This study presents a novel multistage expressway fast charging station (EFCS) planning problem coordinated with the dynamic regional power structure (PS) transformation. Under the prerequisite of the EFCS network's sustainable operation, network accessibility, and orderly construction, a three‐step planning method oriented to the enhancement of energy saving and emission reduction (ESER) benefits and rationalization of facility utilization is developed: (i) EV‐expanded network, (ii) multiagent‐based dynamic traffic assignment (MA‐DTA), and (iii) deployment refinement. Embedding the MA‐DTA and customized refinement strategy into the iterative planning structure enables the integration of operations and planning of the EFCS network. A numerical experiment and an empirical study in the Shandong Peninsula urban agglomeration are conducted. It demonstrates that the method can find a high‐quality solution within acceptable computation time and is applicable to realistic large‐scale EFCS planning. The planning method can effectively play the role of economy and facility in inducing EV users' charging demands, thus enhancing the overall ESER benefits. The integration of operation and planning is proven effective in reasonably matching the supply and demand of facility service and charging loads in a full‐time period. The isolation technologies are adopted to protect the long-span space structures from serious damage under the action of earthquakes. When the isolation system is applied to the space structures, both horizontal and vertical vibration control effects on the structures should be concerned. In this paper, a two-dimensional earthquake isolation and mitigation device (TEIMD) composed of a high-damping viscoelastic (VE) core pad and VE dampers is proposed. Based on the dynamic shear experiment results of the VE damper, the energy dissipation capability of VE material is preferable. Then, the nonlinear equivalent stiffness and equivalent damping ratio of the TEIMD in horizontal and vertical directions are evaluated by using the numerical simulation method. The TEIMDs are arranged on the column top of a double-layer grid structure to assess the isolation effectiveness of the novel device. The dynamic analysis results of the double-layer grid structure with and without TEIMD reveal that this novel device can significantly reduce the acceleration response of the upper grid by relying on the strong energy dissipation performances in the horizontal and vertical directions. This research also verifies the feasibility of applying the TEIMD to the double-layer grid structure, which can effectively dissipate the vertical vibration energy of the structure. Objective To explore the mediating role of self-esteem in the relationship between childhood adverse experiences (ACEs) and depressive symptoms of junior high school students, and the regulatory effect of social support. Methods A total of 1 308 students from 3 junior middle schools in Xiangxi were selected by stratified random cluster sampling from February to April 2023. A questionnaire survey was conducted with the use of Childhood adverse experience questionnaire, Adolescent social support scale, PHQ-9 Depression symptom screening scale, and Rosenberg self-esteem scale. The data were analyzed by correlation analysis, intermediary analysis, and moderated intermediary model test. Results (1) ACEs positively predicted depressive symptoms of junior high school students (β=0.801, t=7.246, P<0.05), while self-esteem and social support negatively predicted depressive symptoms of junior high school students (β=-0.406, t=-14.276, P < 0.05; β =-0.027, t=-2.781, P < 0.05). (2) Self-esteem played a mediating role between ACEs and depressive symptoms (effect=0.468, P < 0.05), and the mediating effect accounted for 31.33% of the total effect. (3) Social support directly regulated the relationship between ACEs and depressive symptoms (β=-0.017, t=-3.043, P<0.01), and social support played a regulatory role in the latter half of the mediating pathway of "ACEs self-esteem and depressive symptoms" (β=-0.004, t=-2.887, P<0.01). Conclusion There is a mediating effect between ACEs and depressive symptoms of junior high school students, and self-esteem is the mediating variable of the relationship between them. Social support buffers the influence of ACEs on the risk of depressive symptoms of junior high school students and strengthens the relationship between self-esteem and depressive symptoms. Therefore, giving more care and social support to junior high school students with ACEs and improving their self-esteem will help prevent and reduce the occurrence of depressive symptoms. Purpose: Chronic inflammation is one of the key mechanisms of depression. Wenyang-Tianjin-Jie Decoction (WTJD) is an effective antidepressant found in the course of diagnosis and treatment, but the mechanism of therapeutic effect is not clear. The study aimed to evaluate the efficacy of WTJD in the kidney yang deficiency (KYD) type of depression rats and reveal its mechanisms. Materials and Methods: We selected forty 6-week-old male Sprague-Dawley rats for the study. We established a KYD [Phellodendron amurense Rupr (Huangbai) solution oral gavage and 4°C environments; 8 weeks] type of depression (chronic unpredictable mild stimulus; 6 weeks) rat model first. After successful modeling, we used WTJD or fluoxetine on rats for 3 weeks. Then we evaluated the depression and KYD behavior. Finally, we observed the expression of key inflammatory factors and proteins in peripheral blood and hippocampus, and further investigated the immune balance of Th17/Treg and Th1/Th2 cells and the activity of their main regulatory pathways JAK2/STAT3 and TLR4/TRAF6/NF-κB. Results: The imbalance of Th17/Treg and Th1/Th2 cells in rats were related to KYD and depressive symptoms. Through this study, we found that WTJD can inhibit the activity of JAK2/STAT3 and TLR4/TRAF6/NF-κB pathways, balance Th17/Treg and Th1/Th2 cell homeostasis, regulate the levels of inflammatory factors in the hippocampus and peripheral blood, and reverse KYD and depression. Conclusion: This study confirmed that WTJD had a reliable effect on depression rats with KYD, and its mechanism was to regulate the immune homeostasis of hippocampal T cells and related inflammatory factors to improve KYD and depression symptoms in rats. Simple Summary: Whirligig beetles inhabit both fresh and brackish waters and possess a pair of split compound eyes. The dorsal eyes are situated above the water surface, whereas the ventral eyes are submerged beneath the water. Due to the difference in optical environments between water and air, we expected a difference in visual features and signal processing between the dorsal and ventral eyes. Using scanning electron microscopy, transmission electron microscopy, and microcomputed tomography, we investigated the functional modifications of different features of the split compound eyes of Dineutus mellyi (Coleoptera: Gyrinidae). Both dorsal and ventral parts of the split compound eyes of D. mellyi are the superposition type, with the ommatidium of dorsal and ventral eyes comprised of a laminated corneal lens, bullet-shaped crystalline cone, upper distal rhabdom, a clear zone and lower distal rhabdom, a basal rhabdom, and an eight retinular cell just above the basement membrane. In contrast to the dorsal eyes, ventral eyes are characterized by a higher field of vision but exhibit similar spatial resolution. The functional anatomy of the split compound eyes of whirligig beetles Dineutus mellyi (Coleoptera: Gyrinidae) was examined by advanced microscopy and microcomputed tomography. We report the first 3D visualization and analysis of the split compound eyes. On average, the dorsal and ventral eyes contain 1913 ± 44.5 facets and 3099 ± 86.2 facets, respectively. The larger area of ventral eyes ensures a higher field of vision underwater. The ommatidium of the split compound eyes is made up of laminated cornea lenses that offer protection against mechanical injuries, bullet-shaped crystalline cones that guide light to the photoreceptive regions, and screening pigments that ensure directional light passage. The