SUMMARY OBJECTIVE: Few studies on physical medicine and rehabilitation analyze the benefit of wheelchair basketball in people with motor disabilities. Given these, this study aimed to investigate the effect of the intervention of wheelchair basketball on urinary tract infection in people with motor disabilities. METHODS: A 12-month experimental follow-up was conducted in a single-center study. A total of 48 male individuals aged 18–55 years were allocated to the control group and experimental group. The experimental group practiced wheelchair basketball for 2 h, twice a week. Intra- and intergroup comparisons were made pre- and post-interventions over urinary tract infection. RESULTS: There was a significant improvement in urinary tract infection and urine culture in pre- and post-intervention antibiograms, respectively. Moreover, the intergroup comparison presented a decrease in infection caused by Klebsiella pneumoniae, as well as an increase in the time variability of partially activated thromboplastin, average corpuscular hemoglobin, and hemoglobin and platelets. In the experimental group, there was an increase in hemoglobin and hematocrit and a decrease in glycated hemoglobin (%HbA1C). On the intragroup comparison, there was a reduction of triiodothyronine (T3), %HbA1C, interleukin-6 pre-intervention, and C-reactive protein post-intervention. CONCLUSIONS: There was a decrease in urinary tract infection and improvement in biochemical, immunological, and microbiological biomarkers evaluated with physical exercise practice by wheelchair basketball, as well as by multiprofessional follow-up and health guidance.

Valsartan is an antihypertensive active pharmaceutical ingredient (API), it is used in the amorphous state in the commercial products. As amorphous materials are metastable, amorphous valsartan can crystallize to valsartan E, promoting changes in the dissolution and bioavailability of the drug. Tablets containing metastable forms of APIs need special conditions for transport and storage in order to avoid crystallization (from amorphous state) or polymorphic transitions (from less stable crystalline structures). A multivariate calibration model based on interval partial least squares (iPLS) regression allied to net analyte signal (NAS) algorithm was built to simultaneously quantify amorphous (VAL-AM) and crystalline (VAL-E) valsartan. Mixtures of VAL-AM and VAL-E were used to produce tablets in order to simulate the crystallization of VAL-AM in a range from 0 to 100% of conversion. The calibration set included 11 samples and 5 samples were used as the external validation set. The following parameters of merit (POM) were obtained for both polymorphs in order to evaluate the calibration model quality: root mean square error (RMSE) for cross validation (RMSECV), validation (RMSEV) and calibration (RMSEC), sensitivity (SEN), selectivity (SEL), analytical sensitivity (γ), inverse analytical sensitivity (γ−1), limit of detection (LOD) and limit of quantification (LOQ).

