Background: Tsetse-transmitted African trypanosomiasis is a debilitating and fatal disease of humans and livestock if left untreated. While knowledge of the spatial distribution patterns of tsetse is essential for the development of risk-based vector control strategies, existing distribution maps in Zambia are more than 40 years old and were based on coarse spatial resolution data. The recently developed vehicle-mounted sticky trap (VST) provides an alternative sampling device to aid in updating existing distribution maps but has not been applied outside an experimental setting and is limited to motorable tracks. Therefore, the objective of the present study was to demonstrate the effectiveness of utilizing the VST for area-wide surveys of Glossina morsitans and to use the occurrence records to predict its spatial distribution in Zambia under current environmental conditions using Maxent. Methodology/Principal findings: Two-sided all-blue VST baited with butanone and 1-octen-3-ol was used to survey 692 and 1020 km of transect routes in G. m. centralis Machado and G. m. morsitans Westwood previously published distribution in Zambia. Maxent species distribution technique was used to predict the potential distribution of the two subspecies using current climatic and environmental data which was then compared to the historical distribution. A total of 15,602 tsetse were captured with G. m. morsitans (58%) being the most abundant. G. m. centralis and G. pallidipes Austin represented 39 and 2% of the catch respectively, and G. brevipalpis Newstead was also detected. The predicted potential distribution for G. m. centralis was 80,863 km2 while that of G. m. morsitans was 70,490 km2 representing a 47 and 29% reduction compared to their historical distributions, respectively. Conclusion/Significance: The VST is effective for sampling G. morsitans outside experimental settings and is recommended for use as an additional tsetse survey tool. The spatial distribution of G. morsitans in Zambia has reduced by 101,051 km2 due to temperature and land cover changes. Mapping tsetse occurrence is therefore a useful predictor of disease risk and provides the basis of evidence-based vector control strategies. In this study, we demonstrate the suitability of using the recently developed vehicle-mounted sticky trap for the rapid generation of occurrence records that can be used in ecological niche models to predict the distribution of G. morsitans. Across an elevation gradient, several biotic and abiotic factors influence community assemblages of interacting species leading to a shift in species distribution, functioning, and ultimately topologies of species interaction networks. However, empirical studies of climate‐driven seasonal and elevational changes in plant‐pollinator networks are rare, particularly in tropical ecosystems. Eastern Afromontane Biodiversity Hotspots in Kenya, East Africa. We recorded plant‐bee interactions at 50 study sites between 515 and 2600 m asl for a full year, following all four major seasons in this region. We analysed elevational and seasonal network patterns using generalised additive models (GAMs) and quantified the influence of climate, floral resource availability, and bee diversity on network structures using a multimodel inference framework. We recorded 16,741 interactions among 186 bee and 314 plant species of which a majority involved interactions with honeybees. We found that nestedness and bee species specialisation of plant‐bee interaction networks increased with elevation and that the relationships were consistent in the cold‐dry and warm‐wet seasons respectively. Link rewiring increased in the warm‐wet season with elevation but remained indifferent in the cold‐dry seasons. Conversely, network modularity and plant species were more specialised at lower elevations during both the cold‐dry and warm‐wet seasons, with higher values observed during the warm‐wet seasons. We found flower and bee species diversity and abundance rather than direct effects of climate variables to best predict modularity, specialisation, and link rewiring in plant‐bee‐interaction networks. This study highlights changes in network architectures with elevation suggesting a potential sensitivity of plant‐bee interactions with climate warming and changes in rainfall patterns along the elevation gradients of the Eastern Afromontane Biodiversity Hotspot. Background: Black screen fly round (BFR) is a mobile sampling method for Glossina morsitans. This technique relies on the ability of operator(s) to capture flies landing on the screen with hand nets. In this study, we aimed to evaluate a vehicle-mounted sticky panel trap (VST) that is independent of the operator's ability to capture flies against BFR, for effective and rapid sampling of G. m. morsitans Westwood and G. m. centralis Machado. We also determined the influence of the VST colour (all-blue, all-black or 1:1 blue-black), orientation and presence of odour attractants on tsetse catch. Methodology/Principal findings: Using randomised block design experiments conducted in Zambia, we compared and modelled the number of tsetse flies caught in the treatment arms using negative binomial regression. There were no significant differences in the catch indices of the three colour designs and for in-line or transversely oriented panels for both subspecies (P > 0.05). When baited with butanone and 1-octen-3-ol, VST caught 1.38 (1.11–1.72; P < 0.01) times more G. m. centralis flies than the un-baited trap. Attractants did not significantly increase the VST catch index for G. m. morsitans (P > 0.05). Overall, the VST caught 2.42 (1.91–3.10; P < 0.001) and 2.60 (1.50–3.21; P < 0.001) times more G. m. centralis and G. m. morsitans respectively, than the BFR. The VST and BFR took 10 and 35 min respectively to cover a 1 km transect. Conclusion/Significance: The VST is several times more effective for sampling G. m. morsitans and G. m. centralis than the BFR and we recommend its use as an alternative sampling tool. The antestia bug, Antestiopsis thunbergii (Gmelin 1790) is a major pest of Arabica coffee in Africa. The bug prefers coffee at the highest elevations, contrary to other major pests. The objectives of this study were to describe the relationship between A. thunbergii populations and elevation, to elucidate this relationship using our knowledge of the pest thermal biology and to predict the pest distribution under climate warming. Antestiopsis thunbergii population density was assessed in 24 coffee farms located along a transect delimited across an elevation gradient in the range 1000–1700 m asl, on Mt. Kilimanjaro, Tanzania. Density was assessed for three different climatic seasons, the cool dry season in June 2014 and 2015, the