Ultrasound‐assisted dehydration of fructose into 5‐hydroxymethylfurfural (5‐HMF) and subsequent oxidation to furandicarboxylic acid (2, 5‐FDCA) is studied in the current work with the main objective being to elucidate the effectiveness of ultrasound for intensified synthesis. The effect of reaction parameters like ultrasound power, duty cycle, reaction time, reaction temperature, solid to solvent ratio, and fructose concentration on the dehydration of fructose into 5‐HMF has been studied. Optimized conditions established were ultrasonic power of 140 W, duty cycle of 60%, reaction time of 60 min, temperature of 100°C, and fructose:dimethylsulfoxide (DMSO) ratio of 3:100 (g/mL), which resulted in the highest 5‐HMF yield of 96% and fructose conversion of 100%. The conventional method carried out at optimized conditions resulted in only 13.5% as 5‐HMF yield. The obtained 5‐HMF was further oxidized to FDCA using Pd/C as the catalyst, H2O/DMSO as solvent, and K2CO3 as a base also using ultrasonic irradiation at 140 W power and 22 kHz frequency in the presence and absence of O2 as oxidant. 100% conversion of 5‐ HMF was obtained in 30 min and 4 h using ultrasound in the presence of O2 and in absence of O2, respectively. 75% conversion of 5‐HMF was observed using the conventional method in 5 h in the presence of O2. Background: To investigate the preventive efficacy of methylsulfonylmethane (MSM) on alveolar bone destruction in rats with periodontitis. Methods: Twenty-four male Sprague-Dawley rats were randomly divided into three groups: control, experimental periodontitis (Ep), and Ep-MSM. Periodontitis was induced by placing 4.0 silk sutures in the subparamarginal position on the necks of the mandibular first molars and applying the suture for 5 weeks. The Ep-MSM group was given 500 mg/body weight/day MSM intraperitoneally for 35 days. At the end of the study, bilateral mandibular samples were taken. Periodontal bone loss was measured through histologic sections. Histomorphometric and immunohistochemical (receptor activator of nuclear factor kappa B ligand (RANKL), osteoprotegerin (OPG)) evaluations were performed on right mandibular tissue samples, and biochemical (interleukin (IL)-1 beta (β)/IL-10, malondialdehyde (MDA), glutathione (GSH), oxidative stress index (OSI)) evaluations were performed on left mandibular tissue samples. Results: No significant difference was found between the groups in IL-1β and IL-1β/IL-10 values ​​(p > 0.05). A significant decrease in IL-10 levels was observed in the Ep-MSM and Ep groups compared with the control group (p < 0.05). MDA levels significantly increased in the Ep and Ep-MSM groups compared with the control group, and GSH levels significantly decreased in the Ep group compared with the other groups (p < 0.05). OSI values ​​were significantly higher only in the Ep group (p < 0.05). RANKL levels showed a significant increase in the Ep group compared with the other groups. OPG levels were significantly increased only in the Ep-MSM group (p < 0.05). Dimethyl sulfoxide (DMSO) is an aprotic solvent widely used in ecotoxicological assays, suitable for solubilizing a wide range of polar and non-polar substances. The aim of this study was to analyze the species-specific biological response of the model organisms Daphnia magna and Ceriodaphnia dubia to 0.5% DMSO, under feeding conditions. Both species were exposed in culture medium, with or without 0.5% DMSO, to the unicellular algae (Raphidocelis subcapitata, Tetradesmus obliquus), either individually or in combination; the algae were pretreated or not pretreated with 0.5% DMSO. The exposure was carried out in acute (24 h and 48 h) and chronic (72 h and 168 h) ecotoxicity tests. The experimental results suggest that a lower DMSO concentration limit (<0.5%) would be appropriate for C. dubia due to its greater sensitivity to the solvent. Finally, considering the ingestion of DMSO-contaminated algae, it can be concluded that, under certain experimental conditions, the solvent ingested through the diet may significantly affect the mortality of both D. magna and C. dubia. Preservation and long-term storage of readily available cell-laden tissue-engineered products are major challenges in expanding their applications in healthcare. In recent years, there has been increasing interest in the development of off-the-shelf tissue-engineered products using the cryobioprinting approach. Here, bioinks are incorporated with cryoprotective agents (CPAs) to allow the fabrication of cryopreservable tissue constructs. Although this method has shown potential in the fabrication of cryopreservable tissue-engineered products, the impact of the CPAs on the viscoelastic behavior and printability of the bioinks at cryo conditions remains unexplored. In this study, we have evaluated the influence of CPAs such as glycerol and dimethyl sulfoxide (DMSO) on the rheological properties of pre-crosslinked alginate bioinks for cryoprinting applications. DMSO-incorporated bioinks showed a reduction in viscosity and yield stress, while the addition of glycerol improved both the properties due to interactions with the calcium chloride used for pre-crosslinking. Further, tube inversion and printability experiments were performed to identify suitable concentrations and cryobioprinting conditions for bioinks containing CPAs & pre-crosslinked with CaCl2. Finally, based on