Cooperation among enterprises can bring overall and individual performance improvement, and a smooth coordination method is indispensable. However, due to the lack of customized coordination methods, cooperation in the downstream oil supply chain cannot be carried out smoothly. This paper intends to propose a multi-party coordination method to promote cooperation between oil shippers and pipeline operator by optimizing oil transportation, oil substitution and pipeline pricing schemes. An integrated game-theoretic modeling and analysis approach is developed to characterize the operation behaviors of all stakeholders in the downstream oil supply chain. The proposed mixed integer nonlinear programming model constrains supply and demand capacity, transportation routes, oil substitution rules and pipeline freight levels. Logarithm transformation and price discretization are introduced for model linear approximation. Simulation experiments are carried out in the oil distribution system in South China. The results show that compared to the business-as-usual scheme, the new scheme saves transportation cost by 3.48%, increases pipeline turnover by 5.7%, and reduces energy consumption and emissions by 7.66% and 6.77%. It is proved that the proposed method improves the revenue of the whole system, achieves fair revenue distribution, and also improves the energy and environmental benefits of the oil supply chain. Background: The purpose of this research was to investigate the effect of uric acid on hypertensive intracerebral hemorrhage (HICH) in young and middle-aged hypertensive patients at high altitude. Methods: High-altitude and low-altitude HICH patients were analyzed retrospectively, and the influence of uric acid on hemorrhage was analyzed by the correlations between age and uric acid, between age and hemoglobin, and between uric acid and hemoglobin. The individuals in the highand low-altitude groups were assigned to a young/middle-aged group and an elderly group, and the correlation between age and uric acid was analyzed. The risk factors for HICH were analyzed by logistic regression analysis. ROC curve analysis was used to investigate the relationship between risk factors and cerebral hemorrhage. Results: Age was negatively correlated with uric acid concentration in patients with HICH at high altitudes but not in patients with HICH at low altitudes. In addition, age was negatively correlated with hemoglobin in patients with HICH at high altitudes but not in patients with HICH at low altitudes. Uric acid was negatively correlated with age in young and middle-aged adults at high altitudes but not in elderly individuals. Diastolic pressure, uric acid and sex were risk factors for HICH in young and middle-aged adults at high altitudes. ROC curve showed that uric acid and diastolic pressure had diagnostic significance. Papillary thyroid carcinoma (PTC) is a highly prevalent cancer that typically exhibits indolent behavior and is associated with a favorable prognosis. The treatment of choice is surgical intervention; however, this approach carries the risk of complications, including scarring and loss of thyroid function. Although active surveillance can mitigate the risk of PTC overtreatment, the possibility of tumor growth and metastasis can elicit anxiety among patients. Ultrasoundguided thermal ablation has emerged as a safe and effective alternative for individuals who are ineligible for or decline surgery. As a new generation of high-performance batteries, lithium-ion batteries have found extensive applications in electric cars, as well as energy storage systems and various other industries. State of charge (SOC) estimation is one of the most important indicators. SOC estimation model of lithium-ion battery based on deep learning neural networks employs diverse external measurement parameters and internal battery parameters as input information, and adopts feed-forward neural network (FNN), convolutional neural network (CNN) and long short-term memory network (LSTM) as predictors to realize the accurate SOC estimation. The model based on deep learning neural networks takes into account the influence of various input parameters and can understand the state of the battery more comprehensively. By using FNN, CNN and LSTM networks, the influence of noise and instability of battery data on SOC estimation can be effectively avoided. Drought stress impairs crop growth and development. BEL1-like family transcription factors may be involved in plant response to drought stress, but little is known of the molecular mechanism by which these proteins regulate plant response and defense to drought stress. Here we show that the BEL1-like transcription factor GhBLH5-A05 functions in cotton (Gossypium hirsutum) response and defense to drought stress. Expression of GhBLH5-A05 in cotton was induced by drought stress. Overexpression of GhBLH5-A05 in both Arabidopsis and cotton increased drought tolerance, whereas silencing GhBLH5- A05 in cotton resulted in elevated sensitivity to drought stress. GhBLH5-A05 binds to cis elements in the promoters of GhRD20-A09 and GhDREB2C-D05 to activate the expression of these genes. GhBLH5- A05 interacted with the KNOX transcription factor GhKNAT6-A03. Co-expression of GhBLH5-A05 and GhKNAT6-A03 increased the transcription of GhRD20-A09 and GhDREB2C-D05. We conclude that GhBLH5-A05 acts as a regulatory factor with GhKNAT6-A03 functioning in cotton response to drought stress by activating the expression of the drought-responsive genes GhRD20-A09 and GhDREB2C-D05. Objective: Hounsfield units (HU), vertebral bone quality (VBQ), and bone mineral density (BMD) can all serve as predictive indicators for thoracolumbar fragility fractures. This study aims to explore which indicator provides better risk prediction for thoracolumbar fragility fractures. Methods: Patients who have received medical attention from The First Affiliated Hospital of Anhui Medical University for thoracolumbar fragility fractures were selected. A total of 78 patients with thoracolumbar fragility fractures were included in the study. To establish a control group, 78 patients with degenerative spinal diseases were matched to the fracture group on the basis of gender, age, and body mass index. The lumbar vertebral HU, the VBQ, and the BMD were obtained for all the 156 patients through computed tomography, magnetic resonance imaging, and dual-energy x-ray absorptiometry (DEXA). The correlations among these parameters were analyzed. The area under curve (AUC) analysis was employed to assess the predictive efficacy and thresholds of lumbar vertebral HU, VBQ, and BMD in relation to the risk of thoracolumbar fragility fractures. Results: Among the cohort of 156 patients, lumbar vertebral HU exhibited a positive correlation with BMD (p < 0.01). Conversely, VBQ showed a negative correlation with HU, BMD (p < 0.05). HU and BMD displayed a favorable predictive efficacy for thoracolumbar fragility fractures (p < 0.01), with HU (AUC = 0.863) showcasing the highest predictive efficacy, followed by the DEXA-measured BMD (AUC = 0.813). VBQ (AUC = 0.602) ranked lowest among the 3 indicators. The thresholds for predicting thoracolumbar fragility fractures were as follows: HU (88), VBQ (3.37), and BMD (0.81). Conclusion: All 3 of these indicators, HU, VBQ, and BMD, can predict thoracolumbar fragility fractures. Notably, lumbar vertebral HU exhibits the highest predictive efficacy, followed by the BMD obtained through DEXA scanning, with VBQ demonstrating the lowest predictive efficacy. Steam flooding is a widely used technique to enhance oil recovery of heavy oil. Thermal viscosity reduction and distillation effect are considered as two main displacement mechanisms in steam flooding process. However, the molecular composition understanding and contribution for oil production are still unclear. In this study, the composition analysis of the heavy oil was investigated in the core scale steam flooding process with the temperature from 120 to 280 °C. The crude oil, produced oils and residual oils were characterized comprehensively by gas chromatography and high-resolution mass spectrometry. It is found that steam flooding preferentially extracts aromatics and remains more resins in the