short rainy season in October 2014 and the warm dry season in January 2015. The pest distribution was predicted over the same transect using three risk indices: the establishment risk index (ERI), the generation index (GI) and the activity index (AI). These indices were computed using simulated life table parameters obtained from temperature-dependent development models and temperature data from 1) field records using data loggers deployed over the transect and 2) predictions for year 2055 extracted from AFRICLIM database. The observed population density was the highest during the cool dry season and increased significantly with increasing elevation. For current temperature, the ERI increased with an increase in elevation and was therefore distributed similarly to observed populations, contrary to the other indices. This result suggests that immature stage susceptibility to extreme temperatures was a key factor of population distribution as impacted by elevation. In the future, distribution of the risk indices globally indicated a decrease of the risk at low elevation and an increase of the risk at the highest elevations. Increasing global human populations and climate change not only increase the demand for food but require it to be supplied in sustainable quantities. One crucial aspect for sustainability is to ensure pollinator services for crop production and ecosystem services., The global distribution of universal bee pollinators and the different degrees of intensity of bee-keeping have resulted in a high variability of impacts. It is therefore essential to separate the different pollinator populations, in particular the cases of honeybees and bumblebees, into wild/feral and managed populations, which can and will interact., Such interactions can be either beneficial or deleterious. A deleterious interaction between wild/feral colonies and the managed populations may result in disease and pest transmission., However, a wild/feral population could easily buffer the effects of newly introduced pathogens increasing the resistance of the population as a whole. Varroa mites that caused the loss of the wild population of Apis mellifera in Europe have recently been established in South Africa. Studies of the mite's introduction into South Africa suggest that African honeybee populations are less affected by the mite and can deal with and survive parasite loads which would kill colonies in the Northern Hemisphere., This observed resilience in Africa and Americas is most likely based on the interactions between wild and managed bees, since a large proportion of the total honeybee population is wild in these regions and therefore not influenced by humans. This allows the wild population to adapt to new parasites/pathogens, without human interference, with the large numbers of wild colonies ensuring that resilience is high. A high ratio of wild to managed colonies could also ensure that beneficial adaptations in the wild population filter through to the managed population., Whether a similar situation prevails in bumblebees with wild and managed populations needs to be more carefully examined, particularly in the context of the global trade in colonies of these bees. In colonies of the honeybee Apis mellifera, the queen is usually the only reproductive female, which produces new females (queens and workers) by laying fertilized eggs. However, in one subspecies of A. mellifera, known as the Cape bee (A. m. capensis), worker bees reproduce asexually by thelytoky, an abnormal form of meiosis where two daughter nucleii fuse to form single diploid eggs, which develop into females without being fertilized. The Cape bee also exhibits a suite of phenotypes that facilitate social parasitism whereby workers lay such eggs in foreign colonies so their offspring can exploit their resources. The genetic basis of this switch to social parasitism in the Cape bee is unknown. To address this, we compared genome variation in a sample of Cape bees with other African populations. We find genetic divergence between these populations to be very low on average but identify several regions of the genome with extreme differentiation. The regions are strongly enriched for signals of selection in Cape bees, indicating that increased levels of positive selection have produced the unique set of derived phenotypic traits in this subspecies. Genetic variation within these regions allows unambiguous genetic identification of Cape bees and likely underlies the genetic basis of social parasitism. The candidate loci include genes involved in ecdysteroid signaling and juvenile hormone and dopamine biosynthesis, which may regulate worker ovary activation and others whose products localize at the centrosome and are implicated in chromosomal segregation during meiosis. Functional analysis of these loci will yield insights into the processes of reproduction and chemical signaling in both parasitic and non-parasitic populations and advance understanding of the process of normal and atypical meiosis. Nutrition plays an important role in physiological stress resistance and by adjusting their intake of key nutrients, such as protein and carbohydrate, many animals can better resist stress., Poor nutrition may contribute to the widespread and on-going declines of honeybee populations by increasing their vulnerability to abiotic (e.g. pesticides) and biotic (e.g. diseases) stressors. However, we do not know how nutrition affects stress resistance in social insects such as honeybees., Here, we examined how exposure to the toxic secondary metabolite nicotine, a neurotoxin that shares structural similarities with the neonicotinoid pesticides, and low temperatures affected nutrient regulation in honeybees using the Geometric Framework of nutrition., Groups of queenless, newly emerged worker bees were given diets containing specific ratios of protein and carbohydrate to determine, first, how toxin exposure and ambient temperature affected their nutrient intake and, secondly, how nutrition affected survival under stress., We find that low temperatures and nicotine interacted to reduce survival in African honeybees that ate low protein, high carbohydrate diets. However, bees fed a high protein diet were better able to survive insult with these interacting stressors., Although protein conferred a survival benefit in honeybees exposed to these dual stressors, when allowed to self-select their diet, caged workers did not shift their intake towards a higher protein food to improve their survival under these stressful conditions., We discuss the possible constraints on nutrient regulation in honeybees and the role that diet could play in their decline. Temperature is an important factor determining the abundance, distribution and