Abstract

Screen-printed electrodes (SPEs) are reliable, portable, affordable, and versatile electrochemical platforms for the real-time analytical monitoring of emerging analytes in the environmental, clinical, and agricultural fields. The aim of this study was to evaluate the electrochemical behavior of gold screen-printed electrodes (SPGEs) modified with molecules containing amino (Tr-N) or α-aminophosphonate (Tr-P) groups for the selective and sensitive detection of the toxic metal ions Pb2+ and Hg2+ in aqueous samples. After optimizing the analytical parameters (conditioning potential and time, deposition potential and time, pH and concentration of the supporting electrolyte), anodic square wave stripping voltammetry (SWASV) was used to evaluate and compare the electrochemical performance of bare or modified electrodes for the detection of Hg2+ and Pb2+, either alone or in their mixtures in the concentration range between 1 nM and 10 nM. A significative improvement in the detection ability of Pb2+ ions was recorded for the amino-functionalized gold sensor SPGE-N, while the presence of a phosphonate moiety in SPGE-P led to greater sensitivity towards Hg2+ ions. The developed sensors allow the detection of Pb2+ and Hg2+ with a limit of detection (LOD) of 0.41 nM and 35 pM, respectively, below the legal limits for these heavy metal ions in drinking water or food, while the sensitivity was 5.84 µA nM−1cm−2 and 10 µA nM−1cm−2, respectively, for Pb2+ and Hg2+. The reported results are promising for the development of advanced devices for the in situ and cost-effective monitoring of heavy metals, even in trace amounts, in water resources. Abstract

New technologies and materials could help in this fight against healthcare-associated infections. As the majority of these infections are caused by antibiotic-resistant bacteria, the development of materials with intrinsic antibacterial properties is a promising field of research. We combined chitosan (CS), with antibacterial properties, with polyhedral oligomeric silsesquioxanes (POSS), a biocompatible polymer with physico-chemical, mechanical, and rheological properties, creating a hydrogel using cross-linking agent genipin. The antibacterial properties of CS and CS-POSS hydrogels were investigated against nosocomial Gram-positive and Gram-negative bacteria both in terms of membrane damage and surface charge variations, and finally, the anti-biofilm property was studied through confocal microscopy. Both materials showed a good antibacterial capacity against all analyzed strains, both in suspension, with % decreases between 36.36 and 73.58 for CS and 29.86 and 66.04 for CS-POSS, and in plates with % decreases between 55.29 and 78.32 and 17.00 and 53.99 for CS and CS-POSS, respectively. The treated strains compared to the baseline condition showed an important membrane damage, which also determined a variation of surface charges, and finally, for both hydrogels, a remarkable anti-biofilm property was highlighted. Our findings showed a possible future use of these biocompatible materials in the manufacture of medical and surgical devices with intrinsic antibacterial and anti-biofilm properties. Abstract

Azepane–based compounds are saturated seven-membered carbocycles where one carbon atom is replaced with a nitrogen atom. The azepane structure is also found in fused-ring derivatives endowed of a wide range of pharmacological activities. The azepane motif is in the top 100 most frequently used ring systems for small molecule drugs. Several synthetic methods toward the preparation of new azepane-based compounds scaffolds have gained significant interest and represent a continuous challenge for synthetic organic chemists. The synthesis of the azepane ring is based on different synthetic approaches such as: ring-closing reactions; ring-expansion reactions of cyclic compounds; and multistep sequences. The azepane based compounds continue to play great synthetic relevance because of their different biological activities such as anti-cancer, antimicrobial, anti-Alzheimer's disease and anticonvulsant activity. New synthetic methods for the construction of azepane derivatives endowed of relevant biological activity and good pharmacological profiles is expected in near future.