residual oil. Viscosity reduction is the dominant mechanism when steam is injected at a low temperature. Large molecular heteroatoms with high carbon number and high double bond equivalent (DBE) are eluted into the produced oil, while compounds with low carbon number and low DBE are remained in the residual oil. As the steam temperature rises, the increased distillation effect results in the extraction of light hydrocarbons from the residual oil to the produced oil. More small heteroatoms with low carbon number and low DBE enter into the produced oil, especially in the none water cut stage. The compositional difference of produced oils is characterized in DBE versus carbon number distribution of the N and O containing compound classes. This work uses a variety of composition analysis methods to clarify the steam flooding mechanism and provides a novel understanding of steam flooding mechanisms with various temperatures and production stages from the molecular perspective. Short-term passenger flow prediction of urban rail transit based on historical data mining can guide the work of station organization, train scheduling, and passenger flow induction effectively and dynamically. We propose the RFBiLSTM prediction model, which combines the random forest algorithm (RF) and bi-directional long short-term memory neural network (BiLSTM). Firstly, the time series features of the flow are obtained by clustering algorithm and correlation analysis. Secondly, RF is used to obtain the importance of the features. Finally, we compare the prediction performance of 7 models and investigate the impact of feature selection on deep learning models. Through case analysis, the prediction accuracy of RF is the highest when using a single model for prediction, and the MAPE is 0.102. When using the combined prediction model, the MAPE of RF-BiLSTM is 0.074. The accuracy of RF-BiLSTM is better compared with the results of a single model. The performance of BiLSTM is improved by 46.6% after the features are optimized using feature selection methods. The ability of wheat to adapt to a wide range of environmental conditions is determined mostly by allelic diversity among genes regulating vernalization requirement. Vrn-1 is a major regulator of this requirement. In this study, two novel alleles of Vrn-A1 were discovered in Chinese cultivars: vrn-A1n was identified in two landraces, Jiunong 2 and Ganchun 16, and Vrn-A1o was detected in Duanhongmangmai. Both novel alleles showed a linked duplication in the promoter region. The common copy of these two alleles was identical to the recessive allele vrn-A1. Compared with the recessive allele vrn-A1, the other copy of vrn-A1n contained a 54-bp deletion in the promoter region and the distinct copy of Vrn-A1o contained an 11-bp deletion in the promoter region. In segregating populations in the greenhouse under nonvernalizing (20–25° C) and long-day (16 h light) conditions, plants with the novel vrn-A1n allele did not head earlier than those with the recessive vrn-A1 allele. However, plants that were either homozygous or heterozygous for the novel Vrn-A1o allele headed earlier than those with the recessive vrn-A1 allele. To identify the novel allele with the small-sized product and facilitate screening, a DNA marker for the novel dominant allele Vrn-A1o was designed. Analysis of the novel-allele distribution showed that two cultivars carrying the vrn-A1n allele were dispersed in the northwestern spring wheat zone, and 12 cultivars carrying the dominant Vrn-A1o allele were widely distributed in the northwestern spring wheat zone, Xinjiang winter and spring wheat zone, Yellow and Huai River valley winter wheat zone, and QinghaiTibetan Plateau spring and winter wheat zone. Petrocodon wui F.Wen & R.B.Zhang (Gesneriaceae), a typically lithophyte occurring in the Danxia areas of north-western Guizhou, China, is described and illustrated as new to science. The new species shows overall similarity with P. chishuiensis Z.B.Xin, F. Wen & S.B.Zhou, which is also its sister species, based on molecular evidence. The new species can be distinguished from P. chishuiensis by the elongated rhizome, the relatively long indumentum on the peduncle, the shape, size and indumentum of calyx lobes, the location of the stamens in the corolla tube and the shape, size and indumentum of the stigma. We provide a diagnosis, detailed description, photographic images and a table with taxonomic notes to distinguish several other morphologically similar Petrocodon species. [ABSTRACT FROM AUTHOR]

Aim: Drug-induced liver injury (DILI) poses significant challenges to clinical practice. Currently, there is no recommended therapy to treat DILI; therefore, it is vital to explore new therapeutic agents. This study aimed to investigate the efficacy and safety of silybin meglumine tablets in treating DILI. Methods: This study analysed 34 296 DILI cases assessed by the updated RUCAM from a nationwide database. A total of 301 patients with RUCAM scores ≥6 were directly enrolled in this study, while an additional 340 patients with RUCAM scores <6 who were adjudged as probable DILI by a panel of three hepatologists were also included in the analysis. The enrolled patients were divided into the silybin meglumine group and the control group. The propensity score matching (PSM) method was used to obtain comparable characteristics between the two groups. Results: There were 129 cases in the silybin meglumine group and 512 cases in the control group. After applying PSM, 129 matched pairs were obtained. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) resumption rates in the silybin meglumine group were significantly higher than the control group (58.91% vs. 20.93%, P ≤ .0001 and 63.49% vs. 37.50%, P ≤ .0001). The univariate and multivariate logistic regression analysis revealed that grouping factor (odds raio [OR], 5.42; 95% confidenxe interval [CI], 3.12–9.39; P < .0001 and OR, 6.10; 95% CI, 2.98– 12.48; P < .0001) and ALT levels (OR, 0.95; 95% CI, 0.93–0.98; P = .0015 and OR, 0.95; 95% CI, 0.92–0.99; P = .0157) were essential influencing factors for ALT normalization. Background and Objectives: The relationship between dietary folate intake and non-alcoholic fatty liver disease (NAFLD) is controversial. This study aimed to investigate the relationship between dietary folate equivalent (DFE) intake and NAFLD in U.S. adults. Methods and Study Design: Data from the National Health and Nutrition Examination Survey (NHANES) 2007-2014 were used. NAFLD was defined as a US fatty liver index (FLI) value ≥30. DFE intake was assessed by two 24-hour dietary recall interviews. Multivariable logistic regression models and restricted cubic spline models were used to investigate the association between DFE intake and NAFLD risk. Results: A total of 6,603 adult participants were included in this study. After adjusting for multiple confounding factors, the odds ratios and 95% confidence intervals of NAFLD for the highest quartile versus lowest quartile of DFE intake was 0.77(0.59-0.99). In stratified analyses by sex, age, and body mass index (BMI), there were statistically significant negative associations between DFE intake and NAFLD risk in women and participants with BMI ≥25. Dose-response analysis indicated a negative linear correlation between DFE intake and NAFLD risk. In recent years, the Corona Virus Disease 2019 (Covid-19) epidemic has raged around the world, with more than 500 million people diagnosed. Relevant medical research and analysis results on Covid-19 indicate that wearing masks is an effective method to prevent and restrain virus transmission. Mask detection stations have been set up in hospitals, railway stations, schools, where there is large crowd flow, but results are not as good as expected. In order to ameliorate pandemic preventing and control measures, a mask wearing detection algorithm YOLOv3-M3 was designed and proposed in this paper. The algorithm can effectively detect people without mask, while consequently reminding them. Firstly, we substituted the feature extraction network of YOLOv3 with MobileNetv3, a lightweight convolutional neural network. Secondly, we utilized K-Means++ to substitute the original ground truth clustering algorithm to improve prediction