Background: The regenerative and adaptive capacity of skeletal muscles reduces with age, leading to severe disability and frailty in the elderly. Therefore, development of effective therapeutic interventions for muscle wasting is important both medically and socioeconomically. In the present study, we aimed to elucidate the potential contribution of fibro-adipogenic progenitors (FAPs), which are mesenchymal stem cells in skeletal muscles, to immobilization-induced muscle atrophy. Methods: Young (2–3 months), adult (12–14 months), and aged (20–22 months) mice were used for analysis. Muscle atrophy was induced by immobilizing the hind limbs with a steel wire. FAPs were isolated from the hind limbs on days 0, 3, and 14 after immobilization for transcriptome analysis. The expression of ST2 and IL-33 in FAPs was evaluated by flow cytometry and immunostaining, respectively. To examine the role of IL-33-ST2 signaling in vivo, we intraperitoneally administered recombinant IL-33 or soluble ST2 (sST2) twice a week throughout the 2-week immobilization period. After 2-week immobilization, the tibialis anterior muscles were harvested and the cross-sectional area of muscle fibers was evaluated. Results: The number of FAPs increased with the progression of muscle atrophy after immobilization in all age-groups. Transcriptome analysis of FAPs collected before and after immobilization revealed that Il33 and Il1rl1 transcripts, which encode the IL-33 receptor ST2, were transiently induced in young mice and, to a lesser extent, in aged mice. The number of FAPs positive for ST2 increased after immobilization in young mice. The number of ST2-positive FAPs also increased after immobilization in aged mice, but the difference from the baseline was not statistically significant. Immunostaining for IL-33 in the muscle sections revealed a significant increase in the number of FAPs expressing IL-33 after immobilization. Administration of recombinant IL-33 suppressed immobilization-induced muscle atrophy in aged mice but not in young mice. Conclusions: Our data reveal a previously unknown protective role of IL-33-ST2 signaling against immobilization-induced muscle atrophy in FAPs and suggest that IL-33-ST2 signaling is a potential new therapeutic target for alleviating disuse muscle atrophy, particularly in older adults. Dependence of the radiative-convective equilibrium structure of the lower atmosphere of Venus on the specification of an atmospheric thermodynamic model is investigated. A series of thermodynamic models including ideal gases, van der Waals gases, and real gases are introduced by the use of the Helmholtz energy given by the EOS-CG mixture model (EOS-CG: Equation of State for Combustion Gases and Combustion Gas-like Mixtures). It is demonstrated that the radiative-convective equilibrium profile for the real gas differs significantly from that for the ideal gas with temperature-dependent specific heat by an increase of about 7 K in the surface temperature. This difference is caused by the fact that the adiabatic lapse rate evaluated with the thermodynamic model of real gas is larger than that of ideal gas, since the non-ideality of gas increases the thermal expansion coefficient, which overwhelms the increases in density and specific heat. It is confirmed that, in order to obtain better calculations of atmospheric circulations including the lower atmosphere of Venus, the ideal gas with a constant specific heat should be abandoned. The ideal gas with a temperature-dependent specific heat may not be enough. A set of line-by-line and correlated k-distribution radiation models are developed aiming for applications in simulations and examinations of Venus and Mars-Iike planetary atmospheres. Our line-by-line model is validated by comparing the results with observations and those of previous studies under conditions of Venus, and present and possible early Mars. The radiation fields calculated by our line-by-line model agree well with observed profiles and are within the acceptable range from those presented in previous studies. The results obtained by our line-by-line model are then processed to generate a series of parameters for our correlated k-distribution model. It is confirmed that the radiation fields calculated with those sets of parameters by our correlated k-distribution model sufficiently agree with those by our line-by-line model for the atmospheres with a wide range of surface pressure. By the use of our correlated k-distribution model implemented with those sets of parameters, we evaluate the radiation field for Venus and calculate radiative-convective equilibrium profiles for Venus and Mars. The obtained vertical thermal structures for Venus are qualitatively consistent with observations, and the behaviors of surface pressure and surface temperature for Mars are similar to those reported by previous studies. Those results demonstrate that our models including the procedure for generating tables of radiation parameters are applicable to examine climates of CO2 dominant atmospheres in our solar and exoplanetary systems. Recent reports revealed that intravenous (iv) busulfan (BU) may not only reduce early nonrelapse mortality (NRM) but also improve overall survival (OS) probability in adults. Therefore, we retrospectively compared outcomes for 460 children with acute leukemia who underwent hematopoietic stem cell transplantation with either iv-BU (n = 198) or oral busulfan (oral-BU) (n = 262) myeloablative conditioning. OS at 3 years was 53.4% ± 3.7% with iv-BU and 55.1% ± 3.1% with oral-BU; the difference was not statistically significant (P = .77). OS at 3 years in 241 acute lymphoblastic leukemia and 219 acute myeloid leukemia patients was 56.4% ± 5.5% with iv-BU and 54.6% ± 4.1 with oral-BU (P = .51) and 51.0% ± 5.0% with iv-BU and 55.8% ± 4.8% with oral-BU (P = .83), respectively. Cumulative incidence of relapse at 3 years with iv-BU was similar to that with oral-BU (39.0% ± 3.6% and 36.4% ± 3.1%, respectively; P = .67). Cumulative incidence of NRM at 3 years was 16.6% ± 2.7% with iv-BU and 18.3% ± 2.5% with oral-BU (P = .51). Furthermore, multivariate analysis showed no significant survival advantage with iv-BU. Allogeneic stem cell transplantation (SCT) is the only curative therapy for patients with refractory or relapsed acute leukemia, although the prognosis remains poor. Few reports have described outcomes of SCT in pediatric patients with refractory acute leukemia. To identify prognostic factors for these patients, we retrospectively evaluated SCT outcomes for advanced acute leukemia in 82 pediatric patients from 3 transplant units in Nagoya City between 1990 and 2008. Median age at transplantation was 8 years (range, 0.5-17 years). Transplantation was performed in the first refractory relapse for 53 patients (64.6%), in the second or subsequent relapse for 16 patients (19.5%), and during primary induction failure for 13 patients (15.9%). Only 4 patients (4.9%) underwent transplantation in the untreated first relapse, and 39 patients (47.6%) received unrelated donor progenitor cells. Of the 82 patients, 61 died (77.9%), with a median survival of 7.1 months (95% confidence interval [CI], 4.2-10.0 months). Median disease-free survival (DFS) was 4.7 months (95% CI, 2.6-6.9 months). In multivariate analysis, peripheral blood blasts, cord blood transplantation, and more than 3 courses of previous salvage chemotherapy were predictive of DFS. These results support the notion that allogeneic SCT offers only a small chance of cure for most pediatric patients with refractory or relapsed acute leukemia, and suggest that reduction of the leukemia burden and earlier optimal timing of transplantation are essential for long-term survival even in patients with refractory acute leukemia. Aplastic anemia (AA) is characterized by a reduced number of hematopoietic stem cells and fatty replacement in the bone marrow. Transcriptional factor GATA-2 plays several