1. Trpv6 channel targeting using monoclonal antibody induces prostate cancer cell apoptosis and tumor regression

Abstract

Abstract TRPV6 calcium channel is a prospective target in prostate cancer (PCa) since it is not expressed in healthy prostate while its expression increases during cancer progression. Despite the role of TRPV6 in PCa cell survival and apoptotic resistance has been already established, no reliable tool to target TRPV6 channel in vivo and thus to reduce tumor burden is known to date. Here we report the generation of mouse monoclonal antibody mAb82 raised against extracellular epitope of the pore region of the channel. mAb82 inhibited TRPV6 currents by 90% at 24 µg/ml in a dose-dependent manner while decreasing store-operated calcium entry to 56% at only 2.4 µg/ml. mAb82 decreased PCa survival rate in vitro by 71% at 12 µg/ml via inducing cell death through the apoptosis cascade via activation of the protease calpain, following bax activation, mitochondria enlargement, and loss of cristae, Cyt C release, pro-caspase 9 cleavage with the subsequent activation of caspases 3/7. In vivo, mice bearing either PC3M trpv6+/+ or PC3M trpv6-/- +pTRPV6 tumors were successfully treated with mAb82 at the dose as low as 100 µg/kg resulting in a significant reduction tumor growth by 31% and 90%, respectively. The survival rate was markedly improved by 3.5 times in mice treated with mAb82 in PC3M trpv6+/+ tumor group and completely restored in PC3M trpv6-/- +pTRPV6 tumor group. mAb82 showed a TRPV6-expression dependent organ distribution and virtually no toxicity in the same way as mAbAU1, a control antibody of the same Ig2a isotype. Overall, our data demonstrate for the first time the use of an anti-TRPV6 monoclonal antibody in vitro and in vivo in the treatment of the TRPV6-expressing PCa tumors.

Abstract

Diabetes poses major economic, social, and public health challenges in all countries worldwide. Besides cardiovascular disease and microangiopathy, diabetes is a leading cause of foot ulcers and lower limb amputations. With the continued rise of diabetes prevalence, it is expected that the future burden of diabetes complications, early mortality, and disabilities will increase. The diabetes epidemic is partly caused by the current lack of clinical imaging diagnostic tools, the timely monitoring of insulin secretion and insulin-expressing cell mass (beta (β)-cells), and the lack of patients’ adherence to treatment, because some drugs are not tolerated or invasively administrated. In addition to this, there is a lack of efficient topical treatment capable of stopping the progression of disabilities, in particular for treating foot ulcers. In this context, polymer-based nanostructures garnered significant interest due to their tunable physicochemical characteristics, rich diversity, and biocompatibility. This review article emphasizes the last advances and discusses the prospects in the use of polymeric materials as nanocarriers for β-cell imaging and non-invasive drug delivery of insulin and antidiabetic drugs in the management of blood glucose and foot ulcers.

Abstract

Improvement of insulin secretion by pancreatic β-cells and preservation of their mass are the current challenges that future antidiabetic drugs should meet for achieving efficient and long-term glycemic control in patients with type 2 diabetes (T2D). The successful development of glucagon-like peptide 1 (GLP-1) analogues, derived from the saliva of a lizard from the Helodermatidae family, has provided the proof of concept that antidiabetic drugs directly targeting pancreatic β-cells can emerge from venomous animals. The literature reporting on the antidiabetic effects of medicinal plants suggests that they contain some promising active substances such as polyphenols and alkaloids, which could be active as insulin secretagogues and β-cell protectors. In this review, we discuss the potential of several polyphenols, alkaloids and venom peptides from snake, frogs, scorpions and cone snails. These molecules could contribute to the development of new efficient antidiabetic medicines targeting β-cells, which would tackle the progression of the disease.

Abstract

Bioactive aminooxime ligands based on optically pure (R)-limonene have been synthesized in two steps. Their ruthenium (II) cationic water-soluble complex was prepared by a reaction between dichloro (para-cymene) ruthenium (II) dimers and aminooxime ligands in a 1:2 molar ratio. Antibacterial and antibiofilm activities of the synthetized complex were assessed against Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, and Enterococcus faecalis. The results revealed that the ruthenium (II) complex has higher antibacterial and antibiofilm activities in comparison with free ligands or the enantiopure (R)-limonene. Moreover, microencapsulation of this complex reduced its cytotoxicity and improved their minimum inhibitory concentration and antibiofilm activity toward the considered bacteria. The ruthenium (II) complex targets the bacterial cell membrane, which leads to rapid leakage of intracellular potassium. Our study suggests that the developed ruthenium (II) complexes could be useful as an alternative to conventional disinfectants.

Abstract

Photoreduction of hexavalent chromium, Cr(VI), identified as carcinogenic and mutagenic element, to Cr(III), believed to be an essential element, using copper sulfide nanostructures (CuS NSs) as photocatalyst was investigated under visible light irradiation (λ > 420 nm). The CuS NSs were synthesized at low temperature using a wet chemical route and fully characterized using various techniques such as scanning electron microscopy, transmission electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy, Raman spectroscopy and UV–vis spectrophotometry. The photocatalytic reduction of Cr(VI) using visible light irradiation was assessed by UV–vis by following the decrease of the characteristic absorbance at ∼352 nm in the presence of CuS NSs. While CuS NSs promoted Cr(VI) full reduction within 60 min, addition of adipic acid or formic acid accelerated significantly the photocatalytic reduction process. Under optimized conditions, CuS NSs rapidly (12 min) reduced Cr(VI) in presence of 0.5 mM adipic acid. Similarly, Cu NSs were found to be efficient for Cr(VI) reduction dissolved in water from a local lake. Stability studies showed that the photocatalyst could be recycled up to four times without any apparent decrease of its catalytic performance. Abstract

Dermaseptin B2 (DRS-B2) is an antimicrobial peptide secreted by Phyllomedusa bicolor, which is an Amazonian tree frog. Here, we show that the adsorption of DRS-B2 on alginate nanoparticles (Alg NPs) results in a formulation (Alg NPs + DRS-B2) with a remarkable antibacterial activity against Escherichia coli ATCC 8739 and E. coli 184 strains, which are sensitive and resistant, respectively, to colistin. The antibacterial activity, obtained with this new formulation, is higher than that obtained with DRS-B2 alone. Of note, the addition of lactic acid or menthol to this new formulation augments its antibacterial activity against the aforementioned Gram-negative bacilli. The safety of DRS-B2, and also that of the new formulation supplemented or not with a small molecule such as lactic acid or menthol has been proven on the human erythrocytes and the eukaryotic cell line types HT29 (human) and IPEC-1 (animal). Similarly, their stability was determined under the conditions mimicking the gastrointestinal tract with different conditions: pH, temperature, and the presence of digestive enzymes. Based on all the obtained data, we assume that these new formulations are promising and could be suggested, after in vivo approval and completing regulation aspects, as alternatives to antibiotics to fight infections caused by Gram-negative bacilli such as E. coli.

