Abstract

Third-harmonic generation microscopy is a powerful label-free nonlinear imaging technique, providing essential information about structural characteristics of cells and tissues without requiring external labelling agents. In this work, we integrated a recently developed compact adaptive optics module into a third-harmonic generation microscope, to measure and correct for optical aberrations in complex tissues. Taking advantage of the high sensitivity of the third-harmonic generation process to material interfaces and thin membranes, along with the 1,300-nm excitation wavelength used here, our adaptive optical third-harmonic generation microscope enabled high-resolution in vivo imaging within highly scattering biological model systems. Examples include imaging of myelinated axons and vascular structures within the mouse spinal cord and deep cortical layers of the mouse brain, along with imaging of key anatomical features in the roots of the model plant Brachypodium distachyon. In all instances, aberration correction led to significant enhancements in image quality.

Abstract

Understanding complex biological systems requires visualizing structures and processes deep within living organisms. We developed a compact adaptive optics module and incorporated it into two- and three-photon fluorescence microscopes, to measure and correct tissue-induced aberrations. We resolved synaptic structures in deep cortical and subcortical areas of the mouse brain, and demonstrated high-resolution imaging of neuronal structures and somatosensory-evoked calcium responses in the mouse spinal cord at great depths in vivo.

24. Fracture toughness characterization through notched small punch test specimens

Abstract

In this work a novel methodology for fracture toughness characterization by means of the small punch test (SPT) is presented. Notched specimens are employed and fracture resistance is assessed through a critical value of the notch mouth displacement {\delta^SPT}. Finite element simulations and interrupted experiments are used to track the evolution of {\delta^SPT} as a function of the punch displacement. The onset of crack propagation is identified by means of a ductile damage model and the outcome is compared to the crack tip opening displacement estimated from conventional tests at crack initiation. The proposed numerical-experimental scheme is examined with two different grades of CrMoV steel and the differences in material toughness captured. Limitations and uncertainties arising from the different damage phenomena observed in the lowest toughness material examined are thoroughly discussed.

Abstract

Background: Element-equivalent matched theranostic pairs facilitate quantitative in vivo imaging to establish pharmacokinetics and dosimetry estimates in the development of preclinical radiopharmaceuticals. Terbium radionuclides have significant potential as matched theranostic pairs for multipurpose applications in nuclear medicine. In particular, 155Tb (t1/2 = 5.32 d) and 161Tb (t1/2 = 6.89 d) have been proposed as a theranostic pair for their respective applications in single photon emission computed tomography (SPECT) imaging and targeted beta therapy. Our study assessed the performance of preclinical quantitative SPECT imaging with 155Tb and 161Tb. A hot rod resolution phantom with rod diameters ranging between 0.85 and 1.70 mm was filled with either 155Tb (21.8 ± 1.7 MBq/mL) or 161Tb (23.6 ± 1.9 MBq/mL) and scanned with the VECTor preclinical SPECT/CT scanner. Image performance was evaluated with two collimators: a high energy ultra high resolution (HEUHR) collimator and an extra ultra high sensitivity (UHS) collimator. SPECT images were reconstructed from photopeaks at 43.0 keV, 86.6 keV, and 105.3 keV for 155Tb and 48.9 keV and 74.6 keV for 161Tb. Quantitative SPECT images of the resolution phantoms were analyzed to report inter-rod contrast, recovery coefficients, and contrast-to-noise metrics. Results: Quantitative SPECT images of the resolution phantom established that the HEUHR collimator resolved all rods for 155Tb and 161Tb, and the UHS collimator resolved rods ≥ 1.10 mm for 161Tb and ≥ 1.30 mm for 155Tb. The HEUHR collimator maintained better quantitative accuracy than the UHS collimator with recovery coefficients up to 92%. Contrast-to-noise metrics were also superior with the HEUHR collimator. Conclusions: Both 155Tb and 161Tb demonstrated potential for applications in preclinical quantitative SPECT imaging. The high-resolution collimator achieves < 0.85 mm resolution and maintains quantitative accuracy in small volumes which is advantageous for assessing sub organ activity distributions in small animals. Abstract

