1. PGAHum: Prior-Guided Geometry and Appearance Learning for High-Fidelity Animatable Human Reconstruction

Abstract

Recent techniques on implicit geometry representation learning and neural rendering have shown promising results for 3D clothed human reconstruction from sparse video inputs. However, it is still challenging to reconstruct detailed surface geometry and even more difficult to synthesize photorealistic novel views with animated human poses. In this work, we introduce PGAHum, a prior-guided geometry and appearance learning framework for high-fidelity animatable human reconstruction. We thoroughly exploit 3D human priors in three key modules of PGAHum to achieve high-quality geometry reconstruction with intricate details and photorealistic view synthesis on unseen poses. First, a prior-based implicit geometry representation of 3D human, which contains a delta SDF predicted by a tri-plane network and a base SDF derived from the prior SMPL model, is proposed to model the surface details and the body shape in a disentangled manner. Second, we introduce a novel prior-guided sampling strategy that fully leverages the prior information of the human pose and body to sample the query points within or near the body surface. By avoiding unnecessary learning in the empty 3D space, the neural rendering can recover more appearance details. Last, we propose a novel iterative backward deformation strategy to progressively find the correspondence for the query point in observation space. A skinning weights prediction model is learned based on the prior provided by the SMPL model to achieve the iterative backward LBS deformation. Extensive quantitative and qualitative comparisons on various datasets are conducted and the results demonstrate the superiority of our framework. Ablation studies also verify the effectiveness of each scheme for geometry and appearance learning.

Abstract

Diabetic ischemic wound treatment remains a critical clinical challenge. Neovascularization plays a significant role in wound healing during all stages of the tissue repair process. Strategies that enhance angiogenesis and neovascularization and improve ischemic pathology may promote the healing of poor wounds, particularly diabetic wounds in highly ischemic conditions. We previously identified a cyclic peptide LXW7 that specifically binds to integrin αvβ3 on endothelial progenitor cells (EPCs) and endothelial cells (ECs), activates vascular endothelial growth factor (VEGF) receptors, and promotes EC growth and maturation. In this study, we designed and synthesized a multi-functional pro-angiogenic molecule by grafting LXW7 and collagen-binding peptides (SILY) to a dermatan sulfate (DS) glycosaminoglycan backbone, named LXW7-DS-SILY, and further employed this multi-functional molecule to functionalize collagen-based extracellular matrix (ECM) scaffolds. We confirmed that LXW7-DS-SILY modification significantly promoted EPC attachment and growth on the ECM scaffolds in vitro and supported EPC survival in vivo in the ischemic environment. When applied in an established Zucker Diabetic Fatty (ZDF) rat ischemic skin flap model, LXW7-DS-SILY-functionalized ECM scaffolds loaded with EPCs significantly improved wound healing, enhanced neovascularization and modulated collagen fibrillogenesis in the ischemic environment. Altogether, this study provides a promising novel treatment to accelerate diabetic ischemic wound healing, thereby reducing limb amputation and mortality of diabetic patients.

