The spectral properties and dynamics of the fluorescence emission of photosystem II core complexes are investigated by single-molecule spectroscopy at 1.6 K. The emission spectra are dominated by sharp zero-phonon lines (ZPLs). The sharp ZPLs are the result of weak to intermediate exciton-vibrational coupling and slow spectral diffusion. For several data sets, it is possible to surpass the effect of spectral diffusion by applying a shifting algorithm. The increased signal-to-noise ratio enables us to determine the exciton-vibrational coupling strength (Huang-Rhys factor) with high precision. The Huang-Rhys factors vary between 0.03 and 0.8. The values of the Huang-Rhys factors show no obvious correlation between coupling strength and wavelength position. From this result, we conclude that electrostatic rather than exchange or dispersive interactions are the main contributors to the exciton-vibrational coupling in this system.

This survey discusses current and emerging isothermal and rapid polymerase chain reaction (PCR) based nucleic acid amplification methods for near-patient diagnostics. To assess the clinical need of rapid diagnostics for infectious diseases based on nucleic acid tests (NATs) we performed and analysed a questionnaire among laboratories participating in corresponding INSTAND ring trials for external quality assurance. The questions concerning new amplification technologies like isothermal nucleic acid amplification, potentially suited to significantly decrease turnaround times, were complemented by questions to evaluate the present status of NATs. Besides end-users, companies were also addressed by sending out a manufacturer specific questionnaire. Analysis of the answers from 48 laboratories in 14 European countries revealed that a much shorter turnaround time is requested for selected pathogens compared to about 2 h or longer when applying temperature cycling amplification, i.e. PCR. In this context, most frequently mentioned were methicillin-resistant Staphylococcus aureus (MRSA), norovirus, influenza A and B viruses, cytomegalovirus (CMV) as well as hepatitis B virus (HBV) and hepatitis C virus (HCV). Photosystem I (PSI) ist ein essentielles Protein in der Photosynthese, das in seiner trimeren Form in Cyanobakterien ca. 300 Chlorophyllmoleküle bindet. In der vorliegenden Arbeit wurde die Fluoreszenz einzelner PSI-Komplexe aus den Cyanobakterien Thermosynechococcus elongatus und Arthrospira platensis mithilfe eines konfokalen Mikroskops bei tiefer Temperatur (1,4 K) untersucht. Glycerin bindet direkt an PSI: Vergleichende Fluoreszenzuntersuchungen von Präparierungen von PSI aus T. elongatus bei 1,4 K in wässrigem Puffer ohne Zusätze, mit Glycerin bzw. in Polyvinylalkohol-Filmen (PVA) zeigten einen deutlichen und gegensätzlichen Einfluss von Glycerin und PVA auf die Konformation von PSI: Glycerin stabilisiert das Proteingerüst und führt zu einer kompakteren Proteinkonformation der PSI-Komplexe, während PVA destabilisierend wirkt und eine unnatürliche Veränderung des Proteingerüsts hervorruft. Durch weitere Experimente konnte gezeigt werden, dass mindestens ein Teil des stabilisierenden Effekts von Glycerin durch dessen Bindung an PSI vermittelt wird. Der Redoxzustand des Reaktionszentrums beeinflußt die Fluoreszenz: Spektren einzelner PSI-Komplexe aus A. platensis zeigen drei unterschiedliche Emissionsbanden, von denen bislang nur zwei aus Ensemblemessungen bekannt waren. Die neu entdeckte Emissionsbande zeichnet sich durch scharfe Nullphononenlinien aus. Die große Ähnlichkeit zu den Spektren von T. elongatus deutet auf eine strukturelle Ähnlichkeit der PSI-Komplexe dieser beiden Organismen hin. In Kooperation mit Eberhard Schlodder (TU Berlin) konnte in Experimenten an Ensembleproben für PSI-Komplexe aus T. elongatus eine bisher unbekannte Emissionsbande nachgewiesen werden, deren Intensität stark vom Redoxzustand des Reaktionszentrums in den PSI-Komplexen abhängt. Sie entspricht einer für PSI-Komplexe aus A. platensis bereits bekannten Emissionsbande. Darüber hinaus wurde die Abhängigkeit der Emission vom Redoxzustand des Reaktionszentrums für beide Organismen erstmals an einzelnen PSI-Komplexen nachgewiesen. Metall-Nanostrukturen ändern die Funktion von PSI-Komplexen: Bio-Nano-Hybridsysteme werden wegen ihres Potentials