precision. In addition, the bounding box regression loss function was revised as CIoU loss function. This loss function solves the issues of overlapping between the ground truth and the anchor box, which has increased the training speed. After experiments, the precision of YOLOv3 algorithm on mAP 0.5 and mAP 0.75 is 93.5% and 71.9%, respectively. Elevating 3.1% and 2.6%, respectively, higher than that of YOLOv3 algorithm, and it was superior to SSD, SSD Lite, YOLOv3-Tiny and other one-stage object detection algorithms. The detection speed can reach 13.6 frame/s, which has met the requirements of pandemic prevention and control in most places and can be deployed on terminal devices for object detection. Objective: To compare and analyze the clinical effects of ultrasound-guided percutaneous nephrolithotripsy and X-ray-guided percutaneous nephrolithotripsy in the treatment of complex renal calculi without hydronephrosis. Methods: Eighty patients with multiple stones without hydronephrosis were admitted at Department of Ultrasound Diagnosis, The first medical center of Chinese PLA General Hospital from January 21, 2020 to December 21, 2020 randomly divided into two groups: experimental group and control group, with 40 cases in each group. Patients in the experimental group were treated with ultrasound-guided percutaneous nephrolithotomy, while those in the control group were treated with X-ray-guided percutaneous nephrolithotomy. The differences in operation time, channel establishment time, channel number, blood loss and stone clearance rate between the two groups were compared and analyzed. Venous blood was drawn before surgery and on the first day after surgery, and serum creatinine, urea nitrogen, blood β2- microglobulin, blood uric acid and other renal indexes were detected. Moreover, renal parenchymal injury was compared between the two groups by renal static imaging, and the incidence of postoperative complications such as pain, fever, urination through incision and injury of surrounding organs were compared and analyzed. Results: The operation time, channel establishment time, channel number and blood loss in the experimental group were significantly lower than those in the control group, with statistically significant differences (p<0.05). The postoperative renal injury score of the experimental group was 1.03±0.37, which was lower than 1.85±0.63 of the control group (p=0.00); Postoperative Cr, BUN, blood β 2-microglobulin and other indicators in the control group were significantly higher than those in the experimental group, with statistically significant differences (p<0.05). The incidence of peripheral organ injury in experimental group was lower than that in control group, with a statistically significant difference (p=0.04). Conclusion: Ultrasound-guided percutaneous nephrolithotomy is a safe and effective treatment regimen, boasting various advantages such as real-time monitoring of the surgical process, more accurate and clear channel establishment, avoidance of large vessel injury, shortening of surgical time, alleviation of kidney injury and reduction of surgical complications, which is more advantageous for the treatment of complex renal calculi. A 22 GHz water-maser survey was conducted towards 178 O-rich asymptotic giant branch (AGB) stars with the aim of identifying maser emission associated with the Sagittarius stellar stream. In this survey, maser emissions were detected in 21 targets, 20 of which were new detections. We studied the Galactic distributions of H2O- and SiO-maser-traced AGBs towards the Sgr orbital plane, and found an elongated structure towards the (l ,  b) ∼ (340°, 40°) direction. In order to verify its association with the Sagittarius tidal stream, we further studied the 3D motion of these sources, but found that, kinematically, these maser-traced AGBs are still Galactic disc sources rather than stream debris. In addition, we found a remarkable outward motion, ∼50 km s−1 away from the Galactic Centre of these maser-traced AGBs, but with no systermatic lag of rotational speed as reported in 2000 for solar-neighbourhood Miras. Oscillatory behaviors, which are ubiquitous in transcriptional regulatory networks, are often subject to inevitable biological noise. Thus, a natural question is how transcriptional regulatory networks can robustly achieve accurate oscillation in the presence of biological noise. Here, we search all two- and three-node transcriptional regulatory network topologies for those robustly capable of accurate oscillation against the parameter variability (extrinsic noise) or stochasticity of chemical reactions (intrinsic noise). We find that, no matter what source of the noise is applied, the topologies containing the repressilator with positive autoregulation show higher robustness of accurate oscillation than those containing the activator- inhibitor oscillator, and additional positive autoregulation enhances the robustness against noise. Nevertheless, the attenuation of different sources of noise is governed by distinct mechanisms: the parameter variability is buffered by the long period, while the stochasticity of chemical reactions is filtered by the high amplitude. Furthermore, we analyze the noise of a synthetic human nuclear factor κB (NF-κB) signaling network by varying three different topologies and verify that the addition of a repressilator to the activator-inhibitor oscillator, which leads to the emergence of high-robustness motif--the repressilator with positive autoregulation--improves the oscillation accuracy in comparison to the topology with only an activator- inhibitor oscillator. Most mammalian cells have an intrinsic circadian clock that coordinates metabolic activity with the daily rest and wake cycle. The circadian clock is known to regulate cell differentiation, but how continuous daily oscillations of the internal clock can control a much longer, multiday differentiation process is not known. Here, we simultaneously monitor circadian clock and adipocyte-differentiation progression live in single cells. Strikingly, we find a bursting behavior in the cell population whereby individual preadipocytes commit to differentiate primarily during a 12-h window each day, corresponding to the time of rest. Daily gating occurs because cells irreversibly commit to differentiate within only a few hours, which is much faster than the rest phase and the overall multiday differentiation process. The daily bursts in differentiation commitment result from a differentiation-stimulus driven variable and slow increase in expression of PPARG, the master regulator of adipogenesis, overlaid with circadian boosts in PPARG expression driven by fast, clock-driven PPARG regulators such as CEBPA. Our finding of daily bursts in cell differentiation only during the circadian cycle phase corresponding to evening in humans is broadly relevant, given that most differentiating somatic cells are regulated by the circadian clock. Pseudomonas fluorescens 2P24 is a beneficial plant root-associated microorganism capable of suppressing several soilborne plant diseases. The capacity of P. fluorescens to aggressively colonize the rhizosphere is an important requirement for its biocontrol trait. We previously found that the PcoI/PcoR quorum-sensing system (QS) is involved in regulating the rhizosphere colonization of P. fluorescens. Here, we revealed a sophisticated regulatory network that connects PcoR, RsaL, and MvaT proteins to fine-tune the PcoI/PcoR QS system. Our data showed that PcoR could directly bind to the promoter region of pcoI thereby inducing the PcoI/PcoR QS system, whereas RsaL binds simultaneously with PcoR to the promoter region of pcoI and represses the PcoR-dependent activation of pcoI gene. In addition, RsaL indirectly downregulates the expression of pcoR. Furthermore, we showed that disruption of mvaT enhanced the expression of pcoI, pcoR, and rsaL, whereas MvaT controls the PcoI/PcoR QS in a RsaL-independent manner. Overall, this study elucidates that PcoR, RsaL, and MvaT regulate the PcoI/PcoR QS through a multi-tiered regulatory mechanism and that PcoR is necessary in the RsaL- and MvaT-mediated repression on the expression of pcoI. The leucine-rich repeat receptor-like kinases (LRR-RLKs) compose a large gene family in plant genomes and implement essential functions in diverse plant physiology progress, including defence against pathogens. However, a systematic analysis of LRR-RLKs has not been accomplished in the economically important cucumber. 