important roles in both hematopoiesis and adipogenesis. Decreased levels of GATA-2 compromise the proliferation and survival of hematopoietic stem cells. GATA-2 suppresses adipocyte differentiation through direct inhibition of adipogenic factors, including peroxisome proliferator-activated receptor−γ (PPARγ). Previous studies have shown that expression of GATA-2 is decreased in marrow CD34-positive cells in AA. To elucidate the mechanisms of fatty marrow replacement, we evaluated the mRNA expression for GATA-2 and PPARγ in mesenchymal stem cells (MSCs) from patients with AA by quantitative real-time polymerase chain reaction. GATA-2 expression by MSCs from AA patients was significantly lower than in normal subjects. Conversely, expression of PPARγ was significantly higher in AA patients. Western blot analysis demonstrated that protein levels of GATA-2 were lower in AA patients than those in normal subjects. Moreover, incubation with interferon-γ induced downregulation of GATA-2 levels in MSCs from normal subjects. These findings indicate that fatty marrow replacement in AA patients can be explained by downregulation of GATA-2 and overexpression of PPARγ in MSCs. JEM-EUSO is a space science mission to explore the extreme energies and physics of the Universe. Its instrument will watch the darkside of the earth and will detect UV photons emitted from the extensive air shower caused by an ultra-high energy cosmic ray (UHECR above 1018 eV), or an extremely high energy cosmic ray (EHECR) particle (e.g. above about 1020 eV). Such a high-rigidity particle as the latter arrives almost in a straight line from its origin through the magnetic fields of our Milky Way Galaxy and is expected to allow us to trace the source location by its arrival direction. This can open the door to new astronomy with charged particles. In its 5 years of operation including the tilted mode, Extreme Universe Space Observatory an Japanese Experiment Module (JEM-EUSO) will detect at least 1000 events with E > 7 × 1019 eV with the Greisen--Zatsepin--Ku'zmin (GZK) suppression spectrum. It can determine the energy spectrum and source locations of GZK to super-GZK regions with a statistical accuracy of several percent. JEM-EUSO is planned to be deployed by H2 transfer vehicle (HTV) and will be attached to the Japanese Experiment Module/Exposure Facility (JEM/EF) of International Space Station. Transplanted donor lymphocytes infused during hematopoietic stem cell transplantation (HSCT) have been shown to cure patients with hematological malignancies. However, less is known about the effects of HSCT on metastatic solid tumors. Thus, a better understanding of the immune cells and their target antigens that mediate tumor regression is urgently needed to develop more effective HSCT approaches for solid tumors. Here we report regression of metastatic renal cell carcinoma (RCC) in patients following nonmyeloablative HSCT consistent with a graft-versus-tumor effect. We detected RCC-reactive donor-derived CD8(+) T cells in the blood of patients following nonmyeloablative HSCT. Using cDNA expression cloning, we identified a 10-mer peptide (CT-RCC-1) as a target antigen of RCC-specific CD8(+) T cells. The genes encoding this antigen were found to be derived from human endogenous retrovirus (HERV) type E and were expressed in RCC cell lines and fresh RCC tissue but not in normal kidney or other tissues. We believe this to be the first solid tumor antigen identified using allogeneic T cells from a patient undergoing HSCT. These data suggest that HERV-E is activated in RCC and that it encodes an overexpressed immunogenic antigen, therefore providing a potential target for cellular immunity. [ABSTRACT FROM AUTHOR]

We measured the temperature and frequency dispersion of the permittivity of vinylidene fluoride and trifluoroethylene (YDF/TrFE) thin films spin-coated on glass substrates with evaporated aluminium electrodes. Assuming the existence of two surface layers with different dielectric properties from the bulk material, a thickness of 2 nm was deduced for each surface layer. The temperature dependence of the frequency dispersion of the permittivity of the surface layers was obtained. Each surface layer has a dielectric relaxation which is essentially the same as the segment mode of the amorphous region of the bulk. It shows no anomaly at the temperature of the paraelectric-ferroelectric phase transition for thick samples. The surface layers appear to be amorphous. [ABSTRACT FROM AUTHOR]

Objective: Individuals with Dravet syndrome (DS) exhibit progressive gait disturbance. No quantitative studies have been conducted to evaluate the effectiveness of medication for gait disturbance. Therefore, the aim of this study was to evaluate the effectiveness of levodopa for pathological gait in people with DS using three‐dimensional gait analysis (3DGA). Methods: Nine individuals with DS, ages 6–20 years, participated in a crossover study of levodopa and were randomly assigned to the levodopa precedence or no levodopa precedence group. Levodopa/carbidopa hydrate was prescribed at a dose of 5 mg/kg/day (body weight <60 kg) or 300 mg/day (body weight ≥60 kg). The medication was taken for 4–6 weeks (4‐week washout period). 3DGA was performed three times before the study, with and without levodopa. A mixed‐effects model was used to evaluate the effectiveness of levodopa. The primary outcome was the change in the Gait Deviation Index (GDI). In addition, spatiotemporal gait parameters, 6‐minute walking distance (6MD), and balance were evaluated. The correlation between the effectiveness of levodopa and age or gait performance before starting levodopa was analyzed. Results: Levodopa improved the GDI by 4.2 points, (p =.029), 6MD by 52 m (p =.002), and balance test result by 4.1 mm (p =.011) in participants with DS. No severe adverse events were observed, with the exception of one participant, who exhibited fever and consequently stopped taking levodopa. Levodopa was more effective in younger participants with a higher baseline gait performance. Simple Summary: Tumor differentiation therapy using retinoids has a long history of clinical development and an established role as a part of the standard treatment for high-risk neuroblastoma. However, basic and clinical science related to this treatment is still immature in multiple respects. In the present review, we systematically expand on the basic and clinical science of retinoids for neuroblastoma treatment, off-label issues related to the treatment and possible countermeasures, and finally, prospects for this therapy. Retinoids are vitamin A derivatives and include trans-retinoic acid, isotretinoin, tamibarotene, and bexarotene, all of which are currently available for clinical use. The clinical development of retinoid therapy for neuroblastoma has a history spanning more than four decades. The most promising agent is isotretinoin, which can contribute to improving event-free survival in patients with high-risk neuroblastoma by approximately 10% when administered over six months as maintenance therapy. Although isotretinoin is regarded as an essential component in the standard clinical management of high-risk neuroblastoma, its use for this purpose in the US and EU is off-label. To promote isotretinoin use in Japan as a treatment for neuroblastoma, our clinical research team is planning to launch an investigator-initiated, registration-directed clinical trial. The present review article discusses the basic science behind retinoid therapy, pre-clinical/clinical evidence on neuroblastoma, the concept of the proposed clinical trial, and prospects for this therapy. Multilineage-differentiating stress-enduring (Muse) cells are endogenous reparative pluripotent stem cells present in the bone marrow, peripheral blood, and organ connective tissues. We assessed the homing and therapeutic effects of systemically administered nafimestrocel, a clinical-grade human Muse