diversity of termite species. Thus, termites are affected by changing climate and have to adopt different means of surviving in order to avoid extinction. Using termite occurrence data, bioclimatic variables and vegetation cover, we modelled and predicted the current and future habitat suitability for mound-building termites in Nigeria. Of the 19 bioclimatic variables and the vegetation index (NDVI) tested, only six were significant and eligible as predictors of habitat suitability for the mound-building termites Macrotermes subhyalinus and M. bellicosus. Under current climatic conditions (2022), the northwest, northeast and central regions are highly suitable for M. subhyalinus, while the distribution of M. bellicosus decreased in the North West, North East and in the Central region. However, regarding habitat suitability for the future (2050), there was a predicted range expansion into suitable areas for the two termite species. The increase in temperature due to global warming has an effect which can either result in migration or sometimes extinction of termite species within an ecosystem. Here, we have predicted habitat suitability for the two mound-building termite species under current and future climatic scenarios, and how the change in climatic variables would lead to an expansion in their range across Nigeria. Simple Summary: Naked mole-rats have a unique social system similar to insects, where there is one reproductive female, one to three reproductive males, and the rest are non-reproductive members of the colony. Within the non-reproductive members, there are dispersers that attempt to leave the colony. We aimed to investigate if naked mole-rat dispersers have consistent behavioral responses to situations and therefore possess animal personality traits. The naked mole-rat dispersers showed consistent responses to different situations, and had consistent differences between individuals. In addition, we recorded how naked mole-rats respond to situations that threaten their survival, such as a new environment and predators. Further investigation into these animal personality traits and how they compare between other colony members, such as workers and soldiers are needed to understand the naked mole-rat social system. Animal personality traits (consistent behavioral differences between individuals in their behavior across time and/or situation) affect individual fitness through facets, such as dispersal. In eusocial naked mole-rat (Heterocephalus glaber) colonies, a disperser morph may arise with distinct morphological, behavioral, and physiological characteristics. This study aimed to quantify the personality traits of a cohort of disperser morphs of naked mole-rat (NMR). Behavioral tests were performed on twelve disperser morphs (six males and six females) in an observation tunnel system that was novel and unfamiliar. Novel stimuli (fresh snakeskin, tissue paper, and conspecific of the same sex) were introduced for fifteen minutes, and the behavioral acts of the individual were recorded. A total of 30 behaviors were noted during the behavioral tests of which eight were used to quantify aggression, boldness, and exploration. The NMR disperser morphs showed consistent individual differences in boldness, and exploration across time and test, indicating a distinct personality. In addition, new naked mole-rat responses including disturbance behaviors; confront, barricade, and stay-away, were recorded. Further investigations into the relationships between animal personality traits and social hierarchy position in entire colonies are needed for more informative results as we further investigate the role of personality in cooperatively breeding societies. The nutritional needs of bees are supplied by nectar carbohydrates and by protein and other nutrients in pollen but little is known of how bees achieve nutritional balance. Using newly emerged caged worker honeybees (Apis mellifera scutellata), we investigated whether bees maintain their intake target when confined to pairs of imbalanced complementary diets varying in protein to carbohydrate (P:C) ratio. Diets were formulated using three protein sources [casein, royal jelly or Feed-Bee® (a natural pollen substitute)] and sucrose. Within each protein type, honeybees switched between complementary diets and converged on the same P:C intake target. However, this target differed between protein types: P:C ratios were 1:12, 1:14 and 1:11 on casein, royal jelly and Feed-Bee® diets, respectively. Except for an early peak in protein consumption on royal jelly diets, these strongly convergent ratios remained constant over the 14 day experiment. This is probably due to the absence of brood, reflected in relatively stable values measured for haemolymph protein concentration and hypopharyngeal gland activation in bees on Feed- Bee® diets. Performance of caged workers was also assessed in terms of survival and ovarian activation. Survival was highest on casein diets and lowest on Feed~Bee® diets but ovarian activation was highest on royal jelly diets and lowest on casein diets. This may be due to additional components in Feed~Bee® and royal jelly (e.g. fatty acids), which are needed to activate the ovaries but also reduce survival. Nutrient intake of broodless workers is directly related to their own physiological requirements, and the strong carbohydrate bias may reflect the high metabolic rate of honeybees even under resting conditions. Honey bees process nectar into honey by active evaporation on the tongue and passive evaporation involving nest ventilation and fanning behaviour, as well as enzymatic action. The elimination of excess water from nectar carries considerable energetic costs. The concentration of the nectar load is assumed to remain constant during transport. However, some of this water elimination may occur before foragers return to the nest and pass their nectar loads to receiver bees. In honey bees captured while foraging in Macadamia orchards, we show that the nectar in their crops has approximately twice the sugar concentration of the fresh nectar in flowers. This was true for four Macadamia cultivars, with up to 75% of the initial water content being removed. There is a further concentration increase in the crops of returning bees captured at the hive entrance. The only possible route of water elimination from the crop is via evaporation from the mouthparts. We calculate the savings in honey processing costs to be on average 35 times more than the reduction in flight costs due to reduced body mass. Laying workers of the Cape honeybee parthenogenetically produce female offspring, whereas queens typically produce males. Beekman et al. confirm this observation, which has repeatedly been reported over the