Cryptosporidiosis is one of the main causes of diarrhea in children and young livestock. The interaction of the parasite with the intestinal host cells has not been characterized thoroughly yet but may be affected by the nutritional demand of the parasite. Hence, we aimed to investigate the impact of C. parvum infection on glucose metabolism in neonatal calves. Therefore, N = 5 neonatal calves were infected with C. parvum on the first day of life, whereas a control group was not (N = 5). The calves were monitored clinically for one week, and glucose absorption, turnover and oxidation were assessed using stable isotope labelled glucose. The transepithelial transport of glucose was measured using the Ussing chamber technique. Glucose transporters were quantified on gene and protein expression level using RT-qPCR and Western blot in the jejunum epithelium and brush border membrane preparations. Plasma glucose concentration and oral glucose absorption were decreased despite an increased electrogenic phlorizin sensitive transepithelial transport of glucose in infected calves. No difference in the gene or protein abundance of glucose transporters, but an enrichment of glucose transporter 2 in the brush border was observed in the infected calves. Furthermore, the mRNA for enzymes of the glycolysis pathway was increased indicating enhanced glucose oxidation in the infected gut. In summary, C. parvum infection modulates intestinal epithelial glucose absorption and metabolism. We assume that the metabolic competition of the parasite for glucose causes the host cells to upregulate their uptake mechanisms and metabolic machinery to compensate for the energy losses., (© 2023. We developed a newborn (NB) mouse Plasmodium yoelii NL infection model to study malaria in early age. Surprisingly, the onset of parasitemia in P. yoelii challenged NB mice was delayed compared to adults and coincided with the weaning date when weanlings switched from maternal milk to normal chow diet. Also, compared to adult mice, parasitemia resolved much later (48 days vs 20 days post challenge) and the peak parasitemia was twice as high in weanlings. Concurrently, weanlings' germinal center reaction was delayed and diminished compared to adult mice. Maternal milk is deficient in para-aminobenzoic acid (PABA), which is required for de novo folate synthesis by Plasmodium. Suggesting a possible role for the protection afforded by PABA-deficient maternal milk, mice fed with a PABA-deficient diet after the weaning continued to control parasitemia. Despite the reduced parasitemia, these mice developed robust T follicular helper (Tfh) responses and were protected from a second P. yoelii challenge. The NB malaria model provides mechanistic insight into the human infant malaria manifestations where a diet solely based on breast-feeding reduces the incidence of severe malaria in infants. NB mice experiments also support further studies to investigate dietary PABA restriction in the management of severe malaria in infants.

The protozoan parasite Cryptosporidium sp. is a leading cause of diarrheal disease in those with compromised or underdeveloped immune systems, particularly infants and toddlers in resource-poor localities. As an enteric pathogen, Cryptosporidium sp. invades the apical surface of intestinal epithelial cells, where it resides in close proximity to metabolites in the intestinal lumen. However, the effect of gut metabolites on susceptibility to Cryptosporidium infection remains largely unstudied. Here, we first identified which gut metabolites are prevalent in neonatal mice when they are most susceptible to Cryptosporidium parvum infection and then tested the isolated effects of these metabolites on C. parvum invasion and growth in intestinal epithelial cells. Our findings demonstrate that medium or long-chain saturated fatty acids inhibit C. parvum growth, perhaps by negatively affecting the streamlined metabolism in C. parvum , which is unable to synthesize fatty acids. Conversely, long-chain unsaturated fatty acids enhanced C. parvum invasion, possibly by modulating membrane fluidity. Hence, gut metabolites, either from diet or produced by the microbiota, influence C. parvum growth in vitro and may also contribute to the early susceptibility to cryptosporidiosis seen in young animals. IMPORTANCE Cryptosporidium sp. occupies a unique intracellular niche that exposes the parasite to both host cell contents and the intestinal lumen, including metabolites from the diet and produced by the microbiota. Both dietary and microbial products change over the course of early development and could contribute to the changes seen in susceptibility to cryptosporidiosis in humans and mice. Consistent with this model, we show that the immature gut metabolome influenced the growth of Cryptosporidium parvum in vitro Interestingly, metabolites that significantly altered parasite growth were fatty acids, a class of molecules that Cryptosporidium sp. is unable to synthesize de novo The enhancing effects of polyunsaturated fatty acids and the inhibitory effects of saturated fatty acids presented in this study may provide a framework for future studies into this enteric parasite's interactions with exogenous fatty acids during the initial stages of infection., (Copyright © 2020 VanDussen et al.)

