Recent studies suggest that coronaviruses circulate widely in Southeast Asian bat species and that the progenitors of the SARS-Cov-2 virus could have originated in rhinolophid bats in the region. Our objective was to assess the diversity and circulation patterns of coronavirus in several bat species in Southeast Asia. We undertook monthly live-capture sessions and sampling in Cambodia over 17 months to cover all phases of the annual reproduction cycle of bats and test specifically the association between their age and CoV infection status. We additionally examined current information on the reproductive phenology of Rhinolophus and other bat species presently known to occur in mainland southeast China, Vietnam, Laos and Cambodia. Results from our longitudinal monitoring (573 bats belonging to 8 species) showed an overall proportion of positive PCR tests for CoV of 4.2% (24/573) in cave-dwelling bats from Kampot and 4.75% (22/463) in flying-foxes from Kandal. Phylogenetic analysis showed that the PCR amplicon sequences of CoVs (n = 46) obtained clustered in Alphacoronavirus and Betacoronavirus. Interestingly, Hipposideros larvatus sensu lato harbored viruses from both genera. Our results suggest an association between positive detections of coronaviruses and juvenile and immature bats in Cambodia (OR = 3.24 [1.46-7.76], p = 0.005). Since the limited data presently available from literature review indicates that reproduction is largely synchronized among rhinolophid and hipposiderid bats in our study region, particularly in its more seasonal portions (above 16° N), this may lead to seasonal patterns in CoV circulation. Overall, our study suggests that surveillance of CoV in insectivorous bat species in Southeast Asia, including SARS-CoV-related coronaviruses in rhinolophid bats, could be targeted from June to October for species exhibiting high proportions of juveniles and immatures during these months. Bats have been identified as the natural hosts of several emerging zoonotic viruses, including paramyxoviruses, such as Hendra and Nipah viruses, that can cause fatal disease in humans. Recently, African fruit bats with populations that roost in or near urban areas have been shown to harbour a great diversity of paramyxoviruses, posing potential spillover risks to public health. Understanding the circulation of these viruses in their reservoir populations is essential to predict and prevent future emerging diseases. Here, we identify a high incidence of multiple paramyxoviruses in urine samples collected from a closed captive colony of circa 115 straw-coloured fruit bats ( Eidolon helvum ). The sequences detected have high nucleotide identities with those derived from free ranging African fruit bats and form phylogenetic clusters with the Henipavirus genus, Pararubulavirus genus and other unclassified paramyxoviruses. As this colony had been closed for 5 years prior to this study, these results indicate that within-host paramyxoviral persistence underlies the role of bats as reservoirs of these viruses.

Rabies transmitted by common vampire bats ( Desmodus rotundus ) has been known since the early 1900s but continues to expand geographically and in the range of species and environments affected. In this review, we present current knowledge of the epidemiology and management of rabies in D. rotundus and argue that it can be reasonably considered an emerging public health threat. We identify knowledge gaps related to the landscape determinants of the bat reservoir, reduction in bites on humans and livestock, and social barriers to prevention. We discuss how new technologies including autonomously-spreading vaccines and reproductive suppressants targeting bats might manage both rabies and undesirable growth of D. rotundus populations. Finally, we highlight widespread under-reporting of human and animal mortality and the scarcity of studies that quantify the efficacy of control measures such as bat culling. Collaborations between researchers and managers will be crucial to implement the next generation of rabies management in Latin America.

Assessing population trends and their underlying factors is critical to propose efficient conservation actions. This assessment can be particularly challenging when dealing with highly mobile, shy and nocturnal animals such as flying-foxes. Here we investigated the dynamics of hunted populations of Pteropus ornatus and P. tonganus in the Northern Province of New Caledonia. First, an ethno-ecological survey involving 219 local experts identified 494 flying-fox roosts. Current status was assessed for 379 of them, among which 125 were no longer occupied, representing a loss of 33% over ca. 40 years. Second, species-specific counts conducted at 35 roosts, and a sample of animals killed by hunters, revealed that the endemic species, P. ornatus, was dominant (68.5%). Between 2010 and 2016, 30 roosts were counted annually during the pre-parturition period. Roosts size averaged 1,425 ± 2,151 individuals (N = 180 counts) and showed high among-year variations (roost-specific CV = 37-162%). If we recorded significant inter-annual variation, we did not detect a significant decline over the 7-yr period, although one roost went possibly extinct. Population size of the two species combined was estimated at 338,000-859,000 individuals distributed over ca. 400 roosts in the Northern Province. Flying-foxes are popular game species and constitute traditional food for all communities of New Caledonia. Annual bags derived from a food survey allowed us to estimate harvesting rates at 5-14%. Such a level of harvesting for species with a 'slow' demography, the occurrence of poaching and illegal trade, suggest the current species use might not be sustainable and further investigations are critically needed., Competing Interests: The authors have declared that no competing interests exist.

Wildfire is an important ecological process that influences species' occurrence and biodiversity generally. Its effect on bats is understudied, creating challenges for habitat management and species conservation as threats to the taxa worsen globally and within fire-prone ecosystems. We conducted acoustic surveys of wildfire areas during 2014-2017 in conifer forests of California's Sierra Nevada Mountains. We tested effects of burn severity and its variation, or pyrodiversity, on occupancy and diversity for the 17-species bat community while accounting for imperfect detection. Occupancy rates increased with severity for at least 6 species and with pyrodiversity for at least 3. Two other species responded negatively to pyrodiversity. Individual species models predicted maximum occupancy rates across burn severity levels but only one species occurred most often in undisturbed areas. Species richness increased from approximately 8 species in unburned forests to 11 in pyrodiverse areas with moderate- to high-severity. Greater accessibility of foraging habitats, as well as increased habitat heterogeneity may explain positive responses to wildfire. Many bat species appear well adapted to wildfire, while a century of fire suppression and forest densification likely reduced habitat quality for the community generally. Relative to other taxa, bats may be somewhat resilient to increases in fire severity and size; trends which are expected to continue with accelerating climate change.

