Water remediation, acknowledged as a significant scientific topic, guarantees the safety of drinking water, considering the diverse range of pollutants that can contaminate it. Among these pollutants, arsenic stands out as a particularly severe threat to human health, significantly compromising the overall quality of life. Despite widespread awareness of the harmful effects of arsenic poisoning, there remains a scarcity of literature on the utilization of biobased polymers as sustainable alternatives for comprehensive arsenic removal in practical concern. Cellulose and chitosan, two of the most prevalent biopolymers in nature, provide a wide range of potential benefits in cutting-edge industries, including water remediation. Nanocomposites derived from cellulose and chitosan offer numerous advantages over their larger equivalents, including high chelating properties, cost-effective production, strength, integrity during usage, and the potential to close the recycling loop. Within the sphere of arsenic remediation, this Review outlines the selection criteria for novel cellulose/chitosan-nanocomposites, such as scalability in synthesis, complete arsenic removal, and recyclability for technical significance. Especially, it aims to give an overview of the historical development of research in cellulose and chitosan, techniques for enhancing their performance, the current state of the art of the field, and the mechanisms underlying the adsorption of arsenic using cellulose/chitosan nanocomposites. Additionally, it extensively discusses the impact of shape and size on adsorbent efficiency, highlighting the crucial role of physical characteristics in optimizing performance for practical applications. Furthermore, this Review addresses regeneration, reuse, and future prospects for chitosan/cellulose-nanocomposites, which bear practical relevance. In this work, we have synthesized four different color (yellow, orange, green, and blue (multicolor)) silver nanostructures (AgNSs) by chemical reduction method where silver nitrate, sodium borohydride and hydrogen peroxide were used as reagents. The as-synthesized multicolor AgNSs were successfully functionalized with bovine serum albumin (BSA) and applied as a colorimetric sensor for the assaying of metal cations (Cr 3+ , Hg 2+ , and K + ). The addition of metal ions (Cr 3+ , Hg 2+ , and K + ) into BSA functionalized AgNSs (BSA-AgNSs) causes the aggregation of BSA-AgNSs, and are accompanied by visual color changes with red or blue shift in the surface plasmon resonance (SPR) band of BSA-AgNSs. The BSA-AgNSs show different SPR characteristic for each metal ions (Cr 3+ , Hg 2+ , and K + ) with exhibiting different spectral shift and color change. The yellow color BSA-AgNSs (Y-BSA-AgNSs) act as a probe for sensing Cr 3+ , orange color BSA-AgNSs (O-BSA-AgNSs) act as probe for Hg 2+ ion assay, green color BSA-AgNSs (G-BSA-AgNSs) act as a probe for the assaying of both K + and Hg 2+ , and blue color BSA-AgNSs (B-BSA-AgNSs) act as a sensor for colorimetric detection of K + ion. The detection limits were found to be 0.26 μM for Cr 3+ (Y-BSA-AgNSs), 0.14 μM for Hg 2+ (O-BSA-AgNSs), 0.05 μM for K + (G-BSA-AgNSs), 0.17 μM for Hg 2+ (G-BSA-AgNSs), and 0.08 μM for K + (B-BSA-AgNSs), respectively. Nitrogen is a vital nutrient for agricultural, and a defieciency of it causes stagnate cotton growth and yield penalty. Farmers rely heavily on N over-application to boost cotton output, which can result in decreased lint yield, quality, and N use efficiency (NUE). Therefore, improving NUE in cotton is most crucial for reducing environmental nitrate pollution and increasing farm profitability. Well-defined management practices, such as the type of sources, N-rate, application time, application method, crop growth stages, and genotypes, have a notable impact on NUE. Different N formulations, such as slow and controlled released fertilizers, have been shown to improve N uptake and, NUE. Increasing N rates are said to boost cotton yield, although high rates may potentially impair the yield depending on the soil and environmental conditions. This study comprehensively reviews various factors including agronomic and environmental constraints that influence N uptake, transport, accumulation, and ultimately NUE in cotton. Furthermore, we explore several agronomic and molecular approaches to enhance efficiency for better N uptake and utilization in cotton. Seed priming technique has a marvelous potential in enhancing seed germination and crop establishment under limited soil moisture conditions, which ultimately increases yield. Therefore, we investigated the effects of seed priming on physiology, growth, yield and antioxidant defense system of pearl millet (Pennisetum glaucum L.) under rain-fed condition. The experiments were conducted under laboratory as well as field conditions comprising three treatments i.e., non-primed seeds (control, T0), priming with tap water (hydropriming) (T1) and priming with 2% KNO3 2% for 6 hours at 25°C followed by shade drying (T2). The results showed that chlorophyll content (10.37-14.15%) and relative water content (RWC) (12.70-13.01%) increased whereas proline (-19.44 to -25%) and soluble sugar (-15.51 to -29.13%) contents decreased on account of seed priming in pearl millet under field conditions. The seed priming significantly improved the plant height, final plant stand and grain weight which resulted in increased yield. Enhanced activities of superoxide dismutase (SOD) (5.89 to 8.10 unit/g/seed/min), catalase (CAT) (22.54 to 39.67 µmol/min/g/seed) and ascorbate peroxidase (APX) (8.92 to 22.10 µmol/cm/min/g) and concomitant decrease in H2O2 and malondialdehyde (MDA) content suggests their role in imparting oxidative tolerance at initial stages of growth in primed seed. The lab studies suggest that the improved yield might be attributes to increased seed germination and seedling vigor. Objective: To gain better insight into the extent of secondary bacterial and fungal infections in hospitalized patients in India, and to assess how these alter the course of coronavirus disease 2019 (COVID-19) so that control measures can be suggested., Methods: In this retrospective, multicentre study, the data of all patients who tested positive for severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) on