the printability analysis & cell recovery results, 10% glycerol was used for cryobioprinting and preservation of cell-laden constructs at −80 °C and the viability of cells within the printed structures were evaluated after recovery. Cell viability results indicate that the addition of 10% glycerol to the pre-crosslinked bioink significantly improved cell viability compared to bioinks without CPAs, confirming the suitability of the developed bioink combination to fabricate tissue constructs for on-demand applications. Background: Paraquat (PQ) is a widely used pesticide, can cause severe intoxication and respiratory failure. Myrtenol (Mrl), an essential oil derived in various plants, exhibits several biological properties, including anti-inflammatory and antioxidant activities. This study aims to investigate the protective potential of Mrl against oxidative stress and inflammation caused by PQ exposure. Methods: Twenty-five Wistar albino rats were divided into the following groups (n = 5 in each group): a control group (treated by dimethyl sulfoxide (DMSO)), a PQ group (exposed to 54 mg/m³ aerosol PQ), and two treatment groups that were exposed to PQ aerosol and administered oral Mrl at doses of 25 mg/kg/day and 50 mg/kg/day, respectively. The final group was exposed to PQ aerosol and treated with oral dexamethasone at a dose of 0.03 mg/kg/day. Various hematological, oxidative, inflammatory, and pathological indices were measured at the conclusion of the treatment period. Results: PQ decreases the levels or activities of superoxide dismutase (SOD), catalase (CAT), and Thiol, while increasing the levels or activities of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and malondialdehyde (MDA). Mrl restored activites of SOD, and CAT, as well as thiol levels to near-control values while reducing TNF-α, IL-6, and MDA levels. Pathological studies further confirmed the therapeutic effects of Mrl. Conclusion: The results of this study demonstrate the promising therapeutic effects of Mrl against inhaled PQ in rats. Microorganisms cause microbiologically influenced corrosion, for the prevention of which bactericide inhibitors are used. The aim of the work was to study in vitro the sensitivity of SRB Desulfovibrio oryzae NUChC SRB1 to different concentrations of dimethyl sulfoxide (DMSO), and evaluate the indicators of the microbial corrosion of steel induced by this bacterium in the presence of the pharmaceutical drugs DMSO and paracetamol. The sensitivity of SRB D. oryzae to 1–100% DMSO (v/v) was studied via the dilution method in Postgate's "C" liquid medium. The corrosion activity of D. oryzae against steel 3 was investigated under DMSO and paracetamol treatment at a final concentration of 45% (v/v) and 0.2% (w/v), respectively, according to the ability of bacteria to form a biofilm on the surface of the steel samples (via the crystal violet method) and the effect on the corrosion rate (via the gravimetric method). It was revealed that DMSO affected D. oryzae NUChC SRB1 and exhibited bactericidal properties (at a concentration range of 10–100%, v/v) and antibiofilm properties (at a concentration of 45%, v/v). Despite its antibiofilm properties confirmed by the reduction in bacterial biofilm mass, anticorrosion features were not observed in the model 35-day conditions of the microbial corrosion of steel in an anaerobic environment with bacterial sulfate reduction. Paracetamol (0.2%, w/v) did not affect biofilm formation by SRB under these conditions, and significantly contributed to an increase in the rate of the microbial corrosion of steel. The prospect of further research is to assess the effect of DMSO and paracetamol on the indicators of microbial corrosion induced by SRB under the influence of the concentrations of these compounds found in wastewater, to clarify the possible additional causes of damage to the equipment of treatment plants. Further research should also be directed at investigating the antimicrobial properties of complexes of compounds with DMSO, which should be considered as an ecological solution to the problem of microbiologically influenced corrosion prevention. Purpose: To investigate the role of dimethyl sulfoxide (DMSO) as a cryoprotectant and solvent in bovine embryogenesis, with a particular focus on its impact on gene expression during early and late developmental stages. Method: Bovine embryos were used to evaluate effects of DMSO on expression of genes critical to apoptosis and developmental processes. Gene expression analysis was performed to assess changes in both pro-apoptotic and anti-apoptotic markers, as well as genes essential for growth and survival. Results: Dimethyl sulfoxide influenced gene expression in a stage-specific manner. During early development, DMSO induced overexpression of the pro-apoptotic gene, BAX, and downregulated the anti-apoptotic gene, BCL2, indicating increased apoptotic activity. Furthermore, the expression of GDF9 and IGF1, which are crucial for growth and survival, was altered, suggesting interference with key developmental pathways. In contrast, late-stage embryos exhibited elevated levels of BCL2 and HSPB1, markers of anti-apoptotic activity, indicating a more complex regulatory role of DMSO at advanced stages of embryogenesis. Conclusion: While DMSO is effective as a cryoprotectant, its impact on gene expression raises concerns about potential