photoreceptive elements, made up of eight retinular cells, exhibit a tri-tiered rhabdom structure, including the upper distal rhabdom, a clear zone that ensures maximum light passage, and an enlarged lower distal rhabdom that ensures optimal photon capture. Rice effective panicle is a major trait for grain yield and is affected by both the genetic tiller numbers and the early tillering vigor (ETV) traits to survive environmental adversities. The mechanism behind tiller bud formation has been well described, while the genes and the molecular mechanism underlying rice-regulating ETV traits are unclear. In this study, the candidate genes in regulating ETV traits have been sought by quantitative trait locus (QTL) mapping and bulk-segregation analysis by resequencing method (BSA-seq) conjoint analysis using rice backcross inbred line (BIL) populations, which were cultivated as late-season rice of double-cropping rice systems. By QTL mapping, seven QTLs were detected on chromosomes 1, 3, 4, and 9, with the logarithm of the odds (LOD) values ranging from 3.52 to 7.57 and explained 3.23% to 12.98% of the observed phenotypic variance. By BSA-seq analysis, seven QTLs on chromosomes 1, 2, 4, 5, 7, and 9 were identified using single-nucleotide polymorphism (SNP) and insertions/deletions (InDel) index algorithm and Euclidean distance (ED) algorithm. The overlapping QTL resulting from QTL mapping and BSA-seq analysis was shown in a 1.39 Mb interval on chromosome 4. In the overlap interval, six genes, including the functional unknown genes Os04g0455650, Os04g0470901, Os04g0500600, and ethylene-insensitive 3 (Os04g0456900), sialyltransferase family domain containing protein (Os04g0506800), and ATOZI1 (Os04g0497300), showed the differential expression between ETV rice lines and late tillering vigor (LTV) rice lines and have a missense base mutation in the genomic DNA sequences of the parents. We speculate that the six genes are the candidate genes regulating the ETV trait in rice, which provides a research basis for revealing the molecular mechanism behind the ETV traits in rice. Background: Sleep deprivation (SD) is commonplace in modern society and there are large individual differences in the vulnerability to SD. We aim to identify the structural network differences based on diffusion tensor imaging (DTI) that contribute to the individual different vulnerability to SD. Methods: The number of psychomotor vigilance task (PVT) lapses was used to classify 49 healthy subjects on the basis of whether they were vulnerable or resistant to SD. DTI and graph theory approaches were used to investigate the topologic organization differences of the brain structural connectome between SD‐vulnerable and ‐resistant individuals. We measured the level of global efficiency and clustering in rich club and non‐rich club organizations. Results: We demonstrated that participants vulnerable to SD had less global efficiency, network strength, and local efficiency but longer shortest path length compared with participants resistant to SD. Lower efficiency was mainly distributed in the right insula, bilateral thalamus, bilateral frontal, temporal, and temporal lobes. Furthermore, a disrupted subnetwork was observed that consisted of widespread connections. Moreover, the vulnerable group showed significantly decreased strength of the rich club compared with the resistant group. The strength of rich club connectivity was found to be correlated negatively with PVT performance (r = −0.395, p = 0.005). We further tested the reliability of the results. Conclusion: The findings revealed that individual differences in resistance to SD are related to disrupted topologic efficiency connectome pattern, and our study may provide potential connectome‐based biomarkers for the early detection of the vulnerable degree to SD. Hepatoblastoma (HB) is a common primary liver malignant tumor in children. Long non-coding RNAs (lncRNAs) are closely engaged in HB progression. The role and regulatory molecule mechanism of lncRNA small nucleolar RNA host gene 1 (SNHG1) in HB remain unclear. Through qRT-PCR or western blot, we found that SNHG1 and proviral integration site for moloney murine leukemia virus 3 (PIM3) were elevated but miR-6838-5p was decreased in HB cells. Cell biology experiments revealed that SNHG1 depletion or miR-6838-5p upregulation suppressed cell proliferation, migration, and invasion of HB cells. Mechanistically, luciferase activity assay validated that miR-6838-5p could interact with SNHG1 or PIM3. SNHG1 up-regulated PIM3 expression via sponging miR-6838-5p. Moreover, miR-6838-5p inhibitor abolished SNHG1 depletion-mediated suppression of malignant behaviors in HB cells. PIM3 overexpression neutralized miR-6838-5p mimics-mediated repression of malignant phenotypes in HB cells. Furthermore, miR-6838-5p overexpression suppressed RhoA activation, which was restored by PIM3 upregulation. What's more, the results at the cellular level were further verified by nude mice tumor formation experiment. In conclusion, SNHG1 regulated miR-6838-5p/PIM3/RhoA axis to promote malignant phenotypes of HB, which might provide novel therapeutic target for HB treatment. [ABSTRACT FROM AUTHOR]

Microwave sintering is a key technology for future lunar habitats, which has been verified by various lunar soil simulant experiments. However, nanophase iron (np-Fe0), a unique component of the real lunar regolith, has been ignored in previous studies of the microwave sintering of lunar soil simulants. In this study, microwave-sintering experiments of lunar soil simulant CLRS-2 with and without np-Fe0 at different temperatures were conducted. A comparison of the microstructure and chemical composition of sintered products indicates that all products of samples with 1.0 wt% np-Fe0 exhibit better properties than those without np-Fe0 after heating at equivalent temperatures. Adding np-Fe0 to the lunar soil simulant CLRS-2 couples well with microwaves, which may increase the real heating temperature in a sample, resulting in the product being solidified at 700 °C and a relatively dense product forming at 900 °C, thus improving the sintering efficiency. This study suggests that microwave sintering is an effective potential technology for future lunar habitats and road hardening because of the wide distribution of np-Fe0 in the lunar regolith. Background: Brain metastases (BMs) are the most frequent intracranial tumours associated with poor clinical outcomes. Radiotherapy is essential in the treatment of these tumours, although the optimal radiation strategy remains controversial. The present study aimed to assess whether whole brain radiation therapy with a simultaneous integrated boost (WBRT + SIB) provides any therapeutic benefit over WBRT alone. Methods: We included and retrospectively analysed 82 patients who received WBRT + SIB and 83 who received WBRT alone between January 2012 and June 2021. Intracranial progression-free survival (PFS), local tumour control (LTC), overall survival (OS), and toxicity were compared between the groups. Results: Compared to WBRT alone, WBRT + SIB improved intracranial LTC and PFS, especially in the lung cancer subgroup. Patients with high graded prognostic assessment score or well-controlled extracranial disease receiving WBRT + SIB had improved intracranial PFS and LTC. Moreover, WBRT + SIB also improved the long-term intracranial tumour control of small cell lung cancer patients. When evaluating toxicity, we found that WBRT + SIB might slightly increase the risk of radiation-induced brain injury, and that the risk increased with increasing dosage. However, low-dose WBRT + SIB had a tolerable radiation-induced brain injury risk, which was lower than that in the high-dose group, while it was comparable to that in the WBRT group. Conclusions: WBRT + SIB can be an efficient therapeutic option for patients with BMs, and is associated with improved intracranial LTC and PFS. Furthermore, low-dose WBRT + SIB (biologically effective dose [BED] ≤ 56 Gy) was recommended, based on the acceptable risk of radiation-induced brain injury and satisfactory tumour control. The tissue stiffness of donor kidneys in transplantation may increase due to pathological changes such as glomerulosclerosis and interstitial fibrosis, and those changes associate worse outcomes in kidney transplantation recipients. Ultrasound elastography is a noninvasive imaging examination with the ability to quantitatively reflect tissue stiffness. Aim of this study was to evaluate the prognostic value of ultrasound elastography for adverse kidney outcome in kidney transplantation recipients. Shear wave elastography (SWE) examinations were performed by two independent operators in kidney transplantation recipients. The primary outcome was a composite of kidney graft deterioration, all-cause re-hospitalization, and all-cause mortality. Survival analysis was calculated by Kaplan-Meier curves with the log-rank test and Cox regression analysis. A total of 161 patients (mean age 46 years, 63.4% men) were followed for a median of 20.1 months. 