Agricultural intensification and associated loss of high-quality habitats are key drivers of insect pollinator declines. With the aim of decreasing the environmental impact of agriculture, the 2014 EU Common Agricultural Policy (CAP) defined a set of habitat and landscape features (Ecological Focus Areas: EFAs) farmers could select from as a requirement to receive basic farm payments. To inform the post-2020 CAP, we performed a European-scale evaluation to determine how different EFA options vary in their potential to support insect pollinators under standard and pollinator-friendly management, as well as the extent of farmer uptake.A structured Delphi elicitation process engaged 22 experts from 18 European countries to evaluate EFAs options. By considering life cycle requirements of key pollinating taxa (i.e. bumble bees, solitary bees and hoverflies), each option was evaluated for its potential to provide forage, bee nesting sites and hoverfly larval resources.EFA options varied substantially in the resources they were perceived to provide and their effectiveness varied geographically and temporally. For example, field margins provide relatively good forage throughout the season in Southern and Eastern Europe but lacked early-season forage in Northern and Western Europe. Under standard management, no single EFA option achieved high scores across resource categories and a scarcity of late season forage was perceived.Experts identified substantial opportunities to improve habitat quality by adopting pollinator-friendly management. Improving management alone was, however, unlikely to ensure that all pollinator resource requirements were met. Our analyses suggest that a combination of poor management, differences in the inherent pollinator habitat quality and uptake bias towards catch crops and nitrogen-fixing crops severely limit the potential of EFAs to support pollinators in European agricultural landscapes.Policy Implications. To conserve pollinators and help protect pollination services, our expert elicitation highlights the need to create a variety of interconnected, well-managed habitats that complement each other in the resources they offer. To achieve this the Common Agricultural Policy post-2020 should take a holistic view to implementation that integrates the different delivery vehicles aimed at protecting biodiversity (e.g. enhanced conditionality, eco-schemes and agri-environment and climate measures). To improve habitat quality we recommend an effective monitoring framework with target-orientated indicators and to facilitate the spatial targeting of options collaboration between land managers should be incentivised.

There is widespread concern regarding declines in bee populations given their importance for the functioning of both natural and managed ecosystems. An increasing number of studies find negative relations between bee species richness and simplification of agricultural landscapes, but the role of land-use intensity and its relative importance compared to landscape simplification remain less clear. We compared the relative effects of nitrogen inputs, as a proxy for land-use intensity, and proportion of natural and semi-natural habitat, as a measure of landscape complexity on total bee species richness, rare species richness and dominant crop-visiting species richness. We used data from 282 grasslands across five countries, covering the entire range of low intensity, no-input systems, to high-input sites (>400 kg N/ha/year). We found consistent negative impacts of increasing land-use intensity at a regional scale on total bee species richness and dominant crop-visiting species across Europe, but no such effects of landscape complexity. In contrast, the richness of rare bee species was not significantly related to increasing land-use intensity. Nevertheless, based on species accumulation curves, grasslands with no nitrogen inputs had higher total bee richness and higher shares of rare species compared with sites with high nitrogen inputs (>125 kg N/ha/year). Our results highlight the importance of retaining grasslands characterised by low land-use intensity across agricultural landscapes to promote bee diversity.

De laatste tijd is er veel aandacht voor de achteruitgang van bijen. Er zijn inmiddels veel initiatieven om wilde bijen een handje te helpen, meestal door het inzaaien van bloemenmengsels. Maar profiteren wilde bijen hier wel van? Welke soorten hebben daar het meest baat bij? Bevordert dit ook de bestuiving van naastgelegen gewassen? En waar moet rekening mee gehouden worden bij het aanleggen en beheren van een goede bloemenstrook? Aan de hand van literatuuronderzoek is naar antwoorden gezocht.

Growing interest in extracellular vesicles (EVs, including exosomes and microvesicles) as therapeutic entities, particularly in stem cell-related approaches, has underlined the need for standardization and coordination of development efforts. Members of the International Society for Extracellular Vesicles and the Society for Clinical Research and Translation of Extracellular Vesicles Singapore convened a Workshop on this topic to discuss the opportunities and challenges associated with development of EV-based therapeutics at the preclinical and clinical levels. This review outlines topic-specific action items that, if addressed, will enhance the development of best-practice models for EV therapies.