developmental consequences. Background/Aim: Diabetic retinopathy (DR), a complication of diabetes, causes damage to retinal blood vessels and can lead to vision impairment. Persistent high blood glucose levels contribute to this damage, and despite ongoing research, effective treatment options for DR remain limited. Dimethyl sulfoxide (DMSO) has shown anti-inflammatory and antioxidant properties in both in vivo and in vitro studies; however, its potential as an anti-inflammatory agent in the context of DR has not yet been explored. This study aimed to assess the effects of subconjunctival injection of DMSO on the progression of DR. Materials and Methods: DR was induced in rats using intraperitoneal injections of streptozotocin (55 mg/kg), confirmed by measuring blood glucose levels and electroretinography (ERG). The rats were divided into five groups: a normal control group (CON), a DR control group receiving PBS injections (DMSO 0), and three DR groups receiving different concentrations of DMSO (98%, 50%, and 10%). Retinal function was evaluated using ERG at weeks 10 and 14, and histological analysis at week 16. Results: The DMSO 50 group had significantly higher B-wave amplitude in ERG compared to the DMSO 0 group (p<0.05). Flicker response amplitudes were also significantly greater in the DMSO 50 and DMSO 10 groups compared to DMSO 0 (p<0.05). Histological examination revealed thinner retinal layers in the DMSO 0 group compared to the CON group, while the DMSO-treated groups showed improved retinal thickness. Antioxidants are contained in fruits and vegetables and are commonly obtained through food. However, it is frequently necessary to supplement the diet with substances that are often poorly soluble in water and sensitive to light and oxygen. For this reason, in this work, luteolin (LUT) and naringenin (NAR), two compounds with antioxidant activity and potential health benefits, were precipitated through the supercritical antisolvent technique using polyvinylpyrrolidone and β-cyclodextrin as the carriers. The precipitation occurred from dimethylsulfoxide using supercritical carbon dioxide as the antisolvent. The influence of pressure (9–12 MPa), active substance/carrier concentration in the solution (20–200 mg/mL), and their ratio (1/1 and 1/2 mol/mol) on morphology, particle mean size, and distribution were investigated. Under the optimized operating conditions, spherical microparticles with a mean diameter equal to 2.7 ± 0.9 μm (for LUT) and 5.5 ± 1.9 μm (for NAR) were obtained. The active ingredients were protected from the external environment by the presence of the carrier, and the dissolution rate was notably increased by processing them with β-cyclodextrin. This study provides a comprehensive analysis of the interactions between dimethyl sulfoxide (DMSO) and two small peptides, diglycine and N-acetyl-glycine-methylamide (NAGMA), in aqueous solutions using FTIR spectroscopy and density functional theory (DFT) calculations. ATR-FTIR spectroscopy and DFT results revealed that DMSO does not form direct bonds with the peptides, suggesting that DMSO indirectly influences both peptides by modifying the surrounding water molecules. The analysis of HDO spectra allowed for the isolation of the contribution of water molecules that were simultaneously altered by the peptide and DMSO, and it also explained the changes in the hydration shells of the peptides in the presence of DMSO. In the DMSO–diglycine system, DMSO contributes to the additional strengthening of water hydrogen bonds in the reinforced hydration sphere of diglycine. In contrast, DMSO has a more moderate effect on the water molecules surrounding NAGMA due to the similarity of their hydration shells, leading to a slight weakening of the hydrogen bonds in the NAGMA hydration sphere. Benzene is a crucial industrial hydrocarbon, posing significant health risks due to its toxic metabolites like hydroquinone (HQ). This study investigates the role of clusterin (CLU) in benzene toxicity by analyzing its protein and mRNA levels, as well as the expression of Bcl-2 and Bax, to evaluate the feasibility of CLU as a biomarker for chronic benzene poisoning. HL-60 cells were induced to differentiate into neutrophil-like cells using 1% Dimethyl Sulfoxide (DMSO). Enzyme-linked immunosorbent assay (ELISA) and RT-PCR were used to analyze CLU protein and mRNA levels. ELISA was employed to detect sCLU protein content in cell culture supernatants, and western blot was used to assess Bcl-2 and Bax expression. The optimal time for 1% DMSO to induce HL-60 cells into neutrophil-like cells was 48 h. As HQ concentration increased, HL-60 cell viability decreased, CLU protein and sCLU protein levels in the supernatant decreased, CLU mRNA levels decreased, Bcl-2 protein expression decreased, and Bax expression increased. HQ exposure reduces CLU protein concentration and mRNA levels in neutrophil-like cells induced from HL-60 cells, indicating that CLU could be a potential biomarker for chronic benzene poisoning. Intratracheal instillation (ITI) is considered the most pragmatic approach for investigating the potential toxicities of various respiratory exposure materials. Various respiratory exposure materials, including nanomaterials, hazardous air pollutants, fine dust, and household biocides, have raised public health concerns because of limited toxicological information and