Abstract

The spin-frustrated conductor Mn$_3$Pt exhibits a characteristic magnetic structure called partial disorder in which some spin sites can form magnetic order through the generation of non-ordered sites that locally relieve the frustration. Here we report the results of a single-crystal inelastic neutron scattering study of this compound. The measured momentum $\vec{Q}$ correlations of diffusive magnetic scattering reveal that the paramagnetic phase exhibits short-range spin fluctuations with the same type of partial disorder. Abstract

目的：体内必須微量元素FeとZnが習慣化した激しい運動によってどの程度変動するのか、また、その体内分布について研究した報告はほとんどない。本研究では雄ラットをトレッドミルで強度が異なる運動をさせた時に、生体内のバイオパラメータはどのように変動するのか、FeとZnが体内でどのような挙動を示すのか、運動強度による体内存在量に違いが生じるのかについて検証を行った。 方法：6週齢Wistar系雄ラットを非運動群（n=4）、低強度運動群（n＝4）、高強度運動群（n=4）に分類し、1日1回、週5日の頻度で運動させた。飼育12週間後、解剖を行い、血液、臓器を採取し、血球成分、FeおよびZn量などを測定した。 結果：赤血球（RBC）、ヘモグロビン（HGB）、ヘマトクリット（HCT）は運動強度に応じて減少し、平均赤血球容積（MCV）、平均赤血球ヘモグロビン量（MCH）は運動強度に応じて増加する傾向が見られた。Fe濃度は脾臓が最も高く、Zn濃度は骨が最も高かった。 結論：高強度運動群は貧血を発症していることが考えられ、激しい運動を行う男性スポーツ選手の貧血によるパフォーマンス低下を軽減させるため、今後、FeおよびZnの体内分布を詳細に検討し、データを蓄積する必要がある。

24. Anti-hyperglycemic Effect of Long-Term Bis(hinokitiolato)zinc Complex ([Zn(hkt)(2)]) Ingestion on Insulin Resistance and Pancreatic Islet Cells Protection in Type 2 Diabetic KK-A(Y) Mice

Abstract

Zinc (Zn) is a trace element with anti-diabetes mellitus (anti-DM) effects. Zn complexes exhibit stronger insulin-like activity than Zn ions. Bis(hinokitiolato)zinc complex ([Zn(hkt)(2)]) was recently reported to be a potent anti-DM candidate. We examined the effects of [Zn(hkt)(2)] on insulin resistance and pancreatic islet cells through in vivo long-term ingestion studies. In an in vivo study, we performed 4-month long-term [Zn(hkt)(2)] administration experiments in KK-A(Y) mice as a type 2 DM animal model. Ingestion of [Zn(hkt)(2)] resulted in lower blood glucose levels compared with the non-treated KK-A(Y) mice (control group). Additionally, [Zn(hkt)(2)] treatment decreased plasma insulin concentration compared with that of the non-treated KK-A(Y) group. [Zn(hkt)2] treatment resulted in a significant suppression of islet cell enlargement and a significantly decreased number of insulin-positive cells compared with the non-treated KK-A(Y) control group. The [Zn(hkt)(2)] treatment group showed the increasing tendency in the amount of Zn levels in peripheral organs; liver, muscle, adipose, and pancreas, compared with the non-treated KK-A(Y) control group. However, the Zn level in the pancreas of the [Zn(hkt)(2)] treatment group did not show the significant increase compared with the non-treated KK-A(Y) control group. This accumulation of Zn in pancreas suggested that [Zn(hkt)(2)] mainly effects on the peripheral tissue, and [Zn(hkt)(2)] has the less effect on the pancreas directly. Thus, we concluded that [Zn(hkt)(2)] exerted the main effect on peripheral organs by ameliorating insulin resistance.

29. Global overexpression of divalent metal transporter 1 delays crocidolite-induced mesothelial carcinogenesis in male mice

Abstract

Exposure to asbestos fiber is central to mesothelial carcinogenesis, for which iron overload in or near mesothelial cells is a key pathogenic mechanism. Alternatively, iron chelation therapy with deferasirox or regular phlebotomy was significantly preventive against crocidolite-induced mesothelial carcinogenesis in rats. However, the role of iron transporters during asbestos-induced carcinogenesis remains elusive. Here, we studied the role of divalent metal transporter 1 (DMT1; Slc11a2), which is a Fe(II) transporter, that is present not only on the apical plasma membrane of duodenal cells but also on the lysosomal membrane of every cell, in crocidolite-induced mesothelial carcinogenesis using DMT1 transgenic (DMT1Tg) mice. DMT1Tg mice show mucosal block of iron absorption without cancer susceptibility under normal diet. We unexpectedly found that superoxide production was significantly decreased upon stimulation with crocidolite both in neutrophils and macrophages of DMT1Tg mice, and the macrophage surface revealed higher iron content 1 h after contact with crocidolite. Intraperitoneal injection of 3 mg crocidolite ultimately induced malignant mesothelioma in ∼50% of both wild-type and DMT1Tg mice (23/47 and 14/28, respectively); this effect was marginally (p = 0.069) delayed in DMT1Tg mice, promoting survival. The promotional effect of nitrilotriacetic acid was limited, and the liver showed significantly higher iron content both in DMT1Tg mice and after crocidolite exposure. The results indicate that global DMT1 overexpression causes decreased superoxide generation upon stimulation in inflammatory cells, which presumably delayed the promotional stage of crocidolite-induced mesothelial carcinogenesis. DMT1Tg mice with low-stamina inflammatory cells may be helpful to evaluate the involvement of inflammation in various pathologies.