27 patients (16.77%) reached the primary endpoint. The mean and median tissue stiffness at the medulla (hazard ratio: 1.265 and 1.229, respectively), estimated glomerular filtration rate (eGFR), and serum albumin level were associated with the primary outcome in univariate Cox regression. Adding mean or median medulla SWE to a baseline model containing eGFR and albumin significantly improved its discrimination (C-statistics: 0.736 for the baseline, 0.766 and 0.772 for the model added mean and median medulla SWE, respectively). The medullary tissue stiffness of kidney allograft measured by shear wave elastography may provide incremental prognostic value to adverse outcomes in kidney transplantation recipients. Plants experience a variety of adverse environments during their vegetative growth and reproductive development, and to ensure that they complete their life cycle successfully, they have evolved specific defense mechanisms to cope with unfavorable environments. Flowering is a vital developmental stage and an important determinant of productivity in the lifetime of plants, which can be vulnerable to multiple abiotic stresses. Exposure to stress during this period can have dramatic effects on flower physiological and morphological development, which may ultimately lead to a substantial loss of yield in seed-producing plants. However, there has been increasing research evidence that diverse abiotic stresses, ranging from drought, low temperature, and heat stress can promote or delay plant flowering. This review focuses on how plants alter developmental direction to balance between survival and productivity under drought and extreme temperature conditions. Starting from the perspective of the functional analysis of key flowering-regulated genes, it is of great help for researchers to quickly gain a deeper understanding of the regulatory effects of abiotic stress on the flowering process, to elucidate the molecular mechanisms, and to improve the regulatory network of abiotic-stress-induced flowering. Additionally, the important agronomic significance of the interaction between abiotic stress and the flowering regulation of perennial plants under climate change conditions is also discussed after summarizing studies on the mechanisms of stress-induced flowering in annual plants. This review aims to clarify the effects of abiotic stresses (mainly drought and temperature) on plant flowering, which are significant for future productivity increase under unfavorable environmental conditions. Forest growing stock volume (GSV) is an essential aspect of ecological carbon stock monitoring. The successive launches of spaceborne microwave satellites have provided a broader way to use microwave remote sensing to monitor forest accumulation. Currently, the inversion parameterization models of active microwave remote sensing stock volume mainly include the interferometric water cloud (IWCM), BIOMASAR, and Siberia. Among them, the IWCM introduces backscattering and coherence, the BIOMASAR model only introduces backscattering, and the Siberia model only introduces coherence. Although these three models combine the backscatter coefficient and coherence of SAR to estimate volume accumulation, the performance of the models has not been evaluated at the same time in the same area. Therefore, this article starts from the perspective of the three combinations of coherence and backscattering, relies on three models that do not require measured data, and evaluates the accuracy of the models' overall inversion of GSV. In addition, we combine precipitation meteorological information, vegetation types, and seasonal variation to separately explore model performance. The comparison results show that the IWCM model is relatively stable in the process of stock volume inversion and is more sensitive to the vegetation types of coniferous and deciduous forests. The influence of seasons and precipitation on the model is weak, and the accuracy of the multi-time-series model is slightly improved. The Siberia model has a good storage volume inversion effect in this study area, but the multiple time series did not improve the model accuracy. The BIOMASAR model is simple, and its performance was slightly inferior in this study area. Precipitation can negatively affect BIOMASAR. The model results for multiple time series outperform those for single time. In summary, the stability of IWCM is more suitable for research with unknown information. The BIOMASAR model is simple, does not require coherence calculations, and is ideal for the estimation of large-scale national or world-level storage distributions. Background: The plant hormone auxin is widely involved in plant growth, development, and morphogenesis, and the TIR1/AFB and AUX/IAA proteins are closely linked to rapid auxin response and signal transmission. However, their evolutionary history, historical patterns of expansion and contraction, and changes in interaction relationships are still unknown. Results: Here, we analyzed the gene duplications, interactions, and expression patterns of TIR1/AFBs and AUX/IAAs to understand their underlying mechanisms of evolution. The ratios of TIR1/AFBs to AUX/IAAs range from 4:2 in Physcomitrium patens to 6:29 in Arabidopsis thaliana and 3:16 in Fragaria vesca. Whole-genome duplication (WGD) and tandem duplication have contributed to the expansion of the AUX/IAA gene family, but numerous TIR1/AFB gene duplicates were lost after WGD. We further analyzed the expression profiles of TIR1/AFBs and AUX/IAAs in different tissue parts of Physcomitrium patens, Selaginella moellendorffii, Arabidopsis thaliana and Fragaria vesca, and found that TIR1/AFBs and AUX/IAAs were highly expressed in all tissues in P. patens, S. moellendorffii. In A. thaliana and F. vesca, TIR1/AFBs maintained the same expression pattern as the ancient plants with high expression in all tissue parts, while AUX/IAAs appeared tissue-specific expression. In F. vesca, 11 AUX/IAAs interacted with TIR1/AFBs with different interaction strengths, and the functional specificity of AUX/IAAs was related to their ability to bind TIR1/AFBs, thus promoting the development of specific higher plant organs. Verification of the interactions among TIR1/AFBs and AUX/IAAs in Marchantia polymorpha and F. vesca also showed that the regulation of AUX/IAA members by TIR1/AFBs became more refined over the course of plant evolution. Conclusions: Our results indicate that specific interactions and specific gene expression patterns both contributed to the functional diversification of TIR1/AFBs and AUX/IAAs. Areca nut has been listed as one of the most addictive substances, along with tobacco, alcohol and caffeine. Areca nut contains seven psychoactive alkaloids; however, the effects of these alkaloids on embryonic development and motor behavior are rarely addressed in zebrafish embryo-larvae. Herein, we investigated the effects of exposure to three alkaloids (arecoline and secondary metabolites—arecaidine and arecoline N-oxide) on the developmental parameters, locomotive behavior, oxidative stress and transcriptome of zebrafish embryos. Zebrafish embryos exposed to different concentrations (0, 0.1, 1, 10, 100 and 1000 μM) of arecoline, arecaidine and arecoline N-oxide showed no changes in mortality and hatchability rates, but the malformation