Predation risk is thought to modify the physiology of prey mainly through the stress response. However, little is known about its potential effects on the immunity of animals, particularly in young individuals, despite the importance of overcoming wounding and pathogen aggression following a predator attack. We investigated the effect of four progressive levels of nest predation risk on several components of the immune system in common blackbird ( Turdus merula ) nestlings by presenting them with four different calls during 1 h: non-predator calls, predator calls, parental alarm calls and conspecific distress calls to induce a null, moderate, high and extreme level of risk, respectively. Nest predation risk induced an increase in ovotransferrin, immunoglobulin and the number of lymphocytes and eosinophils. Thus, the perception of a potential predator per se could stimulate the mobilization of a nestling's immune function and enable the organism to rapidly respond to the immune stimuli imposed by a predator attack. Interestingly, only high and extreme levels of risk caused immunological changes, suggesting that different immunological parameters are modulated according to the perceived level of threat. We also found a mediator role of parasites (i.e. Leucocytozoon ) and the current health status of the individual, as only nestlings not parasitized or in good body condition were able to modify their immune system. Background: Cryptosporidium spp. infections are the most frequent parasitic cause of diarrhea in humans and cattle. However, asymptomatic cases are less often documented than symptomatic cases or cases with experimentally infected animals. Cryptosporidium (C.) hominis infection accounts for the majority of pediatric cases in several countries, while C. parvum is a major cause of diarrhea in neonatal calves. In cattle Cryptosporidium spp. infection can be caused by C. parvum, C. bovis, C.andersoni and C. ryanae, and recently, reports of cattle cases of C. hominis cryptosporidiosis cases suggest that the presence of C. hominis in calves was previously underestimated., Methodology/principal Findings: From February to November 2015, Cryptosporidium spp. infected calves were detected in 29/44 randomly included farms from 5 geographic regions of France. C. hominis and C. parvum were found in 12/44 and 26/44 farms, respectively with higher C. hominis prevalence in the western region. In 9 farms, both C. parvum and C. hominis were detected. Eighty-six of 412 (73/342 asymptomatic and 13/70 symptomatic) one to nine-week-old calves shed C. hominis or C. parvum oocysts (15 and 71 calves, respectively), with no mixed infection detected. The predominant C. hominis IbA9G3 genotype was present in all regions, and more frequent in the western region. An incompletely characterized Ib, and the IbA13G3, IbA9G2 and IbA14G2 genotypes were present only in the western region. For C. parvum, the most frequent genotype was IIaA16G3R1 with no geographic clustering. Most C. hominis infected calves were asymptomatic, with some exceptions of IbA9G2 and IbA9G3 isolates, while C. parvum IIaA16G3R1 was associated with symptoms., Conclusions/significance: Present results indicate for the first time that in several geographic regions of France, C. hominis was present in about one fifth of both asymptomatic and symptomatic infected calves, with isolated genotypes likely associated with human infection. Further investigations are aimed at documenting direct or indirect transmissions between livestock and humans.

Background: The gastrointestinal parasitic nematode Haemonchus contortus is a pathogenic organism resistant to several anthelmintics. This study assessed the efficacy of a medicinal herbal mixture (Herbmix) and organic zinc, as an essential trace element for the proper functioning of both unspecific and specific immune defensive mechanisms, against experimental infections with H. contortus in lambs. All lambs were infected orally with approximately 5000 third-stage larvae of a strain of H. contortus susceptible to anthelmintics (MHco1). Twenty-four female lambs 3-4 months of age were divided into four groups: unsupplemented animals (control), animals supplemented with Herbmix (Hmix), animals supplemented with organic zinc (Zn) and animals supplemented with Herbmix and organic zinc (Hmix+Zn). Eggs per gram (EPG) of faeces were quantified 20, 28, 35, 42, 49, 56, 62 and 70 d post-infection and mean abomasal worm counts were assessed 70 d post-infection. Samples of blood were collected from each animal 7, 35, 49 and 70 d post-infection., Results: Quantitative analyses of the bioactive compounds in Herbmix identified three main groups: flavonoids (9964.7 μg/g), diterpenes (4886.1 μg/g) and phenolic acids (3549.2 μg/g). Egg counts in the lambs treated with Hmix, Zn and Hmix+Zn decreased after 49 d. The EPGs in the Zn and Hmix+Zn groups were significantly lower on day 56 (P < 0.05 and P < 0.01, respectively), and the EPGs and mean worm counts were significantly lower on day 70 in all supplemented groups (P < 0.05 and P < 0.01). Hemograms of complete red blood cells of each animal identified clinical signs of haemonchosis after day 35. Serum calprotectin concentrations and IgA levels were significantly affected by treatment. The treatment influenced serum malondialdehyde concentrations (P < 0.05) and sulfhydryl groups (P < 0.01) of antioxidant status. The mineral status was unaltered in all lambs., Conclusion: A direct anthelmintic impact on the viability of nematodes was not fully demonstrated, but the treatments with herbal nutraceuticals and zinc likely indirectly contributed to the increase in the resistance of the lambs to nematode infection.

Climatic selective pressures are thought to dominate biotic selective pressures at higher latitudes. However, few studies have experimentally tested how these selective pressures differentially act on traits across latitudes because traits can rarely be manipulated independently of the organism in nature. We overcame this challenge by using an extended phenotype-active bird nests-and conducted reciprocal transplant experiments between a subarctic and temperate site, separated by 14° of latitude. At the subarctic site, biotic selective pressures (nest predation) favoured smaller, non-local temperate nests, whereas climatic selective pressures (temperature) favoured larger local nests, particularly at colder temperatures. By contrast, at the temperate site, climatic and biotic selective pressures acted similarly on temperate and subarctic nests. Our results illustrate a functional trade-off in the subarctic between nest morphologies favoured by biotic versus climatic selective pressures, with climate favouring local nest morphologies. At our temperate site, however, allocative trade-offs in the time and effort devoted to nest construction favour smaller, local nests. Our findings illustrate a conflict between biotic and climatic selective pressures at the northern extremes of a species geographical range, and suggest that trade-offs between trait function and trait elaboration act differentially across latitude to create broad geographic variation in traits., (© 2015 The Author(s).)