cell-based product, without immunosuppressants in a neonatal hypoxic–ischemic (HI) rat model. HI injury was induced on postnatal day 7 (P7) and was confirmed by T2-weighted magnetic resonance imaging on P10. HI rats received a single dose nafimestrocel (1 × 106 cells/body) or Hank's balanced salt solution (vehicle group) intravenously at either three days (on P10; M3 group) or seven days (on P14; M7 group) after HI insult. Radioisotope experiment demonstrated the homing of chromium-51-labeled nafimestrocel to the both cerebral hemispheres. The cylinder test (M3 and M7 groups) and open-field test (M7 group) showed significant amelioration of paralysis and hyperactivity at five weeks of age compared with those in the vehicle group. Nafimestrocel did not cause adverse events such as death or pathological changes in the lung at ten weeks in the both groups. Nafimestrocel attenuated the production of tumor necrosis factor-α and inducible nitric oxide synthase from activated cultured microglia in vitro. We report a case of bone metastasis arising from lung cancer, including quantitative values obtained with bone single-photon emission computed tomography/computed tomography (SPECT/CT), which were useful to evaluate the treatment response. The first bone SPECT/CT during pembrolizumab therapy for lung cancer recurrence showed intense 99mTc-HMDP uptake of the right femur head and mild uptake of the left ribs. After the palliative radiotherapy for the right femur head metastasis and chemotherapy, the second bone SPECT/CT showed a decrease in focal uptake of the right femur hip and increasing uptake of the left ribs. There was also new uptake appearance in the sternum, right rib, spine (Th2, Th9, Th12, L4, S1), and bilateral pelvic bone (left ilium, acetabular cartridge, femur, right ilium and ischium). The change of maximum standardized uptake values (SUVmax) for the right femur head and left third and eighth rib bony metastases were −72.6% (from 22.96 to 6.28), +407.7% (from 2.97 to 15.08), and +229.2% (from 2.60 to 8.56), respectively. The change of whole-lesion metabolic bone volume and total bone uptake was +235.4% (from 22.75 to 76.3 cm3) and +219.1% (from 205.0 to 654.09), respectively. Two quantitative bone SPECT/CT images clearly showed the good response of femur head metastasis due to radiotherapy, and progression of other bone metastases regardless of chemotherapy. The existence of liquid water within an oxidized environment on early Mars has been inferred by the Mn-rich rocks found during recent explorations on Mars. The oxidized atmosphere implied by the Mn-rich rocks would basically be comprised of CO2 and H2O without any reduced greenhouse gases such as H2 and CH4. So far, however, it has been thought that early Mars could not have been warm enough to sustain water in liquid form without the presence of reduced greenhouse gases. Here, we propose that H2O2 could have been the gas responsible for warming the surface of the oxidized early Mars. Our one-dimensional atmospheric model shows that only 1 ppm of H2O2 is enough to warm the planetary surface because of its strong absorption at far-infrared wavelengths, in which the surface temperature could have reached over 273 K for a CO2 atmosphere with a pressure of 3 bar. A wet and oxidized atmosphere is expected to maintain sufficient quantities of H2O2 gas in its upper atmosphere due to its rapid photochemical production in slow condensation conditions. Our results demonstrate that a warm and wet environment could have been maintained on an oxidized early Mars, thereby suggesting that there may be connections between its ancient atmospheric redox state and possible aqueous environment. A letter to the editor is presented in response to an article on a study on rabbit antithymocyte globulin and cyclosporine as first-line therapy for children with acquired aplastic anemia.

Methane (CH4) is known to be emitted from lakes to the atmosphere via processes such as diffusion and ebullition (i.e., bubble emission). We developed a practical method for partitioning eddy-covariance CH4 fluxes from a shallow lake into diffusive and ebullitive fluxes using a wavelet analysis based on local scalar similarity between the CH4 concentration and other reference scalars, such as the air temperature or water vapour concentration, in the wavelet time-scale domain, with the hypothesis that similar and dissimilar fluctuation components are related to diffusive and ebullitive CH4 fluxes, respectively. Our method is applied to approximately two weeks of data obtained at a shallow mid-latitude lake. The partitioned diffusive flux has a physically sound relationship with wind speed, supporting the validity of the method. The ratio of the diffusive flux to the total flux is typically 0.11 with flow from an area of steady bubble emission, but otherwise 0.36. Further validation is required using a larger dataset and data from other lakes. The proposed method can be easily applied to historical data because it requires only 10-Hz data of CH4 concentration and other reference scalars, along with an empirical parameter. Neonatal necrotizing enterocolitis (NEC) is a serious disease of premature infants that necessitates intensive care and frequently results in life-threatening complications and high mortality. Dedifferentiated fat cells (DFATs) are mesenchymal stem cell-like cells derived from mature adipocytes. DFATs were intraperitoneally administrated to a rat NEC model, and the treatment effect and its mechanism were evaluated. The NEC model was created using rat pups hand fed with artificial milk, exposed to asphyxia and cold stress, and given oral lipopolysaccharides after cesarean section. The pups were sacrificed 96 h after birth for macroscopic histological examination and proteomics analysis. DFATs administration significantly improved the survival rate from 25.0 (vehicle group) to 60.6% (DFAT group) and revealed a significant reduction in macroscopical, histological, and apoptosis evaluation compared with the vehicle group. Additionally, the expression of C–C motif ligand 2 was significantly decreased, and that of interleukin-6 decreased in the DFAT group. DFAT administration ameliorated 93 proteins mainly related to proteins of fatty acid metabolism of the 436 proteins up-/down-regulated by NEC. DFATs improved mortality and restored damaged intestinal tissues in NEC, possibly by improving the abnormal expression of fatty acid-related proteins and reducing inflammation. Fetal growth restriction (FGR), followed by postnatal early catch-up growth, is associated with an increased risk of metabolic dysfunction, including type 2 diabetes in humans. This study aims to determine the effects of FGR and early catch-up growth after birth on the pathogenesis of type 2 diabetes, with particular attention to glucose tolerance, pancreatic islet morphology, and fibrosis, and to elucidate its mechanism using proteomics analysis. The FGR rat model was made by inducing mild intrauterine hypoperfusion using ameroid constrictors (ACs). On day 17 of pregnancy, ACs were affixed to the uterine and ovarian arteries bilaterally, causing a 20.9% reduction in birth weight compared to sham pups. On postnatal day 4 (P4), the pups were assigned to either the good nutrition (GN) groups with 5 pups per dam to ensure postnatal catch-up growth or poor nutrition groups with 15 pups per dam to maintain lower body weight. After weaning, all pups were fed regular chow food ad libitum (P21). Rats in both FGR groups developed glucose intolerance; however, male rats in the FGR good nutrition (FGR-GN) group also developed hypertriglyceridemia and dysmorphic pancreatic islets with fibrosis. A comprehensive and functional analysis of proteins expressed in the pancreas showed that FGR, followed by early catch-up growth, severely aggravated cell adhesion-related protein expression in male offspring. Thus, FGR and early catch-up growth caused pancreatic islet