Abstract: KM3NeT/ORCA is an underwater neutrino telescope under construction in the Mediterranean Sea. Its primary scientific goal is to measure the atmospheric neutrino oscillation parameters and to determine the neutrino mass ordering. ORCA can constrain the oscillation parameters $\Delta m^{2}_{31}$ and $\theta_{23}$ by reconstructing the arrival direction and energy of multi-GeV neutrinos crossing the Earth. Searches for deviations from the Standard Model of particle physics in the forward scattering of neutrinos inside Earth matter, produced by Non-Standard Interactions, can be conducted by investigating distortions of the standard oscillation pattern of neutrinos of all flavours. This work reports on the results of the search for non-standard neutrino interactions using the first six detection units of ORCA and 433 kton-years of exposure. No significant deviation from standard interactions was found in a sample of 5828 events reconstructed in the 1 GeV$-$1 TeV energy range. The flavour structure of the non-standard coupling was constrained at 90\% confidence level to be $|\varepsilon_{\mu\tau} | \leq 5.4 \times 10^{-3}$, $|\varepsilon_{e\tau} | \leq 7.4 \times 10^{-2}$, $|\varepsilon_{e\mu} | \leq 5.6 \times 10^{-2}$ and $-0.015 \leq \varepsilon_{\tau\tau} - \varepsilon_{\mu\mu} \leq 0.017$. The results are comparable to the current most stringent limits placed on the parameters by other experiments.

Abstract: Indirect dark matter detection methods are used to observe the products of dark matter annihilations or decays originating from astrophysical objects where large amounts of dark matter are thought to accumulate. With neutrino telescopes, an excess of neutrinos is searched for in nearby dark matter reservoirs, such as the Sun and the Galactic Centre, which could potentially produce a sizeable flux of Standard Model particles. The KM3NeT infrastructure, currently under construction, comprises the ARCA and ORCA undersea \v{C}erenkov neutrino detectors located at two different sites in the Mediterranean Sea, offshore of Italy and France, respectively. The two detector configurations are optimised for the detection of neutrinos of different energies, enabling the search for dark matter particles with masses ranging from a few GeV/c$^2$ to hundreds of TeV/c$^2$. In this work, searches for dark matter annihilations in the Galactic Centre and the Sun with data samples taken with the first configurations of both detectors are presented. No significant excess over the expected background was found in either of the two analyses. Limits on the velocity-averaged self-annihilation cross section of dark matter particles are computed for five different primary annihilation channels in the Galactic Centre. For the Sun, limits on the spin-dependent and spin-independent scattering cross sections of dark matter with nucleons are given for three annihilation channels.

Abstract: The KM3NeT Collaboration has tackled a common challenge faced by the astroparticle physics community, namely adapting the experiment-specific simulation software to work with the CORSIKA air shower simulation output. The proposed solution is an extension of the open-source code gSeaGen, allowing for the transport of muons generated by CORSIKA to a detector of any size at an arbitrary depth. The gSeaGen code was not only extended in terms of functionalities but also underwent a thorough redesign of the muon propagation routine, resulting in a more accurate and efficient simulation. Abstract: Neutrinos described as an open quantum system may interact with the environment which introduces stochastic perturbations to their quantum phase. This mechanism leads to a loss of coherence along the propagation of the neutrino $-$ a phenomenon commonly referred to as decoherence $-$ and ultimately, to a modification of the oscillation probabilities. Fluctuations in space-time, as envisaged by various theories of quantum gravity, are a potential candidate for a decoherence-inducing environment. Consequently, the search for decoherence provides a rare opportunity to investigate quantum gravitational effects which are usually beyond the reach of current experiments. In this work, quantum decoherence effects are searched for in neutrino data collected by the KM3NeT/ORCA detector from January 2020 to November 2021. The analysis focuses on atmospheric neutrinos within the energy range of a few GeV to $100\,\mathrm{GeV}$. Adopting the open quantum system framework, decoherence is described in a phenomenological manner with the strength of the effect given by the parameters $\Gamma_{21}$ and $\Gamma_{31}$. Following previous studies, a dependence of the type $\Gamma_{ij} \propto (E/E_0)^n$ on the neutrino energy is assumed and the cases $n = -2,-1$ are explored. No significant deviation with respect to the standard oscillation hypothesis is observed. Therefore, $90\,\%$ CL upper limits are estimated as $\Gamma_{21} < 4.6\cdot 10^{-21}\,$GeV and $\Gamma_{31} < 8.4\cdot 10^{-21}\,$GeV for $n = -2$, and $\Gamma_{21} < 1.9\cdot 10^{-22}\,$GeV and $\Gamma_{31} < 2.7\cdot 10^{-22}\,$GeV for $n = -1$, respectively., Comment: 17 pages, 5 figures