Abstract

Bacterial resistance to antibiotics has become a major public health problem worldwide, with the yearly number of deaths exceeding 700,000. To face this well-acknowledged threat, new molecules and therapeutic methods are considered. In this context, the application of nanotechnology to fight bacterial infection represents a viable approach and has experienced tremendous developments in the last decades. Escherichia coli (E. coli) is responsible for severe diarrhea, notably in the breeding sector, and especially in pig farming. The resulting infection (named colibacillosis) occurs in young piglets and could lead to important economic losses. Here, we report the design of several new formulations based on colistin loaded on alginate nanoparticles (Alg NPs) in the absence, but also in the presence, of small molecules, such as components of essential oils, polyamines, and lactic acid. These new formulations, which are made by concomitantly binding colistin and small molecules to Alg NPs, were successfully tested against E. coli 184, a strain resistant to colistin. When colistin was associated with Alg NPs, the minimal inhibition concentration (MIC) decreased from 8 to 1 µg/mL. It is notable that when menthol or lactic acid was co-loaded with colistin on Alg NPs, the MIC of colistin drastically decreased, reaching 0.31 or 0.62 µg/mL, respectively. These novel bactericidal formulations, whose innocuity towards eukaryotic HT-29 cells was established in vitro, are presumed to permeabilize the bacterial membrane and provoke the leakage of intracellular proteins. Our findings revealed the potentiating effect of the Alg NPs on colistin, but also of the small molecules mentioned above. Such ecological and economical formulations are easy to produce and could be proposed, after confirmation by in vivo and toxicology tests, as therapeutic strategies to replace fading antibiotics.

Abstract

The synthesis of multifunctional photothermal nanoagents for antibiotic loading and release remains a challenging task in nanomedicine. Herein, we investigated a simple, low-cost strategy for the preparation of CuS-BSA nanoparticles (NPs) loaded with a natural enzyme, lysozyme, as an antibacterial drug model under physiological conditions. The successful development of CuS-BSA NPs was confirmed by various characterization tools such as transmission electron microscopy (TEM), X-ray diffraction (XRD), Raman spectroscopy, and X-ray photoelectron spectroscopy (XPS). Lysozyme loading onto CuS-BSA NPs was evaluated by UV/vis absorption spectroscopy, Fourier-transform infrared spectroscopy (FTIR), zeta potential, and dynamic light scattering measurements. The CuS-BSA/lysozyme nanocomposite was investigated as an effective means for bacterial elimination of B. subtilis (Gram-positive) and E. coli (Gram-negative), owing to the combined photothermal heating performance of CuS-BSA and lysozyme release under 980 nm (0.7 W cm−2) illumination, which enhances the antibiotic action of the enzyme. Besides the photothermal properties, CuS-BSA/lysozyme nanocomposite possesses photodynamic activity induced by NIR illumination, which further improves its bacterial killing efficiency. The biocompatibility of CuS-BSA and CuS-BSA/Lysozyme was elicited in vitro on HeLa and U-87 MG cancer cell lines, and immortalized human hepatocyte (IHH) cell line. Considering these advantages, CuS-BSA NPs can be used as a suitable drug carrier and hold promise to overcome the limitations of traditional antibiotic therapy.

Abstract

Vaccination remains one of the most effective tools to prevent infectious diseases. To ensure that the best possible antigenic components are chosen to stimulate a cognitive immune response, boosting antigen presentation using adjuvants is common practice. Nanodiamond-based adjuvants are proposed here as a rapid and versatile platform for antigen conjugation, utilizing peptides common to different pathogenic strains and making this strategy a good candidate for a “ready-to-use” vaccine. Initiation of an inflammatory reaction with a resulting immune response is based on the ability of living organisms to entrap nanostructures such as nanodiamonds with neutrophil extracellular traps (NETs) formation. In this work, coronavirus peptide homological for MERS-CoV, fusion inhibitor, was conjugated to nanodiamonds and used to induce neutrophilic-driven self-limiting inflammation. The resulting adjuvant was safe and did not induce any tissue damage at the site of injection. Mice immunization resulted in IgG titers of ¼,000 within 28 days. Immunization of rabbits resulted in the formation of a high level of antibodies persistently present for up to 120 days after the first immunization (animal lifespan ~3 years). The peptide used for immunization proved to be reactive with sera of convalescent COVID patients, demonstrating the possibility of developing pancoronaviral vaccine candidates.

Abstract

Background: The main goals of cancer treatment are not only to eradicate the tumor itself but also to elicit a specific immune response that overcomes the resistance of tumor cells against chemo- and radiotherapies. Hyperthermia was demonstrated to chemo- and radio-sensitize cancerous cells. Many reports have confirmed the immunostimulatory effect of such multi-modal routines. Methods: We evaluated the interaction of graphene oxide (GO) nanosheets; its derivatives reduced GO and PEGylated rGO, with components of peripheral blood and evaluated its thermal conductivity to induce cell death by localized hyperthermia. Results: We confirmed the sterility and biocompatibility of the graphene nanomaterials and demonstrated that hyperthermia applied alone or in the combination with radiotherapy induced much more cell death in tumor cells than irradiation alone. Cell death was confirmed by the release of lactate dehydrogenase from dead and dying tumor cells. Conclusion: Biocompatible GO and its derivatives can be successfully used in graphene-induced hyperthermia to elicit tumor cell death.

Abstract

Multivalent interactions are omnipresent in biology and confer biological systems with dramatically enhanced affinities towards different receptors. Such multivalent binding interactions have lately been considered for the development of new therapeutic strategies against bacterial and viral infections. Multivalent polymers, dendrimers, and liposomes have successfully targeted pathogenic interactions. While a high synthetic effort was often needed for the development of such therapeutics, the integration of multiple ligands onto nanostructures turned to be a viable alternative. Particles modified with multiple ligands have the additional advantage of creating a high local concentration of binding molecules. This review article will summarize the different nanoparticle-based approaches currently available for the treatment of viral infections.

Abstract

Silver nanoparticles (Ag NPs) exhibit antibacterial activity and are extensively used in numerous applications. The aim of this study was to examine the toxic effect of Ag NPs on the marine microalga, Chlorella vulgaris. The microalgae, at the exponential growth phase, were treated with different concentrations of Ag NPs (50 and 100 nm) for 96 h. X-Ray diffraction (XRD) results indicated that the used NPs are single and pure Ag phase with a mean crystallite size of 21 and 32 nm. Ag NPs were found to have a negative effect on viable cell concentration, a variable effect on chlorophyll a concentration, and increased ROS formation. Transmission electron microscopy (TEM) analysis revealed that Ag NPs were present inside the microalgae cells and formed large aggregates in the culture medium. Ag+ ions, in the form of AgNO3, were also assessed at higher concentrations and found to cause inhibitory effects.

Abstract

Impaired penetration of antibiotics through bacterial biofilms is one of the reasons for failure of antimicrobial therapy. Hindered drug diffusion is caused on the one hand by interactions with the sticky biofilm matrix and on the other hand by the fact that bacterial cells are organized in densely packed clusters of cells. Binding interactions with the biofilm matrix can be avoided by encapsulating the antibiotics into nanocarriers, while interfering with the integrity of the dense cell clusters can enhance drug transport deep into the biofilm. Vapor nanobubbles (VNB), generated from laser irradiated nanoparticles, are a recently reported effective way to loosen up the biofilm structure in order to enhance drug transport and efficacy. In the present study, we explored if the disruptive force of VNB can be used simultaneously to interfere with the biofilm structure and trigger antibiotic release from light-responsive nanocarriers. The antibiotic tobramycin was incorporated in two types of light-responsive nanocarriers—liposomes functionalized with gold nanoparticles (Lip-AuNP) and graphene quantum dots (GQD)—and their efficacy was evaluated on Pseudomonas aeruginosa biofilms. Even though the anti-biofilm efficacy of tobramycin was improved by liposomal encapsulation, electrostatic functionalization with 70 nm AuNP unfortunately resulted in premature leakage of tobramycin in a matter of hours. Laser-irradiation consequently did not further improve P. aeruginosa biofilm eradication. Adsorption of tobramycin to GQD, on the other hand, did result in a stable formulation with high encapsulation efficiency, without burst release of tobramycin from the nanocarriers. However, even though laser-induced VNB formation from GQD resulted in biofilm disruption, an enhanced anti-biofilm effect was not achieved due to tobramycin not being efficiently released from GQD. Even though this study was unsuccessful in designing suitable nanocarriers for simultaneous biofilm disruption and light-triggered release of tobramycin, it provides insights into the difficulties and challenges that need to be considered for future developments in this regard.