During mammalian pregnancy and lactation, the maternal demand for calcium is increased to satisfy fetus and newborn skeletal growth. In addition to the dietary intake, females use the calcium contained in their bones to supply this increased demand, leading to a decrease in maternal bone mineral content. In reproductive insectivorous female bats, bone loss has been described as a physiological cost of reproduction, due to the reported increased risk of bone fracture. This physiological cost may be the mechanism underlying the conflict between increasing litter size and maintaining wing skeletal integrity, which would help to explain the small litter size of most bat species. If bone loss is a linking cost between reproduction and survival in bats, and most bat species have small litter sizes, one would expect to find a loss of bone and an increasing probability of bone fracture during pregnancy and lactation in other non-insectivorous bats. In this study, we tested for the existence of this cost in the Great-fruit eating bat, Artibeus lituratus. We analyzed trabecular structure, bone strength and bone mineral content for the humerus bone, hypothesizing that bone loss during reproduction in females would increase the risk of fracture. Our results showed a decrease of 22-31% in bone trabecular area in lactating females, rapidly compensated following weaning. Bone strength did not differ among reproductive and non-reproductive groups and seems to be more influenced by bone organic components rather than mineral contents. Since we observed bone loss during reproduction yet the humerus strength seems to be unaffected, we suggest that bone loss may not represent a physiological cost during reproduction for this frugivorous bat., Competing Interests: The authors have declared that no competing interests exist.

Metal elements, ubiquitous in the environment, can cause negative effects in long-lived organisms even after low but prolonged exposure. Insectivorous bats living near metal emission sources can be vulnerable to such contaminants. Although it is known that bats can bioaccumulate metals, little information exists on the effects of metal elements on their physiological status. For example, oxidative status markers are known to vary after detoxification processes and immune reactions. Here, for two consecutive summers, we sampled individuals from a natural population of the insectivorous bat, Myotis daubentonii, inhabiting a site close to a metal emission source. We quantified metals and metalloids (As, Ca, Cd, Co, Cu, Mn, Ni, Pb, Se, Zn) from individual fecal pellets. We measured enzymatic antioxidants (GP, CAT, SOD), total glutathione (tGSH) and ratio between reduced and oxidized glutathione (GSH:GSSG) from their red blood cells together with biometrics, hematocrit and parasite prevalence. In general, metal concentrations in feces of M. daubentonii reflected the exposure to ambient contamination. This was especially evident in the higher concentrations of Cd, Co, Cu and Ni close to a smelter compared to a site with less contaminant exposure. Annual differences were also observed for most elements quantified. Sex-specific differences were observed for calcium and zinc excretion. SOD and CAT enzymatic activities were associated with metal levels (principal components of six metal elements), suggesting early signs of chronic stress in bats. Reproductive seasonality in Neotropical bats has been assessed to the better understand their reproductive behavior. This knowledge is especially important for the control of Desmodus rotundus population as it is a transmitter of rabies virus. Therefore, we aimed to evaluate the functional activity of testis and epididymis of D. rotundus in dry and rainy seasons under a morphological approach. We observed an increase in tubular diameter and epithelial height of the seminiferous tubules during the rainy season. In the latter, additionally, stereological analysis of the testis showed increased proportion of seminiferous epithelium and reduced percentage of lumen. The sperm number in caput/corpus epididymis increased in rainy season, whereas sperm count and transit time were reduced in cauda region. These alterations were probably related to the recovery of epithelium activities after mating season in dry season. Despite altered nuclear and cytoplasm parameters of Leydig cells between seasons, the volume and number of these cells were constant. Moreover, no change in serum testosterone levels, daily sperm production, and apoptotic index were observed, which indicates that the reproductive pattern in D. rotundus does not change between seasons. Flying foxes have been widely studied as they are well-known reservoirs of infectious agents. Understanding their population dynamics might help to explain seasonal patterns of disease prevalence, and contribute towards the conservation of flying fox populations. Therefore, this study explored the annual variation in the number of deaths in P. lylei. The study was conducted from 2015-2017, at a Buddhist temple in Thailand, which is the roosting site of P. lylei. The average total number of bat deaths in a month significantly varied between times of a year. A peak was observed during March and May, which ranged in the period of birthing and lactating. There were no significant differences in the average total number of bat deaths in a month between sexes or age classes across times of a years.

We estimated U.S. and Mexican citizens' willingness to pay (WTP) for protecting habitat for a transborder migratory species, the Mexican free-tailed bat (Tadarida brasiliensis mexicana), using the contingent valuation method. Few contingent valuation surveys have evaluated whether households in one country would pay to protect habitat in another country. This study addresses that gap. In our study, Mexican respondents were asked about their WTP for conservation of Mexican free-tailed bat habitat in Mexico and in the United States. Similarly, U.S. respondents were asked about their WTP for conservation in the United States and in Mexico. U.S. households would pay $30 annually to protect habitat in the United States and $24 annually to protect habitat in Mexico. Mexican households would pay $8 annually to protect habitat in Mexico and $5 annually to protect habitat in the United States. In both countries, these WTP amounts rose significantly for increasing the size of the bat population rather than simply stabilizing the current bat population. The ratio of Mexican household WTP relative to U.S. household WTP is nearly identical to that of Mexican household income relative to U.S. household income. This suggests that the perceived economic benefits received from the bats is similar in Mexico and the United States, and that scaling WTP by relative income in international benefit transfer may be plausible.

Monitoring flying-foxes is challenging as their extreme mobility produces highly dynamic population processes, considerable logistic difficulty, and variability in estimated population size. We report on methods for inferring population trend for the population of the spectacled flying-fox (Pteropus conspicillatus) in Australia. Monthly monitoring is conducted at all known roost sites across the species' range in the Wet Tropics Region. The proportion of animals in camps varies seasonally and stochastic environmental events appear to be influential. We develop a state-space model that incorporates these processes and enables inference on total population trends and uses early warning analysis to identify the causes of population dynamics. The model suggests that population growth rate is stable in the absence of cyclones, however, cyclones appear to impact on both survival and reproduction. The population recovered after two cyclones but declined after a third. The modelling estimates a population decline over 15 years of c. 75% (mean r = - 0.12yr -1 and belief of negative trend is c. 83%) suggesting that conservation action is warranted. Our work shows that a state-space modelling approach is a significant improvement on inference from raw counts from surveys and demonstrates that this approach is a workable alternative to other methods.