Ecological intensification aims to enhance important ecosystem processes that contribute to the delivery of the ecosystem services that underpin agricultural production allowing us to reduce our reliance on synthetic inputs. The potential of ecological intensification will depend on many factors, among the most important of which are off-field management and landscape context. These factors, and importantly the interaction between them, are likely to vary across regions and countries.Identifying off-field management approaches that are successful in enhancing ecosystem services will require assessing a range of strategies. The empirical work carried out in task 3.2 provided Original data on the effectiveness of three off-field interventions (hedgerow, set- aside and flower strips) onthe delivery of biocontrol and yield in winter cereals across different European countries. For hedgerows we found that the quality of the hedgerow (flower diversity) generally increased biodiversity of several beneficial groups of insects (e.g. butterflies, tachinids, carabids, spiders), while the delivery of ecosystems services such as pollination and pest control tended to respond more tolandscape factors (proportion of hedgerows or semi-natural habitats in general in the surrounding).For set-aside we found that this intervention increased locally the biodiversity of several beneficial insect groups (literature) but the spillover to winter wheat fields was small with no apparent benefit on the delivery of aphid biocontrol. Finally, we found that wildflower strips helped to reduce aphid pests in winter wheat fields, which, in turn, enhanced crop yield. However, this potential may only be reached in case strips are properly managed, in a way that optimizes floral diversity, and may only be relevant in agricultural landscapes with a low availability of habitat area for natural enemies.Irrespective of the intensity of the agricultural systems, the two most promising interventions to foster biocontrol and support yield in winter wheats are hedgerows and flower strips, but their effect appeared to be stronger in landscapes with low cover of existing semi-natural habitats.

Akkervogels zijn de afgelopen decennia in West-Europa sterk in aantal afgenomen. Een van de belangrijkste oorzaken is de intensivering van de landbouw. Toegenomen gebruik van pesticiden en meststoffen, verandering van gewasrotaties, efficiëntere oogstmethoden en schaalvergroting leiden tot gebrek aan voedsel en nestgelegenheid, en als gevolg daarvan tot een afname van overleving en reproductie (Chamberlain et al., 2000: Donald et., 2001: Newton, 2004). Vooral zaad etende akkervogels, zoals Ringmus (Passer montanus), Kneu (Carduelis cannabina), Veldleeuwerik (Alauda arvensis) en Grauwe gors (Emberiza calandra), die grotendeels afhankelijk zijn van het boerenland voor voedsel en dekking (Bijlsma et al., 2001) hebben het zwaar.

In moderne cultuurlandschappen wordt het voor wilde bijen steeds moeilijker om de benodigde hulpbronnen, zoals bloemen en nestgelegenheid te bemachtigen. Wij willen nagaan of de aanleg van meerjarige bloemenvelden de achteruitgang van wilde bijen in deze landschappen kan tegengaan. De resultaten laten zien dat het verhogen van het aanbod van bloemen kan leiden tot een soortenrijkere gemeenschap van wilde bestuivers, niet alleen in de bloemenvelden zelf maar ook in het omliggende landschap.

An important goal of ecological compensation areas (ECAs) is to increase biodiversity in adjacent intensively managed farmland and the agricultural landscape at large. We tested whether this goal can be achieved in the case of the agri-environmental restoration scheme implemented for Swiss grassland using five large arthropod taxa (bees, true bugs, orthopterans, ground beetles and spiders) representing different ecological and functional groups. The species richness and abundance of all groups and species, respectively, was measured along 100 m transects from ECA-meadows into the adjacent intensively managed grassland at 24 sites. Species richness of all arthropod taxa except ground beetles, and the abundance of 63% of the 234 arthropod species sampled with at least five individuals were higher in ECA-meadows than in their surroundings, while the total abundance of spiders and ground beetles was higher in intensively managed meadows. The abundance of 8% of these species were only increased in the ECA-meadows themselves (“stenotopic” species) but 40% had increased abundance both in the ECA-meadows and the adjacent grassland, declining exponentially with increasing distance from ECA-meadows (“edge species”). The 90%-decay distances for these edge species differed among taxonomic groups (117 ± 18 m for true bugs, 137 ± 24 m for spiders, 152 ± 34 m for bees, 167 ± 5.7 m for orthopterans, 185 ± 34 m for ground beetles; mean ±1 standard error) and independent of taxonomic group were larger for large-sized or predacious species than for small-sized or phytophagous species. Because the average distance between neighbouring ECA-meadows in Swiss grassland is only 73 ± 4 m, the current agri-environment scheme very likely enhances arthropod diversity and possibly associated ecosystem services in the Swiss agricultural landscape at large.

