Background: Alzheimer’s disease (AD) is a multifactorial disorder characterized by cognitive decline. Current available therapeutics for AD have limited clinical benefit. Therefore, preventive therapies for interrupting the development of AD are critically needed. Molecules targeting multifunction to interact with various pathlogical components have been considered to improve the therapeutic efficiency of AD. In particular, herbal medicines with multiplicity of actions produce cognitive benefits on AD. Bugu-M is a multi-herbal extract composed of Ganoderma lucidum (Antler form), Nelumbo nucifera Gaertn., Ziziphus jujuba Mill., and Dimocarpus longan, with the ability of its various components to confer resilience to cognitive deficits. Objective: To evaluate the potential of Bugu-M on amyloid-β (Aβ) toxicity and its in vitro mechanisms and on in vivo cognitive function. Methods: We illustrated the effect of Bugu-M on Aβ25–35-evoked toxicity as well as its possible mechanisms to diminish the pathogenesis of AD in rat cortical neurons. For cognitive function studies, 2-month-old female 3×Tg-AD mice were administered 400 mg/kg Bugu-M for 30 days. Behavioral tests were performed to assess the efficacy of Bugu-M on cognitive impairment. Results: In primary cortical neuronal cultures, Bugu-M mitigated Aβ-evoked toxicity by reducing cytoskeletal aberrations and axonal disruption, restoring presynaptic and postsynaptic protein expression, suppressing mitochondrial damage and apoptotic signaling, and reserving neurogenic and neurotrophic factors. Importantly, 30-day administration of Bugu-M effectively prevented development of cognitive impairment in 3-month-old female 3×Tg-AD mice. Conclusion: Bugu-M might be beneficial in delaying the progression of AD, and thus warrants consideration for its preventive potential for AD.

microRNAs (miRNAs) are ~21–22 nucleotide (nt) RNAs that mediate broad post-transcriptional regulatory networks. However, genetic analyses have shown that the phenotypic consequences of deleting individual miRNAs are generally far less overt compared to their misexpression. This suggests that miRNA deregulation may have broader phenotypic impacts during disease situations. We explored this concept in the Drosophila eye, by screening for miRNAs whose misexpression could modify the activity of pro-apoptotic factors. Via unbiased and comprehensively in vivo phenotypic assays, we identify an unexpectedly large set of miRNA hits that can suppress the action of pro-apoptotic genes hid and grim. We utilize secondary assays to validate that a subset of these miRNAs can inhibit irradiation-induced cell death. Since cancer cells might seek to evade apoptosis pathways, we modeled this situation by asking whether activation of anti-apoptotic miRNAs could serve as “second hits”. Indeed, while clones of the lethal giant larvae (lgl) tumor suppressor are normally eliminated during larval development, we find that diverse anti-apoptotic miRNAs mediate the survival of lgl mutant clones in third instar larvae. Notably, while certain anti-apoptotic miRNAs can target apoptotic factors, most of our screen hits lack obvious targets in the core apoptosis machinery. These data highlight how a genetic approach can reveal distinct and powerful activities of miRNAs in vivo, including unexpected functional synergies during disease or cancer-relevant settings.

The increasing prevalence of overweight and obese children and adolescents has been recognized as a public health threat worldwide. This study aimed to assess the effect of a stepwise lifestyle intervention in children and adolescents.We developed a multidisciplinary clinic aimed at providing lifestyle interventions for obese children and adolescents. The program comprised three stages with stepwise goals: knowledge building (the first 4 weeks), habit consolidation (5-12 weeks), and self-monitoring (13-20 weeks).Of the 63 participants (age 11.6 ± 3.2 years) who entered the first stage of the program, 48, 22, and 15 completed the first, second and third stages (4, 12, and 20 weeks), respectively. In the first stage, significant improvement was noted in body weight, body mass index (BMI), BMI z-score, and waist circumference. Improvements in physical fitness performance were observed at 4 weeks in 3/5 items and at 12 weeks in 4/5 items. The decreases in body weight, BMI and BMI z-score were most prominent in the first two stages. In the third stage, participants maintained a stable body weight. In the 15 subjects who completed the whole program, BMI decreased from 29.3 ± 6.9 to 27.8 ± 6.1 (P = 0.001), and BMI z-score decreased from 3.06 ± 0.96 to 2.69 ± 0.91(P = 0.001).We developed a feasible multidisciplinary program based on knowledge education and individualized training. BMI and physical fitness scores can be used as early indicators of lifestyle change for obese children and adolescents.