Abstract

Abstract Background Parkinson's disease (PD) and cerebral ischemia are chronic and acute neurodegenerative diseases, respectively, and onsets of these diseases are thought to be induced at least by oxidative stress. PD is caused by decreased dopamine levels in the substantia nigra and striatum, and cerebral ischemia occurs as a result of local reduction or arrest of blood supply. Although a precursor of dopamine and inhibitors of dopamine degradation have been used for PD therapy and an anti-oxidant have been used for cerebral ischemia therapy, cell death progresses during treatment. Reagents that prevent oxidative stress-induced cell death are therefore necessary for fundamental therapies for PD and cerebral ischemia. DJ-1, a causative gene product of a familial form of PD, PARK7, plays roles in transcriptional regulation and anti-oxidative stress, and loss of its function is thought to result in the onset of PD. Superfluous oxidation of cysteine at amino acid 106 (C106) of DJ-1 renders DJ-1 inactive, and such oxidized DJ-1 has been observed in patients with the sporadic form of PD. Results In this study, a compound, comp-23, that binds to DJ-1 was isolated by virtual screening. Comp-23 prevented oxidative stress-induced death of SH-SY5Y cells and primary neuronal cells of the ventral mesencephalon but not that of DJ-1-knockdown SH-SY5Y cells, indicating that the effect of the compound is specific to DJ-1. Comp-23 inhibited the production of reactive oxygen species (ROS) induced by oxidative stress and prevented excess oxidation of DJ-1. Furthermore, comp-23 prevented dopaminergic cell death in the substantia nigra and restored movement abnormality in 6-hydroxyldopamine-injected and rotenone-treated PD model rats and mice. Comp-23 also reduced infarct size of cerebral ischemia in rats that had been induced by middle cerebral artery occlusion. Protective activity of comp-23 seemed to be stronger than that of previously identified compound B. Conclusions The results indicate that comp-23 exerts a neuroprotective effect by reducing ROS-mediated neuronal injury, suggesting that comp-23 becomes a lead compound for PD and ischemic neurodegeneration therapies.

Abstract

Japan has many unexcavated tumuli, most of which were buried along with artifacts of precious cultural heritage. For such a tumulus, it is essential to understand how changes in its exterior environment affect its interior environment, and how those interior changes affect the deterioration of the relics buried in the stone chamber. In this study, an underground space was constructed in the forest of the Katsura Campus of Kyoto University to simulate the environment of an unexcavated tumulus, and long-term monitoring was implemented in the simulated stone chamber, including the temperature, humidity, water potential, wetness, and oxygen and carbon dioxide concentrations, along with metal corrosion tests. This article is focused on environmental monitoring, and the results demonstrate that the simulated tumulus has the general characteristics of the hygrothermal environments of an unexcavated tumulus that has small temperature fluctuation and near-saturation humidity. The ceiling of the simulated chamber condensed significantly from October to April, which is related to the variations of the ceiling and floor temperatures. Also, the wetness of the walls in the simulated chamber was affected by rainfall. The oxygen concentration in the simulated stone chamber varied in the range of 13%–19% in 2015, and the variation of carbon dioxide concentration in the simulated stone chamber was contrary to the oxygen concentration and varied in the range of 3%–9% in 2016. The oxygen concentration in the stone chamber was similar to that in the surrounding soil that decreased at times of rainfall, contrary to the fluctuations in the soil water content. Abstract

Vanadium has potential for use in diabetes therapy. Many investigators have reported toxic effects of inorganic vanadium salts; however, there are few reports on toxic effects of oxovanadium(VO(2+)) complexes. Therefore, we studied VO(2+) toxicity by examining histological changes and measuring the vanadium concentration in the testis after repeated oral administration of bis(1-oxy-2-pyridine-thiolato)oxovanadium(VO(2+)) (VO(opt)2) for 2 or 4 weeks in KK-A(y) mice. Severe mineralization and degeneration/necrosis of the seminiferous tubules were detected after either 2 or 4 weeks of administration. Vacuolar changes in Sertoli cells and the seminiferous epithelia, and hyperplasia of Leydig cells were observed in the testes of some animals. Vanadium concentrations in the mineralized testis were much higher than those in the testis of untreated KK-A(y) mice. These results represent the first report of the possibility for seminiferous tubules mineralization induced by VO(opt)2 administration. Therefore, our research provides important information about the potentially toxic effects of VO(2+) complexes.