Abstract: KM3NeT/ORCA is a water Cherenkov neutrino detector under construction and anchored at the bottom of the Mediterranean Sea. The detector is designed to study oscillations of atmospheric neutrinos and determine the neutrino mass ordering. This paper focuses on an initial configuration of ORCA, referred to as ORCA6, which comprises six out of the foreseen 115 detection units of photo-sensors. A high-purity neutrino sample was extracted, corresponding to an exposure of 433 kton-years. The sample of 5828 neutrino candidates is analysed following a binned log-likelihood method in the reconstructed energy and cosine of the zenith angle. The atmospheric oscillation parameters are measured to be $\sin^2\theta_{23}= 0.51^{+0.04}_{-0.05}$, and $ \Delta m^2_{31} = 2.18^{+0.25}_{-0.35}\times 10^{-3}~\mathrm{eV^2} \cup \{-2.25,-1.76\}\times 10^{-3}~\mathrm{eV^2}$ at 68\% CL. Abstract: Results are presented for the measurement of large-scale anisotropies in the arrival directions of ultra-high-energy cosmic rays detected at the Pierre Auger Observatory during 19 years of operation, prior to AugerPrime, the upgrade of the Observatory. The 3D dipole amplitude and direction are reconstructed above $4\,$EeV in four energy bins. Besides the established dipolar anisotropy in right ascension above $8\,$EeV, the Fourier amplitude of the $8$ to $16\,$EeV energy bin is now also above the $5\sigma$ discovery level. No time variation of the dipole moment above $8\,$EeV is found, setting an upper limit to the rate of change of such variations of $0.3\%$ per year at the $95\%$ confidence level. Additionally, the results for the angular power spectrum are shown, demonstrating no other statistically significant multipoles. The results for the equatorial dipole component down to $0.03\,$EeV are presented, using for the first time a data set obtained with a trigger that has been optimized for lower energies. Finally, model predictions are discussed and compared with observations, based on two source emission scenarios obtained in the combined fit of spectrum and composition above $0.6\,$EeV., Comment: Minor modifications, accepted for publication in Astrophysical Journal

7. Berezinsky Hidden Sources: An Emergent Tension in the High-Energy Neutrino Sky?

Abstract: The IceCube Collaboration has recently reported compelling evidence of high-energy neutrino emission from NGC~1068, and also mild excesses for NGC 4151 and CGCG420-015, local Seyfert galaxies. This has increased the interest along neutrino emission from hot-corona surrounding the super massive black holes of Seyfert Galaxies. In this paper, we revisit phenomenological constraints on the neutrino emission from hot-coronae of seyfert galaxies, using an assumption of equi-ripartition between cosmic-rays and magnetic energy densities. We show that not only these sources are consistent with such an assumption but also that the data point towards low beta plasma parameters inside Seyfert Galaxies. We exploit this finding to constrain the Seyfert diffuse neutrino flux and we obtain that, in order not to overproduce neutrinos, not all the sources can be in an equi-ripartition state. We conclude (along with previous findings) that seyfert galaxies cannot explain the diffuse neutrino spectrum above $\sim 100\, \rm TeV$, allowing space for other astrophysical sources., Comment: 17 pages, 1 Table, 6 Figures, 1 appendix

Abstract: Gamma-ray bursts are promising candidate sources of high-energy astrophysical neutrinos. The recent GRB 221009A event, identified as the brightest gamma-ray burst ever detected, provides a unique opportunity to investigate hadronic emissions involving neutrinos. The KM3NeT undersea neutrino detectors participated in the worldwide follow-up effort triggered by the event, searching for neutrino events. In this letter, we summarize subsequent searches, in a wide energy range from MeV up to a few PeVs. No neutrino events are found in any of the searches performed. 9. Constraining the hadronic properties of star-forming galaxies above $1\, \rm GeV$ with 15-years Fermi-LAT data

Abstract: Star-forming and starburst galaxies (SFGs and SBGs) are powerful emitters of non-thermal $\gamma$-rays and neutrinos, due to their intense phases of star-formation activity, which should confine high-energy Cosmic-Rays (CRs) inside their environments. In this paper, using the publicly-available \texttt{fermitools}, we analyse 15.3 years of $\gamma$-ray between $1-1000\, \rm GeV$ data for 70 sources, 56 of which were not previously detected. We find at~$4\sigma$ level an indication of $\gamma$-ray emission for other two SBGs, namely M 83 and NGC 1365. By contrast, we find that, even with the new description of background, the significance for the $\gamma$-ray emission of M 33~(initially reported as discovered) still stands at $\sim \, 4\sigma$ (as already reported by previous works). Along with previous findings, the flux of each detected source is consistent with a $\sim E^{-2.3/2.4}$ spectrum, compatible with the injected CR flux inferred for CRs in the Milky-Way. We notice that the correlation between the calorimetric fraction~$F_{\rm cal}$ of high-energy protons in SFGs and SBGs (the fraction of high-energy protons actually producing high-energy $\gamma$-rays and neutrinos) and the SFR is in accordance with the expected scaling relation for CR escape dominated by advection. We remark that undiscovered sources strongly constrain $F_{\rm cal}$ at 95\% CL, providing fundamental information when we interpret the results as common properties of SFGs and SBGs. Finally, we find that these sources might contribute $(12\pm 3)\%$ to the EGB, while the corresponding diffuse neutrino flux strongly depends on the spectral index distribution along the source class., Comment: 32 pages, 9 figures, 16 Tables. Abstract: The KM3NeT/ARCA neutrino detector is currently under construction at 3500 m depth offshore Capo Passero, Sicily, in the Mediterranean Sea. The main science objectives are the detection of high-energy cosmic neutrinos and the discovery of their sources. Simulations were conducted for the full KM3NeT/ARCA detector, instrumenting a volume of 1 km$^3$, to estimate the sensitivity and discovery potential to point-like neutrino sources and an all-sky diffuse neutrino flux. This paper covers the reconstruction of track- and shower-like signatures, as well as the criteria employed for neutrino event selection. By leveraging both the track and shower observation channels, the KM3NeT/ARCA detector demonstrates the capability to detect the diffuse astrophysical neutrino flux within half a year of operation, achieving a 5$\sigma$ statistical significance. Abstract: The KM3NeT neutrino telescope is currently being deployed at two different sites in the Mediterranean Sea. First searches for astrophysical neutrinos have been performed using data taken with the partial detector configuration already in operation. The paper presents the results of two independent searches for neutrinos from compact binary mergers detected during the third observing run of the LIGO and Virgo gravitational wave interferometers. The first search looks for a global increase in the detector counting rates that could be associated with inverse beta decay events generated by MeV-scale electron anti-neutrinos. The second one focuses on upgoing track-like events mainly induced by muon (anti-)neutrinos in the GeV--TeV energy range. Both searches yield no significant excess for the sources in the gravitational wave catalogs. For each source, upper limits on the neutrino flux and on the total energy emitted in neutrinos in the respective energy ranges have been set. Stacking analyses of binary black hole mergers and neutron star-black hole mergers have also been performed to constrain the characteristic neutrino emission from these categories., Comment: 19 pages, 11 figures