De bestuiving van landbouwgewassen door gehouden en in het wild levende soorten bijen en zweefvliegen vormt een relevante en veelgenoemde ecosysteemdienst, die echter onder toenemende druk staat. De honingbijen die traditioneel landbouwgewassen bestuiven, gaan in aantal sterk achteruit als gevolg van te hoge sterfte, met name gedurende de winterperiode. Momenteel wordt door meerdere instituten, waaronder Wageningen UR, onderzoek uitgevoerd naar de oorzaken van de sterfte van honingbijen. Waarschijnlijk speelt een complex van factoren een rol, waaronder het gebruik van bepaalde insecticiden en de parasitaire varroamijt, een gebrek aan natuurlijke voedselbronnen en/of klimaatsveranderingen. Ook veel wilde bijensoorten nemen in aantal af. Ongeveer de helft van de wilde bijensoorten in Nederland staat op de Rode Lijst. Hoofddoel van het voorliggende onderzoek was het agronomisch en economisch kwantificeren van de bijdrage van bestuivende diensten door wilde en gedomesticeerde (bijgeplaatste) insecten aan de landbouwkundige productie van twee belangrijke Nederlandse insecten-bestoven fruitgewassen: de appel en de blauwe bes. Op deze wijze levert het onderzoek een ‘proof-of-concept’ van het economisch belang van ecosysteemdiensten, en daarmee biodiversiteit, voor de Nederlandse samenleving.

Hoe ziet het landschap eruit als bijen op een kunstbloem met gekleurde pollen foerageren? Als een Van Gogh? Het idee levert onderzoeker David Kleijn een plek op in de finale van de Bio-Art & Design competitie. Synthetic Pollinizer heet het project dat Kleijn samen met de Australische kunstenaar Michael Candy uit wil voeren. En die ‘Kunstbloem’ komt er als het duo bij de eerste drie van de BAD-Award eindigt. Dan ligt er 25.000 euro te wachten om het bestuivingsproject daadwerkelijk uit te voeren.

De bestuiving van landbouwgewassen is het werk van slechts een handjevol soorten bijen. De rest is economisch gezien nutteloos. Als argument voor natuurbeheer is dus meer nodig dan economisch nut.

Insecten leveren een bijdrage aan ecosysteemdiensten vanwege de bestuiving van allerlei gewassen. Maar uit nieuw onderzoek, gepubliceerd in Nature Communications, blijkt dat dat slechts geldt voor een kleine groep algemene soorten. Zeldzame soorten dragen nauwelijks bij aan bestuiving. In het internationale debat over biodiversiteitsbehoud kan de huidige focus op ecosysteemdiensten als argument voor soortbescherming voor zeldzame soorten dus slecht uitpakken.

Over half of the European landscape is under agricultural management and has been for millennia. Many species and ecosystems of conservation concern in Europe depend on agricultural management and are showing ongoing declines. Agri-environment schemes (AES) are designed partly to address this. They are a major source of nature conservation funding within the European Union (EU) and the highest conservation expenditure in Europe. We reviewed the structure of current AES across Europe. Since a 2003 review questioned the overall effectiveness of AES for biodiversity, there has been a plethora of case studies and meta-analyses examining their effectiveness. Most syntheses demonstrate general increases in farmland biodiversity in response to AES, with the size of the effect depending on the structure and management of the surrounding landscape. This is important in the light of successive EU enlargement and ongoing reforms of AES. We examined the change in effect size over time by merging the data sets of 3 recent meta-analyses and found that schemes implemented after revision of the EU's agri-environmental programs in 2007 were not more effective than schemes implemented before revision. Furthermore, schemes aimed at areas out of production (such as field margins and hedgerows) are more effective at enhancing species richness than those aimed at productive areas (such as arable crops or grasslands). Outstanding research questions include whether AES enhance ecosystem services, whether they are more effective in agriculturally marginal areas than in intensively farmed areas, whether they are more or less cost-effective for farmland biodiversity than protected areas, and how much their effectiveness is influenced by farmer training and advice? The general lesson from the European experience is that AES can be effective for conserving wildlife on farmland, but they are expensive and need to be carefully designed and targeted.