189 LRR-RLK genes were identified in the cucumber genome and further divided into 22 subgroups based on the sequence similarities in this study. A total of 31 segmental duplication events and 15 tandem duplication events were present in the genome, indicating that the two duplications were the main driving forces for the expansion of the LRR-RLK family in the cucumber. The expression profile analysis revealed that most of the CsLRR-RLKs were upregulated during a downy mildew infection, and resistant cucumbers comprised more upregulated CsLRR-RLKs than the sensitive lines. Trehalose is the major "blood sugar" of insects and it plays a crucial role in energy supply and as a stress protectant. The hydrolysis of trehalose occurs only under the enzymatic control of trehalase (Treh), which plays important roles in growth and development, energy supply, chitin biosynthesis, and abiotic stress responses. Previous reports have revealed that the vital hormone 20-hydroxyecdysone (20E) regulates Treh, but the detailed mechanism underlying 20E regulating Treh remains unclear. In this study, we investigated the function of HaTreh1 in Helicoverpa armigera larvae. The results showed that the transcript levels and enzymatic activity of HaTreh1 were elevated during molting and metamorphosis stages in the epidermis, midgut, and fat body, and that 20E upregulated the transcript levels of HaTrehl through the classical nuclear re-ceptor complex EcR-B1/USP1. HaTreh1 is a mitochondria protein. We also found that knockdown of HaTreh1 in the fifth- or sixth-instar larvae resulted in weight loss and increased mortality. Yeast two-hybrid, coimmunoprecipitation, and glutathione-S-transferase (GST) pull-down experiments demonstrated that HaTreh1 bound with ATP synthase subunit alpha (HaATPs-α) and that this binding increased under 20E treatment. Background: In recent years, augmented reality (AR), especially markerless augmented reality (MAR), has been used more prevalently to create training games in an attempt to improve humans' cognitive functions. This has been driven by studies claiming that MAR provides users with more immersive experiences that are situated in the real world. Currently, no studies have scientifically investigated the immersion experience of users in a MAR cognitive training game. Moreover, there is an observed lack of instruments on measuring immersion in MAR cognitive training games. Objective: This study, using two existing immersion questionnaires, investigates students' immersion experiences in a novel MAR n-back game. Methods: The n-back task is a continuous performance task that taps working memory (WM) capacity. We compared two versions of n-back training. One was presented in a traditional 2D format, while the second version used MAR. There were 2 experiments conducted in this study that coordinated with 2 types of immersion questionnaires: the modified Immersive Experiences Questionnaire (IEQ) and the Augmented Reality Immersion (ARI) questionnaire. Two groups of students from two universities in China joined the study, with 60 participants for the first experiment (a randomized controlled experiment) and 51 participants for the second. Results: Both groups of students experienced immersion in the MAR n-back game. However, the MAR n-back training group did not experience stronger immersion than the traditional (2D) n-back control group in the first experiment. The results of the second experiment showed that males felt deeply involved with the AR environment, which resulted in obtaining higher levels of immersion than females in the MAR n-back game. Conclusions: Both groups of students experienced immersion in the MAR n-back game. Objective: We used U-shaped convolutional neural network (U_Net) multi-constraint image segmentation method to compare the diagnosis and imaging characteristics of tuberculosis and tuberculosis with lung cancer patients with Computed Tomography (CT). Methods: We selected 160 patients with tuberculosis from the severity scoring (SVR) task is provided by Image CLEF Tuberculosis 2019. According to the type of diagnosed disease, they were divided into tuberculosis combined with lung cancer group and others group, all patients were given chest CT scan, and the clinical manifestations, CT characteristics, and initial suspected diagnosis and missed diagnosis of different tumor diameters were observed and compared between the two groups. Results: There were more patients with hemoptysis and hoarseness in pulmonary tuberculosis combined with lung cancer group than in the pulmonary others group (P<0.05), and the other symptoms were not significantly different (P>0.05). Tuberculosis combined with lung cancer group had fewer signs of calcification, streak shadow, speckle shadow, and cavitation than others group; however, tuberculosis combined with lung cancer group had more patients with mass shadow, lobular sign, spines sign, burr sign and vacuole sign than others group. Conclusion: The symptoms of hemoptysis and hoarseness in pulmonary tuberculosis patients need to consider whether the disease has progressed and the possibility of lung cancer lesions. CT imaging of pulmonary tuberculosis patients with lung cancer usually shows mass shadows, lobular signs, spines signs, burr signs, and vacuoles signs. It can be used as the basis for its diagnosis. Numerous animal lineages have maternally inherited symbionts that are required for host reproduction and growth. Endosymbionts also pose a risk to their hosts because of the mutational decay of their genomes through genetic drift or to selfish mutations that favor symbiont fitness over host fitness. One model for heritable endosymbiosis is the association of aphids with their obligate bacterial symbiont, Buchnera. We experimentally established heteroplasmic pea aphid matrilines containing pairs of closely related Buchnera haplotypes and used deep sequencing of diagnostic markers to measure haplotype frequencies in successive host generations. These frequencies were used to estimate the effective population size of Buchnera within hosts (i.e., the transmission bottleneck size) and the extent of within-host selection. The within-host effective population size was in the range of 10 to 20, indicating a strong potential for genetic drift and fixation of deleterious mutations. Remarkably, closely related haplotypes were subject to strong within-host selection, with selection coefficients as high as 0.5 per aphid generation. In one case, the direction of selection depended on the thermal environment and went in the same direction as between-host selection. In another, a new mutant haplotype had a strong within-host advantage under both environments but had no discernible effect on host-level fitness under laboratory conditions. Thus, within-host selection can be strong, resulting in a rapid fixation of mutations with little impact on host-level fitness. Many bacteria and other organisms carry out fermentations forming acetate. These fermentations have broad importance for foods, agriculture, and industry. They also are important for bacteria themselves because they often generate ATP. Here, we found a biochemical pathway for forming acetate and synthesizing ATP that was unknown in fermentative bacteria. We found that the bacterium Cutibacterium granulosum formed acetate during fermentation of glucose. It did not use phosphotransacetylase or acetate kinase, enzymes found in nearly all acetate-forming bacteria. Instead, it used a pathway involving two different enzymes. The first enzyme, succinyl coenzyme A (succinyl-CoA):acetate CoA-transferase (SCACT), forms acetate from acetyl-CoA. The second enzyme, succinyl-CoA synthetase (SCS), synthesizes ATP. We identified the genes encoding these enzymes, and they were homologs of SCACT and SCS genes found in other bacteria. The pathway resembles one described in eukaryotes, but it uses bacterial, not eukaryotic, gene homologs. To find other instances of the pathway, we analyzed