Abstract: Abstract KM3NeT/ORCA is a water Cherenkov neutrino detector under construction and anchored at the bottom of the Mediterranean Sea. The detector is designed to study oscillations of atmospheric neutrinos and determine the neutrino mass ordering. This paper focuses on an initial configuration of ORCA, referred to as ORCA6, which comprises six out of the foreseen 115 detection units of photo-sensors. A high-purity neutrino sample was extracted, corresponding to an exposure of 433 kton-years. The sample of 5828 neutrino candidates is analysed following a binned log-likelihood method in the reconstructed energy and cosine of the zenith angle. The atmospheric oscillation parameters are measured to be sin 2 θ 23 = 0.51 − 0.05 + 0.04 $$ {\sin}^2{\theta}_{23}={0.51}_{-0.05}^{+0.04} $$ , and Δ m 31 2 = 2.18 − 0.35 + 0.25 × 10 − 3 eV 2 ∪ − 2.25 − 1.76 × 10 − 3 $$ \Delta {m}_{31}^2={2.18}_{-0.35}^{+0.25}\times {10}^{-3}{\textrm{eV}}^2\cup \left\{-2.25,-1.76\right\}\times {10}^{-3} $$ eV2 at 68% CL. The inverted neutrino mass ordering hypothesis is disfavoured with a p-value of 0.25.

Abstract: The Cherenkov Telescope Array and the KM3NeT neutrino telescopes are major upcoming facilities in the fields of $\gamma$-ray and neutrino astronomy, respectively. Possible simultaneous production of $\gamma$ rays and neutrinos in astrophysical accelerators of cosmic-ray nuclei motivates a combination of their data. We assess the potential of a combined analysis of CTA and KM3NeT data to determine the contribution of hadronic emission processes in known Galactic $\gamma$-ray emitters, comparing this result to the cases of two separate analyses. In doing so, we demonstrate the capability of Gammapy, an open-source software package for the analysis of $\gamma$-ray data, to also process data from neutrino telescopes. For a selection of prototypical $\gamma$-ray sources within our Galaxy, we obtain models for primary proton and electron spectra in the hadronic and leptonic emission scenario, respectively, by fitting published $\gamma$-ray spectra. Using these models and instrument response functions for both detectors, we employ the Gammapy package to generate pseudo data sets, where we assume 200 hours of CTA observations and 10 years of KM3NeT detector operation. We then apply a three-dimensional binned likelihood analysis to these data sets, separately for each instrument and jointly for both. We find that the largest benefit of the combined analysis lies in the possibility of a consistent modelling of the $\gamma$-ray and neutrino emission. Assuming a purely leptonic scenario as input, we obtain, for the most favourable source, an average expected 68% credible interval that constrains the contribution of hadronic processes to the observed $\gamma$-ray emission to below 15%., Comment: 20 pages, 16 figures. v2: Matches version published in EPJC

Abstract: The KM3NeT Collaboration is building and operating two deep sea neutrino telescopes at the bottom of the Mediterranean Sea. The telescopes consist of latices of photomultiplier tubes housed in pressure-resistant glass spheres, called digital optical modules and arranged in vertical detection units. The two main scientific goals are the determination of the neutrino mass ordering and the discovery and observation of high-energy neutrino sources in the Universe. Neutrinos are detected via the Cherenkov light, which is induced by charged particles originated in neutrino interactions. The photomultiplier tubes convert the Cherenkov light into electrical signals that are acquired and timestamped by the acquisition electronics. Each optical module houses the acquisition electronics for collecting and timestamping the photomultiplier signals with one nanosecond accuracy. Once finished, the two telescopes will have installed more than six thousand optical acquisition nodes, completing one of the more complex networks in the world in terms of operation and synchronization. The embedded software running in the acquisition nodes has been designed to provide a framework that will operate with different hardware versions and functionalities. The hardware will not be accessible once in operation, which complicates the embedded software architecture. The embedded software provides a set of tools to facilitate remote manageability of the deployed hardware, including safe reconfiguration of the firmware. This paper presents the architecture and the techniques, methods and implementation of the embedded software running in the acquisition nodes of the KM3NeT neutrino telescopes.

