Abstract

The SNO+ experiment is located 2 km underground at SNOLAB in Sudbury, Canada. A low background search for neutrinoless double beta ($0\nu\beta\beta$) decay will be conducted using 780 tonnes of liquid scintillator loaded with 3.9 tonnes of natural tellurium, corresponding to 1.3 tonnes of $^{130}$Te. This paper provides a general overview of the SNO+ experiment, including detector design, construction of process plants, commissioning efforts, electronics upgrades, data acquisition systems, and calibration techniques. The SNO+ collaboration is reusing the acrylic vessel, PMT array, and electronics of the SNO detector, having made a number of experimental upgrades and essential adaptations for use with the liquid scintillator. With low backgrounds and a low energy threshold, the SNO+ collaboration will also pursue a rich physics program beyond the search for $0\nu\beta\beta$ decay, including studies of geo- and reactor antineutrinos, supernova and solar neutrinos, and exotic physics such as the search for invisible nucleon decay. Abstract

A liquid scintillator consisting of linear alkylbenzene as the solvent and 2,5-diphenyloxazole as the fluor was developed for the SNO+ experiment. This mixture was chosen as it is compatible with acrylic and has a competitive light yield to pre-existing liquid scintillators while conferring other advantages including longer attenuation lengths, superior safety characteristics, chemical simplicity, ease of handling, and logistical availability. Its properties have been extensively characterized and are presented here. Abstract

Advances in multimodal characterization methods fuel a generation of increasing immense hyper-dimensional datasets. Color mapping is employed for conveying higher dimensional data in two-dimensional (2D) representations for human consumption without relying on multiple projections. How one constructs these color maps, however, critically affects how accurately one perceives data. For simple scalar fields, perceptually uniform color maps and color selection have been shown to improve data readability and interpretation across research fields. Here we review core concepts underlying the design of perceptually uniform color map and extend the concepts from scalar fields to two-dimensional vector fields and three-component composition fields frequently found in materials-chemistry research to enable high-fidelity visualization. We develop the software tools PAPUC and CMPUC to enable researchers to utilize these colorimetry principles and employ perceptually uniform color spaces for rigorously meaningful color mapping of higher dimensional data representations. Last, we demonstrate how these approaches deliver immediate improvements in data readability and interpretation in microscopies and spectroscopies routinely used in discerning materials structure, chemistry, and properties.

Abstract

This paper reports results from a search for nucleon decay through 'invisible' modes, where no visible energy is directly deposited during the decay itself, during the initial water phase of SNO+. However, such decays within the oxygen nucleus would produce an excited daughter that would subsequently de-excite, often emitting detectable gamma rays. A search for such gamma rays yields limits of $2.5 \times 10^{29}$ y at 90% Bayesian credibility level (with a prior uniform in rate) for the partial lifetime of the neutron, and $3.6 \times 10^{29}$ y for the partial lifetime of the proton, the latter a 70% improvement on the previous limit from SNO. We also present partial lifetime limits for invisible dinucleon modes of $1.3\times 10^{28}$ y for $nn$, $2.6\times 10^{28}$ y for $pn$ and $4.7\times 10^{28}$ y for $pp$, an improvement over existing limits by close to three orders of magnitude for the latter two., Comment: 13 pages, 6 figures

Abstract

SNO+ is a large liquid scintillator-based experiment located 2km underground at SNOLAB, Sudbury, Canada. It reuses the Sudbury Neutrino Observatory detector, consisting of a 12m diameter acrylic vessel which will be filled with about 780 tonnes of ultra-pure liquid scintillator. Designed as a multipurpose neutrino experiment, the primary goal of SNO+ is a search for the neutrinoless double-beta decay (0$\nu\beta\beta$) of 130Te. In Phase I, the detector will be loaded with 0.3% natural tellurium, corresponding to nearly 800 kg of 130Te, with an expected effective Majorana neutrino mass sensitivity in the region of 55-133 meV, just above the inverted mass hierarchy. Recently, the possibility of deploying up to ten times more natural tellurium has been investigated, which would enable SNO+ to achieve sensitivity deep into the parameter space for the inverted neutrino mass hierarchy in the future. Additionally, SNO+ aims to measure reactor antineutrino oscillations, low-energy solar neutrinos, and geoneutrinos, to be sensitive to supernova neutrinos, and to search for exotic physics. A first phase with the detector filled with water will begin soon, with the scintillator phase expected to start after a few months of water data taking. Abstract