morphological abnormalities and fibrosis associated with the disturbance of cell adhesion-related protein expressions. Background Mitigation measures implemented during the coronavirus disease 2019 (COVID-19) pandemic remarkably reduced the incidence of infectious diseases among children. However, a re-emergence of respiratory syncytial virus (RSV) infection was observed in 2021 in Japan. We compared the clinical characteristics of hospitalized patients with RSV infection before and during COVID-19. Methods We retrospectively enrolled children aged <6 years who were hospitalized with RSV infection in 18 hospitals and compared their clinical characteristics before (January 2019 to April 2020, 1675 patients) and during COVID-19 (September 2020 to December 2021, 1297 patients). Results The mean age of patients with RSV infection was significantly higher during COVID-19 than before (17.4 vs 13.7 months, P <.001). Compared with before COVID-19, a 2.6-fold increase in RSV cases in the 2–5 years age group was observed from sentinel surveillance during COVID-19, whereas a 1.2-fold increase was noted in the same age group among hospitalized patients. On average for all patients, consolidation shadows obtained on radiography were less frequently observed (26.1 vs 29.6%, P =.04), and reduced respiratory assistance (42.2% vs 48.7%, P <.001) and hospitalization stay (5.7 vs 6.0 days, P <.001) was required in patients with RSV infection during COVID-19. Conclusions Coronavirus disease 2019 and social activity restriction caused epidemiological changes in pediatric RSV infections, and a majority of patients with RSV infection aged ≥2 years did not develop severe symptoms requiring hospitalization. Juvenile myelomonocytic leukemia (JMML) is a rare heterogeneous hematological malignancy of early childhood characterized by causative RAS pathway mutations. Classifying patients with JMML using global DNA methylation profiles is useful for risk stratification. We implemented machine learning algorithms (decision tree, support vector machine, and naïve Bayes) to produce a DNA methylation-based classification according to recent international consensus definitions using a well-characterized pooled cohort of patients with JMML (n = 128). DNA methylation was originally categorized into three subgroups: high methylation (HM), intermediate methylation (IM), and low methylation (LM), which is a trichotomized classification. We also dichotomized the subgroups as HM/IM and LM. The decision tree model showed high concordances with 450k-based methylation [82.3% (106/128) for the dichotomized and 83.6% (107/128) for the trichotomized subgroups, respectively]. With an independent cohort (n = 72), we confirmed that these models using both the dichotomized and trichotomized classifications were highly predictive of survival. Our study demonstrates that machine learning algorithms can generate clinical parameter-based models that predict the survival outcomes of patients with JMML and high accuracy. Background: Patients with primary refractory and relapsed neuroblastoma have a poor prognosis since safe and effective chemotherapies for these patients are currently limited. The development of new chemotherapy regimens for these patients is imperative to improve survival outcomes. Methods: We retrospectively analyzed 40 patients with refractory (n = 36) or relapsed (n = 4) neuroblastoma who received irinotecan, etoposide, and carboplatin (IREC) as a second‐line treatment. We evaluated their therapeutic response and the toxicity of IREC. We also assessed the impact of UGT1A1 gene polymorphisms, which are involved in irinotecan metabolism, on outcomes and toxicity. Results: A total of 112 cycles of IREC were administered to 40 patients with a median of 2 cycles per patient (range, 1–9). Six (15%) patients (UGT1A1 wild‐type [n = 2] and heterozygous [n = 4]) showed objective responses, including partial response (n = 1), tumor shrinkage (n = 4), and improved findings on their MIBG scan (n = 1). Grade 4 neutropenia, grade 4 leukopenia, and grades 3–4 gastrointestinal toxicity were observed in 110 (98%), 88 (79%), and 3 (3%) cycles, respectively. There was no IREC‐related mortality. Patients with UGT1A1 polymorphisms showed a higher frequency of grade 4 leukopenia, but these patients did not have increased treatment‐related mortality or non‐hematologic toxicity. Conclusions: IREC showed an objective response rate of 15% including 1 case with partial response. IREC was well tolerated regardless of UGT1A1 genotype. Abstract: Terpenoids emitted from forests contribute to the formation of secondary organic aerosols and affect the carbon budgets of forest ecosystems. To investigate seasonal variation in terpenoid flux involved in the aerosol formation and carbon budget, we measured the terpenoid flux of a Larix kaempferi forest between May 2011 and May 2012 by using a relaxed eddy accumulation method. Isoprene was emitted from a fern plant species Dryopteris crassirhizoma on the forest floor and monoterpenes from the L. kaempferi. α-Pinene was the dominant compound, but seasonal variation of the monoterpene composition was observed. High isoprene and monoterpene fluxes were observed in July and August. The total monoterpene flux was dependent on temperature, but several unusual high positive fluxes were observed after rain fall events. We found a good correlation between total monoterpene flux and volumetric soil water content (r = 0.88), and used this correlation to estimate monoterpene flux after rain events and calculate annual terpenoid emissions. Annual carbon emission in the form of total monoterpenes plus isoprene was determined to be 0.93% of the net ecosystem exchange. If we do not consider the effect of rain fall, carbon emissions may be underestimated by about 50%. Our results suggest that moisture conditions in the forest soil is a key factor controlling the monoterpene emissions from the forest ecosystem. [Copyright &y& Elsevier]

Background Febrile neutropenia (FN) is a frequent complication in immunocompromised patients. However, causative microorganisms are detected in only 10% of patients. This study aimed to detect the microorganisms that cause FN using next-generation sequencing (NGS) to identify the genome derived from pathogenic microorganisms in the bloodstream. Here, we implemented a metagenomic approach to comprehensively analyze microorganisms present in clinical samples from patients with FN. Methods FN is defined as a neutrophil count <500 cells/µL and fever ≥37.5°C. Plasma/serum samples of 112 pediatric patients with FN and 10 patients with neutropenia without fever (NE) were sequenced by NGS and analyzed by a metagenomic pipeline, PATHDET. Results The putative pathogens were detected by NGS in 5 of 10 FN patients with positive blood culture results, 15 of 87 FN patients (17%) with negative blood culture results, and 3 of 8 NE patients. Several bacteria that were common in the oral, skin, and gut flora were commonly detected in blood samples, suggesting translocation of the human microbiota to the bloodstream in the setting of neutropenia. The cluster analysis of the microbiota in blood samples using NGS demonstrated that the representative bacteria of each cluster were mostly consistent with the pathogens in each patient. Conclusions NGS technique has great potential for detecting causative pathogens in patients with FN. Cluster analysis, which extracts characteristic microorganisms from a complex microbial population, may be effective to detect pathogens in minute quantities of microbiota, such as those from the bloodstream. A series of ferroelectric switching measurements are performed on the 75/25mol% vinylidene fluoride /trifluoroethylene copolymer thin films with various structures to elucidate the factors governing the switching characteristics of such films. It is shown that the spin-coated and well-annealed samples exhibit a remanent polarization Pr of 