20. Galactic Diffuse Neutrino Emission from Sources beyond the Discovery Horizon

Abstract: The IceCube Neutrino Observatory has recently reported strong evidence for neutrino emission from the Galactic plane. The signal is consistent with model predictions of diffuse emission from cosmic ray propagation in the interstellar medium. However, due to IceCube's limited potential of identifying individual neutrino sources, it is also feasible that unresolved Galactic sources could contribute to the signal. We investigate the contribution of this quasi-diffuse emission and show that the observed Galactic diffuse flux at 100 TeV could be dominated by hard emission of unresolved sources. Particularly interesting candidate sources are young massive stellar clusters that have been considered as cosmic-ray PeVatrons. We examine whether this hypothesis can be tested by the upcoming KM3NeT detector or the planned future facility IceCube-Gen2 with about five times the sensitivity of IceCube., Comment: Matches version published in Physical Review D 109, 043007 (2024)

The purification of mammalian-derived EVs involves intensive processes, and their therapeutic application raises multiple safety and regulatory issues. Plants have the potential to serve as nonconventional sources of therapeutically relevant EVs. In this context, we recently identified hairy roots (HRs) of medicinal plants as a novel biotechnological platform to produce EVs for human health.Methods: Herein, we report the purification, omics profiling, and bioactivity of EVs isolated from HRs of the medicinal plants S. sclarea and S. dominica. EVs were isolated from conditioned media of HR cultures using differential ultracentrifugation (dUC) and size exclusion chromatography (SEC). The isolated EVs were characterized by nanoparticle tracking analysis (NTA) and electron microscopy. The proteomic and metabolomic profiles of the EVs were determined using mass spectrometry. Uptake studies and bioactivity assays, including confocal microscopy, MTT, flow cytometry, ROS quantification, and untargeted metabolomics analyses, were conducted in SH-SY5Y cells treated with the neurotoxin 6-hydroxydopamine (6-OHDA) to evaluate the therapeutic potential of EVs in an in vitro model of Parkinson’s disease.Results: S. sclarea HRs released nanosized round-shaped EVs with a distinctive molecular signature. HR EVs from S. sclarea and S. dominica revealed conserved cargo of secondary metabolites, predominantly triterpenoids, which are known for their antioxidant properties. We showed that HR EVs are safe, enter the cells, and strongly inhibit apoptosis in a cellular model of Parkinson’s disease. Cellular metabolomics revealed that EVs preserved metabolic homeostasis and mitigated cellular oxidative stress when co-administered with 6-OHDA. Mechanistically, HR EVs inhibited 6-OHDA autoxidation and substantially reduced the accumulation of its oxidative products, which are responsible for 6-OHDA-induced toxicity.Conclusion: Collectively, our findings provide compelling evidence that EVs isolated from the hairy roots of Salvia species are promising, non-mammalian alternative for the design of novel therapies targeting neurological disorders. Abstract: Abstract The KM3NeT/ARCA neutrino detector is currently under construction at 3500 m depth offshore Capo Passero, Sicily, in the Mediterranean Sea. The main science objectives are the detection of high-energy cosmic neutrinos and the discovery of their sources. Simulations were conducted for the full KM3NeT/ARCA detector, instrumenting a volume of 1 km $$^3$$ 3 , to estimate the sensitivity and discovery potential to point-like neutrino sources. This paper covers the reconstruction of track- and shower-like signatures, as well as the criteria employed for neutrino event selection. With an angular resolution below 0.1 $$^\circ $$ ∘ for tracks and under 2 $$^\circ $$ ∘ for showers, the sensitivity to point-like neutrino sources surpasses existing observed limits across the entire sky.

Abstract: Abstract The measurement of the flux of muons produced in cosmic ray air showers is essential for the study of primary cosmic rays. Such measurements are important in extensive air shower detectors to assess the energy spectrum and the chemical composition of the cosmic ray flux, complementary to the information provided by fluorescence detectors. Detailed simulations of the cosmic ray air showers are carried out, using codes such as CORSIKA, to estimate the muon flux at sea level. These simulations are based on the choice of hadronic interaction models, for which improvements have been implemented in the post-LHC era. In this work, a deficit in simulations that use state-of-the-art QCD models with respect to the measurement deep underwater with the KM3NeT neutrino detectors is reported. The KM3NeT/ARCA and KM3NeT/ORCA neutrino telescopes are sensitive to TeV muons originating mostly from primary cosmic rays with energies around 10 TeV. The predictions of state-of-the-art QCD models show that the deficit with respect to the data is constant in zenith angle; no dependency on the water overburden is observed. The observed deficit at a depth of several kilometres is compatible with the deficit seen in the comparison of the simulations and measurements at sea level.