increasing consumption. Hence, toxicity studies using ITI in laboratory animals are important to accurately assess the human risks associated with these respiratory-exposed materials. However, data to adequately support the study design of ITI toxicity studies, particularly those examining the effects of commonly used vehicles following repeated exposure are insufficient. Therefore, in this study, we examined the effects of 16 types of commonly used vehicles in toxicity studies following 14-day repeated ITI in mice. General health endpoints (mortality, clinical signs, and body weight) were monitored throughout the study period, and terminal endpoints (gross observation, lung weight, bronchoalveolar lavage fluid analysis, and lung histopathological examination) were assessed after terminal sacrifice. Saline and phosphate-buffered saline elicited the least response, whereas corn oil (50 µL) showed the most severe toxicity findings. In addition, several commonly used vehicles, including distilled water, sodium carboxymethyl cellulose, dimethyl sulfoxide, ethanol, Tween 20, and Tween 80, induced mild-to-severe toxicity in the respiratory system. Based on the results of this study, some commonly used vehicles in toxicity studies should be used with caution when the ITI exposure route is considered. These results provide important background information on the effects of vehicles in ITI toxicity studies along with valuable insights for designing toxicity studies using respiratory exposure materials. The aim of this study was to screen the phytochemicals and assess the antibacterial properties of crude extracts from Vernonia amygdalina leaves against specific pathogenic bacteria. Qualitative and qauntitave analysis was performed on both ethanol and ethyl acetate crude extracts using standardized techniques. The analysis revealed the presence of alkaloids, phenol, flavonoids, terpenoids, saponins, tannins, and cardiac glycosides in the ethanol extract, while saponins and glycosides were absent in the ethyl acetate extract. The quantities of phenols, flavonoids, and alkaloids were determined using Gallic acid, quercetin, and atropine assays, respectively. The ethanol extract contained 154.7 ± 3.6 (mg GAE/g) of total phenols, 33.3 ± 1.8 (mg QE/g) of flavonoids, and 2.4 ± 0.08 (mg AE) of alkaloids, whereas the ethyl acetate extract contained 93.7 ± 3.9 (mg GAE/g) of total phenols, 30.7 ± 8.3 (mg QE/g) of flavonoids, and 1.57 ± 0.3 (mg GAE/g) of alkaloids. This study identified 44 components in the ethanol crude extract and 10 components in the ethyl acetate crude extract. The presence of alkanes, alkenes, amines, carboxylic acids, and alcohols was confirmed by FT-IR analysis. Each crude extract was tested at concentrations of 500 mg/mL, 250 mg/mL, 125 mg/mL, and 62.5 mg/mL using the same method. A concentration of 0.001 mg/mL of Penicillin capsule standard drugs and dimethyl sulfoxide were used as positive and negative controls, respectively. Background: Ketamine HCl, an FDA-approved therapeutic, is administered through various routes, including intranasal delivery. Administering an adequate therapeutic dose of intranasal ketamine HCl is challenging due to the limited volume that can be delivered intranasally given the current commercially available concentrations. Objectives: This study investigates solubilizing strategies to enhance the aqueous solubility of ketamine HCl for intranasal administration. Methods: We assessed the solubility profile of ketamine HCl by evaluating factors such as pH, co-solvents, and surfactants. Additionally, we developed and validated a UV-Vis spectroscopy method for ketamine HCl analysis. Results: Our solubility screening in various organic co-solvents revealed the following order of effectiveness in enhancing solubility: methanol > water > propylene glycol > ethanol > dimethyl sulfoxide (DMSO) > N-methyl-2-pyrrolidone (NMP). Despite methanol's superior solubility, its potential toxicity, coupled with the relatively lower effectiveness of other solvents compared to water, suggests that a co-solvency approach is not advantageous for ketamine HCl. We found that ketamine HCl solubility increased with medium acidity, with pH 3.5 being the optimal for further formulation studies. The impact of pharmaceutical surfactants on ketamine HCl solubility at an acidic pH was also evaluated. Surfactants tested included SDS, PEG 400, PVP, Tween 20, poloxamer 188, and lecithin. Notably, PEG 400 and PVP reduced solubility due to a salting-out effect, whereas Tween 80, lecithin, and poloxamer 188 slightly improved solubility through micelle formation. Among the surfactants tested, 1% SDS emerged as the most effective in enhancing ketamine HCl solubility. Conclusions: These outcomes highlight the potential of these solubilization strategies to address the solubility limitations of ketamine HCl, enabling the preparation of highly concentrated ketamine HCl formulations for intranasal delivery. Several molecules and extracts are known to have bone-specific effects. For example, the long-term use of glucocorticoids like prednisolone causes several negative effects including a loss of bone mass. Molecules like prednisolone are usually dissolved in organic solvent which are known to be toxic for zebrafish embryo in certain concentrations. Nevertheless, solvents like dimethyl sulfoxide (DMSO), ethanol and methanol