rate of zebrafish larvae was significantly increased in a dose-dependent manner and accompanied by changes in body length. Moreover, the swimming activity of zebrafish larvae decreased, which may be due to the increase in reactive oxygen species and the imbalance between oxidation and antioxidation. Meanwhile, transcriptome analysis showed that endoplasmic reticulum stress and the apoptosis p53 signaling pathway were significantly enriched after exposure to arecoline and arecoline N-oxide. However, arecaidine exposure focuses on protein synthesis and transport. These findings provide an important reference for risk assessment and early warning of areca nut alkaloid exposure. Highway bridges in coastal areas are seriously affected by the marine environment, while most of the existing test methods for bridge-reinforced concrete beams considering both corrosion and fatigue factors are carried out in an alternating manner, which cannot reflect the actual service conditions of the bridge structure. This paper focuses on an experimental study of the coupled influence of reinforcement corrosion and fatigue loading in reinforced concrete T-shaped beams. A novel loading test device that can realize the corrosion–fatigue coupling effect is designed, and then six reinforced concrete T-shaped beams are fabricated and tested. For the corrosion–fatigue coupling test beams, the variation law of beam cracks, failure modes, steel strain development law, load-deflection relationship, and fatigue life are analyzed and compared with that of the simple fatigue test beams. The test results show that the cracks of the test beam develop continuously with the fatigue loading times under the corrosion–fatigue coupling environment. The fatigue failure modes are all brittle fractures of the main steel bars, which present the shape of uneven oblique section tearing. The new testing device and approach can provide direct insights into the interaction of reinforcement corrosion and cyclic loading on the fatigue behavior of T-shaped RC beams, which can be further used to understand the long-term performance of bridge structures under complex marine environments. This work is a continuation of our studies on the determination of activity coefficients at infinite dilution ( γ i ∞ ) for organic solutes (i ) in ionic liquids (ILs). New sets of experimental data for 32 organic solutes (alkanes, alkenes, aromatics, chlorinated alkanes, alcohols, and other polar organic solutes) in trialkyl-substituted imidazolium-based IL 1-propyl-2,3-dimethylimidazolium thiocyanate ([PMMIM][SCN]) in the temperature range of 313.15–363.15 K were measured by gas–liquid chromatography (GLC). The partial molar excess enthalpies at infinite dilution ( H ¯ i E , ∞ ), entropies ( T ref S ¯ i E , ∞ ), and Gibbs free energies ( G ¯ i E , ∞ ) of organic solutes in [PMMIM][SCN] at a reference temperature T ref = 298.15 K were obtained from the γ i ∞ values. The density of [PMMIM][SCN] as a function of temperature at pressure p = 101.33 kPa was measured and gas–liquid partition coefficients ( K L ) were calculated. Moreover, the Hildebrand's solubility parameters of [PMMIM][SCN] were determined by the regular solution theory (RST) combined with Flory "combinatorial" equation. However, the mechanisms underlying the exact role of oxidative stress in ALS-associated neural degeneration are not definitively established. Oxidative stress-generated phospholipid peroxides are known to have extensive physiological and pathological consequences to tissues. Here, we discovered that the deficiency of glutathione peroxidase 4 (GPX4), an essential antioxidant peroxidase, led to the accumulation of phospholipid peroxides and resulted in a loss of motor neurons in spinal cords of ALS mice. Mutant human SOD1 G93A transgenic mice were intrathecally injected with neuron-targeted adeno-associated virus (AAV) expressing GPX4 (GPX4 -AAV) or phospholipid peroxidation inhibitor, ferrostatin-1. The results showed that impaired motor performance and neural loss induced by SOD1 G93A toxicity in the lumbar spine were substantially alleviated by ferrostatin-1 treatment and AAV-mediated GPX4 delivery. In addition, the denervation of neuron-muscle junction and spinal atrophy in ALS mice were rescued by neural GPX4 overexpression, suggesting that GPX4 is essential for the motor neural maintenance and function. In comparison, conditional knockdown of Gpx4 in the spinal cords of Gpx4 fl/fl mice triggered an obvious increase of phospholipid peroxides and the occurrence of ALS-like motor phenotype. The NAC transcription factor (TF) family is one of the largest TF families in plants, which has been widely reported in rice, maize and common wheat. However, the significance of the NAC TF family in wild emmer wheat (Triticum turgidum ssp. dicoccoides) is not yet well understood. In this study, a genome-wide investigation of NAC genes was conducted in the wild emmer genome and 249 NAC family members (TdNACs) were identified. The results showed that all of these genes contained NAM/NAC-conserved domains and most of them were predicted to be located on the nucleus. Phylogenetic analysis showed that these 249 TdNACs can be classified into seven clades, which are likely to be involved in the regulation of grain protein content, starch synthesis and response to biotic and abiotic stresses. Expression pattern analysis revealed that TdNACs were highly expressed in different wheat tissues such as grain, root, leaves and shoots. We found that TdNAC8470 was phylogenetically close to NAC genes that regulate either grain protein or starch accumulation. Overexpression of TdNAC8470 in rice showed increased grain starch concentration but decreased grain Fe, Zn and Mn contents compared with wild-type plants. Protein interaction analysis indicated that TdNAC8470 might interact with granule-bound starch synthase 1 (TdGBSS1) to regulate grain starch accumulation. Our work provides a comprehensive understanding of the NAC TFs family in wild emmer wheat and establishes the way for future functional analysis and genetic improvement of increasing grain starch content in wheat. In this article, the secure event-triggered control problem is investigated for multiagent systems (MASs) compromised by Denial-of-Service (DoS) attacks with multiple modes. With the rapid development of information technology, DoS attacks against communication topologies are more and more common in MASs with cyber-physical features. To actively reduce the influences caused by DoS attacks, a secure mode strategy is proposed to make agents switch into the secure mode when attacks occur. In this strategy, each agent accesses or terminates the secure mode with bounded time delays after DoS attacks begin or end. Based on the switched system theory, the secure mode strategy against DoS attacks is modeled as the asynchronous switching between agent models and topologies. Finally, we obtain the sufficient conditions on the frequency and duration of DoS attacks, under which the designed controller can guarantee the consensus of MASs. Perilla frutescens (L.) Britt. (Labiatae), a medicinal plant, has been widely used for the therapy of multiple diseases since about 1800 years ago. It has been demonstrated that the extracts of P. frutescens exert significant anti-inflammatory effects. In this research, two pairs of 7,7′-cyclolignan enantiomers, possessing a cyclobutane moiety, (+)/(−)-perfrancin [(+)/(−)-1] and (+)/(−)-magnosalin [(+)/(−)-2], were separated from P. frutescens leaves. The present study achieved the chiral separation and determined the absolute configuration of (±)-1 and (±)-2. Compounds (+)-1 and (−)-1 have notable anti-inflammatory effects by reducing the secretion of pro-inflammatory factors (NO, TNF-α and IL-6) and the expression of pro-inflammatory mediators (iNOS and COX-2). These findings indicate that cyclolignans are effective substances of P. frutescens with anti-inflammatory activity. Background: The outcome of patients with T4 esophageal squamous cell carcinoma (ESCC) is extremely poor. Two distinct therapeutic options are currently available for T4 esophageal cancers: neochemoradiotherapy followed by surgery (CRT-S) and