One of the foremost goals in vaccine development is the design of effective, heat-stable vaccines that simplify the distribution and delivery while conferring high levels of protective immunity. Here, we describe a method for developing a live, oral vaccine that relies on the biofilm-forming properties of the spore-former bacterium Bacillus subtilis. The amyloid protein TasA is an abundant component of the extracellular matrix of the biofilms formed by B. subtilis that can be genetically fused to an antigen of interest. Spores of the recombinant strain are then prepared and applied via the oral route in an animal model. Due to the intrinsic resistance of the spores, they can bypass the stomach barrier, germinate, and subsequently colonize the gut, where they develop into biofilms, expressing the antigen of interest. We describe here the steps necessary to produce spores, immunization, and downstream analysis of the vaccine efficacy. Keywords: Bacillus subtilis; Biofilm; Endospores; Heterologous; Immunization.

Abstract

The process of eukaryotic ribosome assembly stretches across the nucleolus, the nucleoplasm and the cytoplasm, and therefore relies on efficient nucleocytoplasmic transport. In yeast, the import machinery delivers ~140,000 ribosomal proteins every minute to the nucleus for ribosome assembly. At the same time, the export machinery facilitates translocation of ~2000 pre-ribosomal particles every minute through ~200 nuclear pore complexes (NPC) into the cytoplasm. Eukaryotic ribosome assembly also requires >200 conserved assembly factors, which transiently associate with pre-ribosomal particles. Their site(s) of action on maturing pre-ribosomes are beginning to be elucidated. In this chapter, we outline protocols that enable rapid biochemical isolation of pre-ribosomal particles for single particle cryo-electron microscopy (cryo-EM) and in vitro reconstitution of nuclear transport processes. We discuss cell-biological and genetic approaches to investigate how the ribosome assembly and the nucleocytoplasmic transport machineries collaborate to produce functional ribosomes. Abstract

The SNO+ experiment is located 2 km underground at SNOLAB in Sudbury, Canada. A low background search for neutrinoless double beta ($0\nu\beta\beta$) decay will be conducted using 780 tonnes of liquid scintillator loaded with 3.9 tonnes of natural tellurium, corresponding to 1.3 tonnes of $^{130}$Te. This paper provides a general overview of the SNO+ experiment, including detector design, construction of process plants, commissioning efforts, electronics upgrades, data acquisition systems, and calibration techniques. The SNO+ collaboration is reusing the acrylic vessel, PMT array, and electronics of the SNO detector, having made a number of experimental upgrades and essential adaptations for use with the liquid scintillator. With low backgrounds and a low energy threshold, the SNO+ collaboration will also pursue a rich physics program beyond the search for $0\nu\beta\beta$ decay, including studies of geo- and reactor antineutrinos, supernova and solar neutrinos, and exotic physics such as the search for invisible nucleon decay. Abstract

A liquid scintillator consisting of linear alkylbenzene as the solvent and 2,5-diphenyloxazole as the fluor was developed for the SNO+ experiment. This mixture was chosen as it is compatible with acrylic and has a competitive light yield to pre-existing liquid scintillators while conferring other advantages including longer attenuation lengths, superior safety characteristics, chemical simplicity, ease of handling, and logistical availability. Its properties have been extensively characterized and are presented here. Abstract

