Abstract

Galaxy cluster counts in bins of mass and redshift have been shown to be a competitive probe to test cosmological models. This method requires an efficient blind detection of clusters from surveys with a well-known selection function and robust mass estimates. The Euclid wide survey will cover 15000 deg$^2$ of the sky in the optical and near-infrared bands, down to magnitude 24 in the $H$-band. The resulting data will make it possible to detect a large number of galaxy clusters spanning a wide-range of masses up to redshift $\sim 2$. This paper presents the final results of the Euclid Cluster Finder Challenge (CFC). The objective of these challenges was to select the cluster detection algorithms that best meet the requirements of the Euclid mission. The final CFC included six independent detection algorithms, based on different techniques, such as photometric redshift tomography, optimal filtering, hierarchical approach, wavelet and friend-of-friends algorithms. These algorithms were blindly applied to a mock galaxy catalog with representative Euclid-like properties. The relative performance of the algorithms was assessed by matching the resulting detections to known clusters in the simulations. Several matching procedures were tested, thus making it possible to estimate the associated systematic effects on completeness to $<3$%. All the tested algorithms are very competitive in terms of performance, with three of them reaching $>80$% completeness for a mean purity of 80% down to masses of $10^{14}$ M$_{\odot}$ and up to redshift $z=2$. Based on these results, two algorithms were selected to be implemented in the Euclid pipeline, the AMICO code, based on matched filtering, and the PZWav code, based on an adaptive wavelet approach. Abstract

The Next Generation Virgo Cluster Survey is an optical imaging survey covering 104 deg^2 centered on the Virgo cluster. Currently, the complete survey area has been observed in the u*giz-bands and one third in the r-band. We present the photometric redshift estimation for the NGVS background sources. After a dedicated data reduction, we perform accurate photometry, with special attention to precise color measurements through point spread function-homogenization. We then estimate the photometric redshifts with the Le Phare and BPZ codes. We add a new prior which extends to iAB = 12.5 mag. When using the u*griz-bands, our photometric redshifts for 15.5 \le i \lesssim 23 mag or zphot \lesssim 1 galaxies have a bias |\Delta z| < 0.02, less than 5% outliers, and a scatter \sigma_{outl.rej.} and an individual error on zphot that increase with magnitude (from 0.02 to 0.05 and from 0.03 to 0.10, respectively). When using the u*giz-bands over the same magnitude and redshift range, the lack of the r-band increases the uncertainties in the 0.3 \lesssim zphot \lesssim 0.8 range (-0.05 < \Delta z < -0.02, \sigma_{outl.rej} ~ 0.06, 10-15% outliers, and zphot.err. ~ 0.15). We also present a joint analysis of the photometric redshift accuracy as a function of redshift and magnitude. We assess the quality of our photometric redshifts by comparison to spectroscopic samples and by verifying that the angular auto- and cross-correlation function w(\theta) of the entire NGVS photometric redshift sample across redshift bins is in agreement with the expectations., Comment: Accepted for publication in ApJS. 24 pages, 21 Figures (some with degraded quality to fit the arxiv size limit), 6 Tables

Abstract

[abridged] New near-infrared surveys, using the HST, offer an unprecedented opportunity to study rest-frame optical galaxy morphologies at z>1 and to calibrate automated morphological parameters that will play a key role in classifying future massive datasets like EUCLID or LSST. We study automated parameters (e.g. CAS, Gini, M20) of massive galaxies at 11: bulge-dominated systems have systematically higher concentration and Gini coefficients and are less asymmetric and rounder than disk-dominated galaxies. However, at z>1, galaxies are, on average, 50% more asymmetric and have Gini and M20 values that are 10% higher and 20% lower respectively. In bulge-dominated galaxies, morphological parameters derived from the rest-frame UV and optical wavelengths are well correlated; however late-type galaxies exhibit higher asymmetry and clumpiness when measured in the rest-frame UV. We find that broad morphological classes (e.g. bulge vs. disk dominated) can be distinguished using parameters with high (80%) purity and completeness of 80%. In a similar vein, irregular disks and mergers can also be distinguished from bulges and regular disks with a contamination lower than 20%. However, mergers cannot be differentiated from the irregular morphological class using these parameters, due to increasingly asymmetry of non-interacting late-type galaxies at z>1. Abstract

[abridged] The mass-size relation of early-type galaxies (ETGs) has been largely studied in the last years to probe the mass assembly of the most massive objects in the Universe. In this paper, we focus on the mass-size relation of quiescent massive ETGs (Mstar/Msol > 3*10^10) living in massive clusters (M200 ~ 10^14 Mstar) at 0.8< z <1.5, as compared to those living in the field at the same epoch. Our sample contains ~ 400 ETGs in clusters and the same number in the field. Therefore, our sample is approximately an order of magnitude larger than previous studies in the same redshift range for galaxy clusters. We find that ETGs living in clusters are between ~30-50% larger than galaxies with the same stellar mass residing in the field. We parametrize the size using the mass-normalized size, gamma=Re/Mstar^0.57. The gamma distributions in both environments peak at the same position but the distributions in clusters are more skewed towards larger sizes. Since this size difference is not observed in the local Universe, the size evolution at fixed stellar mass from z~1.5 of cluster galaxies is less steep ((1+z)-0.53pm0.04) than the evolution of field galaxies ((1+z)-0.92pm0.04). The size difference seems to be essentially driven by the galaxies residing in the clusters cores (R<0.5*R200). If part of the size evolution is due to mergers, the difference we see between cluster and field galaxies could be due to higher merger rates in clusters at higher redshift, probably during the formation phase of the clusters when velocity dispersions are lower. We cannot exclude however that the difference is driven by newly quenched galaxies which are quenched more efficiently in clusters. Abstract