Abstract

Necrosis is associated with a profound inflammatory response. The regulation of necrosis-associated inflammation, particularly the mechanisms responsible for resolution of inflammation are incompletely characterized. Nanoparticles are known to induce plasma membrane damage and necrosis followed by sterile inflammation. We observed that injection of metabolically inert nanodiamonds resulted in paw edema in WT and Ncf1** mice. However, while inflammation quickly resolved in WT mice, it persisted over several weeks in Ncf1** mice indicating failure of resolution of inflammation. Mechanistically, NOX2-dependent reactive oxygen species (ROS) production and formation of neutrophil extracellular traps (NETs) were essential for the resolution of necrosis-induced inflammation: Hence, by evaluating the fate of the particles at the site of inflammation, we observed that Ncf1** mice deficient in NADPH-dependent ROS failed to generate granulation tissue therefore being unable to trap the nanodiamonds. These data suggest that NOX2-dependent NETosis is crucial for preventing the chronification of the inflammatory response to tissue necrosis by forming NETosis-dependent barriers between the necrotic and healthy surrounding tissue.

Abstract

International audience; This work describes the preparation of ternary bismuth ferrite oxide nanoparticles (Bi2Fe4O9 NPs) with an enzyme mimetic activity for dopamine (DA) qualitative and quantitative detection. Bi2Fe4O9 NPs were prepared using a facile, low cost, and one-pot hydrothermal treatment. The chemical composition, morphology, and optical properties of Bi2Fe4O9 nanozyme were characterized using different techniques such as Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction pattern (XRD), X-ray photoelectron spectroscopy (XPS), thermo-gravimetric analysis (TGA), dynamic light scattering (DLS), field-emission scanning electron microscopy (FESEM) imaging, FESEM-energy dispersive X-ray spectroscopy (EDS), UV–vis absorption, and fluorescence emission spectroscopy. Bi2Fe4O9 NPs were utilized to catalyze the oxidation of a typical chromogenic peroxidase substrate, 3,3′,5,5′-tetramethylbenzidine (TMB), to form the blue-colored oxidized product (oxTMB), in the presence of hydrogen peroxide (H2O2). All reactions occurred in acetate buffer solution (pH 3.5) to generate hydroxyl radicals (•OH) and the kinetics were followed by UV–vis absorbance at 654 nm. The steady-state kinetic parameters were obtained from the Michaelis-Menten equation and exhibited a good catalytic efficiency of Bi2Fe4O9 NPs as enzyme mimetics. Michaelis–Menten constant (Km) values were estimated as 0.07 and 0.73 mM for TMB and H2O2, respectively. The presented method is efficient, rapid, cost-effective, and sensitive for the colorimetric detection of dopamine with a linear range (LR) from 0.15 to 50 μM and a detection limit (LOD) of 51 nM. The proposed colorimetric sensor was successfully applied for the detection of different concentrations of dopamine in spiked fetal bovine serum (FBS) and horse serum (HS) samples. It is anticipated that Bi2Fe4O9 nanozyme holds great potential in biomedical analysis and diagnostic applications of dopamine-related diseases.

Abstract

International audience; Over the last years, we observed a significant increase in the number of published studies that focus on the synthesis and characterization of deep eutectic solvents (DESs). These materials are of particular interest mainly due to their physical and chemical stability, low vapor pressure, ease of synthesis, and the possibility of tailoring their properties through dilution or change of the ratio of parent substances (PS). DESs, considered as one of the greenest families of solvents, are used in many fields, such as organic synthesis, (bio)catalysis, electrochemistry, and (bio)medicine. DESs applications have already been reported in various review articles. However, these reports mainly described these components’ basics and general properties without focusing on the particular, PS-wise, group of DESs. Many DESs investigated for potential (bio)medical applications comprise organic acids. However, due to the different aims of the reported studies, many of these substances have not yet been investigated thoroughly, which makes it challenging for the field to move forward. Herein, we propose distinguishing DESs comprising organic acids (OA-DESs) as a specific group derived from natural deep eutectic solvents (NADESs). This review aims to highlight and compare the applications of OA-DESs as antimicrobial agents and drug delivery enhancers—two essential fields in (bio)medical studies where DESs have already been implemented and proven their potential. From the survey of the literature data, it is evident that OA-DESs represent an excellent type of DESs for specific biomedical applications, owing to their negligible cytotoxicity, fulfilling the rules of green chemistry and being generally effective as drug delivery enhancers and antimicrobial agents. The main focus is on the most intriguing examples and (where possible) application-based comparison of particular groups of OA-DESs. This should highlight the importance of OA-DESs and give valuable clues on the direction the field can take.

Num período onde a transição digital é evidente em várias áreas do conhecimento, é imprescindível que o ensino acompanhe esta evolução e que implemente, num ambiente controlado, as ferramentas tecnológicas que contribuam para aumentar o interesse dos alunos e também a consolidação das suas aprendizagens. Uma dessas ferramentas é justamente a gamificação. A implementação de gamificação em sala de aula encontra-se numa fase de pleno desenvolvimento, mas é alvo de muita resistência devido ao seu carácter lúdico e por vezes exigente a nível técnico. No entanto, são vários os exemplos onde o seu uso resulta num maior interesse e motivação por parte dos alunos. Apesar de existirem exemplos de aplicabilidade, especialmente no caso da matemática, são ainda raros ou pouco estruturados os exemplos na disciplina de geografia. Este trabalho, através da metodologia adotada na Prática de Ensino Supervisionada pretende demonstrar que a implementação de jogos digitais pode constituir um recurso útil na aprendizagem dos alunos e na sua motivação, através de uma solução harmoniosa enquadrando a componente didática com os conteúdos programáticos. Para além do desenvolvimento de jogos, foi implementado um questionário aos alunos onde eram encorajados a avaliar e a providenciar feedback quanto à utilização de gamificação ao longo do ano letivo como também a sugerir metodologias e aplicações passíveis de serem aplicadas. In a period where the digital transition is clear in several areas of knowledge, it is essential that teaching keeps up with this evolution and implements, in a controlled environment, technological tools that contribute to increase student interest and the consolidation of their learning. One of these tools is precisely gamification. The implementation of gamification in the classroom is in a stage of full development but is subject to much resistance due to its playful and sometimes technically demanding nature. However, there are several examples where its use results in increased student interest and motivation. Although there are examples of applicability, especially in the case of mathematics, examples in geography are still rare or poorly structured. This work, through the methodology adopted in the Supervised Teaching Practice, aims to demonstrate that the implementation of digital games can be a useful resource in student learning and motivation, through a harmonious solution framing the didactic component with the syllabus. In addition to the development of games, a questionnaire was given to students to encourage them to evaluate and provide feedback on the use of gamification throughout the school year, and to suggest methodologies and applications that could be applied.

