Dendritic cell (DC) targeting by DEC205
+ cells effectively promotes the internalization of antigens that may trigger a specific immune response. In this study, we evaluated the ability of a recombinant antibody, anti-DEC205 (rAb ZH9F7), to trigger cellular endocytosis in subpopulations of DCs and targeted cells after intradermal injection and subsequent migration toward lymph nodes. Furthermore, the cellular immune response was evaluated in pigs after intradermal application of the antigenized rAb ZH9F7 combined with porcine circovirus type 2 cap antigen (rAb ZH9F7-Cap). We demonstrated that rAb ZH9F7 recognized conventional type 1 and 2 DCs from the blood and skin and monocytes. It promoted receptor-mediated endocytosis and migration of cDCs and moDCs toward regional lymph nodes. Intradermal application of rAb ZH9F7-Cap induced a higher frequency of IFN-γ-secreting CD4+ CD8+ T lymphocytes and antibodies against Cap protein than that in the control group. In conclusion, the rAb ZH9F7-Cap system promoted the target of skin cDC1 and cDC2, provoking migration to the regional lymph nodes and inducing a Th1 response, as evidenced by the proliferation of double-positive CD4+ CD8+ T cells, which correlates with an enhanced ability to target the cDC1 subset both in vitro and in vivo. RESUMEN Dentro de los componentes de la diversidad biológica se encuentran la diversidad taxonómica y genética. El análisis de ambos resulta indispensable para la elaboración de estrategias de manejo y conservación de áreas prioritarias para la conservación. La península de Hualpén alberga uno de los últimos relictos de bosque costero de la Región del Bío-Bío, concentrado en la Estación de Biología Terrestre (EBT) cedida en comodato a la Universidad de Concepción, por parte de la gobernación de Concepción. En este estudio se evaluó la variabilidad taxonómica y genética de la península con el fin de contribuir al desarrollo de estrategias de conservación y futuro desarrollo de planes de manejo en esta. Para ello se cuantificó la riqueza de especies a lo largo de 20 transectos, localizados dentro de la península. A su vez, se evaluó la diversidad genética con marcadores AFLP para cinco especies no relacionadas filogenéticamente. Se encontró un total de 90 especies, donde el 78% son nativas y el 22% son especies introducidas. La riqueza de especies nativas se concentró en la EBT, presentando diferencias significativas con el resto de los sitios muestreados. El análisis de los marcadores moleculares mostró valores de diversidad genética congruentes con los reportados para especies de similares características biológicas. Además, los resultados de diversidad genética para las especies estudiadas concuerdan con los reportados en otros sitios de su distribución geográfica. Por lo tanto, la EBT concentra altos valores de diversidad específica y genética. Esto podría ser el reflejo de la protección del sitio por parte de la entidad administradora. Considerando la presencia de especies en peligro dentro de la estación biológica y la riqueza de especies en el área, se aconseja incluir a la EBT y algunos lugares aledaños en el sistema de áreas protegidas por el Estado. ABSTRACT Taxonomic and genetic diversity are among the components of biodiversity. Both components need to be taken in consideration to establish management and conservation strategies in high-priority areas for conservation. Pedro del Rio Zañartu park in the peninsula of Hualpen, contains one of the last relicts of the coastal forest of the Bio-Bio Region, mainly concentrated in the Terrestrial Biological Station (EBT) assigned to the University of Concepcion near to the city of Concepcion. In this study, we evaluated both taxonomic and genetic diversity of the peninsula to establish conservation strategies and future management plans in the area. We quantified the number of species along 20 transects within the park and in the surrounding areas. In addition, we measured the genetic diversity in five phylogenetically unrelated species, using AFLP markers. A total of 90 species were recorded, 78 % native and 22 % non-native. The highest species richness was found in the EBT area, having significance difference with the rest of the sampling sites. The genetic analysis showed similar values of genetic diversity than previously reported for species with similar biological traits and for the same studied species, in different zones of their distribution. Therefore, high levels of species richness and genetic diversity were found in the EBT. This could be as a result of the management and protection by the university. Considering the presence of endangered species within the EBT and the spatial distribution of species richness in the area, we recommend that the park Pedro del Rio Zañartu and surrounding areas need to be part of the areas protected by the government.

