RNA pseudoknots play a crucial role in various cellular functions. Established pseudoknots show significant variation in both size and structural complexity. Specifically, three-stemmed pseudoknots are characterized by an additional stem-loop embedded in their structure. Recent findings highlight these pseudoknots as bacterial riboswitches and potent stimulators for programmed ribosomal frameshifting in RNA viruses like SARS-CoV2. To investigate the possible presence of functional three-stemmed pseudoknots in human mRNAs, we employed in-house developed computational methods to detect such structures within a dataset comprising 21,780 full-length human mRNA sequences. Numerous three-stemmed pseudoknots were identified. A selected set of 14 potential instances are presented, in which the start codon of the mRNA is found in close proximity either upstream, downstream, or within the identified three-stemmed pseudoknot. These pseudoknots likely play a role in translational initiation regulation. The probability of their existence gains support from their ranking as the most stable pseudoknot identified in the entire mRNA sequence, structural conservation across homologous mRNAs, stereochemical feasibility as demonstrated by structural modeling, and classification as members of the CPK-1 pseudoknot family, which includes many well-established pseudoknots. Furthermore, in four of the mRNAs, two or three closely spaced or tandem three-stemmed pseudoknots were identified. These findings suggest the frequent occurrence of three-stemmed pseudoknots in human mRNAs. A stepwise co-transcriptional folding mechanism is proposed for the formation of a three-stemmed pseudoknot structure. KH (hnRNP K homology) domains, consisting of approximately 70 amino acid residues, are present in a variety of nucleic-acid-binding proteins. Among these are poly(C)-binding proteins (PCBPs), which are important regulators of mRNA stability and posttranscriptional regulation in general. All PCBPs contain three different KH domains and recognize poly(C)-sequences with high affinity and specificity. To reveal the molecular basis of poly(C)-sequence recognition, we have determined the crystal structure, at 1.6 A resolution, of PCBP2 KH3 domain in complex with a 7-nt DNA sequence (5'-AACCCTA-3') corresponding to one repeat of the C-rich strand of human telomeric DNA. The domain assumes a type-I KH fold in a betaalphaalphabetabetaalpha configuration. The protein-DNA interface could be studied in unprecedented detail and is made up of a series of direct and water-mediated hydrogen bonds between the protein and the DNA, revealing an especially dense network involving several structural water molecules for the last 2 nt in the core recognition sequence. Unlike published KH domain structures, the protein crystallizes without protein-protein contacts, yielding new insights into the dimerization properties of different KH domains. A nucleotide platform, an interesting feature found in some RNA molecules, was identified, evidently for the first time in DNA.

Three-dimensional LiDAR systems that capture point cloud data enable the simultaneous acquisition of spatial geometry and multi-wavelength intensity information, thereby paving the way for three-dimensional point cloud recognition and processing. However, due to the irregular distribution, low resolution of point clouds, and limited spatial recognition accuracy in complex environments, inherent errors occur in classifying and segmenting the acquired target information. Conversely, two-dimensional visible light images provide real-color information, enabling the distinction of object contours and fine details, thus yielding clear, high-resolution images when desired. The integration of two-dimensional information with point clouds offers complementary advantages. In this paper, we present the incorporation of two-dimensional information to form a multi-modal representation. From this, we extract local features to establish three-dimensional geometric relationships and two-dimensional color relationships. We introduce a novel network model, termed MInet (Multi-Information net), which effectively captures features relating to both two-dimensional color and three-dimensional pose information. This enhanced network model improves feature saliency, thereby facilitating superior segmentation and recognition tasks. We evaluate our MInet architecture using the ShapeNet and ThreeDMatch datasets for point cloud segmentation, and the Stanford dataset for object recognition. The robust results, coupled with quantitative and qualitative experiments, demonstrate the superior performance of our proposed method in point cloud segmentation and object recognition tasks. To overcome the inherent bandwidth limitation of non-line-of-sight (NLOS) ultraviolet communication (UVC) channel, a novel space division multiplexing (SDM) framework based on non-coplanar rotation angle is established. The influence of SDM structure parameters on inter-channel interference (ICI) is studied to obtain the optimal antenna configuration. And a Gaussian approximate distribution (GAD) signal detection model is presented, which simplifies the calculation of the Bit error rate (BER) of the Photon multiplier tube (PMT) reception and is suitable for complex ICI analysis. Furthermore, a low ICI scheme based on the relationship between BER and the interference factor is proposed to increase the data transmission rate or reduce the alignment configuration. As a result, the maximum data rate increases to 34 Mbps, or the alignment demand decreases by 33.33% at most. Moreover, these analysis results and techniques can be applied to other NLOS scattering optical wireless communication systems. [ABSTRACT FROM AUTHOR]