Abstract

International audience; Pancreatic islet amyloid deposition is a pathological hallmark of Type 2 diabetes (T2D), contributing to reduced functional β-cell mass. Islet amyloids result not only from the aggregation and fibrillation of human islet amyloid polypeptide (hIAPP), but also from beta-amyloid 42 (Aβ42), the key amyloidogenic peptide linked to Alzheimer's disease. Importantly, Aβ42 and hIAPP aggregates (IAPP:Aβ42) can interact with each other and form some harmful heterocomplex fibrils. While it is well-documented that hIAPP aggregation occurs only when islets are exposed to a diabetic environment, including hyperglycemia and/or elevated concentrations of saturated fatty acids (SFAs), it remains unclear if hIAPP and IAPP:Aβ42 heteromer fibrillations are directly or indirectly triggered by this environment. In this study, we show the interplay between high glucose concentrations and palmitate as the SFA in the aggregation of hIAPP. In addition, we outline that the interaction of hIAPP and Aβ42 leads to the formation of complex protein aggregates, which are toxic to β-cells. Carbon nanocolloids in the form of positively charged carbon quantum dots (CQD-pos) efficiently prevent single amyloid aggregation and the formation of IAPP:Aβ42 heterocomplexes. We provide clear evidence with this study that the diabetogenic environment of islets could directly contribute to the formation of homomeric and heteromeric amyloid aggregates and fibrils in T2D. We also propose carbon nanocolloids as biocompatible nanomaterials for developing innovative therapeutic strategies that prevent the decline of functional β-cell mass.

Abstract

International audience; In the fight against prostate cancer (PCa), TRPM8 is one of the most promising clinical targets. Indeed, several studies have highlighted that TRPM8 involvement is key in PCa progression because of its impact on cell proliferation, viability, and migration. However, data from the literature are somewhat contradictory regarding the precise role of TRPM8 in prostatic carcinogenesis and are mostly based on in vitro studies. The purpose of this study was to clarify the role played by TRPM8 in PCa progression. We used a prostate orthotopic xenograft mouse model to show that TRPM8 overexpression dramatically limited tumor growth and metastasis dissemination in vivo. Mechanistically, our in vitro data revealed that TRPM8 inhibited tumor growth by affecting the cell proliferation and clonogenic properties of PCa cells. Moreover, TRPM8 impacted metastatic dissemination mainly by impairing cytoskeleton dynamics and focal adhesion formation through the inhibition of the Cdc42, Rac1, ERK, and FAK pathways. Lastly, we proved the in vivo efficiency of a new tool based on lipid nanocapsules containing WS12 in limiting the TRPM8–positive cells’ dissemination at metastatic sites. Our work strongly supports the protective role of TRPM8 on PCa progression, providing new insights into the potential application of TRPM8 as a therapeutic target in PCa treatment.

Abstract

Coronaviruses share conservative spike protein (S) on their enveloped membrane surface, where S1 subunit recognizes and binds the cellular receptor, and the S2 subunit mediates membrane fusion. This similarity raises the question: does coronaviral infection by one create protection to others? Convalescent SARS-CoV-2 (COVID-19) sera were tested for cross reactivity with peptides from Middle East respiratory syndrome coronavirus (MERS-CoV) which shares 74% homology. Our results showed significant cross-reactivity with a peptide of the heptad repeat 2 (HR2) domain of the MERS-CoV spike protein. Abstract

Bioactive aminooxime ligands based on optically pure (R)-limonene have been synthesized in two steps. Their ruthenium (II) cationic water-soluble complex was prepared by a reaction between dichloro (para-cymene) ruthenium (II) dimers and aminooxime ligands in a 1:2 molar ratio. Antibacterial and antibiofilm activities of the synthetized complex were assessed against Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, and Enterococcus faecalis. The results revealed that the ruthenium (II) complex has higher antibacterial and antibiofilm activities in comparison with free ligands or the enantiopure (R)-limonene. Moreover, microencapsulation of this complex reduced its cytotoxicity and improved their minimum inhibitory concentration and antibiofilm activity toward the considered bacteria. The ruthenium (II) complex targets the bacterial cell membrane, which leads to rapid leakage of intracellular potassium. Our study suggests that the developed ruthenium (II) complexes could be useful as an alternative to conventional disinfectants.

Abstract

In this contribution, we report a facile functionalization method of carbon dots (CDs) by tetraalkylammonium moieties using diazonium chemistry to improve their antibacterial activity against Gram-positive and Gram-negative bacteria. CDs were modified by novel diazonium salts bearing tetraalkylammonium moieties (TAA) with different alkyl chains (C2, C4, C9, C12) for the optimization of antibacterial activity. The functionalized CDs were characterized by X-ray photoelectron spectroscopy (XPS), Fourier-transform infrared (FTIR) spectroscopy, ultraviolet–visible (UV-vis) absorption spectrophotometry and fluorescence spectroscopy to confirm the covalent binding of TAA onto the CDs’ surface. Variation of the alkyl chain allows to reach the strongest antibacterial effect for CDs-C9 towards Gram-positive S. aureus (MIC 3.09±1.10 µg mL-1) and Gram-negative E. coli (MIC 7.93±0.17 µg mL-1) bacteria. In addition, the antibiofilm properties of the CDs-C9 were assessed; full biofilm inhibition was achieved after 6 h (64 µg mL-1) and 2 h (128 µg mL-1) treatment for S. aureus biofilm and >60% suppression of biofilm mass after 6 h (128 µg mL-1) for E. coli biofilm. CDs-C9 demonstrated good biocompatibility on mouse fibroblast (NIH/3T3), HeLa and U-87 MG cell lines for concentrations up to 256 µg mL-1. Collectively, our work highlights the correlation between the surface chemistry of CDs and their antimicrobial performance.

Abstract

International audience; Carob waste (CW) is an agro-biomass material abundant in nature with potential use for eco-friendly remediation. However, like many biomass-based adsorbents, it suffers from its low adsorption capacity for organic/inorganic pollutants. Therefore, modification using physical and/or chemical means is commonly applied to improve the adsorptive properties of biomass-based adsorbents. In this study, carob waste (CW) and carob waste functionalized with phytic acid (PA-CW), as an ecofriendly product, were applied for the first time for Cr(VI) elimination. Various methods were applied for the material characterization like Fourier-transform infrared spectroscopy, powder X-ray diffraction (PXRD), thermogravimetric analysis (TGA and DTG), X-ray photoelectron spectroscopy (XPS), specific surface area and porosity measurements. The results proved that both CW and PA-CW own appropriate features for efficient adsorption. Bach experiments revealed that the optimum parameters for Cr(VI) (100 mg/mL) removal at 25 °C were pH 2, 0.05 and 0.025 g as adsorbent dose for CW and PA-CW, respectively, over 120 min contact time. The kinetic of adsorption was well-described by the pseudo-second order model, whereas the isotherm modeling fitted well the modified Langmuir model. CW and PA-CW achieved respectively maximum adsorption capacities of 212.4 and 387.9 mg/g, which are among the highest values so far reported for biomass-based adsorbent materials. These results confirmed that CW and PA-CW could be alternative cost-effective adsorbents even for high concentrations of Cr(VI) in industrial wastewaters along with their reduction capacity.