Changes in mRNA expression levels occur during physiological and pathological processes in the cardiovascular system. An increase inDNAtranscription results in increasedmRNAlevels and will subsequently result in increased protein levels that regulate processes inside and outside the cell. To determine alterations in mRNA levels, traditional methods such as Northern blot and ribonuclease protection assay can be used; however, large amounts of RNA are necessary and the methods are very labor intensive. In this chapter, we focus on the newest advancements in reverse transcription polymerase chain reaction (rt-pcr) technology, the real-time PCR or quantitative PCR, using small amounts of RNA to determine expression levels.We discuss the technique in general and describe two different approaches.

Boeren moeten een serie verplichte maatregelen doorvoeren die bedoeld zijn om de Europese landbouw natuur- en milieuvriendelijker te maken. De maatregelen vallen onder drie noemers: gewasrotatie, behoud van permanent grasland, en het inrichten van 5 procent van hun areaal voor ecologische doeleinden, de zogenoemde ecological focus areas (EFA’s). Wat denken experts ervan: gaat de vergroening vruchten afwerpen?

De wereldwijde afname van bijen en ander bestuivers heeft grote consequenties voor de voedselzekerheid. Verdere verslechtering van de leefomstandigheden kan leiden tot het plotseling uitsterven van bestuivende insectensoorten. In Europa dreigt inmiddels een tekort aan honingbijen voor de bestuiving van landbouwgewassen. De vraag naar bestuivers groeit sneller dan het aanbod van bijenvolken, vooral door de groei van het areaal energiegewassen, zoals koolzaad, zonnebloem en soja. Wilde bestuivers zoals bijen en zweefvliegen leveren een belangrijke bijdrage aan de productie van insect-bestoven landbouwgewassen. Wat de bijdrage van wilde bestuivers is ten opzichte van de diensten geleverd door de honingbij is momenteel onbekend in Nederland. De huidige studie had tot doel meer inzicht te krijgen in (1) welke wilde bestuivers tot soorten behoren die voor de - Nederlandse - landbouw relevant kunnen worden geacht, (2) wat bekend is van hun populatieontwikkelingen en wat waarschijnlijk belangrijke factoren zijn die hun achteruitgang kunnen verklaren en (3) indien de achteruitgang van voedselplanten een belangrijke factor zou zijn bij de achteruitgang van wilde bestuivers, welke (natuur)beheermaatregelen dan eventueel denkbaar zijn om de beschikbaarheid van voedselplanten te bevorderen.

Deze studie onderzocht of de achteruitgang van akkervogels in kleinschalige Nederlandse landbouwgebieden tegengegaan kan worden door het vergroten van de voedselbeschikbaarheid voor deze soorten in de winterperiode. De voedselbeschikbaarheid werd experimenteel vergroot in 10 gebieden van ongeveer 100 ha door inzaai van gewasmensgels die niet werden geoogst. Tien gepaarde gebieden met vergelijkbare landschapsstructuur waarin voedselbeschikbaarheid niet werd gemanipuleerd fungeerden als controles. De beschikbaarheid aan zaden, het gebruik van de mengsels door akkervogels, het effect van de mengsels op vogels in de winter en in het broedseizoen werden vervolgens gekwantificeerd om het effect van het vergroten van de voedselbeschikbaarheid vast te stellen.