Background: The abomasal helminth Teladorsagia circumcincta is one of the most economically important parasites affecting sheep in temperate regions. Infection is particularly detrimental to lambs, in which it can cause pronounced morbidity and severe production losses. Due to the spreading resistance of this parasite to all classes of anthelmintic drugs, teladorsagiosis is having an increasingly severe impact on the sheep industry with significant implications for sheep welfare. Protective immunity develops slowly, wanes rapidly and does not appear to be as effective in young lambs. To investigate the development of immunity to T. circumcincta in sheep and lambs, we used cytokine transcript profiling to examine differences in the abomasal mucosa and gastric lymph node of naïve and previously infected sheep and lambs following challenge., Results: The results of these experiments demonstrated that the abomasal mucosa is a major source of cytokines during abomasal helminth infection. A local Th2-type cytokine response was observed in the abomasal mucosa and gastric lymph node of the previously infected sheep and lambs when compared with those of the naïve during the early stages of infection. In contrast, a pro-inflammatory component more was evident in the abomasal mucosa and gastric lymph node of the naïve sheep when compared with those of the previously infected, which was not observed in the lambs., Conclusions: The greater levels of Th2-type cytokine transcripts in both the abomasum and gastric lymph node of the previously infected compared with naïve sheep and lambs emphasises the importance of these mechanisms in the immune response to T. circumcincta infection. Younger lambs appear to be able to generate similar Th2-type responses in the abomasum suggesting that the increased morbidity and apparent lack of resistance in younger lambs following continuous or repeated exposure to T. circumcincta is unlikely to be due to a lack of appropriate Th2-type cytokine production.

Both Cryptosporidium and Giardia are frequently found in the stool of domestic ruminants, especially young animals. Wild ruminants are also host to these protozoa, but the prevalence of these parasites in both free-ranging and captive nondomesticated ruminants needs to be further investigated. Moreover, the role of wild ruminants serving as reservoirs for these zoonotic parasites remains unclear. Therefore, a cross-sectional survey was conducted to estimate the occurrence of Cryptosporidium and Giardia in captive wild ruminants younger than 6 mo and to determine the potential of these animals to serve as reservoirs for these zoonotic parasites. A total of 67 captive wild ruminants belonging to 21 different animal species at the Antwerp Zoo (Belgium), along with 82 American bison (Bison bison) on a commercial breeding farm, were sampled for the detection of Cryptosporidium and Giardia, using a commercial immunofluoresence assay (Merifluor Cryptosporidium/Giardia IFA). The Cryptosporidium prevalence was 7.5% in the Antwerp Zoo animals and 3.7% in the bison from the breeding farm. All but two of the Cryptosporidium-positive animals were younger than 1 mo of age. Molecular characterization by amplification of the 70-kDa heat-shock protein and the 18S ribosomal DNA gene identified Cryptosporidium parvum in four animals of the Antwerp Zoo. The prevalence of Giardia was 8.9% in the Antwerp Zoo animals and 23.2% in the bison calves. Most Giardia-positive animals were older than 1 mo of age. Molecular characterization on the beta-giardin gene and the triose phosphate isomerase gene identified Giardia duodenalis assemblage A in the Antwerp Zoo and both G. duodenalis assemblage A and assemblage E in the bison calves. These findings indicate that both protozoan parasites are prevalent in captive wild ruminants and that these animals can serve as a potential reservoir for zoonotic transmission.

Two freshly-dead female Florida panther (FP) neonates, Puma concolor couguar (=Puma concolor coryi), an 11-day-old and a 17-day-old, were collected in the Florida Panther National Wildlife Refuge (26 degrees 14'N, 81 degrees 36'W), Collier County, Florida. The 2 neonates were siblings and had presumably fed only on milk from the dam since birth. A 12-day-old female FP neonate was collected in the Big Cypress National Preserve (26 degrees 05'N, 81 degrees 15'W), Collier County, Florida and had also fed only on milk from the dam since birth. Milk was the only food item found in the gastrointestinal tract of these neonates. Mesocercariae and diplostomula of Alaria marcianae were collected from the lungs of the 3 neonates, indicating a transmammary route of infection. No mesocercariae, diplostomula, or mature A. marcianae were seen in the stomach or small intestine. The probable paratenic host for the A. marcianae infection in the adult Florida panther is the raccoon (Procyon lotor).

The aim of this study was to demonstrate, through nested PCR (nPCR) method, the occurrence of congenital babesiosis in two foals born of carrier mares. All mares were positive for T. equi based in visualization of intraerytrocytic parasites in blood smears, in indirect fluorescent antibody test (IFAT) and nPCR reactions. Just one mare was nPCR-positive for B. caballi. After the birth, all foals presented nPCR-positive for T. equi, while just one foal presented nPCR-positive for B. caballi. The present study prove the occurrence of congenital babesiosis in new-born foals, however, new studies are necessary to elucidate if the infection occur for transplacental transmission or in parturition moment.