Abstract

PurposeVascular endothelial growth factor receptor 2 (VEGFR-2) and neuropilin-1 (NRP-1) are two prominent synergistic receptors overexpressed on new blood vessels in glioma and may be promising targets for antiglioma therapy. The aim of this study was to design a dual receptor targeting and blood-brain barrier (BBB) penetrating peptide-modified polyethyleneimine (PEI) nanocomplex that can efficiently deliver the angiogenesis-inhibiting secretory endostatin gene (pVAXI-En) to treat glioma.Materials and methodsWe first constructed the tandem peptide TAT-AT7 by conjugating AT7 to TAT and evaluated its binding affinity to VEGFR-2 and NRP-1, vasculature-targeting ability and BBB crossing capacity. Then, TAT-AT7-modified PEI polymer (PPTA) was synthesized, and a pVAXI-En-loaded PPTA nanocomplex (PPTA/pVAXI-En) was prepared. The physicochemical properties, cytotoxicity, transfection efficiency, capacities to cross the BBB and BTB (blood-tumor barrier) and glioma-targeting properties of PPTA/pVAXI-En were investigated. Moreover, the in vivo anti-angiogenic behaviors and anti-glioma effects of PPTA/pVAXI-En were evaluated in nude mice.ResultsThe binding affinity of TAT-AT7 to VEGFR-2 and NRP-1 was approximately 3 to 10 times greater than that of AT7 or TAT. The cellular uptake of TAT-AT7 in endothelial cells was 5-fold and 119-fold greater than that of TAT and AT7 alone, respectively. TAT-AT7 also displayed remarkable efficiency in penetrating the BBB and glioma tissue in vivo. PPTA/pVAXI-En exhibited lower cytotoxicity, stronger BBB and BTB traversing abilities, higher selective glioma targeting and better gene transfection efficiency than PEI/pVAXI-En. More importantly, PPTA/pVAXI-En significantly suppressed the tube formation and migration of endothelial cells, inhibited glioma growth, and reduced the microvasculature in orthotopic U87 glioma-bearing nude mice.ConclusionOur study demonstrates that PPTA/pVAXI-En can be exploited as an efficient dual-targeting nanocomplex to cross the BBB and BTB, and hence it represents a feasible and promising nonviral gene delivery system for effective glioma therapy.

Abstract

RAD51 recombinase plays a critical role for cancer cell proliferation and survival. Targeting RAD51 is therefore an attractive strategy for treating difficult-to-treat cancers, e.g. triple negative breast cancers which are often resistant to existing therapeutics. To this end, we have designed, synthesized and evaluated a panel of new RAD51 inhibitors, denoted IBR compounds. Among these compounds, we have identified a novel small molecule RAD51 inhibitor, IBR120, which exhibited a 4.8-fold improved growth inhibition activity in triple negative human breast cancer cell line MBA-MD-468. IBR120 also inhibited the proliferation of a broad spectrum of other cancer cell types. Approximately 10-fold difference between the IC50 values in normal and cancer cells were observed. Moreover, IBR120 was capable of disrupting RAD51 multimerization, impairing homologous recombination repair, and inducing apoptotic cell death. Therefore, these novel RAD51 inhibitors may serve as potential candidates for the development of pharmaceutical strategies against difficult-to-treat cancers.