Determining the number of cases of an epidemic is the first function of epidemiological surveillance. An important underreporting of cases was observed in many locations during the first wave of the COVID-19 pandemic. To estimate this underreporting in the COVID-19 outbreak of Borriana (Valencia Community, Spain) in March 2020, a cross-sectional study was performed in June 2020 querying the public health register. Logistic regression models were used. Of a total of 468 symptomatic COVID-19 cases diagnosed in the outbreak through anti-SARS-CoV-2 serology, 36 cases were reported (7.7%), resulting in an underreporting proportion of 92.3% (95% confidence interval [CI], 89.5–94.6%), with 13 unreported cases for every reported case. Only positive SARS-CoV-2 polymerase chain reaction cases were predominantly reported due to a limited testing capacity and following a national protocol. Significant factors associated with underreporting included no medical assistance for COVID-19 disease, with an adjusted odds ratio [aOR] of 10.83 (95% CI 2.49–47.11); no chronic illness, aOR = 2.81 (95% CI 1.28–6.17); middle and lower social classes, aOR = 3.12 (95% CI 1.42–6.85); younger age, aOR = 0.97 (95% CI 0.94–0.99); and a shorter duration of illness, aOR = 0.98 (95% CI 0.97–0.99). Abstract

With the ability to correct for the aberrations introduced by biological specimens, adaptive optics-a method originally developed for astronomical telescopes-has been applied to optical microscopy to recover diffraction-limited imaging performance deep within living tissue. In particular, this technology has been used to improve image quality and provide a more accurate characterization of both structure and function of neurons in a variety of living organisms. Among its many highlights, adaptive optical microscopy has made it possible to image large volumes with diffraction-limited resolution in zebrafish larval brains, to resolve dendritic spines over 600μm deep in the mouse brain, and to more accurately characterize the orientation tuning properties of thalamic boutons in the primary visual cortex of awake mice.

Abstract

Volumetric imaging tools that are simple to adopt, flexible, and robust are in high demand in the field of neuroscience, where the ability to image neurons and their networks with high spatiotemporal resolution is essential. Using an axially elongated focus approximating a Bessel beam, in combination with two-photon fluorescence microscopy, has proven successful at such an endeavor. Here, we demonstrate three-photon fluorescence imaging with an axially extended Bessel focus. We use an axicon-based module that allowed for the generation of Bessel foci of varying numerical apertures and axial lengths, and apply this volumetric imaging tool to image mouse brain slices and for in vivo imaging of the mouse brain.

Abstract

El crecimiento demográfico mundial está provocando un aumento en la demanda de alimentos de ori gen animal, entre la que se encuentra la carne y leche de rumiantes. Pero esta necesidad se enfrenta a una ideología que denuesta este sector por considerar su alimentación en clara competencia con los re cursos destinados al consumo humano, al tiempo que contribuye a la degradación de los suelos y es pacios naturales. Ante esta situación, es urgente buscar nuevas fuentes que puedan suplir en parte de terminados componentes de la ración, al tiempo que aporten un valor añadido al producto final (carne/leche). El consumo de harina de insectos, debidamente procesados y bajo una estricta regla mentación que asegure su seguridad, emerge como una posibilidad que podría extenderse al ganado bovino, ovino y caprino. En esta revisión pretendemos ofrecer los últimos estudios en lo que respecta a la legislación europea, los insectos que están permitidos, así como todos aquellos estudios realizados in vivo e in vitro en es tas especies. La bibliografía citada demuestra que, si bien la calidad de la harina de insectos depende de la especie y forma de cría, su introducción como suplemento en la ración, no causa ningún perjui cio en dinámica ruminal ni la producción, pudiendo constituir una solución rentable y respetuosa con el medio ambiente., World population growth is causing an increase in the demand for food of animal origin, including ruminant meat and milk. But this need is confronted by an ideology rejecting this sector because it considers its food to be in direct competition with the resources destined for human consumption while contributing to the degradation of soils and natural spaces. Faced with this situation, it is urgent to look for new sources that can partly supply some components of the ration, while providing an added value to the final product (meat/milk). The consumption of insect meal, duly processed and under strict regulations that ensure its safety, emerges as a possibility that could be extended to cattle, sheep and goats. In this revision, we intend to offer the latest studies regarding European legislation, the allowed insects, and all those studies carried out in vivo and in vitro in these species. All of them show that, although the quality of insect meal depends on the species and rearing method, its introduction as a supplement in the ration, does not cause any damage in ruminal dynamics or production and can be a profitable and environmentally friendly solution