Abstract: In the era of precision measurements of the neutrino oscillation parameters, upcoming neutrino experiments will also be sensitive to physics beyond the Standard Model. KM3NeT/ORCA is a neutrino detector optimised for measuring atmospheric neutrinos from a few GeV to around 100 GeV. In this paper, the sensitivity of the KM3NeT/ORCA detector to neutrino decay has been explored. A three-flavour neutrino oscillation scenario, where the third neutrino mass state $\nu_3$ decays into an invisible state, e.g. a sterile neutrino, is considered. We find that KM3NeT/ORCA would be sensitive to invisible neutrino decays with $1/\alpha_3=\tau_3/m_3 < 180$~$\mathrm{ps/eV}$ at $90\%$ confidence level, assuming true normal ordering. Finally, the impact of neutrino decay on the precision of KM3NeT/ORCA measurements for $\theta_{23}$, $\Delta m^2_{31}$ and mass ordering have been studied. Abstract: Optical communications and networking are fast becoming the solution to support ever-increasing data traffic across all segments of the network, expanding from core/metro networks to 5G/6G front-hauling. Therefore, optical networks need to evolve towards an efficient exploitation of the infrastructure by overcoming the closed and aggregated paradigm, to enable apparatus sharing together with the slicing and separation of the optical data plane from the optical control. In addition to the advantages in terms of efficiency and cost reduction, this evolution will increase the network reliability, also allowing for a fine trade-off between robustness and maximum capacity exploitation. In this work, an optical network architecture is presented based on the physical layer digital twin of the optical transport used within a multi-layer hierarchical control operated by an intent-based network operating system. An experimental proof of concept is performed on a three node network including up to 1000 km optical transmission, open re-configurable optical add & drop multiplexers (ROADMs) and white-box transponders hosting pluggable multi-rate transceivers. The proposed solution is based on GNPy as optical physical layer digital twin and ONOS as intent-based network operating system. The reliability of the optical control decoupled by the data plane functioning is experimentally demonstrated exploiting GNPy as open lightpath computation engine and software optical amplifier models derived from the component characterization. Abstract: Starburst galaxies are well-motivated astrophysical emitters of high-energy gamma-rays. They are well-known cosmic-ray "reservoirs", thanks to their large magnetic fields which confine high-energy protons for $\sim 10^5$ years. Over such long times, cosmic-ray transport can be significantly affected by scatterings with sub-GeV dark matter. Here we point out that this scattering distorts the cosmic-ray spectrum, and the distortion can be indirectly observed by measuring the gamma-rays produced by cosmic-rays via hadronic collisions. Present gamma-ray data show no sign of such a distortion, leading to stringent bounds on the cross section between protons and dark matter. These are highly complementary with current bounds and have large room for improvement with the future gamma-ray measurements in the 0.1-10 TeV range from the Cherenkov Telescope Array, which can strengthen the limits by as much as two orders of magnitude., Comment: 17 pages, 7 figures. v2: version published in Physical Review Letters

Abstract: Abstract Background Tumor-associated macrophages (TAMs) are a prominent immune subpopulation in the tumor microenvironment that could potentially serve as therapeutic targets for breast cancer. Thus, it is important to characterize this cell population across different tumor subtypes including patterns of association with demographic and prognostic factors, and breast cancer outcomes. Methods We investigated CD163+ macrophages in relation to clinicopathologic variables and breast cancer outcomes in the Women’s Circle of Health Study and Women’s Circle of Health Follow-up Study populations of predominantly Black women with breast cancer. We evaluated 611 invasive breast tumor samples (507 from Black women, 104 from White women) with immunohistochemical staining of tissue microarray slides followed by digital image analysis. Multivariable Cox proportional hazards models were used to estimate hazard ratios for overall survival (OS) and breast cancer-specific survival (BCSS) for 546 cases with available survival data (median follow-up time 9.68 years (IQR: 7.43–12.33). Results Women with triple-negative breast cancer showed significantly improved OS in relation to increased levels of tumor-infiltrating CD163+ macrophages in age-adjusted (Q3 vs. Q1: HR = 0.36; 95% CI 0.16–0.83) and fully adjusted models (Q3 vs. Q1: HR = 0.30; 95% CI 0.12–0.73). A similar, but non-statistically significant, association was observed for BCSS. Macrophage infiltration in luminal and HER2+ tumors was not associated with OS or BCSS. In a multivariate regression model that adjusted for age, subtype, grade, and tumor size, there was no significant difference in CD163+ macrophage density between Black and White women (RR = 0.88; 95% CI 0.71–1.10). Conclusions In contrast to previous studies, we observed that higher densities of CD163+ macrophages are independently associated with improved OS and BCSS in women with invasive triple-negative breast cancer. Abstract: Aggressive breast cancers portend a poor prognosis, but current polygenic risk scores (PRSs) for breast cancer do not reliably predict aggressive cancers. Aggressiveness can be effectively recapitulated using tumor gene expression profiling. Thus, we sought to develop a PRS for the risk of recurrence score weighted on proliferation (ROR-P), an established prognostic signature. Using 2363 breast cancers with tumor gene expression data and single nucleotide polymorphism (SNP) genotypes, we examined the associations between ROR-P and known breast cancer susceptibility SNPs using linear regression models. We constructed PRSs based on varying p-value thresholds and selected the optimal PRS based on model r2 in 5-fold cross-validation. We then used Cox proportional hazards regression to test the ROR-P PRS's association with breast cancer-specific survival in two independent cohorts totaling 10,196 breast cancers and 785 events. In meta-analysis of these cohorts, higher ROR-P PRS was associated with worse survival, HR per SD = 1.13 (95% CI 1.06-1.21, p = 4.0 × 10-4). The ROR-P PRS had a similar magnitude of effect on survival as a comparator PRS for estrogen receptor (ER)-negative versus positive cancer risk (PRSER-/ER+). Furthermore, its effect was minimally attenuated when adjusted for PRSER-/ER+, suggesting that the ROR-P PRS provides additional prognostic information beyond ER status. In summary, we used integrated analysis of germline SNP and tumor gene expression data to construct a PRS associated with aggressive tumor biology and worse survival. These findings could potentially enhance risk stratification for breast cancer screening and prevention.