Abstract

International audience; Vaccination remains one of the most effective tools to prevent infectious diseases. To ensure that the best possible antigenic components are chosen to stimulate a cognitive immune response, boosting antigen presentation using adjuvants is common practice. Nanodiamond-based adjuvants are proposed here as a rapid and versatile platform for antigen conjugation, utilizing peptides common to different pathogenic strains and making this strategy a good candidate for a “ready-to-use” vaccine. Initiation of an inflammatory reaction with a resulting immune response is based on the ability of living organisms to entrap nanostructures such as nanodiamonds with neutrophil extracellular traps (NETs) formation. In this work, coronavirus peptide homological for MERS-CoV, fusion inhibitor, was conjugated to nanodiamonds and used to induce neutrophilic-driven self-limiting inflammation. The resulting adjuvant was safe and did not induce any tissue damage at the site of injection. Mice immunization resulted in IgG titers of ¼,000 within 28 days. Immunization of rabbits resulted in the formation of a high level of antibodies persistently present for up to 120 days after the first immunization (animal lifespan ~3 years). The peptide used for immunization proved to be reactive with sera of convalescent COVID patients, demonstrating the possibility of developing pancoronaviral vaccine candidates.

Abstract

International audience; The low mass activity and high price of pure platinum (Pt)-based catalysts predominantly limit their large-scale utilization in electrocatalysis. Therefore, the reduction of Pt amount while preserving the electrocatalytic efficiency represents a viable alternative. In this work, we prepared new PtRu2 nanoparticles supported on sulphur and nitrogen co-doped crumbled graphene with trace amounts of iron (PtRu2/PF) electrocatalysts. The PtRu2/PF catalysts exhibited enhanced electrocatalytic performance and stability for the hydrogen evolution reaction (HER) at pH - 0. Moreover, the prepared PtRu2/PF electrocatalyst displayed higher HER activity than commercial 20% Pt/C. The PtRu2/PF catalyst achieved a current density of 10 mA cm-2 at an overpotential value of only 22 mV for HER, performing better activity than many other Pt-based electrocatalysts. Besides, the PtRu2/PF revealed a good performance for the oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) in alkaline media. The PtRu2/PF catalyst recorded a current density of 10 mA cm-2 at an overpotential of only 270 mV for OER in KOH (1.0 M) solution and an onset potential of 0.96 V vs. RHE (at 1 mA cm-2) for ORR in KOH (0.1 M) solution.

Abstract

International audience; This paper reports on enzyme-like catalytic properties of polyethylene glycol-functionalized poly(N-phenylglycine) (PNPG-PEG) nanoparticles, which have not been explored to date. The developed nanoparticles have the ability to display great inherent peroxidase-like activity at very low concentrations, and are able to catalyze the oxidation of 3,3',5,5'tetramethylbenzidine (TMB) substrate in presence of hydrogen peroxide (H2O2). The oxidized product of TMB has a deep blue color with a maximum absorbance at ~655 nm. The PNPG-PEG nanoparticles exhibit Km values of 0.2828 for TMB and 0.0799 for H2O2, indicating that TMB oxidation takes place at lower concentration of H2O2 in comparison to other nanozymes. Based on the known mechanism of H2O2 oxidation by hexavalent chromium [Cr(VI)] ions to generate hydroxyl radicals (.OH), these nanoparticles were successfully applied for the colorimetric sensing of Cr(VI) ions. The sensor achieved good performance for Cr(VI) sensing with detection limits of 0.012 μM (0.01-0.1 μM linear range) and 0.52 μM (0.05-12.5 μM linear range). The detection scheme was highly selective, and successfully applied for the detection of Cr(VI) in real water samples.

Abstract

International audience; The contamination of water by synthetic organic molecules and trace metals is a growing challenge, in spite of the enormous research efforts being made in the field of water treatment. In this study, reduced graphene oxide-copper sulphide (rGO-CuS) nanocomposites of different rGO/CuS (2/1, 1/1, 1/2) molar ratios were fabricated via a facile one-step hydrothermal method. The nanocomposite materials, named hereafter as 2rGO-CuS, rGO-CuS and rGO-2CuS, were characterized using various analytical techniques, including X-ray diffraction (XRD), Raman spectroscopy, transmission electron microscopy (TEM), energy dispersive X-ray (EDX) spectroscopy, X-ray photoelectron spectroscopy (XPS) and UV-visible spectrophotometry. The photocatalytic performance of the nanocomposites was assessed under visible light irradiation (λ > 420 nm) for the simultaneous photocatalytic reduction of Cr(VI) and phenol degradation. It was found that rGO-2CuS achieved a remarkable enhancement of the photocatalytic activity among the prepared nanocomposites for the degradation of phenol and reduction of Cr(VI). Therefore, the simultaneous photocatalytic phenol degradation and Cr(VI) reduction over rGO-2CuS sample was further investigated. The experimental results revealed that rGO-2CuS catalyst maintained good degradation efficacy of mixed pollutants after 6 runs and dissolved oxygen was found to be essential to promote Cr(VI) reduction and phenol degradation. A detailed photocatalytic activity under visible light irradiation mechanism was proposed based on quenching experiments and fluorescence measurements.

Abstract

International audience; Monitoring blood glucose levels for diabetic patients is critical to achieve tight glycaemic control. As none of the current antidiabetic treatments restore lost functional β-cell mass in diabetic patients, insulin injections and the use of insulin pumps are most widely used in the management of glycaemia. The use of advanced and intelligent chemical engineering, together with the incorporation of micro- and nanotechnological-based processes have lately revolutionized diabetic management. The start of this concept goes back to 1974 with the description of an electrode that repeatedly measures the level of blood glucose and triggers insulin release from an infusion pump to enter the blood stream from a small reservoir upon need. Next to the insulin pumps, other drug delivery routes, including nasal, transdermal and buccal, are currently investigated. These processes necessitate competences from chemists, engineers-alike and innovative views of pharmacologists and diabetologists. Engineered micro and nanostructures hold a unique potential when it comes to drug delivery applications required for the treatment of diabetic patients. As the technical aspects of chemistry, biology and informatics on medicine are expanding fast, time has come to step back and to evaluate the impact of technology-driven chemistry on diabetics and how the bridges from research laboratories to market products are established. In this review, the large variety of therapeutic approaches proposed in the last five years for diabetic patients are discussed in an applied context. A survey of the state of the art of closed-loop insulin delivery strategies in response to blood glucose level fluctuation is provided together with insights into the emerging key technologies for diagnosis and drug development. Chemical engineering strategies centered on preserving and regenerating functional pancreatic β-cell mass are evoked in addition as they represent a permanent solution for diabetic patients.