last 100 years including the notion that natural selection should favor asexual reproduction in Apis mellifera capensis. They attempt to support their arguments with an exceptionally surprising finding that A. m. capensis queens can parthenogenetically produce diploid homozygous queen offspring (homozygous diploid individuals develop into diploid males in the honeybee). Beekman et al. suggest that these homozygous queens are not viable because they did not find any homozygous individuals beyond the third larval instar. Even if this were true, such a lethal trait should be quickly eliminated by natural selection. The identification of sex (both with molecular and morphological markers) is possible but notoriously difficult in honeybees at the early larval stages. Ploidy is however a reliable indicator, and we therefore suggest that these “homozygous” larvae found in queen cells are actually drones reared from unfertilized eggs, a phenomenon well known by honeybee queen breeders. In many species of social Hymenoptera, unmated workers can lay eggs that will produce males by parthenogenesis. Nevertheless, in queenright honey bee colonies (Apis mellifera), worker reproduction is low. One possible mechanism for this difference is worker policing, the removal of worker-laid eggs by other workers. This behavior can evolve in species in which queens are multiply mated, where workers are more closely related to the sons of their mother than those of their sisters. Another possible mechanism of the low level of worker reproduction is worker-laid eggs being less viable than queen-laid eggs. We show that this difference in quality is the case for honey bees. Cape honeybee, Apis mellifera capensis, workers can be social parasites and host colonies can defend themselves by rejection of such workers. Using the pseudo-clonal obligate parasitic lineage of A. m. capensis and wild-type A. m. capensis workers, which are facultative parasites, we show that host colonies significantly increase their defence behaviour towards social parasites upon secondary exposure. Most obligate and facultative social parasites were rejected before they could even produce significant amounts of the queen-like mandibular gland pheromone secretion or activate their ovaries. This suggests that other signals, like cuticular hydrocarbons, could be used by host colonies to identify potential invaders. Honeybee colonies seem to be able to utilise these potential cues, learn from their initial exposure to hive intruders and enable them to improve their defensive behaviour during subsequent infestations, resulting in increased removal rates of parasites. Stress in animals has been linked to behavioural and physiological changes in response to environmental, social and anthropogenic stimuli. Hence, stress-related responses in animals, especially in rodents, have been used as biological indicators of ecosystem health. This study aimed to establish an enzyme immunoassay (EIA) for monitoring adrenocortical activity in free-ranging Namaqua rock mice Micaelamys namaquensis (Rodentia: Muridae) using faeces as a prerequisite for assessing the effects of anthropogenic land use and season on faecal glucocorticoid metabolite (fGCM) concentration. Rodents were live-trapped seasonally across four land use types: an agricultural crop farm, an agricultural livestock farm, a human-populated site and a nature reserve; all situated in Limpopo Province, South Africa. Determined fGCM concentrations from capture and recapture events were used for biologically validating an EIA detecting steroids with a 5α-3β-11β-diol structure. Recapturing resulted in a significant overall 40% elevation of individual fGCM concentrations demonstrating the effectiveness of the chosen EIA to reliably detect glucocorticoid output in the study species. Neither land use type nor season affected fGCM concentrations in the species, suggesting that land use and season-related environmental changes do not necessarily act as stressors for M. namaquensis, presumably due to their adaptive and resilient nature. Such species can be used to identify ecosystems affected by human-mediated disturbances and allow insights into the management and restoration of these threatened ecosystems and their associated species. The global spread of vector-borne diseases remains a worrying public health threat, raising the need for development of new combat strategies for vector control. Knowledge of vector ecology can be exploited in this regard, including plant feeding; a critical resource that mosquitoes of both sexes rely on for survival and other metabolic processes. However, the identity of plant species mosquitoes feed on in nature remains largely unknown. By testing the hypothesis about selectivity in plant feeding, we employed a DNA-based approach targeting trnH-psbA and matK genes and identified host plants of field-collected Afro-tropical mosquito vectors of dengue, Rift Valley fever and malaria being among the most important mosquito-borne diseases in East Africa. These included three plant species for Aedes aegypti (dengue), two for both Aedes mcintoshi and Aedes ochraceus (Rift Valley fever) and five for Anopheles gambiae (malaria). Since plant feeding is mediated by olfactory cues, we further sought to identify specific odor signatures that may modulate host plant location. Using coupled gas chromatography (GC)-electroantennographic detection, GC/mass spectrometry and electroantennogram analyses, we identified a total of 21 antennally-active components variably detected by Ae. aegypti, Ae. mcintoshi and An. gambiae from their respective host plants. Whereas Ae. aegypti predominantly detected benzenoids, Ae. mcintoshi detected mainly aldehydes while An. gambiae detected sesquiterpenes and alkenes. Interestingly, the monoterpenes β-myrcene and (E)-β-ocimene were consistently detected by all the mosquito species and present in all the identified host plants, suggesting that they may serve as signature cues in plant location. This study highlights the utility of molecular approaches in identifying specific vector-plant associations, which can be exploited in maximizing control strategies such as such as attractive toxic sugar bait and odor-bait technology. One remarkable example is that of the ectoparasitic mite Varroa destructor , which has shifted its host from Eastern honey bees (Apis cerana) to Western honey bees (Apis mellifera) and posed a global threat to apiculture. To identify the genetic factors underlying the reproduction of host-shifted V. destructor on the new host. Genome sequencing was conducted to construct the phylogeny of the host-shifted and non-shifted mites and to screen for genomic signatures that differentiated them. Artificial infestation experiment