reverse transcriptase polymerase chain reaction (RT-PCR), admitted to hospital between March 2020 and July 2021, were accessed from the electronic health records of a network of 10 hospitals across five states in North India., Results: Of 19,852 patients testing positive for SARS-CoV-2 on RT-PCR and admitted to the study hospitals during the study period, 1940 (9.8%) patients developed secondary infections (SIs). Patients with SIs were, on average, 8 years older than patients without SIs (median age 62.6 vs 54.3 years; P <0.001). The risk of SIs was significantly ( P <0.001) associated with age, severity of disease at admission, diabetes, admission to the intensive care unit (ICU), and ventilator use. The most common site of infection was urine (41.7%), followed by blood (30.8%) and sputum/bronchoalveolar lavage/endotracheal fluid (24.8%); the least common was pus/wound discharge (2.6%). Gram-negative bacilli (GNB) were the most common organisms (63.2%), followed by Gram-positive cocci (GPC) (19.6%) and fungi (17.3%). Most patients with SIs were on multiple antimicrobials. The most commonly used antibiotics against GNB were beta-lactam/beta-lactamase inhibitors (76.9%), carbapenems (57.7%), cephalosporins (53.9%), and antibiotics against carbapenem-resistant Enterobacteriaceae (47.1%). Empirical use of antibiotics against GPC was seen in 58.9% of patients with SIs, and empirical use of antifungals was observed in 56.9% of patients with SIs. The average length of hospital stay for patients with SIs was almost twice as long as that of patients without SIs (median 13 vs 7 days). Overall mortality among patients with SIs (40.3%) was more than eight times higher than that among patients without SIs (4.6%). Only 1.2% of patients with SIs with mild COVID-19 at admission died, compared with 17.5% of those with moderate COVID-19 at admission and 58.5% of those with severe COVID-19 at admission ( P <0.001). The mortality rate was highest in patients with bloodstream infections (49.8%), followed by those with hospital-acquired pneumonia (47.9%), urinary tract infections (29.4%), and skin and soft tissue infections (29.4%). The mortality rate in patients with diabetes with SIs was 45.2%, compared with 34.3% in those without diabetes ( P <0.001)., Conclusions: SIs complicate the course of patients hospitalized with COVID-19. These patients tend to have a much longer hospital stay, a higher requirement for oxygen and ICU care, and a significantly higher mortality rate compared with those without SIs. The groups most vulnerable to SIs are patients with more severe COVID-19, elderly patients and patients with diabetes. Judicious empirical use of combination antimicrobials in these groups of vulnerable patients can save lives. The impact of elevated temperature at the reproductive stage of a crop is one of the critical limitations that influence crop growth and productivity globally. This study was aimed to reveal how sowing time and changing field temperature influence on the regulation of oxidative stress indicators, antioxidant enzymes activity, soluble sugars (SS), and amino acids (AA) in Indian Mustard. The current study was carried out during the rabi 2017-2018 and 2018-2019 where, five varieties of mustard viz . Pusa Mustard 25 (PM-25) (V1), PM-26 (V2), BPR-541-4 (V3), RH-406 (V4), and Urvashi (V5) were grown under the field conditions on October 30 (normal sowing; S1), November 18 (late sowing; S2) and November 30 (very late sowing; S3) situations. The S1 and S3 plants, at mid-flowering stage, showed a significant variation in accumulation of SS (8.5 and 17.3%), free AA (235.4 and 224.6%), and proline content (118.1 and 133%), respectively, and played a crucial role in the osmotic adjustment under stress. The results showed that S3 sowing, exhibited a significant induction of the hydrogen peroxide (H 2 O 2 ) (110.2 and 86.6%) and malondialdehyde (23.5 and 47.5%) concentrations, respectively, which indicated the sign of oxidative stress in plants. Interestingly, the polyphenol oxidase, peroxidase, superoxide dismutase, and catalase enzyme activities were also significantly increased in S3 plants compared to S1 plants, indicating their significant roles in ameliorating the oxidative stress. Furthermore, the concentration of fatty acid levels such as palmitic, stearic, oleic, and linoleic acids level also significantly increased in S3 plants, which influenced the seed and oil quality. The study suggests that the late sowing significantly impaired the biochemical mechanisms in Indian mustard. The efficiency with which plants use nutrients to create biomass and/or grain is determined by the interaction of environmental and plant intrinsic factors. The major macronutrients, especially nitrogen (N), limit plant growth and development (1.5-2% of dry biomass) and have a direct impact on global food supply, fertilizer demand, and concern with environmental health. In the present time, the global consumption of N fertilizer is nearly 120 MT (million tons), and the N efficiency ranges from 25 to 50% of applied N. The dynamic range of ideal internal N concentrations is extremely large, necessitating stringent management to ensure that its requirements are met across various categories of developmental and environmental situations. Furthermore, approximately 60 percent of arable land is mineral deficient and/or mineral toxic around the world. The use of chemical fertilizers adds to the cost of production for the farmers and also increases environmental pollution. Therefore, the present study focused on the advancement in fertilizer approaches, comprising the use of biochar, zeolite, and customized nano and bio-fertilizers which had shown to be effective in improving nitrogen use efficiency (NUE) with lower soil degradation. Consequently, adopting precision farming, crop modeling, and the use of remote sensing technologies such as chlorophyll meters, leaf color charts, etc. assist in reducing the application of N fertilizer. This study also discussed the role of crucial plant attributes such as root structure architecture in improving the uptake and transport of N efficiency. The crosstalk of N with other soil nutrients plays a crucial role in nutrient homeostasis, which is also discussed thoroughly in this analysis. Current