Rensch's rule, stating that sexual size dimorphism (SSD) becomes more evident and male-biased with increasing body size, has been well supported for taxa that exhibit male-biased SSD. Bats, primarily having female-biased SSD, have so far been tested for whether SSD allometry conforms to Rensch's rule in only three studies. However, these studies did not consider phylogeny, and thus the mechanisms underlying SSD variations in bats remain unclear. Thus, the present study reviewed published and original data, including body size, baculum size, and habitat types in 45 bats of the family Rhinolophidae to determine whether horseshoe bats follow Rensch's rule using a phylogenetic comparative framework. We also investigated the potential effect of postcopulatory sexual selection and habitat type on SSD. Our findings indicated that Rensch's rule did not apply to Rhinolophidae, suggesting that SSD did not significantly vary with increasing size. This pattern may be attributable interactions between weak sexual selection to male body size and strong fecundity selection for on female body size. The degree of SSD among horseshoe bats may be attributed to a phylogenetic effect rather than to the intersexual competition for food or to baculum length. Interestingly, we observed that species in open habitats exhibited greater SSD than those in dense forests, suggesting that habitat types may be associated with variations in SSD in horseshoe bats.

Auditory feedback plays an important role in vocal learning and, more generally, in fine-tuning the acoustic features of communication signals. So far, only a few studies have assessed the developmental onset of auditory feedback. The Lombard effect, a well-studied audio-vocal phenomenon, refers to an increase in vocal loudness of a subject in response to an increase in background noise. Here, we studied the time course of the Lombard effect in developing bats, Phyllostomus discolor We show that infant bats produced louder vocalizations in noise than in silence at an age of only 2 weeks. In contrast, the infant bats' morphology and vocalizations changed gradually until 2 months of age. Furthermore, we found that the Lombard magnitude, i.e. how much the bats increased their vocal loudness in noise relative to silence, correlated positively with the age of the infant bats. Functional diversity is the variability in the functional roles carried out by species within ecosystems. Changes in the environment can affect this component of biodiversity and can, in turn, affect different processes, including some ecosystem services. This study aimed to determine the effect of forest loss on species richness, abundance and functional diversity of Neotropical bats. To this end, we identified six landscapes with increasing loss of forest cover in the Huasteca region of the state of Hidalgo, Mexico. We captured bats in each landscape using mist nets, and calculated functional diversity indices (functional richness and functional evenness) along with species richness and abundance. We analyzed these measures in terms of percent forest cover. We captured 906 bats (Phyllostomidae and Mormoopidae), including 10 genera and 12 species. Species richness, abundance and functional richness per night are positively related with forest cover. Generalized linear models show that species richness, abundance and functional richness per night are significantly related with forest cover, while seasonality had an effect on abundance and functional richness. Neither forest cover nor season had a significant effect on functional evenness. All these findings were consistent across three spatial scales (1, 3 and 5 km radius around sampling sites). The decrease in species, abundance and functional richness of bats with forest loss may have implications for the ecological processes they carry out such as seed dispersal, pollination and insect predation, among others., Competing Interests: The authors have declared that no competing interests exist.

Ecomorphology studies focus on understanding how anatomical and behavioral diversity result in differences in performance, ecology, and fitness. In mammals, the determinate growth of the skeleton entails that bite performance should change throughout ontogeny until the feeding apparatus attains its adult size and morphology. Then, interspecific differences in adult phenotypes are expected to drive food resource partitioning and patterns of lineage diversification. However, Formal tests of these predictions are lacking for the majority of mammal groups, and thus our understanding of mammalian ecomorphology remains incomplete. By focusing on a fundamental measure of feeding performance, bite force, and capitalizing on the extraordinary morphological and dietary diversity of bats, we discuss how the intersection of ontogenetic and macroevolutionary changes in feeding performance may impact ecological diversity in these mammals. We integrate data on cranial morphology and bite force gathered through longitudinal studies of captive animals and comparative studies of free-ranging individuals. We demonstrate that ontogenetic trajectories and evolutionary changes in bite force are highly dependent on changes in body and head size, and that bats exhibit dramatic, allometric increases in bite force during ontogeny. Interspecific variation in bite force is highly dependent on differences in cranial morphology and function, highlighting selection for ecological specialization. While more research is needed to determine how ontogenetic changes in size and bite force specifically impact food resource use and fitness in bats, interspecific diversity in cranial morphology and bite performance seem to closely match functional differences in diet. The molecular events leading to the development of the bat wing remain largely unknown, and are thought to be caused, in part, by changes in gene expression during limb development. These expression changes could be instigated by variations in gene regulatory enhancers. Here, we used a comparative genomics approach to identify regions that evolved rapidly in the bat ancestor, but are highly conserved in other vertebrates. We discovered 166 bat accelerated regions (BARs) that overlap H3K27ac and p300 ChIP-seq peaks in developing mouse limbs. Using a mouse enhancer assay, we show that five Myotis lucifugus BARs drive gene expression in the developing mouse limb, with the majority showing differential enhancer activity compared to the mouse orthologous BAR sequences. These include BAR116, which is located telomeric to the HoxD cluster and had robust forelimb expression for the M. lucifugus sequence and no activity for the mouse sequence at embryonic day 12.5. Developing limb expression analysis of Hoxd10-Hoxd13 in Miniopterus natalensis bats showed a high-forelimb weak-hindlimb expression for Hoxd10-Hoxd11, similar to the expression trend observed for M. lucifugus BAR116 in mice, suggesting that it could be involved in the regulation of the bat HoxD complex. Combined, our results highlight novel regulatory regions that could be instrumental for the morphological differences leading to the development of the bat wing.

