2. Novel Lactic Acid Bacteria Strains from Regional Peppers with Health-Promoting Potential.

Abstract

This study provides a comprehensive investigation of lactic acid bacteria (LAB) isolated from Argentinean Capsicum annum L. This research covers important aspects, including genotypic characterization, bacterial stress tolerance, adhesion ability, safety evaluation, and functional and technological properties. The predominant isolates were identified as Lactilactobacillus curvatus and Lactiplantibacillus plantarum. A Rep-PCR analysis grouped the isolates into 11 clonal groups. Lp. plantarum LVP 40 and LV 46, Levilactobacillus brevis LVP 41, Pediococcus pentosaceus LV P43, and Lt. curvatus LVP44 displayed both safety and resilience against adverse conditions such as a slow pH, bile, and simulated gastric and intestinal juices. Moreover, the LAB strains exhibited high hydrophobicity and auto-aggregation percentages, NaCl tolerance, and a substantial acidifying capacity. LAB supernatants demonstrated promising surfactant and emulsifying properties. Likewise, they differentially inhibited Staphylococcus aureus and Pseudomonas aeruginosa biofilms, showcasing their potential as antipathogenic agents. Noteworthily, some strains displayed considerable co-aggregation with these pathogens, and several isolates showed an effective antimutagenic and detoxifying power, further emphasizing their multifaceted capabilities. Five pepper bacterial strains showcased beneficial properties, suggesting their potential for gut health enhancement. 10. Desechos de vinificación como inhibidores de biopelículas de Staphylococcus aureus

Abstract

Staphylococcus aureus is a well-known pathogen living as biofilm in a wide variety of environments and represents a severe risk of food contamination. The present study focused on the antibiofilm capacity of pomace extracts of different polarities and different varietals against two strains of S. aureus. Wine pomace of three varietals (Bonarda, Tannat, and Malbec) from Cafayate, Argentine, at two concentrations (10 and 100 µg/mL) was used to inhibit biofilm formation (BF) and preformed biofilm of S. aureus ATCC 6538 and LVP63 strains. Extractions were carried out with solvents of increasing polarity (chloroform, ethyl acetate, and ethanol). The ethyl acetate and ethanolic pomace extracts of the Tannat varietal were the most effective in reducing the BF formation in S. aureus without affecting planktonic growth. The extracts of Malbec and Tannat diminish the biofilm metabolic activity up to 70%. Likewise, the polar Tannat extracts and the mixture pomace disrupt the preformed BF in both S. aureus strains. These residues can be used as a natural alternative source for the inhibition of pathogenic bacteria in the food industry and reduce the impact of these agro-industrial wastes on the environment, Staphylococcus aureus es un patógeno conocido que forma biopelículas en una amplia variedad de entornos y representa un riesgo severo de contaminación de los alimentos. El presente estudio se centró en la determinación de las capacidades antibiofilm de extractos de orujo procedentes de diferentes varietales de diferentes polaridades, procedentes de Cafayate, Argentina, frente a dos cepas de S. aureus. Se utilizaron orujos de tres varietales regionales (Bonarda, Tannat y Malbec) para inhibir la formación de biofilm (BF) o tratar una biopelícula formada. Se realizaron extracciones con solventes de polaridad creciente (cloroformo, acetato de etilo y etanol) y se enfrentaron 10 y 100 µg/mL de extractos a las cepas de S. aureus ATCC 6538 y LVP63. Los extractos acetato de etilo y etanólicos de orujo del varietal Tannat fueron los más efectivos para disminuir la formación de BF en S. aureus sin afectar el crecimiento planctónico. Respecto a los efectos sobre la biopelícula preformada, los extractos de los varietales Malbec y Tannat disminuyeron la actividad metabólica hasta un 70% y los extractos polares de Tannat y la mezcla de orujos disgregaron hasta un 65% el BF preformado por ambas cepas S. aureus. Por lo tanto, la reutilización de estos desechos agroindustriales se plantea como una alternativa natural para combatir bacterias patógenas en la industria alimentaria, disminuyendo el impacto de estos residuos agroindustriales en el ambiente y convirtiendo a estos desechos en un bioproducto, económicamente, rentable.