85 mC/m². Unlike that in previous reports, the switching time τs of these samples is independent of thickness down to 50 nm if Au electrodes are used instead of Al electrodes. Thin films with different metal electrodes exhibit anisotropic τs due to a potential difference between metal electrodes produced by their work function difference. A train of unipolar short on-off electric field pulses induce a slow but full polarization reversal whose time evolution supports the nucleation and growth mechanism. Continuous switching under bipolar on-off electric field pulses reveals an accelerated polarization reversal with decreasing pulse width. This means that τs is a function of not only applied electric field but also the preceding poling conditions. Fatigue is significantly improved using Au electrodes instead of Al electrodes. Perinatal hypoxic ischemic encephalopathy (HIE) results in serious neurological dysfunction and mortality. Clinical trials of multilineage-differentiating stress enduring cells (Muse cells) have commenced in stroke using intravenous delivery of donor-derived Muse cells. Here, we investigated the therapeutic effects of human Muse cells in an HIE model. Seven-day-old rats underwent ligation of the left carotid artery then were exposed to 8% oxygen for 60 min, and 72 hours later intravenously transplanted with 1 × 104 of human-Muse and -non-Muse cells, collected from bone marrow-mesenchymal stem cells as stage-specific embryonic antigen-3 (SSEA-3)+ and −, respectively, or saline (vehicle) without immunosuppression. Human-specific probe revealed Muse cells distributed mainly to the injured brain at 2 and 4 weeks, and expressed neuronal and glial markers until 6 months. In contrast, non-Muse cells lodged in the lung at 2 weeks, but undetectable by 4 weeks. Magnetic resonance spectroscopy and positron emission tomography demonstrated that Muse cells dampened excitotoxic brain glutamatergic metabolites and suppressed microglial activation. Muse cell-treated group exhibited significant improvements in motor and cognitive functions at 4 weeks and 5 months. We report here a case of mandibular osteomyelitis in a 63-year-old female in which quantitative values determined using bone SPECT/CT were useful to evaluate response to antibiotic therapy, hyperbaric oxygen therapy, and sequestomy. After finishing therapy, the chief complaints were well relieved, and posttreatment Tc-99m HMDP bone SPECT/CT examination showed decreased uptake. The maximum standardized uptake value (SUV), peak SUV, mean SUV, metabolic bone volume, and total bone uptake of the untreated lesion were 6.26, 5.16, 3.97, and 11.86 mL and 42.21, respectively, which were decreased to 4.65, 3.90, 2.77, and 9.67 mL and 26.80, respectively, following hyperbaric oxygen therapy and antibiotic administration, and were moreover decreased to 4.28, 3.67, 2.75, and 6.24 mL and 17.19, respectively, after sequestomy. In comparison with pretreatment situation, those parameters were decreased by −25.7, −24.4, −30.2, −18.5, and −36.5%, respectively, following hyperbaric oxygen therapy and antibiotic administration, and moreover by −31.6, −28.9, −30.7, −47.4, and −59.3, respectively, after sequestomy, likely reflecting treatment response. Background: Immunosuppression during liver transplantation (LT) enables the prevention and treatment of organ rejection but poses a risk for severe infectious diseases. Immune modulation and antimicrobials affect the plasma microbiome. Thus, determining the impact of immunosuppression on the microbiome may be important to understand immunocompetence, elucidate the source of infection, and predict the risk of infection in LT recipients. We characterized the plasma microbiome of LT recipients at early post-LT and assessed the association between the microbiome and clinical events. Results: In this study, 51 patients who received LT at Nagoya University Hospital from 2016 to 2018 were enrolled. Plasma samples were retrospectively collected at the following time points: 1) within a week after LT; 2) 4 ± 1 weeks after LT; 3) 8 ± 1 weeks after LT; and 4) within 2 days after a positive blood culture. A total of 111 plasma samples were analyzed using shotgun next-generation sequencing (NGS) with the PATHDET pipeline. Relative abundance of Anelloviridae, Nocardiaceae, Microbacteriaceae, and Enterobacteriaceae significantly changed during the postoperative period. Microbiome diversity was higher within a week after LT than that at 8 weeks after LT. Antimicrobials were significantly associated with the microbiome of LT recipients. In addition, the proportion of Enterobacteriaceae was significantly increased and the plasma microbiome diversity was significantly lower in patients with acute cellular rejection (ACR) than non-ACR patients. Sequencing reads of bacteria isolated from blood cultures were predominantly identified by NGS in 8 of 16 samples, and human herpesvirus 6 was detected as a causative pathogen in one recipient with severe clinical condition. Conclusions: The metagenomic NGS technique has great potential in revealing the plasma microbiome and is useful as a comprehensive diagnostic procedure in clinical settings. Direct-imaging techniques of exoplanets have made significant progress recently and will eventually enable monitoring of photometric and spectroscopic signals of Earth-like habitable planets. The presence of clouds, however, would remain as one of the most uncertain components in deciphering such direct-imaged signals of planets. We attempt to examine how the planetary obliquity produces different cloud patterns by performing a series of general circulation model simulation runs using a set of parameters relevant for our Earth. Then we use the simulated photometric lightcurves to compute their frequency modulation that is due to the planetary spin–orbit coupling over an entire orbital period, and we attempt to see to what extent one can estimate the obliquity of an Earth twin. We find that it is possible to estimate the obliquity of an Earth twin within the uncertainty of several degrees with a dedicated 4 m space telescope at 10 pc away from the system if the stellar flux is completely blocked. While our conclusion is based on several idealized assumptions, a frequency modulation of a directly imaged Earth-like planet offers a unique methodology to determine its obliquity. Background: Patients who undergo allogeneic hematopoietic stem cell transplantation (allo-HSCT) can experience musculoskeletal pains in their lower limbs in the early and late post-transplantation period. This study investigated demographics and clinical characteristics of the lower limb pain (LLP) among Japanese survivors of pediatric allo-HSCT.Methods: A total of 143 consecutive Japanese patients who had undergone allo-HSCT less than 18 years of age in a single institute between 2005 and 2015 were reviewed. Patients referred for in-house orthopedic evaluation of their sustained LLPs that impaired ambulation were defined as LLP group. Illness/transplantation-related parameters were compared between LLP group and non-LLP group.Results: Ninety children with a mean age of 8.5 years at transplantation were enrolled. Their median follow-up period following transplantation was 6.3 years (range, 2.1-12.5). There were four patients in LLP group, whose etiologies were AVN of the femoral head and insufficiency fracture (ISF) of the tibia or the medial cuneiform bone. Cumulative dose of steroids that administered from six months before transplantation to six months after discharge from hospitalization for transplantation was significantly higher in LLP group than non-LLP group. Additionally, the two groups differed significantly in terms of hospitalization period after transplantation. LLP caused by AVN of the femoral head manifested between six months and two years, whereas that caused by ISF within the first six months after transplantation.Conclusions: The incidence of sustained