Heads of 64 white-tailed deer (Odocoileus virginianus) fawns, harvested in the vicinity of Magrath, Alberta, Canada, (49 degrees 24'782''N, 112 degrees 52'113''W) were examined for the presence of nasal bots and lice. The deer were collected between 8-30 January 2004 as part of a government-approved herd reduction protocol. The entire surface of each head was scanned visually for the presence of lice. Each head was split longitudinally, and the nasal passages, sinuses, and ethmoid region were washed for recovery of nasal bots. First instar Cephenemyia spp. were recovered from 17 heads (27%). Intensity of infestation ranged from 1-18 larvae (mean intensity 4.8). Among fawns, there were no significant differences in prevalence or mean intensity between the sexes. Two species of nasal bots were identified. Smaller larvae, tentatively identified as C. jellisoni, were present in 16 of 17 infested deer while larger specimens, tentatively identified as C. phobifera, were found in four deer; and in three of the four it co-occurred with C. jellisoni. The presence of C. phobifera in Alberta would represent a range extension for this species, which has not been known to occur west of North Dakota. Thirty-one fawns (48%) were infested with the sucking louse Solenopotes ferrisi. One infested fawn also had one specimen of the chewing louse, Tricholiopeurus lipeuroides.

A longitudinal study was undertaken to determine the prevalence of Cryptosporidium in a dairy farm in Sfax, Tunisia. 480 faecal samples were obtained from 30 calves under one month of age. All faecal samples were analysed for Cryptosporidium oocysts by microscopic examination of smears stained by modified Ziehl Neelsen technique. The parasite was detected in 26 calves (86.7%). Infection was significantly associated with diarrhoea. A molecular characterization, performed in seven calves, confirmed that isolates were C. parvum. This work is the first report on Cryptosporidium in calves in Tunisia.

Liver fluke infection was identified as a probable cause of clinical disease in an approximately 6-mo-old elk (Cervus elaphus) in coastal Oregon. Clinical pathology and necropsy findings are described. The alcohol-fixed flukes that were submitted for identification were similar in size to Fasciola hepatica, but their shape resembled Fascioloides magna in that they lacked a distinctive anterior cone. A few structures consistent with the eggs of F. magna were observed in liver lesions, suggesting that at least some of the worms were sexually maturing. Due to difficulties in morphologic identification associated with improper fixation technique, DNA analysis was used to compare small subunit (SSU) and internal transcribed spacer 2 ribosomal RNA gene sequences of the recovered parasites with those of F. hepatica and F. magna, confirming these small, but sexually mature flukes were F. magna. This is the first publication of the SSU gene sequence for F. magna. Phylogenetic analysis showed that it is related to, but is an outlier, to the genus Fasciola. Due to the high mortality rate associated with this disease outbreak, the overall significance of trematodiasis in the herd is unclear.

Large breeding populations of California sea lions (Zalophus californianus) are located on San Miguel and San Nicolas Islands in the Southern California Bight. In 2001, there was a substantial increase in pup mortality in late summer and fall. From June 2002 to January 2003, 208 freshly dead pups were examined on San Miguel Island, the most western of the Channel Islands off the coast of southern California. Tissues from 186 of these pups were examined histologically. The primary lesions in 133 (72%) of the pups were an enteritis associated with hookworms and infections in major organs. Emaciation/starvation in 43 pups (26%) was the second most important cause of death.

While banding ferruginous pygmy-owls (Glaucidium brasilianum) and Eastern screech-owls (Megascops asio) in south Texas during 2004, we recorded Philornis mimicola (Diptera: Muscidae) and Ornithodoros concanensis (Acari: Argasidae) parasitizing nestlings. Inspection of nestlings revealed 54 P. mimicola and one O. concanensis. Inspection of nest material revealed 111 P. mimicola, including 57 puparia. The effect (e.g., blood loss, anemia) of these hematophagous parasites might have contributed to the demise of at least one Eastern screech-owl nestling. This is the first record of P. mimicola and O. concanensis parasitizing ferruginous pygmy-owls and Eastern screech-owls.

This paper describes a case of Anaplasma phagocytophilum infection in a roe deer (Capreolus capreolus) calf in Norway. The calf was found deserted, paretic, and heavily infested with Ixodes ricinus ticks. It was euthanized and investigated postmortem. Anaplasma phagocytophilum was detected in several tissues by polymerase chain reaction (PCR) and 16S rRNA sequence analyses. Analyses for Borrelia burgdorferi sensu lato and tick-borne encephalitis (TBE) virus infections were negative. This is the first report of a possible paretic condition in A. phagocytophilum infected roe deer.

We examined 267 red foxes (Vulpes vulpes) from the canton of Geneva, Switzerland, for intestinal infections with Echinococcus multilocularis. This region is situated in the core area of the endemic range of this zoonotic cestode in Central Europe. Several factors were taken into account and urbanisation level appeared to be the most explicative to describe observed differences. The prevalence decreased significantly from rural and residential areas (prevalence of 52%, CI 43-62%, and 49%, CI 38-59 %, respectively) to the urban area (prevalence of 31%, CI 19-42%). A few juvenile foxes harboured very high burdens up to more than 120,000 worms and were significantly more heavily infected than adults. The intensity of infection decreased from rural and residential areas to the city, suggesting a lower contamination of the urban environment.

A white-tailed deer (Odocoileus virginianus) fawn was collected in Missouri (USA) and submitted for diagnostic evaluation. Necropsy and histologic examination revealed severe Amblyomma americanum infestation, pronounced icterus, and marked hemosiderin deposition in the liver and spleen. Whole blood evaluation revealed a normocytic normochromic anemia and a piroplasm parasitemia of approximately 70%. The piroplasm was identified as Theileria cervi by polymerase chain reaction and sequencing of the V4 variable region of the 18S rRNA gene from a paraffin-embedded section of lung. Although T. cervi parasitemias have been commonly reported in healthy white-tailed deer, the severe parasitemia in this fawn may have contributed to overt clinical disease, perhaps as part of a combined malnutrition and parasitemia syndrome.