study presents a facile synthesis method for thiol functionalised silica microsphere loaded polymeric hydrogel. Silica microspheres were synthesised as core shell particles by sol-gel method followed by hydrolysis and condensation reaction to form the silica core. The silica was functionalised with thiol and the functionalised silica microsphere was subsequently impregnated into polymeric alginate matrix to form thiol functionalised silica microspheres loaded alginate hydrogel beads (SH-SiO 2 MS-Ca-Alg). The developed components and final products were characterised by BET, FTIR, DLS, SEM-EDS. The developed SH-SiO 2 MS-Ca-Alg hydrogel beads were used for efficient removal of Pb and Cd [72-97 % for Pb; 60-85 % for Cd at concentration range of 0.1-100 μg mL -1 and optimum pH 5-7] from aquatic medium. The sorption capacities evaluated for Pb and Cd from Langmuir isotherm were 127.99 and 70.68 mgg -1 respectively. Uptake kinetics, isotherm, thermodynamics, intraparticle diffusion studies were carried out for both Pb (II) and Cd (II). Zinc is an essential element that is also renowned for widespread contamination and toxicity at high concentrations. The present study was carried out to analyze the responses induced by lower, as well as higher, doses of zinc (0-200 mg/L), in the form of zinc oxide nanoparticles (ZnO NPs) in wheat and maize, for a period of 21 days. Accumulation of zinc increases with increasing Zn doses in both wheat and maize, with higher doses being in wheat (121 mg/kg in root and 66 mg/kg in shoot) than in maize (95 mg/kg in root and 48 mg/kg in shoot). The activity of alpha-amylase showed increase, while that of dehydrogenase decline, in response to ZnO NPs. The length and biomass of plants and photosynthetic pigments increased slightly upon ZnO NPs supply. Malondialdehyde content showed a progressive increase in root and shoot of both plants. However, in response, antioxidant enzymes (superoxide dismutase, ascorbate peroxidase, guaiacol peroxidase, and catalase) showed increase up to lower concentrations (100 mg/L) of ZnO NPs but decline variably at higher levels (150-200 mg/L) in wheat and maize. The results suggest that lower supply of ZnO NPs (100 mg/L) could be stimulatory to the growth of plants and can be recommended as a Zn fertilizer source for crop production.

In the era of rapid climate change, abiotic stresses are the primary cause for yield gap in major agricultural crops. Among them, salinity is considered a calamitous stress due to its global distribution and consequences. Salinity affects plant processes and growth by imposing osmotic stress and destroys ionic and redox signaling. It also affects phytohormone homeostasis, which leads to oxidative stress and eventually imbalances metabolic activity. In this situation, signaling compound crosstalk such as gasotransmitters [nitric oxide (NO), hydrogen sulfide (H 2 S), hydrogen peroxide (H 2 O 2 ), calcium (Ca), reactive oxygen species (ROS)] and plant growth regulators (auxin, ethylene, abscisic acid, and salicylic acid) have a decisive role in regulating plant stress signaling and administer unfavorable circumstances including salinity stress. Moreover, recent significant progress in omics techniques (transcriptomics, genomics, proteomics, and metabolomics) have helped to reinforce the deep understanding of molecular insight in multiple stress tolerance. Currently, there is very little information on gasotransmitters and plant growth regulator crosstalk and inadequacy of information regarding the integration of multi-omics technology during salinity stress. Therefore, there is an urgent need to understand the crucial cell signaling crosstalk mechanisms and integrative multi-omics techniques to provide a more direct approach for salinity stress tolerance. To address the above-mentioned words, this review covers the common mechanisms of signaling compounds and role of different signaling crosstalk under salinity stress tolerance. Background and Aims: Excessive bleeding is a major concern in functional endoscopic sinus surgery (FESS) under general anaesthesia; this can be decreased by various hypotensive agents. This study was conducted to compare the hypotensive effectiveness and haemodynamic stability of dexmedetomidine and clonidine in patients undergoing elective FESS., Methods: In this prospective double-blinded interventional study, 70 adult patients of either sex, 20-50 years of age, posted for elective FESS were randomly assigned to two groups. Group A received a loading dose of intravenous (IV) dexmedetomidine 1 μg/kg, followed by infusion of 1 μg/kg/h, and group B received a loading dose of IV clonidine 2 μg/kg, followed by 1 μg/kg/h infusion. Surgical field quality, emergence time, sedation score, visual analogue score, recovery profile and haemodynamic parameters were recorded. Statistical analysis was done by Student's unpaired t -test to evaluate the significance of normally distributed variables, whereas Mann-Whitney test and Chi-square test were used for ordinal data and categorical variables and proportions, respectively., Results: In both the groups, target mean arterial pressure (MAP) of 65-70 mmHg and improved surgical field quality were achieved. MAP and heart rate (HR) were statistically significantly lower in the dexmedetomidine group with a longer duration of post-operative analgesia ( P = 0.001). None of the groups showed any statistically significant adverse effects., Conclusions: Both dexmedetomidine and clonidine can be used for controlled hypotension to improve surgical field quality in FESS. Background: The present study aimed to evaluate and compare the efficacy of 17% ethylenediaminetetraacetic acid (EDTA), 18% etidronic acid, 10% citric acid, and 7% maleic acid in the removal of smear layer at the apical third of the root canals., Materials and Methods: Sixty single-rooted teeth were equally divided into four study groups ( n = 15), according to the type of irrigant used (17% EDTA, 18% etidronic acid, 10% citric acid, and 7% maleic acid) to remove the smear layer effectively from apical third of root canal. In each group, respective irrigant was used with 5.25% of sodium hypochlorite during instrumentation. Each study sample was then sectioned longitudinally and removal of smear layer was observed using a