definitive chemoradiotherapy (D-CRT). This study aimed to investigate the clinicopathologic characteristics of T4 ESCC in Chinese patients and compare the survival between the two therapeutic options.Methods: We retrospectively analyzed 125 patients with clinically unresectable T4 ESCC in Tianjin Medical University Cancer Institute and Hospital from January 2010 to December 2020. Overall survival (OS), progression-free survival (PFS) and associated factors were analyzed.Results: A total of 106 of 125 T4 ESCC patients were downstaged of the tumor by neoadjuvant CRT. Among 106 patients, 32 patients underwent CRT-S, and 74 patients underwent D-CRT. Patients in the CRT-S group had a higher OS (20.4 months vs. un-reached median OS, p = 0.037) and PFS (8.6 months vs. 21.0 months, p = 0.008) than those in the D-CRT group. In multivariate analysis, treatment was an independent predictor of PFS. After propensity score matching (PSM), 50 patients (CRT-S = 25; D-CRT = 25) were matched. Among these 50 patients, patients in the CRT-S group had a higher OS (15.6 months vs. un-reached median OS, p = 0.025) and PFS (7.2 months vs. 18.8 months, p = 0.026) than those in the D-CRT group. In multivariate analysis, treatment was an independent predictor for PFS.Conclusion: We demonstrated that CRT-S was superior to D-CRT for T4 ESCC patients who were downstaged by neo-CRT with respect to longer OS and PFS. Aiming at solving the trouble that digital image information is easily intercepted and tampered during transmission, we proposed a color image encryption scheme based on alternate quantum random walk and controlled Rubik's Cube transformation. At the first, the color image is separated into three channels: channel R, channel G and channel B. Besides, a random sequence is generated by alternate quantum walk. Then the six faces of the Rubik's Cube are decomposed and arranged in a specific order on a two-dimensional plane, and each pixel of the image is randomly mapped to the Rubik's Cube. The whirling of the Rubik's Cube is controlled by a random sequence to realize image scrambling and encryption. The scrambled image acquired by Rubik's Cube whirling and the random sequence received by alternate quantum walk are bitwise-XORed to obtain a single-channel encrypted image. Finally the three-channel image is merged to acquire the final encrypted image. The decryption procedure is the reverse procedure of the encryption procedure. The key space of this scheme is theoretically infinite. After simulation experiments, the information entropy after encryption reaches 7.999, the NPCR is 99.5978%, and the UACI is 33.4317%. The encryption scheme with high robustness and security has a excellent encryption effect which is effective to resist statistical attacks, force attacks, and other differential attacks. Valine-glutamine sequences (VQs) interact with WRKY transcription factors (TFs), forming VQ-WRKY protein complexes crucial for plant development and response to environmental changes. Cultivated peanut (Arachis hypogaea) is a tetraploid from A. duranensis and A. ipaensis cross. The Arachis spp. WRKY TFs have been identified, but Arachis VQs are largely unknown. This study identified VQs in A. duranensis, A. ipaensis, A. monticola, A. hypogaea cv. Fuhuasheng, A. hypogaea cv. Shitouqi, and A. hypogaea cv. Tifrunner. The study analyzed the homologous relationships between VQs in these Arachis spp. The VQ drought-tolerant genes were detected and VQ-WRKY interactions were determined in A. hypogaea cv. Tifrunner. The results showed that tetraploid Arachis spp. retained duplicated VQs, but lost ancestral VQs after allopolyploidization. The number of VQs in A. monticola, A. hypogaea cv. Fuhuasheng, and A. hypogaea cv. Shitouqi increased relative to their diploid ancestors. RNA-seq and quantitative real-time PCR experiments confirmed that three AhTVQs tolerate drought stress in A. hypogaea cv. Tifrunner. However, evidence of VQ-WRKY interaction for drought stress response is lacking in A. hypogaea cv. Tifrunner. According to the proposal of Kyoto protocol, carbon dioxide emission rights are traded as a commodity, and carbon emission trading market emerges as the times require. As the world's largest carbon emitter, China has established eight pilot markets for carbon emission trading. Selecting the closing price of eight carbon trading markets from the establishment to June 23, 2020, this paper analyzes the daily, weekly, and monthly return series data, using the first-order autoregressive process to adjust the daily income series to eliminate the weak trading market effect and then comprehensively uses the traditional variance ratio test and multiple variance ratio test to analyze the weak-form market efficiency of the eight carbon trading markets. The empirical results show that most of the carbon trading markets are non-weak-form market efficiency, and only Tianjin, Shanghai, and Hubei markets are weak-form market efficiency under the daily trading data. However, with the increase of carbon holding period, the weak-form market efficiency continues to strengthen. The effective fusion of multisource data helps to improve the performance of land cover classification. Most existing convolutional neural network (CNN)-based methods adopt an early/late fusion strategy to fuse the low-/high-level features for classification, which still has two inherent challenges: 1) the conventional convolution operation performs a weighted average operation on each pixel in the receptive field, which will reduce the discriminability of the center pixel due to the influence of the interference pixels and 2) the spatial–spectral features of the hyperspectral image (HSI), the elevation features of light detection and ranging (LiDAR), and the complementary features between the multimodal data are not fully exploited, which results in the reduction of classification accuracy. In this article, an effective multibranch feature fusion network with self- and cross-guided attention (MB2FscgaNet) is proposed for the joint classification of LiDAR and HSI. The main concern of this article is how to accurately estimate more effective spectral–spatial-elevation features and yield more effective transfer in the network. Specifically, MB2FscgaNet adopts a multibranch feature fusion architecture to fully exploit the hierarchical features from LiDAR and HSI level by level. At each level of the network, a self- and cross-guided attention (SCGA) is developed to assign a higher weight to interesting areas and channels of LiDAR and HSI feature maps to obtain refined spectral–spatial-elevation features and provide complementary information cross-guidance between LiDAR and HS. We further designed a spectral supplement module (SeSuM) to improve the discriminative ability of the center pixel. The identification and early warning of potential landslides can effectively reduce the number of casualties and the amount of property loss. At present, interferometric synthetic aperture radar (InSAR) is considered one of the mainstream methods for the large-scale identification and detection of potential landslides, and it can obtain long-term time-series surface deformation data. However, the method of identifying anomalous deformation areas using InSAR data is still mainly manual delineation, which is time-consuming, labor-consuming, and has no generally accepted criterion. In this study, a two-stage detection deep learning network (InSARNet) is proposed and used to detect anomalous deformation areas in Maoxian County, Sichuan Province. Compared with the most commonly used detection models, it is demonstrated that the InSARNet has a better performance in the detection of anomalous deformation in mountainous areas, and all of the quantitative evaluation indexes are higher for InSARNet than for the other models. After the anomalous deformation areas are identified using the proposed model, the possible relationship between the anomalous deformation areas and potential landslides is investigated. By selecting typical seismic sections to carry out detailed structural interpretation, the structural style features of the Wunansha Uplift in the South Yellow Sea basin were systematically