1. Species diversity in Pseudocercospora.

Abstract: Species of Pseudocercospora are commonly associated with leaf and fruit spots on diverse plant hosts in sub-tropical and tropical regions. Pseudocercospora spp. have mycosphaerella-like sexual morphs, but represent a distinct genus in Mycosphaerellaceae (Mycosphaerellales, Dothideomycetes). The present study adds a further 29 novel species of Pseudocercospora from 413 host species representing 297 host genera occurring in 60 countries and designates four epitypes and one lectotype for established names. This study recognises 329 species names, with an additional 69 phylogenetic lineages remaining unnamed due to difficulty in being able to unambiguously apply existing names to those lineages. To help elucidate the taxonomy of these species, a phylogenetic tree was generated from multi-locus DNA sequence data of the internal transcribed spacers and intervening 5.8S nuclear nrRNA gene (ITS), partial actin (actA), and partial translation elongation factor 1-alpha (tef1), as well as the partial DNA-directed RNA polymerase II second largest subunit (rpb2) gene sequences. Novel species described in this study include those from various countries as follows: Australia, Ps. acaciicola from leaf spots on Acacia sp., Ps. anopter from leaf spots on Anopterus glandulosus, Ps. asplenii from leaf spots on Asplenium dimorphum, Ps. australiensis from leaf spots on Eucalyptus gunnii, Ps. badjensis from leaf spots on Eucalyptus badjensis, Ps. erythrophloeicola from leaf spots on Erythrophleum chlorostachys, Ps. grevilleae from leaf spots on Grevillea sp., Ps. lophostemonigena from leaf spots on Lophostemon confertus, Ps. lophostemonis from leaf spots on Lophostemon lactifluus, Ps. paramacadamiae from leaf spots on Macadamia integrifolia, Ps. persooniae from leaf spots on Persoonia sp., Ps. pultenaeae from leaf spots on Pultenaea daphnoides, Ps. tristaniopsidis from leaf spots on Tristaniopsis collina, Ps. victoriae from leaf spots on Eucalyptus globoidea. Results obtained in the present study contribute to a better understanding of the host specificity and distribution in Pseudocercospora spp., many of which represent important pathogens of food or fibre crops, or organisms of quarantine concern. Abstract: Novel species of fungi described in this study include those from various countries as follows: Australia, Aschersonia mackerrasiae on whitefly, Cladosporium corticola on bark of Melaleuca quinquenervia, Penicillium nudgee from soil under Melaleuca quinquenervia, Pseudocercospora blackwoodiae on leaf spot of Persoonia falcata, and Pseudocercospora dalyelliae on leaf spot of Senna alata. Bolivia, Aspicilia lutzoniana on fully submersed siliceous schist in high-mountain streams, and Niesslia parviseta on the lower part and apothecial discs of Erioderma barbellatum onatwig. Brazil, Cyathus bonsai on decaying wood, Geastrum albofibrosum from moist soil with leaf litter, Laetiporus pratigiensis on a trunk of a living unknown hardwood tree species, and Scytalidium synnematicum on dead twigs of unidentified plant. Abstract: Seven Fusarium species complexes are treated, namely F. aywerte species complex (FASC) (two species), F. buharicum species complex (FBSC) (five species), F. burgessii species complex (FBURSC) (three species), F. camptoceras species complex (FCAMSC) (three species), F. chlamydosporum species complex (FCSC) (eight species), F. citricola species complex (FCCSC) (five species) and the F. concolor species complex (FCOSC) (four species). New species include Fusicolla elongata from soil (Zimbabwe), and Neocosmospora geoasparagicola from soil associated with Asparagus officinalis (Netherlands). New combinations include Neocosmospora akasia, N. awan, N. drepaniformis, N. duplosperma, N. geoasparagicola, N. mekan, N. papillata, N. variasi and N. warna. Newly validated taxa include Longinectria gen. nov., L. lagenoides, L. verticilliforme, Fusicolla gigas and Fusicolla guangxiensis. Furthermore, Fusarium rosicola is reduced to synonymy under N. brevis. Finally, the genome assemblies of Fusarium secorum (CBS 175.32), Microcera coccophila (CBS 310.34), Rectifusarium robinianum (CBS 430.91), Rugonectria rugulosa (CBS 126565), and Thelonectria blattea (CBS 952.68) are also announced here