International audience; Aims: In this study, benzalkonium chloride (BAC) microcapsules were developed for surface disinfection purpose and were evaluated against Listeria monocytogenes and Escherichia coli biofilms.Methods and Results: Microcapsules were prepared with two different strategies: uncomplexed BAC‐microcapsules (UBM), containing BAC and maltodextrins and complexed BAC‐microcapsules (CBM) containing BAC complexed by pectin and maltodextrins. The minimum inhibitory concentrations (MICs) of free and microencapsulated BAC was investigated against two food pathogens: L. monocytogenes and E. coli. The antibiofilm activities of UBM and CBM against L. monocytogenes and E. coli biofilms formed on stainless steel at 37°C were evaluated and compared to BAC used under its free form. MICs of encapsulated BAC were up to 4‐fold lower than those of free BAC. The UBM and CBM showed higher antibiofilm effect when compared to the free BAC.Conclusions: Overall, results demonstrated that microencapsulation enhanced the antibacterial activity of BAC against L. monocytogenes and E. coli biofilms.Significance and Impact of the Study: The application of such BAC microcapsule‐based delivery systems can improve surface disinfection procedures and reduce the required BAC concentrations and the related cytotoxicity of this antimicrobial compound.

Abstract

International audience; A novel nanosensor with peroxidase enzyme-mimetic activity, based on CuS-BSA-Cu3(PO4)2 nanoparticles, was developed. CuS-BSA nanoparticles were first synthesized using a facile bio-mineralization assay. Conjugation of Cu3(PO4)2 with CuS-BSA generates CuS-BSA-Cu3(PO4)2 nanoparticles (NPs) of 10 nm in size with high catalytic activity against a peroxidase substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The catalytic action was based on a remarkable color change from colorless TMB into blue oxidized product (oxTMB) with absorption maximum at 654 nm. The enzyme-mimetic activity of CuS-BSA-Cu3(PO4)2 nanoparticles was believed to occur through hydroxyl radical (HOradical dot) generation in presence of H2O2, which was inhibited upon addition of dopamine. Increasing concentrations of dopamine induced a gradual decrease of the nanoparticles' catalytic activity. The developed colorimetric sensor displayed a limit of detection of 0.13 µM for dopamine over 0.05-100 µM linear range and high specificity. The performance of the nanosensor for sensing dopamine in beef meat and blood samples was evaluated and proved to be promising for diagnostic applications without the requirement of complex and expensive instrumentation.

Abstract

International audience; There has been significant interest in the use of peptides as antimicrobial agents, and peptide containing hydrogels have been proposed as biological scaffolds for various applications. Limited stability and rapid clearance of small molecular weight peptides pose challenges to their widespread implementation. As a common approach, antibacterial peptides are physically loaded into hydrogel scaffolds, which leads to continuous release through the passive mode with spatial control but provides limited control over drug dosage. Although utilization of peptide covalent linkage onto hydrogels addresses partially this problem, the peptide release is commonly too slow. To alleviate these challenges, in this work, maleimide-modified antimicrobial peptides are covalently conjugated onto furan-based cryogel (CG) scaffolds via the Diels-Alder cycloaddition at room temperature. The furan group offers a handle for specific loading of the peptides, thus minimizing passive and burst drug release. The porous nature of the CG matrix provides rapid loading and release of therapeutic peptides, apart from high water uptake. Interfacing the peptide adduct containing a CG matrix with a reduced graphene oxide-modified Kapton substrate allows “on-demand” photothermal heating upon near-infrared (NIR) irradiation. A fabricated photothermal device enables tunable and efficient peptide release through NIR exposure to kill bacteria. Apart from spatial confinement offered by this CG-based bandage, the selective ablation of planktonic Staphylococcus aureus is demonstrated. It can be envisioned that this modular “on-demand” peptide-releasing device can be also employed for other topical applications by appropriate choice of therapeutic peptides.

However, the sluggish and complex electro-oxidation of methanol is one of the limiting factors for any practical application. To solve this issue, the use of plasmonic is considered as a promising way to accelerate the methanol oxidation reaction. In this study, we report on a novel approach for achieving enhanced methanol oxidation currents. Perforated gold thin film anodes were decorated with Pt/Ru via electrochemical deposition and investigated for their ability for plasmon-enhanced electrocatalytic methanol oxidation in alkaline media. The novel methanol oxidation anode (AuNHs/PtRu), combining the strong light absorption properties of a gold nanoholes array-based electrode (AuNHs) with surface-anchored bimetallic Pt/Ru nanostructures, known for their high activity toward methanol oxidation, proved to be highly efficient in converting methanol via the hot holes generated in the plasmonic electrode. Without light illumination, AuNHs/PtRu displayed a maximal current density of 13.7 mA/cm2 at -0.11 V vs Ag/AgCl. Enhancement to 17.2 mA/cm2 was achieved under 980 nm laser light illumination at a power density of 2 W/cm2. The thermal effect was negligible in this system, underlining a dominant plasmon process. Fast generation and injection of charge carriers were also evidenced by the abrupt change in the current density upon laser irradiation. The good stability of the interface over several cycles makes this system interesting for methanol electro-oxidation.

Abstract

In this study, graphene oxide (GO), polyethyleneimine (PEI) and potassium hydroxide (KOH) were used to synthesize reduced graphene oxide (rGO/PEI-KOH) nanocomposite. The presence and grafting of PEI molecules on the reduced GO surface were assessed by X-ray photoelectron spectroscopy (XPS) and Fourier transform infrared (FTIR) analyses. The rGO/PEI-KOH nanocomposite was successfully applied for hexavalent chromium, Cr(VI), wastewater elimination. The resulting rGO/PEI-KOH adsorbent was found to be highly effective for Cr(VI) removal at low pH values and achieved a maximum capacity of experimental adsorption of 398.9 mg/g, which is one of the highest sorption capacity of most GO- and PEI-based adsorbents reported in the literature up to date. Studying the adsorption mechanism, the sorption isotherm revealed that the modified-Langmuir model was the best fit and Cr(VI) removal follows a pseudo-second-order kinetics, with the predominance of intraparticle diffusion during the first step of adsorption. XPS analysis indicated the presence of appreciable amount of Cr(III) on the adsorbent surface, which suggests that the adsorbed Cr(VI) ions were effectively reduced to Cr(III) on the rGO/PEI-KOH adsorbent surface (∼70% of the total adsorbed Cr). Cr(VI) adsorption and subsequent reduction to Cr(III) both contributed to the Cr(VI) removal. The results of the present study highlight the benefits of rGO/PEI-KOH like low cost, environmentally friendly, large toxic Cr(VI) ions adsorption capacity and its effective reduction to less-toxic Cr(III).

Abstract

The study reports on the preparation of boron-doped diamond nanospikes (BDD-NSs), supported on porous titanium (Ti) substrate, using reactive ion etching (RIE) with oxygen plasma and their investigation as an electrode material for supercapacitors. The above architecture contributes to an ideal pathway for supercapacitors with good stability and high specific capacitance. As a result, the BDD electrode etched for 5 min (BDD-NSs-5) achieved good performance with a high specific capacitance of 70.6 mF cm−2 at a current density of 0.32 mA cm−2 and a capacitance retention of 84.3% after 15,000 cycles. Moreover, the BDD-NSs-5 electrode was assembled into a symmetric supercapacitor device. The device achieved high energy and power densities of 9.24 mJ cm−2 and 1.26 mW cm−2, respectively, implying enhanced potential for real-time applications. For practical application, the BDD-NSs-5 electrode was used for rotating a home-designed windmill device and the solid-state symmetric supercapacitor device can light up a red LED for 20 s after being charged to 3.0 V.

