The investigation of triggers causing the onset and intensification of Northern Hemisphere Glaciation (NHG) during the late Pliocene is essential for understanding the global climate system, with important implications for projecting future climate changes. Despite their critical roles in the global climate system, influences of land-ocean interactions on high-latitude ice sheets remain largely unexplored. Here, we present a high-resolution Asian dust record from Ocean Drilling Program Site 1208 in the North Pacific, which lies along the main route of the westerlies. Our data indicate that atmosphere-land-ocean interactions affected aeolian dust emissions through modulating moisture and vegetation in dust source regions, highlighting a critical role of terrestrial systems in initiating the NHG as early as 3.6 Myr ago. Combined with additional multi-proxy and model results, we further show that westerly wind strength was enhanced, mainly at low-to-middle tropospheric levels, during major glacial events at about 3.3 and 2.7 Myr ago. We suggest that coupled responses of Earths surface dynamics and atmospheric circulation in the Plio-Pleistocene likely involved feedbacks related to changes in paleogeography, ocean circulation, and global climate.

Pulmonary arterial hypertension (PAH) is a devastating and progressive disease with limited treatment options. Endothelial dysfunction plays a central role in the development and progression of PAH, yet the underlying mechanisms are incompletely understood. The endosome-lysosome system is important to maintain cellular health, and the small GTPase RAB7 regulates many functions of this system. Here, we explored the role of RAB7 in endothelial cell (EC) function and lung vascular homeostasis. We found reduced expression of RAB7 in ECs from patients with PAH. Endothelial haploinsufficiency of RAB7 caused spontaneous pulmonary hypertension (PH) in mice. Silencing of RAB7 in ECs induced broad changes in gene expression revealed via RNA-Seq, and RAB7-silenced ECs showed impaired angiogenesis and expansion of a senescent cell fraction, combined with impaired endolysosomal trafficking and degradation, suggesting inhibition of autophagy at the predegradation level. Furthermore, mitochondrial membrane potential and oxidative phosphorylation were decreased, and glycolysis was enhanced. Treatment with the RAB7 activator ML-098 reduced established PH in rats with chronic hypoxia/SU5416. In conclusion, we demonstrate for the first time to our knowledge the fundamental impairment of EC function by loss of RAB7, causing PH, and show RAB7 activation to be a potential therapeutic strategy in a preclinical model of PH.

Abstract The dose–response of intravenous lidocaine in preventing postoperative vomiting (POV) in children remains unclear. This study investigated whether intravenous lidocaine dose-dependently decreased POV risk within 24 h postoperatively in children undergoing tonsillectomy (with or without adenoidectomy) without severe complications. Patients aged 3–12 years (American Society of Anesthesiologists grade I–II) scheduled for elective tonsillectomy (with or without adenoidectomy) were enroled from December 2021 to March 2022. They were randomly grouped according to the lidocaine dose (A [0 mg kg−1], B [1 mg kg−1], C [1.5 mg kg−1], and D [2 mg kg−1]) and were administered the same induction protocol (sufentanil, propofol, and suxamethonium chloride). Anaesthesia was maintained with sevoflurane. The incidence of POV within 24 h postoperatively was 46, 40, 36, and 20% in groups A, B, C, and D, respectively, with significant differences between groups D and A. Postoperative analgesic rescues in groups A, B, C, and D were 62, 36, 34, and 16%, respectively, with significant differences between groups D and B, C and A, and D and A. No severe adverse events were reported. Intravenous lidocaine has a dose-dependent effect on reducing the risk of POV in children undergoing tonsillectomy (with or without adenoidectomy) without serious adverse events. Abstract Background Human aging and white blood cell (WBC) count are complex traits influenced by multiple genetic factors. Predictors of chronological age have been developed using epigenetic clocks. However, the bidirectional causal effects between epigenetic clocks and WBC count have not been fully examined. Methods This study employed Mendelian randomization (MR) to analyze summary statistics from four epigenetic clocks involving 34,710 participants, alongside data from the Blood Cell Consortium encompassing 563,946 individuals. We primarily explored bidirectional causal relationships using the random-effects inverse-variance weighted method, supplemented by additional MR methods for comprehensive analysis. Additionally, multivariate MR was applied to investigate independent effects of WBC count on epigenetic age acceleration. Abstract To date, endotracheal tube (ETT) remains the mainstream for preterm infants receiving general anesthesia. We aim to compare the perioperative respiratory adverse events between using supraglottic airway device (SAD) and ETT in former preterm infants receiving general anesthesia. Former preterm infants below 52 weeks of postmenstrual age scheduled for herniorrhaphy were randomized to receive SAD or ETT for general anesthesia. Infants with severe congenital cardiopulmonary disease, prolonged oxygen or mechanical ventilation dependence, and recent respiratory tract infection were excluded. Muscle relaxant agents and opioids were avoided in this study. 40 infants were assigned into SAD or ETT groups. Infants in the SAD group had a much lower rate of intraoperative desaturation than those in the ETT group (21.1% vs. 73.7%, p = 0.003). Incidences of other intraoperative and postoperative 24-h respiratory adverse events were similar between groups, including laryngospasm/bronchospasm, cough and stridor during anesthesia, and postoperative apnea, bradycardia, and supplemental oxygen use. All participants were extubated successfully in the operation room. SAD is recommended in former preterm infants receiving general anesthesia for herniorrhaphy in their early infancy as it much decreases the incidence of intraoperative desaturation compared to ETT.