Long-distance migration is a rare phenomenon in European bats. Genetic analyses and banding studies show that females can cover distances of up to 1,600 km, whereas males are sedentary or migrate only short distances. The onset of this sex-biased migration is supposed to occur shortly after rousing from hibernation and when the females are already pregnant. We therefore predicted that the sexes are exposed to different energetic pressures in early spring, and this should be reflected in their behavior and physiology. We investigated this in one of the three Central European long-distance migrants, the common noctule (Nyctalus noctula) in Southern Germany recording the first individual partial migration tracks of this species. In contrast to our predictions, we found no difference between male and female home range size, activity, habitat use or diet. Males and females emerged from hibernation in similar body condition and mass increase rate was the same in males and females. We followed the first migration steps, up to 475 km, of radio-tagged individuals from an airplane. All females, as well as some of the males, migrated away from the wintering area in the same northeasterly direction. Sex differences in long-distance migratory behavior were confirmed through stable isotope analysis of hair, which showed greater variation in females than in males. We hypothesize that both sexes faced similarly good conditions after hibernation and fattened at maximum rates, thus showing no differences in their local behavior. Interesting results that warrant further investigation are the better initial condition of the females and the highly consistent direction of the first migratory step in this population as summering habitats of the common noctule occur at a broad range in Northern Europe. Only research focused on individual strategies will allow us to fully understand the migratory behavior of European bats.

Over the past several decades, several large-scale seemingly unrelated events have unfolded in all corners of the world. Within the oceans, coral reef systems have been facing unprecedented mass bleaching episodes, sea turtles worldwide are currently experiencing an epidemic in the form of fibropapilloma, and global phytoplankton populations have declined by 40%. Within the Earth's terrestrial systems, similar phenomena have appeared in the form of colony collapse disorder (CCD) currently devastating honey bee colonies, White Nose Syndrome decimating bat populations, and the chytrid fungus plaguing amphibian populations. On the surface these events appear to be unrelated yet at the root of each phenomenon there appears an underlying threat - pollution. Bats are the only mammals capable of true flight. Critical adaptations for flight include a pair of dramatically elongated hands with broad wing membranes. To study the molecular mechanisms of bat wing evolution, we perform genomewide mRNA sequencing and in situ hybridization for embryonic bat limbs. We identify seven key genes that display unique expression patterns in embryonic bat wings and feet, compared with mouse fore- and hindlimbs. The expression of all 5'HoxD genes (Hoxd9-13) and Tbx3, six known crucial transcription factors for limb and digit development, is extremely high and prolonged in the elongating wing area. The expression of Fam5c, a tumour suppressor, in bat limbs is bat-specific and significantly high in all short digit regions (the thumb and foot digits). These results suggest multiple genetic changes occurred independently during the evolution of bat wings to elongate the hand digits, promote membrane growth and keep other digits short. Our findings also indicate that the evolution of limb morphology depends on the complex integration of multiple gene regulatory networks and biological processes that control digit formation and identity, chondrogenesis, and interdigital regression or retention.

Morphological evolution can be the result of natural selection favoring modification of developmental signaling pathways. However, little is known about the genetic basis of such phenotypic diversity. Understanding these mechanisms is difficult for numerous reasons, yet studies in model organisms often provide clues about the major developmental pathways involved. The paired-domain gene, PAX9, is known to be a key regulator of development, particularly of the face and teeth. In this study, using a comparative genetics approach, we investigate PAX9 molecular evolution among mammals, focusing on craniofacially diversified (Phyllostomidae) and conserved (Vespertilionidae) bat families, and extend our comparison to other orders of mammal. Open-reading frame analysis disclosed signatures of selection, in which a small percentage of residues vary, and lineages acquire different combinations of variation through recurrent substitution and lineage specific changes. A few instances of convergence for specific residues were observed between morphologically convergent bat lineages. Bioinformatic analysis for unknown PAX9 regulatory motifs indicated a novel post-transcriptional regulatory mechanism involving a Musashi protein. This regulation was assessed through fluorescent reporter assays and gene knockdowns. Results are compatible with the hypothesis that the number of Musashi binding-elements in PAX9 mRNA proportionally regulates protein translation rate. Although a connection between morphology and binding element frequency was not apparent, results indicate this regulation would vary among craniofacially divergent bat species, but be static among conserved species. Under this model, Musashi's regulatory control of alternative human PAX9 isoforms would also vary. The presence of Musashi-binding elements within PAX9 of all mammals examined, chicken, zebrafish, and the fly homolog of PAX9, indicates this regulatory mechanism is ancient, originating basal to much of the animal phylogeny.

Tacaribe virus (TCRV) was first isolated from 11 Artibeus species bats captured in Trinidad in the 1950s during a rabies virus surveillance program. Despite significant effort, no evidence of infection of other mammals, mostly rodents, was found, suggesting that no other vertebrates harbored TCRV. For this reason, it was hypothesized that TCRV was naturally hosted by artibeus bats. This is in stark contrast to other arenaviruses with known hosts, all of which are rodents. To examine this hypothesis, we conducted experimental infections of Jamaican fruit bats (Artibeus jamaicensis) to determine whether they could be persistently infected without substantial pathology. We subcutaneously or intranasally infected bats with TCRV strain TRVL-11573, the only remaining strain of TCRV, and found that low-dose (10(4) 50% tissue culture infective dose [TCID(50)]) inoculations resulted in asymptomatic and apathogenic infection and virus clearance, while high-dose (10(6) TCID(50)) inoculations caused substantial morbidity and mortality as early as 10 days postinfection. Uninoculated cage mates failed to seroconvert, and viral RNA was not detected in their tissues, suggesting that transmission did not occur. Together, these data suggest that A. jamaicensis bats may not be a reservoir host for TCRV.

Neuronal computation of object distance from echo delay is an essential task that echolocating bats must master for spatial orientation and the capture of prey. In the dorsal auditory cortex of bats, neurons specifically respond to combinations of short frequency-modulated components of emitted call and delayed echo. These delay-tuned neurons are thought to serve in target range calculation. It is unknown whether neuronal correlates of active space perception are established by experience-dependent plasticity or by innate mechanisms. Here we demonstrate that in the first postnatal week, before onset of echolocation and flight, dorsal auditory cortex already contains functional circuits that calculate distance from the temporal separation of a simulated pulse and echo. This innate cortical implementation of a purely computational processing mechanism for sonar ranging should enhance survival of juvenile bats when they first engage in active echolocation behaviour and flight.