Abstract

International audience; Herein, we investigated the influence of Fe, Cr and Al incorporation in Ni-based layered double hydroxides (LDHs) supported on nickel foam (NF) electrodes, prepared through a one-step hydrothermal, on their electrochemical performance. The resulting NiFe, NiCr and NiAl (LDHs) were further coated on carbon spheres (CS) supported on NF using a facile two-step hydrothermal process to produce NiFe LDHs@CS/NF, NiCr LDHs@CS/NF and NiAl LDHs@CS/NF. The performance of the prepared materials as binder-free electrodes in supercapacitors was assessed. Among all the prepared electrodes, NiAl LDHs@CS/NF electrode material achieved the largest areal capacity (1042.2 mC cm−2 at 1 mA cm−2), as compared to the areal capacity values attained by NiFe LDHs@CS/NF (705.8 mC cm−2) and NiCr LDHs@CS/NF (814.9 mC cm−2) at 1 mA cm−2. Therefore, a hybrid supercapacitor device comprising NiAl LDHs@CS/NF as the positive electrode and N-doped reduced graphene/NF as the negative electrode was successfully fabricated. The device exhibited favorable flexibility, good mechanical properties and stability; the areal capacity remained ∼75% and ∼67% of the original value after 5000 and 10,000 cycles, respectively. The hybrid supercapacitor attained an energy density of 43 μWh cm−2 at a power density of 0.805 mW cm−2 and was applied successfully to operate a home-made windmill device continuously for 32 s. Moreover, two flexible NiAl LDHs@CS/NF//N-rGO/NF hybrid supercapacitors, connected in series, were able to light up a green, a red and a yellow LED in parallel, lasting for 37 s, 542 s and 199 s, respectively, indicating their potential application for flexible energy storage devices.

Abstract

Coronaviruses are sharing several protein regions notable the spike protein (S) on their enveloped membrane surface, with the S1 subunit recognizing and binding to the cellular receptor, while the S2 subunit mediates viral and cellular membrane fusion. This similarity opens the question whether infection with one coronavirus will confer resistance to other coronaviruses? Investigating patient serum samples after SARS-CoV-2 infection in cross-reactivity studies of immunogenic peptides from Middle East respiratory syndrome coronavirus (MERS-CoV), we were able to detect the production of antibodies also recognizing MERS virus antigens. The cross-reactive peptide comes from the heptad repeat 2 (HR2) domain of the MERS virus spike protein. Indeed, the peptide of the HR2 domain of the MERS spike protein, previously proven to induce antibodies against MERS-CoV is sharing 74% homology with the corresponding sequence of SARS-CoV-19 virus. Sera samples of 47 convalescent SARS-CoV-2 patients, validated by RT-PCR-negative testes 30 days post-infection, and samples of 40 sera of control patients (not infected with SARS-CoV-2 previously) were used to establish eventual cross-bind reactivity with the MERS peptide antigen. Significantly stronger binding (p< 0.0001) was observed for IgG antibodies in convalescent SARS-CoV-2 patients compared to the control group. If used as an antigen, the peptide of the HR2 domain of the MERS spike protein allows discrimination between post-Covid populations from non-infected ones by the presence of antibodies in blood samples. This suggests that polyclonal antibodies established during SARS-CoV-2 infection has the ability to recognize and probably decrease infectiveness of MERS-CoV infections as well as other coronaviruses. The high homology of the spike protein domain suggests in addition that the opposite effect can also be true: coronaviral infections producing cross-reactive antibodies affective against SARS-CoV-19. The collected data prove in addition that despite the core HR2 region being hidden in the native viral conformation, its exposure during cell entry makes it highly immunogenic. Since inhibitory peptides to this region were previously described, this opens new possibilities in fighting coronaviral infections.

Abstract

The electrochemical production of hydrogen is enjoying renewed vigor due to its great promise as an environmentally friendly energy alternative. Herein, we demonstrate the capacity of nanoperforated gold films deposited on flexible substrates coated with nanometer thick Ti/TiO2 adhesion layers for electrochemical water splitting under light irradiation. The strong plasmonic induced electromagnetic field enhancement, which occurs under the illumination of plasmonic electrodes, facilitates the dissociation of water into hydrogen and improves the electrocatalytic hydrogen evolution activity beyond that of platinum. Under illumination at 980 nm at 2 W cm-2, a current density of -10 mA cm-2 was recorded at a low overpotential of h z -0.045 V (vs. RHE); under the same conditions, a Pt electrode achieved -10 mA cm-2 at h z -0.048 V (vs. RHE). Mechanistic investigations revealed that plasmon-excited perforated gold thin layers are an efficient source of hot electrons, contributing to their injection into the underlying TiO2-based adhesion layer. Direct electrochemical reduction of water appears to be a parallel reaction pathway to sustain the hydrogen evolution reaction on the interface. Both reaction pathways decrease the activation energy for the hydrogen evolution reaction similar to the benchmark Pt electrocatalyst.

Abstract

This work reports on the synthesis of organic-inorganic hybrid nanoscale materials, CuS-BSA-Cu-3(PO4)(2). The CuS-BSA-Cu-3(PO4)(2) were successfully applied as artificial colorimetric probes in sensing H2O2, the final outcome of glucose oxidation, and proved to be efficient peroxidase mimics for the catalytic conversion of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB), into a blue colored oxidized product (oxTMB) which can be easily visualized by the naked eye and monitored by a great absorption peak at 654 nm in the UV-vis spectrophotometry. A highly efficient, rapid, sensitive, and selective determination of H2O2 and glucose have been achieved with very low detection limits of 22 nM, and 27.6 nM over 0-8 mu M and 0-1000 mu M linear ranges, respectively. Compared to CuS-BSA, CuS-BSA-Cu-3(PO4)(2) exhibited improved peroxidase-like catalytic activity. Abstract

Regular problems faced due to chemical leaks and oil spills have caused tremendous threats to lakes and sea water. Therefore, there is an urgent need to develop efficient strategies to remove oil and organic pollutants from water surfaces. The aim of this study is to develop a high performance magnetic, superhydrophobic/superoleophilic three-dimensional porous composite material for efficient and selective adsorption of organic pollutants from water. The composite material was fabricated by simple immersion of a commercially available polyurethane (PU) sponge into a solution of high-density polyethylene (HDPE) containing pre-synthesized magnetic (Fe3O4) nanoparticles. The composite material exhibited a water contact of ~155° and an oil contact angle of ~0°. The presence of the Fe3O4 particles allowed magnetic-controlled elimination of underwater oil, while the superhydrophobic character of the functionalized PU sponge permitted efficient separation of oil/water mixtures and demulsification of toluene/water emulsions. Moreover, the HDPE coating not only firmly immobilized the magnetic particles, but also contributed to the excellent stability of the composite sponge which withstood acid, base, salts, seawater or temperature variation from −20 °C to 105 °C. In addition, the composite material maintained its oil adsorption ability over 10 cycles. The ease of fabrication of the magnetic super wetting 3D material sponge along with its durability and reusability makes it an interesting material with potential for practical applications.