was conducted to compare the reproductive difference between the mites, and transcriptome sequencing was conducted to find differentially expressed genes (DEGs) during the reproduction process. The host-shifted and non-shifted V. destructor mites constituted two genetically distinct lineages, with 15,362 high- F ST SNPs identified between them. Oogenesis was upregulated in host-shifted mites on the new host A. mellifera relative to non-shifted mites. The transcriptomes of the host-shifted and non-shifted mites differed significantly as early as 1h post-infestation. The DEGs were associated with nine genes carrying nonsynonymous high- F ST SNPs, including mGluR2-like , Lamb2-like and Vitellogenin 6-like , which were also differentially expressed, and eIF4G , CG5800 , Dap160 and Sas10 , which were located in the center of the networks regulating the DEGs based on protein-protein interaction analysis. The annotated functions of these genes were all associated with oogenesis. These genes appear to be the key genetic determinants of the oogenesis of host-shifted mites on the new host. Over a decade, declines in honey bee colonies have raised worldwide concerns. Several potentially contributing factors have been investigated, e.g. parasites, diseases, and pesticides. Neonicotinoid pesticides have received much attention due to their intensive use in crop protection, and their adverse effects on many levels of honey bee physiology led the European Union to ban these compounds. Due to their neuronal target, a receptor expressed throughout the insect nervous system, studies have focused mainly on neuroscience and behaviour. Through the Geometric Framework of nutrition, we investigated effects of the neonicotinoid thiamethoxam on survival, food consumption and sucrose sensitivity of honey bees (Apis mellifera). Thiamethoxam did not affect protein and carbohydrate intake, but decreased responses to high concentrations of sucrose. Interestingly, when bees ate fixed unbalanced diets, dietary protein facilitated better sucrose detection. Both thiamethoxam and dietary protein influenced survival. These findings suggest that, in the presence of a pesticide and unbalanced food, honey bee health may be severely challenged. The African termite-raiding ant Pachycondyla analis Latreille (Hymenoptera: Formicidae) organizes group raids on termites of the sub-family Macrotermitinae. Termites and ants occupy and share similar habitats, resulting in a co-evolutionary arms race between termites as prey and ants as predators. The present study explored whether P. analis uses semiochemical signaling cues to detect potential termite prey prior to and during raids. Ants’ responses to odors emitted from termites alone, termite gallery soil, and termites inside their galleries were tested using Y-tube olfactometer assays. The results showed that P. analis detected odors of termites and those of their galleries, and odors from termites inside their galleries were more attractive to both minor and major ant workers than odors from termites alone. The composition of these odor sources was identified using gas chromatography- mass spectrometry analysis. While the odors from termite gallery soils were compositionally richer (containing 13 compounds rather than nine from termites alone), those from the termites alone were quantitatively richer, releasing about six times more odors than gallery soil. Most of the compounds in the odor profiles were identified as hydrocarbons. Naphthalene, previously identified as an insect repellent, was also identified as a component of the odors from the gallery soil. Herein, we have expanded on this study by investigating the role of three of the plants, Pithecellobium dulce (Fabaceae), Leonotis nepetifolia (Lamiaceae), and Opuntia ficus-indica (Cactaceae), on the survival, fecundity, and egg viability of the dengue vector Aedes aegypti. Methods: We tested these effects using females that received (i) an initial three rations of blood meals and (ii) no blood meal at all. Two controls were included: age-matched females fed on glucose solution with or without an initial blood meal and those fed exclusively on blood meals. Data were collected daily over a 30-day period. The amino acid contents of Ae. aegypti guts and their respective diets were detected by coupled liquid chromatography-mass spectrometry. Results: Females fed on P. dulce and an exclusively blood meal diet had a shorter survival than those fed on glucose. On the other hand, females fed on L. nepetifolia survived longer than those fed exclusively on blood meals, whereas those fed on O. ficus-indica had the shortest survival time. With an initial blood meal, females fed on L. nepetifolia laid 1.6-fold more eggs while those fed on the other diets laid fewer eggs compared to those fed exclusively on blood meals. The African weaver ant, Oecophylla longinoda, is used as a biological control agent for the management of pests. The ant has several exocrine glands in the abdomen, including Dufour's, poison, rectal, and sternal glands, which are associated with pheromone secretions for intra-specific communication. Previous studies have analyzed the gland secretions of Dufour's and poison glands. The chemistry of the rectal and sternal glands is unknown. We re-analyzed the secretions from Dufour's and poison glands plus the rectal and sternal glands to compare their chemistries and identify additional components. We used the solid-phase microextraction (SPME) technique to collect gland headspace volatiles and solvent extraction for the secretions. Coupled gas chromatography–mass spectrometry (GC-MS) analysis detected a total of 78 components, of which 62 were being reported for the first time. These additional components included 32 hydrocarbons, 12 carboxylic acids, 5 aldehydes, 3 alcohols, 2 ketones, 4 terpenes, 3 sterols, and 1 benzenoid. The chemistry of Dufour's and poison glands showed a strong overlap and was distinct from that of the rectal and sternal glands. Visual and olfactory communications are vital for coordinated group hunting in most animals. To hunt for prey, the group-raiding termite specialist ant Megaponera analis , which lacks good vision, must first confirm the presence or absence of conspecific raiders. Here, we show that M. analis uses olfactory cues for intraspecific communication and showed greater preference for conspecific odors over clean air (blank) or odors from its termite prey. Chemical analysis of ant volatiles identified predominantly short-chained hydrocarbons. Electrophysiological analysis revealed differential sensory detection of the odor compounds, which were confirmed in behavioral olfactometric choice assays with odor bouquets collected from major and minor castes and the 