By acquiring wings, bats are the only mammalian lineage to have achieved flight. To be capable of powered flight, they have unique muscles associated with their wing. However, the developmental origins of bat wing muscles, and the underlying molecular and cellular mechanisms are unknown. Here we report, first, that the wing muscles are derived from multiple myogenic sources with different embryonic origins, and second, that there is a spatiotemporal correlation between the outgrowth of wing membranes and the expansion of wing muscles into them. Together, these findings imply that the wing membrane itself may regulate the patterning of wing muscles. Last, through comparative gene expression analysis, we show Fgf10 signalling is uniquely activated in the primordia of wing membranes. Our results demonstrate how components of Fgf signalling are likely to be involved in the development and evolution of novel complex adaptive traits.

Developmental changes in vocalizations by Pipistrellus abramus were investigated during the first post-natal month. Vocalizations by pups on the day of birth were frequency-modulated ultrasounds from 30.0 ± 4.0 kHz to 19.3 ± 1.9 kHz with multiple harmonics. The terminal frequency of the second harmonic (TF(2)) of pup vocalizations corresponded to that of the fundamental (TF(1)) in adult bats (41.4 ± 2.6 kHz), suggesting that pup vocalizations can be easily detected by the mother. Background: Human activities, such as agriculture, hunting, and habitat modification, exert a significant effect on native species. Although many species have suffered population declines, increased population fragmentation, or even extinction in connection with these human impacts, others seem to have benefitted from human modification of their habitat. Here we examine whether population growth in an insectivorous bat (Tadarida brasiliensis mexicana) can be attributed to the widespread expansion of agriculture in North America following European settlement. Colonies of T. b. mexicana are extremely large (~10(6) individuals) and, in the modern era, major agricultural insect pests form an important component of their food resource. It is thus hypothesized that the growth of these insectivorous bat populations was coupled to the expansion of agricultural land use in North America over the last few centuries., Results: We sequenced one haploid and one autosomal locus to determine the rate and time of onset of population growth in T. b. mexicana. Using an approximate Maximum Likelihood method, we have determined that T. b. mexicana populations began to grow ~220 kya from a relatively small ancestral effective population size before reaching the large effective population size observed today., Conclusions: Our analyses reject the hypothesis that T. b. mexicana populations grew in connection with the expansion of human agriculture in North America, and instead suggest that this growth commenced long before the arrival of humans. As T. brasiliensis is a subtropical species, we hypothesize that the observed signals of population growth may instead reflect range expansions of ancestral bat populations from southern glacial refugia during the tail end of the Pleistocene.

Background: We address three key gaps in research on urban wildlife ecology: insufficient attention to (1) grassland biomes, (2) individual- and population-level effects, and (3) vertebrates other than birds. We hypothesized that urbanization in the North American Prairies, by increasing habitat complexity (via the proliferation of vertical structures such as trees and buildings), thereby enhancing the availability of day-roosts, tree cover, and insects, would benefit synanthropic bats, resulting in increased fitness among urban individuals., Methodology/principal Findings: Over three years, we captured more than 1,600 little brown bats (Myotis lucifugus) in urban and non-urban riparian sites in and around Calgary, Alberta, Canada. This species dominated bat assemblages throughout our study area, but nowhere more so than in the city. Our data did not support most of our specific predictions. Increased numbers of urban bats did not reflect urbanization-related benefits such as enhanced body condition, reproductive rates, or successful production of juveniles. Instead, bats did best in the transition zone situated between strictly urban and rural areas., Conclusions/significance: We reject our hypothesis and explore various explanations. One possibility is that urban and rural M. lucifugus exhibit increased use of anthropogenic roosts, as opposed to natural ones, leading to larger maternity colonies and higher population densities and, in turn, increased competition for insect prey. Other possibilities include increased stress, disease transmission and/or impacts of noise on urban bats. Whatever the proximate cause, the combination of greater bat population density with decreased body condition and production of juveniles indicates that Calgary does not represent a population source for Prairie bats. We studied a highly synanthropic species in a system where it could reasonably be expected to respond positively to urbanization, but failed to observe any apparent benefits at the individual level, leading us to propose that urban development may be universally detrimental to bats.

The fulvous fruit bat (Rousettus leschenaulti) and the greater short-nosed fruit bat (Cynopterus sphinx) are two abundant and widely co-distributed Old World fruit bats in Southeast and East Asia. The former species forms large colonies in caves while the latter roots in small groups in trees. To test whether these differences in social organization and roosting ecology are associated with contrasting patterns of gene flow, we used mtDNA and nuclear loci to characterize population genetic subdivision and phylogeographic histories in both species sampled from China, Vietnam and India. Our analyses from R. leschenaulti using both types of marker revealed little evidence of genetic structure across the study region. On the other hand, C. sphinx showed significant genetic mtDNA differentiation between the samples from India compared with China and Vietnam, as well as greater structuring of microsatellite genotypes within China. Demographic analyses indicated signatures of past rapid population expansion in both taxa, with more recent demographic growth in C. sphinx. Therefore, the relative genetic homogeneity in R. leschenaulti is unlikely to reflect past events. Instead we suggest that the absence of substructure in R. leschenaulti is a consequence of higher levels of gene flow among colonies, and that greater vagility in this species is an adaptation associated with cave roosting.

The primary auditory cortex (AI) of adult Pteronotus parnellii features a foveal representation of the second harmonic constant frequency (CF2) echolocation call component. In the corresponding Doppler-shifted constant frequency (DSCF) area, the 61 kHz range is over-represented for extraction of frequency-shift information in CF2 echoes. To assess to which degree AI postnatal maturation depends on active echolocation or/and reflects ongoing cochlear maturation, cortical neurons were recorded in juveniles up to postnatal day P29, before the bats are capable of active foraging. At P1-2, neurons in posterior AI are tuned sensitively to low frequencies (22-45 dB SPL, 28-35 kHz). Within the prospective DSCF area, neurons had insensitive responses (>60 dB SPL) to frequencies <40 kHz and lacked sensitive tuning curve tips. Up to P10, when bats do not yet actively echolocate, tonotopy is further developed and DSCF neurons respond to frequencies of 51-57 kHz with maximum tuning sharpness (Q(10dB)) of 57. Between P11 and 20, the frequency representation in AI includes higher frequencies anterior and dorsal to the DSCF area. More multipeaked neurons (33%) are found than at older age. In the oldest group, DSCF neurons are tuned to frequencies close to 61 kHz with Q(10dB) values < or =212, and threshold sensitivity, tuning sharpness and cortical latencies are adult-like. The data show that basic aspects of cortical tonotopy are established before the bats actively echolocate. Maturation of tonotopy, increase of tuning sharpness, and upward shift in the characteristic frequency of DSCF neurons appear to strongly reflect cochlear maturation.