RESUMEN Fusarium circinatum es el hongo causante de la enfermedad "cancro resinoso" (pitch canker), que afecta numerosas especies de coníferas. En esta revisión se describe el estado del arte de F. circinatum a nivel mundial y su situación actual en Chile, abarcando antecedentes del patógeno y de la enfermedad. En Chile, el patógeno fue detectado en setos de Pinus radiata a fines de 2001, encontrándose hoy presente en 39 viveros de un total de 196, ubicados entre las regiones de O'Higgins y de Los Ríos. Después de 14 años de la detección de F. circinatum en Chile, el patógeno no ha presentado diseminación en las plantaciones comerciales de P. radiata. Se destacan las medidas para el manejo integrado de la enfermedad, donde la resistencia genética sería la principal estrategia ante un escenario en que el cancro resinoso se presente en las plantaciones comerciales de P. radiata en Chile. ABSTRACT Fusarium circinatum is the fungus that causes the disease "canker" (pitch canker), which affects many species of conifers. This review describes the state of the art of F. circinatum worldwide and the current situation in Chile, covering the history of the pathogen and the disease. In Chile, the pathogen was detected in Pinus radiata hedges in late 2001, being present today in 39 of 196 nurseries, located between the regions of O'Higgins and Los Ríos. After 14 years of the detection of F. circinatum, the pathogen has not attacked commercial plantations of P. radiata. Measures for integrated disease management, where genetic resistance constitutes the main strategy in a scenario in which pitch canker could appear in commercial plantations of P. radiata in Chile.

12. Variabilidad genética mediante AFLP en tres relictos de Gomortega keule (Molina) Baillon: especie endémica chilena en peligro de extinción

Gomortega keule, especie arbórea endémica de Chile y única representante de la familia Gomortegaceae, se encuentra catalogada en peligro de extinción y despierta gran interés científico por su restringida área de distribución. El objetivo de este trabajo fue estimar la variabilidad genética de 24 individuos de G. keule ubicados en tres relictos de la provincia de Concepción, mediante marcadores AFLP (polimorfismos en la longitud de los fragmentos amplificados) para futuros estudios de conservación. Se probaron 16 combinaciones de partidores y se eligieron las tres combinaciones más informativas para detectar diferencias entre individuos. Estas combinaciones generaron un total de 156 fragmentos, siendo un 62 % de ellos polimórficos. El sitio con mayor porcentaje de polimorfismos correspondió a Cerro Neuque, con un 84,6 %, seguido por Coroney y Hualqui con 59,6 y 42,3 %, respectivamente. El análisis de varianza molecular detectó un 73 % de variación dentro de las poblaciones y un 27 % de variación entre ellas. El estudio muestra que la técnica de AFLP puede ser utilizada en G. keule de forma rápida y confiable para identificar poblaciones remanentes y apoyar su conservación tanto ex situ como in situ. Gomortega keule is an endemic woody species to Chile and the only member of the Gomortegaceae family. It is currently an endangered species and is of great scientific interest due to its small area of distribution. The aim of this research was to estimate genetic diversity of 24 individuals of G. keule selected from three isolated relict populations from the province of Concepción using AFLP (Amplified fragment length polymorphism) for future conservation issue. Sixteen combinations of primers for AFLP were tested and three combinations of them were selected as the most informative, revealing differences between individuals. These primer combinations generated 156 fragments, of which 62 % were polymorphic. The relict population with the highest percentage of polymorphisms was located in Cerro Neuque with 84.6 %, followed by Coroney and Hualqui with 59.6 and 42.3 %, respectively. The analysis of molecular variance detected 73 % variation within populations and 27 % variation amoung populations. This study supported that AFLP can be used in G. keule as a fast and reliable test to identified remaining populations of the species. Thus, this technique can be used to support ex situ or in situ conservation of G. keule.