OBJECTIVE: Obesity and metabolic diseases such as metabolic syndrome (MetS) are more prevalent in people with type 2 diabetes mellitus (T2DM), major depressive disorder (MDD), bipolar disorder (BD), and schizophrenia (SZ). MetS components might be associated with neurocognitive and functional impairments in these individuals. The predictive and discriminatory validity of MetS and its components regarding those outcomes were assessed from prospective and transdiagnostic perspectives.; METHODS: Metabolic syndrome components and neurocognitive and social functioning were assessed in 165subjects, including 30 with SZ, 42 with BD, 35 with MDD, 30 with T2DM, and 28healthy controls (HCs). A posteriori, individuals were classified into two groups. The MetS group consisted of those who met at least three of the following criteria: abdominal obesity (AO), elevated triglycerides (TG), reduced high-density lipoprotein cholesterol (HDL), elevated blood pressure (BP), and elevated fasting glucose (FPG); the remaining participants comprised the No-MetS group. Mixed one-way analysis of covariance and linear and binary logistic regression analyses were performed.; RESULTS: Cognitive impairment was significantly greater in the MetS group (n = 82) than in the No-MetS group (n = 83), with small effect sizes (p < 0.05; etap = 0.02 - 0.03). In both groups, the most robust associations between MetS components and neurocognitive and social functioning were observed with TG and FPG (p < 0.05). There was also evidence for a significant relationship between cognition and BP in the MetS group (p < 0.05). The combination of TG, FPG, elevated systolic BP and HDL best classified individuals with greater cognitive impairment (p < 0.001), and TG was the most accurate (p < 0.0001).; CONCLUSIONS: Specific MetS components are significantly associated with cognitive impairment across somatic and psychiatric disorders. Our findings provide further evidence on the summative effect of MetS components to predict cognition and social functioning and allow the identification of individuals with worse outcomes. Transdiagnostic, lifestyle-based therapeutic interventions targeted at that group hold the potential to improve health outcomes. © 2022 The Authors. Background: Systemic, low-grade immune-inflammatory activity, together with social and neurocognitive performance deficits are a transdiagnostic trait of people suffering from type 2 diabetes mellitus (T2DM) and severe mental illnesses (SMIs), such as schizophrenia (SZ), major depressive disorder (MDD), and bipolar disorder (BD). We aimed to determine if immune-inflammatory mediators were significantly altered in people with SMIs or T2DM compared with healthy controls (HC) and whether these biomarkers could help predict their cognition and social functioning 1 year after assessment.; Methods: We performed a prospective, 1-year follow-up cohort study with 165 participants at baseline (TB), including 30 with SZ, 42 with BD, 35 with MDD, 30 with T2DM, and 28 HC; and 125 at 1-year follow-up (TY), and determined executive domain (ED), global social functioning score (GSFS), and peripheral blood immune-inflammatory and oxidative stress biomarkers.; Results: Participants with SMIs and T2DM showed increased peripheral levels of inflammatory markers, such as interleukin-10 (p < 0.01; eta2 p = 0.07) and tumor necrosis factor-alpha (p < 0.05; eta2 p = 0.08); and oxidative stress biomarkers, such as reactive oxygen species (ROS) (p < 0.05; eta2 p = 0.07) and mitochondrial ROS (p < 0.01; eta2 p = 0.08). The different combinations of the exposed biomarkers anticipated 46-57.3% of the total ED and 23.8-35.7% of GSFS for the participants with SMIs.; Limitations: Participants' treatment, as usual, was continued without no specific interventions; thus, it was difficult to anticipate substantial changes related to the psychopharmacological pattern.; Conclusion: People with SMIs show significantly increased levels of peripheral immune-inflammatory biomarkers, which may contribute to the neurocognitive and social deficits observed in SMIs, T2DM, and other diseases with systemic immune-inflammatory activation of chronic development. These parameters could help identify the subset of patients who could benefit from immune-inflammatory modulator strategies to ameliorate their functional outcomes. BACKGROUND: Neurocognition impairments are critical factors in patients with major depressive disorder (MDD), bipolar disorder (BD), and schizophrenia (SZ), and also in those with somatic diseases such as type 2 diabetes mellitus (T2DM). Intriguingly, these severe mental illnesses are associated with an increased co-occurrence of diabetes (direct comorbidity). This study sought to investigate the neurocognition and social functioning across T2DM, MDD, BD, and SZ using a transdiagnostic and longitudinal approach. METHODS: A total of 165 participants, including 30 with SZ, 42 with BD, 35 with MDD, 30 with T2DM, and 28 healthy controls (HC), were assessed twice at a 1-year interval using a comprehensive, integrated test battery on neuropsychological and social functioning. A new imidazolium hydrated salt (DCF–IMI–H2O) of the nonsteroidal anti-inflammatory drug diclofenac (DCF) was synthesized by solvent evaporation. This salt was fully characterized by X-ray diffraction (SCXRD and PXRD), infrared spectroscopy (FT-IR) and thermal analysis (TGA, DSC, and HSM) techniques. The salt formation is driven by deprotonation of the drug's carboxylic group, with proton transfer to the nitrogen atom of the imidazole ring. The water molecule plays an important role in the 3D arrangement observed in the crystal lattice, acting as a bridge between adjacent DCF and coformer molecules. DCF–IMI–H2O exhibits an improved solubility profile when compared with the pure form of DCF, and similar values when compared with sodium and potassium salts. This new salt proved stable under 75% relative humidity for one week and is stable after six-months of storage under ambient conditions. The study of its thermal stability showed that a dehydration process starts at around 45 °C, leading to the probable formation of an unstable anhydrous form, which melts around 92 °C. Spectroscopy studies showed that most of the salt is ionized when in an amorphous state (liquid). These behaviors feature the new compound as an ionic liquid (IL), i.e., organic salt with a melting point below 100 °C. Pharmaceutical ILs have been increasingly exploited in the past few years due to their improved properties regarding drug solubility, formulation and delivery (topical, transdermal and oral). Thus, the salt depicted herein becomes a potent candidate for development of novel/improved DCF delivery systems.

Social functioning impairments were observed in almost all the disorders (p < 0.0001; η2p = 0.29-0.49). Transdiagnostic deficits remained stable across the 1-year follow-up (p < 0.001; η2p = 0.13-0.43) and could accurately differentiate individuals with somatic and psychiatric disorders (χ2 = 48.0, p < 0.0001).ConclusionsThis longitudinal study provides evidence of the overlap in neurocognition deficits across somatic and psychiatric diagnostic categories, such as T2DM, MDD, BD, and SZ, which have high comorbidity. This overlap may be a result of shared genetic and environmental etiological factors. The findings further lay forth promising avenues for research on transdiagnostic phenotypes of neurocognition in these disorders, in addition to their biological bases.