LLP that impairs ambulation following contemporary allo-HSCT is not common in Japanese pediatric survivors. The risk of developing musculoskeletal LLP may increase with a higher steroid dosage in the peri-transplant period. Background: Alveolar capillary dysplasia with misalignment of pulmonary veins (ACD/MPV) is a rare disorder of pulmonary vascular abnormality with persistent pulmonary hypertension of the newborn. The symptom usually presents within hours after birth, leading to an early demise. Heterozygous de novo point mutations and genomic deletions of the FOXF1 (forkhead box F1) gene or its upstream enhancer have been identified in most patients with ACD/MPV. Most cases of ACD/MPV are sporadic; however, familial cases are also reported in 10% of patients. Case presentation: We herein report a case of familial ACD/MPV that showed unusual glomeruloid proliferation of endothelial cells. In this family, three of the four siblings died within two to 3 days after birth because of persistent pulmonary hypertension and respiratory failure. Only the second child remains alive and healthy. An autopsy was performed for the third and fourth children, resulting in a diagnosis of ACD/MPV based on the characteristic features, including misalignment of smaller pulmonary veins and lymphangiectasis. In both of these children, glomeruloid endothelial proliferation of vessels was noted in the interlobular septa. The vessels were immunohistochemically positive for D2–40, CD31, Factor VIII, and ERG, suggestive of differentiation for both lymphatic and blood vessels. Conclusions: Unusual glomeruloid endothelial proliferation was observed in a familial ACD/MPV case. This histologic feature has not been described previously in ACD/MPV or any other pulmonary disease. Although the histogenesis of this histologic feature is unclear, this finding may suggest that ACD/MPV is a compound vascular and lymphovascular system disorder that exhibits various histologic features. We here report on 2 cases of monostotic Paget's disease of bone, one in the ilium and the other in the skull, including quantitative values obtained with bone single photon emission computed tomography/computed tomography (SPECT/CT), which were useful to evaluate the response to bisphosphonate treatment. The quantitative parameters determined with those findings were decreased, with the maximum standardized uptake value (SUV), peak SUV, mean SUV, metabolic bone volume, and total bone uptake in case 1 and case 2 reduced by 48.8 and 60.3%, 46.6 and 58.8%, 24.3 and 60.5%, 87.0 and 11.8%, and 90.2 and 55.8%, respectively, while TRACP-5B and alkaline phosphatase (ALP) were also reduced by 39.5 and 88.6% and by 53.7 and 78.1%, respectively. Hematopoietic stem cell transplantation (HSCT) is the only curative treatment for juvenile myelomonocytic leukemia (JMML), but few large studies of HSCT for JMML exist. Using data from the Japan Society for Hematopoietic Cell Transplantation registry, we analyzed the outcomes of 129 children with JMML who underwent HSCT between 2000 and 2011. The 5-year overall survival (OS) rate and cumulative incidence of relapse were 64% and 34%, respectively. A regimen of busulfan/fludarabine/melphalan was the most commonly used (59 patients) and provided the best outcomes; the 5-year OS rate reached 73%, and the cumulative incidences of relapse and transplantation-related mortality were 26% and 9%, respectively. In contrast, the use of the irradiation-based myeloablative regimen was the most significant risk factor for OS (hazard ratio [HR], 2.92; P =.004) in the multivariate model. In addition, chronic graft-versus-host disease (GVHD) was strongly associated with lower relapse (HR, 0.37; P =.029) and favorable survival (HR, 0.22; P =.006). The current study has shown that a significant proportion of children with JMML can be cured with HSCT, especially those receiving the busulfan/fludarabine/melphalan regimen. Based on the lower relapse and better survival observed in patients with chronic GVHD, additional treatment strategies that focus on enhancing graft-versus-leukemia effects may further improve survival. There have been increasing number of reports of SZT2-related neurological diseases, the main symptoms of which are epilepsy, developmental delay, macrocephaly and a dysmorphic corpus callosum. SZT2 functions as a regulator of mechanistic target of rapamycin complex 1 (mTORC1) signaling in cultured human cell lines and mouse tissues. However, it remains to be determined whether mutations in SZT2 in human patients alter mTORC1 signaling. In this study, we aimed to investigate the functional consequence of biallelic SZT2 variants in Epstein-Barr virus-induced lymphoblastoid cell lines (LCLs) established from two patients with a typical SZT2-related neurodevelopmental disease. Increased phosphorylation of S6 kinase and S6 was identified in patient-derived cell lines under amino acid-starved condition, suggestive of constitutive hyperactivation of mTORC1 signaling. This result was validated by constitutive lysosomal localization of mTOR in patients' LCLs. Furthermore, patients' LCLs display an excessive response to slight amino acid stimulation. Our data suggest the loss-of-function nature of SZT2 mutations in the patients, and consequent hyperactivation of mTORC1 signaling in response to both amino acid starvation and stimulation in their LCLs. By these functional analyses, the pathogenicity of newly identified SZT2 variants can be determined, allowing for more detailed characterization of genotype-phenotype correlations. Objective: Urinary tract infection (UTI), one of the most common bacterial infections occurring during infancy and early childhood, is frequently associated with vesicoureteral reflux (VUR). Although several guidelines recommend performing ultrasonography as a screening test, its utility is not adequate and appropriate screening tests are strongly desirable. In this study, we evaluate the use of magnetic resonance imaging (MRI) as a screening test for VUR in children with UTI. Methods: We prospectively studied 108 patients with suspected UTI between April 2014 and March 2016. UTI was diagnosed on the basis of diffusion-weighted MRI (DW-MRI) and urine culture findings. We measured ureteral dilatation using MRI in 96 patients with UTI and assessed the relationship between ureteral dilatation in MRI and VUR in 46 patients who underwent voiding cystourethrography (VCUG). Results: Among 108 patients, 88 and 8 were diagnosed with upper and lower UTI, respectively. Among 46 patients who underwent VCUG, 23 had VUR (14 low grade and 9 high grade). Patients with ureteral dilatation detected on MRI had VUR more frequently than those without ureteral dilatation (any grades VUR, 71% vs. 32%; P = 0.02; high-grade VUR, 38% vs. 2%, P = 0.007). Overall, ureteral dilatation findings on MRI achieved sensitivity 65.2% and specificity 73.9% as a screening test for VUR. In addition, DW-MRI achieved sensitivity 100% and specificity 81.8% in the diagnosis of upper UTI. To better understand the formation of water-soluble organic aerosols in the forest atmosphere, we measured low molecular weight (LMW) dicarboxylic acids, oxocarboxylic acids, α-dicarbonyls, unsaturated fatty acids (UFAs), and water-soluble organic carbon (WSOC) in aerosols from a Larix kaempferi forest located at the northern slope of Mt. Fuji, Japan, in summer 2012. Concentrations of dicarboxylic acids, oxocarboxylic acids, α-dicarbonyls, and WSOC showed maxima in daytime. Relative abundance of oxalic acid in LMW dicarboxylic acids was on average 52% and its average concentration was 214 ng m −3 . We found that diurnal and temporal variations of oxalic acid are different from those of isoprene and α-pinene, whereas biogenic secondary organic aerosols (BSOAs) derived from isoprene and α-pinene showed similar variations with oxalic acid. The mass concentration ratios of oxalic acid/BSOAs were relatively constant, although