Abstract Background The relationship between surgical sperm retrieval of different etiologies and clinical pregnancy is unclear. We aimed to develop a robust and interpretable machine learning (ML) model for predicting clinical pregnancy using the SHapley Additive exPlanation (SHAP) association of surgical sperm retrieval from testes of different etiologies. Methods A total of 345 infertile couples who underwent intracytoplasmic sperm injection (ICSI) treatment with surgical sperm retrieval due to different etiologies from February 2020 to March 2023 at the reproductive center were retrospectively analyzed. The six machine learning (ML) models were used to predict the clinical pregnancy of ICSI. After evaluating the performance characteristics of the six ML models, the Extreme Gradient Boosting model (XGBoost) was selected as the best model, and SHAP was utilized to interpret the XGBoost model for predicting clinical pregnancies and to reveal the decision-making process of the model. Results Combining the area under the receiver operating characteristic curve (AUROC), accuracy, precision, recall, F1 score, brier score, and the area under the precision-recall (P-R) curve (AP), the XGBoost model has the best performance (AUROC: 0.858, 95% confidence interval (CI): 0.778–0.936, accuracy: 79.71%, brier score: 0.151). The global summary plot of SHAP values shows that the female age is the most important feature influencing the model output. The SHAP plot showed that younger age in females, bigger testicular volume (TV), non-tobacco use, higher anti-müllerian hormone (AMH), lower follicle-stimulating hormone (FSH) in females, lower FSH in males, the temporary ejaculatory disorders (TED) group, and not the non-obstructive azoospermia (NOA) group all resulted in an increased probability of clinical pregnancy. Conclusions The XGBoost model predicts clinical pregnancies associated with testicular sperm retrieval of different etiologies with high accuracy, reliability, and robustness. It can provide clinical counseling decisions for patients with surgical sperm retrieval of various etiologies.

To understand the explosion characteristics of methane-lignite coal dust mixtures and the spatiotemporal evolution of the early flame flow field, explosion pressure, OH* emission spectra, and early flame schlieren images were captured in a 60L constant volume combustion bomb. Schlieren image velocimetry was used to obtain the instantaneous velocity distribution of the methane-coal dust flame front. The results indicate that adding coal dust causes a nonlinear increase in explosion pressure, particularly at low methane concentrations. At a 5 % methane concentration, adding coal dust can increase the maximum explosion pressure by up to 39 % (190 kPa). The trend of the OH* emission spectra corresponds with the maximum explosion pressure. A slight increase in coal dust significantly enhances the OH* emission spectra, especially at lower concentrations, potentially increasing it by nearly five times. However, at higher coal dust concentrations, the OH* emission spectra are significantly reduced. Additionally, within the considered concentration range (≤58.14 g/m³), larger turbulent integral length scales in initial conditions consistently result in the fastest flame acceleration. These findings provide experimental evidence for further exploration of the explosion mechanisms of methane-coal dust mixtures and the development of chemical reaction kinetic models.