have never been tested for specific skeletal effects during development in dose-dependency. Vitality assay, live fluorescence and bone-specific staining were used to evaluate solvents effects compared to prednisolone. DMSO, ethanol and methanol perturb osteogenesis starting from 1%, 1.5% and 3% respectively, concentrations in which vasculature, length and survival rate appear unaffected. This effect may be due to high sensitivity of the osteogenesis process to external chemical stimuli, especially in the trunk. On the contrary, the negative effect of prednisolone on skeletal development appears more specific since it is found at very low concentrations, far from any other developmental defects. The recommended solvent concentration to be used in zebrafish embryos osteogenesis assay was established in 0.5% for DMSO, 2% for methanol and 0.5% for ethanol. When developing immunochemical test systems, it is necessary to obtain specific antibodies. Their quality depends, among other things, on the immunogen used. When preparing hapten–protein conjugates to obtain antibodies for low-molecular-weight compounds, the key factors are the structure of the hapten itself, the presence of a spacer, the size of the carrier protein and the degree of its modification by hapten molecules. This work shows that one additional factor—the conditions for obtaining the hapten–protein conjugate—is overlooked. In this work, we have synthesized conjugates of bisphenol A derivative 4,4-bis(hydroxyphenyl)valeric acid (BVA), the protein carrier soybean trypsin inhibitor (STI), and bovine serum albumin (BSA) in reaction media combining water with two organic solvents: dimethylformamide (DMF) or dimethyl sulfoxide (DMSO). Namely, BSADMF–BVA, STIDMF–BVA, BSADMSO–BVA and STIDMSO–BVA conjugates were obtained. Rabbit polyclonal antibodies against the BSADMF–BVA conjugate demonstrated basically different interactions in the developed ELISA systems using either STIDMF–BVA or STIDMSO–BVA conjugates. The use of the STIDMF–BVA conjugate demonstrated the absence of competition in combination with antisera obtained from BSADMF–BVA in an ELISA. A competitive interaction was observed only with the use of the STIDMSO–BVA conjugate. Under the selected conditions, the detection limit of bisphenol A was 8.3 ng/mL, and the working range of determined concentrations was 18.5–290.3 ng/mL. The obtained data demonstrate the possibility of achieving sensitive immunoassays by simply varying the reaction media for the hapten–protein conjugation, which could provide an additional tool in the development of immunoassays for other low-molecular-weight compounds. Verteporfin (VP) has been used for photodynamic therapy (PDT) for over 20 years, and new applications have brought it back into the spotlight. VP is hydrophobic and requires lipid carriers for clinical delivery as Visudyne. A nanosuspension of VP, termed NanoVP, that requires no carriers is developed, permitting delivery of VP alone in an aqueous solution. NanoVP is produced by solvent–antisolvent precipitation, with dimethyl sulfoxide as the preferable solvent of several screened. The initial formulation has a hydrodynamic diameter of 104 ± 6.0 nm, concentration of 133 ± 10 μm, polydispersity index (Pdi) of 0.12 ± 0.01, and zeta potential of −22.0 ± 0.93 mV. Seeking a concentration >500 μm, a zeta potential <−10 mV, a diameter <64 nm, and a Pdi < 0.2, eight synthesis parameters are probed, identifying three that modified nanoparticle diameter and three that modified nanoparticle dispersity. The diameter is tuned fourfold from 49.0 ± 4.4 to 195 ± 7.1 nm, and the solution concentration is increased by 6.3‐fold to 838 ± 45.0 μm. Finally, the bioavailability and anticancer capacity of NanoVP in glioblastoma are evaluated. Phylanthus species, including P. niruri and P. urinaria have pharmacological potential due to their rich phytochemical composition. People usually used this plant for medicinal treatments. This study aimed to compare the phytochemical and antioxidant and antibacterial activities of two Phyllanthus species. The whole of plants was extracted using maceration method with ethanol as solvent. Phytochemical content analyzed using spectrophotometer. The reagent used for each compounds that were Folin-ciocalteu for phenolics, AlCl3 for flavonoid, dimethyl sulfoxide for chlorophyll and carotenoid. Bioactivity analysis using 1,1-diphenyl-2-picrylhydrazyl method for antioxidant activity and Kirby-Bauer method for antibacterial activity. The highest flavonoid content (12.22 mg QE/gram extract) and total chlorophyll (43.2 µg/ml extract) in P. niruri while phenolic content (80.8 mg GAE/gram extract) in P. urinaria. The carotene of both Phyllanthus were similar (11.9 µg/ml extract. The IC50 values of P. urinaria (6.16 ± 0.42 µg/ml) and P. niruri (17.72 ± 0.80 µg/ml), which indicated very strong antioxidant activity. P. urinaria leaf extract had stronger inhibition against Escherichia coli than Staphylococcus aureus (>20 mm) and P. niruri leaf extract could inhibit E. coli and S. aureus bacteria at all concentrations (11-20 mm). This study found that phenolic compounds strongly influenced the antioxidant and antibacterial abilities of Phyllanthus, while chlorophyll and carotenoids had only a slight influence. The density and viscosity of the pseudo-binary mixtures of eutectic solvent (ES) composed of choline