scanning electron microscope (SEM) at 2000×., Results: A 7% maleic acid revealed better smear layer removal than all other three groups at apical third. A 10% citric acid was found to be more efficient than EDTA and etidronic acid. The intergroup comparison was performed using Mann-Whitney U test, and there was no significant difference between all the study groups, except maleic acid., Conclusion: The present study compared the effect of various irrigants as an adjunct with 5.25% of sodium hypochlorite for root canal irrigation during and after instrumentation. The use of irrigants aids in the removal of the smear layer from the root canals, thereby increasing the success rate of endodontic therapy., Clinical Significance: This study supports the hypothesis that a thorough use of root canal irrigants can efficiently remove the smear layer which is the key for successful root canal treatment. The present study was carried out to investigate the impact of various concentrations of SiO 2 nanoparticles (SiO 2 NP) on the commonly available freshwater fish Oreochromis mossambicus. The 96 h median lethal concentration (LC 50 ) of SiO 2 NP was found to be between 270-280 ppm. In this study, water-soluble emissive carbon dots (CDs) are effectively fabricated with specific optical properties and colors by acid oxidation of muskmelon ( Cucumis melo ) fruit, which are termed as C. melo CDs (CMCDs). The fluorescence properties of CMCDs were tuned by controlling the experimental conditions that allow them to emit different colors, that is, blue (B-), green (G-), and yellow (Y-) CMCDs, with different emission wavelengths at 432, 515, and 554 nm when excited at 342, 415, and 425 nm, respectively. The fabricated multicolor-emissive CDs were confirmed by various analytical techniques. The sizes of B-, G-, and Y-CMCDs were found to be ∼3.5, ∼4.3, and ∼5.8 nm, respectively. The as-prepared CMCDs display stable emissions with quantum yields of 7.07, 26.9, and 14.3% for the three CMCDs, which could act as a promising probe for the selective detection of Hg 2+ ions. Upon the addition of Hg 2+ ions, the fluorescence intensity of G-CMCDs at 515 nm was quenched largely than that of B- and Y-CMCDs. The spectroscopic results display that the G-CMCDs acted as a sensor for the detection of Hg 2+ ions with a wide linear range from 1.0 to 25 μM ( R 2 = 0.9855) with a detection limit of 0.33 μM. This method was successfully applied to detect Hg 2+ ions in biological and water samples. Background: Supraglottic airway devices are used for anesthesia in elective surgical procedures circumventing the need for intubation. We investigated the efficacy and safety of Baska ® mask in comparison to an I-Gel ® device., Methods: In this cross-sectional, observational study, we randomized 100 female patients (age 18-45 years, American Society of Anaesthesiologists grade I or II) undergoing elective short gynecological procedures into two groups, to receive ventilation with either Baska mask ® (group 1, n = 50) or an I-Gel ® device (group 2, n = 50). We excluded patients with obesity, short neck, and known systemic and upper airway disorders. The primary outcome was the oropharyngeal airway seal pressure, and the secondary outcomes were the ease of insertion and the complication rate. The results were analyzed using Mann-Whitney U -test and Fisher's exact test, and correlation analysis was done by Spearman's correlation test., Results: A total of 56 patients underwent dilatation and curettage, whereas the remaining had hysteroscopy in the study. The airway seal pressure achieved was higher with Baska ® mask than I-Gel ® device (35.8 ± 10.3 and 26.9 ± 7.5 of cm H 2 O, respectively; P < 0.0001). The ease of insertion ( P < 0.0001) was better in group 1 and the complication rates were similar in both the groups ( P > 0.05)., Conclusion: Baska ® mask offers a superior airway seal pressure with minimum complications in comparison to an I-Gel ® device. The present study was aimed to analyze the effect of ZnO and TiO 2 nanoparticles (NPs) on Haemato-immunological parameters in adult Tilapia, Oreochromis mossambicus. The nanoparticles size found as 47 nm and 30 nm for ZnO and TiO 2 respectively. The acute toxicity (96 h) of ZnO (LC50: 100-110 ppm) and TiO 2 (LC50: 80-90 ppm) NPs were identified by using probit analysis. RBC, Hb and HCT levels decreased in nanoparticles exposed groups resulted in decreased oxygen carrying capacity of RBC and other erythrocyte indices (MCH, MCV, MCHC). Increased WBC, neutrophils & monocytes and decreased lymphocyte levels were observed as increased concentration of the nanoparticles. The results were found as statistically significant (p < 0.05). In conclusion, the present study depicts that ZnO NPs exhibits more toxicity than TiO 2 NPs. Nanoparticles presence even in low concentration (ppm) cause damage to the connective tissues of fish, so the existing permissible levels of these nanoparticles in water are need to be revised., (Copyright © 2018 Elsevier B.V. Herein, fluorescent gold nanoclusters (Au NCs) were obtained by one-pot synthetic method using bovine serum albumin (BSA) and bromelain as templates. As-synthesized fluorescent Au NCs were stable and showed bright red fluorescence under UV lamp at 365 nm. The fluorescent Au NCs exhibit the emission intensity at 648 nm when excited at 498 nm. Various techniques were used such as spectroscopy (UV-visible, fluorescence, and Fourier-transform infrared), high-resolution transmission electron microscopy, and dynamic light scattering for the characterization of fluorescent Au NCs. The values of I 0 /I at 648 nm are proportional to the concentrations of Hg 2+ ion in the range from 0.00075 to 5.0 μM and of lambda-cyhalothrin in the range from 0.01 to 10 μM with detection limits of 0.0003 and 0.0075 μM for Hg 2+ ion and lambda-cyhalothrin, respectively. The practical application of the probe was successfully demonstrated by analyzing Hg 2+ ion and lambda-cyhalothrin in water samples. In addition, Au NCs used as probes for imaging of Simplicillium fungal cells. These results indicated that the as-synthesized Au NCs have proven to be promising fluorescent material for the sensing of Hg 2+ ion and lambda-cyhalothrin in environmental and for imaging of microorganism cells in biomedical applications.