Flax seed has become an increasingly popular food ingredient because of its nutrient richness as well as potential health benefits. Flax seeds are often ground before consumption, and flax seed cakes are used as animal feed. Aflatoxin production may occur subsequently when the ground seeds are stored in an environment that supports fungal growth. The objectives of this study were to determine the growth of two toxigenic fungi, Aspergillus flavus and A. parasiticus, and to quantify the concentrations of four major aflatoxins (AFB1, AFG1, AGB2, and AFG2) produced by the two fungi on ground flax seeds with water activities (aws) of 0.82, 0.86, 0.90, 0.94, and 0.98, incubated for 30 days at 20, 27, and 35°C. Results of the study showed that A. flavus was able to grow on ground seeds with aw from 0.86 to 0.98 at all three temperatures, and the most rapid growth occurred at aws 0.90 and 0.94 at 27°C. In comparison, A. parasiticus grew on seeds with aw from 0.86 to 0.98 at 27 and 35°C as well as on seeds with aw from 0.86 to 0.90 at 20°C, and the most favorable growth condition was aw from 0.90 to 0.94 at 35°C. A. flavus produced aflatoxins on seeds with aw from 0.90 to 0.94 at 27°C as well as on seeds with aw from 0.86 to 0.98 at 35°C, and the maximum total aflatoxin (298 μg/kg), AFB1 (247 μg/kg), and AFG1 (51 μg/kg) were found on seeds with aw 0.90 at 35°C. In comparison, A. parasiticus produced aflatoxins under a wider range of conditions, which included aw 0.86 at 27 and 35°C, aw 0.90 at 20 and 27°C, aw 0.94 at 27°C, and aw 0.98 at 35°C. The maximum total aflatoxin (364 μg/kg) and maximum AFB1 (324 μg/kg) along with 34 μg/kg AFG1 and 6 μg/kg AFB2 were produced by A. parasiticus on seeds with aw 0.98 incubated at 35°C for 30 days. Linear regression models also indicated that high incubation temperature (35°C) was optimal for overall fungal growth and for formation of high levels of aflatoxin by both fungi. Future studies should also address aflatoxin contamination in flax seed oil. HIGHLIGHTS

This research examines senior high school students’ earth science learning effects, focusing on the influence of science reading beliefs when employing mobile devices. The revision of the Science Reader Belief Inventory (SRBI) was used to examine the connections for high school students’ personal scientific reading beliefs and reading comprehension of earth science learning effectiveness conditions when using mobile devices to learn. The learning outcome was determined using achievement tests. In this research, 97 students from three classes of first-year high school students were enrolled in an eight-week experimental teaching study followed by an achievement test. The major findings are as follows: (1) High school students’ transaction beliefs were stronger than transmission beliefs. Transaction beliefs were significantly correlated with transmission beliefs. (2) Scientific beliefs may take a long time to change. (3) Whereas traditional reading comprehension strategies seem to have relied more heavily on vocabulary development, in an e-learning environment, students tend to rely on sentence-level parsing to understand scientific texts. This research provides a reference for teachers within learning environments in which information is incorporated into technology instruction, and various learning scenarios are used.

The nervous and endocrine systems coordinate with each other to closely influence physiological and behavioural responses in animals. Here we show that WAKE (encoded by wide awake, also known as wake) modulates membrane levels of GABAA receptor Resistance to Dieldrin (Rdl), in insulin-producing cells of adult male Drosophila melanogaster. This results in changes to secretion of insulin-like peptides which is associated with changes in juvenile hormone biosynthesis in the corpus allatum, which in turn leads to a decrease in 20-hydroxyecdysone levels. A reduction in ecdysone signalling changes neural architecture and lowers the perception of the male-specific sex pheromone 11-cis-vaccenyl acetate by odorant receptor 67d olfactory neurons. These finding explain why WAKE-deficient in Drosophila elicits significant male-male courtship behaviour.

This protocol describes the allotransplantation of tumors in Drosophila melanogaster using an auto-nanoliter injection apparatus. With the use of an autoinjector apparatus, trained operators can achieve more efficient and consistent transplantation results compared to those obtained using a manual injector. Here, we cover topics in a chronological fashion: from the crossing of Drosophila lines, to the induction and dissection of the primary tumor, transplantation of the primary tumor into a new adult host and continued generational transplantation of the tumor for extended studies. As a demonstration, here we use Notch intracellular domain (NICD) overexpression induced salivary gland imaginal ring tumors for generational transplantation. These tumors can first be reliably induced in a transition-zone microenvironment within larval salivary gland imaginal rings, then allografted and cultured in vivo to study continued tumor growth, evolution, and metastasis. This allotransplantation method can be useful in potential drug screening programs, as well as for studying tumor-host interactions.