Aims Recently, the cardiovascular outcomes for people using anticoagulation strategies (COMPASS) trial demonstrated that dual therapy reduced cardiovascular outcomes compared with aspirin alone in patients with stable atherosclerotic disease. Methods and We sought to assess the proportion of patients eligible for the COMPASS trial and to compare the epidemiology results and outcome of these patients with those without COMPASS inclusion or with any exclusion criteria in a contemporary, nationwide cohort of patients with stable coronary artery disease. Among the 4068 patients with detailed information allowing evaluation of eligibility, 1416 (34.8%) did not fulfil the inclusion criteria (COMPASS-Not-Included), 841 (20.7%) had exclusion criteria (COMPASS-Excluded), and the remaining 1811 (44.5%) were classified as COMPASS-Like. At 1 year, the incidence of major adverse cardiovascular event (MACE), a composite of cardiovascular death, myocardial infarction, and stroke, was 0.9% in the COMPASS-Not-Included and 2.0% in the COMPASS-Like (P = 0.01), and 5.0% in the COMPASS-Excluded group (P < 0.0001 for all comparisons). Among the COMPASS-Like population, patients with multiple COMPASS enrichment criteria presented a significant increase in the risk of MACE (from 1.0% to 3.3% in those with 1 and >_3 criteria, respectively; P = 0.012), and a modest absolute increase in major bleeding risk (from 0.2% to 0.4%, respectively; P = 0.46). Conclusion In a contemporary real-world cohort registry of stable coronary artery disease, most patients resulted as eligible for the COMPASS. These patients presented a considerable annual risk of MACE that consistently increases in the presence of multiple risk factors.

Abstract

Aims Recently, the cardiovascular outcomes for people using anticoagulation strategies (COMPASS) trial demonstrated that dual therapy reduced cardiovascular outcomes compared with aspirin alone in patients with stable atherosclerotic disease. Methods and results We sought to assess the proportion of patients eligible for the COMPASS trial and to compare the epidemiology and outcome of these patients with those without COMPASS inclusion or with any exclusion criteria in a contemporary, nationwide cohort of patients with stable coronary artery disease. Among the 4068 patients with detailed information allowing evaluation of eligibility, 1416 (34.8%) did not fulfil the inclusion criteria (COMPASS-Not-Included), 841 (20.7%) had exclusion criteria (COMPASS-Excluded), and the remaining 1811 (44.5%) were classified as COMPASS-Like. At 1 year, the incidence of major adverse cardiovascular event (MACE), a composite of cardiovascular death, myocardial infarction, and stroke, was 0.9% in the COMPASS-Not-Included and 2.0% in the COMPASS-Like (P = 0.01), and 5.0% in the COMPASS-Excluded group (P Conclusion In a contemporary real-world cohort registry of stable coronary artery disease, most patients resulted as eligible for the COMPASS. These patients presented a considerable annual risk of MACE that consistently increases in the presence of multiple risk factors.

Abstract

Aims We evaluated the 1-year clinical events, pharmacological management, and quality of life in a contemporary cohort of stable coronary artery disease (CAD) patients managed by cardiologists. Methods and results START (STable Coronary Artery Diseases RegisTry) was a prospective, observational, nationwide study that enrolled 5070 stable CAD patients over 3 months in 183 cardiology centres in Italy. At 1 year, 4790 (94.5%) patients had data on vital status. Death occurred in 107 (2.2%) patients and the cause of death was cardiovascular in 41 (38.3%) of cases. Among the 4775 patients with follow-up data on clinical events available, a hospitalization due to cardiovascular and non-cardiovascular causes occurred in 523 (11.0%) and in 231 (4.8%) of cases, respectively. Over 60% of patients reported as ‘no problems’ in all domains (61.4–84.5%) of the EuroQoL quality of life 5D-5L questionnaire. Among the 3239 patients with clinical visit/telephone interview at follow-up, in whom optimal medical therapy (OMT; aspirin or thienopyridine, β-blocker, and statin) was prescribed at enrolment, 2971 (91.7%) were still receiving OMT at follow-up. At multivariable analysis, only increasing age (odds ratio 0.98; 95% confidence interval 0.97–0.99; P = 0.04) resulted as independent negative predictor of OMT persistence at 1 year from enrolment. Conclusion In this large, contemporary registry, stable CAD patients managed by cardiologists presented a high rate of clinical events at 1 year. Abstract