- Abstract
Notch signaling is a core patterning module for vascular morphogenesis that codetermines the sprouting behavior of endothelial cells (ECs). Tight quantitative and temporal control of Notch activity is essential for vascular development, yet the details of Notch regulation in ECs are incompletely understood. We found that ubiquitin-specific peptidase 10 (USP10) interacted with the NOTCH1 intracellular domain (NICD1) to slow the ubiquitin-dependent turnover of this short-lived form of the activated NOTCH1 receptor. Accordingly, inactivation of USP10 reduced NICD1 abundance and stability and diminished Notch-induced target gene expression in ECs. In mice, the loss of endothelial Usp10 increased vessel sprouting and partially restored the patterning defects caused by ectopic expression of NICD1. Thus, USP10 functions as an NICD1 deubiquitinase that fine-tunes endothelial Notch responses during angiogenic sprouting.

- Abstract
In mammals, postnatal haematopoiesis occurs in the bone marrow (BM) and involves specialized microenvironments controlling haematopoietic stem cell (HSC) behaviour and, in particular, stem cell dormancy and self-renewal. While these processes have been linked to a number of different stromal cell types and signalling pathways, it is currently unclear whether BM has a homogenous architecture devoid of structural and functional partitions. Here, we show with genetic labelling techniques, high-resolution imaging and functional experiments in mice that the periphery of the adult BM cavity harbours previously unrecognized compartments with distinct properties. These units, which we have termed hemospheres, were composed of endothelial, haematopoietic and mesenchymal cells, were enriched in CD150+ CD48- putative HSCs, and enabled rapid haematopoietic cell proliferation and clonal expansion. Inducible gene targeting of the receptor tyrosine kinase VEGFR2 in endothelial cells disrupted hemospheres and, concomitantly, reduced the number of CD150+ CD48- cells. Our results identify a previously unrecognized, vessel-associated BM compartment with a specific localization and properties distinct from the marrow cavity., In mammals, postnatal haematopoiesis occurs in the bone marrow (BM) and involves specialized microenvironments controlling haematopoietic stem cell (HSC) behaviour and, in particular, stem cell dormancy and self-renewal. IntroductionBreast cancer patients with germline pathogenic variants may benefit from risk-reducing surgeries, intensive screening, and targeted cancer therapies. There is a paucity of data regarding prevalence and distribution of germline pathogenic variants in the Brazilian population. Our primary endpoint was the description of prevalence and distribution of germline pathogenic variants among breast cancer patients who underwent next-generation sequencing (NGS) panel testing. Secondary endpoint was the assessment of predictive factors of a positive test.MethodsWe analyzed NGS results, personal, and family history data from a prospectively collected cohort of breast cancer patients from August 2013 to May 2019. Exact logistic regression was used to perform multivariable analysis.ResultsOf 370 breast cancer patients, we found 59 pathogenic variants in 57 (15%) patients. Pathogenic variants were identified in BRCA1 (24%), ATM (14%), BRCA2 (10%), TP53 (8%), PALB2 (8%), CHEK2 (7%), CDH1 (3%), RAD51C (3%), MITF (2%), PMS2 (2%), RAD51D (2%), and TERT (2%). Monoallelic MUTYH pathogenic variants were found in 15%. After multivariable analysis, age of diagnosis (OR 0.89, 95% CI: 0.81–0.95, for each year increase), triple-negative subtype (OR 17.2, 95% CI: 3.74–114.72), and number of breast cancers in the family (OR 2.46, 95% CI 1.57–4.03, for each additional case) were associated with BRCA1 pathogenic variants. In the present study, a quarter of triple-negative breast cancer patients harbored a germline pathogenic variant and two-thirds of those were BRCA1 carriers.ConclusionsPrevalence and distribution of germline pathogenic variants in this Brazilian sample of breast cancer patients are mostly similar to other populations. However, there is a trend to an overrepresentation of TP53 pathogenic variants that merits confirmation in further studies. Early-onset breast cancer patients should be offered genetic counseling, particularly those with triple-negative subtype.