Cisplatin chemotherapy causes myelosuppression and often limits treatment duration and dose escalation in patients. Novel approaches to circumvent or lessen myelotoxicity may improve clinical outcome and quality of life in these patients. Chlorella sorokiniana (CS) is a freshwater unicellular green alga and exhibits encouraging efficacy in immunomodulation and anticancer in preclinical studies. However, the efficacy of CS on chemoprotection remains unclear. We report here, for the first time, that CS extract (CSE) could protect normal myeloid cells and PBMCs from cisplatin toxicity. Also, cisplatin-induced apoptosis in HL-60 cells was rescued through reservation of mitochondrial function, inhibition of cytochrome c release to cytosol, and suppression of caspase and PARP activation. Intriguingly, cotreatment of CSE attenuated cisplatin-evoked hypocellularity of bone marrow in mice. Furthermore, we observed the enhancement of CSF-GM activity in bone marrow and spleen in mice administered CSE and cisplatin, along with increased CD11b levels in spleen. In conclusion, we uncovered a novel mechanism of CSE on myeloprotection, whereby potentially supports the use of CSE as a chemoprotector against cisplatin-induced bone marrow toxicity. Further clinical investigation of CSE in combination with cisplatin is warranted.

Ploidy variation is a cancer hallmark and is frequently associated with poor prognosis in high-grade cancers. Using a Drosophila solid-tumor model where oncogenic Notch drives tumorigenesis in a transition-zone microenvironment in the salivary gland imaginal ring, we find that the tumor-initiating cells normally undergo endoreplication to become polyploid. Upregulation of Notch signaling, however, induces these polyploid transition-zone cells to re-enter mitosis and undergo tumorigenesis. Growth and progression of the transition-zone tumor are fueled by a combination of polyploid mitosis, endoreplication, and depolyploidization. Both polyploid mitosis and depolyploidization are error prone, resulting in chromosomal copy-number variation and polyaneuploidy. Comparative RNA-seq and epistasis analysis reveal that the DNA-damage response genes, also active during meiosis, are upregulated in these tumors and are required for the ploidy-reduction division. Together, these findings suggest that polyploidy and associated cell-cycle variants are critical for increased tumor-cell heterogeneity and genome instability during cancer progression.

Fundamental, bending (ν6, ν7, ν8, ν9), and CC-stretch (ν2, ν3) hot band spectra in the antisymmetric CH stretch (ν4) region near 3330 cm(-1) have been observed and analyzed for jet cooled diacetylene (HC≡C-C≡CH) under sub-Doppler conditions. Diacetylene is generated in situ in the throat of a pulsed supersonic slit expansion by discharge dissociation of acetylene to form ethynyl (C≡CH) + H, followed by radical attack (HC≡CH + C≡C-H) to form HC≡C-C≡CH + H. The combination of (i) sub-Doppler line widths and (ii) absence of spectral congestion permits rotational structure and Coriolis interactions in the ν4 CH stretch fundamental to be observed and analyzed with improved precision. Of particular dynamical interest, the spectra reveal diacteylene formation in highly excited internal vibrational states. Specifically, multiple Π ← Π and Δ ← Δ hot bands built on the ν4 CH stretch fundamental are observed, due to doubly degenerate bending vibrations [cis C≡C-H bend (ν6), trans C-C≡C bend (ν7), trans C≡C-H bend (ν8) and cis C-C≡C bend (ν9)], as well as a heretofore unobserved Σ ← Σ band assigned to excitation of ν2 or 2ν3 CC stretch. Boltzmann analysis yields populations consistent with universally cold rotations (Trot ≈ 15 ± 5 K) and yet superthermal vibrations (Tvib ≈ 85-430 K), the latter of which is quite anomalous for the high collision densities in a slit jet expansion. In order to elucidate the physical mechanism for this excess vibrational excitation, high level ab initio CCSD(T) calculations have been pursued with explicitly correlated basis sets (VnZ-f12; n = 2,3) and extrapolated to the complete basis set (CBS) limit using MOLPRO quantum chemistry software. The results suggest that the extensive hot band structure observed arises from (i) highly exothermic CCH + HCCH addition to yield a strongly bent HCCHCCH radical intermediate (ΔH = -62.6 kcal/mol), followed by (ii) rapid fragmentation over a submerged transition state barrier (ΔH = -18.9 kcal/mol) to form vibrationally hot diacetylene + H products (ΔH = -25.6 kcal/mol), and consistent with crossed molecular beam studies by Kaiser et al. [Phys. Chem. Chem. Phys. 2002, 4, 2950.] Finally, RRKM fragmentation rates for this complex are calculated, which exceed collision frequencies in the slit jet expansion and suggest near unity quantum efficiency for diacetylene formation.

