Traumatic brain injury (TBI) represents a main public health concern during the past decade, attracting considerable interest because of its rising prevalence, wide-ranging risk factors and lifelong familial and societal influence. SUMO2 can conjugate to substrates upon various cellular stresses. Nevertheless, whether and how SUMO2-specific proteases partake in TBI is less understood. The aim of this study is to dissect the effects of SUMO-specific peptidase 5 (SENP5) on accentuating TBI in rats in an effort to unveil its underlying mechanism. SENP5 is overexpressed in hippocampal tissues of TBI rats, and inhibition of SENP5 reduces neurological function scores, decreases brain water content, inhibits apoptosis in hippocampal tissues, and attenuates brain injury caused in rats. Moreover, SENP5 inhibits the SUMOylation level of E2F transcription factor 1 (E2F1) and increases the protein expression of E2F1. Silencing of E2F1 blocks the p53 signaling pathway. Overexpression of E2F1 partially reverses the protective effect of sh-SENP5 on TBI in rats. These findings reveal an essential role of SENP5 and the SUMOylation status of E2F1 in the TBI development.

Pulmonary atresia (PA) is a severe cyanotic congenital heart disease. Although some genetic mutations have been described to be associated with PA, the knowledge of pathogenesis is insufficient. The aim of this research was to use whole-exome sequencing (WES) to determine novel rare genetic variants in PA patients. We performed WES in 33 patients (27 patient–parent trios and 6 single probands) and 300 healthy control individuals. By applying an enhanced analytical framework to incorporate de novo and case–control rare variation, we identified 176 risk genes (100 de novo variants and 87 rare variants). Protein‒protein interaction (PPI) analysis and Genotype-Tissue Expression analysis revealed that 35 putative candidate genes had PPIs with known PA genes with high expression in the human heart. Expression quantitative trait loci analysis revealed that 27 genes that were identified as novel PA genes that could be affected by the surrounding single nucleotide polymorphism were screened. Furthermore, we screened rare damaging variants with a threshold of minor allele frequency at 0.5% in the ExAC_EAS and GnomAD_exome_EAS databases, and the deleteriousness was predicted by bioinformatics tools. For the first time, 18 rare variants in 11 new candidate genes have been identified that may play a role in the pathogenesis of PA. Our research provides new insights into the pathogenesis of PA and helps to identify the critical genes for PA.

With the transformation of urban knowledge economy, cities pay more and more attention on efforts to attract knowledge talents, including young intellectuals. Taking college students in Guangzhou as an example, this research adopts a combination of qualitative and quantitative methods to explore the mobility of young intellectual migrants, and the characteristics and influencing factors of their employment city selection. The research has found the following. First, young intellectual migrants generally tend to choose cities where their undergraduate or postgraduate studies are located and cities with higher grades than their household registration and place of study (i.e., first-tier and new first-tier cities), which reflects their willingness and motivation for upward spatial and social mobility. Second, their employment mobility is a process of the interaction between urban and individual factors. Its influencing factors mainly include urban environment, economy, policy and culture, as well as the type of household registration and the city level of household registration, among others. Third, urban economy development (job opportunities) is the most important factor for attracting young intellectual migrants, along with employment opportunities, wage levels, and others. At the same time, amenities such as cultural facilities and educational institutions, as well as physical features such as climate and natural environment and other living comforts (living opportunities) play an increasingly important role in the employment mobility of young intellectual migrants. Therefore, urban environmental construction needs to pay attention to public cultural facilities, social comfort, transportation convenience, health service facilities, and urban natural and living environment. The urban policies of high-level cities are more attractive to young intellectual migrants, including talent introduction, housing subsidies and urban development policies. Fourth, in terms of personal factors, the type of household registration and the city level of the source of youth intellectual migration have significant impact on their employment mobility decisions. There are differences in the mobility costs between the different types and city level of household registration, which further affects their employment mobility behavior. A detailed exploration of the employment mobility law of young intellectual migrants and its influencing factors is necessary to understand and construct a knowledge-based talent mobility theory based on Chinese context in the new era. It is also the key to talent policy formulation and the realization of the value of knowledge talents.