Air pollution stands as the single largest environmental risk to human health globally, and negative air ions (NAIs) serve as essential indicators for measuring regional air quality. They play a crucial role in alleviating the pressure and risks posed by air pollution. NAIs are notably abundant in forests, where the photosynthesis of vegetation serves as a significant factor influencing their presence. However, a dearth of quantitative research exists regarding the relationship between vegetation photosynthesis and NAIs. Current literature only offers qualitative insights, suggesting that vegetation photosynthesis may foster NAIs production. The mechanisms through which the photosynthetic process influences NAIs and the potential contribution of forest vegetation to NAIs remain unclear, lacking quantitative investigation. In this study, Quercus variabilis, a typical tree species in the warm temperate zone, was chosen as the research subject. We examined the dynamic changes in NAIs and net ecosystem productivity (NEP) from 2019 to 2021. The results demonstrated a strong correlation between NAIs and NEP at the ecosystem scale during the primary vegetation growing season. NEP effectively tracked daily changes in NAIs and exhibited a good fit. Notably, the scatter plots of NAIs and NEP exhibited clear stratification under varying photosynthetically active radiation (PAR) intensities. The correlation between NAIs and NEP was significantly higher under high PAR conditions compared to low PAR conditions, with R2 values of 0.458 and 0.367, respectively. Consequently, NEP emerges as a superior indicator for tracking NAIs variation characteristics at the ecosystem scale compared to PAR. This finding offers a rapid and accurate evaluation method for discerning the spatial and temporal distribution patterns of NAIs within forest ecosystems. Such insights provide a theoretical foundation for further assessing the potential contribution of forest vegetation to NAIs.

Among the growing floating population in China, many older people—who migrate along with their adult children to provide family support—have formed a specific social group: lao-piao-zu . This group of older migrants is a population with particular experiences and challenges, which merits more scholarly attention to their well-being. The study first presents a three-dimensional conceptual framework for understanding multimodal mobile phone communication among older migrants. Guided by this framework and based on a face-to-face survey ( N = 376) conducted in China’s first-tier city Shenzhen, the study specifically examined how various forms of mobile communication could affect psychological well-being (PWB) among these older migrants. The results showed that mobile phone use could increase multimodal communication activities. However, only communication with the hometown could reduce loneliness and promote PWB. We argue that mobile communication has limited effects on older migrants’ PWB. It is a promising channel for maintaining remote ties; however, it should be considered more as a form of compensation than as the optimal option for older people who are away from their hometowns.

IntroductionPerturbation walking (PW) has been shown to improve gait, however its effect on the cortical control of gait might provide insights on neural mechanisms underlying falls in adults with osteoarthritis. The objective of this study is to investigate the effect of PW on prefrontal cortical (PFC) activation in older women with (OA) and without osteoarthritis (HOA). We hypothesized that there would be an increase in PFC activation during PW relative to comfortable walking (CW) and higher increase in PFC activation during PW in HOA compared to OA.MethodsTwenty community-dwelling older women (66.7 ± 5.41 years old) walked on an instrumented treadmill that provided perturbations at pseudo-random intervals between 5–25 s using a counterbalanced design. Functional Near Infrared Spectroscopy was used to quantify PFC oxygenated hemoglobin (HbO2) and deoxyhemoglobin (Hb) levels, while standing prior to the task as a baseline. A linear mixed effects model was conducted to investigate the effects of cohort (HOA vs OA), task (PW vs CW), and their interaction on HbO2 (μM) and Hb (μM) levels.ResultsHbO2 and Hb levels differed significantly between CW and PW tasks for both cohorts (P < 0.001) and demonstrated significant task by cohort interaction (P < 0.05). In addition, we found changes in walking performance (stride time, stride length, stride width and stance time) during and after PW. Spearman correlation demonstrated a strong association between increased stance time, increased body mass index and decreased PFC activation during PW. No other significant results were found.DiscussionThis study found increase in PFC activation during PW and gait adaptation after a short bout of PW in HOA and OA. This increase in PFC activation was higher in HOA compared to OA, particularly during PW tasks, and was consistent with theory of limitations in mobility affecting neural activation in older adults. Further work remains to examine how pain, obesity, and mobility impacts cortical control in older adults with and without osteoarthritis.

Printable colloidal photonic crystals (CPCs) with unique photonic bandgaps and elaborate shapes have attracted significant interest due to their characteristics, such as simplicity of fabrication, adjustable structural colors, photobleaching resistance, and stimulus‐responsiveness. In this review, strategies for printing CPC patterns, including direct use of CPCs as inks, region‐selective modification on responsive (solvent, force, and temperature) CPC papers, and printing combined with lithography, are first summarized. Second, based on the advantages of CPC printing technology, their applications in color displays, coatings, sensors, anticounterfeiting labels, and information storage, are discussed in detail. Finally, the current challenges and outlook regarding CPC printing technology are proposed.