The sub-Doppler CH-symmetric stretch (ν3) infrared absorption spectrum of a hydroxymethyl (CH2OH) radical is observed and analyzed with the radical formed in a slit-jet supersonic discharge expansion (Trot = 18 K) via Cl atom mediated H atom abstraction from methanol. The high sensitivity of the spectrometer and reduced spectral congestion associated with the cooled expansion enable first infrared spectroscopic observation of hydroxymethyl transitions from both ± symmetry tunneling states resulting from large amplitude COH torsional motion. Nuclear spin statistics due to exchange of the two methyl H-atoms aid in unambiguous rovibrational assignment of two A-type Ka = 0 ← 0 and Ka = 1 ← 1 bands out of each ± tunneling state, with additional spectral information obtained from spin-rotation splittings in P, Q, and R branch Ka = 1 ← 1 transitions that become resolved at low N. A high level ab initio potential surface (CCSD(T)-f12b/cc-pvnzf12 (n = 2,3)/CBS) is calculated in the large amplitude COH torsional and CH2 wag coordinates, which in the adiabatic approximation and with zero point correction predicts ground state tunneling splittings in good qualitative agreement with experiment. Of particular astrochemical interest, a combined fit of the present infrared ground state combination differences with recently reported millimeter-wave frequencies permits the determination of improved accuracy rotational constants for the ground vibrational state, which will facilitate ongoing millimeter/microwave searches for a hydroxymethyl radical in the interstellar medium.

Neuroinflammation plays a central role in the pathophysiology of Alzheimer’s disease (AD). Compounds that suppress neuroinflammation have been identified as potential therapeutic targets for AD. Rhinacanthin C (RC), a naphthoquinone ester found in Rhinacanthus nasutus Kurz (Acanthaceae), is currently proposed as an effective molecule against inflammation. However, the exact role of RC on neuroinflammation remains to be elucidated. In the present study, we investigated RC effect on modulating lipopolysaccharides (LPS), amyloid-β peptide (Aβ), or interferon-γ- (IFN-γ-) evoked pathological events in neurons and glia. Our findings demonstrated that RC prevented Aβ-induced toxicity in rat hippocampal neurons and attenuated LPS-activated nitric oxide (NO) production, inducible nitric oxide synthase (iNOS) expression, and NF-κB signaling in rat glia. Likewise, RC suppressed LPS-induced neuroinflammation by reducing NO production and iNOS, IL-1β, CCL-2, and CCL-5 mRNA levels in rat microglia. Further studies using BV-2 microglia revealed that RC inhibited LPS-, Aβ-, and IFN-γ-stimulated IL-6 and TNF-α secretion. Of note, NF-κB and ERK activation was abrogated by RC in BV-2 cell response to Aβ or IFN-γ. Moreover, RC protected neurons from Aβ-stimulated microglial conditioned media-dependent toxicity. Collectively, these data highlight the beneficial effects of RC on neuroprotection and support the therapeutic implications of RC to neuroinflammation-mediated conditions.

A suppressed-Doppler (Δν = 180 MHz) infrared spectrum of monodeuterated ammonium ions (NH3D+) has been obtained for the ν1 (symmetric) and ν4 (degenerate) N–H stretch bands via direct absorption high resolution IR laser spectroscopy in a planar slit jet discharge expansion. The ion is efficiently generated by H3+ protonation of NH2D in a discharge mixture of H2/NH2D, with the resulting expansion rapidly cooling the molecular ions into low rotational states. The first high-resolution infrared spectrum of ν1 is reported, as well as many previously unobserved transitions in the ν4 rovibrational manifold. Simultaneous observation of both ν1 and ν4 permits elucidation of both the vibrational ground and excited state properties of the ion, including rigorous benchmarking of band origins against high-level anharmonic ab initio theory as well as determination of the ν1:ν4 intensity ratio for comparison with bond-dipole model predictions. Ground-state combination differences from this work and earlier studies permit the rotational constants of NH3D+ to be determined to unprecedented accuracy, the results of which support previous laboratory and astronomical assignment of the 10–00 pure rotational transition and should aid future searches for other rotational transitions as well.