Simple Summary: The erucic acid (EA) present in rapeseed oil is known to inhibit growth and lipid utilization in terrestrial animals, but its potential anti-nutritional effects on fish have remained unclear due to a lack of information. Therefore, we conducted this study to investigate the effects of varying levels of EA on growth, health, and lipid metabolism in black carp, Mylopharyngodon piceus. The results demonstrated that high-dose EA inhibited growth, induced lipid accumulation, reduced antioxidant and immune capabilities, and led to oxidative damage by suppressing lipid catabolism and increasing lipogenesis. This study fills a gap in our understanding of the physiological and anti-nutritional effects of EA on fish and provides valuable theoretical insights for enhancing the utilization of rapeseed oil in cultured carnivorous fish. This study investigated the effects of dietary erucic acid (EA) on growth, lipid accumulation, antioxidant and immune abilities, and lipid metabolism in black carp fed six diets containing varying levels of EA (0.00%, 0.44%, 0.81%, 1.83%, 2.74%, and 3.49%), for 8 weeks. Results showed that fish fed the 3.49% EA diet exhibited lower weight gain, compared to those fed the 0.81% EA diet. In a dose-dependent manner, the serum triglycerides and total cholesterol were significantly elevated in the EA groups. The 1.83%, 2.74%, and 3.49% levels of EA increased alanine aminotransferase and aspartate aminotransferase activities, as well as decreased acid phosphatase and alkaline phosphatase values compared to the EA-deficient group. The hepatic catalase activity and transcriptional level were notably reduced, accompanied by increased hydrogen peroxide contents in the EA groups. Furthermore, dietary EA primarily increased the C22:1n-9 and C20:1n-9 levels, while decreasing the C18:0 and C18:1n-9 contents. In the EA groups, expressions of genes, including hsl, cpt1a, cpt1b, and ppara were downregulated, whereas the fas and gpat expressions were enhanced. Additionally, dietary EA elevated the mRNA level of il-1β and reduced the expression of il-10. Collectively, high levels of EA (2.74% and 3.49%) induced lipid accumulation, reduced antioxidative and immune abilities in black carp by inhibiting lipid catabolism and increasing lipogenesis. Aims: To investigate the diagnostic and predictive role of 18F‐FDG PET/CT in patients with autoimmune encephalitis (AE) as a whole group. Methods: Thrty‐five patients (20 females and 15 males) with AE were recruited. A voxel‐to‐voxel semi‐quantitative analysis based on SPM12 was used to analyze 18F‐FDG PET/CT imaging data compared to healthy controls. Further comparison was made in different prognostic groups categorized by modified Rankin Scale (mRS). Results: In total, 24 patients (68.6%) were tested positive neuronal antibodies in serum and/or CSF. Psychiatric symptoms and seizure attacks were major clinical symptoms. In the acute stage, 13 patients (37.1%) demonstrated abnormal brain MRI results, while 33 (94.3%) presented abnormal metabolism patterns. 18F‐FDG PET/CT was more sensitive than MRI (p < 0.05). Patients with AE mainly presented mixed metabolism patterns compared to the matched controls, demonstrating hypermetabolism mainly in the cerebellum, BG, MTL, brainstem, insula, middle frontal gyrus, and relatively hypometabolism in the frontal cortex, occipital cortex, temporal gyrus, right parietal gyrus, left cingulate gyrus (p < 0.05, FWE corrected). After a median follow‐up of 26 months, the multivariable analysis identified a decreased level of consciousness as an independent risk factor associated with poor outcome of AE (HR = 3.591, p = 0.016). Meanwhile, decreased metabolism of right superior frontal gyrus along with increased metabolism of the middle and upper brainstem was more evident in patients with poor outcome (p < 0.001, uncorrected). Conclusion: 18F‐FDG PET/CT was more sensitive than MRI to detect neuroimaging abnormalities of AE. A mixed metabolic pattern, characterized by large areas of cortical hypometabolism with focal hypermetabolism was a general metabolic pattern. To delineate the efficacy and safety profile of hemodiafiltration with endogenous reinfusion (HFR) for uremic toxin removal in patients undergoing maintenance hemodialysis (MHD). Patients who have been on MHD for a period of at least 3 months were enrolled. Each subject underwent one HFR and one hemodiafiltration (HDF) treatment. Blood samples were collected before and after a single HFR or HDF treatment to test uremic toxin levels and to calculate clearance rate. The primary efficacy endpoint was to compare uremic toxin levels of indoxyl sulfate (IS), λ-free light chains (λFLC), and β2-microglobulin (β2-MG) before and after HFR treatment. Secondary efficacy endpoints was to compare the levels of urea, interleukin-6 (IL-6), P-cresol, chitinase-3-like protein 1 (YKL-40), leptin (LEP), hippuric acid (HPA), trimethylamine N-oxide (TMAO), asymmetric dimethylarginine (ADMA), tumor necrosis factor-α (TNF-α), fibroblast growth factor 23 (FGF23) before and after HFR treatment. The study also undertook a comparative analysis of uremic toxin clearance