combined, and the compressional structures (imbricate, opposite/back thrust, and Y-shaped structures), strike-slip faults (positive flower-shaped faults), and extensional normal faults (listric-shaped normal faults) were identified. On this basis, combined with the characteristics of the regional stress field and the background of deep geodynamics, the genetic mechanism and structural evolution of the structural style in the Wunansha Uplift were defined. The stress mechanism of the compressional structures originated from the initial high-speed and low-angle NW subduction of the paleo-Pacific plate during the early Yanshanian movement in the Early Jurassic. The regional strike-slip fault was mainly a positive flower structure with compression and torsion characteristics, and its stress mechanism originated from sinistral shear caused by the Early Cretaceous low-angle NNW subduction of the paleo-Pacific plate. The Tan-Lu fault in eastern China also had sinistral shear characteristics in this period. The extensional normal fault was characterized by a listric shape, which developed along the northern boundary of the Wunansha Uplift, that is, the connection between the Wunansha Uplift and the Southern Depression of the South Yellow Sea basin. The stress mechanism was derived from the transition of the paleo-Pacific plate from low-angle to high-angle subduction during the late Yanshanian movement in the Late Cretaceous. Objective. A retrospective case-control study was performed to observe the effect and clinical significance of early electroacupuncture combined with enhanced recovery after surgery (ERAS) on pain perception and dysfunction after total knee arthroplasty (TKA). Methods. About 100 patients who diagnosed with TKA from February 2019 to April 2021 were enrolled in our hospital. The patients were arbitrarily assigned into control group and study group. The former group was cured with electroacupuncture in the early stage, and the latter group was intervened on the basis of early electroacupuncture combined with the concept of ERAS. The curative effect, the time of getting out of bed for the first time after operation, the time of postoperative rehabilitation, postoperative rehabilitation cost, pain score and knee joint function score, range of motion (ROM) of knee joint, low shear of whole blood viscosity, plasma viscosity, fibrinogen level, and postoperative complications were compared. Results. There exhibited no statistical difference in clinical data. In terms of the treatment effects, there were 27 cases of markedly effective, 22 cases of effective, and 1 case of ineffective in the study group, and the total effective rate was 98.00%; in the control group, 15 cases were markedly effective, 28 cases were effective, and 13 cases were ineffective, and the total effective rate was 86.00%. Compared to the control group, the total effective rate of the study group was higher (P < 0.05). And the first time to get out of bed and the postoperative rehabilitation time in the study group were lower. Compared to the control group (10113.42 ± 524.83) yuan, the postoperative rehabilitation cost in the study group (12401.71 ± 530.77) yuan was higher. In terms of the scores of VAS and HSS, there exhibited no remarkable difference before treatment (P > 0.05). After treatment, the VAS score lessened and the HSS score augmented the study group VAS score (1.76 ± 0.28); the score in the control group was lower compared to the control group (3.45 ± 0.36), and HSS scoring (83.48 ± 11.23) points higher compared to the control group (65.82 ± 10.44) points (P < 0.05). Seven days after operation, the whole blood viscosity, plasma viscosity, and fibrinogen in the study group were lower (P < 0.05). The probability of postoperative complications was compared. In the study group, there were 2 cases of limb swelling and pain, 1 case of joint stiffness, and no swelling and pain complicated with deep venous thrombosis, and the total incidence was 6.00%. In the control group, there were 5 cases of limb swelling and pain, 3 cases of joint stiffness, and 3 cases of swelling and pain complicated with deep venous thrombosis, with a total incidence of 22.00%. Objectives: During embryonic haematopoiesis, haematopoietic stem/progenitor cells (HSPCs) develop from hemogenic endothelial cells (HECs) though endothelial to haematopoietic transition (EHT). However, little is known about how EHT is regulated in human. Here, we report that GFI1 plays an essential role in enabling normal EHT during haematopoietic differentiation of human embryonic stem cells (hESCs). Results: GFI1 deletion in hESCs leads to a complete EHT defect due to a closed chromatin state of hematopoietic genes in HECs. Mechanically, directly regulates important signaling pathways essential for the EHT such as PI3K signaling.etc. Conclutions: Together, our findings reveal an essential role of GFI1 mediated epigenetic mechanism underlying human EHT during hematopoiesis. Both short-distance association knowledge and long-distance interaction knowledge in the knowledge base contain rich semantics. When learning entity and relation representation in a knowledge base, if we can learn short-distance association knowledge and long-distance interaction knowledge at the same time, we can learn the representation method with rich semantic information and keep the original structure of the knowledge base. Among the knowledge contained in a large number of records, some knowledge reflects individual characteristics and can be called local knowledge; others reflect group characteristics and can be called global knowledge. Using different ways to learn local and global knowledge in the deep learning model will better reflect the difference between the two kinds of knowledge at the model level, and make the model have the ability to understand both individual characteristics and overall characteristics. Through layer-by-layer forward propagation and error back propagation algorithms, the entire network is gradually optimized in an "end-to-end" manner. This "end-to-end" approach leaves some means of introducing prior knowledge flexibly into the model. Although it can reduce the burden on researchers, this "data-driven" approach brings the shortcomings of poor interpretability of learning results and weak generalization ability. Combining the specific prior knowledge implicit in the data with the deep learning algorithm can optimize the algorithm in a targeted manner and avoid blind searching in the solution space, so as to obtain a model with better performance and wider use. To this end, this paper investigates combining prior knowledge with deep learning to design efficient algorithms to address the classification of English translation styles. This paper combines local knowledge with global knowledge and deep learning methods and proposes a memory neural network method combining local knowledge and global knowledge. By recording the local knowledge in the local memory module and simultaneously recording the global knowledge in the global memory module, the method effectively learns the latent information in a large number of records. This paper combines short-distance association knowledge with long-distance interaction knowledge and a distributed representation learning method based on deep learning and proposes a deep learning method combining short-distance association knowledge and long-distance interaction knowledge. On the IWSLT English translation task, experiments show that the method significantly improves translation quality, confirming that grammatical dependencies enhance attention by supplementing dependent grammatical information, resulting in more effective and richer context vectors that more accurately represent contextual situations. Additional experimental analysis showed that the model underwent careful parameter selection and analysis. By mining valuable long-distance interactive knowledge in the knowledge base and using it in the distributed representation learning of the knowledge base, while constraining the short-distance related knowledge and constraining the long-distance interactive knowledge, the learned knowledge can be used to effectively