Abstract

International audience; In the last years, carbon-based nanomaterials have attracted considerable attention in a wide range of fields, particularly in biomedicine, owing to their remarkable photo-physical and chemical properties. In this study, we demonstrate that amine-terminated carbon dots (CDs-NH2) functionalized with ampicillin (AMP) offer a new perspective for antibacterial treatment. The amine-functionalized carbon dots were used as a carrier for immobilization and delivery of ampicillin (CDs-AMP) and as a visible light-triggered antibacterial material. Additionally, AMP immobilization on the CDs-NH2 surface improves its stability in solution as compared to free AMP. The AMP conjugated CDs platform combines the antibacterial function of AMP and conserves the intrinsic theranostic properties of CDs-NH2. Therefore, the AMP immobilized onto CDs-NH2 surface together with the generation of moderate quantities of reactive oxygen species under visible light illumination is very effective to inactivate the growth of Escherichia coli.

Abstract

In the present work, a cobalt phthalocyanine-supported reduced graphene oxide material (rGO-CoPc) was used as a catalyst for the degradation of rhodamine B (RhB) and pentachorophenol (PCP) in the presence of peroxymonosulfate (PMS). Inductively coupled plasma (ICP-OES), Raman spectroscopy, Fourier transform infrared (FTIR) spectroscopy, X-ray photoelectron spectroscopy (XPS), thermogravimetric analysis (TGA), and scanning electron microscopy (SEM) were employed to characterize the properties of the composite and Co loading in the material. The catalytic activity of the rGO1-CoPc1 catalyst (rGO:CoPc = 1:1) was evaluated by UV–vis absorption spectroscopy at varying PMS concentrations and catalyst dosages. The complete dye removal was achieved in 9 min by rGO1-CoPc1/PMS system with a high initiation rate constant of 1.019 min−1 using RhB (25 µM), PMS (0.1 mM) and rGO1-CoPc1 (0.5 mg/mL). The removal efficiency was found to be as high as 100% after 11 cycles, suggesting a long-lasting catalytic activity of the of rGO1-CoPc1/PMS system. Quenching experiments suggested that sulfate (SO4 −) radicals are the dominant species in the degradation process although hydroxyl (OH ) radicals are also involved in the reaction mechanism. Furthermore, the catalyst was successfully applied for the efficient degradation of pentachlorophenol (PCP). A full degradation of PCP (25 µM) was achieved within 20 min using rGO1-CoPc1 (0.5 mg/mL) and PMS (0.6 mM). This study demonstrated that the use of rGO-CoPc as a heterogeneous catalyst for PMS activation is useful for the advanced oxidation processes (AOP) for practical wastewater treatment.

Abstract

International audience; A highly efficient, recyclable and magnetically separable core-shell structured CuZnO@Fe3O4 microsphere wrapped with reduced graphene oxide (rGO@CuZnO@Fe3O4) photocatalyst has been developed and used for the photoreduction of carbon dioxide with water to produce methanol under visible light irradiation. Owing to the synergistic effect of the components and to the presence of a thin Fe2O3 layer on Fe3O4, rGO@CuZnO@Fe3O4 4 exhibited higher catalytic activity as compared to the other possible combinations such as CuZnO@Fe3O4 2 and GO@CuZnO@Fe3O4 3 microspheres. The yield of methanol in case of using 2 and 3 as photocatalyst was found to be 858 and 1749 μmol g−1 cat, respectively. However, the yield was increased to 2656 μmol g−1 cat when rGO@CuZnO@Fe3O4 4 was used as photocatalyst under similar experimental conditions. This superior photocatalytic activity of 4 was assumed to be due to the restoration of the sp2 hybridized aromatic system in rGO, which facilitated the movement of electrons and resulted in better charge separation. The synthesized heterogeneous photocatalyst could readily be recovered by external magnet and successfully reused for six subsequent cycles without significant loss in the product yield.

Abstract

Therapeutic options for the highly pathogenic human coronavirus (HCoV) infections are urgently needed. Anticoronavirus therapy is however challenging, as coronaviruses are biologically diverse and rapidly mutating. In this work, the antiviral activity of seven different carbon quantum dots (CQDs) for the treatment of human coronavirus HCoV-229E infections was investigated. The first generation of antiviral CQDs was derived from hydrothermal carbonization of ethylenediamine/citric acid as carbon precursors and postmodified with boronic acid ligands. These nanostructures showed a concentration-dependent virus inactivation with an estimated EC50 of 52 ± 8 μg mL-1. CQDs derived from 4-aminophenylboronic acid without any further modification resulted in the second-generation of anti-HCoV nanomaterials with an EC50 lowered to 5.2 ± 0.7 μg mL-1. The underlying mechanism of action of these CQDs was revealed to be inhibition of HCoV-229E entry that could be due to interaction of the functional groups of the CQDs with HCoV-229E entry receptors; surprisingly, an equally large inhibition activity was observed at the viral replication step.

Abstract

International audience; Drug loaded hydrogels have proven to be versatile controlled-release systems. We report here on heat active hydrogel formation by mixing graphene oxide (GO) or carboxyl enriched reduced graphene oxide (rGO-COOH) with metformin hydrochloride, an insulin sensitizer drug currently used as the first line therapy to treat patients with type 2 diabetes. The driving forces of the gelation process between the graphene-based nanomaterial and metformin are hydrogen bonding and electrostatic interactions, weakened at elevated temperature. Using the excellent photothermal properties of the graphene matrixes, we demonstrate that these supramolecular drug reservoirs can be photothermally activated for transdermal metformin delivery. A sustained delivery of metformin was achieved using a laser power of 1 W cm−2. In vitro assessment of the key target Glucose-6 Phosphatase (G6P) gene expression using a human hepatocyte model confirmed that metformin activity was unaffected by photothermal activation. In vivo, metformin was detected in mice plasma at 1 h post-activation of the metformin loaded rGO-COOH gel.

Abstract

A mandatory step in any sensor fabrication is the introduction of analyte-specific recognition elements to the transducer surface. In this study, the possibility to anchor β-cyclodextrin-modified dopamine to a reduced graphene oxide based electrochemical transducer for the sensitive and selective sensing of folic acid is demonstrated. The sensor displays good electrocatalytic activity toward the oxidation of folic acid. The strong affinity of the surface-confined β-cyclodextrin for folic acid, together with favorable electron transfer characteristics, resulted in a sensor with a detection limit of 1 nM for folic acid and a linear response up to 10 μM. Testing of the sensor on serum samples from healthy individuals and patients diagnosed with folic acid deficiency validated the sensing capability. Graphical abstract.

Abstract

The aim of the present work was to study and compare the effect of growth temperature (20, 30, and 37 degrees C) and surface type (stainless steel and polycarbonate) on the production of virulence factors, such as proteases and siderophores, and the risk of surface contamination associated with Pseudomonas aeruginosa biofilm and planktonic cells. The increase of growth temperature from 20 to 37 degrees C increased (approximately twofold) the electronegative charge and the hydrophobicity of the P. aeruginosa biofilm cell surface. P. aeruginosa biofilm cell adhesion to stainless steel and polycarbonate was 5- and 1.5-fold higher than their planktonic counterparts at 20 and 30 degrees C, respectively. The increase of growth temperature from 20 to 37 degrees C increased the production of proteases (twofold) and siderophores (twofold) and the cytotoxicity (up to 30-fold) against the HeLa cell line in the supernatants of P. aeruginosa planktonic and biofilm cultures. This study also highlighted that biofilm and planktonic P. aeruginosa cells exhibited distinct physiological properties with respect to the production of virulence factors and the cytotoxicity against the Hela cell line. Therefore, effective disinfection procedures should be adapted to inactivate bacteria detached from biofilms.