Declines in insect pollinators across Europe have raised concerns about the supply of pollination services to agriculture. Simultaneously, EU agricultural and biofuel policies have encouraged substantial growth in the cultivated area of insect pollinated crops across the continent. Using data from 41 European countries, this study demonstrates that the recommended number of honeybees required to provide crop pollination across Europe has risen 4.9 times as fast as honeybee stocks between 2005 and 2010. Consequently, honeybee stocks were insufficient to supply >90% of demands in 22 countries studied. These findings raise concerns about the capacity of many countries to cope with major losses of wild pollinators and highlight numerous critical gaps in current understanding of pollination service supplies and demands, pointing to a pressing need for further research into this issue.

7B2 is a neuroendocrine chaperone interacting with the prohormone convertase PC2 in the regulated secretory pathway. Its gene is located near the Prader-Willi syndrome (PWS) region on chromosome 15. In a previous study we were able to show 7B2 immunoreactivity in the supraoptic nucleus (SON) or the paraventricular nucleus (PVN) in only three of five PWS patients. Here we report that in contrast with five other PWS patients, the neurons in the hypothalamic SON and PVN of the two 7B2-immunonegative PWS patients also failed to show any reaction using two antibodies directed against processed vasopressin (VP). On the other hand, even these two cases reacted normally with five antibodies that recognize different parts of the VP precursor. This finding pointed to a processing defect. Indeed, the same patients had no PC2 immunoreactivity in the SON or PVN, whereas PC1 immunoreactivity was only slightly diminished. In conclusion, in the VP neurons of two PWS patients, greatly reduced amounts of 7B2 and PC2 are present, resulting in diminished VP precursor processing

Wolfram syndrome (WS) is characterized by optic atrophy, insulin-dependent diabetes mellitus, vasopressin (VP)-sensitive diabetes insipidus, and neurosensory hearing loss. Here we report a disturbance in VP precursor processing in the supraoptic and paraventricular nuclei of WS patients. In these patients with diabetes insipidus we could hardly detect any cellular immunoreactivity for processed VP in the supraoptic and paraventricular nuclei. On the other hand, in the paraventricular nucleus a considerable number of cells immunoreactive for the VP precursor were present. In addition, the proprotein convertase PC2 and the molecular chaperone 7B2 were absent. As expression of PC2 and 7B2 was detected in the nearby nucleus basalis of Meynert of one WS patient and in the anterior lobe of the other WS patient, the absence of the two proteins in the paraventricular nucleus was not due to mutations in their genes. These results indicate that in WS patients with diabetes insipidus, not only does VP neuron loss occur in the supraoptic nucleus, but there is also a defect in VP precursor processing

Aim: Atherosclerotic plaque characteristics may vary among individuals, in part due to heritable factors. The genetic architecture of plaque phenotypes is largely unknown. A common variant (rs17398575-A) near PIK3CG on chromosome 7 has previously been associated with carotid plaque presence. Animal models suggest that PIK3CG may play a role in plaque formation through neovascularization. We hypothesized that the PIK3CG variant is associated with intraplaque hemorrhage and vessel density in human plaque tissue. Methods: We collected 831 patients in the Athero-Express Biobank Study, all of whom underwent carotid endarterectomy and genotyped them using Affymetrix SNP 5.0. We tested PIK3CG variants for association to intraplaque hemorrhage and vessel density using logistic and linear regression model, respectively, correcting for age, gender and principal components. We used the BiKE cohort to assess the effect of PIK3CG variants on PIK3CG expression in circulating monocytes (N=95) and in carotid plaques (N=126). Results: The reported PIK3CG variant, rs17398575 (risk allele A, frequency = 0.72), was nominally significantly associated with intraplaque hemorrhage (OR=1.40 [1.10-1.69 95% CI], p=0.0271) and vessel density (β=0.095 [0.0415 s.e.m.], p=0.0221). We also report another nearby variant, rs849429 (risk allele A, frequency=0.49), but uncorrelated to rs17398575, associated with intraplaque vessel density (β=-0.164 [0.0361 s.e.m.], p=6.70×10-6). The SNP dependent PIK3CG mRNA expression demonstrated a differential effect in the vascular wall (p=0.783 for rs17398575; p=0.0198 for rs849429) compared to circulating monocytes (p=0.0261 for rs17398575; p=0.350 for rs849429). Conclusion: To our knowledge this is the first report involving the association of common genetic variants to histological plaque phenotypes. These results require further replication in independent cohorts. Further research should focus on elucidating the underlying mechanisms leading to plaque vessel formation and intraplaque hemorrhage, as both have been demonstrated to associate with secondary cardiovascular disease.