The aim of this work is to develop a device based on thin-layer chromatography coupled with surface-enhanced Raman spectroscopy (TLC-SERS) to analyze sulfamethoxazole (SMX) and trimethoprim (TMP) in commercial milk samples using chemometric tools. Samples were eluted in TLC plates, and a central composite design (CCD) of two factors was performed to optimize the gold nanoparticle dispersion on TLC plates for SERS, aiming at the detection of both drugs at concentrations close to their maximum residual limits (MRLs). Following the optimization, hyperspectral images from the SERS were captured of the TLC plates. Multivariate curve resolution (MCR-ALS) and independent component analysis (ICA) chemometric techniques were used to extract the signals of the analytes. All the samples presented recovery values of 81–128% for TMP. The quantification of SMX was not possible due to SERS suppression by an interferent. However, it was possible to detect SMX at a concentration of two times the MRL (8.0 × 10−7 mol·L−1). The results demonstrate that the TLC-SERS device is a potential tool for the quantification of TMP and the detection of SMX in milk.

O estudo teve abordagem plurimetodológica e abrangeu as cinco regiões do Brasil.

Changing wood color with physical treatment is an alternative method to avoid the use of chemicals harmful to the environment. However, despite this advantage, there are few studies on the chemistry and colorimetry of pre-hydrolyzed wood. The objective this research was to evaluate the content of extractives, lignin and holocellulose, lightness (L), red matrix (a*), yellow matrix (b*), saturation (C), and tonality angle (H) on strand board particles of pre-hydrolyzed pine and eucalyptus. Strand board particles measuring 23 x 90 x 0.30 mm in width, length, and thickness, respectively, were pre-hydrolyzed at 130, 150, and 170 °C during 7 or 21 minutes. The temperature had a significant effect on the chemical composition of wood. The pre-hydrolysis temperature of 170 °C resulted in higher chemical modifications. The pre-hydrolysis reduced the equilibrium moisture content of pine and eucalyptus particles, especially at 170 °C. Color modification due to pre-hydrolysis varied according to treatment conditions. Pine particles were more resistant color changing than the eucalyptus particles. [ABSTRACT FROM AUTHOR]

Thermal treatments have the effect of reducing the hygroscopicity and improving the resistance to microbiological attack of wood by the degradation of its chemical constituents. During the treatments, the mass of the wood is reduced, a factor that can affect the quality of the materials according to their use. The objective was to verify the effect of the thermal treatment variables on the thermogravimetric properties and the chemical composition of Eucalyptus grandis. The treatments were carried out in a vacuum oven with three atmosphere conditions - vacuum; N2; vacuum+N2 at temperatures of 140, 180 and 220 °C for 6 hours. It was observed that the mass loss during treatments differed only according to the temperatures used. The extractive content, total lignin and holocellulose presented significant changes only at 220°C in all three atmospheres. In the thermogravimetric analysis, the greatest value of residual mass was found in the treatment that used nitrogen and 220 °C, thus demonstrating that this treatment was more invasive, leading to the conclusion that the vacuum application can help to reduce the degradation of the constituents of the eucalypti wood. wood, which can lead to the production of thermally treated wood without great losses in the mechanical properties.

Brazilian productivity of eucalyptus trees is one of the highest in the world, and it has sustainable and viable conditions to supply the demand for woody biomass in a large quantity and diversity. This favorable situation makes Brazil one of the countries with the greatest potential for the production development of torrefied woody biomass on a commercial scale. Torrefaction is a mild pyrolysis applied to increase the energy quality of the feedstock. The aim of this study was to evaluate the Brazilian potential of woody biomass torrefaction for energy purposes. An experimental study was performed evaluating this thermal treatment of eucalyptus wood chips on a semi-continuous screw reactor developed by a Brazilian university. The results showed the increases in the energy quality of eucalyptus wood chips as a function of torrefaction temperature and time. This thermal treatment was technically feasible for the hygroscopicity reduction and the increases of fixed carbon and calorific values of the woody biomass.

ABSTRACT This study had the objective of comparing two methodologies of sampling, Simple Random Sampling (ACS) and Stratified Random Sampling (ACE) to determine the optimum number of roundwood samples to obtain the moisture content of the population. In order to achieve this goal, different percentages of allowable error (5,10,15 and 20%) were considered for each sampling methodology. In the conduction of this study, the samples were randomly taken from a lot of 250 steres of wood, 144 roundwood of three meters of length and distributed in four classes of diameter. Later, the moisture content of these samples was determined. And, from these values, the population estimates (average, standard deviation, variance, coefficient of variation, and standard error) by ACS and ACE methods, helped to determine the optimum number of roundwood (n) to be sampled from different percentages of allowable error adopted in this study at 95% probability. According to the results, the amount of roundwood to be sampled from ACS for each allowable error 5, 10, 15 and 20% was respectively 214, 55, 25 and 14. For the ACE (proportional allocation) the amount of roundwood was 141, 35, 16 and 9 for ACE (optimal allocation) this number was 136, 34, 15 and 8. It was concluded that the most indicated sampling method for this study, considering the allowable error, was the ACE method.