Euclid Collaboration; International audience; Galaxy cluster counts in bins of mass and redshift have been shown to be a competitive probe to test cosmological models. This method requires an efficient blind detection of clusters from surveys with a well-known selection function and robust mass estimates, which is particularly challenging at high redshift. The Euclid wide survey will cover 15 000 deg2 of the sky, avoiding contamination by light from our Galaxy and our solar system in the optical and near-infrared bands, down to magnitude 24 in the H-band. The resulting data will make it possible to detect a large number of galaxy clusters spanning a wide-range of masses up to redshift ∼2 and possibly higher. This paper presents the final results of the Euclid Cluster Finder Challenge (CFC), fourth in a series of similar challenges. The objective of these challenges was to select the cluster detection algorithms that best meet the requirements of the Euclid mission. The final CFC included six independent detection algorithms, based on different techniques, such as photometric redshift tomography, optimal filtering, hierarchical approach, wavelet and friend-of-friends algorithms. These algorithms were blindly applied to a mock galaxy catalog with representative Euclid-like properties. The relative performance of the algorithms was assessed by matching the resulting detections to known clusters in the simulations down to masses of M200 ∼ 1013.25 M⊙. Several matching procedures were tested, thus making it possible to estimate the associated systematic effects on completeness to < 3%. All the tested algorithms are very competitive in terms of performance, with three of them reaching > 80% completeness for a mean purity of 80% down to masses of 1014 M⊙ and up to redshift z = 2. Based on these results, two algorithms were selected to be implemented in the Euclid pipeline, the Adaptive Matched Identifier of Clustered Objects (AMICO) code, based on matched filtering, and the PZWav code, based on an adaptive wavelet approach.

Abstract

This is the third and final part of our update on the latest advances in cardiac valvular replacement. Part 1 was dedicated to cardiac valvular replacement, and Part 2 focused on transcatheter cardiac valvular treatment. This part concerns annuloplasty prostheses for valvular reconstructive surgery. The number of patients undergoing surgical heart valve repair has been increasing, particularly in high-volume centers. Annuloplasty is now considered the gold standard in mitral valve regurgitation repair secondary to degenerative, ischemic and idiopathic dilated cardiomyopathy disease. The techniques of mitral valve reconstruction have been well established, but controversies remain regarding the type of annuloplasty ring to be used. The available annuloplasty rings include rigid, flexible, complete, partial, and semi-rigid/flexible. The choice of annuloplasty ring has been the focus of extensive investigation and debate, but to date it still largely remains a matter of "surgeon's preference" rather than an evidence-based selection. Functional tricuspid regurgitation was traditionally treated by the classic De Vega annuloplasty, but has since evolved after the development of prosthetic tricuspid annuloplasty. Head-to-head comparisons have demonstrated superior long-term outcomes with device-based annuloplasty compared to suture-based surgery, but the type of ring to be used (flexible versus rigid) has recently been questioned, without reaching definitive conclusions. In contrast to mitral and tricuspid valve repair, aortic repair is more difficult with respect to specific valve features. Annuloplasty is considered to play a key role in controlling aortic regurgitation and preventing recurrence after valve repair. Various modifications of annuloplasty have been advocated (internal/external, with/without ring (suture), rigid/flexible ring). but none of them has become a de facto standard. This paper describes the various rings that are available to help orient surgeons and to serve as a reference for students.

Abstract

According to the JNC7 report, prehypertension category includes subjects with systolic blood pressure between 120 and 139 mmHg and/or diastolic blood pressure between 80 and 89 mmHg that would be at risk for developing hypertension and its untoward sequelae as myocardial infarction and cerebrovascular disease. Moreover, ambulatory blood pressure monitoring made it possible to detect subjects with masked hypertension, who are at risk of greater target organ damage than those with normal ambulatory or home blood pressure. The aim of this study was to evaluate the risk of cardiac, cerebral and vascular events in a group of prehypertensive subjects, with and without masked hypertension.We studied 204 consecutive asymptomatic prehypertensive subjects without history and signs of cardiovascular disease or diabetes. All the subjects underwent clinical evaluation, electrocardiogram, routine laboratory tests and ambulatory blood pressure monitoring. They were followed-up for a maximum of 237 months or until a cardiovascular event occurred.Twenty-seven cardiovascular events (13.2%) occurred, including 4 abdominal aortic aneurysms. Age (p0.0001), total cholesterol (p=0.004), smoking (p=0.03) and clinically overt hypertension development (p=0.011) were related to cardiovascular events. Prognosis was not related to masked hypertension.The results of this study suggest that, in subjects with prehypertension, followed for 20 years, traditional cardiovascular risk factors and development of clinically overt hypertension could be more relevant than ambulatory hypertension in the prediction of an adverse outcome.

Control of hypertension is unsatisfactory among older women. Data about Mediterranean countries are not currently reported.The aim of the present study was to describe the features of blood pressure (BP) control and the clustering of other cardiovascular (CV) risk factors in Mediterranean post-menopausal hypertensive women.We consecutively selected 516 post-menopausal female patients (mean age 69±11 years) with drug-treated essential hypertension (ESH/ESC grade 1 and 2) for this cross-sectional study. All patients were divided in 4 groups:60 years; 60-69 years; 70-79 years; ≥ 80 years.The Kruskal-Wallis analysis of variance showed a significant difference among the 4 age groups both for systolic BP (p0.001) and diastolic BP (p0.01). Mann-Whitney test for multiple comparisons of each age group vs. octogenarians demonstrated that there is a significant incremental trend of SBP through the age decades. Mean diastolic BP values were significantly higher in younger patients (age decades60 and 60-69 years, p0.01 and p0.05 respectively), while in patients aged 70-79 years there was no difference vs. octogenarians. Dyslipidemia was the more prevalent clustered risk factor with a peak rate of 49% in patients aged 60-69 years, statistically different (p0.05) from octogenarians. Global BP control (i.e. treated BP140/90 mmHg) was low (33.5% in the whole population) and there was no trend through age decades.BP control varied across age groups, but was poor. Nevertheless, the studied population appeared to be at low cardiovascular risk, due to a modest clustering of traditional risk factors.