In the present study, freshwater fish Oreochromis mossambicus were exposed to sub lethal concentrations (120, 150 and 180 ppm) of Aluminium oxide nanoparticles (Al 2 O 3 NPs) for 96 h. Histological abnormalities were not observed in the organs of control fishes whereas severe damages and extensive architectural loss was found in the brain, gill, intestine, kidney and muscle tissues of treated fishes with more pronounced effects in 180 ppm. The yeast bc1 complex (complex III) and cytochrome oxidase (complex IV) are mosaics of core subunits encoded by the mitochondrial genome and additional nuclear-encoded proteins imported from the cytosol. Both complexes build in the mitochondrial inner membrane various supramolecular assemblies. The formation of the individual complexes and their supercomplexes depends on the activity of dedicated assembly factors. We identified a so far uncharacterized mitochondrial protein (open reading frame YDR381C-A ) as an important assembly factor for complex III, complex IV, and their supercomplexes. Therefore, we named this protein Co x i nteracting (Coi) 1. Deletion of COI1 results in decreased respiratory growth, reduced membrane potential, and hampered respiration, as well as slow fermentative growth at low temperature. In addition, coi1Δ cells harbour reduced steady-state levels of subunits of complexes III and IV as well as of the assembled complexes and supercomplexes. Interaction of Coi1 with respiratory chain subunits seems transient, as it appears to be a stoichiometric subunit neither of complex III nor of complex IV. Background: Different studies have implicated the exposure to systemic conditions in the aetiology of cardiovascular diseases like chronic inflammation including chronic periodontitis., Aim: The present study has been conducted to examine whether biomarker sTNF-R was elevated in apical periodontitis as sTNF-R is a systemic marker of inflammation and has been identified as risk factors for cardiovascular diseases., Material and Methods: sTNF-R levels were measured in 52 patients with apical periodontitis (M:F::25:27), aged 20-45 years and in 20 control patients without periodontitis (M:F::10:10, aged 20-48 years). Measurement of sTNF-R1 and sTNF-R2 was carried out in duplicate with standardised, commercially available enzyme immunoassays (R&D Systems Europe, Abingdon, UK)., Results: The mean sTNF-R1 and sTNF-R2 levels in periodontitis were 820 (240) pg/ml (413 - 1620 pg/ml) and 1309 (403) pg/ml (540 - 2430 pg/ml), while in normal sTNF-R1 and sTNF - R2 levels were 740 (340) pg/ml (407-1240 pg/ml) and 1283 (414) pg (480 - 2340 pg/ml) respectively. Results indicated a positive high relationship between cardiovascular markers such as sTNF-R1 and sTNF - R2 and apical periodontitis., Conclusion: Elevated levels of sTNF-R1 and sTNF - R2 in apical periodontitis patients indicate an increased independent risk of coronary heart disease.

In this work, tyrosine-protected gold nanoparticles (Tyr-Au NPs) were fabricated by one-step reduction of Au 3+ ion using Tyr as a reducing and capping agent under microwave irradiation. The Tyr-Au NPs were successfully used as a dual probe for colorimetric and fluorescence turn-on assays of spermine and spermidine in biological samples. Upon addition of spermine and spermidine, the characteristic surface plasmon resonance (SPR) band of Tyr-Au NPs was red-shifted to 596 and 616nm and the emission peak (Tyr) at 410nm was gradually increased with increasing concentration of both analytes, confirming the aggregation of Tyr-Au NPs induced by spermine and spermidine, which results to restore fluorescence of Tyr on the surfaces of Au NPs. In addition, it shows high selectivity for sensitive detection of prostatic cancer biomarkers spermine and spermidine in real clinical applications with reduced sample preparations., (Copyright © 2016 Elsevier B.V. One-step green microwave synthetic approach was developed for the synthesis of copper nanoclusters (Cu NCs) and used as a fluorescent probe for the sensitive detection of thiram and paraquat in water and food samples. Unexpectedly, the prepared Cu NCs exhibited strong orange fluorescence and showed emission peak at 600 nm, respectively. Under optimized conditions, the quenching of Cu NCs emission peak at 600 nm was linearly proportional to thiram and paraquat concentrations in the ranges from 0.5 to 1000 µM, and from 0.2 to 1000 µM, with detection limits of 70 nM and 49 nM, respectively. In addition, bioimaging studies against Bacillus subtilis through confocal fluorescence microscopy indicated that Cu NCs showed strong blue and green fluorescence signals, good permeability and minimum toxicity against the various bacteria species, which demonstrates their potential feasibility for chemical species sensing and bioimaging applications., (Copyright © 2016 Elsevier B.V. This study was framed to investigate the (60)Co gamma radiation induced morphological and histological variations in freshwater prawn Macrobrachium rosenbergii. The LD50 value of (60)Co gamma irradiated M. rosenbergii observed (by probit analysis) at 30 Gy. Prawns were irradiated to four different dose levels (3 mGy, 30 mGy, 300 mGy and 3,000 mGy) using Theratron Phoenix TeleCobalt Unit [P-33] and one control group (without irradiation) maintained separately. Irradiated groups exhibited several morphological variations such as discoloration; damaged rostrum; opaque coloration in cephalothorax; black bands and dot formation in abdomen; deformed uropods and telson in tail regions when compared with control group. The Hepato Somatic Index reflected the severity of radiation on hepatopancreas. Histological variations in gills, hepatopancreas and muscles of irradiated groups were observed. In gills, structural changes such as swollen and fused lamellae, abnormal gill tips, hyperplasic, necrotic and clavate-globate lamellae were observed in gamma irradiated prawns. Accumulation of hemocytes in hemocoelic space, interstitial