-coding genes, 22 tRNA and 2 rRNA genes. Except for the 8 tRNA and ND6 genes, all other mitochondrial genes are encoded on the heavy strand. We also analyzed the phylogenetic position of L. mandarinus in respect to the tribe Arvicolini using the sequence of complete Cytb gene, 2rRNA genes and 12 protein-coding genes, and maximum likelihood and Bayesian methods. Background/Aims: Notch signaling pathway regulates cancer cell growth. RBPJ is a key transcription factor downstream of Notch receptor activation, whereas the role of RBPJ in carcinogenesis of prostate cancer is ill-defined. Methods: Here, we evaluated the effects of RBPJ inhibition on the growth of prostate cancer cells. We knocked down RBPJ in prostate cancer cells by a short hairpin interfering RNA (shRNA). We measured cell growth by an MTT assay. We analyzed the levels of cell-cycle-associated proteins by Western blot. Results: We found that shRNA for RBPJ efficiently inhibited RBPJ expression in prostate cancer cells, resulting in a significant decrease in the cell growth. Further, RBPJ-mediated cell-growth inhibition appeared to be resulting from alteration of cell-cycle inhibitors p21 and p27, cellcycle activators CDK2, CDK4 and CyclinD1, and apoptosis-suppressor Bcl-2. Evapotranspiration and soil moisture content (SMC) are key elements of the hydrological cycle. Accurate prediction of the dynamic processes of evapotranspiration and soil water is essential for irrigation and water management. Here, the boosted regression tree (BRT) method was employed to quantify environmental controls on actual evapotranspiration (ET a), potential evapotranspiration (ET 0), and SMC using monitoring data from the Wudaogou hydrological experimental station. The results indicated that: (1) the BRT algorithm was effective in predicting the relative control of different environmental factors on ET a , ET 0 , and SMC; and (2) vapor pressure deficit (VPD) was the most important factor affecting daily ET 0 , and sunshine duration (SSD) also played a nonnegligible role. The results further explained the phenomenon of the "evaporation paradox" in the study area. SSD could be a leading control on daily ET a , followed by VPD, leaf area index (LAI). (3) Among the underground factors, groundwater level (GL) and LAI played a dominant role in the relative contribution to SMC. Among the aboveground factors, relative humidity (RH) and soil temperature (TS) have a relatively large influence on SMC. (4) The differences in SMC at different depths were determined by multiple influencing factors, including LAI, VPD, and precipitation (P). This study also underscores the importance of vegetation variations to hydrological cycle processes. Abstract: In micro-wire electrical discharge machining (micro-WEDM), the machined kerf width varies with different machining parameters, which greatly influences the machining precision. In order to study the kerf variations in micro-WEDM, the mathematical model of wire lateral vibration in machining process is established and its analytical solution is obtained in this paper. The model is practically verified on a self-developed micro-WEDM machine. Under this model, a 30.8μm width slot is achieved on a stainless steel workpiece with ∅30μm wire-tool. The acoustic softening effect in metals during plastic deformation has been widely investigated in the past decades. However, the mechanism of such an acoustic plasticity remains controversial. As a result, several models were proposed to understand the acoustic softening effect in terms of stress superposition, thermal activation theory, crystal plastic theory and other mechanisms associated with dislocation evolution. In this study, we proposed a mechanism that the athermal dislocation dynamics may heterogeneously change at microstructure level during ultrasonic vibration assisted (UVA) deformation. Specifically, the work required to eject dislocations from grain boundaries may be altered by the acoustic energy absorption difference of a dislocation in the grain interior and one in the grain boundary. As a result, the acoustic softening effect on the Hall-Petch behavior was modeled by incorporating a power function of acoustic energy density into the dislocation ejection work. To validate the developed model, UVA micro-tension tests were conducted on pure titanium specimens. Results showed that the Hall-Petch slope decreased due to ultrasonic vibration, and the ultrasound-induced decrease of the Hall-Petch slope increased with plastic deformation. Note that our model predictions matched well with the experimental results at the lower strains, providing an alternative insight into the acoustic softening effect on the Hall-Petch behavior. Microstructure examinations showed that the superimposed ultrasonic vibration in micro-tension could lead to the retardation of texture evolution, the decreases in kernel average misorientation (KAM) value, low-angle grain boundary (LAGB) fraction and dislocation density in titanium foils, which somewhat supported our model assumptions in terms of the acoustic energy dependent reduction in dislocation density and the enhanced dislocation ejection causing