Abstract

A multicentre, randomised, open trial with a 2 x 2 factorial design was conducted to compare the benefits and risks of two thrombolytic agents, streptokinase (SK, 1·5 MU infused intravenously over 30-60 min) and alteplase (tPA, 100 mg infused intravenously over 3 h) in patients with acute myocardial infarction admitted to coronary care units within 6 h from onset of symptoms. The patients were also randomised to receive heparin (12 500 U subcutaneously twice daily until discharge from hospital, starting 12 h after beginning the tPA or SK infusion) or usual therapy. All patients without specific contraindications were given atenolol (5-10 mg iv) and aspirin (300-325 mg a day). The end-point of the study was the combined estimate of death plus severe left ventricular damage. 12 490 patients were randomised to four treatment groups (SK alone, SK plus heparin, tPA alone, tPA plus heparin). No specific differences between the two thrombolytic agents were detected as regards the combined end-point (tPA 23·1%; SK 22·5%; relative risk 1·04, 95% Cl 0·95-1·13), nor after the addition of heparin to the aspirin treatment (hep 22·7%, no hep 22·9%; RR 0·99, 95% Cl 0·91-1·08). The outcome of patients allocated to the four treatment groups was similar with respect to baseline risk factors such as age, Killip class, hours from onset of symptoms, and site and type of infarct. The rates of major in-hospital cardiac complications (reinfarction, post-infarction angina) were also similar. The incidence of major bleeds was significantly higher in SK and heparin treated patients (respectively, tPA 0·5%, SK 1·0%, RR 0·57, 95% Cl 0·38-0·85; hep 1·0%, no hep 0·6%, RR 1·64, 95% Cl 1·09-2·45), whereas the overall incidence of stroke was similar in all groups. SK and tPA appear equally effective and safe for use in routine conditions of care, in all infarct patients who have no contraindications, with or without post-thrombolytic heparin treatment. Abstract

[abridged] New near-infrared surveys, using the HST, offer an unprecedented opportunity to study rest-frame optical galaxy morphologies at z>1 and to calibrate automated morphological parameters that will play a key role in classifying future massive datasets like EUCLID or LSST. We study automated parameters (e.g. CAS, Gini, M20) of massive galaxies at 11: bulge-dominated systems have systematically higher concentration and Gini coefficients and are less asymmetric and rounder than disk-dominated galaxies. However, at z>1, galaxies are, on average, 50% more asymmetric and have Gini and M20 values that are 10% higher and 20% lower respectively. In bulge-dominated galaxies, morphological parameters derived from the rest-frame UV and optical wavelengths are well correlated; however late-type galaxies exhibit higher asymmetry and clumpiness when measured in the rest-frame UV. We find that broad morphological classes (e.g. bulge vs. disk dominated) can be distinguished using parameters with high (80%) purity and completeness of 80%. In a similar vein, irregular disks and mergers can also be distinguished from bulges and regular disks with a contamination lower than 20%. However, mergers cannot be differentiated from the irregular morphological class using these parameters, due to increasingly asymmetry of non-interacting late-type galaxies at z>1. Abstract

Duchenne muscular dystrophy (DMD) is a severe X-linked neuromuscular childhood disorder that causes progressive muscle weakness and degeneration. A lack of dystrophin in DMD leads to inflammatory response, autophagic dysregulation, and oxidative stress in skeletal muscle fibers that play a key role in the progression of the pathology. β-glucans can modulate immune function by modifying the phagocytic activity of immunocompetent cells, notably macrophages. Mitochondrial function is also involved in an important mechanism of the innate and adaptive immune responses, owing to high need for energy of immune cells. In the present study, the effects of 1,3-1,6 β-glucans on five-day-old non-dystrophic and dystrophic (sapje) zebrafish larvae were investigated. The effects of the sonication of β-glucans and the dechorionation of embryos were also evaluated. The results showed that the incidence of dystrophic phenotypes was reduced when dystrophic embryos were exposed to 2 and 4 mg L−1of 1,3-1,6 β-glucans. Moreover, when the dystrophic larvae underwent 8 mg L−1treatment, an improvement of the locomotor performances and mitochondrial respiration were observed. In conclusion, the observed results demonstrated that 1,3-1,6 β-glucans improve locomotor performances and mitochondrial function in dystrophic zebrafish. Therefore, for ameliorating their life quality, 1,3-1,6 β-glucans look like a promising diet supplement for DMD patients, even though further investigations are required.