chloride and ethylene glycol ([ChCl/EG]) with dimethylsulfoxide (DMSO) were measured. In order to understand the effect of the mole ratio of ChCl:EG, two ChCl/EG ESs with the mole ratio of 1:3 and 1:4 (abbreviated as [ChCl/EG](1:3) and [ChCl/EG](1:4) in this work) were prepared. The measurements were carried out by digital vibrating U-tube density meter and Ubbelohde capillary viscometer from 303.15 to 323.15 K at atmospheric pressure (98.5 kPa). The Jouyban–Acree model was applied to correlate the experimental density and viscosity data of DMSO/[ChCl/EG](1:3) and DMSO/[ChCl/EG](1:4) mixtures. In addition, based on the experimental data, the derived properties of the mixtures, such as excess molar volume and viscosity deviation, were calculated. In this work, we describe the synthesis, crystal structures and magnetic properties of four air-stable mononuclear lanthanide(III) complexes with the N-(2,4,6-trimethylphenyl)oxamate (Htmpa) of formula: n-Bu4N[Nd(Htmpa)4(H2O)]·4H2O (1), n-Bu4N[Gd(Htmpa)4(H2O)]·3DMSO·2H2O (2), n-Bu4N[Tb(Htmpa)4(H2O)]·3DMSO·1H2O (3) and n-Bu4N[Dy(Htmpa)4(H2O)]·3DMSO·2H2O (4) (n-Bu4N+ = n-tetrabutylammonium; DMSO = dimethylsulfoxide). Their crystal structures reveal the occurrence of calixarene-type monoanionic species containing all-cis-disposed Htmpa ligands and one water molecule coordinated with the respective LnIII ion (Ln = Nd, Gd, Tb and Dy), featuring a nine-coordinated environment with muffin (MFF-9) (1) or spherical-capped square antiprism (CSAPR-9) (2–4) geometry. The major difference between their crystal structures is related to the nature of crystallization solvent molecules, either water (1) or both DMSO and water (2–4). The intermolecular hydrogen bonds among the self-complementary Htmpa ligands in all four compounds mediated a 2 D supramolecular network in the solid state. Direct-current (dc) magnetic properties for 1–4 show typical behavior for the ground state terms of the LnIII ions [4I9/2 (Nd); 8S7/2(Gd), 7F6 (Tb), 6H15/2 (Dy)]. Alternating-current (ac) magnetic measurements reveal the presence of slow magnetic relaxation without the presence of a dc field only for 4. This study explores the innovative potential of native lignin as a sustainable biopolyol for synthesizing polyurethane aerogels with variable microstructures, significant specific surface areas, and high mechanical stability. Three types of lignin—Organosolv, Aquasolv, and Soda lignin—were evaluated based on structural characteristics, Klason lignin content, and particle size, with Organosolv lignin being identified as the optimal candidate. The microstructure of lignin polyurethane samples was adjustable by solvent choice: Gelation in DMSO and pyridine, with high affinity to lignin, resulted in dense materials with low specific surface areas, while the use of the low-affinity solvent e.g acetone led to aggregated, macroporous materials due to microphase separation. Microstructural control was achieved by use of DMSO/acetone and pyridine/acetone solvent mixtures, which balanced gelation and phase separation to produce fine, homogeneous, mesoporous materials. Specifically, a 75% DMSO/acetone mixture yielded mechanically stable lignin polyurethane aerogels with a low envelope density of 0.49 g cm−3 and a specific surface area of ~300 m2 g−1. This study demonstrates a versatile approach to tailoring lignin polyurethane aerogels with adjustable textural and mechanical properties by simple adjustment of the solvent composition, highlighting the critical role of solvent–lignin interactions during gelation and offering a pathway to sustainable, high-performance materials. Electrospinning has emerged as a versatile technique for producing nanofibers for diverse applications. The fibers produced are highly regarded for their biocompatibility, exceptional surface-to-volume ratios, porosity, and adjustable composition properties, making them promising scaffolds for tissue engineering. In the literature, annatto-loaded cellulose acetate has already been successfully used for electrospinning. However, N,N-dimethylformamide (DMF) was utilized as a solvent in the experiments, which is considered a carcinogenic and mutagenic substance. The aim of this study is to analyze whether comparable results can be achieved with a low toxicity solvent consisting of acetone and dimethyl sulfoxide (DMSO). In this study, we investigate the electrospinning process of cellulose acetate (CA) and cellulose acetate combined with annatto extract (CA/A). Initially, various polymer concentrations ranging from 12% to 16% (w/v) were characterized by rheological measurements to determine their suitability for electrospinning. The morphology and diameter distribution of the electrospun fibers were analyzed using scanning electron microscopy (SEM). Contact angle measurements were carried out to determine the wettability of the electrospun meshes. The rheological investigations conducted revealed that increasing the polymer content of CA raises viscosity, while the incorporation of annatto decreases it. Uniform fibers are achieved at polymer concentrations of 14% and above. The electrospun nanofibers exhibit uniform morphology and diameters in the nanometer range, indicating the successful fabrication of annatto-loaded CA nanofibers. The contact angle measurements showed hydrophilic behavior on all investigated meshes. The incorporation of annatto