sequences of all biochemically characterized homologs of SCACT and SCS (103 enzymes from 64 publications). Homologs with similar enzymatic activity had similar sequences, enabling a largescale search for them in genomes. We searched nearly 600 genomes of bacteria known to form acetate, and we found that 6% encoded homologs with SCACT and SCS activity. This included .30 species belonging to 5 different phyla, showing that a diverse range of bacteria encode the SCACT/SCS pathway. The protein–protein interaction (PPI) is a basic strategy for life to operate. The analysis of PPIs in multicellular organisms is very important but extremely challenging because PPIs are particularly dynamic and variable among different development stages, tissues, cells, and even organelles. Therefore, understanding PPI needs a good resolution of time and space. More importantly, understanding in vivo PPI needs to be realized in situ. Proximity-based biotinylation combined with mass spectrometry (MS) has emerged as a powerful approach to study PPI networks and protein subcellular compartmentation. TurboID, the newly engineered promiscuous ligase, has been reported to label proximate proteins effectively in various species. In Drosophila, we systematically apply TurboID-mediated biotinylation in a wide range of developmental stages and tissues, and demonstrate the feasibility of TurboID-mediated labeling system in desired cell types. For a proof-of-principle, we use the TurboID-mediated biotinylation coupled with MS to distinguish CTP synthase with or without the ability to form filamentous cytoophidia, retrieving two distinct sets of proximate proteomes. Therefore, this makes it possible to map PPIs in vivo and in situ at a defined spatiotemporal resolution, and demonstrates a referable resource for cytoophidium proteome in Drosophila. A new method for producing biomass seedling trays with cow manure was developed based on heating to above the glass transition point of the lignin in cow dung. The maize seedling tray, manufactured through heat compression molding, could potentially meet the demand for transplanting maize seedlings. A scanning electron microscope and universal testing machine were used to compare the changes in the internal structure and mechanical properties of the seedling tray before and after the seeding period through a compression molding technique at normal temperature before seeding and a heat compression molding technique after seeding. The results showed that the strength of the seedling tray was mainly derived from the mechanical setting force of the fiber laminated in the seedling tray. Meanwhile, the moisture in the seedling tray hindered the lignin from filling in the gaps between the stem fibers in cow manure, as well as it reduced the protective effect of lignin on the laminated inlay structure of the stem fibers in cow manure. Therefore, under the premise that the material could be completely filled in the mold, the study concluded that a lower moisture content resulted in better strength and water resistance of the seedling tray. Caenorhabditis elegans expresses human Werner syndrome protein (WRN) orthologs as two distinct proteins: MUT-7, with a 30-50 exonuclease domain, and CeWRN-1, with helicase domains. How these domains cooperate remains unclear. Here, we demonstrate the different contributions of MUT-7 and CeWRN-1 to 22G small interfering RNA (siRNA) synthesis and the plasticity of neuronal signaling. MUT-7 acts specifically in the cytoplasm to promote siRNA biogenesis and in the nucleus to associate with CeWRN-1. The import of siRNA by the nuclear Argonaute NRDE-3 promotes the loading of the heterochromatin-binding protein HP1 homolog HPL-2 onto specific loci. This heterochromatin complex represses the gene expression of the guanylyl cyclase ODR-1 to direct olfactory plasticity in C. elegans. Our findings suggest that the exonuclease and helicase domains of human WRN may act in concert to promote RNA-dependent loading into a heterochromatin complex, and the failure of this entire process reduces plasticity in postmitotic neurons. Background: A diverse physician workforce improves the quality of care for all patients, and there is a need for greater diversity in orthopaedic surgery. It is important that medical students of diverse backgrounds be encouraged to pursue the specialty, but to do so, we must understand students' perceptions of diversity and inclusion in orthopaedics. We also currently lack knowledge about how participation in an orthopaedic clinical rotation might influence these perceptions.Questions/purposes: (1) How do the perceptions of diversity and inclusion in orthopaedic surgery compare among medical students of different gender identities, races or ethnicities, and sexual orientations? (2) How do perceptions change after an orthopaedic clinical rotation among members of demographic groups who are not the majority in orthopaedics (that is, cis-gender women, underrepresented racial minorities, other racial minorities, and nonheterosexual people)?Methods: We surveyed students from 27 US medical schools who had completed orthopaedic rotations. We asked about their demographic characteristics, rotation experience, perceptions of diversity and inclusion in orthopaedics, and personal views on specialty choice. Questions were derived from diversity, equity, and inclusion climate surveys used at major academic institutions. Cis-gender men and cis-gender women were defined as those who self-identified their gender as men or women, respectively, and were not transgender. Forty-five percent (59 of 131) of respondents were cis-men and 53% (70 of 131) were cis-women; 49% (64 of 131) were white, 20% (26 of 131) were of underrepresented racial minorities, and 31% (41 of 131) were of other races. Eighty-five percent (112 of 131) of respondents were heterosexual and 15% (19 of 131) reported having another sexual orientation. We compared prerotation and postrotation perceptions of diversity and inclusion between majority and nonmajority demographic groups for each demographic domain (for example, cis-men versus cis-women). We also compared prerotation to postrotation perceptions within each nonmajority demographic group. To identify potential confounding variables, we performed univariate analysis to compare student and rotation characteristics across the demographic groups, assessed using an alpha of 0.05. No potential confounders were identified. Statistical significance was assessed at a Bonferroni-adjusted alpha of 0.0125. Our estimated response percentage was 26%. To determine limitations of nonresponse bias, we compared all early versus late responders and found that for three survey questions, late responders had a more favorable perception of diversity in orthopaedic surgery, whereas for most questions, there was no difference.Results: Before rotation, cis-women had lower agreement that diversity and inclusion are part of orthopaedic culture (mean score 0.96 ± 0.75) compared with cis-men (1.4 ± 1.1) (mean difference 0.48 [95% confidence interval 0.16 to 0.81]; p = 0.004), viewed orthopaedic surgery as less diverse (cis-women 0.71 ± 0.73 versus cis-men 1.2 ± 0.92; mean difference 0.49 [95% CI 0.20 to 0.78]; p = 0.001) and more sexist (cis-women 1.3 ± 0.92 versus cis-men 1.9 ± 1.2; mean difference 0.61 [95% CI 0.23 to 0.99]; p = 0.002), believed they would have to work harder than others to be valued equally (cis-women 2.8 ± 1.0 versus cis-men 1.9 ± 1.3; mean difference 0.87 [95% CI 0.45 to 1.3]; p < 0.001), and were less likely to pursue orthopaedic surgery (cis-women 1.4 ± 1.4 versus cis-men 2.6 ± 1.1; mean difference 1.2 [95% CI 0.76 to 1.6]; p < 0.001). Before rotation, underrepresented minorities had less agreement that diversity and inclusion are part of orthopaedic surgery culture (0.73 ± 0.72) compared with white students (1.5 ± 0.97) (mean difference 0.72 [95% CI 0.35 to 1.1]; p < 0.001). Many of these differences between nonmajority and majority demographic groups ceased to exist after rotation. Compared with their own prerotation beliefs, after rotation, cis-women believed more that diversity and inclusion are part of orthopaedic surgery culture (prerotation mean score 0.96 ± 0.75 versus postrotation mean