Long noncoding RNAs (lncRNAs) account for the largest portion of RNA from the transcriptome, yet most of their functions remain unknown. Here, we performed two independent high-throughput CRISPRi screens to understand the role of lncRNAs in monocyte function and differentiation. The first was a reporter-based screen to identify lncRNAs that regulate TLR4-NFkB signaling in human monocytes and the second screen identified lncRNAs involved in monocyte to macrophage differentiation. We successfully identified numerous noncoding and protein-coding genes that can positively or negatively regulate inflammation and differentiation. To understand the functional roles of lncRNAs in both processes, we chose to further study the lncRNA LOUP [lncRNA originating from upstream regulatory element of SPI1 (also known as PU.1)], as it emerged as a top hit in both screens. Not only does LOUP regulate its neighboring gene, the myeloid fate-determining factor SPI1, thereby affecting monocyte to macrophage differentiation, but knockdown of LOUP leads to a broad upregulation of NFkB-targeted genes at baseline and upon TLR4-NFkB activation. LOUP also harbors three small open reading frames capable of being translated and are responsible for LOUPs ability to negatively regulate TLR4/NFkB signaling. This work emphasizes the value of high-throughput screening to rapidly identify functional lncRNAs in the innate immune system.

Background and Aims: This investigation aims to conduct a comprehensive review of educational interventions targeting stroke prevention to provide effective preventive measures and optimize resource utilization in adults and the elderly populations. Methods: A comprehensive literature search was conducted on PubMed, SCOPUS, and Embase for articles published online or in print until February 22, 2022. Inclusion criteria for studies were limited to the studies that examined stroke education or training interventions aimed at improving knowledge among adults aged 30 years and above, with a particular focus on older adults. Results: A review of 97,848 papers was conducted, resulting in the inclusion of 19 papers. Of these, six were randomized controlled trials (RCTs), six were non‐randomized studies, five were campaign studies, one was a cross‐sectional study, one was a pilot study, and one was a prospective study. The provided information describes various interventions and educational programs related to stroke awareness, prevention, and management. The intervention subjects were categorized as awareness of warning signs and symptoms of stroke (n = 14), comprehensive awareness campaigns (n = 5), multilevel strategies for stroke education (n = 4), community‐based nursing education and rehabilitation program (n = 5), multimedia campaign for 9‐1‐1 awareness (n = 3), and self‐management interventions (n = 1). Conclusion: The categorized interventions, addressing awareness of warning signs and symptoms, comprehensive awareness campaigns, multilevel strategies, community‐based nursing education and rehabilitation programs, multimedia campaigns for 9‐1‐1 awareness, and self‐management interventions, collectively enrich our understanding of the multifaceted approaches to stroke education. Background: MicroRNAs (miRNAs) regulate gene expression and various cellular activities. They also hold significant importance in the progression and development of human malignancies. Among these, miRNA-484 and the Fis-1 gene have been identified as having substantial roles in lung cancer. This study aims to ascertain miRNA-484 and Fis-1 gene expression levels in non-small cell lung cancer (NSCLC) patients. Method: In this case-control study, 45 pairs of tumor tissues and their corresponding healthy margin tissues were surgically obtained from NSCLC patients and promptly preserved in liquid nitrogen after excision. Total RNA extraction was performed using TRIzol, followed by cDNA synthesis using a designated kit. Afterward, we used quantitative reverse transcription polymerase chain reaction (qRT-PCR) to measure the expression levels of miRNA-484 and the Fis-1 gene. Furthermore, the clinico-pathological characteristics of the NSCLC patients were assessed. Results: Our findings revealed an upregulation of miRNA-484 expression and downregulation of Fis-1 gene expression in NSCLC tissues compared with non-tumor tissues. Additionally, significant correlations were observed between miRNA-484 and Fis-1 gene expression levels and clinicopathological features of the patients, including factors such as lymph node involvement and distant metastasis. Conclusion: These findings suggest the potential utility of Fis-1 and miR-484 as prognostic and diagnostic markers in NSCLC. [ABSTRACT FROM AUTHOR]