Abstract

Introducing superconductivity into two-dimensional (2D) films with nontrivial topology has been intensively pursued as one of the feasible scenarios to realize 1D topological superconductor. Prevailing endeavors mostly exploit the external gating or proximity effect of a traditional superconductor, by which the critical temperatures ( T c ) are limited to several Kelvin range. Here, we report on the discovery of interface-enhanced superconductivity in monolayer 1T′-MoTe2 film. A thermally driven phase transition from Mo6Te6 nanowires to 1T′-MoTe2 films, grown on SrTiO3(001) surface by the molecular beam epitaxial methods, is demonstrated. A combined study of scanning tunneling microscopy/spectroscopy, electrical transport and magnetization measurements indicates the T c of MoTe2 film is around 30 K, two orders of magnitude larger than its 3D counterpart crystal. This study shows that interfacial engineering is an efficient way to tune monolayer 1T′-MoTe2 film into superconducting states, and thus may pave the way toward higher- T c 1D intrinsic topological superconductivity. Nymphanthus calcicola S.R. Yi & Gang Yao, sp. nov. (Figs. 1‒2) Diagnosis: Nymphanthus calcicola is most similar to N. bodinieri (H. Lév.) R.W. Bouman in morphology, but differs from the latter by having cylindrical and twisting branchlets (vs. angled and straight branchlets), triangular-subulate stipules (vs. triangular stipules), larger flowers (female flowers that are 6.5–8.0 mm in diameter and male flowers 5.5–6.5 mm in diameter) (vs. female flowers that are ca. 4 mm in diameter and male flowers 4–4.5 mm in diameter), longer pedicles of male flowers (10–20 mm long vs. 3–7 mm long), depressed globose ovary (vs. globose ovary), deeply bifid stigmas (vs. shallowly bifid stigmas), and obscurely grooved fruits (vs. 6-groovded fruits). Description: Shrubs, 30–60 cm tall, dioecious, glabrous; stems cylindrical, brownish; branchlets cylindrical, twisting, reddish-brown, 10–30 cm long, swollen at junction with stem. Leaves distichous, 4–8 pairs along each branchlet; leaf blades elliptic to lanceolate, 2.7–5.0 × 1.3–2.0 cm, thick papery to subleathery, margin slightly revolute, apex acuminate to acute, base broadly cuneate to slightly round, adaxially green, abaxially pale; midrib adaxially inconspicuous, abaxially slightly raised; lateral veins 5–7 pairs, adaxially obscurely, adaxially evidently; petioles 0.5–2.2 mm long; Stipules triangular-subulate, 2.5–3.5 mm long, 1.2–1.5 mm wide basely, reddish brown. Inflorescence an axillary fascicle. Male flowers up to 15-clustered, 5.5–6.5 mm in diameter, pedicels slender, glabrous, yellow-green to pale pink, 1.0–2.0 cm long; sepals 4, triangular, 2.5–3.0 × 2.5–3.0 mm, apex acute, margin entire or microwave; disc glands 4, reniform, 1.5–1.8 × 0.8–1.0 mm, margin entire; stamens 4, filaments connate at base, anthers 4, 1-locular, dehisced horizontally. Female flowers usually solitary or rarely 2–5-clustered, 6.5–8.0 mm in diameter, pedicels 1–2 cm long, pink, smooth and glabrous; sepals 6, in 2 series, broadly ovate-triangular, 2.3–2.6 × 2.5–3.0 mm, apex acute, margin entire or microwave; disk glands 6, reniform, 1.2–1.4 × 0.7–0.9 mm, margin entire; ovary depressed globose, glabrous, 1.2–1.5 mm in diameter, 0.8–1.0 high, 3-loculus, ovules 2 per loculus; style 3, free; stigmas deeply bifid nearly to the base, lobes 0.5–0.6 mm, glabrous. Capsule subglobose, 4.0– 4.5 mm in diameter, yellowish-brown, calyx persistent; seeds 6, 2.2–2.5 × 1.2–1.6 mm, reniform-triangular, laterally compressed. Phenology: —Flowering from February to March and fruiting from May to June. Etymology: — Nymphanthus calcicola is named after its habitat, which is characterized by the calcareous substrates in limestone hills. Chinese name: —Hui Yan Ye Xia Zhu (灰岩叶下t) Habitat and distribution: — Nymphanthus calcicola is found only in Ludugou, Guankou village, Hanjia town, Pengshui county, Chongqing, China (Fig. 3). It grows under the damp forest of limestone hills, at the elevation of 275‒350 m. It is considered as Critically Endangered (CR) according to current information and the IUCN Red List criteria (IUCN Standards and Petitions Committee, 2019) for it is only found from the type locality of less than 2 km 2 according to the current investigation results. Taxonomic discussion: —Morphologically, the new species has 4 sepals in staminate flowers (Fig. 1G & H), 4 stamens (Fig. 1G & M), connate filaments (Fig. 1M) and horizontally dehisced anthers (Fig. 1M), indicating that it is a member of Nymphanthus. The new species Nymphanthus calcicola resembles N. bodinieri (Fig. 2), a species distributed in Chongqing and Guizhou province, China (Fig. 3). However, Nymphanthus calcicola is a dioecious shrub and has cylindrical and twisting branchlets (Fig. 1B & F), leaf blades that are acuminate or acute at apex and broadly cuneate to slightly round at base (Fig. 1A–F), inconspicuous midrib not raised adaxially (Fig. 1A, B, D & F), triangular-subulate stipules, female flowers that are 6.5–8.0 mm in diameter (Fig. 1I–J) and male flowers 5.5–6.5 mm in diameter (Fig. 1G–H), longer pedicels of male flowers that are 10–20 mm in length (Fig. 1B–C), oblate-spherical ovary (Fig. 1N), stigmas that are deeply bifid near to the base (Fig. 1I & L), and obscurely grooved fruits (Fig. 1K).