Classification �� Nectriaceae, Hypocreales, Sordariomycetes. On SNA and CLA, sporulation abundant from aerial conidiophores and sporodochia. Notes �� Fusarium chuoi resides in the Asian clade of the Fusarium fujikuroi species complex (FFSC: O��Donnell et al. 1998, Yilmaz et al. 2021, Crous et al. 2021b). Based on nucleotide searches using the Fusarium Pairwise ID engine on the Fusarioid-ID database (www.fusarium.org, Crous et al. 2021) the closest hit using the ITS sequence was Fusarium siculi (strain CBS 142422; identities = 449/450 (99 %), no gaps). The closest hit using the LSU sequence was F. siculi (strain CBS 142422; identities = 804/805 (99 %), no gaps). Closest hit using the cmdA sequence was Fusarium fractiflexum (strain NRRL 28852; identities = 426/434 (98 %), no gaps). Closest hit using the rpb1 sequence was F. fujikuroi (strain NRRL 13566; identities = 687/702 (98 %), no gaps). Closest hit using the rpb2 sequence was Fusarium globosum (strain CBS 428.97; identities = 856/867 (98 %), no gaps). Closest hit using the tef1 sequence was F. fractiflexum (strain NRRL 28852; identities = 619/643 (96 %), 2 gaps (0.3 %)). The phylogenetic results, however, showed that F. chuoi is not directly related to any of the previously described species of FFSC (see Suppl. material FP1353), clustering as the second basal-most species of that clade after F. sacchari. Asian Fusarium spp. in the FFSC are characterised by mono-and polyphialides producing oval to ellipsoid, rarely pyriform to globose (i.e., F. annulatum, F. fujikuroi and F. globosum) microconidia organized in chains or false heads; 3��5-septate sporodochial conidia and lacking chlamydospores. The elaborate, profusely branched aerial conidiophores of F. chuoi are comparable to those of F. concentricum, F. lumajangense, F. mangiferae and F. sacchari, all the latter species producing oval, ellipsoidal to allantoid microconidia on false heads.Aerial conidiophores of F. chuoi, however, mostly produce macroconidia, while microconidia grouped on false heads are restricted to short, mostly unbranched and prostrate conidiophores formed on the surface on the culture media. Several Asian species of the FFSC have been reported from Musa spp. i.e., F. annulatum, F. concentricum, F. fujikuroi, F. lumajangense and F. sacchari (Leslie & Summerell 2006, Maryani et al. 2019, Farr & Rossman 2021). The two strains representing F. chuoi were isolated as endophytes from asymptomatic seeds of wild banana (Musa itinerans), which had been collected predispersal and stored in the Millennium Seed Bank for ~2.5 years at -20 �� C prior to isolation. Abstract: Recent publications have argued that there are potentially serious consequences for researchers in recognising distinct genera in the terminal fusarioid clade of the family Nectriaceae. Thus, an alternate hypothesis, namely a very broad concept of the genus Fusarium was proposed. In doing so, however, a significant body of data that supports distinct genera in Nectriaceae based on morphology, biology, and phylogeny is disregarded. A DNA phylogeny based on 19 orthologous protein-coding genes was presented to support a very broad concept of Fusarium at the F1 node in Nectriaceae. Here, we demonstrate that re-analyses of this dataset show that all 19 genes support the F3 node that represents Fusarium sensu stricto as defined by F. sambucinum (sexual morph synonym Gibberella pulicaris). The backbone of the phylogeny is resolved by the concatenated alignment, but only six of the 19 genes fully support the F1 node, representing the broad circumscription of Fusarium. Furthermore, a re-analysis of the concatenated dataset revealed alternate topologies in different phylogenetic algorithms, highlighting the deep divergence and unresolved placement of various Nectriaceae lineages proposed as members of Fusarium. Species of Fusarium s. str. are characterised by Gibberella sexual morphs, asexual morphs with thin- or thick-walled macroconidia that have variously shaped apical and basal cells, and trichothecene mycotoxin production, which separates them from other fusarioid genera. Here we show that the Wollenweber concept of Fusarium presently accounts for 20 segregate genera with clear-cut synapomorphic traits, and that fusarioid macroconidia represent a character that has been gained or lost multiple times throughout Nectriaceae. Thus, the very broad circumscription of Fusarium is blurry and without apparent synapomorphies, and does not include all genera with fusarium-like macroconidia, which are spread throughout Nectriaceae (e.g., Cosmosporella, Macroconia, Microcera). Abstract: This article is the 13th contribution in the Fungal Diversity Notes series, wherein 125 taxa from four phyla, ten classes, 31 orders, 69 families, 92 genera and three genera incertae sedis are treated, demonstrating worldwide and geographic distribution. Fungal taxa described and illustrated in the present study include three new genera, 69 new species, one new combination, one reference specimen and 51 new records on new hosts and new geographical distributions. Three new genera, Cylindrotorula (Torulaceae), Scolecoleotia (Leotiales genus incertae sedis) and Xenovaginatispora (Lindomycetaceae) are introduced based on distinct phylogenetic lineages and unique morphologies.

