Abstract

Diatoms are among the dominant phytoplankters in the worl's ocean, and their external silica investments, resembling artificial photonics crystal, are expected to play an active role in light manipulation. Digital holography allowed studying the interaction with light of Coscinodiscus wailesii cell wall reconstructing the light confinement inside the cell cytoplasm, condition that is hardly accessible via standard microscopy. The full characterization of the propagated beam, in terms of quantitative phase and intensity, removed a long-standing ambiguity about the origin of the light. The data were discussed in the light of living cell behavior in response to their environment.

Abstract

Gastrointestinal emergencies (GE) are frequently encountered in emergency department (ED), and patients can present with wide-ranging symptoms. more than 3 million patients admitted to US hospitals each year for EGS diagnoses, more than the sum of all new cancer diagnoses. In addition to the complexity of the urgent surgical patient (often suffering from multiple co-morbidities), there is the unpredictability and the severity of the event. In the light of this, these patients need a rapid decision-making process that allows a correct diagnosis and an adequate and timely treatment. The primary endpoint of this Italian nationwide study is to analyze the clinicopathological findings, management strategies and short-term outcomes of gastrointestinal emergency procedures performed in patients over 18. Secondary endpoints will be to evaluate to analyze the prognostic role of existing risk-scores to define the most suitable scoring system for gastro-intestinal surgical emergency. The primary outcomes are 30-day overall postoperative morbidity and mortality rates. Secondary outcomes are 30-day postoperative morbidity and mortality rates, stratified for each procedure or cause of intervention, length of hospital stay, admission and length of stay in ICU, and place of discharge (home or rehabilitation or care facility). In conclusion, to improve the level of care that should be reserved for these patients, we aim to analyze the clinicopathological findings, management strategies and short-term outcomes of gastrointestinal emergency procedures performed in patients over 18, to analyze the prognostic role of existing risk-scores and to define new tools suitable for EGS. This process could ameliorate outcomes and avoid futile treatments. These results may potentially influence the survival of many high-risk EGS procedure.

Abstract

Clostridioides (previously Clostridium) difficile infection (CDI) is a common cause of antibiotic-associated diarrhea, whose symptoms range from mild diarrhea to life-threatening pseudomembranous colitis. CDI is characterized by significant recurrence rate following initial resolution and recurrent C. difficile infection (rCDI) represents an onerous burden for the healthcare systems. Conventional antibiotic-based approaches are generally used for the treatment of rCDI but the effective therapy remains elusive. Recently, the faecal microbiota transplantation (FMT) has emerged as an alternative therapeutic strategy against rCDI, with high treatment success rate. In 2018, the Italian National FMT Program was launched, with the aim to provide high quality standards in FMT application to adults with rCDI not responding to antibiotic therapy. Here, we sketch out the key characteristics and the progress of the Italian National FMT Program during the CO-VID-19 pandemic.

Abstract

Pro-nerve growth factor (proNGF) is the predominant form of NGF in the brain and its levels increase in neurodegenerative diseases. The balance between NGF receptors may explain the contradictory biological activities of proNGF. However, the specific role of the two main proNGF variants is mostly unexplored. proNGF-A is prevalently expressed in healthy brain, while proNGF-B content increases in the neuro-degenerating brain. Recently we have investigated in vitro the biological action of native mouse proNGF variants. To gain further insights into the specific functions of the two proNGFs, here we intranasally delivered mouse-derived proNGF-A and proNGF-B to the brain parenchyma of healthy and diabetic rats, the latter characterized by dysfunction in spatial learning and memory, in the septo-hippocampal circuitry and by relative increase in proNGF-B hippocampal levels. Exogenous proNGF-B induces depression of hippocampal DG-LTP and impairment of hippocampal neurogenesis in healthy animals, with concomitant decrease in basal forebrain cholinergic neurons and cholinergic fibers projecting to the hippocampus. proNGF-A, while ineffective in healthy animals, rescues the diabetes-induced impairment in DG-LTP and hippocampal neurogenesis, promoting the concomitant recovery of the basal forebrain cholinergic phenotype. Our experimental evidences suggest that the balance between different proNGFs may influence the development and progression of neurodegenerative diseases.