The acquisition of immunity to Eimeria maxima by chicks infected 18 hr after hatch with a single dose of 100 oocysts was investigated. In the first experiment, birds were moved each day to clean cages in order to prevent the possibility of secondary infection resulting from ingestion of oocysts passed in their feces. Immunity was measured at 4 wk of age by calculation of oocyst production following challenge with 500 oocysts or weight gain following challenge with 100,000 oocysts. Large numbers of oocysts were produced by infected birds following challenge, although numbers were significantly less than those from birds that had been reared in the absence of infection (susceptible controls). The weight gain of infected birds following challenge was significantly greater than that of susceptible controls but less than that of unchallenged controls. Thus, only partial protection had been acquired, whether parasite replication or body weight gain was used to assess the extent of immunity development. In a second experiment, acquisition of immunity at 4 wk by chicks infected 18 hr after hatch with 100 oocysts of E. maxima and reared in floor pens in contact with their droppings was investigated. Infected birds produced no oocysts following challenge, and weight gains were not significantly different from the unchallenged controls, which indicates that full immunity had developed by 4 wk. It is concluded that if oocysts of Eimeria species are used to vaccinate day-old chicks, reinfection by oocysts present in the litter is necessary for the establishment of protective immunity.

In recent years, the northern goshawk (Accipiter gentilis) has colonized suburban and urban areas in Berlin, Germany, and elsewhere in Europe. Because of the high proportion of feral pigeons (Columba livia f. domestica) in their diet, urban goshawks are suspected to have a high infection rate with Trichomonas gallinae. Therefore, from 1998 to 2001, we examined 269 nestlings from 90 nests for infection with T. gallinae by culture of swabs taken from the oropharynx and checked their oropharynx for the presence of caseous lesions indicative of trichomonosis. In 80% of the nest sites (n=90), at least one nestling was infected. The nestling infection rate with T. gallinae was 69.7% (n=33) in 1998, 73.0% (n=89) in 1999, 55.8% (n=77) in 2000, and 62.9% (n=70) in 2001. In total, 65.1% of the northern goshawk nestlings were culture positive for T. gallinae. Prevalence increased with the age of nestlings (chi2=12.4, n=269, df=5, P=0.03) and tended to increase with brood size (chi2=9.345, n=269, df=4, P=0.053). Caseous lesions were present in 12 nestlings (4.5%), but only 10 of these were culture positive for T. gallinae. Two nestlings (0.7%) had large caseous lesions (diameter>1 cm) characteristic of late-stage trichomonosis and died shortly after examination. It is suggested that the combination of a high prevalence of T. gallinae with a low rate of pathologicic changes is the result of an evolutionary-adapted parasite-host relationship.

Seasonal population trends and effects of Oestrus ovis in naturally infected sheep were studied over 13 months, in the Western of the Pampeana region. At weaning, 140 growing lambs were randomly allocated to two groups: UG, untreated group and TG treated every 4 weeks with closantel (10 mg/kg). Successive Oestrus free tracer lambs (TL) by previous treatment (n = 65) were slaughtered after 20-30 exposition days for larval counts. Likewise, other group PL of 117 permanent untreated lambs was slaughtered from four to 17 months of age. Weighing and assessment of health signs of UG and TG and blood samples were monthly carried out. The prevalence of infection in permanent group varied from 33% to 100%. Mean number of larvae in PL was 6.1 with 3 L1, 1.4 L2 and 1.6 L3 during spring-summer and 17.9 with 16.9 L1, 0.5 L2 and 0.4 L3 during autumn-winter months. In PL the proportions of larvae in each of the different larval stages was similar during spring and summer, but a significant (P < 0.01) increase of L1 was detected during autumn and winter. The prevalence in tracer lambs was 100% during summer time and larvae were absent from 25-May to 25-October. Mean larval burdens of positive TL varied from 6.4 to one Oestrus and a significant peak (P < 0.05) of larvae was seen from December to March. Since March to November only L1 was recovered from TL. TG group showed a reduction in nasal discharge and in antibody ELISA levels, but no difference was observed in live weight gain between TG and UG. These results show a high prevalence during summer and that the perpetuation of Oestrus is ensured by an autumn period of arrested development and the over wintering larvae in the sheep heads.

Lice have been described on goats in commercial farming systems in South Africa but not from flocks on communal grazing. During a longitudinal survey on the causes of goat kid mortality, conducted in Jericho district, North West Province, lice were collected from communally grazed indigenous goats. These lice were prepared for and viewed by scanning electron microscopy, and micro-morphological taxonomic details are described. Three species of lice were found in the study area and identified as Bovicola caprae, Bovicola limbatus and Linognathus africanus. Sucking and biting lice were found in ten of the 12 herds of goats examined. Lice were found on both mature goats and kids. Bovicola caprae and L. africanus were the most common biting and sucking lice respectively in all herds examined. Scanning electron microscopy revealed additional features which aided in the identification of the louse species. Photomicrographs were more accurate aids to identification than the line drawings in the literature and facilitated identification using dissecting microscope.

