21. Directional and Eye-Tracking Light Field Display with Efficient Rendering and Illumination.

Abstract

Current efforts with light field displays are mainly concentrated on the widest possible viewing angle, while a single viewer only needs to view the display in a specific viewing direction. To make the light field display a practical practice, a super multi-view light field display is proposed to compress the information in the viewing zone of a single user by reducing the redundant viewpoints. A quasi-directional backlight is proposed, and a lenticular lens array is applied to achieve the restricted viewing zone. The eye-tracking technique is applied to extend the viewing area. Experimental results show that the proposed scheme can present a vivid 3D scene with smooth motion parallax. Only 16.7% conventional light field display data are required to achieve 3D display. Furthermore, an illumination power of 3.5 watt is sufficient to lighten a 31.5-inch light field display, which takes up 1.5% of the illumination power required for planar display of similar configuration. Abstract

SUMMARY: The formation of adventitious roots (ARs) derived from hypocotyl is the most important morphological adaptation to waterlogging stress in Cucumis sativus (cucumber). Our previous study showed that cucumbers with the gene CsARN6.1, encoding an AAA ATPase domain‐containing protein, were more tolerant to waterlogging through increased AR formation. However, the apparent function of CsARN6.1 remained unknown. Here, we showed that the CsARN6.1 signal was predominantly observed throughout the cambium of hypocotyls, where de novo AR primordia are formed upon waterlogging treatment. The silencing of CsARN6.1 expression by virus‐induced gene silencing and CRISPR/Cas9 technologies adversely affects the formation of ARs under conditions of waterlogging. Waterlogging treatment significantly induced ethylene production, thus upregulating CsEIL3 expression, which encodes a putative transcription factor involved in ethylene signaling. Furthermore, yeast one‐hybrid, electrophoretic mobility assay and transient expression analyses showed that CsEIL3 binds directly to the CsARN6.1 promoter to initiate its expression. CsARN6.1 was found to interact with CsPrx5, a waterlogging‐responsive class‐III peroxidase that enhanced H2O2 production and increased AR formation. These data provide insights into understanding the molecular mechanisms of AAA ATPase domain‐containing protein and uncover a molecular mechanism that links ethylene signaling with the formation of ARs triggered by waterlogging. Significance Statement: The AAA ATPase domain‐containing protein CsARN6.1 is required for waterlogging stress tolerance, where it regulates adventitious root (AR) formation. However, the functional relevance of this gene remained unknown. Abstract

Cucumber glossiness is an important visual quality trait that affects consumer choice. Accumulating evidence suggests that glossy trait is associated with cuticular wax accumulation. However, the molecular genetic mechanism controlling cucumber glossiness remains largely unknown. Here, we report the map-based cloning and functional characterization of CsGLF1, a locus that determines the glossy trait in cucumber. CsGLF1 encodes a homolog of the Cys2His2-like fold group (C2H2) -type zinc finger protein 6 (ZFP6) and its deletion leads to glossier pericarp and decreased cuticular wax accumulation. Consistently, transcriptomic analysis demonstrated that a group of wax biosynthetic genes were downregulated when CsZFP6 was absent. Further, transient expression assay revealed that CsZFP6 acted as a transcription activator of cuticular wax biosynthetic genes. Taken together, our findings demonstrated a novel regulator of fruit glossiness, which will provide new insights into regulatory mechanism of fruit glossiness in cucumber. [ABSTRACT FROM AUTHOR]

Abstract

Cucumber (Cucumis sativus L.) is an agriculturally and economically important vegetable crop worldwide. The thickness of mesocarp is an important quality pertaining to edibility. The development of mesocarp, however, has received little attention. In this study, we selected one thick mesocarp line D8 to analyze the gene expression profiles of mesocarp at 0, 3, 6, and 9 days after pollination (DAP) by RNA sequencing. Genes mainly related to ribosomes, carbon metabolism, biosynthesis of amino acids, and oxidative phosphorylation were up-regulated at 3 DAP. From 3 to 6 DAP, the genes related to oxidative phosphorylation, photosynthesis, and phagosome were highly induced. From 6 to 9 DAP, the genes related to plant hormone signal transduction were highly induced. Differentially expressed genes linked to cyclin, hormone biosynthesis and signaling, carbon metabolism, and transcription factors MYB and WRKY were common up-regulated at 3, 6, 9 DAP in comparison with 0 DAP. The auxin-responsive and cytokinin-responsive genes were significantly up-regulated in mesocarp development. The higher expression levels of the auxin-responsive gene indole-3-acetic acid-amido synthetase 3.6 and the cytokinin response regulator 3/4b in the thick mesocarp cultivar D8 than in the thin mesocarp cultivar XUE1 might have explained the different growth rates of the mesocarp.

50. RNA-seq-based comparative transcriptome analysis reveals the role of CsPrx73in waterlogging-triggered adventitious root formation in cucumber

Abstract

Abiotic stressors like waterlogging are detrimental to cucumber development and growth. However, comprehension of the highly complex molecular mechanism underlying waterlogging can provide an opportunity to enhance cucumber tolerance under waterlogging stress. We examined the hypocotyl and stage-specific transcriptomes of the waterlogging-tolerant YZ026A and the waterlogging-sensitive YZ106A, which had different adventitious rooting ability under waterlogging. YZ026A performed better under waterlogging stress by altering its antioxidative machinery and demonstrated a greater superoxide ion (O 2−) scavenging ability. KEGG pathway enrichment analysis showed that a high number of differentially expressed genes (DEGs) were enriched in phenylpropanoid biosynthesis. By pairwise comparison and weighted gene co-expression network analysis analysis, 2616 DEGs were obtained which were categorized into 11 gene co-expression modules. Amongst the 11 modules, black was identified as the common module and yielded a novel key regulatory gene, CsPrx73. Transgenic cucumber plants overexpressing CsPrx73enhance adventitious root (AR) formation under waterlogging conditions and increase reactive oxygen species (ROS) scavenging. Silencing of CsPrx73expression by virus-induced gene silencing adversely affects AR formation under the waterlogging condition. Our results also indicated that CsERF7-3, a waterlogging-responsive ERF transcription factor, can directly bind to the ATCTA-box motif in the CsPrx73promoter to initiate its expression. Overexpression of CsERF7-3enhanced CsPrx73expression and AR formation. On the contrary, CsERF7-3-silenced plants decreased CsPrx73expression and rooting ability. In conclusion , our study demonstrates a novel CsERF7-3–CsPrx73module that allows cucumbers to adapt more efficiently to waterlogging stress by promoting AR production and ROS scavenging.