Pulmonary arterial hypertension (PAH) features pathogenic and abnormal endothelial cells (ECs), and one potential origin is clonal selection. We studied the role of p53 and toll-like receptor 3 (TLR3) in clonal expansion and pulmonary hypertension (PH) via regulation of bone morphogenetic protein (BMPR2) signaling. ECs of PAH patients had reduced p53 expression. EC-specific p53 knockout exaggerated PH, and clonal expansion reduced p53 and TLR3 expression in rat lung CD117+ ECs. Reduced p53 degradation (Nutlin 3a) abolished clonal EC expansion, induced TLR3 and BMPR2, and ameliorated PH. Polyinosinic/polycytidylic acid [Poly(I:C)] increased BMPR2 signaling in ECs via enhanced binding of interferon regulatory factor-3 (IRF3) to the BMPR2 promoter and reduced PH in p53-/- mice but not in mice with impaired TLR3 downstream signaling. Our data show that a p53/TLR3/IRF3 axis regulates BMPR2 expression and signaling in ECs. This link can be exploited for therapy of PH.

With the rapid development of the field of life sciences, traditional 2D cell culture and animal models have long been unable to meet the urgent needs of modern biomedical research and new drug development. Establishing a new generation of experimental models and research models is of great significance for deeply understanding human health and disease processes, and developing effective treatment measures. As is well known, long research and development cycles, high risks, and high costs are the “three mountains” facing the development of new drugs today. Organoids and organ-on-chips technology can highly simulate and reproduce the human physiological environment and complex reactions in vitro, greatly improving the accuracy of drug clinical efficacy prediction, reducing drug development costs, and avoiding the defects of drug testing animal models. Therefore, organ-on-chips have enormous potential in medical diagnosis and treatment.

Accurately estimating shrub biomass in arid and semi-arid regions is critical for understanding ecosystem productivity and carbon stocks at both local and global scales. Due to the short and sparse features of shrubs, capturing the shrub biomass accurately by satellite observations is challenging. Previous studies mostly used ground samples and satellite observations to estimate shrub biomass by establishing a direct connection, which was often hindered by the limited number of ground samples and spatial scale mismatch between samples and observations. Unmanned aerial vehicles (UAVs) provide opportunities to obtain more samples that are in line with the aspects of satellite observations (i.e., scale) for regional-scale shrub biomass estimations accurately with low costs. However, few studies have been conducted based on the air-space-ground-scale connection assisted by UAVs. Here we developed a framework for estimating 10 m shrub biomass at a regional scale by integrating ground measurements, UAV, Landsat, and Sentinel-1/2 observations. First, the spatial distribution map of shrublands and non-shrublands was generated in 2023 in the Helan Mountains of Ningxia province, China. This map had an F1 score of 0.92. Subsequently, the UAV-based shrub biomass map was estimated using an empirical model between the biomass and the crown area of shrubs, which was aggregated at a 10 m × 10 m grid to match the spatial resolution of Sentinel-1/2 images. Then, a regional-scale estimation model of shrub biomass was developed with a random forest regression (RFR) approach driven by ground biomass measurements, UAV-based biomass, and the optimal satellite metrics. Finally, the developed model was used to produce the biomass map of shrublands over the study area in 2023. The uncertainty of the resultant biomass map was characterized by the pixel-level standard deviation (SD) using the leave-one-out cross-validation (LOOCV) method. The results suggested that the integration of multi-scale observations from the ground, UAVs, and satellites provided a promising approach to obtaining the regional shrub biomass accurately. Our developed model, which integrates satellite spectral bands and vegetation indices (R2 = 0.62), outperformed models driven solely by spectral bands (R2 = 0.33) or vegetation indices (R2 = 0.55). In addition, our estimated biomass has an average uncertainty of less than 4%, with the lowest values (30%) and biomass production (>300 g/m2). This study provides a methodology to accurately monitor the shrub biomass from satellite images assisted by near-ground UAV observations as well as ground measurements.