Abstract

International audience; The NGVS-IR project (Next Generation Virgo Cluster Survey-Infrared) is a contiguous, near-infrared imaging survey of the Virgo cluster of galaxies. It complements the optical wide-field survey of Virgo (NGVS). In its current state, NGVS-IR consists of K-s-band imaging of 4 deg(2) centered on M87 and J- and K-s-band imaging of similar to 16 deg(2) covering the region between M49 and M87. We present observations of the central 4 deg(2) centered on Virgo's core region. The data were acquired with WIRCam on the Canada-France-Hawaii Telescope, and the total integration time was 41 hr distributed over 34 contiguous tiles. A survey-specific strategy was designed to account for extended galaxies while still measuring accurate sky brightness within the survey area. The average 5s limiting magnitude is K-s = 24.4 AB mag, and the 50% completeness limit is K-s = 23.75 AB mag for point-source detections, when using only images with better than 0.” 7 seeing (median seeing 0.” 54). Star clusters are marginally resolved in these image stacks, and Virgo galaxies with mu(Ks) similar or equal to 24.4 AB mag arcsec(-2) are detected. Combining the K-s data with optical and ultraviolet data, we build the uiK(s) color-color diagram, which allows a very clean color-based selection of globular clusters in Virgo. This diagnostic plot will provide reliable globular cluster candidates for spectroscopic follow-up campaigns, needed to continue the exploration of Virgo's photometric and kinematic substructures, and will help the design of future searches for globular clusters in extragalactic systems. We show that the new uiK(s) diagram displays significantly clearer substructure in the distribution of stars, globular clusters, and galaxies than the gzK(s) diagram-the NGVS + NGVS-IR equivalent of the BzK diagram that is widely used in cosmological surveys. Equipped with this powerful new tool, future NGVS-IR investigations based on the uiK(s) diagram will address the mapping and analysis of extended structures and compact stellar systems in and around Virgo galaxies.

31. Modeling sacsin depletion in Danio Rerio offers new insight on retinal defects in ARSACS.

Abstract

Biallelic mutations in the SACS gene, encoding sacsin, cause early-onset autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS), a neurodegenerative disease also characterized by unique and poorly understood retinal abnormalities. While two murine models replicate the phenotypic and neuronal features observed in patients, no retinal phenotype has been described so far. In a zebrafish knock-out strain that faithfully mirrors the main aspects of ARSACS, we observed impaired visual function due to photoreceptor degeneration, likely caused by cell cycle defects in progenitor cells. RNA-seq analysis in embryos revealed dysfunction in proteins related to fat-soluble vitamins (e.g., TTPA, RDH5, VKORC) and suggested a key role of neuroinflammation in driving the retinal defects. Abstract

CLN8 and other neuronal ceroid lipofuscinoses (NCLs) often lead to cognitive decline, emotional disturbances, and social deficits, worsening with disease progression. Disrupted lysosomal pH, impaired autophagy, and defective dendritic arborization contribute to these symptoms. Using a cln8 -/- zebrafish model, we identified significant impairments in locomotion, anxiety, and aggression, along with subtle deficits in social interactions, positioning zebrafish as a useful model for therapeutic studies in NCL. Our findings show that trehalose, an autophagy enhancer, ameliorates anxiety, and modestly improves social behavior and predator avoidance in mutant zebrafish. This finding aligns animal models with clinical reports suggestive of behavioral improvements in NCL patients. Trehalose holds promise as a therapeutic agent for CLN8, warranting further research into its neuroprotective mechanisms and clinical applications.

33. Dapagliflozin ameliorates Lafora disease phenotype in a zebrafish model.

Abstract

Lafora disease (LD) is an ultra-rare and still incurable neurodegenerative condition. Although several therapeutic strategies are being explored, including gene therapy, there are currently no treatments that can alleviate the course of the disease and slow its progression. Recently, gliflozins, a series of SGLT2 transporter inhibitors approved for use in type 2 diabetes mellitus, heart failure and chronic kidney disease, have been proposed as possible repositioning drugs for the treatment of LD. With this in mind, we tested dapagliflozin (50 µM), canagliflozin (2.5 µM) and empagliflozin (200 µM) in our epm2a -/- zebrafish model, investigating their effects on pathological behaviour. In the case of dapagliflozin, we also investigated the possible mechanisms of action. Overall, the gliflozins reduced or rescued neuronal hyperexcitability and locomotor impairment. Dapagliflozin also reduced spontaneous seizure-like events in epm2a -/- larvae. At the biochemical and molecular level, dapagliflozin was found to slightly reduce glycogen content, and suppress inflammation and oxidative stress. It also ameliorates autophagic homeostasis and improves lysosomal markers. In conclusion, our preclinical study showed that dapagliflozin was able to ameliorate part of the pathological phenotype of epm2a -/- zebrafish larvae and could potentially be a suitable drug for repurposing in LD. Abstract

Autism spectrum disorder (ASD) is a multifactorial neurodevelopmental condition with several identified risk factors, both genetic and non-genetic. Among these, prenatal exposure to valproic acid (VPA) has been extensively associated with the development of the disorder. The zebrafish, a cost- and time-effective model, is useful for studying ASD features. Using validated VPA-induced ASD zebrafish models, we aimed to provide new insights into VPA exposure effects during embryonic development and to identify new potential biomarkers associated with ASD-like features. Dose-response analyses were performed in vivo to study larval phenotypes and mechanisms underlying neuroinflammation, mitochondrial dysfunction, oxidative stress, microglial cell status, and motor behaviour. Wild-type and transgenic Tg(mpeg1:EGFP) zebrafish were water-exposed to VPA doses (5 to 500 µM) from 6 to 120 h post-fertilisation (hpf). Embryos and larvae were monitored daily to assess survival and hatching rates, and numerous analyses and tests were conducted from 24 to 120 hpf. VPA doses higher than 50 µM worsened survival and hatching rates, while doses of 25 µM or more altered morphology, microglial status, and larval behaviours. VPA 50 µM also affected mRNA expression of inflammatory cytokines and neurogenesis-related genes, mitochondrial respiration, and reactive oxygen species accumulation. The study confirmed that VPA alters brain homeostasis, synaptic interconnections, and neurogenesis-related signalling pathways, contributing to ASD aetiopathogenesis. Further studies are essential to identify novel ASD biomarkers for developing new drug targets and tailored therapeutic interventions for ASD.