complete the knowledge base distributed representation for discovering new relations. The rapid-charging capability of lithium-ion batteries (LIBs) can effectively alleviate the load on new energy vehicles. The electrode material's structure can be severely harmed by rapid charging and discharging, necessitating the use of electrode materials that exhibit excellent performance in fast-charging. In order to enhance the rapid charging capability of LIBs while maintaining a large capacity, we utilized nanotechnology and carbon coating to create Ge-CNFs, which are rod-shaped germanium/carbon nanofibers. These Ge-CNFs serve to mitigate the volume fluctuations of germanium. Ge-CNFs-(700) demonstrated remarkable electrochemical characteristics and durability, maintaining a 795.9 mAh∙g−1 capacity even after 100 cycles at 0.1 A∙g−1, a capacity of 281.6 mAh∙g−1 capacity after 5000 cycles at 5 A∙g−1, and achieved a Coulombic Efficiency of 99.76 %. Furthermore, the Ge-CNFs-(700)/LCO full cell achieving a discharge capacity of 124 mAh∙g−1 after 100 cycles when operated at a current density of 0.2 A∙g−1. The main reason for the excellent electrochemical performance was the ability of CNFs to reduce crushing by serving as a buffer to absorb mechanical stress resulting from volume changes when lithium ions are intercalated/deintercalated. The electrospinning technique used to create a rod-shaped structure, along with its outstanding electrochemical capabilities, showcases the promise of germanium-based materials for rapid charging in electric vehicles. • Combined the advantages of nanotechnology and carbon coating to design one-dimensional rod-shaped Ge-CNFs nanofibers. • Ge-CNFs-(700) exhibited ultra-high fast-charging performance and stability, retaining 281.6 mAh∙g−1 after 5000 cycles at 5 A∙g−1. • Ge-CNFs-(700)/LCO full cell achieving an excellent discharge capacity. Estrogen receptor 1 (ESR1) has been recently demonstrated as a potential diagnostic biomarker for thoracic aortic aneurysm (TAA). However, its precise role in the progression of TAA remains unclear. In this study, TAA models were established in ApoE-knockout mice and primary mouse vascular smooth muscle cells (VSMCs) through treatment with angiotensin (Ang) II. Our findings revealed a downregulation of ESR1 in Ang II-induced TAA mice and VSMCs. Upregulation of ESR1 mitigated expansion and cell apoptosis in the mouse aorta, reduced pathogenetic transformation of VSMCs, and reduced inflammatory infiltration and oxidative stress both in vitro and in vivo. Furthermore, we identified macrophage migration inhibitory factor (MIF) as a biological target of ESR1. ESR1 bound to the MIF promoter to suppress its transcription. Artificial MIF restoration negated the mitigating effects of ESR1 on TAA. Additionally, we discovered that murine double minute 2 (MDM2) was highly expressed in TAA models and mediated protein degradation of ESR1 through ubiquitination modification. Silencing of MDM2 reduced VSMC dedifferentiation and suppressed oxidative stress. However, these effects were reversed upon further silencing of ESR1. In conclusion, this study demonstrates that MDM2 activates MIF by mediating ESR1 degradation, thus promoting VSMC dedifferentiation and oxidative stress during TAA progression. • ESR1 is downregulated in TAA and correlates with oxidative stress. • ESR1 upregulation alleviates aorta expansion and oxidative stress in TAA mice. • ESR1 upregulation alleviates Ang II-induced VSMC dedifferentiation and oxidative stress. • MDM2 mediates ESR1 protein degradation to rescue MIF transcription. • MDM2 silencing attenuates VSMC transition, whereas MIF upregulation augments it. The Fabry-Perot resonant cavity has been recognized as a crucial mechanism for confining Terahertz (THz) waves. However, the efficiency of the Fabry-Perot device has been hampered by the absence of lossless materials. In this study, we present a Fabry-Perot resonant cavity with enhanced efficiency, utilizing one-step laser-induced graphene without complex fabrication process. Firstly, the THz response of the samples induced by varying laser power and scanning velocity is systematically studied and measured through Terahertz frequency-domain spectroscopy (THz-FDS). Subsequently, Fourier transform is applied to facilitate quantitative analysis of the resonance intensity. Finally, the optimal resonance intensity of the device is achieved by dynamically adjusting the laser power and scanning velocity. Transmission lines are affected by aeolian vibration, sandstorms and acid rain, which cause structural damage and eventually lead to line breaking accidents. In this paper, the structural health monitoring technology of conductors is researched, and a structural monitoring system for conductors is designed. First, based on the structure of the ACSR (Aluminum Conductor Steel Reinforced) conductor and the dynamic equation, the relation between the natural frequencies and the structure of the conductor is obtained. Then, an online health monitoring system of the conductor structure is proposed, and key issues, such as the power supply solution of sensors, data preprocessing technology, multiple‐sensor synchronization method, are analyzed in detail. Finally, the structural health monitoring of overhead transmission lines designed in this paper is tested on 110‐kV experimental transmission lines. The absence of tumor antigens leads to a low response rate, which represents a major challenge in immune checkpoint blockade (ICB) therapy. Pyroptosis, which releases tumor antigens and damage‐associated molecular patterns (DAMPs) that induce antitumor immunity and boost ICB efficiency, potentially leads to injury when occurring in normal tissues. Therefore, a strategy and highly efficient agent to induce tumor‐specific pyroptosis but reduce pyroptosis in normal tissues is urgently required. Here, a smart tumor microenvironmental reactive oxygen species (ROS)/glutathione (GSH) dual‐responsive nano‐prodrug (denoted as MCPP) with high paclitaxel (PTX) and photosensitizer purpurin 18 (P18) loading is rationally designed. The ROS/GSH dual‐responsive system facilitates the nano‐prodrug response to high ROS/GSH in the tumor microenvironment and achieves optimal drug release in tumors. ROS generated by P18 after laser irradiation achieves controlled release and induces tumor cell pyroptosis with PTX by chemo‐photodynamic therapy. Pyroptotic tumor cells release DAMPs, thus initiating adaptive immunity, boosting ICB efficiency, achieving tumor regression, generating immunological memory, and preventing tumor recurrence. Mechanistically, chemo‐photodynamic therapy and control‐release PTX synergistically induce gasdermin E (GSDME)‐related pyroptosis. It is speculated that inspired chemo‐photodynamic therapy using the presented nano‐prodrug strategy can be a smart strategy to trigger pyroptosis and augment ICB efficiency. [ABSTRACT FROM AUTHOR]

This study employed traditional and advanced echocardiographic techniques to assess comprehensively age- and sex-related changes in cardiovascular structure and function in wildtype (WT) mice. Forty-five normal adult wildtype mice were apportioned to groups based on age and sex: 2-month (young) male or female, and 24-month (old) male or female (n = 13, 13, 13, and 6, respectively). Each underwent 2-dimensional (2D) imaging echocardiography, Doppler, tissue Doppler imaging echocardiography, and speckle-tracking echocardiography (STE) for comparison of cardiovascular structure and function parameters. Compared to the young mice, the old had significantly higher body weight (BW), and lower diastolic and mean arterial pressure. The left ventricular (LV) end-diastolic and end-systolic volumes, and left ventricular mass, were significantly higher in the old mice. Within each sex, the cardiac diastolic and systolic function parameters were comparable between the young and old. Isovolumetric relaxation time (IVRT)/diastolic time interval (DT) and the maximum drop rate of pressure in LV (− dP/dtmax) were significantly lower in the old mice, while the LV relaxation time constant (Tau) was significantly higher. Spearman's rank correlation showed a positive association