a large variation in the concentrations of toluene that is an anthropogenic volatile organic compound was observed. These results suggest that formation of oxalic acid is associated with the oxidation of isoprene and α-pinene with O 3 and other oxidants in the forest atmosphere. In addition, concentrations of UFAs were observed, for the first time, to decrease dramatically during daytime in the forest. Mass concentration ratios of azelaic acid to UFAs showed a positive correlation with O 3 , suggesting that UFAs are oxidized to yield azelaic acid, which may be further decomposed to oxalic acid in the forest atmosphere. We found that contributions of oxalic acid to WSOC are significantly high ranging from 3.7 to 9.7% (average 6.0%). Allogeneic hematopoietic stem cell transplantation (allo-HSCT) remains the best therapeutic option for childhood high-risk acute leukemia. However, which donor source is optimal for children lacking an identical sibling remains unclear. To evaluate the clinical impact of donor source on allo-HSCT in childhood acute leukemia, we analyzed data from 577 children who underwent allo-HSCT after a myeloablative regimen during first or second complete remission from 2005 to 2012, using registry data of the Japan Society for Hematopoietic Cell Transplantation, and we compared outcomes of 7/8 to 8/8 HLA allelic–matched unrelated bone marrow transplantation (UR-BMT, n = 218) and 4/6 to 6/6 HLA allelic–matched unrelated cord blood transplantation (UR-CBT, n = 200) to those of HLA-identical related bone marrow transplantation (ID-BMT, n = 159). The median follow-up of survivors was 40.0 months. Three-year overall survival (OS) and leukemia-free survival (LFS) rates for ID-BMT, UR-BMT, and UR-CBT were 74.8% and 69.0%, 75.0% and 69.6%, and 71.8% and 63.8%, respectively. The multivariate analysis demonstrated that OS and LFS for the 3 groups are comparable, although UR-CBT carries a greater risk of nonrelapse mortality (hazard ratio, 2.20; P  = .03, compared to ID-BMT) in the myeloablative setting for childhood high-risk acute leukemia. Earlier reports suggested that umbilical cord blood transplantation (UCBT) for aplastic anemia (AA) was feasible in alternative transplantation. To identify differences in outcomes of UCBT and HLA-matched or mismatched unrelated bone marrow transplantation (UBMT) in adults with AA, we analyzed registry data of the Japan Society for Hematopoietic Cell Transplantation and compared results of UCBT (n = 69) to 8/8-matched (n = 101), 7/8-matched (n = 65), or 6/8-matched (n = 37) UBMT. The transplantation period was from 2002 to 2012, and patients 16 years or older with AA were eligible. Median ages were 49, 35, 28, and 30 years for UCBT, 8/8-matched, 7/8-matched, and 6/8-matched UBMT, respectively. In multivariate analysis, risk of mortality was lower for 8/8-matched UBMT compared with that of UCBT (hazard ratio [HR], .55; 95% confidence interval [CI], .32 to .94; P  = .029), adjusted for age and graft-versus-host disease (GVHD) prophylaxis, which were other associated factors. Mortality risks of 7/8-matched UBMT (HR, .55; 95% CI, .29 to 1.02) or 6/8-matched UBMT (HR, .67; 95% CI, .32 to 1.39) were not significantly different from those of UCBT. Risks of grade 3 or 4 acute and chronic GVHD were not different among the 4 groups. The most prevalent cause of death was graft failure in UCBT and 6/8-matched UBMT and infection in 8/8-matched and 7/8-matched UBMT. Under 40 years old,survival of UCBT was similar to that of UBMT (76%, 79%, 83%, and 83% for UCBT and 8/8-matched, 7/8-matched, and 6/8-matched UBMT, respectively, at 3 years), adjusted for transplantation period, which was another associated factor; however, for ages over 40 years, that of UCBT tended to be lower (47%, 64%, 64%, and 75% for UCBT, 8/8-matched, 7/8-matched, and 6/8-matched UBMT, respectively, at 3 years). Juvenile myelomonocytic leukemia (JMML), an overlap of myelodysplastic / myeloproliferative neoplasm, is an intractable pediatric myeloid neoplasm. Epigenetic regulation of transcription, particularly by CpG methylation, plays an important role in tumor progression, mainly by repressing tumor-suppressor genes. To clarify the clinical importance of aberrant DNA methylation, we studied the hypermethylation status of 16 target genes in the genomes of 92 patients with JMML by bisulfite conversion and the pryosequencing technique. Among 16 candidate genes, BMP4, CALCA, CDKN2A, and RARB exhibited significant hypermethylation in 72% (67/92) of patients. Based on the number of hypermethylated genes, patients were stratified into three cohorts based on an aberrant methylation score (AMS) of 0, 1–2, or 3–4. In the AMS 0 cohort, the 5-year overall survival (OS) and transplantation-free survival (TFS) were good (69% and 76%, respectively). In the AMS 1–2 cohort, the 5-year OS was comparable to that in the AMS 0 cohort (68%), whereas TFS was poor (6%). In the AMS 3–4 cohort, 5-year OS and TFS were markedly low (8% and 0%, respectively). Epigenetic analysis provides helpful information for clinicians to select treatment strategies for patients with JMML. For patients with AMS 3–4 in whom hematopoietic stem cell transplantation does not improve the prognosis, alternative therapies, including DNA methyltransferase inhibitors and new molecular-targeting agents, should be established as treatment options. Therapy for BK virus (BKV)–associated hemorrhagic cystitis (BKV-HC) is limited after hematopoietic stem cell transplantation (HSCT). We examined whether choreito, a formula from Japanese traditional Kampo medicine, is effective for treating BKV-HC. Among children who underwent allogeneic HSCT between October 2006 and March 2014, 14 were diagnosed with BKV-HC (median, 36 days; range, 14 to 330 days) after HSCT, and 6 consecutive children received pharmaceutical-grade choreito extract granules. The hematuria grade before treatment was significantly higher in the choreito group than in the nonchoreito group ( P = .018). The duration from therapy to complete resolution was significantly shorter in the choreito group (median, 9 days; range, 4 to 17 days) than in the nonchoreito group (median, 17 days; range, 15 to 66 days; P = .037). In 11 children with macroscopic hematuria, the duration from treatment to resolution of macroscopic hematuria was significantly shorter in the choreito group than in the nonchoreito group (median, 2 days versus 11 days; P = .0043). The BKV load in urine was significantly decreased 1 month after choreito administration. No adverse effects related to choreito administration were observed. Congenital macrothrombocytopenia (CMTP) is a heterogeneous group of rare platelet disorders characterized by a congenital reduction of platelet counts and abnormally large platelets, for which CMTP-causing mutations are only found in approximately half the cases. We herein performed whole-exome sequencing and targeted Sanger sequencing to identify mutations that cause CMTP, in which a dominant mode of transmission had been suspected but for which no known responsible mutations have been documented. In 13 Japanese CMTP-affected pedigrees, we identified six (46%) affected by ACTN1 variants cosegregating with CMTP. In the entire cohort, ACNT1 variants accounted for 5.5% of the dominant forms of CMTP cases and represented the fourth most common cause in Japanese individuals. Individuals with ACTN1 variants presented with moderate macrothrombocytopenia with anisocytosis but were either asymptomatic or had only a modest bleeding tendency. ACTN1 encodes a-actinin-1, a member of the actin-crosslinking protein superfamily that participates in the organization of the cytoskeleton. In vitro transfection experiments in Chinese hamster ovary cells demonstrated that altered α-actinin-1 disrupted the normal actin-based cytoskeletal structure. Moreover, transduction of mouse fetal liver-derived megakaryocytes with disease-associated ACTN1 variants