29. Determination of Phenylalanine by a Novel Enzymatic PHD/SPE Biosensor

Abstract

In this article, we report the first example of a novel potentiometric sensor for the real-time monitoring of phenylalanine (Phe). The developed enzymatic biosensor consists of a commercial screen-printed electrode (SPE) platform with the gold working electrode covalently functionalized with the enzyme Phe dehydrogenase (PHD). The PHD/SPE membrane-free biosensor was fabricated by immobilizing the PHD enzyme to the gold electrode using 3-3’-dithiopropionic acid of $N$ -succinimide ester (DSP) as a linker capable of creating a thiol bond with the gold electrode and an amide bond with the amine group present on the PHD enzyme. The ability of the enzyme to catalyze the transformation of Phe to pyruvic acid, NADH, and ammonia, under the conditions used for the enzymatic biosensor, was first assessed by UV-Vis analysis under batch conditions. Under the experimental conditions adopted, the enzymatic reaction was almost completed in 40–60 min. The biosensor was then tested for the Phe monitoring using the open circuit potential (OCP) potentiometric technique, recording the potential of the biosensor as the reaction proceeded over time. The potentiometric response with respect to the Ag/AgCl reference electrode was found to be linear over a relatively wide concentration range (0– $5000 \mu \text{M}$ ). The reported results demonstrated the feasibility of developing a novel membrane-free enzymatic biosensor for Phe monitoring in patients affected with phenylketonuria (PKU).

Abstract

Functionalized polyhedral oligosilsesquioxanes (POSS) containing an isoxazolidine nucleus have been synthesized by microwave assisted 1,3-dipolar cycloaddition of N-methyl-C-alkoxycarbonyl nitrone 1 with POSS containing olefin moieties. The results of cycloaddition processes were rationalized by computational studies at the DFT level. The covalent conjugation of chitosan with the cycloadduct 3a leads to composite material CS-POSS 7 which was gelified using genipin as cross linking agent. The suitability of the system for bone tissue engineering purposes was evaluated by in vitro drug release studies using ketoprofen as a model drug and cytotoxicity assays performed on human fetal osteoblastic cells. The preliminary biological tests showed the lack of cytotoxicity of the hybrid material and suggest its potential role in bone tissue engineering applications.

The cycloaddition reaction between C-(2-thiazolyl) nitrones and allylic alcohol takes place with complete exo selectivity when the reactions are carried out in presence of one equiv of zinc triflate. The use of a chiral dipolarophile allowed application to the methodology to the synthesis of a hitherto unknown enantiomerically pure isoxazolidinyl analogue of the C-nucleoside tiazofurin.

Abstract

A novel, efficient entry to 2'-(dimethylamino)-2'-3'-dideoxynucleosides was described. The synthetic approach involves the 1,3-dipolar cycloaddition of C-alkoxycarbonyl nitrones to allyl acetate, reductive ring opening to substituted lactones, DIBALH reduction to the corresponding 3-(dimethylamino)tetrahydro-2-furanols and coupling with silylated thymine. Formal synthesis of d4T and analogous unsaturated 2',3'-dideoxynucleosides was made possible by removal of the dimethylamino group via Cope elimination.

Abstract

Bioabsorbable materials have received increasing attention as innovative systems for the development of osteoconductive biomaterials for bone tissue engineering. In this paper, chitosan-based composites were synthesized adding hydroxyapatite and/or magnetite in a chitosan matrix by in situ precipitation technique. Composites were characterized by optical and electron microscopy, thermogravimetric analyses (TGA), x-ray diffraction (XRD), and in vitro cell culture studies. Hydroxyapatite and magnetite were found to be homogeneously dispersed in the chitosan matrix and the composites showed superior biocompatibility and the ability to support cell attachment and proliferation; in particular, the chitosan/hydroxyapatite/magnetite composite (CS/HA/MGN) demonstrated superior bioactivity with respect to pure chitosan (CS) and to the chitosan/hydroxyapatite (CS/HA) scaffolds.

Abstract

Two different routes which exploit the reactivity of a phosphonated nitrone are employed to obtain a new template of C-4?-truncated phosphonated nucleosides (TPCOANs) in good yields. The [beta]-anomers of TPCOANs exhibit the ablility to inhibit the reverse trancriptase of different retroviruses at concentrations in the nanomolar range, with potency comparable with that of tenofovir.