score 1.2 ± 0.96; mean difference 0.60 [95% CI 0.22 to 0.98]; p = 0.002) and that orthopaedic surgery is friendlier (prerotation 2.3 ± 1.2 versus postrotation 2.6 ± 1.1; mean difference 0.41 [95% CI 0.14 to 0.69]; p = 0.004), more diverse (prerotation 0.71 ± 0.73 versus postrotation 1.0 ± 0.89; mean difference 0.28 [95% CI 0.08 to 0.49]; p = 0.007), less sexist (prerotation 1.3 ± 0.92 versus postrotation 1.9 ± 1.0; mean difference 0.63 [95% CI 0.40 to 0.85]; p < 0.001), less homophobic (prerotation 2.1 ± 1.0 versus postrotation 2.4 ± 0.97; mean difference 0.27 [95% CI 0.062 to 0.47]; p = 0.011), and less racist (prerotation 2.3 ± 1.1 versus postrotation 2.5 ± 1.1; mean difference 0.28 [95% CI 0.099 to 0.47]; p = 0.003). Compared with before rotation, after rotation cis-women believed less that they would have to work harder than others to be valued equally on the rotation (prerotation 2.8 ± 1.0 versus postrotation 2.5 ± 1.0; mean difference 0.31 [95% CI 0.12 to 0.50]; p = 0.002), as did nonheterosexual students (prerotation 2.4 ± 1.4 versus postrotation 1.8 ± 1.3; mean difference 0.56 [95% 0.21 to 0.91]; p = 0.004). Underrepresented minority students saw orthopaedic surgery as less sexist after rotation compared with before rotation (prerotation 1.5 ± 1.1 versus postrotation 2.0 ± 1.1; mean difference 0.52 [95% CI 0.16 to 0.89]; p = 0.007).Conclusion: Even with an estimated 26% response percentage, we found that medical students of demographic backgrounds who are not the majority in orthopaedics generally perceived that orthopaedic surgery is less diverse and inclusive than do their counterparts in majority groups, but these views often change after a clinical orthopaedic rotation.Clinical Relevance: These perceptions may be a barrier to diversification of the pool of medical student applicants to orthopaedics. However, participation in an orthopaedic surgery rotation is associated with mitigation of many of these negative perceptions among diverse students. Medical schools have a responsibility to develop a diverse workforce, and given our findings, schools should promote participation in a clinical orthopaedic rotation. Pseudomonas fluorescens 2P24 is a rhizosphere bacterium that protects many crop plants against soilborne diseases caused by phytopathogens. The PcoI/PcoR quorum-sensing (QS) system and polyketide antibiotic 2,4-diacetylphloroglucinol (2,4-DAPG) are particularly relevant to the strain's biocontrol potential. In this study, we investigated the effects of c-di-GMP on the biocontrol activity of strain 2P24. The expression of the Escherichia coli diguanylate cyclase (YedQ) and phosphodiesterase (YhjH) in P. fluorescens 2P24 significantly increased and decreased the cellular concentration of c-di-GMP, respectively. The production of the QS signals N-acyl homoserine lactones (AHLs) and 2,4-DAPG was negatively regulated by c-di-GMP in 2P24. The regulatory proteins RsmA and RsmE were positively regulated by c-di-GMP. Genomic analysis revealed that 2P24 has 23 predicted proteins that contain c-di-GMP-synthesizing or -degrading domains. Among these proteins, C0J56_12915, C0J56_13325, and C0J56_27925 contributed to the production of c-di-GMP and were also involved in the regulation of the QS signal and antibiotic 2,4-DAPG production in P. fluorescens. Overexpression of C0J56_12915, C0J56_13325, and C0J56_27925 in 2P24 impaired its root colonization and biocontrol activities. Renal interstitial fibrosis (RIF) is characterized by excessive extracellular matrix deposition and involves epithelial-mesenchymal transition (EMT). The lncRNA taurine-upregulated gene 1 (TUG1) participates in EMT in several cancers; however, the effect and underlying mechanism of TUG1 in RIF-related EMT remain unclear. Here, we explored the mechanisms by which TUG1 modulates RIF. An in vivo model of renal fibrosis was established by unilateral ureteral obstruction in Balb/c mice. Human renal proximal tubular epithelial (HK-2) cells treated with transforming growth factor (TGF)-β1 were used to induce the in vitro model. Morphological changes and TUG1 expression were assessed. HK-2 cells were transfected with siRNA to silence TUG1. Western blot analysis, immunofluorescence staining, cell proliferation, and migration assays were performed to examine TGF-β1-induced changes in EMT markers and EMT-like cell behaviors. TUG1 and β-catenin (CTNNB1) levels were significantly upregulated, whereas miR-141-3p was significantly downregulated, during EMT in vitro and in vivo. TUG1 knockdown or miR-141-3p overexpression supported the epithelioid morphology of HK-2 cells while enhancing the downregulation of E-cadherin and upregulation of vimentin, α-smooth muscle actin, and β-catenin levels in TGF-β1-treated HK-2 cells. TUG1 knockdown promoted the proliferation and decreased the migration of HK-2 cells and enhanced the downregulation of miR-141-3p levels in TGF-β1-treated HK-2 cells. TUG1 directly targeted miR-141-3p, and miR-141-3p was directly bound to CTNNB1. Downregulation of miR-141-3p inhibited TUG1 silencing-induced suppression of EMT. In conclusion, TUG1 promotes EMT in TGF-β1-induced HK-2 cells via upregulation of β-catenin levels by sponging miR-141-3p, suggesting a novel therapeutic candidate for RIF. [ABSTRACT FROM AUTHOR]

Objective: Lumbar discectomy is commonly performed for symptomatic lumbar disc herniation. We aimed to examine prescribing patterns and risk factors for chronic opioid use following lumbar discectomy. Methods: Using a private insurance claims database, patients were identified who underwent primary lumbar discectomy from 2010–2015 and had 1-year of continuous enrollment postoperatively. Patients were excluded with spinal fusion. The strength of opioid prescriptions was quantified using morphine milligram equivalents daily (MMED). Univariate and multivariate logistic regression models were built to examine risk factors associated with chronic postoperative opioid use. Results: A total of 5,315 patients were included in the study (mean age, 59 years; 50% female). 1,198 of patients (23%) used chronic opioids postoperatively. Chronic opioid use declined significantly from 27% in 2010 to 17% in 2015, p<0.001. In addition, there were significantly fewer patients receiving high and very high-dose opioid prescriptions from 2010–2015, p<0.001. The median duration that patients used opioids postoperatively was 211 days in 2010 (interquartile range [IQR], 29–356 days), and decreased significantly to 44 days (IQR, 10–294 days) in 2015. The strongest factors associated with chronic opioid use were preoperative opioid use (odds ratio [OR], 4.0), drug abuse (OR, 2.6), depression (OR, 1.6), surgery in the west (OR, 1.6) or south (OR, 1.6), anxiety (OR, 1.5), or 30-day readmission (OR, 1.4). Conclusion: Chronic opioid use following primary lumbar discectomy has declined from 2010–2015. A variety of factors are associated with chronic opioid use. L-homoserine, which is one of the few amino acids that is not produced on a large scale by microbial fermentation, plays a significant role in the synthesis of a series of valuable chemicals. In this study, systematic metabolic engineering was applied to target E. coli W3110 for production of L-homoserine. Initially, the basic L-homoserine producer was engineered through the strategies overexpressing thrA (encoding homoserine dehydrogenase), removing the degradative and competitive pathways by knocking out metA (encoding homoserine O-succinyltransferase) and thrB (encoding homoserine kinase), reinforcing the transport system and redirecting the carbon flux by deleting iclR (encoding isocitrate lyase regulator). The resulted strain constructed by these strategies yielded 3.21 g/L of L-homoserine in batch cultures. Moreover, based on CRISPR-Cas9/dCas9 mediated gene repression for 50 genes, the iterative genetic modifications of biosynthesis pathways stepwise improved the L-homoserine yield. The rational integration of glucose uptake and recovery of L-glutamate increased L-homoserine production to 7.25 g/L in shake-flask cultivation. Furthermore, the intracellular metabolic analysis further provided target for strain modification by introducing the anaplerotic route afforded by pyruvate carboxylase to oxaloacetate formation, which