La madera es un recurso natural renovable que proporciona materia prima para la construcción, generación de energía, fibras para la producción de pulpa y papel, paneles y tableros, y más recientemente para biocombustibles y biomateriales, lo que la hace el quinto producto comercial más importante del mundo. Diferencias en las propiedades físicas, químicas y anatómicas de la madera la hacen un material altamente variable y complejo. Estas propiedades influyen de forma directa en su valor comercial y derivan del tamaño, forma y arreglo de los diferentes tipos de células, y de la estructura y composición química de la pared celular de las distintas células del xilema. Éstas a su vez, están controladas por distintos factores ambientales y genéticos que regulan los procesos de biosíntesis de los diferentes componentes de la madera. Para aplicar herramientas moleculares y genéticas que permitan optimizar los procesos de selección de especies forestales con características superiores, es necesario entender los mecanismos que regulan estas propiedades, lo cual incluye comprender la dinámica de la estructura de la pared celular desde la formación de la célula cambial inicial hasta la formación de las células diferenciadas finales que conforman el xilema. La presente revisión explora las principales características anatómicas y el proceso de xilogénesis, enfocado particularmente en angiospermas, discutiendo los aspectos genéticos involucrados en la determinación de las características anatómicas de la madera, como dimensiones de los elementos celulares y características de la fibra, basándose en diferentes estudios realizados con especies de los géneros Populus y Eucalyptus, además de la planta modelo Arabidopsis thaliana. Wood is a renewable natural resource that provides raw material for construction, power generation, fibers for pulp and paper production, panels and boards, and more recently, biofuels and biomaterials, making it the fifth most important commercial product in the world. The wood is a highly variable and complex material that has different chemical, physical and anatomical properties that are influencing their commercial value. These properties depend on the size, shape and arrangement of the different cell types, and of the structure and chemical composition of the xylem cell wall. At the same time, these properties are controlled by different environmental and genetic factors that regulate the biosynthesis processes of the different wood components. To apply molecular and genetic tools to optimize the selection processes of forest species with superior traits, it is necessary to understand the mechanisms that regulate these properties, as the dynamics of cell wall structure from the initial cambial cell formation to the final differentiated cells formation that compose the xylem. This review explores the main anatomical traits and the xylogenesis processes, particularly focused in angiosperms, discussing the genetic aspects involved in the determination of wood anatomical characteristics, as cellular elements and fiber characteristics, based on different studies with species of the Populus and Eucalyptus genus, besides the model plant Arabidopsis thaliana.

14. Establishment and optimization of ISSR and SAMPL molecular markers as a tool for breeding programs of Pinus radiata

Pinus radiata is a conifer of recognized economic value. Studies at genetic and molecular level to support breeding programs of radiata pine have been conducted with difficulty due to its large and complex genome. The development of molecular markers has allowed performing studies on genetic variation, identification of clones and construction of linkage maps. In order to increase the genetic molecular markers in P. radiata, two inter simple sequence repeat (ISSR) markers and twelve selective amplifications of microsatellite polymorphic loci (SAMPL) primer combinations were tested, using a first-generation full-sib family of 86 individuals. Polymerase chain reaction (PCR) products were visualized by capillary electrophoresis and polymorphism was detected by presence or absence of a particular fragment in one P. radiata parental. A total of 18 polymorphic fragments were found for two ISSR primers tested, with an average segregation distortion of 33 %. SAMPL markers yielded 85 polymorphic fragments with eight primer combinations, with an average of 22.3 % segregation distortion. Both methodologies showed a good reproducibility, with easy implementation and useful for genetic analysis on P. radiata. Pinus radiata es una conífera de reconocido valor económico, en la cual los estudios a nivel genético y molecular, han sido conducidos con dificultad en programas de mejoramiento debido a su genoma complejo y de gran tamaño. El desarrollo de marcadores moleculares ha permitido realizar estudios de variación genética, validar clones y construir mapas de ligamiento. Para lograr incrementar la disponibilidad de marcadores moleculares en P. radiata, se examinaron dos marcadores ISSR (inter simple sequence repeat) y 12 combinaciones de partidores SAMPL (selective amplification of microsatellite polymorphic loci), usando una familia F1 de 86 hermanos completos de primera generación. Los productos de PCR (polymerase chain reaction) se visualizaron mediante electroforesis capilar y la selección de fragmentos polimórficos se efectuó a base de la presencia o ausencia de un fragmento en uno de los parentales de la familia de P. radiata. Un total de 18 fragmentos polimórficos fueron encontrados para los dos partidores ISSR, con una distorsión de segregación promedio de 33 %. Los marcadores SAMPL generaron 85 fragmentos polimórfcos empleando ocho combinaciones, con un promedio de 22,3 % de distorsión de segregación. Ambas metodologías mostraron buena reproducibilidad, fácil implementación y utilidad para análisis genético en P. radiata.