One of the unusual aspect of the life cycle of Strongyloides stercoralis is the occurrence of autoinfective third-stage larvae (L3a). These are the causative agents of severe hyperinfective strongyloidiasis. When 6-wk-old gerbils are infected with 1,000 infective third-stage larvae (L3i), no L3a are seen during the course of the infection. However, in neonatal gerbils (1-13 days of age) infected with 1,000 L3i, a burst of autoinfection takes place between 15 and 30 days postinfection (PI). Only occasional L3a can be found in neonatally infected gerbils after 4 wk PI. This autoinfective burst is not seen in neonatal gerbils infected with 200 L3i.

Congenital toxoplasmosis was evaluated in Wistar and Holtzman rats using two strains of Toxoplasma gondii isolated in Brazil. Pregnant rats were inoculated by subcutaneous or intraperitoneal routes with 10(6) or 8 x 10(6) tachyzoites of N strain (virulent for mice) and by subcutaneous or oral routes with 10(2) or 1.2 x 10(3) cysts of P strain (avirulent for mice). The tissues of rat pups born from these rats were bioassayed for T. gondii infection. T. gondii was not observed in the pups born from rats inoculated with N strain. In the animals inoculated with P strain, congenital toxoplasmosis occurred in 22.8% (Wistar rats inoculated with 10(2) cysts by the subcutaneous route), 11.4% (Wistar rats inoculated with 10(2) cysts by the oral route), 21.2% (Wistar rats inoculated with 1.2 x 10(3) cysts by the oral route) and 2.9% of fetal infection (Holtzman rats inoculated with 10(2) cysts by the oral route). None of the pups born from chronically infected mother were infected with T. gondii.

Six groups of six pregnant ewes each were inoculated with 170,000 or 1,700,000 tachyzoites of Neospora caninum on gestation day 65, 90, or 120. All ewes seroconverted, and none showed signs of illness other than abortion. Regardless of the inoculum dose, all ewes inoculated on gestation day 65 aborted; ewes inoculated on gestation day 90 aborted, gave birth to weak lambs, or gave birth to clinically normal lambs; and all ewes inoculated on gestation day 120 gave birth to clinically normal lambs. Using an immunohistological procedure that stains bradyzoites, we observed protozoal cysts in brains of 11 of 29 (38%) aborted fetuses, in one of four (25%) weak lambs, and in seven of 18 (39%) clinically normal lambs. Cysts were not observed in extraneural tissues from two clinically normal lambs that had cysts in the brain. No evidence of infection was observed in tissues of five ewes examined using an immunohistological procedure that stains N. caninum tachyzoites and bradyzoites. Multifocal nonsuppurative encephalitis was observed in 46 of 51 (90%) aborted, weak, or clinically normal lambs. Cerebral necrosis, dystrophic mineralization, and meningitis were also commonly identified in live and aborted lambs (even when severely autolyzed). Nonsuppurative, necrotizing placentitis was observed in 15 of 17 (88%) placentas. Nonsuppurative myositis was common in fetuses but not in live lambs. Inflammation occurred less frequently in liver and lung. Clinical and pathological features of neosporosis in sheep closely resemble those of bovine neosporosis and ovine toxoplasmosis. Although abortion caused by naturally occurring neosporosis in sheep has not been reported, diagnosticians should carefully distinguish between neosporosis and toxoplasmosis in cases of ovine protozoal abortion unless future investigations exclude the likelihood of naturally acquired neosporosis in sheep.

We investigated whether administration of interferon-gamma (IFN-gamma) to pregnant rats, infected or not with Trypanosoma cruzi, was likely to protect their offspring from trypanosomal infection. Upon mating with syngeneic sires, four groups of 70-day-old female 1 rats were subjected to one of the following procedures: treatment with recombinant rat (Rr)IFN-gamma 50,000 IU/rat five times/week for three weeks; infection with 1 x 10(6) trypomastigotes of T. cruzi at 7, 14, and 21 days after mating plus IFN-gamma treatment as given to the former group; the same protocol but IFN-gamma injections being replaced by injection with physiologic saline. Offspring were nursed by their mothers until weaning and then infected with a similar dose of T. cruzi. Pregnant rats showed no exacerbated infection but a self-resolving mild disease, regardless of whether or not they had received IFN-gamma. Maternal infection with T. cruzi and/or IFN-gamma treatment did not affect gestational outcome. Offspring born to both groups of IFN-gamma-treated mothers were almost fully protected from acute infection, and showed higher levels of anti-T. cruzi IgG antibodies when compared with young born to their respective IFN-gamma-untreated mothers. Measurements of IFN-gamma serum activities indicated that ameliorated acute disease in offspring whose mothers were given IFN-gamma during gestation, was not associated with increased levels of endogenously produced IFN-gamma.

An unidentified Toxoplasma gondii-like disseminated infection was found in tissues of a 2-day-old hartebeest. Protozoal tachyzoites were about 3.5 x 1.7 Microns, had electron-dense rhoptries, and divided by endodyogeny. The organism did not stain with polyclonol antibodies to T. gondii, Neospora caninum, or Sarcocystis cruzi.