Abstract: ObjectivesPhysical inactivity and sedentary behaviour are associated with higher breast cancer risk in observational studies, but ascribing causality is difficult. Mendelian randomisation (MR) assesses causality by simulating randomised trial groups using genotype. We assessed whether lifelong physical activity or sedentary time, assessed using genotype, may be causally associated with breast cancer risk overall, pre/post-menopause, and by case-groups defined by tumour characteristics.MethodsWe performed two-sample inverse-variance-weighted MR using individual-level Breast Cancer Association Consortium case-control data from 130 957 European-ancestry women (69 838 invasive cases), and published UK Biobank data (n=91 105-377 234). Genetic instruments were single nucleotide polymorphisms (SNPs) associated in UK Biobank with wrist-worn accelerometer-measured overall physical activity (nsnps=5) or sedentary time (nsnps=6), or accelerometer-measured (nsnps=1) or self-reported (nsnps=5) vigorous physical activity.ResultsGreater genetically-predicted overall activity was associated with lower breast cancer overall risk (OR=0.59; 95% confidence interval (CI) 0.42 to 0.83 per-standard deviation (SD;~8 milligravities acceleration)) and for most case-groups. Genetically-predicted vigorous activity was associated with lower risk of pre/perimenopausal breast cancer (OR=0.62; 95% CI 0.45 to 0.87,≥3 vs. 0 self-reported days/week), with consistent estimates for most case-groups. Greater genetically-predicted sedentary time was associated with higher hormone-receptor-negative tumour risk (OR=1.77; 95% CI 1.07 to 2.92 per-SD (~7% time spent sedentary)), with elevated estimates for most case-groups. Results were robust to sensitivity analyses examining pleiotropy (including weighted-median-MR, MR-Egger).ConclusionOur study provides strong evidence that greater overall physical activity, greater vigorous activity, and lower sedentary time are likely to reduce breast cancer risk. More widespread adoption of active lifestyles may reduce the burden from the most common cancer in women.

Abstract: Abstract Identifying women at high risk of osteoporotic fracture from aromatase inhibitor (AI) therapy for breast cancer is largely based on known risk factors for healthy postmenopausal women, which might not accurately reflect the risk in breast cancer patients post-AI therapy. To determine whether a polygenic score associated with fracture in healthy women is also significant in women treated with AIs for breast cancer, we used data from a prospective observational cohort of 2152 women diagnosed with hormonal receptor positive breast cancer treated with AIs as the initial endocrine therapy and examined a polygenic score of heel quantitative ultrasound speed of sound (gSOS) in relation to incident osteoporotic fracture after AI therapy during a median 6.1 years of follow up after AI initiation. In multivariable models, patients with the second and third highest tertiles (T) versus the lowest tertile of gSOS had significantly lower risk of fracture (T2: adjusted HR = 0.61, 95% CI: 0.46-0.80; T3: adjusted HR = 0.53, 95% CI: 0.40-0.70). The lower risk of fracture in patients with the highest tertile of gSOS remained significant after further adjustment for BMD at the hip (T3: adjusted HR = 0.62, 95% CI: 0.42-0.91). In conclusion, our analysis showed gSOS as a novel genetic predictor for fracture risk independent of BMD among breast cancer patients treated with AIs. Future studies are warranted to evaluate the performance of incorporating gSOS in prediction models for the risk of AI-related fracture in breast cancer patients.

Abstract: The gamma-ray emission from Starburst and Starforming Galaxies (SBGs and SFGs) strongly suggest a correlation between star-forming activity and gamma-ray luminosity. However, the very nature of cosmic-ray (CR) transport and the degree of their confinement within SBG cores are still open questions. We aim at probing the imprints left by CR transport on gamma-ray and neutrino observations of point-like SFGs and SBGs, looking into quantitative ways to discriminate among different transport models. Moreover, following the reported scenarios, we quantitatively assess the SBGs and SFGs contribution to the Extra-galactic Gamma-Ray Background (EGB data) and the IceCube diffuse observations (HESE data). We analyse the 10-year Fermi-LAT spectral energy distributions of 13 nearby galaxies with two different CR transport models, taking into account the corresponding IR and UV observations. We generate mock gamma-ray data to simulate the CTA performance in detecting these sources. In the way, we propose a test to discriminate between the two CR models, quantifying the statistical confidence at which one model can be preferred over the other. We point out that current data already give a slight preference to CR models which are dominated by advection in their nucleus. Moreover, we show that CTA will allow us to firmly disfavour models dominated by diffusion over self-induced turbulence, compared to advection-dominated models, with Bayes factors which can be as large as $10^7$ for some of the SBGs. Finally, we estimate the diffuse gamma-ray and neutrino fluxes of SFGs and SBGs, showing that they can explain $25\%$ of the diffuse HESE data, while remaining consistent with gamma-ray limits on non-blazar sources., Comment: 9 pages, 5 figures, 3 tables

Abstract: The KM3NeT Collaboration is currently constructing a multi-site high-energy neutrino telescope in the Mediterranean Sea consisting of matrices of pressure-resistant glass spheres, each holding a set of 31 small-area photomultipliers. The main goals of the telescope are the observation of neutrino sources in the Universe and the measurement of the neutrino oscillation parameters with atmospheric neutrinos. Both extraterrestrial and atmospheric neutrinos are detected through the Cherenkov light induced in seawater by charged particles produced in neutrino interactions in the surrounding medium. A relative time synchronization between photomultipliers of the order of 1 ns is needed to guarantee the required angular resolution of the detector. Due to the large detector volumes to be instrumented by KM3NeT, a cost reduction of the different systems is a priority. To this end, the inexpensive Nanobeacon has been designed and developed by the KM3NeT Collaboration to be used for detector time-calibration studies. At present, more than 600 Nanobeacons have been already produced. The characterization of the optical pulse and the wavelength emission profile of the devices are critical for the time calibration. In this paper, the main features of the Nanobeacon design, production and operation, together with the main properties of the light pulse generated are described.