extract in the electrospun fibers holds promise for various applications, including biomedical scaffolds, scaffolds for cultured meat, filtration membranes and food packaging materials. Abstract Dimethyl sulfoxide (DMSO) is a widely used solvent in drug research. However, recent studies indicate that even at low concentration DMSO might cause structural changes of proteins and RNA. The pyrazolopyrimidine antiviral OBR-5-340 dissolved in DMSO inhibits rhinovirus-B5 infection yet is inactive against RV-A89. This is consistent with our structural observation that OBR-5-340 is only visible at the pocket factor site in rhinovirus-B5 and not in RV-A89, where the hydrophobic pocket is collapsed. Here, we analyze the impact of DMSO in RV-A89 by high-resolution cryo-electron microscopy. Our 1.76 Å cryo-EM reconstruction of RV-A89 in plain buffer, without DMSO, reveals that the pocket-factor binding site is occupied by myristate and that the previously observed local heterogeneity at protein–RNA interfaces is absent. These findings suggest that DMSO elutes the pocket factor, leading to a collapse of the hydrophobic pocket of RV-A89. Consequently, the conformational heterogeneity observed at the RNA-protein interface in the presence of DMSO likely results from increased capsid flexibility due to the absence of the pocket factor and DMSO-induced affinity modifications. This local asymmetry may promote a directional release of the RNA genome during infection.

Background and Objectives: Dimethylsulfoxide(DMSO) is gold standard for cryopreservation of hematopoietic progenitor cells (HPC) to reconstitute hematopoiesis in autologous stem cell transplants (auto-HSCT). Higher DMSO-related adverse effects (AEs) are reported in pediatric patients due to lower body weight with the recommended maximal dose of 1g/kg bodyweight. This study compares adding Hhydroxyethyl starch Vs Pentastarch to DMSO in order to reduce the DMSO related adverse effects. Adding a non-permeating cryoprotectant like Hydroxyethyl starch (HES) lowers DMSO concentration. HES is known to cause pruritus and nephrotoxicity. Pentastarch has lower molecular weight hence faster renal elimination with fewer reported AEs. Adding a non-permeating cryoprotectant like Hydroxyethyl starch (HES) lowers DMSO concentration. Methods: This study is a single-centre retrospective-comparative analysis from October 2022 to February 2024, comparing cryopreservation outcomes using standard cryoprotectant-DMSO+ HES+albumin solution (CPS-1) with pentastarch+albumin+DMSO solution (CPS-2). During the study period, 20 pediatric patients who underwent auto-HSCT requiring cryopreserved HPC-A were included. Results: Average CD34+ recovery with CPS-1 and CPS-2 were 86.58±13.42 and 87.5±13.2% (P-value=0.879). Median time to neutrophil engraftment was comparable (10 days) and no significant difference in platelet engraftment was observed, median 13.5 and 15 days with CPS-1 and CPS-2. Post-cryopreservation product volume was lesser with CPS-2 compared to CPS-1 (271±34.77ml and 78±24.7; P< 0.0001). Perovskite solar cells (PSCs) offer an efficient, inexpensive alternative to current photovoltaic technologies, with the potential for manufacture via high-throughput coating methods. However, challenges for commercial-scale solution-processing of metal-halide perovskites include the use of harmful solvents, the expense of maintaining controlled atmospheric conditions, and the inherent instabilities of PSCs under operation. Here, we address these challenges by introducing a high volatility, low toxicity, biorenewable solvent system to fabricate a range of 2D perovskites, which we use as highly effective precursor phases for subsequent transformation to α-formamidinium lead triiodide (α-FAPbI3), fully processed under ambient conditions. PSCs utilising our α-FAPbI3 reproducibly show remarkable stability under illumination and elevated temperature (ISOS-L-2) and "damp heat" (ISOS-D-3) stressing, surpassing other state-of-the-art perovskite compositions. We determine that this enhancement is a consequence of the 2D precursor phase crystallisation route, which simultaneously avoids retention of residual low-volatility solvents (such as DMF and DMSO) and reduces the rate of degradation of FA+ in the material. Our findings highlight both the critical role of the initial crystallisation process in determining the operational stability of perovskite materials, and that neat FA+-based perovskites can be competitively stable despite the inherent metastability of the α-phase. The use of harmful solvents to fabricate stable devices hampers the commercialization of perovskite solar cells. In the current work, electrostatic interactions were used to immobilize the horseradish peroxidase (HRP) onto five types of ceramic materials (C) with different concentrations of oxidized metals (C1–C5). The highest immobilization efficiency (70 and 77%) was detected at 6 mg C3 and 18 enzyme units. Scanning Electron Microscope (SEM), Energy Dispersive X-ray (EDX) and Fourier Transform Infrared (FTIR) analysis of C3-HRP confirmed the immobilization of the enzyme. After ten reuses, the reusability analysis showed that (66%) of the C3-HRP enzyme activity was retained. For C3-HRP, the optimum pH and temperature of the soluble enzyme were shifted from 7.0 and 30 °C to 6.0 and 50 °C. Up to 40 °C and 50 °C, respectively, the soluble HRP