Porcine epidemic diarrhea virus (PEDV) is a significant pathogen affecting swine, causing severe economic losses worldwide. This study explores the regulatory role of miRNA-328-3p to ZO-1 expression and its impact on PEDV proliferation via the PLC-β1-PKC pathway in IPEC-J2 cells. We found that miRNA-328-3p can target ZO-1, influencing its expression and subsequently affecting the integrity of tight junctions in the cells. Overexpression of PLC-β1, combined with miRNA-328-3p silencing, enhanced ZO-1 expression, while PLC-β1 knockdown combined with miRNA-328-3p overexpression inhibited ZO-1 expression. Furthermore, PLC-β1 overexpression increased both viral genome expression and PEDV titers, whereas its silencing had the opposite effect. Notably, our data indicated a negative correlation between PLC-β1 and PKC expression, and PKC silencing attenuated the upregulatory effect of PLC-β1 on ZO-1. These findings suggest that PLC-β1 modulates ZO-1 expression through the PKC pathway, providing new insights into the molecular mechanisms of PEDV infection and potential therapeutic targets. Background: Regional lifestyles influence allergic disease risks, with dietary factors being crucial. However, observational studies on micro- and macronutrient impacts are inconsistent. Methods: We used a two-sample Mendelian randomization (MR) design with the inverse variance weighted (IVW) method and sensitivity analyses to evaluate the effects of various micronutrients (e.g. zinc, selenium, phosphorus) and macronutrient intake on allergic diseases. Results: Zinc was inversely associated with allergic asthma (OR = 0.856, p = 0.014) and atopic dermatitis (OR = 0.918, p = 0.039). Selenium was inversely associated with allergic rhinitis (OR = 0.863, p = 0.037). Phosphorus was linked to an increased risk of allergic urticaria (OR = 4.396, p = 0.0025). Other nutrients showed no significant associations. Conclusion: Certain micronutrients, like zinc, selenium, and phosphorus, may play significant roles in allergic disease development, warranting further research to confirm these findings. Objective To investigate the safety of the ultra-early (0~6 h) activity program for patients after minimally invasive radical treatment of cervical cancer. Methods A non-randomized concurrent controlled trial was conducted on 118 patients undergoing minimally invasive radical surgery for cervical cancer in our department from June to December 2019. According to patient's wish, they were divided into the ultra-early (0~6 h) postoperative activity group (n=58) and the conventional activity group (n=60). The patients in both groups received the perioperative program of rapid gynecological rehabilitation. After the operation, the conventional activity group adopted the conventional nursing plan, and the ultra-early activity group adopted the ultra-early postoperative activity plan according to the characteristics of patients with one-to-one guidance and assistance by professional health education nurses. If a patient suffered from conscious weakness, palpitation, dizziness, pale or cold sweats, oxygen saturation < 94%, or heart rate reaching the target heart rate of Karvonen formula, the activity program would be stopped immediately when any of them presented. Results The time to get out of bed for the first time and the activity scores were better in the ultra-early activity group than the conventional activity group (P=0.011). There were no statistically significant differences in SpO2, pulse, pain scores, and incidence of complications between the 2 groups before and after activities. The ultra-early activity group had obviously better average activity in 3 d post-operatively during hospitalization (P=0.000), shorter times to first postoperative exhaust (P=0.020) and to first oral feeding (P=0.010), and shorter length of hospital stay (P=0.010) when compared with the conventional activity group. Conclusion All patients in the ultra-early activity group are able to get out of bed within 24 h post-operatively, and attain better activity level in 3 d after operation than those of the conventional activity group. Ultra-early postoperative activity program can effectively improve the early out-of-bed activity rate after cervical cancer surgery and have no effect on the occurrence of postoperative complications.

Objective To observe the gastrointestinal function recovery in the patients with different feeding time after gynecological laparoscopic surgery. Methods A total of 132 patients undergoing laparoscopic surgery in our department from December 2020 to April 2021 were enrolled in this study. Five of them were excluded and 127 cases were finally enrolled, including 64 cases in the experimental group (taking food immediately after returning to the ward after surgery) and 63 cases in the control group (taking food 6 h after returning). The recovery of gastrointestinal function, the incidences of abdominal distention, nausea and vomiting, and the average length of postoperative hospital stay were recorded and compared between the 2 groups. Results The experimental group had significantly shorter time of first postoperative bowl sound (7.44±3.99 vs 10.48±4.62), first exhaust (11.50±6.75 vs 16.30±7.44 h), and first defecation (21.38±9.22 vs 28.68±12.99 h), and shorter average length of postoperative hospital stay (4.28±1.55 vs 5.59±4.07 d) when compared with the control group (P < 0.05). The incidences of abdominal distention, nausea and vomiting in postoperative patients were obviously lower in the experimental group than the control group (25.0% vs 46.0%, 14.1% vs 34.9%, P < 0.05). Early oral feeding after laparoscopic operation improved the recovery of intestinal function and shorten the postoperative hospital stay. Conclusion Postoperative early food-taking program is safe for patients undergoing gynecological laparoscopic surgery. It is beneficial to improve the recovery of gastrointestinal function, reduce the incidence of postoperative abdominal distention, nausea and vomiting, and promote early discharge of patients from hospital.

Models used to study the fracture process of concrete are often considered 2D, ignoring the influence of specimen width. However, during the fracture process in pre-cracked concrete beams, the crack length varies along the thickness direction, especially in reinforced concrete. To study the influence of specimen width on reinforced concrete fracture behavior, a 3D numerical method was used to simulate the crack propagation processes of lightly reinforced concrete beams based on Fracture Mechanics. Nonlinear spring elements with different stress-displacement constitutive laws were employed to characterize the softening behavior of concrete and the bond-slip behavior between the steel bars and concrete, respectively. It is assumed that the crack begins to propagate when the maximum stress intensity factor at the crack tip along the beam width reaches the initial fracture toughness of concrete. To verify the validity of the proposed method, the completed crack propagation processes of lightly reinforced concrete three-point bending notched beams were simulated, and the calculated load-crack mouth opening displacement curves showed a reasonable agreement with the experimental data. Moreover, the impact of the 2D reinforced concrete beam model on the crack propagation process was analyzed. The results indicate that at the initial loading stage, the external load P obtained from the 2D model is significantly larger than the result from the presented 3D model. [ABSTRACT FROM AUTHOR]