Phenyl radical has been studied via sub-Doppler infrared spectroscopy in a slit supersonic discharge expansion source, with assignments for the highest frequency b2 out-of-phase C-H symmetric stretch vibration (ν19) unambiguously confirmed by ≤6 MHz (0.0002 cm(-1)) agreement with microwave ground state combination differences of McMahon et al. [Astrophys. J. 2003, 590, L61-64]. Least squares analysis of over 100 resolved rovibrational peaks in the sub-Doppler spectrum to a Watson Hamiltonian yields precision excited-state rotational constants and a vibrational band origin (ν0 = 3071.8915(4) cm(-1)) consistent with a surprisingly small red-shift (0.9 cm(-1)) with respect to Ar matrix isolation studies of Ellison and co-workers [J. Am. Chem. Soc. 2001, 123, 1977]. Nuclear spin weights and inertial defects confirm the vibrationally averaged planarity and (2)A1 rovibronic symmetry of phenyl radical, with analysis of the rotational constants consistent with a modest C2v distortion of the carbon backbone frame due to partial sp rehybridization of the σ C radical-center. Most importantly, despite the number of atoms (N = 11) and vibrational modes (3N - 6 = 27), phenyl radical exhibits a remarkably clean jet cooled high-resolution IR spectrum that shows no evidence of intramolecular vibrational relaxation (IVR) phenomena such as local or nonlocal perturbations due to strongly coupled nearby dark states. This provides strong support for the feasibility of high-resolution infrared spectroscopy in other aromatic hydrocarbon radical systems.

The transcription factor FliA, also called sigma 28, is a major regulator of bacterial flagellar biosynthesis genes. Growing evidence suggest that in addition to motility, FliA is involved in controlling numerous bacterial behaviors, even though the underlying regulatory mechanism remains unclear. By using a transcriptional fusion to gfp that responds to cyclic (c)-di-GMP, this study revealed a higher c-di-GMP concentration in the fliA deletion mutant of Pseudomonas aeruginosa than in its wild-type strain PAO1. A comparative analysis of transcriptome profiles of P. aeruginosa PAO1 and its fliA deletion mutant revealed an altered expression of several c-di-GMP-modulating enzyme-encoding genes in the fliA deletion mutant. Moreover, the downregulation of PA4367 (bifA), a Glu-Ala-Leu motif-containing phosphodiesterase, in the fliA deletion mutant was confirmed using the β-glucuronidase reporter gene assay. FliA also altered pyocyanin and pyorubin production by modulating the c-di-GMP concentration. Complementing the fliA mutant strain with bifA restored the motility defect and pigment overproduction of the fliA mutant. Our results indicate that in addition to regulating flagellar gene transcription, FliA can modulate the c-di-GMP concentration to regulate the swarming motility and phenazine pigment production in P. aeruginosa.

The sensitivity of vibronic calculations to electronic structure methods and basis sets is explored and compared to accurate relative intensities of the vibrational bands of phenylacetylene in the S(1)(A(1)B(2)) ← S(0)(X(1)A(1)) transition. To provide a better measure of vibrational band intensities, the spectrum was recorded by cavity ringdown absorption spectroscopy up to energies of 2000 cm(-1) above the band origin in a slit jet sample. The sample rotational temperature was estimated to be about 30 K, but the vibrational temperature was higher, permitting the assignment of many vibrational hot bands. The vibronic structure of the electronic transition was simulated using a combination of time-dependent density functional theory (TD-DFT) electronic structure codes, Franck-Condon integral calculations, and a second-order vibronic model developed previously [Johnson, P. M.; Xu, H. F.; Sears, T. J. J. Chem. Phys. 2006, 125, 164331]. The density functional theory (DFT) functionals B3LYP, CAM-B3LYP, and LC-BLYP were explored. The long-range-corrected functionals, CAM-B3LYP and LC-BLYP, produced better values for the equilibrium geometry transition moment, but overemphasized the vibronic coupling for some normal modes, while B3LYP provided better-balanced vibronic coupling but a poor equilibrium transition moment. Enlarging the basis set made very little difference. The cavity ringdown measurements show that earlier intensities derived from resonance-enhanced multiphoton ionization (REMPI) spectra have relative intensity errors.