Abstract

Introduction: Background: refeeding syndrome (RS) is a metabolic complication associated with nutritional support. The lack of management protocols for patients on nutritional support may lead to the development of RS or undernourishment. Objective: to evaluate adherence to the algorithm for total enteral nutritional support (ASNET), and the efficacy of reaching nutrient requirements in patients at risk of RS. Methods: a cohort study: screening and nutritional evaluation of patients and risk of RS were performed. Adults admitted to hospital for noncritical illness who received exclusively enteral nutrition (EN) were eligible. Patients with RS, intestinal failure or chronic diarrhea were excluded. Adherence to ASNET was evaluated along with the efficacy of nutritional support to reach the optimal protein and energy requirement (OPER) and any associated complications. Patient follow-up ended when the feeding route was changed or upon discharge. Results: a total of 73 patients were included (mean age, 62 ± 16 yrs; BMI, 18.5 ± 4.2 kg/m2), and 55 % were men. All had nutritional risk of developing RS (57, low; 15, high; 1, very high); 34 % had adequate adherence to ASNET, and 33 % managed to cover the total requirement between 4 and 6 days. OPER was reached by 38 % by the fourth day of EN, and adequate adherence to ASNET increased the probability of achieving it (RR, 2.2; 95 % CI, 1.6-3.2, p < 0.0001) without increasing the associated complications. Nonetheless, 36 % developed complications, of whom 96 % did not adhere to ASNET. Conclusion: adherence to ASNET in patients at risk of RS allowed the achievement of OPER safely by day four with fewer associated complications. [ABSTRACT FROM AUTHOR]

Abstract

Senecio punae Cabrera (Asteraceae–Senecioneae) is an endemic species from Puna semi-desert region of Argentina.The major compound, 4,5-eremophil7(11)9-dien-8-one, also known as dehydrofukinone (D), was isolated from the diethyl ether extract (E) of the plant. The phytochemical constituents from S. punae are presented here for the first time, as well as bioassays employing D, a molecule with versatility to carry out many different biological activities. E and D showed acute molluscicidal activity against the bilharzia vector snail Biomphalaria peregrina with LD50 values of 68.6 and 16.7 µg/mL, respectively. D exerted moderate and strong effects against promastigotes of Leishmania amazonensis and L. braziliensis, with IC50 values of 34.3±1.2 and 9.93±0.17 µg/mL, respectively. Antibacterial effects were also found. Diethyl ether extract (E) and dehydrofukinone (D) were slightly active against Pseudomonas aeruginosa and Staphylococcus aureus at 50 and 25 µg/mL, respectively. However, D and E improved the effectiveness of oxacillin at 6 µg/mL against S. aureus growth and its biofilm, but did not affect beneficial lactobacilli strains. Synergistic effects were also measured between D (23 µg/mL) and ciprofloxacin (0.25 µg/mL) against two P. aeruginosa strains (FIC index = 0.38), as well as against two S. aureus strains (FIC indexes = 0.38 and 0.25). Finally, when 250 µg of either dehydrofukinone (D) or diethyl ether extract (E) were incorporated into the diet of the pest Oryzaephillus surinamensis, an alteration in the feeding behavior of the insect (Repellency indexes = +60 and +10, respectively) was observed. Abstract

The essential oil of an Argentine collection of the annual herb Acanthospermum hispidum DC. (Asteraceae), which contains the sesquiterpenoids beta-caryophyllene (35.2%), alpha-bisabolol (11.4%), and germacrene D (11.1%) as major constituents, displayed molluscicidal effects on an adult population of Biomphalaria peregrina Orbigny (LD50 37.8 μg/mL) and produced alteration of the feeding and oviposition behavior of the polyphagous insect Spodoptera frugiperda Smith when incorporated in the larval diet at 250 μg/g. In addition, a selective antibacterial activity was observed against pathogenic strains of Staphylococcus aureus and Enterococcus faecalis (MICs 62.5 and 125 μg/mL), but was inactive at 125 μg/mL against three beneficial lactic acid bacteria. Synergistic interactions were further validated by FIC index determination of the combination of the antibiotics oxacillin and gentamicin and the essential oil on the four pathogenic strains evaluated.