ABSTRACT The objective of this study was to determine the effects of tree age and of pith-bark and base-top positions on anatomical characteristics, and the effect of age on the chemical components of Schizolobium amazonicum (Paricá wood). The material was obtained from commercial plantations in the states of Maranhão and Pará with ages of 5, 7, 9 and 11 years. The first three logs (2.7 m length each) were obtained from the trees of each age. Only the effect of age was analyzed for the chemical characterization of the wood. The dimensions of the fibers increased along the pith-bark direction, while wall thickness decreased and the length of the fibers increased with aging. The width of the fibers and the lume diameter increased in the base-top direction. Paricá wood presented low extractive and hemicellulose content, and high cellulose content. The anatomical characteristics were more affected by the factors under study than the chemical composition, evidencing that the chemical elements of the wood are more stable with aging in relation to the anatomical characteristics.

The objective of this study was to determine the influence of the chemical properties (elemental composition, levels of extractives, lignin total and holocellulose) and the syringyl/guaiacyl, wood of different Eucalyptus genetic materials in the production of charcoal. It was used in the study four hybrid clones, at age of 7 years, 6 trees per clone, totaling 24 sampling units. There was significant difference, at 5% significance, between treatments for elemental composition, levels of extractives, lignin total and holocellulose and the wood syringyl/guaiacyl, as well as for gravimetric yields in charcoal, condensable gases and non-condensable gases, levels of fixed carbon and volatiles, and for the higher calorific value of charcoal. For the ash content and relative apparent density of charcoal were not observed significant differences at the same level of significance. Concluded that there is variability in the wood quality of four evaluated genetic materials, having the same, distinct correlations, thus the yield and the quality of charcoal. All genetic material presented satisfactorily gravimetric yield in charcoal and quality. The woods of clones with low syringyl/guaiacyl ratio showed an increase in yield in charcoal. The chemical composition of woods from the four clones, in general, showed no significant correlations, the 5% significance level, at the yield and quality of charcoal.

The city centre of São Paulo is increasingly a key site for local housing movements to challenge the rules and practices of spatial injustice in Brazil. In a context where housing for low-income groups is in short supply and, critically, continues to be characterized by highly skewed social and spatial distribution, occupied buildings have emerged as laboratories for the production of novel ways of inhabiting the city. This paper addresses on-going inner-city occupations as possible alternatives to the prevailing modes of conceiving and imagining urban renewal. As such, it outlines an approach to regeneration that engages processes of self-production and self-management as a means to achieve dignified homes and contest exclusionary urbanization.

ABSTRACT Growth in world demand for wood implies a search for new fast growing species with silvicultural potential, and in this scenario for native species such as Paricá . Thus, the objective of this study was determining the physical and mechanical wood properties of the Schizolobium amazonicum species (known as Paricá in Brazil). Trees were collected from commercial plantations located in the north of Brazil with ages of 5, 7, 9 and 11 years. Four logs from trees of each age in the longitudinal direction of the trees were obtained, and later a diametrical plank of each log was taken to manufacture the specimens which were used to evaluate some physical and mechanical properties of the wood. The basic density of Paricá was reduced in the basetop direction and no difference between the radial positions was observed, while the average basic density of this wood was characterized as low. The region close to the bark showed less longitudinal contraction and also greater homogeneity of this property along the trunk, while for tangential contraction the smallest variation was found in the region near the pith. Paricá wood contraction was characterized as low. Age influenced most of the mechanical properties, where logs from the base had the highest values of mechanical strength.

ABSTRACT The objective of this study was to evaluate the effect of the final torrefaction temperature on the energy properties of wood. It was applied four treatments with three replicates, consisting of untreated wood particles and particles torrefied at temperatures of 170, 220 and 260ºC. Better physical, chemical and thermal properties of the particles were observed as a function of the torrefaction temperature. Comparing with the control group, the particles torrefied at the highest final temperature (260ºC) showed significant differences in bulk density - from 239 to 396kg/m3; the equilibrium moisture content changed from 12.3 to 5.7%; fixed carbon content - from 13.0 to 24.8%; heating value - from 4,465 to 4,945kcal/kg; and energy density - from 882 to 1,727Mcal/m3. It was concluded that torrefaction had a positive effect on the energy properties of the wood.