The Black-tailed Godwit, Limosa limosa, is a near-threatened wader bird with a European breeding distribution that is concentrated in Western Europe. This report summarizes the legal aspects of Black-tailed Godwit conservation and gives a concise overview of the implementation and effectiveness of conservation action that is taken in Belgium, Germany and the Netherlands. In all three countries, conservation initiatives are rarely effective enough to halt the ongoing decline of the species, although there are successful local exceptions. The review concludes with the recommendation to formulate a coherent strategy for Black-tailed Godwit conservation starting with the identification of favourable reference values in each of the three countries

Het grote oppervlak dat de landbouw inneemt maakt dat zelfs beperkte positieve effecten van agrarisch natuurbeheer een grote impact kunnen hebben. Anderzijds komen slechts weinig zeldzame soorten voor op landbouwgronden. De financiële middelen voor natuurbeheer staan momenteel onder druk. Er is behoefte aan inzicvhten die kunnen leiden tot een efficiëntere inzet van het beschikbare budget. Dit artikel geeft een overzicht van de (kosten)effectiviteit van agrarisch natuurbeheer en doet op basis daarvan aanbevelingen voor aanpassingen die kunnen leiden tot een slagvaardiger agrarisch natuurbeheer.

Het verklaren van populatietrends is een belangrijk doel geweest van een breed scala aan ecologische studies. Dergelijke studies worden bemoeilijkt doordat bij zeldzame soorten een bepaalde eigenschap of gedrag zowel oorzaak als gevolg kan zijn van de achteruitgang van een soort. Wij omzeilden dit probleem door eigenschappen van soorten te vergelijken aan de hand van exemplaren in natuurhistorische musea die waren verzameld toen de soorten nog algemeen voorkwamen. Over de rol van voedselvoorkeur en -specialisatie als veroorzaker van de achteruitgang van hommelsoorten wordt al lange tijd gediscussieerd. Wij vergeleken de samenstelling van stuifmeelladingen van vijf hommelsoorten met stabiele populaties en vijf soorten met afnemende populaties met behulp van exemplaren in musea die voor 1950 waren verzameld in België, Engeland en Nederland.

Onderzoek is uitgevoerd naar overzomerende Grauwe ganzen op Texel, gericht op het effect van aantal regulerende maatregelen op de populatieomvang, op de landbouwschade en op de schade aan enkele voor Texel belangrijke natuurwaarden. Er is gebruik gemaakt van uitgevoerde tellingen door het jaar heen en specifieke tellingen naar het aantal jonge en volwassen ganzen in het broedseizoen. Daarnaast is gebruik gemaakt van terugmeldingen aan gehalsbande exemplaren. Voor het vaststellen van effecten aan de natuur is in een experimentele aanpak een vergelijking gemaakt tussen wel en niet door ganzen gebruikte delen (exclosures), betrekking hebbend op duinplassen en matig-voedselrijke graslandvegetatie. Voor de landbouwschade is gebruik gemaakt van gegevens die door het Faunafonds worden verzameld.

Onderzoek naar wilde bestuivende insecten in appel- en perenboomgaarden in de Betuwe tussen Zaltbommel en Echteld. De achterliggende onderzoeksvragen waren: 1. Welke bijen- en zweefvliegsoorten verzamelen stuifmeel op landbouwgewassen? 2. Op welke afstanden van hun (vermoedelijke) nestelplaatsen worden deze bijensoorten nog aangetroffen op landbouwgewassen? 3. Gebruiken deze soorten in dezelfde gebieden daarnaast andere pollenbronnen? Het huidige rapport doet verslag van een onderzoek om de eerste twee vragen te beantwoorden. De derde vraag komt in een latere fase aan de orde. De keuze om appel en peren te onderzoeken was ingegeven door het grote belang van deze fruitsoorten in de Nederlandse landbouw.