the improvement of plastic compatibility. • A Hall-Petch model based on a dislocation ejection mechanism was developed with the acoustic plasticity involved. • Ultrasonic vibration induced the decease of the Hall-Petch slope which increased with plastic deformation. • The proposed model can capture the decrease in strain hardening rate in UVA micro-tension of titanium. • The model predictions matched well with the acoustic softening effect on the Hall-Petch slope at lower strains. • Microstructure examinations showed dislocation density reduction and enhanced plastic compatibility in UVA foils. Background: Circulating tumor cells (CTCs) play a key role in cancer progression, especially metastasis, due to the rarity and heterogeneity of CTCs, fewer researches have been conducted on them at the molecular level. However, through the Gene Expression Omnibus (GEO) database, this kind of minority researches can be well integrated, the gene expression differences between CTCs and primary tumors can be identified, and molecular targets for CTCs can be found. Methods: We analyzed 7 sets of gene chips (GSE82198, GSE99394, GSE31023, GSE65505, GSE67982, GSE76250, GSE50746) obtained by GEO. Analysis of differentially expressed genes (DEGs) between CTCs and corresponding primary tumors by NetworkAnalyst. Metascape tool for Gene Ontology (GO) / Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis of differential genes and visual display. Cytoscape performs protein-protein interaction (PPI) analysis and obtains the hub genes. Renal cancer patients' clinical specimens to verify the correctness of enrichment results. Prognostic analysis of hub genes in kidney cancer patients using the Kaplan–Meier plotter survival analysis tool. Results: We obtained a total of 589 DEGs. The GO / KEGG enrichment results indicate that the DEGs are mainly concentrated in cell adhesion, epithelial-mesenchymal transition (EMT), and apoptosis. Renal cancer clinical specimens suggest that CTCs have epithelial and mesenchymal types. At the same time, PSMC2 can be used as a poor prognostic indicator for renal cancer patients. Conclusions: In summary, our study suggests that compared with primary tumors, CTCs mainly change cell adhesion, EMT, and apoptosis. PSMC2 can be used as a poor prognostic factor. The Mandarin vole (Lasiopodomys mandarinus), a typical subterranean rodent, has undergone hematological adaptations to tolerate the hypoxic/hypercapnic underground environment. Hemoglobin (Hb) genes encode respiratory proteins functioning principally in oxygen binding and transport to various tissues and organs. To investigate the evolution of α- and β-hemoglobin (Hb) in subterranean rodent species, we sequenced Hb genes of the Mandarin vole and the related aboveground Brandt's vole (L. brandtii). Sequencing showed that in both voles, α-globin was encoded by a cluster of five functional genes in the following linkage order: HBZ, HBA-T1, HBQ-T1, HBA-T2, and HBQ-T2; among these, HBQ-T2 is a pseudogene in both voles. The β-globin gene cluster in both voles also included five functional genes in the following linkage order: HBE, HBE/HBG, HBG, HBB-T1, and HBB-T2. Phylogenetic analysis revealed that the Mandarin vole underwent convergent evolution with its related aboveground species (Brandt's vole) but not with other subterranean rodent species. Selection pressure analyses revealed that α- and β-globin genes are under strong purifying selection (ω < 1), and branch-site analyses identified positive selection sites on HBAT-T1 and HBB-T1 in different subterranean rodent species. This suggests that the adaptive evolution of these genes enhanced the ability of Hb to store and transport oxygen in subterranean rodent species. Developing bio-inspired functional surfaces on engineering metals is of extreme importance, involving different industrial sectors, like automotive or aeronautics. In particular, micro-embossing is one of the efficient and large-scale processes for manufacturing bio-inspired textures on metallic surfaces. However, this process faces some problems, such as filling defects and die breakage due to size effect, which restrict this technology for some components. Electrically assisted micro-forming has demonstrated the ability of reducing size effects, improving formability and decreasing flow stress, making it a promising hybrid process to control the filling quality of micro-scale features. This research focuses on the use of different current densities to perform embossed micro-channels of 7 μm and sharklet patterns of 10 μm in textured bulk metallic glass dies. These dies are prepared by thermoplastic forming based on the compression of photolithographic silicon molds. The results show that large areas of bio-inspired textures could be fabricated on magnesium alloy when current densities higher than 6 A/mm2 (threshold) are used. The optimal surface quality scenario is obtained for a current density of 13 A/mm2. Additionally, filling depth and depth–width ratio nonlinearly increases when higher current densities are used, where the temperature is a key parameter to control, keeping it below the temperature of the glass