Bands in the CH 2 b ˜ 1 B 1 – a ˜ 1 A 1 transition between 12 500 and 13 000 cm−1 were recorded at Doppler-limited resolution using a transient frequency-modulation (FM) laser absorption spectrometer. Rotational levels in seven upper vibronic states: a ˜ ( 0 , 11 , 0 ) 1 , 3 , a ˜ ( 2 , 6 , 0 ) 1 , b ˜ ( 0 , 2 , 0 ) 4 , b ˜ ( 1 , 1 , 0 ) 1 , 2 , and b ˜ ( 0 , 3 , 0 ) 1 were assigned with the assistance of optical–optical double resonance (OODR) and ground state combination differences. Perturbations due to rotation-vibration coupling, anharmonic couplings, l-resonance, as well as the Renner–Teller effect are discussed as sources of the irregular rotational patterns observed in the b ˜ ( 0 , 3 , 0 ) 1 , b ˜ ( 1 , 1 , 0 ) 1 , and a ˜ ( 0 , 11 , 0 ) 3 vibronic levels.

The origin band in the b̃(1)B(1)-ã(1)A(1) transition of CH(2) near 1.2 μm has been recorded at Doppler-limited resolution using diode laser transient absorption spectroscopy. The assignments of rotational transitions terminating in upper state levels with K(a) = 0 and 1, were confirmed by ground state combination differences and extensive optical-optical double resonance experiments. The assigned lines are embedded in a surprisingly dense spectral region, which includes a strong hot band, b̃(0,1,0) K(a) = 0 - ã(0,1,0) K(a) = 1 sub-band lines, with combination or overtone transitions in the ã(1)A(1) state likely responsible for the majority of unassigned transitions in this region. From measured line intensities and an estimate of the concentration of CH(2) in the sample, we find the transition moment square for the 0(00) ← 1(10) transition in the b̃(1)B(1)(0,0,0)(0)-ã(1)A(1)(0,0,0)(1) sub-band is 0.005(1) D(2). Prominent b̃(1)B(1)(0,1,0)(0)-ã(1)A(1)(0,1,0)(1) hot band lines were observed in the same spectral region. Comparison of the intensities of corresponding rotational transitions in the two bands suggests the hot band has an intrinsic strength approximately 28 times larger than the origin band. Perturbations of the excited state K(a) = 0 and 1 levels are observed and discussed. The new measurements will lead to improved future theoretical modeling and calculations of the Renner-Teller effect between the ã and b̃ states in CH(2).

The vibrational structure of the S(1)-S(0) electronic band of phenylacetylene has been recorded by 1 + 1 resonance-enhanced multiphoton ionization, accompanied by slow electron velocity map imaging photoelectron spectroscopy at each resonant vibrational band. Assignments of the S(1) vibrations (up to 2000 cm(-1) above the band origin) are based upon the relative intensities of the vibronic bands calculated by complete second-order vibronic coupling, vibration-rotation (Coriolis and Birss) coupling calculations, and the vibrational structure of the S(1) resonant photoelectron spectra. Although this is an allowed electronic transition, the relative intensities of the a(1) bands are often largely determined by vibronic coupling rather than simple Franck-Condon factors, and second-order coupling is substantial. Nonsymmetric vibrations have intensities obtained through either vibronic or Coriolis coupling, and the calculations have been instrumental in discriminating between alternate possibilities in the assignments. Strong vibronic effects are expected to be present in the spectra of most monosubstituted benzenes, and the calculations presented here show that theoretical treatments based upon electronic structure calculations will generally be useful in the analysis of their spectra.

