2. Economic and engineering aspects of disposables-based bioprocessing

Biopharmaceutical companies face a fast moving competitive market with high product failure rates. Disposables-based bioprocessing meets some of these current pressures and concerns by employing single-use, pre-sterilised, pre-validated components instead of traditional stainless-steel fixed equipment. The advantages include increased flexibility, smaller initial investments and potential reduction of time to market. This thesis provides an engineering study of the use of disposable equipment as an alternative to conventional systems. A costing framework was developed to compare disposables-based and conventional plants. The use of disposable equipment was shown to result in a 70% increase in running costs, substantially offset by a 40% reduction in the capital investment required. The production of a Fab' antibody fragment from an E. coli fermentation was used as the illustrative case study. Sensitivity analysis to different variables was made to confirm the results. The study showed a loss in yield in different unit operations in the disposable process could be compensated for by a reduction of the materials costs. It was also predicted that the use of disposables could reduce time to market by up to 1.5 years. The running costs associated with the single use of microfiltration membranes were shown to have a high impact on the overall cost indicating that minimisation of membrane area was crucial. Experimental work focused on this unit operation, aimed at controlling transmission. Transmission was shown to decrease rapidly and the causes for this decrease were explained and modelled. A strategy that maintains % transmission at high values was developed and evaluated theoretically and experimentally. The method comprises short intermediate rinsing steps, capable of restoring the membrane properties. The resulting reduced filtration areas were shown to enhance further the economic attractiveness of the disposable approach. Overall disposables-based bioprocessing was shown to be economically and technologically competitive with conventional engineering approaches.

Relatório de Estágio do Mestrado em Ensino de História no 3º ciclo do Ensino Básico e no Ensino Secundário apresentado à Faculdade de Letras O presente relatório de estágio tem dois objetivos, sendo o primeiro, dar a conhecer as várias etapas percorridas durante o estágio pedagógico, inserido no Mestrado em Ensino de História no 3º Ciclo do Ensino Básico e no Ensino Secundário, e, o segundo, através de um projeto de estudo, responder à questão de investigação: Qual a importância do conhecimento tácito para o processo de aprendizagem em História? O estudo foi desenvolvido com os alunos das três turmas do 7º ano e com uma turma do 9º ano, na Escola E. B. 2, 3 Carlos de Oliveira, durante o estágio pedagógico realizado no ano letivo de 2019/2020. Como referido acima, este estudo teve como principal objetivo responder à questão de investigação. De forma a responder a esta questão foram elaborados três objetivos: Identificar as ideias tácitas dos alunos sobre as temáticas exploradas no programa do 7º ano de escolaridade, verificar a importância das ideias tácitas substantivas para o desenvolvimento do conhecimento histórico e desenhar experiências de aprendizagem que promovam a utilização e/ou desconstrução das ideias tácitas no processo de aprendizagem da História. Para alcançar estes objetivos foi feita uma recolha de dados através de questionários que foram distribuídos pelas turmas em estudo. Como estratégia metodológica foi utilizada a Design Based Research (DBR) por permitir o desenvolvimento de teorias auxiliares à compreensão da forma como os alunos aprendem. Através dos resultados obtidos, podemos concluir que os alunos levam para a escola ideias tácitas concebidas a partir do meio social e cultural em que estão inseridos. Havendo a forte possibilidade de que estas ideias influenciam o processo de aprendizagem, fazendo com os alunos mesmo após estudarem uma determinada temática continuem com ideias destorcidas em relação à mesma. Foi assim pensada uma estratégia pedagógica para que os alunos de uma forma mais independente façam o percurso pela aprendizagem confrontando as suas ideias com a informação nova adquirida através de imagens, documentos e vídeos, seguindo um guião de pesquisa fornecido pelo professor. Tendo em conta a pandemia vivida (COVID-19) e aproveitando o ensino à distância inserindo mais os meios digitais a favor da aprendizagem. The present internship report as two objectives, the first one is letting know the various steps taken during the pedagogical internship, inserted in the Masters in History Teaching in the 3rd Cycle of Basic Education and Secondary Education, and, the second, through a study project, answer the research question: What is the importance of tacit knowledge for the learning process in History? This study was developed with students from the three classes of the 7th grade and with a class of the 9th grade, at ‘Carlos Oliveira’ Basic School, during the pedagogical internship held in the school year 2019/2020. As mention above, this study had as main objective to answer the research question. In order to answer this question, three objectives were elaborated: Identify the student’s tacit ideas about the themes explored in the 7th grade school program, verify the importance of substantive tacit ideas for the development of historical knowledge and design learning experiences that promote the use and/or deconstruction of tacit ideas in the process of History learning. To achieve these objectives, was made a data collection through questionnaires that were distributed to the groups under study. As a methodological strategy was used the Design Based Research (DBR) because it allows the development of auxiliar theories to understand the way how the students learn. Through the results obtained, we can conclude that students take to school tacit ideas conceived from the social and cultural environment in which they are inserted. There is a strong possibility that these ideas influence the learning process, making students even after studying a certain topic continue with ideas distorted in relation to it. So, a pedagogical strategy was devised so that students, in a more independent way, make the journey through learning, confronting their ideas with the new information acquires through images, documents and videos, following a research guide provided by the teacher. Considering the pandemic experienced (COVID-19) and taking advantage of distance learning by inserting more digital media in favour of learning.

Diamond Open Access - Abstract
Science Europe, cOAlition S, OPERAS, and the French National Research Agency (ANR) present an Action Plan for Diamond Open Access to further develop and expand a sustainable, community-driven Diamond OA scholarly communication ecosystem. It focuses on efficiency, quality standards, capacity building, and sustainability, and it addresses the alignment and development of common resources for the whole Diamond OA ecosystem, including journals and platforms, while respecting the cultural, multilingual, and disciplinary diversity that constitutes the strength of the sector. The Action Plan intends to create an inclusive worldwide community that has the tools to strengthen existing Diamond OA journals and platforms and increase their visibility.
