Bacterial microcompartments enclose a biochemical pathway and reactive intermediate within a protein envelope formed by the shell proteins. Herein, the orientation of the propanediol?utilization (Pdu) microcompartment shell protein PduA in bacterial microcompartments and in synthetic nanotubes, and the orientation of PduB in synthetic nanotubes are revealed. When produced individually, PduA hexamers and PduB trimers, tessellate to form flat sheets in the crystal, or they can self?assemble to form synthetic protein nanotubes in solution. Modelling the orientation of PduA in the 20 nm nanotube so as to preserve the shape complementarity and key interactions seen in the crystal structure suggests that the concave surface of the PduA hexamer faces out. This orientation is confirmed experimentally in synthetic nanotubes and in the bacterial microcompartment produced in vivo. The PduB nanotubes described here have a larger diameter, 63 nm, with the concave surface of the trimer again facing out. The conserved concave surface out characteristic of these nano?structures reveals a generic assembly process that causes the interface between adjacent subunits to bend in a common direction that optimizes shape complementarity and minimizes steric clashes. This understanding underpins engineering strategies for the biotechnological application of protein nanotubes.

Summary Gammaproteobacteria are important gut microbes but only persist at low levels in the healthy gut. The ecology of G ammaproteobacteria in the gut environment is poorly understood. Here, we demonstrate that choline is an important growth substrate for representatives of G ammaproteobacteria. Using P roteus mirabilis as a model, we investigate the role of choline metabolism and demonstrate that the cut C gene, encoding a choline‐trimethylamine lyase, is essential for choline degradation to trimethylamine by targeted mutagenesis of cut C and subsequent complementation experiments. P roteus mirabilis can rapidly utilize choline to enhance growth rate and cell yield in broth culture. Importantly, choline also enhances swarming‐associated colony expansion of P . mirabilis under anaerobic conditions on a solid surface. Comparative transcriptomics demonstrated that choline not only induces choline‐trimethylamine lyase but also genes encoding shell proteins for the formation of bacterial microcompartments. Subsequent analyses by transmission electron microscopy confirmed the presence of such novel microcompartments in cells cultivated in liquid broth and hyper‐flagellated swarmer cells from solid medium. Together, our study reveals choline metabolism as an adaptation strategy for P . mirabilis and contributes to better understand the ecology of this bacterium in health and disease.

Amino terminal acetylation is a ubiquitous protein modification affecting the majority of eukaryote proteins to regulate stability and function. We describe an optimised recombinant expression system for rapid production of aminoterminal-acetylated proteins within bacteria. We go on to describe the\ud system’s use in a fluorescence based in vivo assay for use in the highthroughput screen to identify drugs that impact amino-terminal acetylation\ud dependent oligomerisation. These new tools and protocols will allow researchers to enhance routine recombinant protein production and identify\ud new molecules for use in research and clinical applications.

B12 is unique among the vitamins as it is biosynthesized only by certain prokaryotes. The complexity of its synthesis relates to its distinctive cobalt corrin structure, which is essential for B12 biochemistry and renders coenzyme B12 (AdoCbl) so intriguingly suitable for enzymatic radical reactions. However, why is cobalt so fit for its role in B12‐dependent enzymes? To address this question, we considered the substitution of cobalt in AdoCbl with rhodium to generate the rhodium analogue 5′‐deoxy‐5′‐adenosylrhodibalamin (AdoRbl). AdoRbl was prepared by de novo total synthesis involving both biological and chemical steps. AdoRbl was found to be inactive in vivo in microbial bioassays for methionine synthase and acted as an in vitro inhibitor of an AdoCbl‐dependent diol dehydratase. Solution NMR studies of AdoRbl revealed a structure similar to that of AdoCbl. However, the crystal structure of AdoRbl revealed a conspicuously better fit of the corrin ligand for RhIII than for CoIII, challenging the current views concerning the evolution of corrins.

A partir de uma revisão sistemática para identificar aspectos metodológicos, características bibliométricas e as abordagens da violência contra a mulher em artigos sobre o tema, publicados entre 2003 e 2007 e disponíveis nas bases de dados PsycInfo, Pubmed, Scielo e Lilacs, foram selecionados 171 artigos que abordaram a violência contra a mulher por parceiro íntimo. Observou-se que estes artigos foram publicados, em sua maioria, no idioma inglês (84,2%), em revistas das ciências médicas (49,1%) e com enfoque metodológico quantitativo (86,5%). Foi identificada uma diversidade significativa de países onde os estudos foram realizados, com destaque para a América do Norte (43,9%). Dentre os sujeitos, foram mais pesquisadas mulheres entre 20 e 39 anos, casadas (72,5%) e cujo agressor constituía o parceiro íntimo atual (97,1%). Os sujeitos foram abordados principalmente em amostras por domicílios (30,4%) e em serviços de saúde (29,8%). Em relação à abordagem da violência, houve prevalência da associação dos diferentes tipos, que foram descritos predominantemente por atos e/ou comportamentos de abuso/agressão. Destacou-se a violência física. Foi observada a invisibilidade das formas subjetivas da violência, apesar de ocorrerem sobrepostas aos outros tipos e serem consideradas fatores de risco às agressões físicas. Os resultados sugerem que futuras pesquisas associem diferentes enfoques metodológicos e detalhem as especificidades dos sujeitos, assim como investiguem os abusos emocionais e o controle, e sua relação com a dinâmica de violência.