Abstract: The KM3NeT research infrastructure is under construction in the Mediterranean Sea. KM3NeT will study atmospheric and astrophysical neutrinos with two multi-purpose neutrino detectors, ARCA and ORCA, primarily aimed at GeV-PeV neutrinos. Thanks to the multi-photomultiplier tube design of the digital optical modules, KM3NeT is capable of detecting the neutrino burst from a Galactic or near-Galactic core-collapse supernova. This potential is already exploitable with the first detection units deployed in the sea. This paper describes the real-time implementation of the supernova neutrino search, operating on the two KM3NeT detectors since the first months of 2019. A quasi-online astronomy analysis is introduced to study the time profile of the detected neutrinos for especially significant events. The mechanism of generation and distribution of alerts, as well as the integration into the SNEWS and SNEWS 2.0 global alert systems are described. The approach for the follow-up of external alerts with a search for a neutrino excess in the archival data is defined. Abstract: This article presents the potential of a combined analysis of the JUNO and KM3NeT/ORCA experiments to determine the neutrino mass ordering. This combination is particularly interesting as it significantly boosts the potential of either detector, beyond simply adding their neutrino mass ordering sensitivities, by removing a degeneracy in the determination of $\Delta m_{31}^2$ between the two experiments when assuming the wrong ordering. The study is based on the latest projected performances for JUNO, and on simulation tools using a full Monte Carlo approach to the KM3NeT/ORCA response with a careful assessment of its energy systematics. From this analysis, a $5\sigma$ determination of the neutrino mass ordering is expected after 6 years of joint data taking for any value of the oscillation parameters. This sensitivity would be achieved after only 2 years of joint data taking assuming the current global best-fit values for those parameters for normal ordering., Comment: 24 pages, 9 figures

Abstract: Starburst galaxies (SBGs) and more in general starforming galaxies represent a class of galaxies with a high star formation rate (up to 100 solar masses/year). Despite their low luminosity, they can 19 be considered as guaranteed "factories" of high energy neutrinos, being "reservoirs" of accelerated 20 cosmic rays and hosting a high density target gas in the central region. In this contribution 21 we present a novel multimessenger study of these sources and the possibility of observing their 22 neutrino signals with the KM3NeT/ARCA telescope. Abstract: KM3NeT/ORCA is a next-generation neutrino telescope optimised for atmospheric neutrino oscillations studies. In this paper, the sensitivity of ORCA to the presence of a light sterile neutrino in a 3+1 model is presented. After three years of data taking, ORCA will be able to probe the active-sterile mixing angles $\theta_{14}$, $\theta_{24}$, $\theta_{34}$ and the effective angle $\theta_{\mu e}$, over a broad range of mass squared difference $\Delta m^2_{41} \sim [10^{-5}, 10]$ $\rm{eV}^2$, allowing to test the eV-mass sterile neutrino hypothesis as the origin of short baseline anomalies, as well as probing the hypothesis of a very light sterile neutrino, not yet constrained by cosmology. ORCA will be able to explore a relevant fraction of the parameter space not yet reached by present measurements.

42. Could nearby star-forming galaxies light up the point-like neutrino sky?

Abstract: Star-forming and starburst galaxies, which are well-known cosmic-rays reservoirs, are expected to emit gamma-rays and neutrinos predominantly via hadronic collisions. In this Letter, we analyze the 10-year Fermi-LAT spectral energy distributions of 13 nearby galaxies by means of a physical model which accounts for high-energy proton transport in starburst nuclei and includes the contribution of primary and secondary electrons. In particular, we test the hypothesis that the observed gamma-ray fluxes are mostly due to star-forming activity, in agreement with the available star formation rates coming from IR and UV observations. Through this observation-based approach, we determine the most-likely neutrino counterpart from star-forming and starburst galaxies and quantitatively assess the ability of current and upcoming neutrino telescopes to detect them as point-like sources. Remarkably, we find that the cores of the Small Magellanic Cloud and the Circinus galaxy are potentially observable by KM3NeT/ARCA with 6 years of observation. Moreover, most of the nearby galaxies are likely to be just a factor of a few below the KM3NeT and IceCube-Gen2 point-like sensitivities. Abstract: Recently Dolgov and Pozdnyakov proposed a new baryogenesis mechanism in which baryon asymmetry is produced without violating baryon number at the Lagrangian level. In this scenario, baryon asymmetry is generated by absorption of a new particle X carrying baryon number onto Primordial Black Holes (PBHs). Assuming CP-violation, the particle X is absorbed at a different rate than the antiparticle $\bar{X}$, producing an asymmetry in the baryonic number. We independently test this scenario, finding that it suffers from two fundamental issues.\\ At the phenomenological level, strong absorption by PBHs initially increases the baryon asymmetry. However, at later times such asymmetry is completely absorbed by PBHs. In order to overcome this issue, we account for PBH evaporation, which provides a natural way of halting the absorption while keeping a finite baryon asymmetry. We provide a systematic study of the parameter space, identifying the regions leading to the production of the baryon asymmetry without violating the known constraints on PBHs concentration. At the theoretical level, a model realizing the CP-violation postulated in this scenario is difficult to realize. We show, by implementing a minimal model, that the framework proposed in the original work in order to produce CP-violation, even if qualitatively correct, is quantitatively in disagreement with the observed baryon asymmetry, namely this mechanism produces only a fraction of the total baryon asymmetry., Comment: 8 pages, 3 figures