Abstract

AIM: Enhanced Recovery After Surgery (ERAS) guidelines represent one of the most important steps forward in colorectal surgery in the last ten years. Despite the well-known and demonstrated positive impact on the clinical outcomes that this pathway provides, a cultural revolution in patient management is needed. This is not easy to obtain, especially in small and peripheral centers. In Italy, the diffusion of minimally invasive surgery and "fast-track" perioperative management of the patient is rapidly spreading, even in the central and southern regions. However, in these regions, the percentage of laparoscopic colorectal procedures is dramatically less than in the north of Italy. In this context, the idea of a research group based in Rome focused on the development and spreading of ERAS protocols in the Lazio Region was developed.METHODS: A research group, based in Rome, was founded in December 2016 to evaluate the diffusion of the ERAS program over the main colorectal centers of the region. This "Lazio Network" began with a group of surgeons and anesthesiologists from 5 hospitals. After one and half years, the project now includes 17 hospitals in the region. A multicenter database was created, including consecutive patients who underwent laparoscopic colorectal resection following the ERAS program in the participating centers between January 2016 and December 2017.RESULTS: Data for more than 1200 patients were collected over the observed period. The rate of minimally invasive surgery was higher compared to the regional rate (90% vs. 30%), adherence to the ERAS pathway was around 60% of the items per patient. A clinical study will result from this database. Abstract

Vetiver is the only grass cultivated worldwide for the root essential oil, which is a mixture of sesquiterpene alcohols and hydrocarbons, used extensively in perfumery and cosmetics. Light and transmission electron microscopy demonstrated the presence of bacteria in the cortical parenchymatous essential oil-producing cells and in the lysigen lacunae in close association with the essential oil. This finding and the evidence that axenic Vetiver produces in vitro only trace amounts of oil with a strikingly different composition compared with the oils from in vivo Vetiver plants stimulated the hypothesis of an involvement of these bacteria in the oil metabolism. We used culture-based and culture-independent approaches to analyse the microbial community of the Vetiver root. Results demonstrate a broad phylogenetic spectrum of bacteria, including α-, β- and γ-Proteobacteria, high-G+C-content Gram-positive bacteria, and microbes belonging to the Fibrobacteres/Acidobacteria group. We isolated root-associated bacteria and showed that most of them are able to grow by using oil sesquiterpenes as a carbon source and to metabolize them releasing into the medium a large number of compounds typically found in commercial Vetiver oils. Several bacteria were also able to induce gene expression of a Vetiver sesquiterpene synthase. These results support the intriguing hypothesis that bacteria may have a role in essential oil biosynthesis opening the possibility to use them to manoeuvre the Vetiver oil molecular structure. © 2008 The Authors.

30. Combined effects of axial load and concrete strength variation on the seismic performance of existing RC buildings

It is well known that the axial load plays an important role in the evaluation of the structural capacity of RC columns. In existing buildings this problem can be even more significant than in new ones, since the material can easily present poor mechanical properties. The paper is aimed at the investigation of the role of the axial load variation on the seismic performance of RC columns of a case-study, i.e. a doubly symmetric 4-storey RC building. The effects of the axial load variation have been checked on the first storey columns, by comparing the seismic response, measured in terms of chord rotation and shear force, with the corresponding capacity. The sensitivity of the seismic performance to the axial load is evaluated with special attention on the type of analysis adopted to determine the seismic response and on considering a wide range of values for the concrete strength. The study points out a non-negligible effect of the axial load variation on the seismic response of the case-study building, especially when combined to concrete strength variability., Applied Mechanics