Ein charakteristisches Merkmal der Alzheimer-Krankheit ist die Ablagerung von Amyloid-beta (A beta) Protein im Gehirn. Die Proteinaggregate bilden Plaques, in deren Umgebung eine chronische Entzündungsreaktion nachgewiesen werden kann. Welche Rolle diese plaqueassoziierte Inflammation für die Pathogenese der Alzheimerschen Krankheit spielt, ist unklar. Es wird diskutiert, dass es sich um einen Versuch des Immunsystems handelt, A beta durch Prozessierung und Phagozytose aus dem Gehirn zu entfernen. Durch die dadurch entstehende chronische Entzündung könnten Nervenzellen in der Umgebung von Plaques geschädigt werden ("bystander attack"). Ein Schritt zu einem besseren Verständnis der plaqueassoziierten Entzündungsprozesse und ihrer pathogenetischen Bedeutung ist die Aufklärung der zugrunde liegenden molekularen Mechanismen. Daher beschäftigt sich die vorliegende Arbeit mit drei zentralen Fragen: (1) Welche Moleküle sind an der plaqueassoziierten Entzündungsreaktion beteiligt? (2) Sind ausgewählte Kandidatenmoleküle (Toll-like Rezeptoren, SOCS) in der Umgebung von Plaques reguliert? (3) Welche Rolle spielen Entzündungsmediatoren aus Endothelzellen in der Nähe von Plaques? Die plaqueassoziierten Entzündungsprozesse wurden im Gehirn von alten APP23 transgenen Mäusen, einem Modell der Alzheimer-Erkrankung, untersucht. Plaques, plaquenahes Gewebe und plaquefreies Gewebe wurden mittels Laser Mikrodissektion ausgeschnitten und anschließend mit Hilfe von Mikroarrays und quantitativer RT-PCR analysiert. Dazu wurden zwei methodische Ansätze gewählt: Im ersten Teil der Arbeit wurden in einem hypothesenfreien Ansatz neue regulatorische Kandidatenmoleküle identifiziert, unter ihnen der Rezeptor Trem2. Dabei konnten sowohl eine erhöhte plaqueassoziierte mRNA als auch eine erhöhte Protein Expression von Trem2 sowie dessen Lokalisation auf Mikrogliazellen nachgewiesen werden. Trem2 spielt anscheinend eine Rolle bei der Herbeiführung eines mikroglialen Aktivierungsstatus, der die Phagozytose unterstützt, während die Entstehung von proinflammatorischen Zytokinen unterdrückt wird. Im zweiten Teil der vorliegenden Arbeit wurde mit einem hypothesenbasierten Ansatz die plaqueassoziierte mRNA Expression von Molekülen untersucht, die bereits in anderen Untersuchungen mit der Alzheimer-Erkrankung in Zusammenhang gebracht wurden. Dabei konnte eine erhöhte plaqueassoziierte mRNA Expression der Toll-like Rezeptoren Tlr2, 4 und 9 sowie einzelner SOCS in APP23 transgenen Mäusen nachgewiesen werden. Tlr2 und 4 sind in der Lage, Mikrogliazellen und andere phagozytierende Zellen zu aktivieren und werden mit der Aufnahme und Beseitigung von Amyloid-beta in Verbindung gebracht. Im dritten Teil der Arbeit wurde ebenfalls mit einem hypothesenbasierten Ansatz die mRNA Expression von plaqueassoziiertem Endothelgewebe überprüft. Dabei konnte eine erhöhte mRNA Expression von MIP-1 alpha und CXCL10, zwei Entzündungsmediatoren aus der Familie der Chemokine, in plaqueassoziiertem Endothelgewebe gezeigt werden. Es kann daher davon ausgegangen werden, dass auch das Endothelgewebe an der Entzündungsreaktion beteiligt ist. Die Ergebnisse dieser Arbeit tragen zu einem besseren Verständnis der plaqueassoziierten Entzündungsreaktion im Rahmen der Alzheimer-Erkrankung bei. Die Aufklärung der Pathogenese der Alzheimer-Erkrankung ist entscheidend, um in den nächsten Jahren und Jahrzehnten neue und effektive Medikamente zur Behandlung dieser schweren Demenzerkrankung zu entwickeln.