transition to avoid melting or an early breakage of the die. This work puts forward numerical and experimental investigations on laser dicing of glass-anisotropic single-crystal silicon double-layer wafer using laser induced thermal-crack propagation (LITP). A semiconductor continuous wave laser working at the defocusing mode serves as volumetric heat source for glass layer while as surface heat source for silicon layer. Based on the classical fracture theory, a static seam-type crack is introduced under the circumstance of ABAQUS to simulate the crack fracture modes in glass layer as well as silicon layer with crystal planes of (100), (110) and (111) during laser dicing in different dicing directions. In the experiments, processing parameters are kept the same as the simulations and typical dicing directions obtained from simulations are also used. The surface morphologies of crack edges are measured by the optical microscope and surface profiler. Through the comparison of numerical and experimental results it is discovered that for the specific substrate, the evolution of crack edge qualities in different dicing directions and different layers can be interpreted based on the corresponding stress distribution and stress intensity factor (SIF) ratio explicitly. And most important of all, the anisotropy of silicon layer has significant influence on the fracture mode and edge quality of crack in both layers. [ABSTRACT FROM AUTHOR]

The size effects in metal forming have been found to be crucial in micro-scale plastic deformation or micro-forming processes, which lead to attenuation of the material's formability due to the increasing heterogeneity of the plastic flow. The use of an electric field during micro-scale plastic deformation has shown to relieve size effects, enhance the material's formability, modify the microstructure, etc. Consequently, these electric-assisted (EA) micro-forming processes seem to bring many potential benefits that need to be investigated. Accordingly, here we investigated the influence of an electric field on the size effects to describe the fracture behavior in uniaxial micro-tension tests of an AZ31 alloy with various grain sizes. In order to decouple the thermal-mechanical and microstructure changes, room temperature (RT), oven-heated (OH), air-cooled (AC), and EA uniaxial micro-tension tests were conducted. The size effects contribution on the fracture stress and strain showed a similar trend in all the testing configurations. However, the smallest fracture stresses and the largest fracture strains were denoted in the EA configuration. EBSD examination shows that current-induced dynamic recrystallization (DRX) and texture evolution could be negligible under the studied conditions. The kernel average misorientation (KAM) maps give the larger plastic deformation in the EA specimens due to the reduction of plastic micro-heterogeneity. Finally, the fracture morphology indicates that the current-induced ductility enhancement may be attributed to the arrest of micro-crack propagation and the inhibition of void initiation, growth, and coalescence caused by lattice melting and expansion. Any object can be considered as a spatial distribution of atoms and molecules; in this sense, we can manufacture any object as long as the precise distribution of atoms and molecules is achieved. However, the current point-by-point methods to precisely manipulate single atoms and single molecules, such as the scanning tunneling microscope (STM), have difficulty in manipulating a large quantity of materials within an acceptable time. The macroscopic manipulation techniques, such as magnetron sputtering, molecular beam epitaxy, and evaporation, could not precisely control the distribution of materials. Herein, we take a step back and present a universal method of controlling the distribution of multimaterails during macroscopic manipulation via microtopography-guided substrates. For any given target distribution of multimaterials in a plane, the complicated lateral distribution of multimaterials was firstly transformed into a simple spatial lamellar body. Then, a deposition mathematical model was first established based on a mathematical transformation. Meanwhile, the microtopographic substrate can be fabricated according to target distribution based on the deposition mathematical model. Following this, the deposition was implemented on the substrate according to the designed sequence and thickness of each material, resulting in the formation of the deposition body on the substrate. Finally, the actual distribution was obtained on a certain section in the deposition body by removing the upside materials. The actual distribution can mimic the target one with a controllable accuracy. Furthermore, two experiments were performed to validate our method. As a result, we provide a feasible and scalable solution for controlling the distribution of multimaterials, and point out the direction of improving the position accuracy of each material. We may achieve real molecular manufacturing and nano-manufacturing if the position accuracy of distribution approaches the atomic level. [ABSTRACT FROM AUTHOR]