To further study the reservoir characteristics and adsorption capacity of the Taiyuan Formation shale in the South North China Basin (SNCB), the pore structure and adsorption capacity of shale are discussed using various analysis tests, including elemental geochemistry, organic geochemistry, mineral composition, low-temperature nitrogen adsorption (LTNA), and methane adsorption experiments. The results indicate that the Taiyuan Formation shale formed in a poor oxygen and anaerobic sedimentary environment in still water. The average value of total organic carbon (TOC) content is 2.37%. The organic matter type mainly consists of type III kerogen. The vitinite reflectance (Ro) ranges from 3.11% to 3.50%. The clay mineral content varies greatly, averaging at 40.7%, while the quartz content averages at 37.7%. The Taiyuan Formation shale mainly develops interparticle (InterP) pores, followed by organic pores, intraparticle (IntraP) pores, solution pores, and microfractures. BET specific surface area (SSA) is between 9.47 m2/g and 22.14 m2/g, while pore volume (PV) ranges from 0.0098 cm3/g to 0.022 cm3/g, indicating favorable conditions for shale gas storage. According to the results of the CH4 adsorption experiment, Langmuir volume from Taiyuan Formation shales exhibits 1.35~4.30 cm3/g, indicating excellent adsorption capacity. TOC content shows a positive correlation with both Langmuir volume and BET SSA from Taiyuan Formation shales, suggesting that TOC plays a crucial role in controlling microscopic pores and gas adsorption capacity. Organic matter enhances the shale adsorption capacity by providing abundant pore SSA. Due to formation compaction, the pore size of clay minerals decreases, leading to an increase in pore SSA, while kaolinite exhibits weak hydrophilic ability. Consequently, with the increase in clay minerals and kaolinite content, the shale adsorption capacity is enhanced to a certain extent. However, an increase in the carbonate mineral content may result in a decrease in the proportion of clay minerals, therefore reducing the CH4 adsorption capacity of shale.

Tai Chi is a Chinese martial art that provides an adaptive and accessible exercise for older adults with varying functional capacity. While Tai Chi is widely recommended for its physical benefits, wider adoption in at-home practice presents challenges for practitioners, as limited feedback may hamper learning. This study examined the feasibility of using a wearable sensor, combined with machine learning (ML) approaches, to automatically and objectively classify Tai Chi expertise. We hypothesized that the combination of wrist acceleration profiles with ML approaches would be able to accurately classify practitioners’ Tai Chi expertise levels. Twelve older active Tai Chi practitioners were recruited for this study. The self-reported lifetime practice hours were used to identify subjects in low, medium, or highly experienced groups. Using 15 acceleration-derived features from a wearable sensor during a self-guided Tai Chi movement and 8 ML architectures, we found multiclass classification performance to range from 0.73 to 0.97 in accuracy and F1-score. Based on feature importance analysis, the top three features were found to each result in a 16–19% performance drop in accuracy. These findings suggest that wrist-wearable-based ML models may accurately classify practice-related changes in movement patterns, which may be helpful in quantifying progress in at-home exercises.

Cardiac sarcomas, characterized by their rarity and aggressive nature, pose substantial diagnosis and treatment challenges. This Commentary emphasizes a multidisciplinary approach integrating clinical insights, advanced imaging techniques, histopathological analysis, and molecular diagnostics to achieve effective personalized patient management. It discusses the critical role of surgical resection in managing localized disease, complemented by adjuvant therapies such as chemotherapy and radiotherapy, to improve patient outcomes. This Commentary highlights emerging treatments including targeted therapies and immunotherapy, and calls for sustained research efforts to better understand the molecular underpinnings of these tumors and develop more effective treatments.

In order to solve the problems of serious dust pollution and difficult dust removal in fully mechanized coal mining face, we conduct research on the concentration and particle size field of dust in the fully mechanized mining face of Shoushan No.1 Mine, propose the dust source multi-level and multi-dimensional closed atomization control dust removal technology. The results show that the air flow occurs around and across the shearer drum, and the wind speed is up to 2.5 m/s, the wind speed tends to be stable at about 10 m away from the shearer; a high-speed wind flow zone was formed in 9 m area downwind of the center of the front drum, with the average wind speed exceeding 2.0 m/s. The high concentration dust belt with a length of 38 m is formed in the goaf of the unmoved frame area within 10 m away from it, and the concentration is between 1 629 mg/m3 and 2 257 mg/m3. The length of the high concentration dust belt near the center height of the front roller reaches 40 m, and the dust concentration in this range is between 1 590 mg/m3 and 2 845 mg/m3. In the pavement area, the particle size ranges from 0.25 μm to 10 μm, causing great disturbance to the breathing of the downhole workers. After the application of dust source multi-stage and multi-dimensional closed atomization control dust removal technology, the average dust reduction rate of on-site exhaled dust was 84.68%, and the average dust reduction rate of total dust was 88.52%.