Abstract

CLN8 is an endoplasmic reticulum cargo receptor and a regulator of lysosome biogenesis whose loss of function leads to neuronal ceroid lipofuscinosis. CLN8 has been linked to autophagy and lipid metabolism, but much remains to be learned, and there are no therapies acting on the molecular signatures in this disorder. The present study aims to characterize the molecular pathways involved in CLN8 disease and, by pinpointing altered ones, to identify potential therapies. To bridge the gap between cell and mammalian models, we generated a new zebrafish model of CLN8 deficiency, which recapitulates the pathological features of the disease. We observed, for the first time, that CLN8 dysfunction impairs autophagy. Using autophagy modulators, we showed that trehalose and SG2 are able to attenuate the pathological phenotype in mutant larvae, confirming autophagy impairment as a secondary event in disease progression. 36. Zebrafish Feed Intake: A Systematic Review for Standardizing Feeding Management in Laboratory Conditions.

Abstract

Zebrafish are one of the most used animal models in biological research and a cost-effective alternative to rodents. Despite this, nutritional requirements and standardized feeding protocols have not yet been established for this species. This is important to avoid nutritional effects on experimental outcomes, and especially when zebrafish models are used in preclinical studies, as many diseases have nutritional confounding factors. A key aspect of zebrafish nutrition is related to feed intake, the amount of feed ingested by each fish daily. With the goal of standardizing feeding protocols among the zebrafish community, this paper systematically reviews the available data from 73 studies on zebrafish feed intake, feeding regimes (levels), and diet composition. Great variability was observed regarding diet composition, especially regarding crude protein (mean 44.98 ± 9.87%) and lipid content (9.91 ± 5.40%). Interestingly, the gross energy levels of the zebrafish diets were similar across the reviewed studies (20.39 ± 2.10 kilojoules/g of feed). In most of the reviewed papers, fish received a predetermined quantity of feed (feed supplied). The authors fed the fish according to the voluntary intake and then calculated feed intake (FI) in only 17 papers. From a quantitative point of view, FI was higher than when a fixed quantity (pre-defined) of feed was supplied. Also, the literature showed that many biotic and abiotic factors may affect zebrafish FI. Finally, based on the FI data gathered from the literature, a new feeding protocol is proposed. In summary, a daily feeding rate of 9-10% of body weight is proposed for larvae, whereas these values are equal to 6-8% for juveniles and 5% for adults when a dry feed with a proper protein and energy content is used.

Abstract

Milk oligosaccharides are a complex class of carbohydrates that act as bioactive factors in numerous defensive and physiological functions, including brain development. Early nutrition can modulate nervous system development and can lead to epigenetic imprinting. We attempted to increase the sialylated oligosaccharide content of zebrafish yolk reserves, with the aim of evaluating any short-term effects of the treatment on mortality, locomotor behavior, and gene expression. Wild-type embryos were microinjected with saline solution or solutions containing sialylated milk oligosaccharides extracted from human and bovine milk. The results suggest that burst activity and larval survival rates were unaffected by the treatments. Locomotion parameters were found to be similar during the light phase between control and treated larvae; in the dark, however, milk oligosaccharide-treated larvae showed increased test plate exploration. Thigmotaxis results did not reveal significant differences in either the light or the dark conditions. The RNA-seq analysis indicated that both treatments exert an antioxidant effect in developing fish. Moreover, sialylated human milk oligosaccharides seemed to increase the expression of genes related to cell cycle control and chromosomal replication, while bovine-derived oligosaccharides caused an increase in the expression of genes involved in synaptogenesis and neuronal signaling. These data shed some light on this poorly explored research field, showing that both human and bovine oligosaccharides support brain proliferation and maturation.

38. Dietary Supplementation with a Blend of Hydrolyzable and Condensed Tannins Ameliorates Diet-Induced Intestinal Inflammation in Zebrafish ( Danio rerio ).

Abstract

The current study evaluated the effects of hydrolyzable and condensed tannins from chestnut and quebracho wood, respectively (TSP, Silvafeed ® ), on zebrafish with intestinal inflammation induced by a plant-based diet (basal diet). Four experimental diets were prepared as follows: the basal diet + 0 TSP, the basal diet + TSP at 0.9 g/kg of feed, the basal diet + TSP at 1.7 g/kg of feed, and the basal diet + TSP at 3.4 g/kg of feed. Eighty-four zebrafish ( Danio rerio ) were fed for 12 days with the experimental diets. In zebrafish fed the basal diet, intestine integrity appeared to be altered, with damaged intestinal villi, high immunoexpression of tumor necrosis factor-α (TNFα) and cyclooxygenase 2 (COX2), and high expression of the cox2 , interleukin 1 (il-1b ), interleukin 8 ( cxcl8-l1 ), and tnfα genes. The tannin treatment partially restored intestinal morphology and downregulated the expression of cytokines. The best activity was detected with 1.7 and 3.4 g/kg of feed. In the guts of all groups, Proteobacteria, Fusobacteria, Firmicutes, and Bacteroidetes were the most represented phyla. The most represented genera were Plesiomonas and Sphingomonas , belonging to the Proteobacteria phylum; Cetobacterium , belonging to the Fusobacteria phylum; and Lactobacillus , belonging to the Firmicutes phylum. No significant differences were detected among groups, except for a slight decrease in the Fusobacteria phylum and slight increases in the Shewanella and Bacteroides genera with TSP. 39. A Review on the Bioactivity of Cannabinoids on Zebrafish Models: Emphasis on Neurodevelopment.