2 most dominant volatiles and n- undecane n- tridecane. A comparative analysis of the cuticular hydrocarbon profile with those of the short-chained odor bouquet of different populations shows a high divergence in the long-chained profile and a much-conserved short-chained odor bouquet. This suggests that there is less selection pressure for divergence and individual recognition in the short- than the long-chained odor profiles. We conclude that olfactory communication serves as an alternative to visual or sound communication, especially during group raids in M. analis when ants are not in direct contact with one another. Background: A blend of compounds (pentanoic acid, guaiacol, δ-octalactone and geranylacetone) identified in waterbuck (Kobus defassa) body odour referred to as waterbuck repellent compounds (WRC) and a synthetic repellent 4-methylguaiacol have previously been shown to repel tsetse flies from the morsitans group. However, these repellents have not been evaluated on palpalis group tsetse flies. In this study, we evaluated the effect of these repellents on catches of Glossina fuscipes fuscipes (major vector of human sleeping sickness) in biconical traps and on sticky small targets which are visually attractive to palpalis group flies. The attractive devices were baited with different doses and blends of the repellent compounds. We also assessed the effect of removal of individual constituents in the synthetic blend of WRC on catches of G. f. fuscipes. Methodology/Principal findings: The study was conducted in western Kenya on four islands of Lake Victoria namely Big Chamaunga, Small Chamaunga, Manga and Rusinga. The tsetse fly catches from the treatments were modeled using a negative binomial regression to determine their effect on catches. In the presence of WRC and 4-methylguaiacol (released at ≈2 mg/h and ≈1.4 mg/h respectively), catches of G. f. fuscipes were significantly reduced by 33% (P<0.001) and 22% (P<0.001) respectively in biconical traps relative to control. On sticky small targets the reduction in fly catches were approximately 30% (P<0.001) for both 4-methylguiacol and WRC. In subtractive assays, only removal of geranylacetone from WRC significantly increased catches (by 1.8 times; P <0.001) compared to the complete blend of WRC. Conclusions/Significance: We conclude that WRC and 4-methylguaiacol reduce catches of G. f. fuscipes at stationary visually attractive traps and suggest that they may serve as broad spectrum repellents for Glossina species. We recommend further studies to investigate the effects of these compounds on reduction of G. f. fuscipes attracted to human hosts as this may lead to development of new strategies of reducing the prevalence and incidence of sleeping sickness. Background: African trypanosomosis, primarily transmitted by tsetse flies, remains a serious public health and economic challenge in sub-Saharan Africa. Interventions employing natural repellents from non-preferred hosts of tsetse flies represent a promising management approach. Although zebras have been identified as non-preferred hosts of tsetse flies, the basis for this repellency is poorly understood. We hypothesized that zebra skin odors contribute to their avoidance by tsetse flies. Methodology/Principal findings: We evaluated the effect of crude zebra skin odors on catches of wild savannah tsetse flies (Glossina pallidipes Austen, 1903) using unbaited Ngu traps compared to the traps baited with two known tsetse fly management chemicals; a repellent blend derived from waterbuck odor, WRC (comprising geranylacetone, guaiacol, pentanoic acid and δ-octalactone), and an attractant comprising cow urine and acetone, in a series of Latin square-designed experiments. Coupled gas chromatography-electroantennographic detection (GC/EAD) and GC-mass spectrometry (GC/MS) analyses of zebra skin odors identified seven electrophysiologically-active components; 6-methyl-5-hepten-2-one, acetophenone, geranylacetone, heptanal, octanal, nonanal and decanal, which were tested in blends and singly for repellency to tsetse flies when combined with Ngu traps baited with cow urine and acetone in field trials. The crude zebra skin odors and a seven-component blend of the EAD-active components, formulated in their natural ratio of occurrence in zebra skin odor, significantly reduced catches of G. pallidipesby 66.7% and 48.9% respectively, and compared favorably with the repellency of WRC (58.1%– 59.2%). Repellency of the seven-component blend was attributed to the presence of the three ketones 6-methyl-5-hepten-2-one, acetophenone and geranylacetone, which when in a blend caused a 62.7% reduction in trap catch of G. pallidipes. Conclusions/Significance: Our findings reveal fundamental insights into tsetse fly ecology and the allomonal effect of zebra skin odor, and potential integration of the three-component ketone blend into the management toolkit for tsetse and African trypanosomosis control. Varroa destructor is an ectoparasitic pest of honeybees, and a threat to the survival of the apiculture industry. Several studies have shown that unlike European honeybees, African honeybee populations appear to be minimally affected when attacked by this mite. However, little is known about the underlying drivers contributing to survival of African honeybee populations against the mite. We hypothesized that resistant behavioral defenses are responsible for the survival of African honeybees against the ectoparasite. We tested this hypothesis by comparing grooming and hygienic behaviors in the African savannah honeybee Apis mellifera scutellata in Kenya and A. mellifera hybrids of European origin in Florida, USA against the mite. Grooming behavior was assessed by determining adult mite infestation levels, daily mite fall per colony and percentage mite damage (as an indicator of adult grooming rate), while hygienic behavior was assessed by determining the brood removal rate after freeze killing a section of the brood. Our results identified two additional undescribed damaged mite categories along with the six previously known damage categories associated with the grooming behavior of both honeybee subspecies. Adult mite infestation level was approximately three-fold higher in A. mellifera hybrids of European origin than in A. m. scutellata, however, brood removal rate, adult grooming rate and daily natural mite fall were similar in both honeybee subspecies. Unlike A. mellifera hybrids of European origin, adult grooming rate and brood removal rate did not correlate with mite infestation levels on adult worker honeybee of A. m. scutellata though they were more aggressive towards the mites than their European counterparts. Background: Lack of effective vaccines and therapeutics for