43. Development of a 99mTc-labeled tetrazine for pretargeted SPECT imaging using an alendronic acid-based bone targeting model

Abstract

Pretargeting, which is the separation of target accumulation and the administration of a secondary imaging agent into two sequential steps, offers the potential to improve image contrast and reduce radiation burden for nuclear imaging. In recent years, the tetrazine ligation has emerged as a promising approach to facilitate covalent pretargeted imaging due to its unprecedented kinetics and bioorthogonality. Pretargeted bone imaging with TCO-modified alendronic acid (Aln-TCO) is an attractive model that allows the evaluation of tetrazines in healthy animals without the need for complex disease models or targeting regimens. Recent structure-activity relationship studies of tetrazines evaluated important parameters for the design of potent tetrazine-radiotracers for pretargeted imaging. However, limited information is available for 99mTc-labeled tetrazines. In this study, four tetrazines intended for labeling with fac-[99mTc(OH2)3 (CO)3]+ were synthesized and evaluated using an Aln-TCO mouse model. 3,6-bis(2-pyridyl)-1,2,4,5-Tz without additional linker showed higher pretargeted bone uptake and less background activity compared to the same scaffold with a PEG8 linker or 3-phenyl-1,2,4,5-Tz-based compounds. Additionally, improved bone/blood ratios were observed in pretargeted animals compared to animals receiving directly labeled Aln-TCO. The results of this study implicate 3,6-bis(2-pyridyl)-1,2,4,5-Tz as a promising scaffold for potential 99mTc-labeled tetrazines.

44. Fluid Intelligence, Working Memory, Reading Fluency and Comprehension in Chilean School Children

Abstract

Introduction: Existing studies of the variables that can impact reading comprehension at intermediate levels of Primary Education are not entirely conclusive. The present study analyzes the contribution to reading comprehension made by domain-general skills, such as fluid intelligence (Gf) and working memory (WM), and domain-specific skills, such as accuracy and speed. Method: A sample of 98 Chilean third graders participated in the study. Measures of fluid intelligence, working memory, reading fluency (speed and accuracy) and reading comprehension were administered. Results: The study revealed the significant contribution made by the general and specific domains to reading comprehension. The unique contribution of Gf and WM to the prediction of reading comprehension was similar to that of reading speed. Accuracy was not found to make a significant contribution. Discussion and Conclusions: The implications and relevance of the findings with regard to psycho-pedagogical and educational practice at intermediate levels of Primary School are discussed.

Abstract

Early detection and intervention play a crucial role in the prognosis of children with specific learning disabilities. Due to the complexity of the writing process and its persistence into adulthood, tools for early detection are essential. Curriculum-based measures (CBMs) are quick, reliable, and evidence-based tools used for early assessments. This systematic review aims to summarize the technical features of CBM in writing for kindergarten students following the PRISMA guidelines. A total of 311 articles from PsycINFO, SCOPUS, ERIC, and WOS were examined, and five studies met the inclusion criteria. Production measures and transcription skills were the most studied. The reviewed tasks demonstrated criterion validity, reliability, sensitivity to the timing of assessment, or diagnostic accuracy in at least one of their measures, with a greater presence of the dependent production measures. The findings emphasize the scarcity of evidence-based tools for identifying early writing difficulty, underscoring the need for rigorous research in early childhood education. [ABSTRACT FROM AUTHOR]