between IVRT/DT and − dp/dtmax (male r = 0.663; female r = 0.639). Among the males, the maximum rise rate of pressure in LV (+ dp/dtmax), and systolic global longitudinal strains and rates (S-GLS, S-GLSR) were significantly different between the young and old. Spearman's rank correlation showed positive association between S-GLS, S-GLSR and + dp/dtmax (r = 0.709 and r = 0.499). Regarding vascular structure, the ascending aorta systolic and diastolic diameters were significantly higher in the old mice compared with the young. The male mice had progressive, age-related aortic stiffness. Ageing in mice leads to changes in cardiovascular structure and cardiac diastolic function, but systolic function is relatively well preserved in females. Changes in cardiac function and arterial stiffness were more significant in males than females. Background: Non-alcoholic fatty liver disease (NAFLD) is characterized by ectopic accumulation of triglycerides in the liver. Emerging evidence has demonstrated that lipophagy regulates lipid mobilization and energy homeostasis in the liver. Sirtuin 3 (SIRT3), a mitochondrial NAD+-dependent deacetylase, modulates the activities of several substrates involving in autophagy and energy metabolism. Honokiol (HK) is a natural lignan from the plants of Magnolia genus that exhibits potent liver protective property. Methods: AML12 was challenged with 500 μM palmitic acid and 250 μM oleic acid mixture solution to induce lipotoxicity. C57BL/6J mice were fed with a choline-deficient high fat diet (CDHFD) to generate liver steatosis. The expression of autophagy-related and AMP-activated protein kinase (AMPK) pathway proteins was evaluated by Western blotting and immunofluorescence staining. Intracellular lipid accumulation was validated by Nile red staining. Molecular docking analysis was performed on AutoDock 4.2. Results: HK (5 and 10 μM) was found to attenuate lipid accumulation through promoting SIRT3-AMPK-mediated autophagy, mainly on lipid droplets. HK had hydrophobic interaction with amino acid residues (PHE294, GLU323 and VAL324) and NAD+. Moreover, HK improved mitochondrial function to enhance lipolysis, through decreasing the acetylated long-chain acyl-CoA dehydrogenase level. In CDHFD-fed mice, HK (2.5 and 10 mg/Kg) treatment obviously prevented lipid accumulation in the liver. And co-treatment of the AMPK inhibitor, Compound C, almost abolished the above changes. Conclusions: These results suggest that HK could ameliorate lipotoxicity in hepatocytes by activating SIRT3-AMPK-lipophagy axis, which might be a potential therapeutic agent against NAFLD. Sonodynamic therapy (SDT), presenting spatial and temporal control of ROS generation triggered by ultrasound field, has attracted considerable attention in tumor treatment. However, its therapeutic efficacy is severely hindered by the intrinsic hypoxia of solid tumor and the lack of smart design in material band structure. Here in study, fine α-Fe2O3 nanoparticles armored with Pt nanocrystals (α-Fe2O3@Pt) was investigated as an alternative SDT agent with ingenious bandgap and structural design. The Schottky barrier, due to its unique heterostructure, suppresses the recombination of sono-induced electrons and holes, enabling superior ROS generation. More importantly, the composite nanoparticles may effectively trigger a reoxygenation phenomenon to supply sufficient content of oxygen, favoring the ROS induction under the hypoxic condition and its extra role played for ultrasound imaging. In consequence, α-Fe2O3@Pt appears to enable effective tumor inhibition with imaging guidance, both in vitro and in vivo. This study has therefore demonstrated a highly potential platform for ultrasound-driven tumor theranostic, which may spark a series of further explorations in therapeutic systems with more rational material design. Background: This study aimed to probe and verify aberrantly methylated and expressed genes in hepatoblastoma and to analyze their interactions with tumor immune microenvironment.Methods: Aberrantly methylated and expressed genes were obtained by comprehensively analyzing gene expression and DNA methylation profiles from GSE81928, GSE75271 and GSE78732 datasets. Their biological functions were predicted by the STRING and Metascape databases. CIBERSORT was utilized for inferring the compositions of tumor-infiltrating immune cells (TIICs) in each sample. Correlation between hub genes and immune cells was then analyzed. Hub genes were validated in hepatoblastoma tissues via western blot or immunohistochemistry. After transfection with sh-NOTUM, migration and invasion of HuH-6 and HepG2 cells were investigated. The nude mouse tumorigenesis model was constructed.Results: Totally, 83 aberrantly methylated and expressed genes were determined in hepatoblastoma, which were mainly involved in metabolic and cancer-related pathways. Moreover, their expression was liver-specific. 13 hub genes were screened, which were closely related to immune cells in hepatoblastoma tissues. Among them, it was confirmed that AXIN2, LAMB1 and NOTUM were up-regulated and SERPINC1 was down-regulated in hepatoblastoma than normal tissues. NOTUM knockdown distinctly weakened migration and invasion of HuH-6 and HepG2 cells and tumor growth in vivo.Conclusions: This study identified aberrantly methylated and expressed signatures that were in relation to immune microenvironment in hepatoblastoma. Targeting NOTUM hub gene could suppress migration and invasion of hepatoblastoma cells. Immunotherapies such as immune checkpoint blockade (ICB) and adoptive cell therapy (ACT) have revolutionized cancer treatment, especially in patients whose disease was otherwise considered incurable. However, primary and secondary resistance to single agent immunotherapy often results in treatment failure, and only a minority of patients experience long-term benefits. This review article will discuss the relationship between cancer immune response and mechanisms of resistance to immunotherapy. It will also provide a comprehensive review on the latest clinical status of combination therapies (e.g., immunotherapy with chemotherapy, radiation therapy and targeted therapy), and discuss combination therapies approved by the US Food and Drug Administration. It will provide an overview of therapies targeting cytokines and other soluble immunoregulatory factors, ACT, virotherapy, innate immune modifiers and cancer vaccines, as well as combination therapies that exploit alternative immune targets and other therapeutic modalities. Finally, this review will include the stimulating insights from the 2020 China Immuno-Oncology Workshop co-organized by the Chinese American Hematologist and Oncologist Network (CAHON), the China National Medical Product Administration (NMPA) and Tsinghua University School of Medicine. Background. SHC SH2 domain-binding protein 1 (SHCBP1), one of the members of Src homolog and collagen homolog (Shc) family, has been reported to be overexpressed in several malignant cancers and involved in tumor progression. However, the expression of SHCBP1 in nasopharyngeal carcinoma (NPC) remains unclear, and its clinical significance remains to be further elucidated. Methods. The expression of SHCBP1 mRNA in 35 pair samples of NPC and adjacent normal tissues of NPC was detected by RT-qPCR. The expression level of SHCBP1 protein and mRNA in the selected cells was detected by western blot and RT-qPCR, respectively. The effects of SHCBP1 on NPC in vitro were observed by MTT method, colony formation assay, apoptosis assay, cell cycle assay, wound healing assay, transwell migration assay, and transwell invasion assay. Results. SHCBP1 was highly expressed in clinical tissues and NPC cell lines, and SHCBP1 knockdown significantly inhibited NPC cell proliferation. Overexpression of SHCBP1 promoted NPC cell proliferation, migration, and invasion in NPC cell lines. Silencing SHCBP1 expression can delay cell cycle and inhibit cell apoptosis. Conclusion. Our results suggest that SHCBP1 may promote proliferation and metastasis of NPC cells, which represents that SHCBP1 may act as a new indicator for predicting the prognosis of NPC and a new target for clinical treatment.