and C3-HRP remained steady. The kinetic analysis revealed that the Km and Vmax of soluble HRP and C3-HRP were, respectively, 5.5 mM, 0.66 units, and 8 mM, 0.52 units for hydrogen peroxide (H2O2) and 35.5 mM, 3.4 units and 40 mM, 1.1 units for guaiacol. Compared to soluble-HRP, the C3-HRP exhibited a greater oxidizing affinity toward several phenolic compounds (Guaiacol, o-dianisidine, o–phenylenediamine, pyrogallol, p-aminoantipyrine). In comparison with soluble-HRP, the C3-HRP showed increased stress tolerance with Triton X-100, urea, metals, isopropanol, and dimethyl sulfoxide. Background: The emergence of multidrug resistant Candida species to available drugs has led to renewed interest in the use of herbal medicines globally. This study scientifically verified antifungal effectiveness of five commonly used plant species in Pader district, against selected pathogenic candida strains. Methods: Powdered roots of Momordica foetida, Sansevieria dawei and Distimake dissectus; and stem barks of Khaya anthotheca and Mitragyna rubrostipulata were extracted sequentially using petroleum ether and methanol, respectively; and total water extraction at 24.4 °C (maceration), 60 °C (decoction) and boiling water at 87 °C (hot water infusion). Extracts and their combinations, positive controls (amphotericin B, and fluconazole) and negative control (80% dimethyl sulfoxide, verified to be tolerable concentration to the tested Candida species) were screened and verified for their antifungal activity against Candida albicans (ATCC: American Type Culture Collection reference strain 10231, ATCC 90028, 0770a and 0796), C. glabrata (VVc 004, ATCC 2950) and C. tropicalis (ATCC 750 and 0210) using agar well diffusion and broth micro-dilution, respectively. Results: Aqueous extract (24.4 °C) of M. rubrostipulata (ZOI: 18.00 ± 1.00 to 38.33 ± 0.17; MIC: 3.13 ± 0.00 to 20.83 ± 4.17; MFC: 12.50 ± 0.00 to 200.00 ± 0.00), methanol extract of K. anthotheca (10.11 ± 0.31 to 15.11 ± 0.65; 1.04 ± 0.26 to 12.50 ± 0.00; 12.50 ± 0.00 to 100.00 ± 0.00), and combination of aqueous extract (60 °C) of D. dissectus + methanol extract of K. anthotheca (7.89 ± 0.26 to 19.67 ± 0.37; 0.78 ± 0.00 to 50.00 ± 0.00; 12.50 ± 0.00 to 200.00 ± 0.00) exhibited broad spectrum antifungal activities and were fungistatic against all tested Candida species, which comprised 8 clinical/control and susceptible/resistant strains. None of the conventional drugs used demonstrated broad spectrum antifungal activity across all tested Candida species/strains. Conclusion: Methanol extract of K. anthotheca, aqueous extract (24.4 °C) of M. rubrostipulata, and combination of aqueous extract (60 °C) of D. dissectus + methanol extract of K. anthotheca could be effective in the treatment of candidiasis. They demonstrated potential broad spectrum antifungal activity against different species and strains of tested Candida than the fluconazole and amphotericin B drugs. Their fungistatic nature showed their ability to inhibit fungal growth. Hence, these extracts/extract combination can offer better treatment option for candidiasis if they are standardized and also their active curative compounds isolated and made into antifungal drugs. Primary hepatocytes are widely utilized for investigating drug efficacy and toxicity, yet variations between batches and limited proliferation capacity present significant challenges. HepaRG cells are versatile cells, capable of maintaining an undifferentiated state and differentiating through dimethyl sulfoxide treatment, allowing for molecular analysis of hepatocyte plasticity. To elucidate the underlying molecular mechanisms of HepaRG cell plasticity, we used CYP3A4G/7R HepaRG cells engineered to express DsRed under the control of the fetus-specific CYP3A7 gene and EGFP under the adult-specific CYP3A4 gene promoter. In time-lapse imaging of CYP3A4G/7R HepaRG cells, we observed CYP3A7-DsRed expression transitioning from negative to positive during proliferation period and CYP3A4-GFP expression activating during differentiation. The de-differentiation potency of differentiated CYP3A4G/7R HepaRG cells was assessed using inhibitors and cytokines. It was found that Y-27632 (Y), A-83-01 (A), and CHIR99021 (C) (collectively referred to as YAC), which are known to promote liver regeneration in mice, did not induce CYP3A7-DsRed expression. Instead, these inhibitors increased CYP3A4-GFP expressing population. Furthermore, CHIR99021 alone increased CYP3A4-GFP-positive cells, while Wnt3a treatment increased CYP3A7-DsRed-positive cells, suggesting that Wnt signaling plays distinct roles in HepaRG cells. It was apparent that de-differentiated cells had increased CYP3A4 activity after a second round of differentiation, compared to differentiated cells after the first round. Transcriptomic analysis of HepaRG cells revealed distinct profiles between proliferative, differentiated, and de-differentiated states, highlighting their robust plasticity. Notably, hepatoblastic cells de-differentiated by YAC or C displayed transcriptome patterns similar to undifferentiated cells, whereas CYP3A7-DsRed and CYP3A4-GFP exhibited expression patterns different from those of undifferentiated cells. These findings underscore the dynamic nature of HepaRG cells while cautioning against solely relying on CYP3 family gene expression as a marker of differentiation. [ABSTRACT FROM AUTHOR]