Vocal imitation--the ability to learn a previously unknown acoustic signal from a tutor--is considered to be a key innovation in the evolution of speech. This faculty is very rare and patchily distributed within the animal kingdom, suggesting multiple instances of convergent evolution. It has long been predicted that bats should be capable of vocal imitation and our results provide evidence for this phenomenon. We report that pups of the bat Saccopteryx bilineata learn a complex vocalization through vocal imitation. During ontogeny, pups of both sexes imitate territorial song from adult males, starting with simple precursor songs that develop into genuine renditions. The resemblance of pup renditions to their acoustic model is not caused by physical maturation effects, is independent of pups' gender and relatedness towards adult males and becomes more pronounced during ontogeny, showing that auditory experience is essential for vocal development. Our findings indicate that the faculty of vocal imitation is more widespread than previously thought and emphasize the importance of research on audiovocal communication in bats for a better understanding of the evolutionary origin of vocal imitation.

Nonfunctional visual genes are usually associated with species that inhabit poor light environments (aquatic/subterranean/nocturnal), and these genes are believed to have lost function through relaxed selection acting on the visual system. Indeed, the visual system is so adaptive that the reconstruction of intact ancestral opsin genes has been used to reject nocturnality in ancestral primates. To test these assertions, we examined the functionality of the short and medium- to long-wavelength opsin genes in a group of mammals that are supremely adapted to a nocturnal niche: the bats. We sequenced the visual cone opsin genes in 33 species of bat with diverse sensory ecologies and reconstructed their evolutionary history spanning 65 million years. We found that, whereas the long-wave opsin gene was conserved in all species, the short-wave opsin gene has undergone dramatic divergence among lineages. The occurrence of gene defects in the short-wave opsin gene leading to loss of function was found to directly coincide with the origin of high-duty-cycle echolocation and changes in roosting ecology in some lineages. Our findings indicate that both opsin genes have been under purifying selection in the majority bats despite a long history of nocturnality. However, when spectacular losses do occur, these result from an evolutionary sensory modality tradeoff, most likely driven by subtle shifts in ecological specialization rather than a nocturnal lifestyle. Our results suggest that UV color vision plays a considerably more important role in nocturnal mammalian sensory ecology than previously appreciated and highlight the caveat of inferring light environments from visual opsins and vice versa.

Mammals have evolved a variety of morphological adaptations that have allowed them to compete in their natural environments. The developmental genetic basis of this morphological diversity remains largely unknown. Bats are mammals that have the unique ability of powered flight. We have examined the molecular embryology of bats and investigated the developmental genetic basis for their highly derived limbs used for flight. Initially, we developed an embryo staging system for a model chiropteran, Carollia perspicillata, the short-tailed fruit bat that has subsequently been used for staging other bat species. Expression studies focusing on genes that regulate limb development indicate that there are similarities and differences between bats and mice. To determine the consequences of these expression differences, we have conducted an enhancer switch assay by gene targeting in mouse embryonic stem cells to create mice whose genes are regulated by bat sequences. Our studies indicate that cis-regulatory elements contribute to the morphological differences that have evolved among mammalian species.

Group fission is an important dispersal mechanism for philopatric adults. In Cypress Hills Interprovincial Park, Saskatchewan, tree-roosting big brown bats (Eptesicus fuscus) exhibit fission-fusion roosting behaviour. During 2004-2007, the majority of females previously resident to roosting area 1 (RA1) moved to a new roosting area (RA4). We examined how genetic relationships, inferred from data for microsatellite loci and mitochondrial DNA, influenced new roost area (RA) selection during 2006 when colony members were split between the RAs. We found that females who moved to RA4 had higher average relatedness than those that remained in RA1. We found that nearly all females belonging to matrilines with high average relatedness moved to RA4 while females from matrilines with low average relatedness were split between the two RAs. These results suggest that closely related maternal kin preferentially move to new RAs. However, daily roosting preferences within a RA are not based on genetic relationships probably because daily roosting associations between kin and non-kin are used to ensure adequate roost group size. Studying the effects of kinship on the fission and movements of groups not only enhances our understanding of social behaviour and population genetics but also informs conservation decisions.

1. Landscape connectivity may greatly influence the distribution of animals when it alters their movements and their ability to reach food patches. Depending on their foraging behaviour, organisms may or may not adapt to anthropogenic changes in landscape connectivity and may eventually undergo local extinctions. 2. Recent studies underlined the need to use indicators of functional landscape connectivity based on the behaviour and movement abilities of studied animals to better link landscape structure to ecological processes in disturbed and fragmented areas. 3. The objectives of this study were: to elaborate an index of functional connectivity for Rhinophylla pumilio, a Neotropical understorey frugivorous bat; to use this index to investigate the possible mechanisms controlling its distribution and sustainability in a fragmented landscape; and to test whether this index could be applied to other species of the same guild. 4. We pursued a 10-year bat mist-net survey, coupled to local estimates of food availability, in a mature forest of French Guiana that was recently fragmented by the completion of a reservoir lake. The 18 sampling sites range from undisturbed continuous forest sites to small remote forest fragments. A connectivity value, based on radio-tracking surveys, was attributed to each site. Connectivity measures mean forest cover within neighbouring landscape units, weighted by the probability that bats would use them, as estimated by frequency distribution of flight distance data. 5. The abundance of R. pumilio was positively correlated with landscape connectivity and not correlated with local food availability. Its foraging strategy has evolved in response to the highly scattered distribution of its fruit resource. In spite of its high mobility, R. pumilio apparently failed to exploit a food resource that is distributed patchily over a low-connective habitat because its foraging movements are not well adapted to habitat disruptions. 6. The connectivity index contributed to explain general tendencies of abundance variations in other understorey frugivorous bats, although the spatial scale we examined was probably too small for these species. We make recommendations to adapt a functional connectivity index to species whose large-scale movements are difficult to survey.