resulted in accumulating 8.54 g/L L-homoserine (0.33 g/g glucose, 62.4% of the maximum theoretical yield) in shake-flask cultivation. The epithelial mesenchymal transition (EMT) plays significant roles in the progression of cancer and fibrotic disease. Moreover, this process is reversible, resulting in mesenchymal epithelial transition (MET), which plays an important role in cancer metastasis. There is a lack of methods to trace and target EMT cells using synthetic biology circuits, which makes it difficult to study the cell fate or develop targeted treatments. In this study, we introduced responsive EMT sensing circuits, which sense the EMT using specific promoters that respond to transcription factors typical of EMT activation (EMT-TFs). The transcriptional strength of EMT-sensing promoters decreased more than 13-fold in response to the overexpression of the EMT-TF. Then, the NOT gate circuits were built by placing the tetR transcription repressor under the control of EMT sensing promoters and expressed an output signal using the constitutive CMV promoter modified with tetO sites This circuit is named EMT sensing and responding circuits. When the EMT transcription factors was present, we observed a 5.8-fold signal increase in the system. Then, we successfully distinguished mesenchymal breast cancer cells from epithelial cancer cells and repressed the proliferation of EMT tumor cells using our circuits. The EMT sensing and responding circuits are promising tools for the identification of EMT cells, which is crucial for EMT-related disease therapy and investigating the mechanisms underlying the reversible EMT process. This is the first report on the coating of diamond dicing blades with metallic glass (MG) coating to reduce chipping when used to cut Si, SiC, sapphire, and patterned sapphire substrates (PSS). The low coefficient-of-friction (CoF) of Zr-based MG-coated dicing blades was shown to reduce the number and size of chips, regardless of the target substrate. Overall, SiC, sapphire and PSS were most affected by chipping, due to the fact that higher cutting forces were needed for the higher hardness of SiC, sapphire and PSS. Compared to the bare blade, the MG coating provided the following reductions in chipping area: Si (~ 23%), SiC (~ 36%), sapphire (~ 45%), and PSS (~ 33%). The proposed coating proved particularly effective in reducing chips of larger size (> 41 µm in chipping width), as indicated by an ~ 80% reduction when cutting sapphire. Small variations in kerf angle and depth demonstrate the durability of the coated blades, which would no doubt enhance consistency in dicing performance and extend the blade lifespan. Here, we present coupled geochemical and Sr-Nd-Pb-S isotopic data of Early Cretaceous primitive gabbros from the North China craton. Strikingly, these rocks have highly unradiogenic lead compositions (206Pb/204Pb = 16.58 ± 0.24) and anchor one extreme end member (low 206Pb/204Pb and 143Nd/144Nd) in the global array of oceanic-island volcanics. Our study shows that they originated from an Archean fluid-metasomatized refractory peridotite source, in which highly unradiogenic lead was preferentially released with subducted Archean seawater and sequestered into recrystallized sulfides at shallow mantle depths. Sulfide/silicate partition coefficients for lead show a negative pressure dependence: Lead is more enriched in sulfide with decreasing pressure. Sulfide-bearing and iron-poor harzburgite as well as dunite residues at shallow mantle are expected to develop low U/Pb (and thereby low time-integrated 206Pb/204Pb) relative to a deeper upper-mantle source. There is a growing interest in the role of gut microbiota in the pathophysiology of several diseases, including coronary artery diseases (CAD). Gut microorganisms may produce beneficial effects in myocardial ischemia either directly in the form of exogenous administration or indirectly by acting on fiber-rich food to produce important cardioprotective components. The harmful effects of gut microbiota in CAD are due to alteration in their composition with a significant decrease in Bacteroidetes and an increase in Firmicutes, Escherichia, Shigella, and Enterococcus. The altered microbiota may produce potentially toxic metabolites, including trimethylamine-N-oxide (TMAO). Indeed, the fasting plasma levels of TMAO are directly correlated to increased risk of major cardiovascular events in CAD patients, and it is proposed as a potential biomarker to predict the onset of major cardiovascular events. It is concluded that the change in the composition of gut microbiota in CAD patients may predispose to more harmful effects. The cell wall mechanical properties are an important indicator for evaluating the overall mechanical properties of natural bamboo fibers. Using the nanoindentation technique, the variation of the mechanical properties of the fiber cell wall of Bambusa pervariabilis culms with different ages and different positions (both radial and longitudinal) was studied. Moreover, x-ray diffraction (XRD) was employed to measure the microfibril angle (MFA), and the correlation between the MFA and the mechanical properties of the fiber cell wall. The results showed that there was a remarkable difference in the fiber cell wall mechanical properties at different ages and at different radial and longitudinal positions. However, at different ages and at different positions, the absolute value of variation of MFA was less than 1° and was very minor. Furthermore, there was no significant correlation between the fiber cell wall mechanics and MFA, indicating that the mechanical property of the fiber cell walls might be synergistically affected by many factors. The distribution of film cooling effectiveness in supersonic mainstream of circle- rectangular Convergent-Divergent transition has been numerically investigated under different pressure ratios. The shock wave exerted superior influence on film cooling. In supersonic main flow, extra compression waves formed in upstream of the film holes, resulted by the obstruction of the multiple cooling jets. This exerted extra pressure to the boundary-layer, induced adverse pressure gradient, and led to weakening of the film flow attachment ability and decreasing of local cooling effectiveness. Bow oblique shock wave occurred in front of holes, the two oblique bow shaped low pressure zones formed on both sides of the hole, and low cooling effectiveness zones appeared accordingly. The inefficient region at the leading edge of the hole destroyed the film developing between holes, decreased the cooling effectiveness accumulation in the rear part. The decrease of hole incline angle caused an increase of cooling effectiveness, which reduced reverse velocity gradient caused by shock wave in the boundary-layer and improved film attachment. Effect of film hole diameter to discharge coefficients and film cooling effectiveness of the lamilloy were experimentally investigated at the blowing ratios ranging from 0.6 to 2.5. Generally, the cooling effectiveness on the test surface is increased with the film hole diameters and blowing ratios increasing, and the variation tendency along the streamwise through the centerline of film holes also keeps consistent. In the upstream, all configurations have a relatively lower cooling effectiveness, however, with the flows to the downstream, cooling effectiveness grows gradually, and the peak values of effectiveness appeared periodically, the position matches the film hole positions. Besides, between the film holes, fluctuation exists simultaneously. By comparison, the cooling effectiveness and discharge coefficients are both increased with the blowing ratio with same film hole diameters, Moreover, under the same blowing ratios, the cooling effectiveness and discharge coefficients are getting higher when hole diameters increase. In the mouse, septation occurs postnatally and is thought to require the alveolar myofibroblast (AMF). Though abundant during alveologenesis, markers for AMFs are minimally detected in the adult. After septation, the alveolar walls thin to allow efficient gas exchange. Both loss of AMFs or retention and differentiation into another cell type during septal thinning have been