between a single HFR and HDF treatment. Meanwhile, the lever of serum albumin and branched-chain amino acids before and after a single HFR or HDF treatment were compared. In terms of safety, the study was meticulous in recording vital signs and the incidence of adverse events throughout its duration. The study enrolled 20 patients. After a single HFR treatment, levels of IS, λFLC, β2-MG, IL-6, P-cresol, YKL-40, LEP, HPA, TMAO, ADMA, TNF-α, and FGF23 significantly decreased (p < 0.001 for all). The clearance rates of λFLC, β2-MG, IL-6, LEP, and TNF-α were significantly higher in HFR compared to HDF (p values: 0.036, 0.042, 0.041, 0.019, and 0.036, respectively). Compared with pre-HFR and post-HFR treatment, levels of serum albumin, valine, and isoleucine showed no significant difference (p > 0.05), while post-HDF, levels of serum albumin significantly decreased (p = 0.000). HFR treatment effectively eliminates uremic toxins from the bloodstream of patients undergoing MHD, especially protein-bound toxins and large middle-molecule toxins. Simple Summary: Carbohydrate utilization is closely associated with level, chemical structure or processing method. Selecting an optimal carbohydrate source needs to take into consideration all these factors. This study investigated the effects of different carbohydrate sources, including monosaccharide and gelatinized or ungelatinized polysaccharide starch, on growth performance, hepatic glucose metabolism, antioxidant capacity, and immunity in juvenile largemouth bass. The results showed that dietary starch exhibited better growth performance independent of gelatinization. Ungelatinized potato starch was proved to be the more effective carbohydrate source for increasing hepatic glycolysis and ungelatinized tapioca starch better improved antioxidant and immune capacities. This study provides new information for selecting carbohydrate sources in artificial feed and helps improve fish health in farmed largemouth bass. This study investigated the effects of glucose (GLU), tapioca starch (TS), gelatinized tapioca starch (GTS), potato starch (PS) and gelatinized potato starch (GPS) on growth and physiological responses in juvenile largemouth bass Micropterus salmoides. After 8 weeks, fish fed with starch diets had better weight gain and growth rates. Counts of red blood cells and monocytes were increased in the PS and GPS groups, compared to GLU group. Contents of serum triglyceride and total cholesterol were markedly elevated in the TS, PS and GPS groups. There were lower levels of serum glucose, insulin and cholecystokinin, and higher agouti-related peptide contents in the PS group compared to GLU group. PS and GPS could enhance glycolysis and TCA cycle by increasing their enzyme activities and transcriptional levels. Additionally, starch sources markedly heightened mRNA levels of key genes involved in the respiratory electron transport chain. Additionally, elevated mRNA levels of key antioxidant genes were shown in the TS and GTS groups. Moreover, TS and PS could promote immunity by upregulating transcriptional levels of the complement system, lysozyme and hepcidin. Taken together, starch exhibited better growth via increasing glycolysis and TCA cycle compared with GLU, and PS could improve antioxidant and immune capacities in largemouth bass. To explore how endophytic microbial compositions in amaranth roots are influenced by various fertilization methods and to determine whether these microbes are associated with amaranthin formation, we conducted an analysis of the endophytic microbial community structure. The roots of amaranth plants subjected to different fertilization treatments—conventional fertilization without potassium (NP), conventional fertilization without phosphorus (NK), conventional fertilization without nitrogen (PK), and balanced fertilization (NPK)—were examined. The results showed that the proportions of Streptomyces, Actinospica, and Burkholderia-Caballeronia-Paraburkholderia in the amaranth roots under the balanced fertilization (NPK) treatment were all greater than those in the amaranth roots under the nitrogen (PK), phosphorus (NK), and potassium (NP) deficiency fertilization treatments. In contrast, the proportions of Phenylobacterium, Acrocalymma, Neocosmospora, Fusarium, Acidovorax, Gibellulopsis, Cladosporium, Dactylonectria, and Gibberella in the amaranth roots under the nutrient deficiency fertilization (NP, NK, and PK) treatments were higher than those in the amaranth roots under the balanced fertilization treatment. Additionally, a significantly positive correlation was found between Streptomyces and the amaranthin content. Furthermore, Acrocalymma, Neocosmospora, and Fusarium exhibited significantly negative correlations with the amaranthin content. The above results suggested that endophytes could easily colonize in amaranth roots as beneficial microorganisms under balanced fertilization conditions. In other words, the balanced fertilization (N, P and K fertilizers are 188.0, 53.0 and 50.0 kg·hm−2, respectively) could recruit more beneficial endogenous microorganisms in amaranth roots for improving their growth and quality.