本研究旨在發現高中學生不同的「生涯發展組型」（career development patterns, CDP），進而對CDP 進行效度考驗，並探析其在升學及生涯輔導上的意義。本研究分三部分：一、組型建構：採取量化取向之調查法，根據20 所高中1,443 名學生在「工作價值觀量表」、「生涯探索量表」與「多元智能量表」上的評量結果，加上學科能力測驗（五科）成績，以總共三十種變項進行因素分析和典型相關分析，綜合研判組合成為八種有意義的CDP，包括實用組型、研究組型、藝術組型、社會組型、企業組型、事務組型、人文組型和數理組型；二、組型驗證：以4 所高中327 名學生為樣本，以「我的特質」及「學生特質」檢核表為效標，考驗兩者與CDP 之相關及高、低分組之差異，結果發現，八種CDP 與效標間大多呈現相對應的顯著正相關，也具有良好的同時效度，惟學生自評的效度高於教師的評定；三、組型應用：首先詮釋各CDP 的內涵，然後透過專家及資深輔導教師的諮詢座談，修正各組型內涵。最後，進行優勢組型（組型分數T≧60）之人數分布分析與個案分析，結果發現，優勢組型的分布存在著很大的個別差異，或屬於單一優勢，或屬於多重優勢，而各校均有優勢學生（合占約四成）。 根據研究發現，研究者提出若干對CDP 的應用及高中生升學與生涯輔導的建議。 The purposes of this study are twofolds: 1) To explore the “career development patterns” (CDPs) of senior high school students (SHSs) and define the meaning of the CDP; 2) To exmine the validity of the CDP and the application of the CDP to academic and career counseling. There are three parts of this study: 1) The construction of CDPs: 1,443 SHSs from 20 senior high schools took the Work Value Inventory, the Career Exploration Inventory, the Multiple Intelligences Scales, and the Academic Achievement Tests. Eight CDPs were then identified by means of factor analysis, cannonnical correlation analysis and comprehensive judgement based on 30 variables of the five measures. The eight CDPs were Realistic, Investigative, Artistic, Social, Enterprising, Conventional, Humanistic, and Scienticic; 2) The validitaion of CDPs: 327 students from 4 SHSs served as the subjects of validitaion. The “My Charactistics” and the “My Students’ Charactistics” checklists and rating scales were devised for students and teachers and exployed as the criteria of concurrent validity. It was found that most of the criterion measures were significantly correlated to CDP measures and had significant differential effects on two extreme CDP groups. However, students’ self-reports were found more effective than teachers’ ratings, and the concret and checklist method was more effective than the subjective over-all rating, especially for teachers; 3) The application of CDPs: The interpretation of CDPs was made and further revised by focus-group discussion, followed by identifing the subjects with superior CDPs (T≧60) at four senior high schools. It was found that there were 41.6% of the total subjects with one or more superior CDPs. However, there existed salient inter- and intra-individual differences in this regard. Finally, applications and suggestions of CDPs in academic and career counseling are given according to the research findings.

Sub-Doppler infrared rovibrational transitions in the symmetric (v1) and antisymmetric (v6) NH stretch modes of the isotopomerically substituted ND2H2+ ammonium cation are reported for the first time in a slit jet discharge supersonic expansion spectrometer. The partially H/D substituted cation is generated by selective isotopic exchange of ND3 with H2O to form NHD2, followed by protonation with H3+ formed in the NHD2/H2/Ne slit-jet discharge expansion environment. Rotational assignment for ND2H2+ is confirmed rigorously by four line ground state combination differences, which agree to be within the sub-Doppler precision in the slit jet (∼9 MHz). Observation of both b-type (ν1) and c-type (ν6) bands enables high precision determination of the ground and vibrationally excited state rotational constants. From an asymmetric top Watson Hamiltonian analysis, the ground state constants are found to be A″ = 4.856 75(4) cm−1, B″ = 3.968 29(4) cm−1, and C″ = 3.446 67(6) cm−1, with band origins at 3297.5440(1) and ...

Organ formation requires a delicate balance of positive and negative regulators. In Drosophila eye development, wingless (wg) is expressed at the lateral margins of the eye disc and serves to block retinal development. The T-box gene optomotor-blind (omb) is expressed in a similar pattern and is regulated by Wg. Omb mediates part of Wg activity in blocking eye development. Omb exerts its function primarily by blocking cell proliferation. These effects occur predominantly in the ventral margin. Our results suggest that the primary effect of Omb is the blocking of Jak/STAT signaling by repressing transcription of upd which encodes the Jak receptor ligand Unpaired.

The sub-Doppler CH-symmetric stretch (ν3) infrared absorption spectrum of a hydroxymethyl (CH2OH) radical is observed and analyzed with the radical formed in a slit-jet supersonic discharge expansion (Trot = 18 K) via Cl atom mediated H atom abstraction from methanol. The high sensitivity of the spectrometer and reduced spectral congestion associated with the cooled expansion enable first infrared spectroscopic observation of hydroxymethyl transitions from both ± symmetry tunneling states resulting from large amplitude COH torsional motion. Nuclear spin statistics due to exchange of the two methyl H-atoms aid in unambiguous rovibrational assignment of two A-type Ka = 0 ← 0 and Ka = 1 ← 1 bands out of each ± tunneling state, with additional spectral information obtained from spin-rotation splittings in P, Q, and R branch Ka = 1 ← 1 transitions that become resolved at low N. A high level ab initio potential surface (CCSD(T)-f12b/cc-pvnzf12 (n = 2,3)/CBS) is calculated in the large amplitude COH torsional and CH2 wag coordinates, which in the adiabatic approximation and with zero point correction predicts ground state tunneling splittings in good qualitative agreement with experiment. Of particular astrochemical interest, a combined fit of the present infrared ground state combination differences with recently reported millimeter-wave frequencies permits the determination of improved accuracy rotational constants for the ground vibrational state, which will facilitate ongoing millimeter/microwave searches for a hydroxymethyl radical in the interstellar medium.