sinuses filled with abnormal infiltrated hemocytes, the tubular epithelium with ruptured basal laminae, abnormal and coagulated lumen, necrotic tubules, thickened basal laminae, tissue debris, necrotic hepatocytes were observed in irradiated prawn hepatopancreas. In muscle, shrinkage of muscular fiber and necrotic musculature were observed in irradiated prawns. These structural alterations of the organs it is felt could affect the vital physiological functions such as respiration, osmotic and ionic regulation in gills and muscles; absorption, storage and secretion of the hepatopancreas which in turn could adversely affect the growth and survival of freshwater prawn M. rosenbergii., (Copyright © 2013 Elsevier B.V. The thermodynamic parameters viz. the standard free energy (∆Gº), Standard enthalpy change (∆Hº) and standard entropy change (∆Sº) were determined using the obtained values of distribution coefficient (kd) of U (VI) in two different types of soils (agricultural and undisturbed) by conducting a batch equilibrium experiment with aqueous media (groundwater and deionised water) at two different temperatures 25°C and 50°C. The obtained distribution coefficients (kd) values of U for undisturbed soil in groundwater showed about 75% higher than in agricultural soil at 25°C while in deionised water, these values were highly insignificant for both soils indicating that groundwater was observed to be more favorable for high surface sorption. At 50°C, the increased kd values in both soils revealed that solubility of U decreased with increasing temperature. Batch adsorption results indicated that U sorption onto soils was promoted at higher temperature and an endothermic and spontaneous interfacial process. The high positive values of ∆Sº for agricultural soil suggested a decrease in sorption capacity of U in that soil due to increased randomness at solid-solution interface. The low sorption onto agricultural soil may be due to presence of high amount of coarse particles in the form of sand (56%). Geochemical modeling predicted that mixed hydroxo-carbonato complexes of uranium were the most stable and abundant complexes in equilibrium solution during experimental.

Present study was designed to observe the effects of (60)Co gamma radiation in behavioral and histological changes in the gills of giant fresh water prawn Macrobrachium rosenbergii. The adult prawns were irradiated with four different dose levels (3mGy, 30mGy, 300mGy and 3000mGy) and the control group (without irradiation) was maintained separately. Behavioral changes like hyperactivity, loss of balance, reduced swimming rate, slower rate of food intake and convulsions were observed in higher dose levels of 300mGy and 3000mGy. The histological alterations such as accumulated haemocytes in haemocoelic spaces, abnormal gill tips, lifted lamellar epithelium, swollen and fused lamellae, hyperplasic, necrotic, clavate-globate and complete disorganization of lamellae were observed in (60)Co gamma irradiated prawns. Significantly more considerable histological alterations were observed in the highest dose level of 3000mGy, but no mortality was evidenced. The irradiation effect of (60)Co at the three dose level of 3 mGy, 30 mGy and 300 mGy on the histology of liver of the freshwater fish Oreochromis mossambicus was investigated. The liver of O. mossambicus was dissected out and processed for light microscopy studies. (60)Co exposed O. mossambicus were found to result in several alterations in the histoarchitecture of liver. The alterations included mild congestion of blood vessels, structural alteration, cellular swelling, vacuolation and necrotic liver cells, indicating a definite response to (60)Co irradiation. The results suggest that the liver of O. mossambicus exposed to (60)Co were structurally altered with increasing dose levels. It is to record that alteration in the liver does not affect the physiology, behaviour or lethality of the individuals. Self regulating mechanisms would have influenced the liver to remain sustained. During this work, size fractionation technique "ultra filtration" is used in speciation studies of trace elements in the coastal sea water. Filtration is the most commonly used method to fractionate trace metal species, but often only "dissolved" and "particulate" fraction. The purpose of the present study is to determine colloidal and suspended particulate concentrations of Fe, Zn, Cu, Ni, and Mn in sea water. Suspended particulate matter were separated in three different size groups namely (>2.7 microm, <2.7->0.45 microm and <0.45->0.22 microm) by suction filtration using cellulose acetate and nitrate filter membranes. Thereafter to concentrate the solution with colloidal particle <0.22 microm-1.1 nm (0.5 k Nominal Molecular Weight cut-off Limit {NMWL}), the solution obtained from filtration through <0.22 microm, is sequentially passed through the ultra-filtration membranes having pore diameters of 14 nm (300 k NMWL), 3.1 nm (50 k NMWL), 2.2 nm (30 k NMWL), 1.6 nm (10 k NMWL) and 1.1 nm (0.5 k NMWL) by using Stirred Ultra-filtration Cells, operating in concentration mode. The concentration of Fe, Zn, Cu, Ni, and Mn were measured in suspended and dissolved fraction by ion chromatography, ICP-AES and Atomic Absorption Spectrometer. The salinity of the solution in various dissolved fractions of sequential filtration varies between 30.89-34.22 parts per thousand. The maximum concentrations of colloidal Zn, Cu, Ni and Mn in dissolved fraction were in <2.2->1.6 nm fraction. In case of Fe, colloidal fractions <2.2->1.6 nm and <1.6-<1.1 nm shows higher concentration. The concentration of Zn, Cu, Ni and Mn increase with decrease in size in suspended particulate matter, while the reverse is observed in case of Fe. This size separation data that specifies the partitioning of metals between dissolved and suspended solid phases is necessary for developing physically based models of metal transport in aquatic system.