Abstract

Walter Dellinger's 1994 Memorandum Opinion for the Office of Legal Counsel ("OLC") defining when the President may decline to enforce a law of Congress remains one of his lasting contributions to the theory and practice of the separation of powers. Despite articulating a departmentalist vision for relations between the political branches, however, the Memo accepts judicial supremacy as a given. It emphasizes that the Supreme Court has the final word on contested constitutional questions and that enforcement judgments must hew to judicial doctrine. For decades, scholars, officials, and advocates of various ideological stripes have challenged judicial supremacy, even as it has remained central to executive branch practice. Typically, these challenges underscore the authority and even primacy of Congress, the representative legislature, over constitutional meaning. But some critics have called on the President to reject judicial dominance, too. In this tribute to Dellinger's storied life and career, I offer a hypothetical follow-up opinion to his 1994 Memo, entitled Presidential Authority to Decline to Follow Supreme Court Opinions. I suggest how OLC might define the circumstances under which it would be permissible for the Executive to continue enforcing a law that the Supreme Court has declared unconstitutional on the ground that the Court got it wrong. Even for those who would regard such a call as implausible or dangerous, the varieties of resistance available to the President and the Executive Branch warrant attention as part of the larger debate over whose province and duty it is to say what the law means. Abstract

Eating disorders (EDs) manifest as persistent disruptions in eating habits or related behaviors, significantly impacting physical health and psychosocial well-being. Nutritional assessment in ED patients is crucial for monitoring treatment efficacy. While dual-energy X-ray absorptiometry (DEXA) remains standard, interest in alternative methods such as bioelectrical impedance vector analysis (BIVA) and Nutritional Ultrasound® (NU) has risen due to their affordability and portability. Additionally, hand dynamometry offers a user-friendly approach to assessing grip strength (HGS), indicative of nutritional status. A prospective study was carried out to evaluate the utility of BIVA, NU®, and HGS in 43 female AN patients. Measurements were taken at baseline and hospital discharge. A total of 41 patients completed the study. After the intervention, numerous BIVA-related parameters such as fat (3.5 ± 2 kg vs. 5.3 ± 2.7 kg, p < 0.001) and free fat mass (33.9 ± 3.8 kg vs. 37.5 ± 4.1 kg, p < 0.001) were partially restored. Similarly, Nutritional Ultrasound® showed promising results in assessing body composition changes such as total abdominal fat tissue (0.5 ± 0.3 cm vs. 0.9 ± 0.3 cm, p < 0.05). In the same way, rectus femoris cross-sectional area values correlated with clinical outcomes such as free fat mass (0.883, p < 0.05) and appendicular muscle mass (0.965, p < 0.001). HGS reached the normality percentile after the intervention (21.6 ± 9.1 kg vs. 25.9 ± 12.3 kg, p < 0.05), demonstrating a significant association between grip strength and body composition parameters such as free fat mass (0.658, p < 0.001) and appendicular muscle mass (0.482, p < 0.001). Incorporating BIVA-, NU®-, and HGS-enhanced nutritional assessment into the treatment of AN patients offers cost-effective, portable, and non-invasive alternatives to DEXA. These techniques offer valuable insights into changes in body composition and nutritional status, which, in turn, facilitate treatment monitoring and contribute to improved patient outcomes. Abstract

Although the effect of teacher expectations on students' academic performance has been verified, few studies have focused on establishing which factors affect the formation of these expectations, especially ones that consider previous student performance and the professional experience of the teacher as explanatory factors. The aim of this study was to examine the effect of these factors on the formation of teacher expectations. The sample comprised 21 teachers from 11 schools, with low, medium, and high experience levels, as well as 363 firstyear primary-school students from Chile (ages 6-7), whose academic performance was measured at the start of the school year. Teacher expectations were measured part-way through the school year. A mixed ANOVA analysis made it possible to assess whether teacher expectation varies depending on previous performance and teacher experience. The results indicated that previous performance and teacher experience both have a significant effect on how expectations form.