important arboviral diseases such as Rift Valley fever (RVF) and dengue, necessitates continuous monitoring of vector populations for infections in them. Plant-based lures as surveillance tools has the potential of targeting mosquitoes of both sexes and females of varied physiological states; yet such lures are lacking for vectors of these diseases. Here, we present evidence of the effectiveness of linalool oxide (LO), a single plant-based lure previously developed for malaria vectors in trapping RVF vectors, Aedes mcintoshi and Aedes ochraceus, and dengue vector, Aedes aegypti. Methods: For RVF vectors, we used CDC traps to evaluate the performance of LO against three vertebrate-based lures: CO2 (dry ice), BioGent (BG) lure, and HONAD (a blend of aldehydes) in 2 experiments with Completely Randomized design: 1) using unlit CDC traps baited separately with LO, HONAD and BG-lure, and unlit CDC trap + CO2 and lit CDC trap as controls, 2) similar treatments but with inclusion of CO2 to all the traps. For dengue vectors, LO was evaluated against BG lure using BG sentinel traps, in a 3 × 6 Latin Square design, first as single lures and then combined with CO2 and traps baited with CO2 included as controls. Trap captures were compared between the treatments using Chi square and GLM. Results: Low captures of RVF vectors were recorded for all lures in the absence of CO2 with no significant difference between them. When combined with CO2, LO performance in trapping these vectors was comparable to BG-lure and HONAD but it was less effective than the lit CDC trap. In the absence of CO2, LO performed comparably with the BGlure in trapping female Ae. aegypti, but with significantly higher males recorded in traps baited with the plant-based lure. When CO2 was added, LO was significantly better than the BG-lure with a 2.8- fold increase in captures of male Ae. aegypti. Conclusions: These results highlight the potential of LO as a generalist plant-based lure for mosquito disease vectors, pending further assessment of possible specificity in their response profile to the different stereoisomers of this compound. Background: Effective surveillance and estimation of the biting fraction of Aedes aegypti is critical for accurate determination of the extent of virus transmission during outbreaks and inter-epidemic periods of dengue and chikungunya fever. Here, we describe the development and use of synthetic human odor baits for improved sampling of adult Ae. aegypti, in two dengue and chikungunya fevers endemic areas in Kenya; Kilifiand Busia counties. Methods: We collected volatiles from the feet and trunks of twofemale and two male volunteers aged between 25 and 45 years. We used coupled gas chromatography- electroantennographic detection (GC/EAD) analysis to screen for antennally-active components from the volatiles and coupled GC-mass spectrometry (GC/MS) to identify the EAD-active components. Using randomized replicated designs, we compared the efficacies of Biogents (BG) sentinel traps baited with carbon dioxide plus either single or blends of the identified compounds against the BG sentinel trap baited with carbon dioxide plus the BG commercial lure in trapping Ae. aegypti. The daily mosquito counts in the different traps were subjected to negative binomial regression following the generalized linear models procedures. Results: A total of ten major EAD-active components identified by GC/MS as mainly aldehydes and carboxylic acids, were consistently isolated from the human feet and trunk volatiles from at least two volunteers. Field assays with synthetic chemicals of the shared EAD-active components identified from the feet and trunk gave varying results. Ae. aegypti were more attracted to carbon dioxide baited BG sentinel traps combined with blends of aldehydes than to similar traps combined with blends of carboxylic acids. When we assessed the efficacy of hexanoic acid detected in odors of the BG commercial lure and volunteers plus carbon dioxide, trap captures of Ae. aegypti doubled over the trap baited with the commercial BG lure. However, dispensing aldehydes and carboxylic acids together in blends, reduced trap captures of Ae. aegypti by ∼45%-50%. Conclusions: Our results provide evidence for roles of carboxylic acids and aldehydes in Ae. aegypti host attraction and also show that of the carboxylic acids, hexanoic acid is a more effective lure for the vector than the BG commercial lure. Background: Methods currently used in sampling adult Aedes aegypti, the main vector of dengue and chikungunya viruses are limited for effective surveillance of the vector and accurate determination of the extent of virus transmission during outbreaks and inter - epidemic periods. Here, we document the use of natural human skin odours in baited traps to improve sampling of adult Ae. aegypti in two different endemic areas of chikungunya and dengue in Kenya – Kilifi and Busia Counties. The chemistry of the volatiles released from human odours and the Biogent (BG)-commercial lure were also compared. Methods: Cotton socks and T-shirts were used to obtain natural human skin volatiles from the feet and trunk of three volunteers (volunteers 1 and 2 in Kilifi and volunteers 2 and 3 in Busia). Using Latin square design, we compared the efficacies of BG sentinel traps baited with carbon dioxide plus (a) no bait, (b) human feet volatiles, (c) human trunk volatiles each against (c) a control (Biogent commercial lure) at the two sites. Coupled gas chromatography-mass spectrometry (GC-MS) was used to identify and compare candidate attractants released by the commercial lure and human odours. Results: Ae. aegypti captured in the trap baited with feet odours from volunteer 2 and trunk odours from the same volunteer were significantly higher than in the control trap in Busia and Kilifi respectively, [IRR = 5.63, 95% CI: 1.15 - 28.30, p = 0.030] and [IRR = 3.99, 95% CI: 0.95-16.69, p = 0.049]. At both sites, Ae. aegypti captures in traps baited with either the feet or trunk odours from volunteers 1 and 3 were not significantly different from the control. Major qualitative differences were observed between the chemical profiles of human odours and the commercial BG-lure. Aldehydes, fatty acids and ketones dominated human odour profiles, whereas the BG-lure released mainly hexanoic acid. Conclusions: Our results suggest that additional candidate attractants are present in human skin volatiles which can help to improve the efficacy of lures for trapping and surveillance of Ae. aegypti. In honeybees, worker reproduction is mainly regulated by pheromones produced by the brood and the queen. The source of one of the queen pheromones influencing worker reproduction has been located in the mandibular glands. In nonlaying