Adenosine deaminase activity (ADA) was estimated in cerebrospinal fluid (CSF) of 30 patients of tuberculous meningitis (TBM) and 10 patients each of partially treated pyomeningitis (PTM), aseptic meningitis (AM) and pyogenic meningitis (PM). Mean ADA levels in CSF of TBM patients were higher (18.22 U/L) as compared to 6.28 U/L, 3.43 U/L and 7.98 U/L in PTM, AM and PM respectively. This difference of ADA values in CSF between TBM and other types of meningitis was statistically significant (p<0.01) different. Sensitivity and specificity of ADA levels in CSF of children to diagnose tuberculous meningitis was 66.6% and 90% respectively at 10 U/L cut off of ADA levels in CSF. ADA levels in CSF could also differentiate PTM, AM and PM from TBM with a specificity of 90%, 100% and 80% respectively.

In the present study, we found that oxidized low density lipoprotein, but not low density lipoprotein, down-regulated base excision repair activity in extracts of mouse monocyte cell line PU5-1.8. An enzyme required in this pathway, DNA polymerase beta, was also down-regulated. In contrast, treatment of monocytes with a combination of ascorbate and alpha-tocopherol up-regulated base excision repair activity and expression of DNA polymerase beta. Co-treatment of monocytes with antioxidants plus oxidized low density lipoprotein prevented down-regulation by oxidized low density lipoprotein. Oxidative DNA damage, as measured by 8-hydroxyguanine accumulation in genomic DNA, was found in cells treated with oxidized low density lipoprotein; 8-hydroxyguanine was not found in the cells treated with low density lipoprotein, antioxidants or oxidized low density lipoprotein plus antioxidants. These results establish a linkage between the DNA base excision repair pathway, oxidative DNA damage and oxidized low density lipoprotein treatment in mouse monocytes. Since oxidized low density lipoprotein is implicated in chronic disease conditions such as atherogenesis, these findings facilitate understanding of genetic toxicology mechanisms related to human health and disease.

DNA polymerase beta is required in mammalian cells for the predominant pathway of base excision repair involving single nucleotide gap filling DNA synthesis. Here we examine the relationship between oxidative stress, cellular levels of DNA polymerase beta and base excision repair capacity in vitro , using mouse monocytes and either wild-type mouse fibroblasts or those deleted of the DNA polymerase beta gene. Treatment with an oxidative stress-inducing agent such as hydrogen peroxide, 3-morpholinosydnonimine, xanthine/xanthine oxidase or lipopolysaccharide was found to increase the level of DNA polymerase beta in both monocytes and fibroblasts. Base excision repair capacity in vitro , as measured in crude cell extracts, was also increased by lipopolysaccharide treatment in both cell types. In monocytes lipopolysaccharide-mediated up-regulation of the base excision repair system correlated with increased resistance to the monofunctional DNA alkylating agent methyl methanesulfonate. By making use of a quantitative PCR assay to detect lesions in genomic DNA we show that lipopolysaccharide treatment of fibroblast cells reduces the incidence of spontaneous DNA lesions. This effect may be due to the enhanced DNA polymerase beta-dependent base excision repair capacity of the cells, because a similar decrease in DNA lesions was not observed in cells deficient in base excision repair by virtue of DNA polymerase beta gene deletion. Similarly, fibroblasts treated with lipopolysaccharide were more resistant to methyl methanesulfonate than untreated cells. This effect was not observed in cells deleted of the DNA polymerase beta gene. These results suggest that the DNA polymerase beta-dependent base excision repair pathway can be up-regulated by oxidative stress-inducing agents in mouse cell lines.

Base excision repair (BER) is a cellular defense mechanism repairing modified bases in DNA. Recently, a G:U repair reaction has been reconstituted with several purified enzymes from Escherichia coli (Dianov, G., and Lindahl, T.(1994) Curr. Biol. 4, 1069-1076). Using bovine testis crude nuclear extract, we have shown that G:U is repaired efficiently in vitro, and DNA polymerase beta (beta-pol) is responsible for the single nucleotide gap-filling synthesis (Singhal, R. K., Prasad, R., and Wilson, S. H.(1995) J. Biol. Chem. 270, 949-957). To investigate potential interaction of beta-pol with other BER protein(s), we developed affinity chromatography matrices by cross-linking purified rat beta-pol or antibody against beta-pol to solid supports. Crude nuclear extract from bovine testis was applied to these affinity columns, which were then extensively washed. Proteins that bound specifically to the affinity columns were co-eluted in a complex with beta-pol. This complex had a molecular mass of approximately 180 kDa and was able to conduct the complete uracil-initiated BER reaction. The BER complex contained both beta-pol and DNA ligase I. An antibody to beta-pol was able to shift the complex in sucrose gradients to a much larger molecular mass (>300 kDa) that again contained both beta-pol and DNA ligase I. Furthermore, DNA ligase I and beta-pol were co-immunoprecipitated from the testis nuclear extract with anti beta-pol IgG. Thus, we conclude that beta-pol and DNA ligase I are components of a multiprotein complex that performs BER.