This paper reports results from a search for nucleon decay through 'invisible' modes, where no visible energy is directly deposited during the decay itself, during the initial water phase of SNO+. However, such decays within the oxygen nucleus would produce an excited daughter that would subsequently de-excite, often emitting detectable gamma rays. A search for such gamma rays yields limits of $2.5 \times 10^{29}$ y at 90% Bayesian credibility level (with a prior uniform in rate) for the partial lifetime of the neutron, and $3.6 \times 10^{29}$ y for the partial lifetime of the proton, the latter a 70% improvement on the previous limit from SNO. Abstract

This paper reports results from a search for nucleon decay through 'invisible' modes, where no visible energy is directly deposited during the decay itself, during the initial water phase of SNO+. However, such decays within the oxygen nucleus would produce an excited daughter that would subsequently de-excite, often emitting detectable gamma rays. A search for such gamma rays yields limits of $2.5 \times 10^{29}$ y at 90% Bayesian credibility level (with a prior uniform in rate) for the partial lifetime of the neutron, and $3.6 \times 10^{29}$ y for the partial lifetime of the proton, the latter a 70% improvement on the previous limit from SNO. Abstract

SNO+ is a large liquid scintillator-based experiment located 2km underground at SNOLAB, Sudbury, Canada. It reuses the Sudbury Neutrino Observatory detector, consisting of a 12m diameter acrylic vessel which will be filled with about 780 tonnes of ultra-pure liquid scintillator. Designed as a multipurpose neutrino experiment, the primary goal of SNO+ is a search for the neutrinoless double-beta decay (0$\nu\beta\beta$) of 130Te. In Phase I, the detector will be loaded with 0.3% natural tellurium, corresponding to nearly 800 kg of 130Te, with an expected effective Majorana neutrino mass sensitivity in the region of 55-133 meV, just above the inverted mass hierarchy. Recently, the possibility of deploying up to ten times more natural tellurium has been investigated, which would enable SNO+ to achieve sensitivity deep into the parameter space for the inverted neutrino mass hierarchy in the future. Additionally, SNO+ aims to measure reactor antineutrino oscillations, low-energy solar neutrinos, and geoneutrinos, to be sensitive to supernova neutrinos, and to search for exotic physics. A first phase with the detector filled with water will begin soon, with the scintillator phase expected to start after a few months of water data taking. Abstract

The new opportunities of modern assays of molecular biology can only be exploited fully if the results can be accurately correlated to the tissue phenotype under investigation. This is a general problem of non-in situ techniques, whereas results from in situ techniques are often difficult to quantitate. The use of bulk tissue, which is not precisely characterized in terms of histology, has long been the basis for molecular analysis. It has, however, become apparent, that this simple approach is not sufficient for a detailed analysis of molecular alterations, which might be restricted to a specific tissue phenotype (e.g., tumor or healthy tissue, stromal or epithelial cells). Microdissection is a method to provide minute amounts of histologically characterized tissues for molecular analysis with non-in situ techniques and has become an indispensable research tool. Here, we describe a very simple technique for microdissection of tissues that can easily and cost effectively be established.

Abstract

The following method describes the identification of candidate immunogenic peptides through their ability to recall an immune T-cell activation from peripheral blood mononuclear cells (PBMCs) of individuals with defined HLA-peptide restrictions that have been previously exposed to the antigen. Isolated PBMCs are plated out at a concentration of 1 x 10(6) cells/ml in a 200 microl medium and incubated overnight to reduce cytokine gene expression due to cell manipulation. After starving, cells are either directly stimulated with individual peptides or not stimulated and incubated from 3 to 12 h according to experimental conditions. Quantitative real-time PCR (qrt-PCR) is performed on reverse-transcribed complementary DNA (cDNA) from total RNA that is isolated from peptide-cultured PBMCs. To perform high quality qrt-PCR, primers and probes are designed to span exon-intron junctions in order to prevent amplification of genomic DNA and to produce amplicons <150 base pairs (bp). Real-time monitoring of fluorescent emission from the cleavage of sequence specific probe by the nuclease activity of Taq polymerase (TaqMan method) defines threshold cycles during exponential phases of amplification. Standard curves of copy numbers of the gene of interest are normalized using as reference copy numbers of control genes.