Unplanned natural and anthropogenic disasters provide unique opportunities for investigating the influence of perturbations on population vital rates and species recovery times. We investigated the potential effects of a major pesticide spill by comparing annual survival rates using mark-recapture techniques on a riparian bat species, Yuma Myotis (Myotis yumanensis). Demography and population dynamics for most bat species remain poorly understood despite advances in mark-recapture estimation and modeling techniques. We compared survival and population growth rates of two roost populations exposed to a large chemical (metam sodium) spill in the upper Sacramento River in Northern California with two roost populations outside the contaminated area from 1992 to 1996. Hypotheses about long-term effects of the spill on female juvenile and adult survival were tested using an information-theoretic approach (AIC). Working hypotheses included effects of age, chemical spill, and time trend on survival. Female adult survival was higher than female juvenile survival across all sites, suggesting stage-specific mortality risks. Model-averaged estimates of female juvenile survival in the contaminated area (0.50-0.74) were lower than in control roosts (0.60-0.78) for each year in the study, suggesting that the spill may have reduced juvenile survival for several years. Female adult survival (0.73-0.89) did not appear to be strongly affected by the spill during the years of the study. There was an increase in survival for both stage-classes across all populations during the study period, which may have been caused by the end of an extended drought in California in the winter of 1993. The spill-affected population was in decline for the first year of the study as indicated by an estimated growth rate (lambda) < 1, but population growth rates increased during the four-year period.

In recent times, the apparent population decline of the southern bent-wing bat (Miniopterus schreibersii bassanii) at Bat Cave, Naracoorte has been ascribed to pesticide use in the region, following the finding of organochlorine and orgaonophosphate insecticide residues in bat guano. Adult southern bent-wing bats were collected from Bat Cave and Starlight Cave in 2003. Organochlorine contaminants were detected in all carcass samples: p,p'-DDE was by far the most dominant contaminant with concentrations ranging from 11,000 to 59,000ngg(-1), followed by p,p'-DDT (110-1600ngg(-1)), p,p'-DDD (35-620ngg(-1)), summation operatorPCBs (33-490ngg(-1)), summation operatorchlordane and related compounds (7.9-270ngg(-1)), HCB (1.6-120ngg(-1)), HP epox. (3.1-230ngg(-1)), TCPMOH (3.8-38ngg(-1)), summation operatorHCHs (1.4-9.6ngg(-1)), and TCPMe (0.1-4.2ngg(-1)) (all values on lipid-weight basis). No significant difference in DDE, DDD, DDT, summation operatorDDT, summation operatorPCB, trans-chlordane, heptachlor epoxide, trans-nonachlor, alpha-HCH, beta-HCH, gamma-HCH, TCPMOH or TCPMe concentrations were observed either between sexes within sites, or between sites (p>0.05). However, there were significant differences in HCB and oxychlordane concentrations between sexes and between sites (p<0.05), between site differences in cis-nonachlor concentrations in male bats (p<0.05), and cis-chlordane concentrations between sexes at Starlight Cave, and between males of each site (p<0.05). There were also significant differences in the liver concentrations of some metals between sexes within sites (Ag, Cd, Co, Cu, Pb, Se, Zn), and between sites (Ag, Cd, Co, Cu, Hg, Pb, Se, V, Zn). Clustering or grouping of sites was observed when the OC data was expressed on a lipid-weight basis. These inter-site differences in OC concentrations reflect local exposure over a period of time, and do not unambiguously support any suggestion that we are witnessing incipient speciation. However, for conservation purposes, it may be prudent to assume that there are two sub-populations of M. s. bassani feeding in different locations in this region of southern Australia, rather than the single homogeneous population suggested by genetic studies.

Postnatal development of the mustached bat's cochlea was studied by measuring cochlear microphonic and compound action potentials. In adults, a cochlear resonance is involved in enhanced tuning to the second harmonic constant frequency component (CF2) of their echolocation calls at approximately 61 kHz This resonance is present immediately after birth in bats that do not yet echolocate. Its frequency is lower (46 kHz) and the corresponding threshold minimum of cochlear microphonic potentials is broader than in adults. Long-lasting ringing of the cochlear microphonic potential after tone stimulus offset that characterizes the adult auditory response close to CF2 is absent in newborns. In the course of the first 5 postnatal weeks, there is a concomitant upward shift of CF2 and the frequency of cochlear threshold minima. Up to the end of the third postnatal week, sensitivity of auditory threshold minima and the Q value of the cochlear resonance increase at a fast rate. Between 2 and 4 wk of age, two cochlear microphonic threshold minima are found consistently in the CF2 range that differ in their level-dependent dynamic growth behavior and are 1.5-5.7 kHz apart from each other. In older animals, there is a single minimum that approaches adult tuning in its sharpness. The data provide evidence to show that during maturation of the cochlea, the frequency and the sensitivity of the threshold minimum associated with CF2 increases and that these increases are associated with the fusion of two resonances that are partly dissociated in developing animals.