zur Stromerzeugung und Wasserstoffproduktion intensiv erforscht. Untersuchungen von PSI-Komplexen aus T. elongatus auf hexagonal angeordneten periodischen Metallnanostrukturen - sog. Fischer-Pattern - aus Gold und Silber zeigten eine Verstärkung der Fluoreszenzemission, die bei Silber stärker ausfiel als bei Gold. Die zusätzlich nachgewiesene Abhängigkeit der Verstärkung von der Wellenlänge deutet auf eine Veränderung der Funktion der PSI-Komplexe durch die Metallnanostrukturen hin. Ein neuartiges konfokales Tieftemperaturmikroskop für Einzelmolekülspektroskopie: Das Herzstück des von Grund auf neu entwickelten konfokalen Tieftemperaturmikroskops für Einzelmolekülspektroskopie bilden Piezoaktuatoren zur Probenpositionierung und Piezoscanner zur Rasterung der Probe. Ein neuartiges Probentransfersystem erlaubt den Einbau bzw. das Wechseln einer Probe bei tiefen Temperaturen bis hinunter zu 4,2 K. Ein vergleichbares System wurde bisher nicht beschrieben. Eine parallel entwickelte Steuerungssoftware dient der Steuerung aller Komponenten und der Erfassung von Messdaten. Darüber hinaus wurde eine Auswertungssoftware zur Analyse der erfassten Daten entwickelt, mit der die Daten gesichtet und statistisch ausgewertet werden können. Photosystem I (PSI) is a protein essential for photosynthesis harbouring in its trimeric form in cyanobacteria about 300 chlorophyll molecules. In this work, I report on fluorescence measurements of single PSI complexes from the cyanobacteria Thermosynechococcus elongatus and Arthrospira platensis, performed with a confocal microscope at low temperature (1.4 K). Glycerol binds directly to PSI: Fluorescence measurements comparing preparations of PSI from T. elongatus at 1.4 K in buffer without additives, with glycerol, and in poly(vinyl alcohol) (PVA) films showed a clear and opposite effect of glycerol and PVA on the conformation of PSI. Whereas glycerol stabilises the protein scaffold and induces a more compact protein conformation of the PSI complexes, PVA destabilises and leads to a nonnatural protein conformation. Further experiments showed clearly that the stabilising effect of glycerol can be attributed at least in part to a direct interaction with PSI. Fluorescence is affected by the redox state of the reaction center: Spectra of single PSI complexes of A. platensis showed three distinctive emission bands, only two of which had been known from ensemble measurements. This newly discovered emission band is characterised by sharp zero phonon lines. The striking similarity to spectra from T. elongatus points towards structural similarities of the PSI complexes of both organisms. Bulk spectra of T. elongatus preparations recorded in collaboration with Eberhard Schlodder (TU Berlin) led to the discovery of a new, previously unknown emission band whose emission intensity depends strongly on the redox state of the reaction centre. This band corresponds to the one known for PSI complexes from A. platensis. For the first time this dependence of fluorescence on the redox state of the reaction centre could be demonstrated for single PSI complexes. Metal nanostructures change the function of PSI complexes: Due to their potential use for power generation and hydrogen production, bio-nano hybrid systems are currently intensely investigated. PSI complexes from T. elongatus showed amplified emission on hexagonal periodic metal nanostructures (Fischer pattern) made of gold and silver, respectively. The amplification was stronger with silver compared to gold. Additionally, the amplification depended on the wavelength, pointing towards a change in function of the PSI complexes due to the metal nanostructures. A new confocal low-temperature microscope for single molecule spectroscopy: At the heart of the newly developed confocal low-temperature microscope for single molecule spectroscopy are piezo actuators for sample positioning and piezo scanners for sample scanning. A new system for sample transfer allows for change of samples at very low temperatures down to 4.2 K. Comparable systems have not been described so far. The setup is operated by software developed in parallel, allowing for controlling all components and acquiring data. Furthermore, a toolbox for data analysis was developed to display the data and perform statistical analyses.