Abstract

International audience; Glial cells express acetylcholine receptors. In particular, rat Schwann cells express different muscarinic receptor subtypes, the most abundant of which is the M2 subtype. M2 receptor activation causes a reversible arrest of the cell cycle. This negative effect on Schwann cell proliferation suggests that these cells may possibly progress into a differentiating program. In this study we analyzed the in vitro modulation, by the M2 agonist arecaidine, of transcription factors and specific signaling pathways involved in Schwann cell differentiation. The arecaidine-induced M2 receptor activation significantly upregulates transcription factors involved in the promyelinating phase (e.g., Sox10 and Krox20) and downregulates proteins involved in the maintenance of the undifferentiated state (e.g., c-jun, Notch-1, and Jagged-1). Furthermore, arecaidine stimulation significantly increases the expression of myelin proteins, which is accompanied by evident changes in cell morphology, as indicated by electron microscopy analysis, and by substantial cellular re-distribution of actin and cell adhesion molecules. Moreover, ultrastructural and morphometric analyses on sciatic nerves of M2/M4 knockout mice show numerous degenerating axons and clear alterations in myelin organization compared with wild-type mice. Therefore, our data demonstrate that acetylcholine mediates axon-glia cross talk, favoring Schwann cell progression into a differentiated myelinating phenotype and contributing to compact myelin organization.

Abstract

Diatoms are among the dominant phytoplankters in the world's oceans, and their external silica investments, resembling artificial photonic crystals, are expected to play an active role in light manipulation. Digital holography allowed studying the interaction with light of Coscinodiscus wailesii cell wall reconstructing the light confinement inside the cell cytoplasm, condition that is hardly accessible via standard microscopy. The full characterization of the propagated beam, in terms of quantitative phase and intensity, removed a long-standing ambiguity about the origin of the light confinement. 36. Valutazione mediante approccio ecosistemico dei Pleuronectiformes bersaglio delle flotte da pesca artigianale nella Fascia Costiera fra Capo San Marco e Capo Granitola

Abstract

Obiettivo primario del progesto è stata la valutazione a fini gestionali dei Pleuronettiformi bersaglio della flottiglia artigianale che opera mediante tremagli e monofilo nell'ampio golfo tra Capo Granitola e Capo San Marco. Dall'aprile 2005 al marzo 2009 sono state rilevate le catture e lo sforzo di pesca della flottiglia artigianale di Marinella di Selinunte mediante interviste allo sbarco statisticamente programmate su base campionaria nello spazio e nel tempo. L'analisi della cattura del relativo sforzo e della cpue è stata effettuata a partire dal 2002 fino al 2008. La cpue relativa alla cattura totale oscilla fra il massimo del 2002 pari a 5,3 e il minimo del 2005 pari a 2,7. Nel 2008, ultimo anno di monitoraggio, la cpue è risultata 4,7. La cpue relativa alla cattura di Pegusa impar nel periodo 2002-2007 è risultata compresa fra 0,35 e 0,25 cioè praticamente costante. Nel 2008 la cpue e crollata a 0,15. La riproduzione avviene in aprile e maggio.La taglia alla prima maturità sessuale è risultata di circa 156 mm. I parametri di crescita L?, K e t0 rispettivamente sono: 255 mm, 0,27 anni-1 e -1,5 anni; l'esemplare più anziano aveva sei anni di età. La specie sembra ben sopportare l'attività di pesca anche se nell'ultimo anno di osservazione la cpue è drasticamente diminuita. Scophthalmus rhombus tra il 2002 e il 2004 è stato poco catturato e la cpue è risultata 0,01 nel 2002 e 0,03 nei due anni successivi. Nel 2005 la cpue è balzata a 0,20 per poi scendere a 0,16 ed a 0,14 nei due anni successivi. Infine nel 2008 è crollata a 0,07. Giugno e luglio sono i mesi in cui avviene la riproduzione. I parametri di crescita L?, K e t0 rispettivamente sono: 395 mm, 0,26 anni-1 e -1,3 anni; l'esemplare più anziano aveva sei anni di età. L'andamento fortemente oscillante della cpue sembra indicare che sia l'attività di pesca che le condizioni climatiche influenzano pesantemente la popolazione di Scophthalmus rhombus. Sembra utile ed opportuno proporre: un monitoraggio a lungo termine delle risorse e dell'attività di pesca; uno studio oceanografico mirato alla valutazione di eventuali cambiamenti climatici in relazione alla riproduzione ed alla mortalità naturale.