The effect of dietary lactose and Lactobacillus acidophilus on the colonization of Salmonella enteritidis (phage type 4) in newly hatched leghorn chicks concurrently infected with Eimeria tenella was studied. There were six groups of birds in each of two replicate trials: uninfected controls, birds infected with S. enteritidis, birds infected with S. enteritidis and E. tenella, birds infected with S. enteritidis and E. tenella that were fed with dietary lactose (5%), birds infected with S. enteritidis and E. tenella that were provided with L. acidophilus, and birds infected with S. enteritidis and E. tenella that were fed both L. acidophilus and dietary lactose (5%). Chickens were necropsied at 2, 5, and 8 days after S. enteritidis inoculation for bacteriological examination. The cecal population of S. enteritidis was significantly increased by infection with E. tenella. Lactose alone and the combination of lactose and L. acidophilus significantly reduced the population of S. enteritidis in the ceca of birds infected with E. tenella (P < 0.05), but the combination was more effective than lactose alone (P < 0.05). Adding anaerobic culture of L. acidophilus alone did not significantly decrease the population of S. enteritidis in the ceca. No significant reduction of colonization of S. enteritidis in the liver and spleen of birds with coccidiosis was observed.

Neonatal Jersey calves (n = 96) were used to evaluate effects of housing (individual hutches or wooden pens in a barn) and colostrum feeding (calves were separated from the dam and fed 2 L of colostrum in nipple-bottles or allowed to nurse the dam for 3 d) on the prevalence of selected organisms in feces. Prevalence of Cryptosporidium and Eimeria were reduced, and prevalence of rotavirus tended to be reduced, when calves were housed in hutches. Prevalence of coronavirus was unaffected by treatment. Weekly prevalence of Giardia was increased when calves were left to nurse the dam for 3 d. Mean prevalence of Cryptosporidia (wk 1 to 4), Eimeria (wk 4 to 5), Giardia, rotavirus, and coronavirus (wk 1 to 5) were 34.7, 20.6, 27.1, 15.8, and 4.9%, respectively. Escherichia coli (K99 positive) were observed in 3 of 174 samples cultured. Methods of housing and colostrum feeding affected acquisition of enteropathogens in this study.

Faecal and blood samples were taken from 10-30% of calves, 36 hours to 14 days old, in 47 dairy herds in different regions of Sweden from September 1987 to October 1988 (Olsson et al. 1993). Faecal samples from 279 calves were analysed for the presence of Escherichia coli (K99+), rotavirus and Cryptosporidium sp. Twenty (7.2%) of these samples were from diarrhoeic calves. An ELISA was developed and used for the rotavirus analysis. E. coli K99+ was detected in 11.5%, Cryptosporidium sp. in 6.1% and rotavirus in 5.4% of the faecal samples. The presence of rotavirus alone and the combination rotavirus and E. coli (K99+) was found to be associated with diarrhoea (p < 0.001 and p < 0.01 respectively). Blood samples from 327 calves were analysed for the level of total protein and gamma-globulin. In 43 of these samples (13%) gamma-globulin did not separate from the beta 2-region by electrophoresis. The mean total protein concentration was 53.6 g/l in calves free from diarrhoea. The mean gamma-globulin concentration, adjusted to 7 days age was 5.9 g/l. The 20 diarrhoeic calves had lower levels of both total protein and gamma-globulin, compared with calves without diarrhoea, but the difference was not significant. One litre more of colostrum at the first feed increased the level of total protein of the calves' sera by 1.4 g/l (p = 0.05). Calves born between May and September had a 2.0 g/l higher (p < 0.001) serum concentration of gamma-globulin than calves born between October and April.

A rat model of congenital toxoplasmosis is described for the study of protective immunity and chemotherapy during pregnancy. Six Sprague-Dawley 7- to 15-day gestational rats were inoculated orally (three rats, trial A) or subcutaneously (three rats, trial B) with 10,000 infective oocysts of the CT-1 strain of Toxoplasma gondii. The tissues of rat pups born from these rats were bioassayed for T. gondii infection. T. gondii was recovered from 30 of 33 (90.9%, trial B) and 23 of 28 (82.17%, trial A) rat pups bioassayed at 1 to 134 days of age. Two 10- to 14-day gestational pregnant rats were inoculated subcutaneously (trial C) with 10,000 infective bradyzoites from tissue cysts; 10 of 23 (43.8%) of the newborn pups were infected with T. gondii. Congenital infection occurred only when rats were infected during pregnancy. None of three rats in trial A that had given birth to congenitally infected rat pups produced congenitally infected rat pups during the second pregnancy.

The absence of larvae in lambs of six infected and controlled ewes immediately killed at birth without suckling may indicate the absence of prenatal infection. The ewes had larvae in their milk immediately prior to lambing and several hours afterwards. The persistence of infection, even after intestinal infection was removed with thiabendazole at 50 mg/kg body weigh, may be due to the pool of inhibited larvae. All the suckling lambs had patent infections within 4 to 5 days. Infection of suckling lambs with Strongyloides papillosus via milk from their ewes demonstrates the transmammary route of infection of this nematode.

One of seven female coyotes (Canis latrans) captured in Webb County, Texas during September 1986 and confined and mated in holding facilities at Millville, Utah whelped the following spring. The maternal female (greater than 5-yr-old) and her five neonates were killed at 22 days postparturition. All were infected with adult Ancylostoma caninum and were passing eggs in their feces. Also, the neonates and maternal female were infected with immature and adult Alaria marcianae, respectively. These findings suggested that the transmammary route is an important transmission mechanism for acquisition of these species of helminths in coyotes. The lack of overdispersion in the frequency distribution of these parasites and infection of the entire litter indicated that transmission from the infected female was nonselective among the pups.