proposed. Using a novel Fgf18:CreERT2 allele to lineage trace AMFs, we demonstrate that most AMFs are developmentally cleared during alveologenesis. Lung mesenchyme also contains other poorly described cell types, including alveolar lipofibroblasts (ALF). We show that Gli1:CreERT2 marks both AMFs as well as ALFs, and lineage tracing shows that ALFs are retained in adult alveoli while AMFs are lost. We further show that multiple immune cell populations contain lineage-labeled particles, suggesting a phagocytic role in the clearance of AMFs. The demonstration that the AMF lineage is depleted during septal thinning through a phagocytic process provides a mechanism for the clearance of a transient developmental cell population. Paraphlomis kuankuoshuiensis (Lamiaceae), a new species found in the limestone areas of northern Guizhou, China, is described and illustrated in this paper. Based on its tubular-campanulate calyx, this taxon should be a member of sect. Paraphlomis Prain. The new species resembles P. patentisetulosa C.Y. Wu & H. W. Li, P. hispida C.Y. Wu, and P. hirsutissima C.Y. Wu & H.W. Li, but differs from these three taxa in the following aspects: the stems are very short (<7 cm), with one or two short internodes, giving the impression of having a tuft of basal leaves; it has sparsely setose hairs on the outer surface of the calyces and short fruiting calyces. The low-pathogenic H7N9 influenza viruses that emerged in 2013 acquired an insertion of four amino acids in their hemagglutinin cleavage site and thereby became highly pathogenic to chickens in 2017. Previous studies indicated that these highly pathogenic H7N9 viruses are virulent in chickens but have distinct pathotypes in mice. A/chicken/Guangdong/SD098/2017 (CK/SD098) is avirulent, with a 50% mouse lethal dose (MLD50) of >7.5 log10 50% egg infectious dose (EID50), whereas A/chicken/Hunan/S1220/2017 (CK/S1220) is virulent in mice, with an MLD50 of 3.2 log10 EID50. In this study, we explored the genetic determinants that contribute to the difference in virulence between these two H7N9 viruses by generating a series of reassortants and mutants in the CK/S1220 virus background and testing their virulence in mice. We found that the reassortant CK/1220-SD098-NP, carrying the nucleoprotein (NP) of CK/SD098, was avirulent in mice, with an MLD50 of >107.5 EID50. The NPs of these two viruses differ by two amino acids, at positions 286 and 437. We further demonstrated that the amino acid mutations A286V and T437M of NP independently slowed the process of NP import to and export from the nucleus and thus jointly impaired the viral life cycle and attenuated the virulence of these H7N9 viruses in mice. Based on a recently developed approach that can recognize both persistent blocking and ridge events effectively, the contributions of the frequency of these persistent events (FOPE) over different regions in Eurasia to precipitation over eastern China were investigated. The results reveal that, the FOPE over the longitudinal range of 110°-130°E, near the Stanovoy Mountains and the Okhotsk Sea, is significantly correlated with precipitation over the middle and lower reaches of the Yangtze River (MLRYR) during summer, particularly in August. The preceding full July (or 1-20 July) mean Balkhash Lake-Caucasus geopotential height index, which measures the combined effect of the Balkhash Lake and Caucasus geopotential height anomalies, is closely related to the August geopotential height anomaly around the Stanovoy Mountains and the Okhotsk Sea, and can therefore reflect the August 110°-130°E FOPE. The predictability based on this preceding atmospheric signal seems to be attributable to slow-varying atmospheric processes on a subseasonal (20-day mean) time scale. On this time scale, the Balkhash Lake and Caucasus geopotential height anomalies occur prior to, and seem to modulate, the geopotential height anomaly around the Stanovoy Mountains and the associated 110°-130°E FOPE through an eastward extension and through exciting a positive-negative-positive pattern in 500-hPa geopotential heights, respectively. As a result of the slow-varying atmospheric processes, this preceding atmospheric signal performs well in predicting the August 110°-130°E FOPE, which also facilitates the prediction of the MLRYR precipitation. The thermal tolerance of an organism limits its ecological and geographic ranges and is potentially affected by dependence on temperature-sensitive symbiotic partners. Aphid species vary widely in heat sensitivity, but almost all aphids are dependent on the nutrient-provisioning intracellular bacterium Buchnera, which has evolved with aphids for 100 million years and which has a reduced genome potentially limiting heat tolerance. We addressed whether heat sensitivity of Buchnera underlies variation in thermal tolerance among 5 aphid species. We measured how heat exposure of juvenile aphids affects later survival, maturation time, and fecundity. At one extreme, heat exposure of Aphis gossypii enhanced fecundity and had no effect on the Buchnera titer. In contrast, heat suppressed Buchnera populations in Aphis fabae, which suffered elevated mortality, delayed development and reduced fecundity. Likewise, in Acyrthosiphon kondoi and Acyrthosiphon pisum, heat caused rapid declines in Buchnera numbers, as well as reduced survivorship, development rate, and fecundity. Fecundity following heat exposure is severely decreased by a Buchnera mutation that suppresses the transcriptional response of a gene encoding a small heat shock protein. Similarly, absence of this Buchnera heat shock gene may explain the heat sensitivity of Ap. fabae. Fluorescent in situ hybridization revealed heat-induced deformation and shrinkage of bacteriocytes in heat-sensitive species but not in heat-tolerant species. Sensitive and tolerant species also differed in numbers and transcriptional responses of heat shock genes. We describe and illustrate Sedum lipingense (Crassulaceae), a new species of stonecrop found in the limestone areas of SE Guizhou, China. Based on the presence of adaxially gibbous carpels and follicles, this taxon belongs to sect. Sedum S.H. Fu. The new species superficially resembles S. subtile Miquel and S. bulbiferum Makino but differs from these two taxa in its development of a basal leaf rosette during florescence. Three new species of the genus Oecleopsis Emeljanov, 1971 from China, O. acerbus Lv & Chen, sp. nov. and O. panxianensis Lv & Chen, sp. nov. from Guizhou Province, and O. digitatus Lv & Chen, sp. nov. from Sichuan Province, are described and illustrated. With these additions, the number of species in the genus is increased to 18. The increasing proportion of distributed photovoltaics (DPVs) and electric vehicle charging stations in low-voltage distribution networks (LVDNs) has resulted in challenges such as distribution transformer overloads and voltage violations. To address these problems, we propose a coordinated planning method for flexible interconnections and energy storage systems (ESSs) to improve the accommodation capacity of DPVs. First, the power-transfer characteristics of flexible interconnection and ESSs are analyzed. The equipment costs of the voltage source converters (VSCs) and ESSs are also analyzed comprehensively, considering the differences in installation and maintenance costs for different installation locations. Second, a bilevel programming model is established to minimize the annual comprehensive cost and yearly total PV curtailment capacity. Within this framework, the upper-level model optimizes the installation locations and capacities of the VSCs and ESSs, whereas the lower-level model optimizes the operating power of the VSCs and ESSs. The proposed model is solved using a non-dominated sorting genetic algorithm with an elite strategy (NSGA-II). The effectiveness of the proposed planning method is validated through an actual LVDN scenario, which demonstrates its advantages in enhancing PV accommodation capacity. In addition, the economic benefits of various planning schemes with different flexible interconnection topologies and different PV grid-connected forms are quantitatively analyzed, demonstrating the adaptability of the proposed coordinated planning method.