Abstract

For the determination of the chirality of amino acids and other compounds containing primary amino groups, derivatization with the chiral Marfey’s reagent has kept its place as a complementary method beside direct chromatographic analysis with chiral stationary phases. Here, we describe the use of Marfey’s reagent for common amino acid analysis. The simplicity of pre-column derivatization of amino acids with this reagent, the ease of separation of the derivatized amino acids with non-dedicated reverse-phase highperformance liquid chromatography (RP-HPLC) equipment and the high sensitivity of their chromatographic detection at 340 nm make the described method an attractive alternative to the dedicated amino acid analyzer.

Abstract

Drosophila melanogaster has long been a prime model organism for developmental biologists. During their work, they have established a large collection of techniques and reagents. This in turn has made fruit flies an attractive system for many other biomedical researchers who have otherwise no background in fly biology. This review intends to help Drosophila neophytes in setting up a fly lab. It briefly introduces the biological properties of fruit flies, describes the minimal equipment required for working with flies, and offers some basic advice for maintaining fly lines and setting up and analyzing experiments.

Abstract

SNO+ is a large liquid scintillator-based experiment located 2km underground at SNOLAB, Sudbury, Canada. It reuses the Sudbury Neutrino Observatory detector, consisting of a 12m diameter acrylic vessel which will be filled with about 780 tonnes of ultra-pure liquid scintillator. Designed as a multipurpose neutrino experiment, the primary goal of SNO+ is a search for the neutrinoless double-beta decay (0$\nu\beta\beta$) of 130Te. In Phase I, the detector will be loaded with 0.3% natural tellurium, corresponding to nearly 800 kg of 130Te, with an expected effective Majorana neutrino mass sensitivity in the region of 55-133 meV, just above the inverted mass hierarchy. Recently, the possibility of deploying up to ten times more natural tellurium has been investigated, which would enable SNO+ to achieve sensitivity deep into the parameter space for the inverted neutrino mass hierarchy in the future. Additionally, SNO+ aims to measure reactor antineutrino oscillations, low-energy solar neutrinos, and geoneutrinos, to be sensitive to supernova neutrinos, and to search for exotic physics. A first phase with the detector filled with water will begin soon, with the scintillator phase expected to start after a few months of water data taking. 42. Inorganic and black carbon aerosols in the Los Angeles Basin during CalNex

Abstract

We evaluate predictions from the Community Multiscale Air Quality (CMAQ version 4.7.1) model against a suite of airborne and ground-based meteorological measurements, gas- and aerosol-phase inorganic measurements, and black carbon (BC) measurements over Southern California during the CalNex field campaign in May/June 2010. Ground-based measurements are from the CalNex Pasadena ground site, and airborne measurements took place onboard the Center for Interdisciplinary Remotely-Piloted Aircraft Studies (CIRPAS) Navy Twin Otter and the NOAA WP-3D aircraft. BC predictions are in general agreement with observations at the Pasadena ground site and onboard the WP-3D, but are consistently overpredicted when compared to Twin Otter measurements. Adjustments to predicted inorganic mass concentrations, based on predicted aerosol size distributions and the AMS transmission efficiency, are shown to be significant. Owing to recent shipping emission reductions, the dominant source of sulfate in the L.A. Basin may now be long-range transport. Sensitivity studies suggest that severely underestimated ammonia emissions, and not the exclusion of crustal species (Ca^(2 +), K^+, and Mg^(2 +)), are the single largest contributor to measurement/model disagreement in the eastern part of the L.A. Basin. Despite overstated NO_x emissions, total nitrate concentrations are underpredicted, which suggests a missing source of HNO_3 and/or overprediction of deposition rates. Adding gas-phase NH_3 measurements and size-resolved measurements, up to 10 μm, of nitrate and various cations (e.g. Na^+, Ca^(2 +), K^+) to routine monitoring stations in the L.A. Basin would greatly facilitate interpreting day-to-day fluctuations in fine and coarse inorganic aerosol.