Novel nanocomposites based on type I collagen (Col) containing a small amount (17.4, 43.5, and 174 ppm) of gold nanoparticles (AuNPs, approximately 5 nm) were prepared in this study. The pure Col and Col-AuNP composites (Col-Au) were characterized by the UV-Vis spectroscopy (UV-Vis), surface-enhanced raman spectroscopy (SERS) and atomic force microscopy (AFM). The interaction between Col and AuNPs was confirmed by infrared (IR) spectra. The effect of AuNPs on the biocompatibility of Col, evaluated by the proliferation and reactive oxygen species (ROS) production of mesenchymal stem cells (MSCs) as well as the activation of monocytes and platelets, was investigated. Results showed that Col-Au had better biocompatibility than Col. Upon stimulation by vascular endothelial growth factor (VEGF) and stromal derived factor-1α (SDF-1α), MSCs expressed the highest levels of αvβ3 integrin/CXCR4, focal adhesion kinase (FAK), matrix metalloproteinase-2 (MMP-2), and Akt/endothelial nitric oxide synthase (eNOS) proteins when grown on the Col-Au (43.5 ppm) nanocomposite. Taken together, Col-Au nanocomposites may promote the proliferation and migration of MSCs and stimulate the endothelial cell differentiation. These results suggest that Col-Au may be used to construct tissue engineering scaffolds for vascular regeneration.

Rovibrational spectroscopy of the fundamental OH stretching mode of the trans-HOCO radical has been studied via sub-Doppler high-resolution infrared laser absorption in a discharge slit-jet expansion. The trans-HOCO radical is formed by discharge dissociation of H2O to form OH, which then combines with CO and cools in the Ne expansion to a rotational temperature of 13.0(6) K. Rigorous assignment of both a-type and b-type spectral transitions is made possible by two-line combination differences from microwave studies, with full rovibrational analysis of the spectrum based on a Watson asymmetric top Hamiltonian. Additionally, fine structure splittings of each line due to electron spin are completely resolved, thus permitting all three ε(aa), ε(bb), ε(cc) spin-rotation constants to be experimentally determined in the vibrationally excited state. Furthermore, as both a- and b-type transitions for trans-HOCO are observed for the first time, the ratio of transition dipole moment projections along the a and b principal axes is determined to be μ(a)/μ(b) = 1.78(5), which is in close agreement with density functional quantum theoretical predictions (B3LYP/6-311++g(3df,3pd), μ(a)/μ(b) = 1.85). Finally, we note the energetic possibility in the excited OH stretch state for predissociation dynamics (i.e., trans-HOCO → H + CO2), with the present sub-Doppler line widths providing a rigorous upper limit of2.7 ns for the predissociation lifetime.

Nascent emission and laser-induced dispersed fluorescence spectra of products or intermediates from the multiphoton photolysis reaction of bromomethanes (CHBr(3), CHBr(2)Cl, CHBrCl(2), and CH(2)Br(2)) at 266 nm were recorded in a slow flow cell. Electronically excited species including CH (A(2)Delta, B(2)Sigma(-), and C(2)Sigma(+)), C(2) (d(3)Pi(g)), and atomic Br ((4)D(J) and (4)P(J)) were observed in the nascent emission spectra. Free radicals such CHBr or CHCl were also successfully found using laser-induced dispersed fluorescence spectroscopy. The reactive intermediate, CHBr, was seen only in the photolysis of CHBr(3), whereas CHCl was only discovered when the precursor was CHBr(2)Cl or CHBrCl(2). More experiments including the power dependence and temporal waveform measurements were conducted. The present study reports the first direct measurements of the intermediate products in the multiphoton photodissociation reaction of these bromomethanes at 266 nm. Nascent emission spectra following the photolysis at longer near-ultraviolet wavelengths (280 and 355 nm) were also acquired. On the bassis of these results, the multiphoton photodissociation mechanism of these bromomethanes at 266 nm can be confirmed.