Twenty-eight piglets coming from a "specific pathogen free" herd were inoculated at three days of age with 50 000 or 100 000 sporulated oocysts of Isospora suis. Fecal samples were examined for oocyst shedding daily and several clinical parameters were recorded. Ten piglets were used as normal controls. Groups of piglets were euthanized from three days to 12 days postinoculation and routine necropsies were performed. Bacteriological, virological, parasitological and histopathological examinations were made on the intestinal tracts. The incubation period was four to five days. Clinical signs and microscopic intestinal lesions observed in the experimentally infected animals were similar to those reported in spontaneous cases of porcine neonatal coccidiosis. Lesions of villous atrophy in the small intestine seemed to result from the destruction of villous epithelial cells mainly during the peak of asexual reproduction which occurred around four to five days postinoculation. Intracellular coccidial organisms were difficult to find during the late atrophic and villous regrowth stages of the intestinal lesions. The prepatent period varied from four to seven days and the most common was five days. Eighty percent of the piglets kept alive more than four days postinoculation have shed oocysts. Piglets dosed with old sporulated oocysts (ten months old) shed many more oocysts than those infected with a fresh inoculum (less than two months old). The patent period was not determined precisely with the design of the experiment but some of the infected piglets shed oocysts for at least five days.

Three 3-day-old kittens were inoculated subcutaneously and orally with Neospora caninum tachyzoites. A littermate and the queen were not inoculated with N. caninum and served as controls. Kitten 1 died between 14 and 16 days postinoculation (DPI) and was eaten by the mother. Kitten 2 died 17 DPI and kitten 3 was euthanized 29 DPI in a moribund condition. The control littermate and the dam remained healthy. Granulomatous skeletal myositis and nonsuppurative encephalomyelitis were the main lesions and were associated with numerous N. caninum tachyzoites in kittens 2 and 3. Cysts were found in kitten 3. Oocysts were not found in any cats. Neither lesions nor parasites were found in control cats.

Transmammary infection with Alaria marcianae mesocercariae was demonstrated using mice as model, paratenic hosts. Prenatal transmission was ruled out because neonates removed immediately postpartum from infected dams were never infected. Mesocercarial distribution in virgin females and in females examined immediately postpartum showed no marked preference for the mammary glands. In contrast, infection of neonates that were allowed to suckle on infected dams was absolute, and the number of mesocercariae in the mammary glands of postparturient dams that suckled their young was increased significantly. These experimental observations were coupled with other observations on paratenic hosts to outline the pathways open to mesocercariae in gravid hosts. The term amphiparatenic host is coined for those host species that are paratenic hosts as adults, but as juveniles can serve as definitive hosts.

It was demonstrated that the tissue reaction around the developing C. bovis in neonatally infected calves (4 days after birth) differs quantitatively and partly also qualitatively from the reaction in calves infected at the age of several months. In neonatal infection, the foci caused by cysticerci were 3-4-times smaller on day 14 p.i., and after 34 days, the cysts were quite transparent and similar to those occurring in older calves not before 42-50 days p.i. The tissue eosinophilia was lacking and the activation of fibroblasts and collagen formation was markedly weak. There was a surprisingly low number of cells of lymphoid type. The growth of the cysticercus and its morphological differentiation was only insignificantly more rapid. The serological tests (IHA, MPA, IFR) repeated four times (10-34 days p.i.). were negative in one calf, whereas in the other one, slaughtered on day 14 p.i., positive results were obtained only in IFR (1:40).

Neospora caninum-like organisms were found in histologic sections of spinal cord of 4 paralyzed calves as reported by Parish et al. (1987). Tachyzoites divided by endodyogeny. Tissue cysts were up to 62 microns wide and the cyst wall was up to 2.5 microns thick. The organism in calves was structurally distinct from Toxoplasma gondii and Sarcocystis species and reacted positively with anti-N. caninum serum in an immunoperoxidase test.

Histochemical studies on the cysticercus and surrounding tissue reaction were performed at various intervals after experimental infection. It was found that acid mucosubstances and proteins with SH- and SS-groups appeared first in the granulation tissue around the cysticercus (on about day 14 p.i.) and only later (on day 28 p.i.) in the tegument of the cysticercus where they were localized in the rim of microtriches. This envelope consisting of mucosubstances and proteins seems to be identical with the electron-dense substance found on the surface of developing cysticercus during electron-microscopical studies. It is considered to be a mimicry enabling the cysticerci to survive even in an immunologically unfavourable environment. Phospholipids were found in activated fibroblasts and in some cells of macrophage type on days 21 and 30 p.i. and in a large number in subtegumental cells of cysticercus on days 28-34 p.i. This phenomenon seems to be correlated with the increased activity of subtegumental cells of the larva in this period. In morphologically fully differentiated cysticerci, the reaction for phospholipids in subtegumental cells and distal cytoplasm was only feebly positive. Phospholipids were not detected in the rim of microtriches at any time after infection.

Residual ovine nematode pasture infections were assessed by grazing groups of ewes and their lambs on permanent sheep and cattle pastures and by the use of tracer lambs. Ostertagia spp., Cooperia oncophora, Nematodirus spp., Chabertia ovina and Trichuris spp. eggs and/or larvae survived on pastures overwinter. Second generation Ostertagia larvae were present in greatest numbers on pasture during the latter part of August and early September. The failure of a significant build-up of Cooperia oncophora was attributed to negligible worm egg output of this species in sheep. A build-up of Nematodirus spp. on pasture was not detected in this study.