Adult mustached bats employ Doppler-sensitive sonar to hunt fluttering prey insects in acoustically cluttered habitats. The echolocation call consists of 4-5 harmonics, each composed of a long constant frequency (CF) component flanked by brief frequency modulations (FM). The 2nd harmonic CF component (CF2) at 61 kHz is the most intense, and analyzed by an exceptionally sharply tuned auditory system. The maturation of echolocation calls and the development of Doppler-shift compensation was studied in Cuba where large maternity colonies are found in hot caves. In the 1st postnatal week, infant bats did not echolocate spontaneously but could be induced to vocalize CF-FM signals by passive body motion. The CF2 frequency emitted by the smallest specimens was at 48 kHz (i.e., 0.4 octaves lower than the adult signal). CF-FM signals were spontaneously produced in the 2nd postnatal week at a CF2 frequency of 52 kHz. The CF2 frequencies of induced and spontaneous calls shifted upward to reach a value of 60.5 kHz in the 5th postnatal week. Standard deviations of CF2 frequency were large (up to +/-1.5 kHz) in the youngest bats and dropped to values of +/-250 Hz at the end of the 3rd postnatal week. Some individuals in the 4th and 5th postnatal weeks emitted with adultlike frequency precision of about +/-100 Hz. In the youngest bats, the 1st harmonic CF component (CF1) was up to 22 dB stronger than CF2. Adultlike relative levels of CF1 (-28 dB relative to CF2) were reached in the 5th postnatal week. In spontaneously emitted CF-FM calls, the duration of the CF2 component gradually increased with age from 5 ms to maximum values of 18 ms. Durations of the CF2 component in induced calls averaged 7 +/- 2.6 ms in the 1st postnatal week and 8.2 +/- 1.5 ms in the 5th postnatal week. There were no age-related changes in duration of the terminal FM sweep (3 +/- 0.4 ms) in both induced and spontaneous calls. The magnitude of the terminal FM sweep in spontaneous calls was not correlated with age (mean 13.5 +/- 2 kHz). Values for induced calls slightly increased with age from 11 +/- 2 to 13 +/- 2 kHz. The emission rate of induced CF-FM signals increased with age from values of 2.5 +/- 2 to 17 +/- 5 pulses/s. Values for spontaneously emitted calls were 4.4 +/- 3 and 9 +/- 4.5 pulses/s, respectively. Doppler-shift compensation, as tested in the pendulum task, emerged during the 4th postnatal week in young bats that were capable of very brief active flights, but before the time of active foraging outside the cave.

This study documents the postnatal growth, age estimation and development of the foraging behaviour of the fulvous fruit bat Rousettus leschenaulti under captive conditions. At birth, the young were naked and pink with closed eyes and folded pinnae. By day four of age, their eyes had opened and the pups began to move. The mean length of forearm in 5-day-old pups was 24.9 mm and body mass was 10.8 g, equivalent to 32.3% and 14.2% of the values from postpartum females. The length of forearm and body mass increased linearly until 45 and 50 days, respectively, and thereafter maintained an apparent stability. The epiphyseal gap of the fourth metacarpal-phalangeal joint increased until 15 days, then decreased linearly until 75 days and thereafter closed. Age was estimated quantitatively, based on linear changes observed in the length of the forearm and epiphyseal gap. Pups began to roost separately, but adjacent to their mothers when 30 days old and flew clumsily when they were about 40 days old. After attaining clumsy flight, the young bats made independent foraging attempts feebly by biting and licking small fruit pieces. Young bats were engaged in suckling as well as ingesting fruits when they were about 50 days old. Between 55 and 65 days, they flew well and fed on fruits. At the age of 75 days, the young bats were completely weaned and at two months, their foraging behaviour was similar to that of their mothers. There was no significant difference in the growth pattern of the young maintained in captivity compared with those under natural conditions.

Development projects in tropical forests can impact biodiversity. Assessment and monitoring programs based on the principles of adaptive management assist managers to identify and reduce such impacts. The small mammal community is one important component of a forest ecosystem that may be impacted by development projects. In 1996, a natural gas exploration project was initiated in a Peruvian rainforest. The Smithsonian Institution's Monitoring and Assessment of Biodiversity program cooperated with Shell Prospecting and Development Peru to establish an adaptive management program to protect the region's biodiversity. In this article, we discuss the role of assessing and monitoring small mammals in relation to the natural gas project. We outline the conceptual issues involved in establishing an assessment and monitoring program, including setting objectives, evaluating the results and making appropriate decisions. We also summarize the steps taken to implement the small mammal assessment, provide results from the assessment and discuss protocols to identify appropriate species for monitoring.

This note examines the effect of parasitism on host size, the preference of the parasite for a specific host body area, and the seasonal abundance for the 3 most abundant bat flies (i.e., Trichobius joblingi Wenzel, a parasite of the bat Carollia perspicillata [Linnaeus], and Aspidoptera falcata Wenzel and Megistopoda proxima [Séguy], parasites on Sturnira lilium [Geoffroy]). Trichobius joblingi and A. falcata are moderately dorsoventrally flattened and were collected on the wing membranes of their hosts, and M. proxima is moderately laterally compressed, has long, thin hind legs, and was collected in the body fur of the host. These 3 parasites also showed distinct seasonal patterns. There was a significant negative correlation between the simultaneous occurrence of A. falcata and M. proxima on the host. Parasitism by M. proxima was correlated with a significant weight loss in male S. lilium, which may reflect the large size, high activity, and constant feeding of this parasite, thereby causing a significant negative effect on the host. Sex ratios favoring male flies could be explained by the tendency of female flies to leave the host immediately before the bat leaves the shelter in search for food or immediately after bats are collected but could also be a consequence of higher mortality among females, especially gravid ones. Finally, collecting may have influenced the skewed sex ratio because male flies, being more active, were more evident to the collector.

Little red flying foxes (Pteropus scapulatus) are seasonal breeders: they mate in late spring/early summer, and young are born the following autumn. In captivity, males housed outdoors in a normal breeding colony in natural daylight showed a single cycle of testis growth and regression each year. During reproductive quiescence, testicular volume was approximately 2 cm3; recrudescence began soon after the winter solstice; testicular volume was maximum at approximately 6.5 cm3 at the spring equinox; and regression was complete by the end of summer. To test whether photoperiod entrains or synchronizes the cycle, groups of males were housed indoors, without females, at constant temperature, and artificial lighting was timed to either mimic naturally changing daylength or provide alternating 3-month periods of short (11 h light:13 h dark) or long (16 h light:8 h dark) days (two groups, three months out of phase with each other). During 18 months, the applied photoperiod protocol had no effect on the frequency of testicular cycles (which remained at one per year), the time course of recrudescence and regression (as described above for normal outdoor control males), or the completeness of growth and regression stages. These results suggest that male P. scapulatus are not reproductively photoresponsive.