Synthesis of DNA by DNA polymerase-beta is distributive on single-stranded DNA templates, but short DNA gaps with a 5' PO4 in the gap are filled processively to completion. In vitro studies have suggested a role of beta-polymerase in different types of DNA repair. However, the significance of these studies to the in vivo role of beta-polymerase has remained unclear. Because genetic studies are essential for determining the physiological role of a gene, we established embryonic fibroblast cell lines homozygous for a deletion mutation in the gene encoding DNA polymerase-beta. Extracts from these cell lines were found to be defective in uracil-initiated base-excision repair. The beta-polymerase-deleted cells are normal in viability and growth characteristics, although they exhibit increased sensitivity to monofunctional DNA-alkylating agents, but not to other DNA-damaging agents. Both the deficiency in base-excision repair and hypersensitivity to DNA-alkylating agents are rescued following stable transfection with a wild-type beta-polymerase minitransgene. These studies demonstrate that beta-polymerase functions specifically in base-excision repair in vivo.

DNA polymerase beta (beta-polymerase) has been implicated in short-patch DNA synthesis in the DNA repair pathway known as base excision repair. The native 39 kDa enzyme is organized into four structurally and functionally distinct domains. In an effort to examine this enzyme as a potential therapeutic target, we analyzed the effect of various beta-polymerase domains on the activity of the enzyme in vitro. We show that the 14 kDa N-terminal segment of beta-polymerase, which binds to both single- and double-stranded DNA, but lacks DNA polymerase activity, inhibits beta-polymerase activity in vitro. Most importantly, the 8, 27 and 31 kDa domains of beta-polymerase do not inhibit beta-polymerase activity, demonstrating that the inhibition by the 14 kDa domain is specific. The inhibition of beta-polymerase activity in vitro is abolished by increasing the concentrations of both of the substrates (template-primer and deoxynucleoside triphosphate). In contrast, an in vitro base excision repair assay is inhibited in a domain specific manner by the 14 kDa domain even in the presence of saturating substrates. The inhibition of beta-polymerase activity by the 14 kDa domain appears specific to beta-polymerase as this domain does not inhibit either mammalian DNA polymerase alpha or Escherichia coli polymerase I (Klenow fragment). These data suggest that the 14 kDa domain could be used as a potential inhibitor of intracellular beta-polymerase and that it may provide a means for sensitizing cells to therapeutically relevant DNA damaging agents.

The G:U mismatch in genomic DNA mainly arises from deamination of cytosine residues and is repaired by the base excision repair pathway. We found that a bovine testis crude nuclear extract conducts uracil-initiated base excision repair in vitro. A 51-base pair synthetic DNA substrate containing a single G:U mismatch was used, and incorporation of dCMP during repair was exclusively to replace uracil. A neutralizing polyclonal antibody against DNA polymerase beta (beta-pol) inhibited the repair reaction. ddCTP also inhibited the repair reaction, whereas aphidicolin had no significant effect, suggesting that activity of beta-pol was required. Next, the base excision repair system was reconstituted using partially purified components. Several of the enzymatic activities required were resolved, such that DNA ligase and the uracil-DNA glycosylase/apurinic/apyrimidinic endonuclease activities were separated from the DNA polymerase requirement. We found that purified beta-pol could restore full DNA repair activity to the DNA polymerase-depleted fraction, whereas purified DNA polymerases alpha, delta, and epsilon could not. These results with purified proteins corroborated results obtained with the crude extract and indicate that beta-pol is responsible for the single-nucleotide gap filling reaction involved in this in vitro base excision repair system.

We have shown by activity gel that overexpression in E. coli of a yeast chromosome 3 open reading frame (ORF) designated YCR14C and bearing homology to mammalian DNA polymerases beta results in a new DNA polymerase in the host cells. The molecular mass of this enzyme corresponded to the YCR14C-predicted 67 kDa protein, and NH2-terminal amino acid sequencing confirmed that the expressed protein was encoded by the yeast ORF. This new yeast DNA polymerase was purified to homogeneity from E.coli. In a fashion similar to that of mammalian beta-polymerases, the purified yeast enzyme exhibited distributive DNA synthesis on DNA substrate with a single-stranded template and processive gap-filling synthesis on a short-gapped DNA substrate. Activity of this yeast beta-polymerase-like enzyme was sensitive to the beta-polymerase inhibitor ddNTP and resistant to both 1 mM NEM and neutralizing antibody to E. coli DNA polymerase I. These results, therefore, indicate that YCR14C encodes a DNA beta-polymerase-like enzyme in yeast, and we name it DNA polymerase IV. Yeast strains harboring a deletion mutation of the pol IV gene are viable, they exhibit no increase in sensitivity to ultraviolet light, ionizing radiation or alkylating agents, and sporulation and spore viability are not affected in the mutant.

Mammalian DNA polymerase beta (beta-pol) is considered to be part of the DNA repair machinery. However, there have been conceptual problems with this idea because beta-pol does not completely fill some gapped substrates, and unlike other DNA polymerases, beta-pol adds just one dNMP for each cycle of binding to the DNA substrate, incorporation, and product release (distributive synthesis). To examine the questions of complete gap filling and gap recognition by beta-pol, we designed template-primer substrates with a range of gap sizes from 1 to 53 template residues between the 3'-OH primer and a downstream polynucleotide. M13mp18(+) single-stranded DNA was used as template, and synthetic deoxyoligonucleotides were used as primers and downstream polynucleotides. We find that beta-pol can completely fill gaps and that the gap-filling activity on substrates with gaps of up to 6 nucleotides is highly processive rather than distributive; processive synthesis to fill a gap strictly required 5'-phosphate on the 5'-moiety of the gap. Similar specificity for filling short gaps was not observed with other DNA polymerases. These results on substrate specificity and the mechanism of beta-pol suggest distinct requirements for the role of beta-pol in vivo.