45. Clinical, genetic, and functional characterization of four patients carrying partial loss-of-function mutations in the steroidogenic acute regulatory protein (StAR)

Abstract

Context: Nonclassic congenital lipoid adrenal hyperplasia (lipoid CAH) is a recently recognized disorder caused by mutations in the steroidogenic acute regulatory protein (StAR) that retain partial function. Affected individuals can present with a phenotype of late onset adrenal insufficiency with only mild or minimally disordered sexual development. Objectives: The aim was to delineate the clinical spectrum of StAR mutations and correlate phenotype with StAR activity. Patients: Four patients had nonclassic/atypical lipoid CAH. Adrenal insufficiency was manifested at birth in two patients and at 11 months and 4 yr in the other two. Three were 46,XY with underdeveloped genitalia. Methods: The StAR gene was sequenced, mutations were recreated in expression vectors, and StAR activity was measured as pregnenolone production in COS-1 cells cotransfected with the cholesterol side-chain cleavage system. StAR mutants were expressed as N-62 StAR in bacteria, and purified proteins were tested for activity with isolated steroidogenic mitochondria and for cholesterol-binding capacity. Results: DNA sequencing identified mutations on all alleles. Missense mutations were R188C, G221D, L260P, and F267S; we also tested R192C described by others. The respective activities of R188C, R192C, G221D, L260P, and F267S were 8.0, 39.4, 2.4, 3.1, and 6.1% of wild-type in transfected cells, and 12.8, 54.8, 6.3, 1.8, and 9.5% with isolated mitochondria. Cholesterol binding capacities of R188C, R192C, G221D, L260P, and F267S were 6.7, 55.3, 10.2, 4.6, and 20.9%. These data are correlated to the three-dimensional structure of StAR. Conclusions: There is a broad clinical spectrum of StAR mutations; StAR activities in vitro correlate well with clinical phenotypes.

Abstract

N-arachidonoyl ethanolamine (AEA) and 2-arachidonoyl glycerol (2-AG) are endogenous cannabinoids binding to the cannabinoid receptors CB1 and CB2 to modulate neuronal excitability and synaptic transmission in primary afferent neurons. To investigate the compartmentalization of the machinery for AEA and 2-AG signalling, we studied their partitioning into lipid raft fractions isolated from a dorsal root ganglion X neuroblastoma cell line (F-11).F-11 cells were homogenized and fractionated using a detergent-free OptiPrep density gradient. All lipids were partially purified from methanolic extracts of the fractions on solid phase cartridges and quantified using liquid chromatography tandem mass spectrometry (LC/MS/MS). Protein distribution was determined by Western blotting.Under basal conditions, the endogenous cannabinoid AEA was present in both lipid raft and specific non-lipid raft fractions as was one of its biosynthetic enzymes, NAPE-PLD. The 2-AG precursor 1-stearoyl-2-arachidonoyl-sn-glycerol (DAG), diacylglycerol lipase alpha (DAGLalpha), which cleaves DAG to form 2-AG, and 2-AG were all co-localized with lipid raft markers. CB1 receptors, previously reported to partition into lipid raft fractions, were not detected in F-11 membranes, but CB2 receptors were detected at high levels and partitioned into non-lipid raft fractions.The biochemical machinery for the production of 2-AG via the putative diacylglycerol pathway is localized within lipid rafts, suggesting that 2-AG synthesis via DAG occurs within these microdomains. The observed co-localization of AEA, 2-AG, and their synthetic enzymes with the reported localization of CB1 raises the possibility of intrinsic-autocrine signalling within lipid raft domains and/or retrograde-paracrine signalling.