Abstract: Couch smut, caused by Ustilago cynodontis, is an economically important disease of Cynodon dactylon, the most widely planted warm season turfgrass in Australia. The disease is distributed across all states and mainland territories of Australia, and is found worldwide wherever the host plant is present. The most characteristic disease symptoms are expressed at the flowering stage, when the inflorescence is partly or entirely destroyed and covered by a mass of black powdery spores. However, there are also less obvious impacts of the disease on plant growth such as a more erect growth habit, and a reduced rate of stolon extension and root development, leading to lower levels of tolerance of the turf to wear during usage, and greater amounts of wastage during harvest due to the roll breaking at points of infection. Ustilago cynodontis was first described nearly 130 years ago but pathogen biology and disease epidemiology are still poorly understood, hindering development of disease management strategies. We review the current knowledge and understanding of couch smut biology, the history of its occurrence, distribution and impact, and existing disease management practices to identify the gaps in knowledge that require further research.

Abstract: In 2011, patches of lodged plants of Helianthus annuus (commercial sunflower) with stem lesions were observed in a commercial crop at Kingsthorpe, Queensland, Australia. Several species of Diaporthe were consistently isolated from the lesions. Diaporthe novem was identified by DNA sequence analysis. In following surveys, Diaporthe novem was found to be associated with other crop species including Cicer arietinum (chickpea), Glycine max (soybean), Lupinus alba (lupin), Sorghum bicolor (sorghum) Vicia faba (faba bean) and Vigna radiata (mungbean), and with the weed species Datura stramonium (common thornapple), Helianthus annuus (wild-type sunflower), Malva parviflora (small flowered mallow), Rapistrum rugosum (turnip weed), Sambucus gaudichaudiana (wild elderberry), Sisymbrium orientale (indian hedge mustard), Sonchus oleraceus (sowthistle), Verbena sp., Vicia sativa (common vetch), and Xanthium strumarium (noogoora burr). In pathogenicity tests, isolates of D. novem from sunflower were highly virulent when re-inoculated on commercial sunflower varieties. This study has identified D. novem as a frequent cause of stem canker of sunflower in the eastern cropping areas of Australia, and extends the known host range of D. novem.

Abstract: In July 2016, peanuts (Arachis hypogaea L. cvs. Holt, Kairi) with symptoms of stem and peg dieback were collected from Kingaroy, Childers, and Bundaberg regions in Queensland, Australia. Pycnidia of Diaporthe spp. were visible in brown lesions on stems and on live and shriveled pegs. Affected tissues were surface sterilized in 1% v/v NaOCl for 3 min, rinsed with sterile distilled water, incubated on water agar with 100 µg/ml streptomycin sulfate at 23 to 25°C under ambient light. After 1 to 14 days, conidia were streaked onto potato dextrose agar with 100 µg/ml streptomycin sulfate (PDAS) and incubated as above. After 24 h, single germinating conidia were transferred to PDAS. Mycelial DNA was extracted from 80 single conidial isolates using the Bioline Isolate II Genomic DNA extraction kit. Primers for the internal transcribed spacer region (ITS) and partial region of the translation elongation factor 1-alpha (TEF) were as described in Thompson et al. (2015). Both ITS and TEF loci were amplified with Biolabs Hot Start Taq 2x Master Mix Enzyme. PCR products were purified using Bioline Sure Clean Plus and sequenced by Macrogen Incorporated (Seoul, Korea) with the amplification primers. Sequences were assembled with Geneious v.9.1. Species morphological descriptions are in Thompson et al. 2011, 2015, and Gao et al. 2016. Using BLAST sequence analysis and culture morphology, species were identified as D. kongii (four isolates), D. masirevicii (42), D. ueckerae (syn. D. miriciae) (27), or Diaporthe sp. (seven) (Gao et al. 2016; Thompson et al. 2011, 2015). A representative isolate of D. kongii (BRIP 64964d), D. masirevicii (BRIP 64786a), and D. ueckerae (BRIP 64785a) was deposited into the Queensland Plant Pathology Herbarium (Brisbane, Australia) and was used for pathogenicity testing.