The benefits from the development of transgenic trees are expected from the improvement of traits as growth and form, wood quality, industrial processes, disease and insect resistance, herbicide tolerance, ecological restoration, rooting ability, etc. One of the first reported field trials with genetically modified forest trees was established in Belgium in 1988 and the characteristic evaluated was herbicide tolerance in poplars. Since then, there have been more than 200 reported trials, involving at least 15 forest species. The majority of the field trials have been carried out in the USA (64%). More than 50% of the field trials are done with Populus species and the main target traits are herbicide tolerance (31%), followed by marker genes (23%) and insect resistance (14%). Until today, there is only one report on commercial-scale production of transgenic forest trees which is Populus nigra with the Bt gene release in China in 2002 and established on commercial plantations in 2003. Operational application of GMO's in forestry depends on technical, economical, political and public aspects, but the development of adequate regulatory frameworks and public acceptance of transgenic trees will define the future of this technology in forestry.

16. Transformation von Nicotiana benthamiana mit verschiedenen Sequenzen des BWYV (Beet western yellows virus) zur Virus Resistenztestung

N. benthamiana plants were transformed by means of A. tumefaciens with either the viral replicase (ORF1/2), one of two smaller sequences involving the 5' and 3' ends (5'3'S and 5'3'AS) of the genome of BWYV or with the pBin19 vector alone (Kp). In total ca. 120 kanamycin (Km) resistant plantlets were generated. Of these 115 were positive by NPTII ELISA and/or PCR for the transgene (50, 24, 25 and 16 plants containing the ORF1/2, 5'3'S, 5'3'AS and Kp, respectively). Plants were transferred to the greenhouse and allowed to self-pollinate. Seeds from the primary transformants (To) were collected and tested for segregation on different Km concentrations (100 to 300 mg/l). Seeds from ca. 30 of the lines tested did not germinate or had a poor growth at these antibiotic concentrations. According to different criteria such as levels of NPTII ELISA of To lines, PCR, growth at 200 mg/l of Km and phenotype some lines were choosen as candidates to be tested for resistance against BWYV. 5 lines of each construct were randomly selected. These lines were analysed by Northern blot for expression of the transgene and/or the nptII gene. Different levels of expression of the transgene were found. Greenhouse resistance tests were performed twice (from June to August 1999 and August to October 1999). 15 transgenic N. benthamiana lines (5 per construct) as well as two controls (vector transformed and untransformed plants) were tested. Km resistant plantlets were transferred to the greenhouse and 20 plants from each line were inoculated with BWYV by means of green peach aphids (Myzus persicae), while other 20 were kept as uninoculated control. At 4, 6 and 8 weeks post infection (wpi) an BWYV ELISA of the inoculated plants was carried out and the height of each plant was measured, while the weight was determined at the end of the experiment (8 wpi). All 5'3'AS lines tested were susceptible to BWYV. Although plants from line SV 77 had low ELISA values at all times tested their final w, N. bethamiana Pflanzen wurden mittels Agrobakterien mit der viralen Replikase (ORF1/2), einer fusionierte Sequenz, die das 5' und 3' Ende (5'3'S und 5'3'AS) des BWYV enthält, oder nur mit dem pBin19 Vektor (Kp) transformiert. Infolge unterschiedlicher Kriterien wie NPTII ELISA der To Linien, PCR, Wachstum auf 200 mg/l Km und Phänotyp wurden je 5 Linien pro Konstrukt als Kandidaten für den Resistenztest gegen BWYV ausgewählt. Diese Linien wurden im Northern blot auf die Expression der viralen Gensequenzen analysiert. Unterschiedliche Expressionsniveaus dieser Gene wurden gefunden. Gewächshaus-Resistenztests wurden zweimal (von Juni bis August 1999 und August bis Oktober 1999) durchgeführt. 15 transgene Linien (5 je Konstrukt) und jeweils 2 Kontrollen (mit dem Vektor-transformierten und nicht transformierte Pflanzen) wurden getestet. Km-resistente Pflanzen wurden in das Gewächshaus überführt und 20 Pflanzen jeder Linie mittels Grüner Pfirsichblattlaus (M. persicae) mit BWYV inokuliert, während andere 20 nicht inokulierte Pflanzen als Kontrolle dienten. Jeweils 4, 6 und 8 Wochen nach Infektion erfolgten ein BWYV ELISA-Test der inokulierten Pflanzen, das Vermessen der Wuchshöhe und am Ende des Experiments (8 Wochen nach Infektion) das Bestimmen des Gewichts. Alle getesteten 5'3'AS Linien waren anfällig gegen BWYV. Obwohl die Pflanzen der Linie SV 77 bei allen Tests einen niedrigen ELISA-Wert hatten, waren das Gewicht und die Höhe ähnlich