Abstract

For centuries, the cannabis plant has been used as a source of food, fiber, and medicine. Recently, scientific interest in cannabis has increased considerably, as its bioactive compounds have shown promising potential in the treatment of numerous musculoskeletal and neurological diseases in humans. However, the mechanisms that underlie its possible effects on neurodevelopment and nervous-system functioning remain poorly understood and need to be further investigated. Although the bulk of research on cannabis and cannabinoids is based on in vitro or rodent models, the zebrafish has now emerged as a powerful in vivo model for drug-screening studies and translational research. We here review the available literature on the use of cannabis/cannabinoids in zebrafish, and particularly in zebrafish models of neurological disorders. A critical analysis suggests that zebrafish could serve as an experimental tool for testing the bioactivity of cannabinoids, and they could thus provide important insights into the safety and efficacy of different cannabis-extract-based products. The review showed that zebrafish exhibit similar behaviors to rodents following cannabinoid exposure. Abstract

Mutations in the EPM2A gene encoding laforin cause Lafora disease (LD), a progressive myoclonic epilepsy characterized by drug-resistant seizures and progressive neurological impairment. To date, rodents are the only available models for studying LD; however, their use for drug screening is limited by regulatory restrictions and high breeding costs. To investigate the role of laforin loss of function in early neurodevelopment, and to screen for possible new compounds for treating the disorder, we developed a zebrafish model of LD. Our results showed the epm2a -/- zebrafish to be a faithful model of LD, exhibiting the main disease features, namely motor impairment and neuronal hyperexcitability with spontaneous seizures. The model also showed increased inflammatory response and apoptotic death, as well as an altered autophagy pathway that occurs early in development and likely contributes to the disease progression. Early administration of trehalose was found to be effective for rescuing motor impairment and neuronal hyperexcitability associated with seizures. Our study adds a new tool for investigating LD and might help to identify new treatment opportunities.

41. In Vivo Evaluation of Cannabis sativa Full Extract on Zebrafish Larvae Development, Locomotion Behavior and Gene Expression.

Abstract

Historically, humans have been using Cannabis sativa for both recreational and medical purposes. Nowadays, cannabis-based products have gained scientific interest due to their beneficial effects on several syndromes and illnesses. The biological activity of cannabinoids is essentially due to the interaction with the endocannabinoid system, and zebrafish ( Danio rerio ) is a very well-known and powerful in vivo model for studying such specific interactions. The aim of the study was to investigate the effects of different doses of a Cannabis sativa whole extract [dissolved in dimethyl sulfoxide (DMSO)] on zebrafish eggs' hatchability, embryo post-hatching survival, larvae locomotion behavior and mRNA gene expression. The results showed the absence of toxicity, and no significant differences were observed between treatments for both embryo hatching and survival rate. In addition, larvae exposed to the cannabis extract at the highest dose [containing 1.73 nM and 22.3 nM of ∆ 9 -tetrahydrocannabinol (THC) and cannabidiol (CBD), respectively] showed an increased locomotion compared to the control and DMSO treated groups. Moreover, qRT-PCR analysis showed that the highest dosage of cannabis induced an over-expression of cnr1 and cnr2 cannabinoid receptors. In conclusion, the exposition of zebrafish larvae to the whole extract of Cannabis sativa showed no negative effects on embryo development and survival and enhanced the larvae's locomotor performances. These findings may open up possible Cannabis sativa applications in human pharmacology as well as in other animal sectors.

Abstract

Autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS) is a multisystem hereditary ataxia associated with mutations in SACS , which encodes sacsin, a protein of still only partially understood function. Although mouse models of ARSACS mimic largely the disease progression seen in humans, their use in the validation of effective therapies has not yet been proposed. Recently, the teleost Danio rerio has attracted increasing attention as a vertebrate model that allows rapid and economical screening, of candidate molecules, and thus combines the advantages of whole-organism phenotypic assays and in vitro high-throughput screening assays. Through CRISPR/Cas9-based mutagenesis, we generated and characterized a zebrafish sacs -null mutant line that replicates the main features of ARSACS. The sacs -null fish showed motor impairment, hindbrain atrophy, mitochondrial dysfunction, and reactive oxygen species accumulation. As proof of principle for using these mutant fish in high-throughput screening studies, we showed that both acetyl-DL-leucine and tauroursodeoxycholic acid improved locomotor and biochemical phenotypes in sacs -/- larvae treated with these neuroprotective agents, by mediating significant rescue of the molecular functions altered by sacsin loss. Taken together, the evidence here reported shows the zebrafish to be a valuable model organism for the identification of novel molecular mechanisms and for efficient and rapid in vivo optimization and screening of potential therapeutic compounds. Abstract