caused a disorganized actin-based cytoskeleton in megakaryocytes, resulting in the production of abnormally large proplatelet tips, which were reduced in number. This study determined the energy content and immunoglobulin G (IgG) concentration of Awassi ewe colostrum and examined the effects of parity and litter size on them. Colostral samples were collected from 57 ewes from the 1st to 7th parities with a litter size of 1 or 2, within 12 h after lambing. Concentrations of colostral fat, lactose, and protein were measured as energy sources, and IgG was measured as representative of the immunoglobulin content of the colostrum. Mean values obtained from analyses of the ewe colostrum samples were 7.4 ± 2.1 kJ/g for the energy value and 60.9 ± 21.4 mg/mL for IgG. Ewe parity did not influence colostral energy content, whereas the IgG concentration of the colostrum obtained from the primiparous ewes was higher than that from the multiparous ewes (P < 0.05). Among the multiparous ewes, the colostrum obtained from ewes carrying twins showed higher energy content and IgG concentrations than that from those carrying a single lamb (P < 0.05). We conducted a soil warming experiment in a cool-temperate forested peatland in northern Japan during the snow-free seasons of 2007-2011, to determine whether the soil warming would change the heterotrophic respiration rate and its temperature sensitivity. We elevated the soil temperature by 3°C at 5-cm depth by using overhead infrared heaters and continuously measured hourly soil CO2 fluxes with a 15-channel automated chamber system. The 15 chambers were divided into three groups each with five replications for the control, unwarmed-trenched and warmed-trenched treatments. Soil warming enhanced heterotrophic respiration by 82% (mean of four seasons (2008-2011) observation±SD, 6.84±2.22µmol C m-2 s-1) as compared to the unwarmed-trenched treatment (3.76±0.98 µmol C m-2 s-1). The sustained enhancement of heterotrophic respiration with soil warming suggests that global warming will accelerate the loss of carbon substantially more from forested peatlands than from other upland forest soils. Soil warming likewise enhanced temperature sensitivity slightly (Q10, 3.1±0.08 and 3.3±0.06 in the four-season average in unwarmed- and warmedtrenched treatments, respectively), and significant effect was observed in 2009 (p<0.001) and 2010 (p<0.01). However, there was no significant difference in the basal respiration rate at 10°C (R10, 2.2±0.52 and 2.8±1.2 µmol C m-2 s-1) between treatments, although the values tended to be high by warming throughout the study period. These results suggest that global warming will enhance not only the heterotrophic respiration rate itself but also its Q10 in forests with high substrate availability and without severe water stress, and predictions for such ecosystems obtained by using models assuming no change in Q10 are likely to underestimate the carbon release from the soil to the atmosphere in a future warmer environment. Juvenile myelomonocytic leukemia (JMML) is a rare pediatric myeloid neoplasm char-acterized by excessive proliferation of myelomonocytic cells. Somatic muta-tions in genes involved in GM-CSF signal transduction, such as NRAS, KRAS, PTPN11, NF1, and CBL, have been identi-fied in more than 70% of children with JMML. In the present study, we report 2 patients with somatic mosaicism for oncogenic NRAS mutations (G12D and G12S) associated with the development of JMML. The mutated allele frequencies quantified by pyrosequencing were vari-ous and ranged from 3%-50% in BM and other somatic cells (ie, buccal smear cells, hair bulbs, or nails). Both patients experi-enced spontaneous improvement of clinical symptoms and leukocytosis due to JMML without hematopoietic stem cell transplantation. These patients are the first reported to have somatic mosaicism for oncogenic NRAS mutations. We measured the methane flux of a forest canopy throughout a year using a relaxed eddy accumulation (REA) method. This sampling system was carefully validated against heat and CO fluxes measured by the eddy covariance method. Although the sampling system was robust, there were large uncertainties in the measured methane fluxes because of the limited precision of the methane gas analyzer. Based on the spectral characteristics of signals from the methane analyzer and the diurnal variations in the standard deviation of the vertical wind velocity, we found the daytime and nighttime precision of half-hourly methane flux measurements to be approximately 1.2 and 0.7 μg CH m s, respectively. Additional uncertainties caused by the dilution effect were estimated to affect the accuracy by as much as 0.21 μg CH m s on a half-hourly basis. Diurnal and seasonal variations were observed in the measured fluxes. The biological emission from plant leaves was not observed in our studies, and thus could be negligible at the canopy-scale exchange. The annual methane sink was 835 ± 175 mg CH m year (8.35 kg CH ha year), which was comparable to the flux range of 379-2,478 mg CH m year previously measured in other Japanese forest soils. This study indicated that the REA method could be a promising technique to measure canopy scale methane fluxes over forests, but further improvement of precision of the analyzer will be required. EBV-associated T/NK-cell lymphoproliferative disease (T/NK-LPD) is defined as a systemic illness characterized by clonal proliferation of EBV-infected T or NK cells. We prospectively enrolled 108 nonimmunocompromised patients with this disease (50 men and 58 women; median onset age, 8 years; age range, 1-50 years) evidenced by expansion of EBV+ T/NK cells in the peripheral blood; these were of the T-cell type in 64 cases and of the NK-cell type in 44, and were clinically categorized into 4 groups: 80 cases of chronic active EBV disease, 15 of EBV-associated hemophagocytic lymphohistiocytosis, 9 of severe mosquito bite allergy, and 4 of hydroa vacciniforme. These clinical profiles were closely linked with the EBV+ cell immunophenotypes. In a median follow-up period of 46 months, 47 patients (44%) died of severe organ complications. During the follow-up, 13 patients developed overt lymphoma or leukemia characterized by extranodal NK/T-cell lymphoma and aggressive NK-cell leukemia. Fifty-nine received hematopoietic stem cell transplantation, 66% of whom survived. Age at onset of disease (≥ 8 years) and liver dysfunction were risk factors for mortality, whereas patients who received transplantation had a better prognosis. These data depict clinical characteristics of systemic EBV+ T/NK-LPD and provide insight into the diagnostic and therapeutic approaches for distinct disease. [ABSTRACT FROM AUTHOR]

Synthesis studies using multiple-site datasets for eddy covariance potentially contain uncertainties originating from the use of different flux calculation options, because the choice of the process for calculating half-hourly fluxes from raw time series data is left to individual researchers. In this study, we quantified the uncertainties associated with different flux calculation methods at seven sites. The differences in the half-hourly fluxes were small, generally of the order less than a few percentiles, but they were substantial for the annual fluxes. After the standardisation under current recommendations in the FLUXNET communities, we estimated the uncertainties in the annual fluxes associated with the flux calculations to be 2.692.7 W m-2 (the mean 90% 9 confidence interval) for the sensible heat flux, 72937 g C m-2 yr-1 for net ecosystem exchange (NEE), 1296% for evapotranspiration, 1296% for gross primary productivity and 16910% for ecosystem respiration. The selfheating correction strongly influenced the annual carbon balance (143993 g C m-2 yr-1), not only for cold sites but also for warm sites, but did not fully account for differences between the open- and closed-path systems (4139189 g C m-2 yr-1).