workers, this gland’s profile is dominated by fatty acids that are incorporated into the food given to the brood and to nest mates. After queen loss and onset of reproductive activity, workers are able to synthesize different fatty acids, which are normally only produced by queens and that contribute to their reproductive success. Apis mellifera capensis workers have the ability to rapidly produce queen-like mandibular profiles that could represent an important factor in their ability to behave as facultative intraspecific social parasites. Indeed, A. m. capensis workers can take over reproduction from the host queens in colonies of other subspecies. Here, we show that in the presence of their own queen, the mandibular gland profile of A. m. capensis workers is dominated by the precursor of the major compound of the queen pheromone. This is a unique trait among honeybee workers and suggests that A. m. capensis workers are primed for reproduction and that this phenomenon represents a pheromonal predisposition to social parasitism. We identified geographical variation in the ratio of queen- to worker-specific compounds in the mandibular gland profile of A. m. capensis workers, which corresponds with the introgression with the neighboring subspecies A. m. scutellata. To explore reasons for the unusually high rates of worker ovary activation in Apis cerana, we investigated the reproductive physiology of workers in conspecific and mixed-species colonies of A. cerana and its sibling species Apis mellifera under queenright and queenless conditions. In both queenless and queenright conspecific colonies, the proportion of A. cerana workers with activated ovaries was significantly greater than the proportion of A. mellifera workers with activated ovaries. Apis cerana workers showed significantly greater ovary activation in queenright mixed-species colonies than in conspecific queenright colonies. Moreover, there was significantly greater ovary activation in A. cerana workers in mixed-species colonies headed by A. mellifera queens than A. mellifera workers in mixed-species colonies headed by A. cerana queens. Apis mellifera workers in conspecific queenless colonies showed significantly greater ovary activation than those in mixed-species queenless colonies. Quantification of the chemical components of mandibular gland pheromones of queens of the 2 species showed that they are similar. Combined, our results show that queen signals have been preserved between the 2 species and that workers respond to the heterospecific signals from queens. However, despite the similarity in the queen mandibular gland fatty acid secretions between the 2 species, workers respond differentially toward hetero- and conspecific queens. Background: Small targets comprising panels of blue and insecticide-treated black netting material each 0.25 × 0.25 m have been shown to attract and kill Glossina fuscipes fuscipes Newstead, 1910 (Diptera: Glossinidae) thereby reducing its population density by over 90% in field trials. However, their attractive ability has not been fully exploited for sampling purposes. Therefore, in this study we assessed the effectiveness of using sticky small targets as sampling tools for G. f. fuscipes in western Kenya. We also determined the influence of colour on the landing response of female and male flies on sticky small targets. Methods: Using a series of randomised block experiments, the numbers of tsetse flies caught with sticky small targets were compared with those caught with biconical traps. A negative binomial regression was used to model fly catches. Odds ratios as measures of association between the landing response on the blue or black panel of the sticky small target and the sex of flies were obtained from a multiple logistic regression. Results: The results showed that sticky small targets caught 13.5 and 3.6 times more female and male tsetse flies than biconical traps (Z = 9.551, P < 0.0001 and Z = 5.978, P < 0.0001, respectively). Females had a 1.7 times likelihood of landing on the black panel than males (Z = 2.25, P = 0.025). Conclusion: This study suggests that sticky small targets are an effective sampling tool for G. f. fuscipes. Therefore, we recommend the use of sticky small targets as an alternative to biconical traps for observational and experimental investigations of G. f. fuscipes. [ABSTRACT FROM AUTHOR]

Insecticides are thought to be among the major factors contributing to current declines in bee populations. However, detoxification mechanisms in healthy, unstressed honey bees are poorly characterised. Alkaloids are naturally encountered in pollen and nectar, and we used nicotine as a model compound to identify the mechanisms involved in detoxification processes in honey bees. Nicotine and neonicotinoids have similar modes of action in insects. Our metabolomic and proteomic analyses show active detoxification of nicotine in bees, associated with increased energetic investment and also antioxidant and heat shock responses. The increased energetic investment is significant in view of the interactions of pesticides with diseases such as Nosema spp which cause energetic stress and possible malnutrition. Understanding how healthy honey bees process dietary toxins under unstressed conditions will help clarify how pesticides, alone or in synergy with other stress factors, lead to declines in bee vitality. The internal temperature of flowers may be higher than air temperature, and warmer nectar could offer energetic advantages for honeybee thermoregulation, as well as being easier to drink owing to its lower viscosity. We investigated the responses of Apis mellifera scutellata (10 colonies) to warmed 10% w/w sucrose solutions, maintained at 20-35°C, independent of low air temperatures, and to 20% w/w sucrose solutions with the viscosity increased by the addition of the inert polysaccharide Tylose (up to the equivalent of 34.5% sucrose). Honeybee crop loads increased with nectar temperature, as did the total consumption of sucrose solutions over 2 h by all bees visiting the feeders. In addition, the preference of marked honeybees shifted towards higher nectar temperatures with successive feeder visits. Crop loads were inversely proportional to the viscosity of the artificial nectar, as was the total consumption of sucrose solutions over 2 h. Marked honeybees avoided higher nectar viscosities with successive feeder visits. Bees thus showed strong preferences for both warmer and less viscous nectar, independent of changes in its sugar concentration. Bees may benefit from foraging on nectars that are warmer than air temperature for two reasons that are not mutually exclusive: reduced thermoregulatory costs and faster ingestion times due to the lower viscosity.