Abstract: The next generation of water Cherenkov neutrino telescopes in the Mediterranean Sea are under construction offshore France (KM3NeT/ORCA) and Sicily (KM3NeT/ARCA). The KM3NeT/ORCA detector features an energy detection threshold which allows to collect atmospheric neutrinos to study flavour oscillation. This paper reports the KM3NeT/ORCA sensitivity to this phenomenon. The event reconstruction, selection and classification are described. The sensitivity to determine the neutrino mass ordering was evaluated and found to be 4.4 $\sigma$ if the true ordering is normal and 2.3 $\sigma$ if inverted, after three years of data taking. The precision to measure $\Delta m^2_{32}$ and $\theta_{23}$ were also estimated and found to be $85\cdot10^{-6}$ eV$^2$ and $(^{+1.9}_{-3.1})^{\circ}$ for normal neutrino mass ordering and, $75\cdot10^{-6}$ eV$^2$ and $(^{+2.0}_{-7.0})^{\circ}$ for inverted ordering. Finally, a unitarity test of the leptonic mixing matrix by measuring the rate of tau neutrinos is described. Three years of data taking were found to be sufficient to exclude $\nu_{\tau}$ and $\bar{\nu}_{\tau}$ event rate variations larger than 20% at $3\sigma$ level., Comment: Accepted for publication at EPJ-C

Abstract: The KM3NeT research infrastructure is under construction in the Mediterranean Sea. It consists of two water Cherenkov neutrino detectors, ARCA and ORCA, aimed at neutrino astrophysics and oscillation research, respectively. Instrumenting a large volume of sea water with $\sim$ 6,200 optical modules comprising a total of $\sim$ 200,000 photomultiplier tubes, KM3NeT will achieve sensitivity to $\sim$ 10 MeV neutrinos from Galactic and near-Galactic core-collapse supernovae through the observation of coincident hits in photomultipliers above the background. In this paper, the sensitivity of KM3NeT to a supernova explosion is estimated from detailed analyses of background data from the first KM3NeT detection units and simulations of the neutrino signal. The KM3NeT observational horizon (for a $5\,\sigma$ discovery) covers essentially the Milky-Way and for the most optimistic model, extends to the Small Magellanic Cloud ($\sim$ 60 kpc). Detailed studies of the time profile of the neutrino signal allow assessment of the KM3NeT capability to determine the arrival time of the neutrino burst with a few milliseconds precision for sources up to 5$-$8 kpc away, and detecting the peculiar signature of the standing accretion shock instability if the core-collapse supernova explosion happens closer than 3$-$5 kpc, depending on the progenitor mass. Abstract: PurposeTo determine the contribution of germline pathogenic variants (PVs) in hereditary cancer testing panel genes to invasive lobular carcinoma (ILC) of the breast.Materials and methodsThe study included 2,999 women with ILC from a population-based cohort and 3,796 women with ILC undergoing clinical multigene panel testing (clinical cohort). Frequencies of germline PVs in breast cancer predisposition genes (ATM, BARD1, BRCA1, BRCA2, BRIP1, CDH1, CHEK2, PALB2, PTEN, RAD51C, RAD51D, and TP53) were compared between women with ILC and unaffected female controls and between women with ILC and infiltrating ductal carcinoma (IDC).ResultsThe frequency of PVs in breast cancer predisposition genes among women with ILC was 6.5% in the clinical cohort and 5.2% in the population-based cohort. In case-control analysis, CDH1 and BRCA2 PVs were associated with high risks of ILC (odds ratio [OR] > 4) and CHEK2, ATM, and PALB2 PVs were associated with moderate (OR = 2-4) risks. BRCA1 PVs and CHEK2 p.Ile157Thr were not associated with clinically relevant risks (OR < 2) of ILC. Compared with IDC, CDH1 PVs were > 10-fold enriched, whereas PVs in BRCA1 were substantially reduced in ILC.ConclusionThe study establishes that PVs in ATM, BRCA2, CDH1, CHEK2, and PALB2 are associated with an increased risk of ILC, whereas BRCA1 PVs are not. The similar overall PV frequencies for ILC and IDC suggest that cancer histology should not influence the decision to proceed with genetic testing. Similar to IDC, multigene panel testing may be appropriate for women with ILC, but CDH1 should be specifically discussed because of low prevalence and gastric cancer risk.

48. Starburst galaxies strike back: a multi-messenger analysis with Fermi-LAT and IceCube data

Abstract: Starburst galaxies, which are known as "reservoirs" of high-energy cosmic-rays, can represent an important high-energy neutrino "factory" contributing to the diffuse neutrino flux observed by IceCube. In this paper, we revisit the constraints affecting the neutrino and gamma-ray hadronuclear emissions from this class of astrophysical objects. In particular, we go beyond the standard prototype-based approach leading to a simple power-law neutrino flux, and investigate a more realistic model based on a data-driven blending of spectral indexes, thereby capturing the observed changes in the properties of individual emitters. We then perform a multi-messenger analysis considering the extragalactic gamma-ray background (EGB) measured by Fermi-LAT and different IceCube data samples: the 7.5-year High-Energy Starting Events (HESE) and the 6-year high-energy cascade data. Along with starburst galaxies, we take into account the contributions from blazars and radio galaxies as well as the secondary gamma-rays from electromagnetic cascades. Remarkably, we find that, differently from the highly-constrained prototype scenario, the spectral index blending allows starburst galaxies to account for up to $40\%$ of the HESE events at $95.4\%$ CL, while satisfying the limit on the non-blazar EGB component. Moreover, values of $\mathcal{O}(100~\mathrm{PeV})$ for the maximal energy of accelerated cosmic-rays by supernovae remnants inside the starburst are disfavoured in our scenario.