Duchenne muscular dystrophy (DMD) is a severe X-linked neuromuscular childhood disorder that causes progressive muscle weakness and degeneration. A lack of dystrophin in DMD leads to inflammatory response, autophagic dysregulation, and oxidative stress in skeletal muscle fibers that play a key role in the progression of the pathology. β-glucans can modulate immune function by modifying the phagocytic activity of immunocompetent cells, notably macrophages. Mitochondrial function is also involved in an important mechanism of the innate and adaptive immune responses, owing to high need for energy of immune cells. In the present study, the effects of 1,3-1,6 β-glucans on five-day-old non-dystrophic and dystrophic ( sapje ) zebrafish larvae were investigated. The effects of the sonication of β-glucans and the dechorionation of embryos were also evaluated. The results showed that the incidence of dystrophic phenotypes was reduced when dystrophic embryos were exposed to 2 and 4 mg L -1 of 1,3-1,6 β-glucans. Moreover, when the dystrophic larvae underwent 8 mg L -1 treatment, an improvement of the locomotor performances and mitochondrial respiration were observed. In conclusion, the observed results demonstrated that 1,3-1,6 β-glucans improve locomotor performances and mitochondrial function in dystrophic zebrafish. Therefore, for ameliorating their life quality, 1,3-1,6 β-glucans look like a promising diet supplement for DMD patients, even though further investigations are required.

Abstract

Duchenne muscular dystrophy (DMD), caused by mutations in the dystrophin gene, is an inherited neuromuscular disorder that causes loss of muscle mass and motor skills. In the era of genomic medicine, there is still no known cure for DMD. In clinical practice, there is a growing awareness of the possible importance of nutrition in neuromuscular diseases. This is mostly the result of patients' or caregivers' empirical reports of how active substances derived from food have led to improved muscle strength and, thus, better quality of life. In this report, we investigate several nutraceutical principles in the sapje strain of zebrafish, a validated model of DMD, in order to identify possible natural products that, if supplemented in the diet, might improve the quality of life of DMD patients. Gingerol, a constituent of fresh ginger, statistically increased the locomotion of mutant larvae and upregulated the expression of heme oxygenase 1, a target gene for therapy aimed at improving dystrophic symptoms. Although three other compounds showed a partial positive effect on locomotor and muscle structure phenotypes, our nutraceutical screening study lent preliminary support to the efficacy and safety only of gingerol. Gingerol could easily be proposed as a dietary supplement in DMD.

45. Social Preference Tests in Zebrafish: A Systematic Review.

Abstract

The use of animal models in biology research continues to be necessary for the development of new technologies and medicines, and therefore crucial for enhancing human and animal health. In this context, the need to ensure the compliance of research with the principles Replacement, Reduction and Refinement (the 3 Rs), which underpin the ethical and human approach to husbandry and experimental design, has become a central issue. The zebrafish ( Danio rerio ) is becoming a widely used model in the field of behavioral neuroscience. In particular, studying zebrafish social preference, by observing how an individual fish interacts with conspecifics, may offer insights into several neuropsychiatric and neurodevelopmental disorders. The main aim of this review is to summarize principal factors affecting zebrafish behavior during social preference tests. We identified three categories of social research using zebrafish: studies carried out in untreated wild-type zebrafish, in pharmacologically treated wild-type zebrafish, and in genetically engineered fish. We suggest guidelines for standardizing social preference testing in the zebrafish model. The main advances gleaned from zebrafish social behavior testing are discussed, together with the relevance of this method to scientific research, including the study of behavioral disorders in humans. The authors stress the importance of adopting an ethical approach that considers the welfare of animals involved in experimental procedures. 46. Short communication: Technological and seasonal variations of vitamin D and other nutritional components in donkey milk.

Abstract

Vitamin D is an essential nutrient that plays a crucial role in calcium homeostasis and bone metabolism and also acts as a hormone. Although several studies on the content of vitamin D in bovine milk have been conducted, little information is available regarding donkey milk. In the context of the nutritional assessment of donkey milk, the aim of this study was to assess the vitamin D content in donkey milk and its chemical profile, with particular reference to seasonal and technological modifications after pasteurization. The study was conducted on a dairy farm that produces donkey milk for human consumption located in central Italy. At sampling time, an aliquot of total bulk milk production was sampled before and after pasteurization (63°C for 30 min without homogenization) with a total of 20 raw and 20 pasteurized milk samples. The samples were collected for 10 mo, every 15 d, from May to February 2017. All the samples were analyzed for the chemical composition and vitamin D 2 and D 3 content by HPLC after saponification. The donkey milk analyzed showed a higher average vitamin D content (raw milk: vitamin D 2 = 1.68, vitamin D 3 = 0.60 μg/100 mL; pasteurized milk: vitamin D 2 = 1.38, vitamin D 3 = 0.30 μg/100 mL) than reported for bovine and human milk. The results of the effect of pasteurization on milk did not highlight significant differences in the total content of vitamin D. However, vitamin D 3 has a poor thermal stability, which led to a significant reduction in content in pasteurized milk compared with raw milk. The total vitamin D content of donkey milk did not show significant variations between seasons; however, a higher concentration of vitamin D 3 was found in spring and summer. In conclusion, raw and pasteurized donkey milk showed a high content of vitamin D, which could be useful in meeting the deficiencies of this vitamin in humans. Further investigations are needed to improve the vitamin D content in donkey milk by increasing its endogenous synthesis or its transfer in milk and to clarify other variability factors., (Copyright © 2018 American Dairy Science Association.