Tesis (Magíster en Terapia Ocupacional, Mención Salud Física) A nivel mundial existe una tendencia global de envejecimiento de la población de la que Chile no está ajeno. Estas personas que van envejeciendo, en la mayoría de los casos se encuentran en situación de dependencia severa, la cual viene acompañada como consecuencia de un cuidador, el cual debe tomar grandes responsabilidades, dar apoyo y realizar todas las actividades de la vida diaria de la persona dependiente. La mayoría de los cuidadores son familiares o cuidadores “no formales” que deben lidiar con reorganizar su propia rutina y la de la persona que cuida, ya que de un momento a otro deben sacrificar su propia vida por cuidar a otro. Además los roles de cada cuidador y de toda su familia también son un factor importante en la vida de una persona, los que se ven afectados significativamente en la vida cotidiana. Sabemos que dichas responsabilidades y agobiadas rutinas ocupacionales progresivamente van deteriorando la calidad de vida de estos cuidadores provocando el conocido “estrés del cuidador”. Existen investigaciones que dan cuenta de ello pero no se conoce sobre el proceso de resiliencia que ellos viven a diario, el cual se va formando a través del discurso social y de las propias experiencias del día a día. Pensando en esto, creemos que a través de un pensamiento socio construccionista podremos dar sentido a nuestra tesis, pensando en la idea de que a través del discurso e intercambio de experiencias creamos un lenguaje en común y nuevas formas de actuar. Por tal motivo nos preguntamos ¿Cuáles son las áreas en las que se ve afectada la vida cotidiana y construyen resiliencia en el tiempo los cuidadores de personas en situación de dependencia severa? El principal propósito de la investigación fue identificar las percepciones de los cuidadores en relación al proceso de resiliencia que han adquirido en el tiempo, además de identificar los roles y rutinas alterados en el proceso de cuidar a una persona dependiente. La metodología empleada tiene un enfoque cualitativo de forma que describiremos todas las interrogantes. Para la recolección de datos, se utilizó la entrevista semi estructurada, la observación y la realización de un grupo de discusión entre los entrevistados. Ahora la forma en la que analizaremos e interpretaremos la información entregada es a través de la técnica codificación de tipo abierta y axial. Como principales resultados, se obtiene que la existencia de cambios en las rutinas y roles, incluyen percepciones positivas y negativas asociadas, potenciando o no la resiliencia como tal.

20. Beyond the outcomes: generic change indicators in a video-feedback intervention with a depressed mother and her baby: a single case study.

Child and dyadic psychotherapy have been scarcely investigated from the psychotherapy process research perspective. Thus, content and mechanisms related to therapeutic change have been overlooked by research. This study aimed at testing the applicability of the generic change indicators (GCI) model to identify moments of change in the parent during dyadic interventions, as well as to explore, describe and illustrate the therapeutic process through a brief mentalization-informed intervention with a depressive mother and her baby, using video-feedback as its main strategy, which has ample evidence about its effectiveness. We conducted a single case qualitative study using the GCI model. The mother's ongoing change was determined by identifying episodes of change (EC) and moments of change (MC). Each MC was then labelled with one of the 19 GCIs. GCIs were observed from the intervention's start. GCI's hierarchical levels were increasing over the intervention, in association with the video-feedback situation. Our findings suggest that the GCI model is feasible to observe and understand dyadic interventions, contributing to the growing body of evidence supporting psychotherapists' training and supervision.
